Seven plant species (lucerne, maize, oat, sugarbeet, sorghum, tomato, wheat) and 12 Pythium and Phytophthora species were used in a comparative study designed to investigate the effects of plant and oomycete inter-specific variation on zoospore encystment density and pathogenicity. Zoospores showed differential encystment behaviour and they encysted more on dicotyledonous than on monocotyledonous plants. Pythium aphanidermatum, P. deliense, and Phytophthora nicotianae were the most aggressive species. Sugarbeet was the most severely attacked plant species followed by tomato while oat plants were relatively unaffected. The relationship between zoospore encystment on roots and disease severity depended on the oomycete-plant combination. Correlation analysis between zoospore encystment density and disease severity indicated low and no significant levels (p.05) of association for most plant-oomycete combinations.  相似文献

Tomato agar and soybean agar were found to be comparable to or in some cases better than the popular V8 vegetable juice agar in supporting linear growth of Phytophthora cactorum, Phytophthora capsici, Phytophthora parasitica, Pythium aphanidermatum, and Pythium splendens; sporangium production of P. capsici, P. palmivora, and Pythium splendens; and oospore formation of P. cactorum, P. parasitica, Pythium aphanidermatum and Pythium splendens. These two media with readily accessible ingredients can be used to substitute for V8 juice agar in pathological and physiological studies of these two important groups of fungi in countries where V8 vegetable juice is not available or difficult to obtain.  相似文献

Aim:  To find sustainable alternatives to the application of synthetic chemicals for oomycete pathogen suppression.
Methods and Results:  Here, we present experiments on an Arabidopsis thaliana model system in which we studied the antagonistic properties of rhizobacterium Paenibacillus polymyxa strains towards the oomycete plant pathogens Phytophthora palmivora and Pythium aphanidermatum . We carried out studies on agar plates, in liquid media and in soil. Our results indicate that P. polymyxa strains significantly reduced P. aphanidermatum and P. palmivora colonization in liquid assays. Most plants that had been treated with P. polymyxa survived the P. aphanidermatum inoculations in soil assays.
Conclusions:  The antagonistic abilities of both systems correlated well with mycoidal substance production and not with the production of antagonistic substances from the biocontrol bacteria.
Significance and Impact of the Study:  Our experiments highlight the need to take biofilm formation and niche exclusion mechanisms into consideration for biocontrol assays performed under natural conditions.  相似文献

Burkholderia cepacia AMMDR1 is a biocontrol agent that protects pea and sweet corn seeds from Pythium damping-off in field experiments. The goal of this work was to understand the effect of B. cepacia AMMDR1 on Pythium aphanidermatum and Aphanomyces euteiches zoospore homing events and on infection of pea seeds or roots. In vitro, B. cepacia AMMDR1 caused zoospore lysis, prevented cyst germination, and inhibited germ tube growth of both oomycetes. B. cepacia AMMDR1 also reduced the attractiveness of seed exudates to Pythium zoospores to nondetectable levels. However, when present at high levels on seeds, B. cepacia AMMDR1 had little net effect on zoospore attraction, probably because it also enhanced seed exudation. Seed-applied B. cepacia AMMDR1 dramatically reduced the incidence of infection by Pythium zoospores in situ compared with an antibiosis-deficient Tn5 mutant strain. This mutant strain also decreased Pythium infection incidence to some extent, but only when the pathogen inoculum potential was low. B. cepacia AMMDR1 did not affect attraction of Aphanomyces zoospores or Aphanomyces root rot incidence. These results suggest that B. cepacia AMMDR1 controls P. aphanidermatum largely through antibiosis, but competition for zoospore-attracting compounds can contribute to the effect. Differences in suppression of Aphanomyces and Pythium are discussed in relation to differences in the ecology of the two pathogens.  相似文献

A molecular phylogeny of Phytophthora and related oomycetes   总被引:11,自引:0,他引:11  
Phylogenetic relationships among 50 Phytophthora species and between Phytophthora and other oomycetes were examined on the basis of the ITS sequences of genomic rDNA. Phytophthora grouped with Pythium, Peronospora, and Halophytophthora, distant from genera in the Saprolegniales. Albugo was intermediate between these two groups. Unlike Pythium, Phytophthora was essentially monophyletic, all but three species forming a cluster of eight clades. Two clades contained only species with nonpapillate sporangia. The other six clades included either papillate and semipapillate, or semipapillate and nonpapillate types, transcending traditional morphological groupings, which are evidently not natural assemblages. Peronospora was related to P. megakarya and P. palmivora and appears to be derived from a Phytophthora that has both lost the ability to produce zoospores and become an obligate biotroph. Three other Phytophthoras located some distance from the main Phytophthora-Peronospora cluster probably represent one or more additional genera.  相似文献

