The blue wildebeest (Connochaetes taurinus) is distributed throughout southern and east Africa while the black wildebeest (Connochaetes gnou) is endemic to South Africa and was driven to near extinction in the early 1900s due to hunting pressure and disease outbreaks. Extensive translocation of both species throughout South Africa is threatening the genetic integrity of blue and black wildebeest. To effectively manage these species, genetic tools that can be used to detect hybrid individuals, identify genetically unique subpopulations and determine the levels of genetic diversity are required. In this study, 11 microsatellite markers were developed for wildebeest through next-generation sequencing. The microsatellite loci displayed 2.00–4.14 alleles, unbiased heterozygosity values ranged from 0.32 to 0.60 and observed heterozygosity values ranged from 0.26 to 0.52. The comparatively high level of polymorphism observed in the microsatellite markers indicates that these markers can contribute significantly to our knowledge of population genetic structure, relatedness, genetic diversity and hybridization in these species.  相似文献

The feeding niches and trophic ecology of two South African grazers, blue wildebeest Connochaetes taurinus and black wildebeest Connochaetes gnou, are compared using stable carbon and nitrogen isotope data from feces and tooth dentine collagen. As sympatric, closely related taxa predicted to occupy similar trophic positions, the blue and black wildebeest provide a good model for studying the mechanisms of coexistence and macroevolution in mammals. Data from feces collected from a single reserve in the Free State Province reveal different trophic behaviors between two herds of blue wildebeest and between both compared with a single herd of black wildebeest. These data suggest that sympatric coexistence of blue and black wildebeest is facilitated by differential niche occupation at family group or herd levels, rather than between species. However, such separation does not occur over longer time scales: results from dentine collagen support the hypothesis that the two species are indistinct in terms of trophic behavior, although blue wildebeest show more feeding flexibility, probably because of their wider habitat tolerance range. Similarities in premaxillary width of males and females of both species also suggest that both species are adapted to similar feeding styles. Thus, it is unlikely that changes in trophic behavior provided the trigger for divergence of the black from the blue wildebeest lineage in the Middle Pleistocene. We argue that the case of these two species represents an example of speciation that was not driven by resource competition, as is often assumed for many turnover events in mammalian evolution. We briefly discuss a previous suggestion that links black wildebeest evolution to their more territorial breeding behavior associated with Middle-to-Late Pleistocene landscape changes in southern Africa.  相似文献

Many ungulates, including wildebeest, seek shade and orient their bodies relative to incoming solar radiation in order to reduce environmental heat loads. Blue (Connochaetes taurinus) and black wildebeest (Connochaetes gnou), which co-exist artificially in some reserves in South Africa, are thought to adopt different thermoregulatory behaviors to mitigate high environmental heat loads. However, whether or not blue and black wildebeest use different behaviors to reduce heat loads in regions where they co-occur has never previously been examined. We compared the shade seeking and solar orientation behavior of free-ranging blue and black wildebeest in summer at three locations in South Africa where both species co-occur. We found that blue wildebeest exhibited more shade seeking behavior than did black wildebeest at all times of day, at all study sites. Black wildebeest remained in the sun but were more likely than blue wildebeest to orient their bodies parallel to the sun at all study sites, a behavior which reduces the amount of surface area exposed to incoming radiation. Black wildebeest were most likely to employ parallel solar orientation during the hottest times of the day when the sun was not directly overhead (i.e., solar noon ± 1 hour). We thus demonstrate that co-occurring blue and black wildebeest use different thermoregulatory behaviors to reduce high heat loads. It is possible that the lack of shade in the historical distribution of black wildebeest led to selective pressure for reliance on solar orientation. Differences in thermoregulatory behavior can affect species-specific heat loads, habitat use, body mass, fitness and grazing activity. Such differences may also allow blue and black wildebeest to inhabit separate microclimates within the same habitat, provided there is sufficient heterogeneity in vegetation structure, potentially facilitating reproductive isolation.  相似文献

