Freshwater sponges play a major role in freshwater ecological system as important filter-feeding organisms and bioindicators. There are only few data about their ecological diversity and population genetic structure available, though a deeper knowledge is needed to propose proper conservation and effective management. The aim of this study was to assess data on distribution patterns of freshwater sponges to study the connectivity of genotypes of Ephydatia fluviatilis in a river system. We sampled specimens from River-Sieg system (River Agger and River Sieg, Germany). We hypothesized that strong anthropogenic influence would cause a uniform distribution of population structures. The genetic structure of E. fluviatilis populations was analysed with a set of eleven microsatellite loci from seven locations in River-Sieg system. Besides of E. fluviatilis, three other species co-occurred (Ephydatia mülleri, Spongilla lacustris, Eunapius fragilis). In contrast to our hypothesis, we observed an overall correlation between genetic and geographic distances among populations of this sessile species, which follows a clear isolation-by-distance pattern. A significant microsatellite polymorphism and high levels of genetic divergence between populations (FST) in upstream reaches were present. These results will provide important information for conservation management of populations with limited dispersal ability in connected river systems.  相似文献

Steelhead (Oncorhynchus mykiss) populations have declined dramatically in many parts of their range in North America, most critically in Southern California, where these anadromous trout are now classified as ‘Endangered’ under the United States Endangered Species Act. The widespread introduction of hatchery rainbow trout, the domesticated freshwater resident form of the species O. mykiss, is one factor threatening the long-term persistence of native steelhead and other trout populations. To identify where native fish of coastal steelhead lineage remained, we performed a population genetic analysis of microsatellite and SNP genotypes from O. mykiss populations at the extreme southern end of their range in Southern California, USA and Baja California, Mexico. In the northern part of this region, nearly all populations appeared to be primarily descendants of native coastal steelhead. However, in the southern, more urbanized part of this region, the majority of the sampled populations were derived primarily from hatchery trout, indicating either complete replacement of native fish or a strong signal of introgression overlaying native ancestry. Nevertheless, these genetically introgressed populations represent potentially critical genetic resources for the continued persistence of viable networks of O. mykiss populations, given the limited native ancestry uncovered in this region and the importance of genetic variation in adaptation. This study elucidates the geographic distribution of native trout populations in this region, and serves as a baseline for evaluating the impacts of hatchery trout on native O. mykiss populations and the success of steelhead conservation and recovery efforts.  相似文献

We studied the invasive warty cabbage Bunias orientalis (Brassicaceae) in three geographically distinct areas. Using inter-simple sequence repeat fingerprinting, we analyzed warty cabbages, including non-native populations, from the eastern Baltic and western Siberian regions and native populations from southwestern Russia. The eastern Baltic region and western Siberia represent the two opposite directions of B. orientalis spread in climatically different zones. The genetic structures of the native and non-native B. orientalis populations were assessed through analysis of molecular variance (AMOVA) and the Bayesian clustering method and by determining the main measures of genetic diversity. AMOVA revealed considerable population differentiation in both the native and invasive ranges. Our results did not indicate a decrease in genetic diversity in the non-native populations of B. orientalis. Similar measures of genetic diversity and genetic structure were determined in the invasive populations in two geographically and ecologically distinct, non-native regions located in Europe and Asia. In both of these regions, higher genetic diversity was detected in the non-native populations than in the native region populations, which may be due to multiple introductions. However, Bayesian clustering analysis revealed slightly different sources of invasive populations in the two non-native regions. Genetic diversity patterns revealed the lack of isolation by distance between populations and confirmed the influence of anthropogenic factors on the spread of B. orientalis. The significance of native populations as germplasm resources for breeding is discussed.  相似文献

Spread of smooth cordgrass (Spartina alterniflora) in China is an exceptional example of unanticipated outcomes arising from intentional introductions. It has been proposed that in China, management strategies used to establish S. alterniflora inadvertently promoted evolutionary outcomes that have contributed to other Spartina invasions. In this study, we assessed whether S. alterniflora in China exhibits genetic signatures of mechanisms known to promote invasion success, including large founding populations, evolved self-fertility, ‘superior source ecotypes’, and post-introduction admixture. This involved comparing microsatellite genotype and chloroplast haplotype variation among Chinese populations to other invasive S. alterniflora populations as well as native range populations, inclusive of samples from all reported source areas. We found distinct signatures of source population contributions to Chinese populations, as well as evidence of post-introduction admixture, and no evidence of limitations from a genetic bottleneck. Measures of inbreeding were well below what has been found in other non-native populations that have evolved self-fertility. Differences in genetic diversity among sites were similar to latitudinal patterns in the native range, but could be attributable to introduction history. Comparisons to other invasive populations indicate that a combination of common and idiosyncratic processes have contributed to the success of S. alterniflora in China and elsewhere, with intentional introductions promoting mechanisms that accelerate rates of spread and widespread invasion.  相似文献