A bacterial cell that has a single polar flagellum alternately repeats forward swimming, in which the flagellum pushes the cell body, and backward swimming, in which the flagellum pulls the cell body. We have reported that the backward swimming speeds of Vibrio alginolyticus are on average greater than the forward swimming speeds. In this study, we quantitatively measured the shape of the trajectory as well as the swimming speed. The trajectory shape in the forward mode was almost straight, whereas that in the backward mode was curved. The same parameters were measured at different distances from a surface. The difference in the motion characteristics between swimming modes was significant when a cell swam near a surface. In contrast, the difference was indistinguishable when a cell swam >60 microm away from any surfaces. In addition, a cell in backward mode tended to stay near the surface longer than a cell in forward mode. This wall effect on the bacterial motion was independent of chemical modification of the glass surface. The macroscopic behavior is numerically simulated on the basis of experimental results and the significance of the phenomenon reported here is discussed.  相似文献

Video measurements were used to monitor the temperature acclimatizedswimming speeds (24 hours exposure) of 11 species of marinedinoflagellates, some represented by different clones, on atemperature gradient plate. Although the inherent variabilityamong individuals within a population under the same treatmentwas high, each species or clone could be represented by a responsescatter plot that characterized its temperature-dependent swimmingability. A curve-fitting treatment of the data demonstratedthe similarity of the swimming speed versus temperature responsesfor repetitive trials on a single clone or for different clonesand the diversity of the swimming speed versus temperature responsesamong different species. Comparisons among populations includedviable temperature range, maximum swimming speed and responsecurve shape. All species swam over a broader temperature rangethan that over which growth was detected. Maximum swimming speedwithin the measured group occurred at a cell length of -35 µgThis possible optimum in cell size may result from the hydrodynamiccharacteristics of dinoflagellate swimming. Swimming speed variationamong dinoflagellate species can influence the competitive interactionswithin the group or with other kinds of phytoplankton and canaffect predator-prey interactions with herbivores.  相似文献

Metabolic profiles from four stages of differentiation of the fungus Phytophthora palmivora were obtained by gas chromatography/mass spectrometry. The profiles showed the presence of sterols in the asexual reproduction stage of the organism, and confirmed their virtual absence from the mycelial stages. The zoospore stage was characterized by the presence of polyunsaturated fatty acids of C20 and C22 chain length. The transition from zoospore to cyst was also marked by the appearance of disaccharides and by a decrease in the amount of phosphate present. There were also distinctive shifts in the proportions and the total amounts of amino acids present, with gamma-aminobutyrate and alanine increasing as germination took place. These distinctive profiles identify some of the metabolic changes which accompany differentiation in this fungus.  相似文献

Summary The role of tubular mastigonemes in the reversal of thrust of the anterior flagellum ofPhytophthora cinnamomi was analysed using mastigoneme-specific monoclonal antibodies and immunoflu-orescence and video microscopy. Exposure of live zoospores ofP. cinnamomi to the mastigoneme-specific Zg antibodies caused alterations in the arrangement of mastigonemes on the flagellar surface and at Zg concentrations above 0.3 /ml, mastigonemes became detached from the flagellum. As a consequence of antibody binding to the mastigonemes there were concentration-dependent perturbations in zoospore swimming behaviour and anterior flagellum beat pattern. With increasing antibody concentration zoospores swam more slowly and other parameters of their swimming pattern, such as the wavelength of the swimming helix and the frequency of rotation, were also reduced. The effects of Zg antibodies were specific at two levels: control immunoglobulins or antibodies that bound to other flagellar surface components did not have an effect on motility, and Zg antibodies did not interfere with the motility of zoospores of oomycete species to which they did not bind. The effects of antibody-induced disruption of mastigoneme arrangement strongly support previous hypotheses that tubular mastigonemes are responsible for thrust reversal by the anterior flagellum, enabling it to pull the cell through the surrounding medium.  相似文献