We recorded the body axis orientation of free-living black wildebeest relative to incident solar radiation and wind. Observations were made on three consecutive days, on six occasions over the course of 1 year, in a treeless, predominantly cloudless habitat. Frequency of orientation parallel to incident solar radiation increased, and perpendicular to incident solar radiation decreased, as ambient dry-bulb temperature or solar radiation intensity increased, or wind speed decreased. We believe these changes were mediated via their effect on skin temperature. Parallel orientation behavior was more prominent when the wildebeest were standing without feeding than it was when they were feeding. We calculate that a black wildebeest adopting parallel orientation throughout the diurnal period would absorb 30% less radiant heat than the same animal adopting perpendicular orientation. Parallel orientation was reduced at times when water was freely available, possibly reflecting a shift from behavioral to autonomic thermoregulatory mechanisms. The use of orientation behavior by black wildebeest is well developed and forms part of the suite of adaptations that help them to maintain heat balance while living in a shadeless, often hot, environment.  相似文献

Rumen contents from four blue wildebeest (Connochaetes taurinus) and six black wildebeest (Connochaetes gnou) were collected from two locations during two winter culling seasons. A total of 16 species of protozoa were found in blue wildebeest with Dasytricha ruminantium, Opisthotrichum janus and Ostracodinium gracile occurring in all four animals. In black wildebeest, 23 species of protozoa were observed with only Diplodinium bubalidis (caudal spine morphotype) and Ostracodinium damaliscus being present in all animals. In the blue wildebeest, the total number of species in an individual animal varied from 9 to 11 and in the black wildebeest, the number ranged from 4 to 14. Concentrations of cells per ml ranged from 1110 to 5880 in the blue wildebeest and 3120 to 6240 in the black wildebeest. This study is the first report on protozoa species in the blue and black wildebeest. A new species of Entodinium is described, Entodinium taurinus n. sp., observed in the rumen contents of three blue wildebeest. Several physical parameters of the digestive tract were also measured, including distribution, pH and density of gastrointestinal contents in different sections of the total tract. Artificial interspecific hybrids between large scale loach P. dabryanus and tetraploid pond loach M. anguillicaudatus (Cobitidae, Cypriniformes) are viable. To detect the occurrence of possible natural hybridization, genetic analyses by using microsatellite markers were performed for natural populations of large scale loach and pond loach, the reciprocal laboratory hybrids, and “supposed hybrids” with ambiguous morphology. The fertility of the artificial hybrids was also tested. At one diagnostic microsatellite (Mac50), one out of 20 “supposed hybrids” was identified to be F1 hybrid between the two loach species because it had the same genotype as that of the laboratory hybrids. The triploid hybrids between the two species were confirmed to be female-sterile. The results show that rare hybridization has occurred between diploid large scale loach and tetraploid pond loach in nature although it may have little effect in genetic introgression. This study is helpful for fish conservation and encourages further investigation on natural hybridization and introgression of loaches.  相似文献

The nychthemeral activity patterns of a population of female black wildebeest inhabiting a shadeless environment were surveyed periodically over 1 year. The wildebeest fed mostly at night, with the proportion of feeding at night increasing when ambient conditions were hotter. Inactive periods were spent mostly lying during cooler weather but standing as days became hotter. We suggest that the entire suite of behavioural adjustments is beneficial to heat exchange with the environment. Behaviour patterns were markedly different during one warm weather survey, from the other warm weather surveys, when an 8-month dry spell had just been broken. We suggest that this may reflect the availability of water for autonomic thermoregulation, a consequent decreased reliance on behavioural thermoregulation, and a release of the thermal constraints on foraging. Our results help to explain the ability of black wildebeest to maintain body core temperature within a very narrow range despite being exposed to an environment with large nychthemeral variations in thermal conditions and offering little in the way of microclimate selection.  相似文献

Over the past few decades, use of molecular markers for species delimitation has drastically increased. Schoenoplectiella Lye has been recognized as a taxonomically difficult genus because of its morphological simplicity and frequent interspecific hybridization. The main objective of this study was to clarify the taxonomic identities of eight Schoenoplectiella species by use of molecular markers. We also evaluated the genetic relationships among S. × trapezoidea, known as a natural hybrid, and its close relatives. We used six microsatellite markers for 44 individuals from 31 natural populations of eight Schoenoplectiella species in South Korea. Six microsatellite marker combinations generated 59 amplification-detectable bands, of which 33 were specifically detected in one or more individuals of each species. Cluster analysis revealed that the grouping was consistent with the taxonomically recognized species. Our results do not support the hybrid origin of S. × trapezoidea. Rather, they suggest that this species is more closely related to S. hotarui. The informative microsatellite markers enabled us to clarify the distinctions among Schoenoplectiella species from South Korea and to identify the genetic relationships among these species. The molecular signatures found suitable for accurate identification of Schoenoplectiella species can be reliably used for studies of the phylogeny and evolution of this genus.  相似文献