The freshwater pearl mussel Margaritifera margaritifera L. is one of the most endangered freshwater mussels in the world. Effective conservation of threatened species requires not only ecological, but also genetic information from the target species and populations. Since low genetic diversity can reduce the ability of a species to adapt to environmental changes, maintaining genetic diversity has been identified as one of the key elements in successful conservation programs. We examined genetic variation of the freshwater pearl mussel from the River Vuokkijoki, Karelia, Russia. We sequenced a fragment of the cytochrome c oxidase subunit I gene (COI) from 22 individuals and compared the data to 32 previously published COI sequences available in GenBank. We identified 10 different COI haplotypes in the sequenced samples, three of which had not been previously reported. Our results show that the River Vuokkijoki has high genetic diversity and suggest that the colonization of this northern freshwater pearl mussel population might have occurred from multiple and even distant refugia. Therefore, the freshwater pearl mussel population of the River Vuokkijoki is valuable for the conservation of the whole species.  相似文献

Factors promoting the invasion success of introduced populations have been receiving increased attention in studies of biological invasions. Previous reports have indicated that successful invasions may be attributable to reduced genetic diversity in the invasive species. However, there is large variation in the magnitude and direction of the impact of exotic species that have remained unexplained. Here, we present a structured meta-analysis of papers investigating the genetic diversity of native and introduced populations of exotic insects using nuclear microsatellites and mitochondrial DNA sequences. The results indicate that invasion by exotic insects had an overall reducing effect on the genetic diversity of the invading population, with nonzero effect sizes for the number of alleles (NA), observed heterozygosity (Ho), expected heterozygosity (He) and nucleotide diversity (Nd). However, when analyzing different orders (e.g., Lepidoptera, Hemiptera), the effect sizes of NA, Ho and Nd in Lepidoptera were found to bracket zero, as did the effect size of He in Hemiptera. These results suggest an asymmetric reduction in the genetic diversity of introduced populations of exotic insects, indicating diverse mechanisms underlying their successful invasion.  相似文献

Genotypic and phenotypic characterization of Bacillus spp. from polluted freshwater has been poorly addressed. The objective of this research was to determine the diversity and enzymatic potentialities of Bacillus spp. strains isolated from the Almendares River. Bacilli strains from a polluted river were characterized by considering the production of extracellular enzymes using API ZYM. 14 strains were selected and identified using 16S rRNA, gyrB and aroE genes. Genotypic diversity of the Bacillus spp. strains was evaluated using pulsed field gel electrophoresis. Furthermore, the presence of genetic determinants of potential virulence toxins of the Bacillus cereus group and proteinaceous crystal inclusions of Bacillus thuringiensis was determined. 10 strains were identified as B. thuringiensis, two as Bacillus megaterium, one as Bacillus pumilus and one as Bacillus subtilis. Most strains produced proteases, amylases, phosphatases, esterases, aminopeptidases and glucanases, which reflect the abundance of biopolymeric matter in Almendares River. Comparison of the typing results revealed a spatio-temporal distribution among B. thuringiensis strains along the river. The results of the present study highlight the genotypic and phenotypic diversity of Bacillus spp. strains from a polluted river, which contributes to the knowledge of genetic diversity of Bacilli from tropical polluted freshwater ecosystems.  相似文献

The Patagonian silverside Odontesthes hatcheri is a native fish restricted to streams and lakes of Patagonia (Argentina and Chile). Stocking programs to enhance recreational fisheries in man-made reservoirs have introduced a nonnative, closely-related species (the pejerrey O. bonariensis) in Patagonia almost a century ago, and yet little is known about the invasiveness of this species. To evaluate the impact of these introductions we analyze genetic data (microsatellite markers and mitochondrial DNA) to quantify the incidence of hybridization between these two species and assess potential effects on native population structure. Phylogeographic analyses reveal weak geographic differentiation among populations of O. hatcheri, in agreement with previous studies for other freshwater fishes in Patagonia strongly influenced by Quaternary glaciations and hydrographic basin changes since the last glaciation. However, many populations have unique genetic pools. In several areas, introductions resulted in extensive hybridization, with high frequencies of F2 and backcrossed hybrids in natural populations, and in some cases O. bonariensis has completely displaced the native populations. The negative impact of these introductions on native populations is correlated to temperature, a critical parameter in the face of global warming, suggesting that invasiveness of O. bonariensis may increase in the future. Our results advise against continuing stocking programs to preserve the integrity of natural populations of the Patagonian silverside.  相似文献

