Our previous work demonstrated that exogenous gibberellins (GAs) applications during rapid fruit growth significantly increases
sink demand and results in a larger fruit in Japanese pear. In an attempt to unravel the mechanism of increased sink demand
by applied GAs, the histology, cell wall components of the flesh, and carbon accumulation in the fruit were assessed for Japanese
pear (Pyrus pyrifolia, cultivar ‘Kousui’), as were the activities of sucrose- and sorbitol-cleaving enzymes. Our results show that most vascular
tissues occurred in core tissue with very little vascular tissue in the flesh. Application of a mixture of GA3 + GA4 in lanolin paste significantly increased the amount of ethanol-insoluble solids, e.g., total pectins, hemicellulose, and
cellulose in the cell walls. There was a significantly increased sink demand (assessed by 13C accumulation in the fruit) by the applied GAs, and this increased sink strength was closely related to increased activities
of cell wall-bound invertase in the core, neutral invertase and NAD-dependent sorbitol dehydrogenase in the flesh during rapid
fruit growth. As well, concentrations of sorbitol and sucrose in the flesh were decreased by GA application, while glucose
concentration increased. Most importantly, the fact that sink activity can be increased by GA application implies that endogenous
GAs are likely to be important modulators for sugar metabolism. Hence, selecting for genotypes with elevated GA production
in the growing fruit and increased activities of key enzymes for sugar metabolism could result in increased fruit size. 相似文献

Fruit and stem water potentials of tomato plants were measured continuously for several days using automated psychrometers. A linear voltage displacement transducer was used to simultaneously measure diameter changes on an adjacent fruit. A strong correlation was observed between the water potential gradient of the fruit and stem, and changes in fruit diameter. Fruit diameter increased when the apoplasmic water potential gradient favoured solution flow into the fruit and fruit shrinkage occurred only when the water potential gradient was inverted. Based on our data and other published data (Ehret & Ho 1986; Lee 1989a) on phloem transport in tomato, we have concluded that low stem water potentials have an immediate and direct effect on phloem turgor; reducing the driving force for sap flow into the fruit. Since fruit water potential remained relatively constant, the diurnal variation in stem water potential was sufficient to account for the correlation with changes in fruit diameter. There are consequences with respect to predicting the accumulation of dry matter in tomato fruit. 相似文献

Sugar uptake by protoplasts isolated from tomato fruit tissues during various stages of fruit growth 总被引:4,自引:0,他引:4
The uptake of radioactive glucose and sucrose by protoplasts isolated from pericarp and placenta tissues of tomato ( Lycopersicon esculentum cv. Counter) fruit was investigated in relation to the dry matter accumulation rates of these tissues. Uptake of glucose by protoplasts isolated from pericarp tissue was highest in fruit of around 20 g fresh weight or 25 days after anthesis. Sucrose uptake by pericarp protoplasts was lower than that of glucose and did not show a peak of uptake. The maximum rate of glucose uptake by protoplasts from the pericarp was at the time when the tomato fruit was accumulating dry matter at the highest rate. Glucose uptake by placenta protoplasts was lower and at a similar level as sucrose.
Protoplast uptake of glucose, but not of sucrose, was partially inhibited by (1) p -chloromercuribenzene sulphonic acid, a sulphydryl group modifier; (2) erythrosin B, an H+ -ATPase inhibitor; and (3) valinomycin, a K+ -ionophore, suggesting that membrane transport of glucose by tomato fruit sink cells may be a carrier-mediated, energy-dependent process.

Summary The paper reviews information, much of it obtained from studies using the East Malling root observation laboratories, on the growth and development of the fruit tree root system. The production of new white root varies from year-to-year, generally being highest in the early years. As trees age, woody roots constitute an increasing fraction of total root length although the contribution made by new root growth to the total root length of established trees is also affected by soil management, being higher for trees under grass than under herbicide. Soil management also affects the balance of short (lateral) to long (extension) roots; under grass there are more lateral roots.Calculation of the rate of water uptake per unit root length needed at various times in the year to meet transpirational demand, suggests that woody roots, which recent experimental work has shown to be capable of absorbing water, must be responsible for much of total water supply.Measurements of VA mycorrhizal infection in field-grown trees indicated, for part of the season, higher per cent infection in trees grown under irrigated grass than under herbicide management. It is suggested that this, which is associated with raised leaf phosphorus levels, may be due at least partly to higher numbers of lateral roots, the root type which becomes infected. The growth and functioning of the root system under field conditions depend upon the production and integration of a range of root types. 相似文献