Dreissenids display a high diversity of shell morphology, and it is frequently difficult to ascribe some individuals from mixed populations to one of the two species, Dreissena polymorpha (Pallas, 1771) or D. bugensis (Andrusov, 1897). Presumably, such individuals may be interspecific hybrids. We have analyzed species-specific allozyme loci of the typical representatives of these two mussel species and putative interspecific hybrids. A natural interspecific hybrid between D. polymorpha and D. bugensis was discovered for the first time by genetic methods. It has been demonstrated that D. bugensis was a maternal parent.  相似文献

The genus Prosopis (Leguminosae, Mimosoideae), comprises 44 species widely distributed in arid and semi-arid zones. Prosopis pallida (Humb. & Bonpl. ex Willd.) Kunth and P. juliflora (Sw.) DC. are the two species that are truly tropical apart from P. africana, which is native to tropical Africa (Pasiecznik et al. 2004), and they have been introduced widely beyond their native ranges. However, taxonomic confusion within the genus has hampered exploitation and better management of the species. The present study focusses primarily on evaluating the genetic relationship between Prosopis species from the section Algarobia, containing most species of economic importance, though P. tamarugo from section Strombocarpa is also included for comparison. In total, 12 Prosopis species and a putative P. pallida × P. chilensis hybrid were assessed for their genetic relationships based on RAPD markers and microsatellite transferability. The results show that P. pallida and P. juliflora are not closely related despite some morphological similarity. Evidence also agrees with previous studies which suggest that the grouping of series in section Algarobia is artificial.  相似文献

Coenagrionid damselflies are in general decline in the British Isles. Numerous factors have been implicated in the loss of these species including recent speculation that hybridisation between congeners may result in species decline. Here we use a panel of 12 microsatellite loci to examine levels of genetic divergence and the likely occurrence of hybridisation in five populations of Coenagrion puella and C. pulchellum using samples from four sites in south-east England. Coenagrion puella and C. pulchellum were highly genetically divergent, and there was no evidence of hybridisation between any of the populations examined, even where C. puella and C. pulchellum were sympatric. There was some suggestion that C. pulchellum was less genetically diverse than C. puella, though this may have been a result of ascertainment bias associated with cross-species application of microsatellite markers. We conclude that there is no evidence that hybridisation between C. puella/pulchellum could be responsible for the on-going demographic decline in C. pulchellum. Nevertheless, further genetic studies such as this one are likely to provide estimates of diversity, population structure and dispersal capacity that will be invaluable in future conservation management strategies for coenagrionid damselflies. Black poplar (Populus nigra L.) is a major species for European riparian forests but its abundance has decreased over the decades due to human influences. For restoration of floodplain woodlands, the remaining black poplar stands may act as source population. A potential problem is that P. nigra and Populus deltoides have contributed to many interspecific hybrids, which have been planted in large numbers. As these Populus x canadensis clones have the possibility to intercross with wild P. nigra trees, their offspring could establish themselves along European rivers. In this study, we have sampled 44 poplar seedlings and young trees that occurred spontaneously along the Rhine river and its tributaries in the Netherlands. Along these rivers, only a few native P. nigra L. populations exist in combination with many planted cultivated P. x canadensis trees. By comparison to reference material from P. nigra, P. deltoides and P. x canadensis, species-specific AFLP bands and microsatellite alleles indicated that nearly half of the sampled trees were not pure P. nigra but progeny of natural hybridisation that had colonised the Rhine river banks. The posterior probability method as implemented in NewHybrids using microsatellite data was the superior method in establishing the most likely parentage. The results of this study indicate that offspring of hybrid cultivated poplars compete for the same ecological niche as native black poplars. Hybridization between introduced and endemic ungulates, resulting from anthropogenic actions, has been reported for several species. Several studies of such events contain the common themes of extralimital movements, problematic phenotypic and genetic detection, and imperfect management. In southern Africa, the endemic black wildebeest (Connochaetes gnou) currently faces a serious threat of hybridization and introgression. This species survived near extinction and consequent genetic bottlenecks in the late 1800s and in the 1930s. Initiatives by private farmers followed by conservation authorities led to a dramatic recovery in numbers of this species. However, in an ironic twist, the very same advances in conservation and commercial utilisation which led to the recovery of numbers are now themselves threatening the species. Injudicious translocation has brought the species into contact with its congener, the blue wildebeest (Connochaetes taurinus), and in recent times, hybridization between the species has occurred at numerous localities in South Africa. Consequently, a significant proportion of the national black wildebeest population potentially carries a proportion of introgressed blue wildebeest genetic material. We discuss completed and ongoing attempts to find molecular markers to detect hybrids and highlight the difficulty of detecting advanced backcrosses. Additional avenues of research, such as work on morphology (cranial and postcranial elements), estimating of the probability of introgression and modelling of diffusion rates are also introduced. In addition to the difficulty in detecting hybrid animals or herds, the lack of consensus on the fate of hybrid herds is discussed. Finally, in an environment of imperfect information, we caution against implementation of management responses that will potentially induce a new genetic bottleneck in C. gnou.  相似文献

