The regulatory role of cyclic AMP in various cellular activities is well known. It has been documented that both the notochord and extracellular matrix materials (ECM) induce somite chrondrogenesis. We believe that the ECM modulates the intracellular cAMP level during chondrogenic differentiation. The studies indicated that notochordal induction, which resulted in somite chondrogenesis (reflected by increased sulfated glycosaminoglycan synthesis) reduced the intracellular cAMP level in somites. Addition of forskolin and dibutyryl cAMP resulted in increased intracellular cAMP levels and decreased synthesis of sulfated glycosaminoglycans (decreased chondrogenesis). In the case of dibutyryl cAMP, the inhibition of sulfated glycosaminoglycan synthesis was related to the length of exposure time. Thus, the inverse relationship between cAMP content and enhanced chondrogenesis supports the theory that, in somites, a decrease in the intracellular cAMP level may be necessary to trigger chondrogenic differentiation.  相似文献

The role of hyaluronic acid in limb morphogenesis (chondrogenesis) has been well defined. In the present study, we found that hyaluronic acid synthesis in somite explants steadily increased until day 6, then decreased, and inclusion of notochord did not accelerate the rate of synthesis. Analysis of hyaluronidase activity in the somite explants indicated an increase in the enzyme level in day-6 cultures. Again, inclusion of notochord did not alter this pattern. The decrease in hyaluronic acid after day 6 and the increase in sulfated proteoglycan synthesis from day 6 resemble the pattern described during limb development. Subsequent studies showed that, with time, the size of the hyaluronic acid synthesized by somites increased and, again, inclusion of notochord did not influence this pattern. The results indicate that unstimulated somites are capable of synthesizing cartilage-specific proteoglycans in a relatively restricted manner, and the inclusion of notochord resulted in accelerated synthesis of stable proteoglycan aggregates typical of differentiated chondrocytes. Metabolic events in somites related to hyaluronic acid are not influenced by the notochord.  相似文献

In the present study the level of cAMP was measured during in vitro chondrogenesis of wing mesenchyme of stage 24 chick embryos and was found to increase significantly from 6.3 pmol/mg protein at the end of the first day of culture to 9.7 pmol/mg protein on the second day, when chondrogenic expression is first detected by the appearance of an Alcian blue staining extracellular matrix. Nonchondrogenic cultures derived from wings of stage 19 embryos had a lower level of cAMP (4.4 +/- 0.07 pmol/mg protein). The level of cAMP in intact wings was 4.5 +/- 0.4 pmol/mg protein and did not change between stages 19 through 25. The correlatin between increased levels of cAMP and the onset of chondrogenesis is consistent with a role of cAMP in the expression of differentiated functions in chondrocytes, as well as in some other cell types.  相似文献

The present study represents a first step in investigating the possible involvement of cyclic AMP in the stimulation of somite chondrogenesis elicited by extracellular matrix components produced by the embryonic notochord. Dibutyryl cyclic AMP (db-cAMP) at 1.0 mM severely impairs “spontaneous” somite chondrogenesis, i.e., inhibits the formation of the small amount of cartilaginous matrix normally formed by embryonic somites in vitro in the absence of inducing tissues. This inhibition of cartilaginous matrix formation is reflected in a 30–40% reduction in sulfated glycosaminoglycan (GAG) accumulation. 8-Bromo-cyclic AMP also severely inhibits cartilage formation and sulfated GAG accumulation by somite explants. This impairment is limited to cyclic AMP derivatives; dibutyryl cyclic GMP, 5′-AMP, and 2′,3′-AMP have no effect. The inhibitory effect of cyclic AMP derivatives is mimicked by the cyclic AMP-phosphodiesterase inhibitor, theophylline, and potentiated by the addition of both db-cAMP and theophylline. Dibutyryl cyclic AMP and/or theophylline also inhibit the stimulation of cartilaginous matrix formation and sulfated GAG accumulation normally elicited by the embryonic notochord, reducing accumulation to a level similar to that found in somite explants without notochord. The inhibition of chondrogenesis by cyclic AMP in notochord-somite explants appears to result from an inability of somites to respond and not from an effect on the inductive capacity of the notochord, since db-cAMP has no detectable effect on the synthesis of molecules (sulfated GAG and collagen) by the notochord that have been implicated in its inductive activity. Finally, db-cAMP and/or theophylline inhibit the stimulation of somite chondrogenesis normally elicited by purified Type I collagen substrates. Dibutyryl cyclic AMP and theophylline reduce sulfated GAG accumulation by somites cultured on collagen to a level even below that accumulated by somites cultured in the absence of collagen, i.e., on Millipore filters.  相似文献