Metabolic labeling of immature jackbean cotyledons with 14C-amino acids was used to determine the processing steps involved in the assembly of concanavalin A. Pulse-chase experiments and analyses of immunoprecipitated lectin forms indicated a complex series of events involving seven distinct species. The structural relatedness of all of the intermediate species was confirmed by two-dimensional mapping of 125I-tryptic peptides. An initial glycosylated precursor was deglycosylated and cleaved into smaller polypeptides, which subsequently reannealed over a period of 10-27 h. NH2-terminal sequencing of the abundant precursors confirmed that the intact subunit of concanavalin A was formed by the reannealing of two fragments, since the alignment of residues 1-118 and 119-237 was reversed in the final form of the lectin identified in the chase and the precursor first labeled. When the tissue was pulse-chased in the presence of monensin, processing of the glycosylated precursor was inhibited. The weak bases NH4Cl and chloroquine were without effect. Immunocytochemical studies showed that monensin treatment caused the accumulation of immunoreactive material at the cell surface and indicated that the ionophore had induced the secretion of a component normally destined for deposition within the protein bodies. Consideration of the tertiary structure of the glycosylated precursor and mature lectin showed that the entire series of processing events could occur without significant refolding of the initial translational product. Proteolytic events included removal of a peptide from the surface of the precursor molecule that connected the NH2- and COOH-termini of the mature protein. This processing activated the carbohydrate-binding activity of the lectin. The chase data suggest the occurrence of a simultaneous cleavage and formation of a peptide bond, raising the possibility that annealment of the fragments to give rise to the mature subunit involves a transpeptidation event rather than cleavage and subsequent religation. 相似文献

以单性结实的国庆1号温州蜜柑和自花结实的华农本地早橘为材料,研究了果实生长发育过程中果实不同部位的吲哚乙酸(IAA)、脱落酸(ABA)、赤霉素(GA1/3)和玉米素核苷(ZR)含量的变化。结果表明:(1)国庆1号果皮IAA、GA1/3和ZR含量在幼果阶段均相对较高,随后果皮和果肉IAA含量均趋下降,而在果实膨大期内果肉ABA和果皮、果肉GA1/3、ZR含量均出现上升峰值,果实成熟采收时果皮和果肉ABA含量均明显回升。(2)华农本地早种子、果皮和果肉IAA及其种子ABA含量均在果实膨大期内出现明显峰值,在幼果阶段至果实膨大初期内种子GA1/3和ZR含量均居较高并出现明显上升,对应的果皮、果肉4种内源激素水平均相对较低且变幅小。还就两结实类型柑橘果实生长发育与其内源IAA、ABA、GA1/3和ZR含量动态的关系进行了讨论。 相似文献

The greening of callus was achieved by modulating the medium's growth regulator concentrations under continuous light. Canavalia lineata (L.) DC. calluses formed chlorophyll when they were exposed to continuous light in the presence of benzylaminopurine and indole-3-acetic acid. Canavanine and canaline were detected in the green callus. But only canaline was detected in the white callus grown in the dark. Feedings of canaline to suspension cultures showed that the green suspended cells were capable of de novo biosynthesis of canavanine, but the white suspended cells were not. Exogeneously supplied canavanine was used to produce canaline and homoserine by the white suspended cells. Arginase activity was induced by the addition of arginine or canavanine to the medium, and canaline reductase activity was induced by the addition of canaline but not with ornithine in the white suspended cells.Abbreviations BA

