Diamondback moth, Plutella xylostella, larvae were infected with a primary pathogen, Bacillus thuringiensis kurstaki (Btk) in single strain and mixed infections. Mixed infections comprised Btk and a non-pathogenic isolate, either Bacillus thuringiensis tenebrionis (Btt) or Bacillus cereus (Bc). All strains reproduced in larval cadavers, but there was evidence of competition between different isolates within hosts. Non-pathogenic isolates (Btt, Bc) had growth rates that were faster than Btk in vivo, whereas Btk outcompeted Btt in vitro. Passage through insects increased the in vitro competitive ability of Btk against Btt.  相似文献

A set of DNA markers was developed that successfully identifies Bacillus thuringiensis ssp. kurstaki (Btk) when screened against other Bacillus species and subspecies. These subspecies-specific primer sets allowed detection and characterization of Btk within an environmental background that contained many Bacillus species. Because Btk is used as an active ingredient in many commercial formulations, yet is not naturally widely distributed in North America or Europe, these markers will prove useful in investigations on the environmental persistence and ecological fate of Btk.  相似文献

The cotton bollworm Helicoverpa armigera is one of the most devastating insect pests. A set of protease enzymes allows this species to feed on different host plant species. Control measures in agriculture often involve the application of the pathogenic bacterium Bacillus thuringiensis subsp. kurstaki (Btk). In the present study, the effects of sublethal Btk doses are evaluated with respect to the food utilization indices and proteolytic activities of Helicoverpa armigera. Accordingly, the H. armigera larvae are fed with artificial diet containing sublethal Btk doses (LC5, LC10, LC15, LC20 and LC25) and a Btk‐free diet as control. All but one of the food utilization indices we measured is observed to increase significantly with increasing Btk doses. By contrast, the specific activity of total protease, chymotrypsin and elastase enzymes decrease significantly with an increasing Btk concentration. We conclude that Btk was not toxic to H. armigera larvae and any damage that it causes can be compensated for by H. armigera larvae via various mechanisms. In conclusion, increased nutritional indices in the larvae fed with Btk diet represent an important issue that needs to be considered to avoid the pest establishing Bt resistant populations. Meanwhile, the lack of effect of Btk sublethal concentrations on trypsin enzyme specific activity can bolster this challenge.  相似文献

Fomites are known to play a role in the transmission of pathogens. Quantitative analysis of the parameters that affect sample recovery efficiency (SRE) at the limit of detection of viruses on fomites will aid in improving quantitative microbial risk assessment (QMRA) and infection control. The variability in SRE as a function of fomite type, fomite surface area, sampling time, application media, relative humidity (rH), and wetting agent was evaluated. To quantify the SRE, bacteriophage P22 was applied onto fomites at average surface densities of 0.4 ± 0.2 and 4 ± 2 PFU/cm2. Surface areas of 100 and 1,000 cm2 of nonporous fomites found in indoor environments (acrylic, galvanized steel, and laminate) were evaluated with premoistened antistatic wipes. The parameters with the most effects on the SRE were sampling time, fomite surface area, wetting agent, and rH. At time zero (the initial application of bacteriophage P22), the SRE for the 1,000-cm2 fomite surface area was, on average, 40% lower than that for the 100-cm2 fomite surface area. For both fomite surface areas, the application medium Trypticase soy broth (TSB) and/or the laminate fomite predominantly resulted in a higher SRE. After the applied samples dried on the fomites (20 min), the average SRE was less than 3%. A TSB wetting agent applied on the fomite improved the SRE for all samples at 20 min. In addition, an rH greater than 28% generally resulted in a higher SRE than an rH less than 28%. The parameters impacting SRE at the limit of detection have the potential to enhance sampling strategies and data collection for QMRA models.  相似文献

We conducted laboratory experiments to examine the effects of single versus double exposures of spruce budworm, Choristoneura fumiferana (Lepidoptera: Tortricidae) female larvae to various concentrations of a Bacillus thuringiensis variety kurstaki (Btk) commercial formulation (Foray 48B). Our main objective was to document the vulnerability to Btk and the sublethal responses of fifth-instar larvae that survived from a first ingestion of Btk during their fourth stadium and to compare them with insects treated either during their fifth or fourth stadium only. As reported in the literature, fifth-instar larvae were more vulnerable than fourth-instar larvae, but only at low and medium concentrations. Fifth-instar larvae that had survived Btk ingestion during their fourth stadium were more vulnerable to a high concentration of Btk and had a shorter feeding inhibition period than those that had not been exposed during their fourth stadium. Compared with a single treatment at the fourth stadium, a double exposure to Btk further reduced the population by 20-30%, depending on the concentration applied. The second treatment also induced another feeding inhibition period and increased larval development time by 14%. The impact of the different treatments on pupal weight depended on whether treated insects exhibited supernumerary instars. In the absence of developmental polymorphism, a higher concentration, a late, or a double exposure to Btk significantly reduced pupal weight.  相似文献