Recent studies have shown that in the developing chick embryo, at physiological level retinoic acid (RA) causes mirror-image duplication of limb skeletal elements. This has led to the suggestion that RA could be the endogenous morphogen or isgnal substance. In this study, in order to explore the effect of RA on somite chondrogenesis, we have standardized a serum-free chemically defined medium that supports the growth of somite explants in vitro. The results indicate that in somites RA at 10 ng/ml level induces cell proliferation, DNA and sulfated proteoglycan synthesis, and at higher concentrations is toxic. The results further show that RA induced stimulation of somite chondrogenesis is sclerotomal specific and the dermamyotemal portion of the somites does not exihibit a similar response. Retinoic acid also increases heparan sulfate synthesis and aggregation of isolated sclerotomal cells in culture. These results thus suggest that in amplifying chondrogenesis, RA acts at all phases such as cell proliferation (may increase cell viability) and aggregation, increased DNA synthesis and increased synthesis of matrix components. In otherwords, RA seems to initiate a chain of inter-related events.  相似文献

Anterior somites cultured with (NSA) or without (SA) notochord, and posterior somites cultured with (NSP) or without notochord (SP) were compared with respect to changes in their DNA content, their potential to synthesize the active sulfate principle phosphoadenosine phosphosulfate (PAPS), and their ability to accumulate 35S-sulfate.Chondrogenesis was observed in the NSA, NSP, and SP explants, but was rarely noted in the SA explants. A decrease in DNA content during the initial 48 hr of culture was common to all explants. After this initial decrease, DNA content increased most in those explants forming cartilage. The synthesis of PAPS by cell-free extracts of each type of somite explant also decreased during the initial period of culture. Only extracts of those explants undergoing chondrogenesis showed increases in PAPS synthesis with continued culture. Each type of somite explant accumulated 35S-sulfate into chondroitin sulfate during the first hours of culture. The non-chondrogenic SA explants accumulated little 35S-sulfate during the period of culture. At varying times after 24 hr the chondrifying explants (NSA, SP, and NSP) initiated an increased rate of accumulation of 35S-sulfate.Cartilage nodules, increases in DNA content, PAPS synthesis and 35S-sulfate accumulation occurred within the same 24 hr period, during the 2nd day in NSP explants, the 3rd day in NSA explants, and between the 3rd and 4th day for SP explants. A hypothesis of in vitro somite chondrogenesis based on differential cell viability is presented.  相似文献

In the present investigation, evidence is presented directly implicating proteoglycans produced by the embryonic notochord in the control of somite chondrogenesis. It has been demonstrated by several histochemical techniques that during the period of its interaction with somites, the notochord synthesizes perinotochordal proteoglycans, and these proteoglycans have been shown to contain chondroitin 4-sulfate (40%), chondroitin 6-sulfate (40%), and heparan sulfate (20%). Dissection of notochords from embryos with the aid of a brief treatment with trypsin results in the removal of perinotochordal extracellular matrix materials including proteoglycans, while dissection of notochords without the aid of enzyme treatment or with a low concentration of collagenase results in their retention. There is a considerable increase in the rate and amount of cartilage formation and a corresponding 2 to 3-fold increase in the amount of sulfated glycosaminoglycan accumulated by somites cultured in association with notochords dissected under conditions in which perinotochordal materials are retained. Treatment of collagenase-dissected or freely dissected notochords with highly purified enzymes (chondroitinase ABC, AC, and testicular hyaluronidase) which specifically degrade proteoglycans causes a loss of histochemically detectable perinotochordal proteoglycans. These notochords are considerably impaired in their ability to support in vitro somite chondrogenesis. In addition, when trypsin-treated notochords are cultured (“precultured”) for 24 hr on nutrient agar (in the absence of somites), perinotochordal material reaccumulates. Somites cultured in association with such “precultured” notochords exhibit considerable increase in the amount of cartilage formed and a 2- to 3-fold increase in the amount of sulfated glycosaminoglycan accumulated as compared to somites cultured in association with trypsin-treated notochords which have not been “precultured.” This observation indicates that trypsin-treated notochords reacquire their ability to maximally stimulate in vitro somite chondrogenesis by resynthesizing and accumulating perinotochordal material. Finally, “precultured” notochords treated with chondroitinase to remove perinotochordal proteoglycans are considerably impaired in their ability to support in vitro somite chondrogenesis. These observations are consonant with the concept that proteoglycans produced by the embryonic notochord play an important role in somite chondrogenesis.  相似文献