Concentrations and characteristics of procyanidins and other phenolics in apples during fruit growth 总被引:3,自引:0,他引:3
Apples (Malus domestica Borkh.) of two table and two cider cultivars were collected during fruit growth and maturation from the end of cell proliferation. Concentrations of flavonoids (flavan-3-ols, dihydrochalcones and flavonols) in the fruit flesh decreased sharply between circa 35 and circa 100 days after flowering. For hydroxycinnamic acids, the decrease appeared slower. In a second experiments apples of the cider cultivars Kermerrien and Avrolles were sampled every 2 weeks from 40 days after flowering to overripeness for a detailed characterisation of polyphenol accumulation kinetics in the fruit flesh. Most polyphenol synthesis had occurred at 40 days after full bloom, though it persisted at a low (Kermerrien) to very low (Avrolles) level during all the fruit growth. All qualitative characteristics of the polyphenols were remarkably stable. The degree of polymerisation of the procyanidins increased slightly in Avrolles and decreased in Kermerrien. This was accompanied by a relative increase in procyanidin B2, while size-exclusion chromatography of Kermerrien polyphenol extracts showed the disappearance of a highly polymerised fraction. 相似文献

After fruit development has been triggered by pollination, the abscission zone (AZ) in the pedicel strengthens its adhesion to keep the fruit attached. Unpollinated flowers are shed at their respective AZs, whereas an enlargement of the same tissue is observed in pollinated flowers. After the fruit has developed and is fully ripened, shedding occurs easily at the AZ, indicating an acceleration of abscission. Cell wall degradation and synthesis may play important roles in these processes; however, little is understood. In this report, we have visualized changes in polysaccharide distribution in the AZs of pollinated versus unpollinated flowers and in the ripened fruits using immunohistochemistry. During floral abscission, a large increase was observed in LM15 labeling of xyloglucan specifically at the AZ in the abscising pedicel. LM5 and LM6 labeling of galactan and arabinan, respectively, also increased—LM5 throughout the pedicel and LM6 at the basal side of the AZ. The results suggest that xyloglucan, pectic galactan and arabinan play key roles in the abscission process. During fruit abscission, unlike in floral abscission, no AZ-specific cell wall polysaccharide deposition was observed; however, high autofluorescence was seen in the AZ of over-ripe fruit pedicels, suggesting secondary cell wall synthesis and lignification of the AZ prior to fruit abscission. 相似文献

To determine the distribution of the epidermal growth factor (EGF) receptor (EGFR) on the surface of cells responding to EGF as a chemoattractant, an EGFR-green fluorescent protein chimera was expressed in the MTLn3 mammary carcinoma cell line. The chimera was functional and easily visualized on the cell surface. In contrast to other studies indicating that the EGFR might be localized to certain regions of the plasma membrane, we found that the chimera is homogeneously distributed on the plasma membrane and becomes most concentrated in vesicles after endocytosis. In spatial gradients of EGF, endocytosed receptor accumulates on the upgradient side of the cell. Visualization of the binding of fluorescent EGF to cells reveals that the affinity properties of the receptor, together with its expression level on cells, can provide an initial amplification step in spatial gradient sensing. 相似文献

The effectiveness of the inhibitor, canavanine, was evaluated by examining its action in Canavalia ensiformis and Glycine max. Isolated roots were grown in culture tubes containing White's medium plus canavanine or arginine. A differential effect of canavanine on the incorporation of precursors of DNA, RNA, and protein was found, which is assumed to be related to the ability of the plant to utilize canavanine in reactions typically involving arginine. Canavanine was not found to affect DNA, RNA, or protein synthesis in Canavalia ensiformis, a plant in which this amino acid is synthesized naturally. In the canavanine sensitive species, Glycine max, of the same subfamily Papilionoideae, canavanine was observed to inhibit strongly DNA, RNA, and protein synthesis. A primary inhibition of the RNA synthesizing system is suggested. The data indicate the canavanine inhibitions are more complex than a simple competition with arginine in protein synthesis. 相似文献

Maximum shoot induction from stem explants ofCanavalia lineata was obtained with an agar-solidified PC medium containing 10 M benzylaminopurine and 1 M naphthaleneacetic acid. Rooting of thesein vitro produced shoots was achieved with hormone-free PC medium. Canavanine was produced almost exclusively in the leaves and was not detected in the roots ofin vitro propagatedC. lineata. To exclude the possibility of imminent translocation of canavanine from the root to leaf, adventitious roots were induced from leaf explants in PC medium supplemented with 1 M kinetin and 20 M indole-3-acetic acid and subcultured in medium lacking growth regulators, and the roots excised from germinated seedlings were cultured in hormone-free PC medium. All the roots were incapable of accumulation of canavanine. These results suggest that leaves ofC. lineata are the possible site of canavanine synthesis. 相似文献