The round goby Neogobius melanostomus has recently invaded several major freshwater systems in Europe and North America. Despite numerous studies predicting an impact on native fish assemblages, few have attempted to demonstrate it. In this case study, we monitored the effect of N. melanostomus colonisation on abundance and habitat utilisation of both young-of-the-year (YOY) native fish and YOY western tubenose goby Proterorhinus semilunaris in a typical, medium-sized European river. Colonisation by N. melanostomus had no apparent effect on either native fish abundance and species richness or P. semilunaris abundance. Moreover, after colonisation, both native fish and P. semilunaris occupied similar niches (i.e. microhabitats) to those occupied before colonisation. While niche use of YOY N. melanostomus and P. semilunaris overlapped significantly, YOY native fish utilised different habitats from the gobiids. We suggest that N. melanostomus did not compete for resources with YOY fish in our study area due to lack of niche overlap and/or surplus resources. As N. melanostomus rapidly dominated the fish assemblage at our site, we further suggest that utilisation of an empty niche, rather than competitive superiority, was the main factor facilitating its success.  相似文献

The elucidation of species diversity and connectivity is essential for conserving coral reef communities and for understanding the characteristics of coral populations. To assess the species diversity, intraspecific genetic diversity, and genetic differentiation among populations of the brooding coral Seriatopora spp., we conducted phylogenetic and population genetic analyses using a mitochondrial DNA control region and microsatellites at ten sites in the Ryukyu Archipelago, Japan. At least three genetic lineages of Seriatopora (Seriatopora-A, -B, and -C) were detected in our specimens. We collected colonies morphologically similar to Seriatopora hystrix, but these may have included multiple, genetically distinct species. Although sexual reproduction maintains the populations of all the genetic lineages, Seriatopora-A and Seriatopora-C had lower genetic diversity than Seriatopora-B. We detected significant genetic differentiation in Seriatopora-B among the three populations as follows: pairwise F ST = 0.064–0.116 (all P = 0.001), pairwise G′′ST = 0.107–0.209 (all P = 0.001). Additionally, only one migrant from an unsampled population was genetically identified within Seriatopora-B. Because the peak of the settlement of Seriatopora larvae is within 1 d and almost all larvae are settled within 5 d of spawning, our observations may be related to low dispersal ability. Populations of Seriatopora in the Ryukyu Archipelago will probably not recover unless there is substantial new recruitment from distant populations.  相似文献

An emerging insight in invasion biology is that intra-specific genetic variation, human usage, and introduction histories interact to shape genetic diversity and its distribution in populations of invasive species. We explore these aspects for the tree species Paraserianthes lophantha subsp. lophantha, a close relative of Australian wattles (genus Acacia). This species is native to Western Australia and is invasive in a number of regions globally. Using microsatellite genotype and DNA sequencing data, we show that native Western Australian populations of P. lophantha subsp. lophantha are geographically structured and are more diverse than introduced populations in Australia (New South Wales, South Australia, and Victoria), the Hawaiian Islands, Portugal, and South Africa. Introduced populations varied greatly in the amount of genetic diversity contained within them, from being low (e.g. Portugal) to high (e.g. Maui, Hawaiian Islands). Irrespective of provenance (native or introduced), all populations appeared to be highly inbred (F IS ranging from 0.55 to 0.8), probably due to selfing. Although introduced populations generally had lower genetic diversity than native populations, Bayesian clustering of microsatellites and phylogenetic diversity indicated that introduced populations comprise a diverse array of genotypes, most of which were also identified in Western Australia. The dissimilarity in the distribution and number of genotypes in introduced regions suggests that non-native populations originated from different native sources and that introduction events differed in propagule pressure.  相似文献