Teratogenic effects of the anticonvulsant drugs valproic acid (VPA) and diphenylhydantoin (DPH) on the development of mouse embryos during early organogenesis were studied using the whole embryo culture technique. Embryos with one to seven somites were exposed in vitro to 50-375 micrograms/ml VPA or 15-135 micrograms/ml DPH for up to 42 hours and compared to control embryos cultured in 80% rat serum without either drug. For both VPA- and DPH-treated embryos, a dose-dependent increase in the frequency of abnormal embryos and a decrease in viability were found. VPA and DPH produced a similar pattern of defects. Drug-induced anomalies included open neural tubes in the cranial regions, abnormal body curvature, craniofacial deformities, and yolk sac defects. Ultrastructural changes were noted in the neuroepithelium of exencephalic VPA-treated embryos. Growth and development were retarded in embryos exposed to greater than 35 micrograms/ml DPH or greater than 50 micrograms/ml VPA as indicated by the decrease in protein and DNA content and the reduction in somite number, crown-rump length, and yolk sac diameter. On a molar basis DPH was potentially more teratogenic than VPA, which correlates with the higher lipid solubility of DPH. With VPA, susceptibility to the drug depended on the developmental stage; e.g., at 150 micrograms/ml VPA the frequency of malformations was 70% in embryos with one to four somites as compared to 35% in embryos with five to seven somites.  相似文献

The ionophore, A23187, a drug that facilitates Ca2+ uptake leading to elevation of cytoplasmic Ca2+ levels, at concentrations of 0.25-1.0 microgram/ml severely impairs "spontaneous" somite chondrogenesis, i.e., inhibits the formation of the small amount of cartilaginous matrix normally formed by embryonic somites in vitro in the absence of inducing tissues. This inhibition is reflected in a considerable reduction in sulfated glycosaminoglycan (GAG) accumulation by A23187-treated somite explants. Furthermore, A23187 inhibits the striking stimulation of cartilaginous matrix formation and sulfated GAG accumulation normally elicited by the embryonic notochord and collagen substrates. In fact, 1.0 microgram/ml of A23187 reduces sulfated GAG accumulation by somites cultured in association with notochord or on collagen to a level even below that accumulated by somites cultured in the absence of these inductive agents. Although these results must be interpreted with caution, they provide incentive for considering a possible regulatory role for Ca2+ in the chondrogenic response of somites to extracellular matrix components produced by the embryonic notochord.  相似文献

The stimulation of somite chondrogenesis by extracellular matrix components was studied by monitering the synthesis of cartilage-specific large proteoglycan aggregates. Chick embryonic sternal proteoglycans were separated into various components: monomers, hyaluronic acid, link protein and glycosaminoglycan side chains. The effects of these components, either individually or in various combinations, on somite chondrogenesis was examined. Proteoglycan monomers, alone or in a mixture with other components, induced chondrogenesis. The other components did not have any stimulating effect of their own. The results of these induction studies were also observed on a Sepharose CL–2B column and correlated using electron microscopy. Stimulation of somites resulted in an increase in the amount of proteoglycan aggregation (material excluded from the column) and was in agreement with the morphological appearance of the matrix in that there was increased accumulation of large proteoglycan granules. A matrix mixture of collagen and proteoglycans showed significant stimulation. When the matrix environment of the somites was altered to be unfavorable to the explants (medium containing hyaluronic acid) there was altered synthesis of cartilage-specific molecules. The results presented in this report strongly suggest that the composition of the extracellular matrix material is critical for somite chondrogenesis.  相似文献

