Doryctobracon brasiliensis (Szépligeti) is a parasitoid larval–pupal of fruit flies and has great potential to be used in biological control programmes as it feeds on other Anastrepha species in addition to Anastrepha fraterculus (Wiedemann). This study investigated the biology of D. brasiliensis at different temperatures to design a life fertility table and determine thermal requirements. The parasitoids were multiplied in larvae of A. fraterculus in air‐conditioned chambers at 15, 18, 20, 22, 25, 28 and 30°C, 70 ± 20% RH and photophase of 12 h. We determined the number of offspring, sex ratio, longevity of males and females and duration of egg–adult period. The temperature range 18–22°C ensures higher fecundity and at 20°C, and the average number of offspring per female was 152.77 parasitoids. The sex ratio of offspring produced was reduced with increasing temperatures. Longevity of males and females of D. brasiliensis was reduced by increasing temperatures. At 15, 28 and 30°C, there was no development of immature stages. For the temperature range 18–25°C, the duration of egg–adult period of D. brasiliensis was inversely proportional to temperature. At 20 and 22°C, we observed the highest values of net reproduction rate (Ro) and finite reason of increase (λ), meaning that at the estimated optimum temperature (21°C), the population of D. brasiliensis increased 47 times each generation. The lower temperature threshold for development was 10.01°C and the thermal constant (K) 303.21 degree/days. This information confirms that D. brasiliensis is better suited to temperate environments, which implies a significant potential for the use of D. brasiliensis in the control of A. fraterculus, because most areas occupied by this pest are in temperate regions. In addition, D. brasiliensis is useful in mass rearing systems in laboratory.  相似文献

The vegetative morphology and life history of Halopteris filicina (Grateloup) Kutzing, collected from Korea, were examined in laboratory culture. Field plants attaining 3–5 cm in height were epilithic, tufted, yellowish-brown, and produced numerous erect axes with alternately distichous branches from compact basal discs. They were cultured under a 12:12 h LD photoperiod at 10°-C, 15°C and 20°C to observe the influence of temperature on reproduction. At 10°C plants grew only vegetatively, whereas at 15°C and 20°C they produced unilocular sporangia. Unispores released from sporangia developed into monoecious, anisogamous gametophytes that formed plurilocular female and male gametangia on the same lateral branches. The zygotes, by fusion of female macrogametes and male microgametes, developed into sporophytes bearing unilocular sporangia, whereas the unfused female gametes germinated parthenogenetically. This species was confirmed to have an isomorphic life history, basically similar to the other species of Sphacelariales.  相似文献

Twenty Aspergillus strains were evaluated for production of extracellular cellulolytic and xylanolytic activities. Aspergillus brasiliensis, A. niger and A. japonicus produced the highest xylanase activities with the A. brasiliensis and A. niger strains producing thermostable β-xylosidases. The β-xylosidase activities of the A. brasiliensis and A. niger strains had similar temperature and pH optima at 75°C and pH 5 and retained 62% and 99%, respectively, of these activities over 1 h at 60°C. At 75°C, these values were 38 and 44%, respectively. Whereas A. niger is a well known enzyme producer, this is the first report of xylanase and thermostable β-xylosidase production from the newly identified, non-ochratoxin-producing species A. brasiliensis.  相似文献

A method of inoculating sugar beet plants (Beta vulgaris L.) with Ramularia beticola Faut. & Lamb, is described. Following inoculation, disease development in relation to temperature and plant age was studied for more than a month. The incubation period was 18 days at 10°C compared to 14 days at 17°C. At 25°C no symptoms appeared. Both temperature and plant age significantly influenced disease level and rate of disease development. Plants incubated at 17°C were more severely diseased 33 days after inoculation than plants incubated at 10°C. Young plants (3 weeks at inoculation) Were more susceptible than older plants (5 and 8 weeks at inoculatson) under growth chamber conditions. In the field, symptoms of Ramularia leaf spot appear relatively late in the season and young leaves are rarely attacked. The inconsistency of these observations is discussed.  相似文献