An emerging insight in invasion biology is that intra-specific genetic variation, human usage, and introduction histories interact to shape genetic diversity and its distribution in populations of invasive species. We explore these aspects for the tree species Paraserianthes lophantha subsp. lophantha, a close relative of Australian wattles (genus Acacia). This species is native to Western Australia and is invasive in a number of regions globally. Using microsatellite genotype and DNA sequencing data, we show that native Western Australian populations of P. lophantha subsp. lophantha are geographically structured and are more diverse than introduced populations in Australia (New South Wales, South Australia, and Victoria), the Hawaiian Islands, Portugal, and South Africa. Introduced populations varied greatly in the amount of genetic diversity contained within them, from being low (e.g. Portugal) to high (e.g. Maui, Hawaiian Islands). Irrespective of provenance (native or introduced), all populations appeared to be highly inbred (F IS ranging from 0.55 to 0.8), probably due to selfing. Although introduced populations generally had lower genetic diversity than native populations, Bayesian clustering of microsatellites and phylogenetic diversity indicated that introduced populations comprise a diverse array of genotypes, most of which were also identified in Western Australia. The dissimilarity in the distribution and number of genotypes in introduced regions suggests that non-native populations originated from different native sources and that introduction events differed in propagule pressure.  相似文献

Interspecific hybridization has been implicated in population declines for some waterfowl species within the mallard complex, and hybridization with mallards (Anas platyrhynchos) is currently considered the largest threat to mottled ducks (A. fulvigula), one North American member of that complex. We assessed genetic variation among 225 mottled ducks and mallards using five microsatellite loci, and detected significant overall differences between these species within two geographic areas. We characterized hybridization in Florida, where mottled ducks are endemic and mallards are beginning to appear on the breeding grounds, and in South Carolina, where mottled ducks were introduced outside their native range. We used Bayesian genetic mixture analysis in an attempt to distinguish between these closely related species. In Florida, we detected two distinct genetic groups, and 10.9% of our samples from Florida mottled ducks were inferred to have been hybrids. In contrast only 3.4% of Florida mallards were inferred to have been hybrids, suggesting asymmetric hybridization. Populations from different geographic areas within Florida exhibited hybridization rates ranging from 0% to 24%. These data indicate a genetic component would be appropriate in actively managing interspecific hybridization in Florida mottled ducks. In contrast, South Carolina mottled ducks and mallards cannot be differentiated.  相似文献