Little is known about the behaviour patterns and swimming speed strategies of anadromous upriver migrating fish. We used electromyogram telemetry to estimate instantaneous swimming speeds for individual sockeye (Oncorhynchus nerka) and pink salmon (O. gorbuscha) during their spawning migrations through reaches which spanned a gradient in river hydraulic features in the Fraser River, British Columbia. Our main objectives were to describe patterns of individual-specific swim speeds and behaviours, identify swimming speed strategies and contrast these between sexes, species and reaches. Although mean swimming speeds did not differ between pink salmon (2.21 BL s–1) and sockeye salmon (1.60 BL s–1), sockeye salmon were over twice as variable (mean CV; 54.78) in swimming speeds as pink salmon (mean CV; 22.54). Using laboratory-derived criteria, we classified swimming speeds as sustained (<2.5 BL s–1), prolonged (2.5–3.2 BL s–1), or burst (>3.2 BL s–1). We found no differences between sexes or species in the proportion of total time swimming in these categories – sustained (0.76), prolonged (0.18), burst (0.06); numbers are based on species and sexes combined. Reaches with relatively complex hydraulics and fast surface currents had migrants with relatively high levels of swimming speed variation (e.g., high swimming speed CV, reduced proportions of sustained speeds, elevated proportions of burst speeds, and high rates of bursts) and high frequency of river crossings. We speculate that complex current patterns generated by river constrictions created confusing migration cues, which impeded a salmon's ability to locate appropriate pathways.  相似文献

We tested the impact of bacterial swimming speed on the survival of planktonic bacteria in the presence of protozoan grazers. Grazing experiments with three common bacterivorous nanoflagellates revealed low clearance rates for highly motile bacteria. High-resolution video microscopy demonstrated that the number of predator-prey contacts increased with bacterial swimming speed, but ingestion rates dropped at speeds of >25 microm s(-1) as a result of handling problems with highly motile cells. Comparative studies of a moderately motile strain (<25 microm s(-1)) and a highly motile strain (>45 microm s(-1)) further revealed changes in the bacterial swimming speed distribution due to speed-selective flagellate grazing. Better long-term survival of the highly motile strain was indicated by fourfold-higher bacterial numbers in the presence of grazing compared to the moderately motile strain. Putative constraints of maintaining high swimming speeds were tested at high growth rates and under starvation with the following results: (i) for two out of three strains increased growth rate resulted in larger and slower bacterial cells, and (ii) starved cells became smaller but maintained their swimming speeds. Combined data sets for bacterial swimming speed and cell size revealed highest grazing losses for moderately motile bacteria with a cell size between 0.2 and 0.4 microm(3). Grazing mortality was lowest for cells of >0.5 microm(3) and small, highly motile bacteria. Survival efficiencies of >95% for the ultramicrobacterial isolate CP-1 (< or =0.1 microm(3), >50 microm s(-1)) illustrated the combined protective action of small cell size and high motility. Our findings suggest that motility has an important adaptive function in the survival of planktonic bacteria during protozoan grazing.  相似文献

Using the mitochondrial DNA miniprep technique, the identity of sixteen morphologically unusual cultures allocated to Phytophthora nicotianae, Phytophthora mexicana or Phytophthora porri was determined by comparison with a library of mtDNA band patterns obtained from reference cultures. Seven cultures were identified as Phytophthora nicotianae (including those assigned to Phytophthora mexicana and Phytophthora porri), six as strains of Phytophthora palmivora with small, ovoid, weakly caducous sporangia, and one as Phytophthora citrophthora. Some cultures of P. nicotianae had a low percentage of caducous sporangia. Percentage sporangium caducity, but not sporangium L : B ratio, is considered a useful taxonomic criterion for separating species morphologically similar to Phytophthora nicotianae. One culture from tobacco in New Zealand had a highly unusual morphology and a unique DNA band pattern, but was not identifiable. One culture from Acacia mearnsii in South Africa had a unique DNA band pattern which was identical to that of an isolate from Annona squamosa from Australia previously identified as Phytophthora palmivora, the precise identity of which is still unclear. The identity of most isolates from diseased durian was found to be Phytophthora palmivora, confirming its role as the main pathogen, but P. nicotianae was also identified from this host. We studied postexercise physiology and behaviour of smallmouth bass (Micropterus dolomieu) that voluntarily ascended experimental raceways of varying length (20-50 m) against water velocities ranging from 8 to 120 cm/s. Our first objective was to link mean swimming speed to metabolism using patterns in postexercise muscle glycogen, muscle lactate, and plasma lactate. Our second objective was to examine several behavioural indices (attempt rate, success rate, and recovery time between an ascent and a subsequent attempt) and determine whether patterns in these data reflected those from the physiological measurements. Postexercise muscle glycogen and plasma lactate data suggest that smallmouth bass powered swimming speeds up to 70-80 cm/s using energy from aerobic processes. However, lactate did not begin to accumulate in the white muscle until speeds in excess of 120-130 cm/s were reached. The behavioural parameters measured did not indicate the presence of a physiological threshold at 70-80 cm/s; however, patterns in all factors changed appreciably when fish maintained speeds in excess of 120-130 cm/s. Therefore, it is clear that behaviour and physiology are tightly linked in this species and that maximum aerobic swimming capacity may not limit performance (or re-performance) during short-duration swims.  相似文献