A priority issue in ecology and biogeography is understanding the patterns in species diversity and the causal factors of their distribution, which allows the generation of information for conservation strategies. The longitudinal distribution of fishes and their relationships with environmental variables were studied in the Guayalejo-Río Tamesí system (northeastern Mexico) from February 2000 to July 2001. A total of 5918 fish were caught in 27 collections along an altitudinal gradient in the main river course, from high mountain (1500 masl) to coastal plain near Tampico. Forty-three native and five exotic species, belonging to 35 genera in 23 families, were identified. Cluster analyses identified four major fish habitats in the river system. A distinctive euryhaline marine fish habitat (1) occurs near the mouth with native and two exotic species. Two other habitats consist essentially of freshwater fish species that are distributed along the longitudinal gradient. One of these habitats (habitat 4) shows greater diversity, as per the Shannon index value, and also includes amphidromous fish, in addition to two exotic freshwater fish; the other (habitat 2) includes freshwater, euryhaline and three exotic species. The changes in the frequency of occurrence and the abundance of Gambusia vittata, Astyanax mexicanus, and Xiphophorus variatus contribute to explaining differences between these habitats. Another habitat (3) is represented by two sampling sites located near the mouth and consist of freshwater and euryhaline fish and three exotic cyprinids with broad salinity tolerance. The low abundance and richness of exotic species suggest little impact on native fish fauna in this river. The fish assemblage of the Guayalejo-Tamesí river system species changes along a longitudinal gradient with the addition, replacement and presence of indicator species. Upstream fish fauna is mostly composed of freshwater species, some of them generalists that inhabit the entire longitudinal gradient, others that are restricted to certain sites, and the remainder of species is an assemblage composed of a mixture of euryhaline freshwater and marine species near the mouth.  相似文献

Identification of population units is crucial for management and monitoring programs, especially for endangered wild species. The roughskin sculpin (Trachidermus fasciatus Heckel) is a small catadromous fish and has been listed as a second class state protected aquatic animal since 1988 in China. To achieve sustainable conservation of this species, it is necessary to clarify the existing genetic structure both between and within populations. Here, population genetic structure among eight populations of T. fasciatus were analyzed by using 16 highly polymorphic microsatellites. High levels of genetic variation were observed in all populations. All pairwise F ST estimates were significant after false discovery rate correction (overall average F ST = 0.054). Furthermore, both STRUCTURE and discriminant analysis of principal components (DAPC) analysis showed that the eight populations were grouped into six clusters. BAYESASS analysis showed generally low recent and asymmetric migration among populations. All these results suggested significant genetic structure across populations. However, there was no isolation by distance relationship among populations, likely resulting from barriers to gene flow created by habitat fragmentation. Our results highlight the need for in situ conservation efforts for T. fasciatus across its entire distribution range, through maximizing habitat size and quality to preserve overall genetic diversity and evolutionary potential.  相似文献

Geometric morphometrics and molecular methods are effective tools to study the variability of stingless bee populations and species that merit protection given their worldwide decline. Based on previous evidence of cryptic lineages within the Scaptotrigona genus, we tested the existence of multiple evolutionary lineages within the species S. mexicana and we investigated the status of S. pectoralis. By analyzing their population structure, we found differences between the Pacific and Atlantic populations of each of these species, although geometric morphometrics of the wing only confirmed these results in S. mexicana. There was a tendency towards enhanced genetic differentiation over larger distances in the Atlantic populations of both species but not in the Pacific populations. These results revealed a pattern of differentiation among evolutionary units and a specific distribution of genetic diversity within these Scaptotrigona species in Mesoamerica, suggesting the need for future taxonomic revisions, as well as activities aimed at management and conservation.  相似文献

The South African abalone (Haliotis midae) is a gastropod mollusc of economic importance. In recent years natural populations have come under considerable pressure due to overharvesting and ecological shifts. The spatial genetic structure of H. midae has been determined; however there has not been a temporal assessment of abalone population dynamics around the South African coast. Using a population genomics approach this study aimed to assess fluctuations in genetic diversity among wild and cultured South African abalone populations through time and space. Various estimates of genetic diversity and population differentiation were calculated using EST-derived SNP markers. All populations had comparable levels of genetic diversity and the long-term effective population size appears to be sufficiently large for the wild populations, despite evidence of recent bottlenecks. Population differentiation was for the most part geographically correlated, with spatial genetic structure maintained across temporal samples. Significant genetic differentiation was, however, detected among temporal samples taken from the same locality. There was evidence for comparatively small short-term effective population sizes that could explain large changes in allele frequencies due to stochastic effects. Temporal heterogeneity could also be explained by changes in selection pressures over time. H. midae populations could, therefore, be more dynamic than previously estimated and this could have implications for effective conservation and fisheries management.  相似文献