The feeding ecology of the spotted hyena Crocuta crocuta was studied in the central and eastern part of the Etosha National Park, Namibia. Hyenas mainly hunted migratory ungulates such as springbok Antidorcas marsupialis, zebra Equus burchelli, and blue wildebeest Connochaetes taurinus, but also resident species such as the greater kudu Tragelaphus strepsiceros and gemsbok Oryx gazella. There were, however, major differences in the species most frequently killed by hyenas in central and eastern Etosha. The preferred prey species of spotted hyenas in central Etosha was springbok, whereas regarding the abundance of zebra and wildebeest, these two species were rather avoided. In contrast, the prey species preferred by hyenas in eastern Etosha was kudu. Zebra and gemsbok were taken in proportion to their abundance, whereas wildebeest and springbok seemed to be rather avoided. Differences in prey selection and preferences were also reflected in differences in hunting group sizes. In eastern Etosha, where spotted hyenas frequently hunted larger prey, hunting group sizes were significantly larger compared to those in the center of the park.  相似文献

The seeds of Theobroma cacao (cacao) are the source of cocoa, the raw material for the multi-billion dollar chocolate industry. Cacao’s two most important traits are its unique seed storage triglyceride (cocoa butter) and the flavor of its fermented beans (chocolate). The genome of T. cacao is being sequenced, and to expand the utility of the genome sequence to the improvement of cacao, we are evaluating Theobroma grandiflorum, the closest economically important species of Theobroma for its potential use in a comparative genomic study. T. grandiflorum differs from cacao in important agronomic traits such as flavor of the fermented beans, disease resistance to witches’ broom and abscission of mature fruits. By comparing genomic sequences and analyzing viable inter-specific hybrids, we hope to identify the key genes that regulate cacao’s most important traits. We have investigated the utility in T. grandiflorum of three types of markers (microsatellite markers, single-strand conformational polymorphism markers and single nucleotide polymorphism (SNP) markers) developed in cacao. Through sequencing of amplicons of 12 diverse individuals of both cacao and T. grandiflorum, we have identified new intra- and inter-specific SNPs. Two markers which had no overlap of alleles between the species were used to genotype putative inter-specific hybrid seedlings. Sequence conservation was significant and species-specific differences numerous enough to suggest that comparative genomics of T. grandiflorum and T. cacao will be useful in elucidating the genetic differences that lead to a variety of important agronomic trait differences.  相似文献

Little is known about the population ecology of the recently described bottlenose dolphin species Tursiops australis. The classification of this species is still under debate, but this putative species is thought to be comprised of small and genetically distinct populations (including sub-populations under increasing anthropogenic threats) and is likely endemic to coastal southern Australia. Mitochondrial DNA (mtDNA) control region sequences and microsatellite loci were used to assess genetic variation and hierarchical population structure of coastal T. cf. australis across a range of spatial scales and environmental discontinuities between southern Western Australia (WA) and central South Australia (SA). Overall, genetic diversity was similar to that typically found for bottlenose dolphins, although very low mtDNA diversity was found in Gulf St. Vincent (GSV) dolphins. We found historical genetic subdivision and likely differences in colonisation between GSV and Spencer Gulf, outer- and inner-gulf locations, and SA/WA and previously identified Victorian/Tasmanian populations. A hierarchical metapopulation structure was revealed along southern Australia, with at least six genetic populations occurring between Esperance, WA and southern Tasmania. In addition, fine-scale genetic subdivision was observed within each SA/WA population. In general, contemporary migration was limited throughout southern Australia, but an important gene flow pathway was identified eastward along the Great Australian Bight. Management strategies that promote gene flow among populations should be implemented to assist with the maintenance of the inferred metapopulation structure. Further research into the population ecology of this species is needed to facilitate well-informed management decisions.  相似文献

Hybridization between alien and native species is biologically very important and could lead to genetic erosion of native taxa. Solidago × niederederi was discovered over a century ago in Austria and described by Khek as a natural hybrid between the alien (nowadays regarded also as invasive) S. canadensis and native S. virgaurea. Although interspecific hybridization in the genus Solidago is considered to be relatively common, hybrid nature of S. × niederederi has not been independently proven using molecular tools, to date. Because proper identification of the parentage for the hybrid Solidago individuals solely based on morphological features can be misleading, in this paper we report an additive polymorphism pattern expressed in the ITS sequences obtained from individuals representing S. × niederederi, and confirm the previous hypothesis that the parental species of this hybrid are S. canadensis and S. virgaurea. Additionally, based on variability at the cpDNA rpl32-trnL locus, we showed that in natural populations hybridization occurs in both directions.  相似文献