Experiments on the swimming kinetics and behaviour of weather loach Misgurnus anguillicaudatus showed that horizontal swim speed was significantly greater than swim speeds when ascending to or descending from the water surface to gulp air. Vertical swimming speeds during ascending or descending were similar. Misgurnus anguillicaudatus swam unsteadily during vertical movements compared with horizontal movements.  相似文献

Single-strand-conformation polymorphism (SSCP) of ribosomal DNA of 29 species (282 isolates) of Phytophthora was characterized in this study. Phytophthora boehmeriae, Phytophthora botryosa, Phytophthora cactorum, Phytophthora cambivora, Phytophthora capsici, Phytophthora cinnamomi, Phytophthora colocasiae, Phytophthora fragariae, Phytophthora heveae, Phytophthora hibernalis, Phytophthora ilicis, Phytophthora infestans, Phytophthora katsurae, Phytophthora lateralis, Phytophthora meadii, Phytophthora medicaginis, Phytophthora megakarya, Phytophthora nicotianae, Phytophthora palmivora, Phytophthora phaseoli, Phytophthora pseudotsugae, Phytophthora sojae, Phytophthora syringae, and Phytophthora tropicalis each showed a unique SSCP pattern. Phytophthora citricola, Phytophthora citrophthora, Phytophthora cryptogea, Phytophthora drechsleri, and Phytophthora megasperma each had more than one distinct pattern. A single-stranded DNA ladder also was developed, which facilitates comparison of SSCP patterns within and between gels. With a single DNA fingerprint, 277 isolates of Phytophthora recovered from irrigation water and plant tissues in Virginia were all correctly identified into eight species at substantially reduced time, labor, and cost. The SSCP analysis presented in this work will aid in studies on taxonomy, genetics, and ecology of the genus Phytophthora.  相似文献

疫病是我国植胶区的主要病害。近年来,作者从云南西双版纳和广东海南岛的橡胶树和胶园土共分离出57株疫霉菌种。通过分类研究,共鉴定出4个种:恶疫霉 Phytophthoracactorum(Leb.& Cohn)Schroeter,辣椒疫霉 P.capsici Leoman,柑桔褐腐疫霉 P.citrophthora(Sm.& Sm.)Leonian,和棕榈疫霉 P.palmivora(Butl.)Butler。其中辣椒疫霉是首次在橡胶树上发现。我国橡胶树疫霉的种群结构与东南亚和南亚的有所不同,除棕榈疫霉外,其余3种在东南亚和南亚均未发现。而东南亚常见种:簇囊疫霉(P.botryosa)、橡胶疫霉(P.heveae)和蜜色疫霉(P.meadii),在我国却迄今尚未发现或有待证实。以前报道分离自胶园土壤中的芋疫霉(P.colocasiae),可能系柑桔褐腐疫霉之误。绝大多数分离物经配对培养均可产生性器官:辣椒疫霉的A~1交配型和A~2交配型大致相等;柑桔褐腐疫霉和棕榈疫霉的A~2交配型则明显多于A~1交配型。  相似文献

Starfish seminal plasma has such characteristics as higher concentration of potassium ions, higher osmolality, and lower pH compared with those of sea water. These factors independently inhibited the movement of spermatozoa. Sperm movement was recorded by taking photographs of the swimming paths under a dark field microscope. Spermatozoa either did not move or swam with decreased beat frequency in isolated seminal plasma or in the presence of certain fractions of seminal plasma obtained by gel-filtration or by partitions. In normal sea water (containing calcium ions), a low pH of less than 6 resulted in a decrease in the number (%) of swimming spermatozoa, though the speed of propulsion and the beat frequency was not affected. On the other hand, in a calcium-free sea water, a large proportion of spermatozoa moved in both low and high pH conditions, but in low pH the speed of propulsion was reduced and many spermatozoa (31 %) swam in smaller circles.  相似文献

The hyphal branching systems at the periphery of a fungal colony growing on cleared V-8 juice agar medium were analyzed quantitatively. A comparative study of Phytophthora cactorum, cinnamomi, P. megasperma, Pythium aphanidermatum, Saprolegnia ferax, Mucor hiemalis and Rhizopus homothallicus indicated that the characteristics of a hyphal branching system might be useful in taxa delineation.  相似文献