The peacock hind Cephalopholis argus (family Serranidae), locally known as ‘roi’, was introduced from French Polynesia to Hawaii in the mid-twentieth century as a food fish. However, because of its association with ciguatera fish poisoning, it is rarely fished for food. Previous research indicates that roi could have a negative impact on native reef fish assemblages because of their high densities and prey consumption rates. However, it is unclear whether roi add to the cumulative mortality of prey (predation hypothesis), or whether predation is instead compensatory (doomed surplus hypothesis). This study experimentally assessed the effects of roi on reef fish populations through a long-term (5.5 year) predator removal experiment. A Before-After-Control-Impact study design was used to assess changes in fish assemblages following the removal of roi on 1.3 ha of patch reef. Increases in the density of prey-sized fish (<15 cm TL) were observed 18 months after roi removal. However, those effects did not translate into sustained increases in prey. While increases in potential competitors, wrasses (family Labridae), particularly the piscivorous ringtail wrasse Oxycheilinus unifasciatus, were observed on roi-free reefs, the fish assemblage did not diverge substantially in composition. Native reef fish appeared to resist the potential negative impacts of predation by roi, possibly through a refuge in size for some fish families. Management to protect intact fish assemblage size-structure could serve to bolster native resistance to invading species. In considering the threats facing coral reefs, and the possible solutions, roi removal alone will not likely replenish native fishery resources.  相似文献

The Chinese weevil, Hylobitelus xiaoi Zhang, is a major pest affecting the pine industry throughout southern China, but its dispersal is still poorly understood. We aimed to investigate its dispersal by assessing the genetic structure of seven geographical populations, using the distribution of two mitochondrial genes (cytochrome c oxidase subunit I and cytochrome b). A 1212-bp fragment of the two mitochondrial genes was sequenced for 70 individuals from seven geographical populations. The gene sequences included 861 conserved sites, 351 variable sites, 254 parsim-info sites, and 97 singleton sites. These polymorphic sites defined 24 haplotypes with a haplotype diversity of 0.825. Nucleotide diversity was low (0.04568). The differentiation parameter (0.619) was much greater than the coefficient of genetic differentiation (0.285). An analysis of molecular variance suggested that most of the variation was due to within-population differences (61.68%). This molecular data also demonstrated that there is significant divergence among the seven sampled populations of H. xiaoi. This is probably due to habitat fragmentation, preventing effective gene flow between sites. Our molecular evidence supports the view that the Chinese weevil is a native pest, and that outbreaks of H. xiaoi are caused by large plantations of the exotic slash pine.  相似文献

The conservation of narrow endemic species relies on accurate information regarding their population structure. Juglans hopeiensis Hu (Ma walnut), found only in Hebei province, Beijing, and Tianjin, China, is a threatened tree species valued commercially for its nut and wood. Sequences of two maternally inherited mitochondrial markers and two maternally inherited chloroplast intergenic spacers, three nuclear DNA sequences, and allele sizes from 11 microsatellites were obtained from 108 individuals of J. hopeiensis, Juglans regia, and Juglans mandshurica. Haplotype networks were constructed using NETWORK. Genetic diversity, population differentiation, and analysis of molecular variance (AMOVA) were used to determine genetic structure. MEGA was used to construct phylogenetic trees. Genetic diversity of J. hopeiensis was moderate based on nuclear DNA, but low based on uniparentally inherited mitochondrial DNA and chloroplast DNA. Haplotype networks showed that J. hopeiensis haplotypes were different than haplotypes found in J. regia and J. mandshurica. Allelic variants in nuclear genes that were shared among J. hopeiensis populations were not found in J. regia or J. mandshurica. Sampled populations of J. hopeiensis showed clear genetic structure. The maximum parsimony (MP) tree showed J. hopeiensis to be distinct from J. mandshurica but threatened by hybridization with J. regia and J. mandshurica. J. hopeiensis populations are strongly differentiated from sympatric Juglans species, but they are threatened by small population sizes and hybridization.  相似文献

The rosy bitterling, Rhodeus ocellatus, is a small freshwater fish belonging to the family Cyprinidae. This species lives in ponds where freshwater mussels are abundant and female lays eggs inside mussels. To understand whether the reproduction mode influenced the phylogeography, our study examined the genetic structure of R. ocellatus using sequences from the mitochondrial DNA (cytochrome b gene and control region) and nuclear DNA (the first intron of ribosomal protein S7 gene). In total, 213 specimens were collected from twelve populations in south of Yangtze River, including Yangtze River, Taiwan and Hainan Islands. The phylogenetic analyses based on mitochondrial and nuclear sequences both showed the lack of a population genetic structure, but all results, including the approximate Bayesian computation approaches, showed that these two markers revealed incongruent historical signals. Our study found that (1) the discordance between these two markers were accounted for admixtures by introduced; (2) the phylogeographic pattern of R. ocellatus and that of other freshwater fish were identical; (3) the reproduction mode was due to that the gene flows among populations were limited; and (4) R. ocellatus colonized in south of the Yangtze River (including) after southeast coastal districts formed and before Taiwan Island reached its present shape by comparing our results with that of the previous studies.  相似文献