In a 2-yr study, we investigated the efficacy of large-scale application of the biological insecticide Bacillus thuringiensis variety aizawai x kurstaki (Btk) in a Swedish spruce, Picea abies L. (Karst.), seed orchard for controlling damage caused by four lepidopteran species: Dioryctria abietella Den et. Schiff. (Pyralidae), Eupithecia abietaria G?tze, E. analoga Djakonov (Geometridae), and Cydia strobilella (L.) (Tortricidae). The frequencies of these species, and Strobilomyia anthracina Czerny (Diptera: Anthomyiidae), were regularly monitored throughout the vegetative growth season to map their temporal distribution patterns and to quantify occurrences of species that may have been present in the cones at some stage during the season but migrated before the final sampling. This investigation revealed that E. abietaria occurred in similar numbers to D. abietella and has probably been overlooked as a potentially serious pest in spruce seed orchards in Sweden. To determine the number, timing, and rate of Btk required to control the lepidopterans, spraying was conducted at different phases of flowering and cone development, and three rates of Btk were applied. The Btk treatment reduced cone damage caused by D. abietella and Eupithecia spp. by one-half in 2002, a year with an intermediate number of cones, but the effect was weaker in 2003, when the cone crop was low. Damage caused by C. strobilella was not affected by the treatment. The different rates of Btk application did not affect the results, and repeated spraying seemed to be effective during 2002 but not in 2003.  相似文献

Bacillus thuringiensis (Bt) is an efficient entomopathogen used for pest control. While Bt pathogenesis has been thoroughly studied, less is known about the biological effects of sublethal Bt exposure. To address this knowledge gap, we document sublethal effects of Bacillus thuringiensis var. kurstaki (Btk) on the biology and development of Helicoverpa armigera (Hübner) under laboratory conditions. The results obtained revealed that duration of the different life stages in treated neonates of H. armigera was significantly affected by sublethal treatments. Furthermore, fecundity was also negatively affected in female moths developed from Btk‐treated neonates, with the rate of egg hatchability reaching zero in the LC25. We also present data supporting that the effect of sublethal Btk concentrations could carry over to the next generation. Sublethal Btk concentrations reduced the net reproduction rate (R0), and there were also significant differences among the values of this parameter at all treatments tested when compared with controls. The intrinsic and finite rates of increase (rm and λ, respectively) were significantly lower in insects treated with sublethal Btk concentrations compared with control insects. Consequent with the reduce rate of development observed for H. armigera treated with Btk, the mean generation time (T) and doubling time (DT) were significantly higher in insects exposed to any Btk concentration tested compared with control. Our data demonstrate significant fitness costs resulting from sublethal Btk exposure in H. armigera. These observations underline the importance of considering the role of sublethal effects when attempting to evaluate the impact of Bt treatment on an insect pest population and its progeny.  相似文献

Bacillus thuringiensis subspecies kurstaki (Btk) and subspecies berliner (Btb) both produce lepidopteran-specific larvicidal protoxins with different activities against the same insect species. Toxic activity resides in the amino-terminal half of both protoxins, whereas the carboxy-terminal half of the molecules is not required for toxicity. The protoxins are 90% homologous, with a major cluster of differences in the amino-terminal half, and a 26 consecutive amino-acid insertion within the carboxy-terminal half of the Btk protoxin. Protoxin chimeras composed of the amino-terminal half of one subspecies and the carboxy-terminal half of the other were generated. Wild-type and chimeric protoxins were compared in bioassays against tobacco hornworm larvae. The amino-terminal half, the toxin itself, dictates specific larvicidal activity.  相似文献