Previous studies have demonstrated that collagen substrates stimulate in vitro somite chondrogenesis, and that agents that elevate intracellular cyclic AMP levels in hibit the ability of somites to respond to the inductive influence of collagen. In the present investigation, radiommunoassay was utilized to compare the cyclic AMP content of somite explants cultured on purified Type I collagen substrates with control explants cultured on Millipore filters. During the period of culture, the cyclic AMP content of collagen-treated explants is significantly lower than the cyclic AMP content of control explants. The cyclic AMP content of collagen-treated explants is 66% of control values as early as one hour following the initiation of culture, and the cyclic AMP content of collagen-treated explants remains lower than controls throughout the 3-day cultured period. The greatest difference in the cyclic AMP content of collagen-treated and control explants is observed at the seventeenth hour of culture, at which time the cyclic AMP content of collagen-treated explants is 56% of controls. These results combined with previous studies provides support for the hypothesis that collagen elicits a reduction in the cyclic AMP content of embroyic somites and that this reduction is necessary to trigger chondrogenic differentiation.  相似文献

Antibodies to surface immunoglobulins activate inositol phospholipid hydrolysis in B-lymphocytes, but very little is known concerning their effects on cAMP levels. In other cells, products from the hydrolysis of phosphatidylinositol 4,5-bisphosphate can increase and/or potentiate cAMP accumulation. In this study we have examined whether goat anti-mouse IgM (mu-chain-specific) stimulates and/or potentiates increases in the cAMP levels of splenocytes from athymic nude mice. Goat anti-mouse IgM, by itself, stimulated a 60% increase in cAMP within 2 min. Pretreating the cell suspensions at 37 degrees C with anti-IgM produced opposite effects on the forskolin- and prostaglandin E1 (PGE1)-induced increase in cAMP. Anti-IgM (25 micrograms/ml) potentiated the rise in cAMP induced by 100 microM forskolin 76%, but it decreased the response to 50 nM PGE1 by 30%. Direct activation of protein kinase C (Ca2+/phospholipid-dependent enzyme) by 12-O-tetradecanoylphorbol 13-acetate and/or sn-1,2-dioctanoylglycerol resulted in a similar pattern of responses. A 3-min preincubation with 97 nM 12-O-tetradecanoylphorbol 13-acetate potentiated the forskolin-induced response from 1.7 +/- 0.1 to 4.3 +/- 0.6 pmol of cAMP/10(6) cells but reduced the PGE1 response from 0.98 +/- 0.06 to 0.51 +/- 0.03 pmol of cAMP/10(6) cells. Similarly, preincubating the cells for 3 min with 5 microM sn-1,2-dioctanoylglycerol increased the forskolin response from 1.7 +/- 0.1 to 5.1 +/- 0.2 pmol of cAMP/10(6) cells but reduced the response to PGE1 from 1.15 +/- 0.03 to 0.75 +/- 0.04 pmol of cAMP/10(6) cells. Thus, activation of protein kinase C by hydrolysis products of inositol phospholipids, 12-O-tetradecanoylphorbol 13-acetate, or exogenous diacylglycerols modified adenylate cyclase itself and sites upstream of adenylate cyclase such as the receptor or G proteins coupling the receptor to the cyclase. Furthermore, modification of the PGE1 response by anti-IgM provides a mechanism by which antigen can differentially regulate T- and B-cells responding to macrophage-produced prostaglandins during an immune response.  相似文献

This study correlates endogenous levels of cAMP and cGMP with their immunohistochemical localization during chondrogenic differentiation of C57B1/6J mouse limb mesenchyme in vivo and in vitro. A transient decrease in cGMP but not cAMP was found from days 12 to 13 in vivo correlating with early stages of chondrogenesis in the developing limb. Intracellular levels of both cAMP and cGMP in high density limb mesenchyme cultures increased 25% after 24 hr in culture when aggregate and nodule formation was detectable. When cells were seeded at different initial plating densities to delay the onset of aggregate and nodule formation, increased levels of intracellular cAMP correlated temporally with the appearance of nodules. Both cyclic AMP and cGMP were immunohistochemically localized in perichondrial cells and chondrocytes in vivo and in vitro. Therefore, (1) cAMP levels correlated temporally with the appearance of chondrogenic cells and (2) cAMP and cGMP were immunohistochemically localized to chondrogenic cells. These data indicate that fluctuations of both cAMP and cGMP levels may be involved in limb cartilage differentiation. Although increases in both nucleotides were found to correlate with the onset of chondrogenesis in vitro, in vivo data suggest that the amount of cAMP relative to cGMP rather than the absolute amount of an individual cyclic nucleotide may be more significant in modulating differentiation.  相似文献