Seven cDNAs, designated PcExp1 to PcExp7 , encoding expansin homologues, were isolated from mature pear fruit and their expression profiles were investigated in ripening fruit and other tissues, and in response to ethylene. Accumulation of PcExp2 , - 3, - 5 and - 6 mRNA increased markedly with fruit softening and then declined at the over-ripe stage. Treatment of fruit at an early ripening stage with 1-methylcyclopropene (MCP), an inhibitor of ethylene action, suppressed ethylene biosynthesis, fruit softening and the accumulation of the expansin mRNAs. Conversely, propylene treatment at the preclimacteric stage induced accumulation of the same four expansin genes, as well as ethylene production and fruit softening. The expression patterns correlated with alteration in the rate and extent of fruit softening. The abundance of PcExp1 mRNA increased at the late expanding phase of fruit development and further increased during ripening, whereas PcExp4 mRNA levels were constant throughout fruit growth and ripening. The MCP and propylene treatments had little effect on PcExp1 and PcExp4 expression. PcExp7 was expressed in young but not mature fruit. PcExp4 and PcExp6 mRNA was also detected in flowers. The accumulation of PcExp4, -5, -6 and - 7 mRNA was more abundant in young growing tissues, but not in fully expanded tissues, suggesting roles for these genes in cell expansion. These results demonstrate that characteristically, multiple expansin genes show differential expression and hormonal regulation during pear fruit development and at least six expansins show overlapping expression during ripening. 相似文献

An expansin gene, LeExp2, was isolated from auxin-treated, etiolated tomato (Lycopersicon esculentum cv T5) hypocotyls. LeExp2 mRNA expression was restricted to the growing regions of the tomato hypocotyl and was up-regulated during incubation of hypocotyl segments with auxin. The pattern of expression of LeExp2 was also studied during tomato fruit growth, a developmental process involving rapid cell enlargement. The expression of genes encoding a xyloglucan endotransglycosylase (LeEXT1) and an endo-1, 4-beta-glucanase (Cel7), which, like LeExp2, are auxin-regulated in etiolated hypocotyls (C. Catalá, J.K.C. Rose, A.B. Bennett [1997] Plant J 12: 417-426), was also studied to examine the potential for synergistic action with expansins. LeExp2 and LeEXT1 genes were coordinately regulated, with their mRNA accumulation peaking during the stages of highest growth, while Cel7 mRNA abundance increased and remained constant during later stages of fruit growth. The expression of LeExp2, LeEXT1, and Cel7 was undetectable or negligible at the onset of and during fruit ripening, which is consistent with a specific role of these genes in regulating cell wall loosening during fruit growth, not in ripening-associated cell wall disassembly. 相似文献

Depolymerization of cell wall xyloglucan has been proposed to be involved in tomato fruit softening, along with the xyloglucan modifying enzymes. Xyloglucan endotransglucosylase/hydrolases (XTHs: EC 2.4.1.207 and/or EC 3.2.1.151) have been proposed to have a dual role integrating newly secreted xyloglucan chains into an existing wall-bound xyloglucan, or restructuring the existing cell wall material by catalyzing transglucosylation between previously wall-bound xyloglucan molecules. Here, 10 tomato (Solanum lycopersicum) SlXTHs were studied and grouped into three phylogenetic groups to determine which members of each family were expressed during fruit growth and fruit ripening, and the ways in which the expression of different SlXTHs contributed to the total XET and XEH activities. Our results showed that all of the SlXTHs studied were expressed during fruit growth and ripening, and that the expression of all the SlXTHs in Group 1 was clearly related to fruit growth, as were SlXTH12 in Group 2 and SlXTH6 in Group 3-B. Only the expression of SlXTH5 and SlXTH8 from Group 3-A was clearly associated with fruit ripening, although all 10 of the different SlXTHs were expressed at the red ripe stage. Both total XET and XEH activities were higher during fruit growth, and decreased during fruit ripening. Ethylene production during tomato fruit growth was low and experienced a significant increase during fruit ripening, which was not correlated either with SlXTH expression or with XET and XEH activities. We suggest that the role of XTH during fruit development could be related to the maintenance of the structural integrity of the cell wall, and the decrease in XTHs expression, and the subsequent decrease in activity during ripening may contribute to fruit softening, with this process being regulated through different XTH genes. 相似文献