The bacterium Bacillus thuringiensis (Bt) Berliner and the braconid wasp Habrobracon hebetor Say are valuable biocontrol agents attacking larval stages of Helicoverpa armigera (Hübner). Little information is available regarding combination of these biocontrol agents during integrated management of H. armigera. To address this knowledge gap, we document sublethal effects of Bacillus thuringiensis var. kurstaki (Btk) on the H. hebetor attacking H. armigera larvae infected with Btk. The results revealed that the duration of different life stages and fecundity of H. hebetor was significantly affected by sublethal treatments with Btk. We also present data supporting that sublethal concentrations of Btk could adversely affect life table parameters of H. hebetor. Sublethal treatments reduced the net reproductive rate (R 0) and there were also significant differences among the values of this parameter at all treatments tested. The intrinsic (r m ) and finite (λ) rates of increase were also significantly lower in parasitoid wasps reared on the treated larvae of H. armigera compared to control. These findings will be useful to develop appropriate strategies for assessing the risks of Btk to the parasitoids and safe deployment of both organisms in integrated pest management programs for sustainable crop production.  相似文献

Biomonitoring is an appropriate tool for assessing the levels of atmospheric pollution, having several advantages compared with the use of direct measurements of contaminants (e.g., in airborne particulate matter, atmospheric deposition, precipitation), related primarily to the permanent and common occurrence of the chosen organisms in the field, the ease of sampling, and trace element accumulation. Furthermore, biomonitors may provide a measure of integrated exposure over an extended period of time and are present in remote areas and no expensive technical equipment is involved in collecting them. They accumulate contaminants over the exposure time and concentrate them, thus facilitating analytical measurements. Based on largescale biomonitoring surveys, polluted areas can be identified, and by applying appropriate statistical tools, information can be obtained on the type of pollution sources and on the transboundary transport of atmospheric pollutants. The International Atomic Energy Agency is including the research on biomonitors in its projects on health-related environmental studies. Biomonitoring activities from several coordinated research projects on air pollution are presented, and results from an international workshop are discussed. In addition, activities in supporting improvement quality in the participating laboratories are outlined.  相似文献

The DT40 cell-line has been extensively used to create deletion mutants, one of which lacks Bruton’s tyrosine kinase (Btk). Btk is a cytoplasmic tyrosine kinase important for B-lymphocyte maturation. It was previously shown that there are differences in gene expression between wild-type and Btk-deficient animals. Global gene expression profiling of the avian B-lymphoma DT40 cell-line was used as a model to differentiate among Btk knockout (KO) and Btk KO cells reconstituted with human Btk. Differences in the gene expression pattern showed statistically significant changes between parental DT40 and all the Btk KO cell populations irrespective of whether they are reconstituted or not. These results imply that in the process of generating a knockout cell-line, sub-clones are selected, which have multiple changes in their gene expression pattern (p < 0.01). Although other parameters could also influence the expression profile, this potentially has important implications when interpreting microarray data from gene-deleted cell-lines.  相似文献

Liquid suspensions and dry formulations of a granulovirus (family Baculoviridae, genus Granulovirus, PoGV) derived from infected larvae and the bacterium Bacillus thuringiensis subsp. kurstaki (Berliner) (Btk) were evaluated for control of the potato tuberworm, Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae), in stored tubers. Laboratory bioassays at 25 degrees C showed that both PoGV and a wettable powder (WP) formulation of Btk incorporated with carriers (water, talc, sand, diatomaceous earth, and kaolin clay), were effective against neonate larvae. Depending on the technique, 100% larval mortality was achieved at concentrations as low as 0.025 larval equivalents (LE) PoGV per kg tuber and 150 mg Btk WP per kg tuber. However, 100% mortality was never achieved with tests on preinfested tubers, ostensibly due to the higher dosage required to kill older instars inside tubers. The most effective PoGV formulations were dipping (water) and talc, with dipping most effective for postinfestation treatments, causing up to 91.6% mortality at 0.4 LE per kg. There was no significant effect of formulation in the Btk treatments. The protective effects of residues were also evaluated under longer-term storage conditions. Batches of tubers treated with PoGV or Btk via dipping (up to 0.1 LE and 150 mg WP per kg tuber) were stored in cages containing an initial potato tuberworm infestation (10% of tubers). Although potato tuberworm populations were reduced by up to 98.4% after 2 mo at 25 degrees C, no treatments prevented the development and reproduction of the F1 generation. The sprouting of stored tubers seemed to be a limiting factor for sustained control. No significant treatment effects were detected in similar cages held at 12 degrees C for 4.5 mo. Improved strategies for the application of PoGV and Btk for long-term potato tuberworm control in tuber stores, including the use of chemical sprout suppressants, are discussed.  相似文献