The enteric nervous system (ENS) is mainly derived from vagal neural crest cells (NCC) that arise at the level of somites 1-7. To understand how the size and composition of the NCC progenitor pool affects ENS development, we reduced the number of NCC by ablating the neural tube adjacent to somites 3-6 to produce aganglionic gut. We then back-transplanted various somite lengths of quail neural tube into the ablated region to determine the 'tipping point', whereby sufficient progenitors were available for complete ENS formation. The addition of one somite length of either vagal, sacral or trunk neural tube into embryos that had the neural tube ablated adjacent to somites 3-6, resulted in ENS formation along the entire gut. Although these additional cells contributed to the progenitor pool, the quail NCC from different axial levels retained their intrinsic identities with respect to their ability to form the ENS; vagal NCC formed most of the ENS, sacral NCC contributed a limited number of ENS cells, and trunk NCC did not contribute to the ENS. As one somite length of vagal NCC was found to comprise almost the entire ENS, we ablated all of the vagal neural crest and back-transplanted one somite length of vagal neural tube from the level of somite 1 or somite 3 into the vagal region at the position of somite 3. NCC from somite 3 formed the ENS along the entire gut, whereas NCC from somite 1 did not. Intrinsic differences, such as an increased capacity for proliferation, as demonstrated in vitro and in vivo, appear to underlie the ability of somite 3 NCC to form the entire ENS.  相似文献

The relation between the logarithm of the volume of the embryo and the number of somites was studied in the mouse and rat in the period of about 0-43 somites, a period characterized by a very rapid volume increase. A linear relation was found, and the variability of the individual observations around the straight line was rather small. Using a previously defined age estimate, i.e. the 'developmental age' based on the embryonic volume, we also found a linear relation between this age estimate and the number of somites. With the help of this straight line, the developmental age can be estimated from the number of somites with an accuracy of less than 0.1 day. Therefore, the developmental age can be considered a better age estimate than those based on the usual methods. The time needed to form a new somite was found to be constant in both species during the period studied. The length of the period differed, and was 1.68 h in the mouse and 2.24 h in the rat. However, the volume of the embryo at the time when a specific somite was formed (for instance the 25th) appeared to be the same in both species. Comparison of the volume of embryos, embedded in paraffin and Epon and sectioned, enabled us to estimate the shrinkage in Epon embryos. The volume after processing was approximately 62% of the original volume.  相似文献

A disordered somite pattern could be produced artificially when the segmental lateral plate of chickembryo was replaced by dissociated cells of quail segmental pate.The artificially disordered somitepattern formed at either place was used in our work as a model to analyze the mechanism of thedevelopment and differentiation of somite on chick embryo.Our conclusions include the following:1.Although the formation of somites from the dissociated segmental plate cells does not requirespecial environment,the development and differentiation of the somltes require a special environmentwhich is related to the neural tube and notochord.The effect of this special environmental factor maydecrease gradually with the increase of the distance from neural tube to lateral plate.2.The somites located on paraxial area at different distances to the axis have different fates indevelopment.3.The formation of epithelial vesicles is the property of somite cells and the epithelial vesicle is thestructural basis of somite differentiation.If and factor interferes with the differentiation of thesomite,the epithelial vesicle of the somite will be degenerated within certain period of time.4.During resegmentation of the somite,the number,size and arrangement of sclerotome in situ donot depend on the somite from which they are derived.5.Somite cells do not transdifferentiate into kidney tubule directly from their original epithelialvesicles,but are reorganized from the free cells dispersed from the disrupted somites.6.The establishment of cell commitment may involve several steps.Before commitment isestablished the of cell commitment is labile.7.The differentiation of sclerotome starts with the rupture of epithelial wall of somites and thedirection of its movement depends not only on the notochord but also on their position with respectto the neural tube and notochord.8.The disordered somite pattern doesn't influence the segmentation of dorsal root ganglia in situ,but causes the formation of the ectopic dorsal root ganglia.Key Words:Somite differentiation;Artificial disordered somite pattern;Chimeral somite;Resegmentation of sclerotome;Distribution of dorsal root ganglia  相似文献