Transgenic plants that produce pesticidal proteins have the potential to release these products into the environment when the plants are incorporated into soil. This could result in novel exposure of soil organisms to these pesticidal proteins. There is a lack of knowledge about the fate and persistence of transgenic pesticidal products in the soil. A model system of transgenic cotton, which produces Bacillus thuringiensis kurstakiδ-endotoxin (Bt toxin), was used to address this issue. Methods were developed to quantify Btk toxin in soil and soil/plant litter by extraction of the Btk toxin with an aqueous buffer and quantification by ELISA. The highest recovery of Btk toxin from soil was obtained with a high salt, high pH buffer. In addition, for certain soil types, addition of a non-ionic detergent, Tween-20, was needed for optimal recovery. Recovery of Btk toxin from soil ranged from 60% for a low clay content, low organic matter soil to 27% for a high clay content, high organic matter soil. The limit of detection of this method is 0.5 ng of extractable toxin per g dry weight soil. The method was shown to be useful in tracking over time the persistence of both purified and transgenic Btk toxin in laboratory experiments.  相似文献

Despite tremendous advances in the therapy of rheumatoid arthritis (RA), there remains interest in oral agents that may offer benefits that are similar to, or better than, those of biologic therapies. In their paper, Chang and colleagues demonstrate the effectiveness of a Bruton tyrosine kinase (Btk) inhibitor in two models of RA. Btk inhibition impacts several pathways affecting both B-cell and macrophage activation, making it a promising target in RA. However, other kinase inhibitors have failed to transition from animal models to human therapy, so it remains to be seen whether a Btk inhibitor will have a role in the RA treatment armamentarium.  相似文献

The U.S. Department of Justice (DOJ) mail facility in Landover, Maryland, was contaminated with Bacillus anthracis spores as a result of the 2001 anthrax bioterrorism attacks through the U.S. postal system. Surface environmental sampling within the facility indicated that the contamination was due to receipt of mail that had come in contact with Bacillus anthracis spores from the source letters at the Brentwood postal facility in Washington, DC. The DOJ adopted a two-pronged approach for remediating the facility, using aqueous chlorine dioxide to decontaminate hard, nonporous surfaces and paraformaldehyde to fumigate two pieces of mail equipment. Before the start of the remediation activities, all porous materials were removed from the mail area. Since all postremediation environmental samples were negative for growth of Bacillus anthracis spores, the remediation was judged to be effective. The facility remained closed for almost 4(1/2) months. The cleanup activities took about 2(1/2) months, with source reduction activities being the most time-consuming. Of the seven facilities that performed fumigations to remediate Bacillus anthracis contamination, the DOJ mail facility was the second building to be reopened.  相似文献

Inanimate surfaces, or fomites, can serve as routes of transmission of enteric and respiratory pathogens. No previous studies have evaluated the impact of surface disinfection on the level of pathogen transfer from fomites to fingers. Thus, the present study investigated the change in microbial transfer from contaminated fomites to fingers following disinfecting wipe use. Escherichia coli (108 to 109 CFU/ml), Staphylococcus aureus (109 CFU/ml), Bacillus thuringiensis spores (107 to 108 CFU/ml), and poliovirus 1 (108 PFU/ml) were seeded on ceramic tile, laminate, and granite in 10-μl drops and allowed to dry for 30 min at a relative humidity of 15 to 32%. The seeded fomites were treated with a disinfectant wipe and allowed to dry for an additional 10 min. Fomite-to-finger transfer trials were conducted to measure concentrations of transferred microorganisms on the fingers after the disinfectant wipe intervention. The mean log10 reduction of the test microorganisms on fomites by the disinfectant wipe treatment varied from 1.9 to 5.0, depending on the microorganism and the fomite. Microbial transfer from disinfectant-wipe-treated fomites was lower (up to <0.1% on average) than from nontreated surfaces (up to 36.3% on average, reported in our previous study) for all types of microorganisms and fomites. This is the first study quantifying microbial transfer from contaminated fomites to fingers after the use of disinfectant wipe intervention. The data generated in the present study can be used in quantitative microbial risk assessment models to predict the effect of disinfectant wipes in reducing microbial exposure.  相似文献

Many of the lepidopterous insects which attack sweet corn, Zea mays L., are susceptible to insecticidal proteins produced by Bacillus thuringiensis ssp. kurstaki (Berliner) (Btk). Transgenic sweet corn expressing a synthetic cry gene for production of a Btk-insecticidal protein may provide a more environmentally acceptable means of sweet corn production. Eight transgenic sweet corn hybrids containing a synthetic gene for CryIA(b) protein production (BT11 event) were evaluated for resistance to the corn earworm, Helicoverpa zea (Boddie), and fall armyworm, Spodoptera frugiperda (J. E. Smith). Laboratory tests revealed that all Btk sweet corn hybrids were highly resistant to leaf and silk feeding by neonate 3 and 6 d old corn earworm larvae. Ear damage in the field to the Btk sweet corn hybrids caused by corn earworm was negligible. All Btk sweet corn hybrids, except Btk 95-0901, were moderately resistant to leaf and silk feeding by the fall armyworm. Survival and weight gain were reduced when neonates were fed excised whorl leaves of the Btk plants. Weight gain, but not survival, was reduced when 3- and 6-d-old fall armyworm larvae were fed excised whorl leaves of the Btk plants. Btk sweet corn hybrids appear to be ideal candidates for use in integrated pest management (IPM) programs for both the fresh and processing sweet corn markets, and their use should drastically reduce the quantity of insecticides currently used to control these pests in sweet corn. With appropriate cultural practices, it is highly unlikely that Btk sweet corn will contribute to the development of resistance to Btk proteins in these insects because of the high toxicity of the Cry proteins expressed in these sweet corn hybrids and the harvest of sweet corn ears from fields before larvae can complete development.  相似文献

The efficacy of methanolic seed extracts of Annona squamosa (As) and Prosopis juliflora (Pj) individually and in combination with spore-crystal mixture of Bacillus thuringiensis kurstaki (Btk) was tested against third instar larvae of Helicoverpa armigera. The combination of botanicals with Btk significantly decreased LC50 values in less time as compared to individual treatments. As and Pj in combination with Btk significantly affected feeding and dietary utilization of H. armigera significantly than in individual treatments. Physiological effects found in larvae fed on Btk and botanicals included extended larval and pupal durations, decreased adult longevity and fecundity, difficulty in moulting between various instars, formation of larval–pupal intermediates and abnormal pupae. As was found to have a higher interaction with Btk than Pj. Further, Btk showed strong synergistic action with As and Pj, which are the promising candidates for development of alternative bioinsecticide in IPM programs.  相似文献

A fraction of Bruton's tyrosine kinase (Btk) co-localizes with actin fibers upon stimulation of mast cells via the high affinity IgE receptor (FcepsilonRI). In this study, a molecular basis of the Btk co-localization with actin fibers is presented. Btk and other Tec family tyrosine kinases have a pleckstrin homology (PH) domain at their N termini. The PH domain is a short peptide module frequently found in signal-transducing proteins and cytoskeletal proteins. Filamentous actin (F-actin) is shown to be a novel ligand for a subset of PH domains, including that of Btk. The actin-binding site was mapped to a 10-residue region of the N-terminal region of Btk. Basic residues in this short stretch are demonstrated to be involved in actin binding. Isolated PH domains induced actin filament bundle formation. Consistent with these observations, Btk binds F-actin in vitro and in vivo. Wild-type Btk protein is in part translocated to the cytoskeleton upon FcepsilonRI cross-linking, whereas Btk containing a mutated PH domain is not. Phosphatidylinositol 3,4, 5-trisphosphate-mediated membrane translocation of Btk was enhanced in cytochalasin D-pretreated, FcepsilonRI-stimulated mast cells. These data indicate that PH domain-mediated F-actin binding plays a role in Btk co-localization with actin filaments.  相似文献

The mortality of Tuta absoluta (Meyrick) larvae that were fed on leaves of Lycopersicon hirsutum f. glabratum (PI 134417, insect resistant) and Lycopersicon esculentum (cultivar Santa Clara, susceptible) treated with Bacillus thuringiensis var. kurstaki (Btk) was evaluated. Feeding on untreated PI 134417 was detrimental to the survival of T. absoluta larvae. When Btk was applied to the two Lycopersicon plants, mortality occurred in all T. absoluta instars. Application of Btk on tomato leaves had synergistic or additive effects with the resistant genotype on larval survival. This effect was dependent on the instar at which the larvae were fed Btk-treated leaves. Delayed Btk application may cause higher insect mortality if the insects become more susceptible to the pathogen after a longer period of feeding on the resistant crop.  相似文献