Somites, along with adjacent neural tube and overlying ectoderm, were extirpated unilaterally from embryos of Chelydra serpentina. Mesoderm of three somites was removed from various levels. The operations included the last formed somite and were done on embryos with 12 to 22 pairs of somites. In practice it was found that ventromedial portions of the somites were not included in the extirpation. The animals were preserved before pigmentation became heavy. The cartilaginous skeleton was stained selectively. The extirpations resulted in depletions of ribs consonant with relating the second rib to the fourteenth somite. The somites behaved as mosaics; they did not reconstitute each other nor did they regenerate after partial extirpation. The rudiments for the ribs were separable from the rudiments of the vertebrae, the sclerotomes, and were found to arise from a more lateral portion of the somite. The scutes are ectodermal derivatives, which are held to be dependent upon underlying somitic mesoderm for their differentiation. The extirpations resulted in abnormalities and depletions of scutes.  相似文献

Acetylcholinesterase (AChE) activity, localized histochemically, appeared in the nuclei of presumptive somitic mesodermal cells prior to the onset of somitogenesis. AChE activity appeared in a rostro-caudal sequence, in cells located the equivalent of five somite lengths caudal to the last formed somite. To investigate whether AChE activity was required for somitogenesis, several inhibitors of AChE activity were tested for their ability to block somitogenesis. Diisopropylfluorophosphate (DFP), a broad spectrum inhibitor of serine proteases and related enzymes, was the only AChE inhibitor tested that disrupted somitogenesis. Gastrulae at 50% epiboly exposed continuously to DFP at concentrations between 40 microM and 90 microM completed epiboly, but exhibited a dose-dependent decrease in the number of somites formed, and a parallel decrease in the caudal extent of somite innervation, by 24 hours post-fertilization (h). Fifteen somite (15h) embryos exposed to DFP at the ED50 of 70 microM for 3 hours, followed by recovery to 24h, developed abnormal somites. Approximately five normal somites formed after drug treatment before the first abnormal somite formed. The abnormal somites corresponded in location to that area of the presumptive somitic mesoderm that would have initiated AChE activity while the DFP was present. While exposed to 70 microM DFP, presumptive somites formed and motoneurons extended processes that had initiated AChE activity at the time of treatment with DFP, although at a slower than normal rate. However, embryos exposed to 1 mM DFP for 30 minutes at both the 5 and 15 somite stages, followed by recovery to 24h, developed the normal number of somites but were reduced in the caudal extent of somite innervation, and occasionally developed abnormal primary motoneurons. As with the abnormal somites, the abnormal motoneurons would have initiated AChE activity while the DFP was present. Presumptive somitic mesoderm unable to initiate AChE activity due to inhibition by DFP developed abnormally. Effects of the phorbol ester 12-O-tetradecanoylphorbol-13-acetate (TPA) on chondrogenesis and concentrations of prostaglandin E2 (PGE2) and cyclic AMP (cAMP) were investigated in micromass cultures of chick limb mesenchyme derived from the distal tip of stage 25 limb buds. TPA completely inhibited chondrogenesis during the first 4 days of culture; however, a few small cartilage nodules formed by day 6. Relative to control cultures, both PGE2 and cAMP concentrations were altered by TPA treatment during the 6-day period of cell culture. Concentrations of both compounds increased in control cells during the first 24 h of culture and then declined during the remaining 5 days. In TPA-treated cells both PGE2 and cAMP levels increased progressively during the 6 days of days of cell culture, each being elevated at day 6 by twofold over control cells. The results suggest the presence of regulatory pathways important in chondrogenesis which occur independent of those initiated by PGE2 and the cAMP system.  相似文献