Ten soft white spring wheat (Triticum aestivum L.) F1 hybrids were grown under three temperature regimes, and anthers were cultured at two plating densities to investigate the effect of plant growth conditions, plating density, and genotype on embryo induction and plant regeneration. Anthers from plants grown at high temperature (25 °/18 °C) or from plants transferred from low (15 °/12 °C) to high temperature generally produced more embryos and green shoots, with a lower frequency of albinos, than did anthers from plants grown at low temperature. However, plating densities of 10 versus 20 anthers per milliliter, had little effect on anther response. Four of the five hybrids with `Fielder' as the female parent produced more embryos and green shoots than did hybrids with `AC Reed' as the female parent. Laboratory cultures of Ectocarpus siliculosus originating from New Zealand showed a defect in gametangium formation. Nuclear divisions in gametangium initials are not followed by cell wall formation. In the resulting multinucleate cells nuclear DNA increases dramatically, and nuclear membranes disintegrate. Eventually, the entire structure is filled with hexagonal particles of approximately 130 nm diameter. These were isolated and shown by EM to consist of a dense core surrounded by a 3-layered shell. They are released into the culture medium when the host cells burst. Ectocarpus gametes from healthy cultures could be infected by these particles. The resulting partheno-sporophytes developed pathological symptoms, suggesting that the particles are viruses. The expression of the defect is temperature dependent. At 10°C all gametangia are abnormal, while between 15 and 20 °C defective and normal gametangia and gametes are formed on the same plant. Partheno-sporophytes developing from such gametes carry the viral particles expressed in deformed unilocular and plurilocular sporangia.  相似文献

Ornithine decarboxylase in Paracoccidioides brasiliensis, a dimorphic human pathogenic fungus, was more active at 37° C in the yeast phase and at 30° C in the mycelial phase. In contrast to other fungal systems, yeast growth and mycelium-to-yeast transition in P. brasiliensis were accompanied by a high activity of ornithine decarboxylase at the onset of the budding process, the activity of which was inhibited by 1,4-diamino-2-butanone. The activity of ornithine decarboxylase remained at a basal level during vegetative growth of both the mycelial phase and the late stage of yeast phase, and also through the yeast-to-mycelium transition. Parasitoid wasps of the genus Trichogramma are used worldwide as biological control agents against lepidopteran pests. Trichogramma wasps develop inside eggs of a wide range of host species, most of them moths. They are generally considered as diurnal insects. Here, we investigated whether Trichogramma wasps can also successfully parasitise host eggs at night under controlled laboratory conditions. Eggs of the moth Ephestia kuehniella were offered under dark conditions (scotophase) to females of Trichogramma brassicae and Trichogramma evanescens either from 9:00 PM to 9:00 AM or from 11:00 AM to 5:00 PM at four different temperatures (5°C, 10°C, 15°C and 20°C). Both species are known to parasitise E. kuehniella eggs in the photophase during daytime. The results show that T. brassicae did not parasitise eggs in the scotophase at night and only very few in the artificially induced scotophase during daytime from 10°C to 20°C. In contrast, T. evanescens parasitised more eggs in the dark both at night and artificially induced scotophase during daytime. Parasitism in the scotophase already started at 5°C, with more eggs being parasitised and more offspring being produced at higher temperatures. T. evanescens displayed higher parasitism activity in the induced scotophase during daytime than in the scotophase at night. The present study suggests that Trichogramma are capable of successfully parasitising host eggs at night, even at low temperatures, but that nocturnal activity with respect to parasitism varies between wasp species.  相似文献

Morphology of field material and life history in culture were studied in Scytosiphon canaliculatus (Setchell et Gardner) comb. nov. from northern Japan. Erect gametophytes of S. canaliculatus are cylindrical, tubular, up to 7 mm wide and 40 cm long, and without regular constrictions. S. canaliculatus has pronounced anisogamy and ascocysts accompanied with plurilocular gametangia. The life history of S. canaliculatus showed an alternation between erect gametophytes and crustose prostrate sporophytes bearing unilocular sporangia. Since field sporophytes of S. canaliculatus were found to be identical with Hapterophycus canaliculatus Setchell et Gardner (Ralfsiaceae, Phaeophyceae), it is proposed to transfer H. canaliculatus to the genus Scytosiphon. In the field, gametophytes with plurilocular gametangia appeared in spring and disappeared in summer. Sporophytes with unilocular sporangia were collected in late autumn and winter. Unilocular sporangia were produced at 15°C in short-day culture conditions and unispores developed into erect gametophytes at 5–15°C. It is suggested that the seasonal Occurrence of gametophytes in the field is due to the seasonal formation of unilocular sporangia, which is regulated by temperature and photoperiod.  相似文献

In addition to its role in vernalization, temperature is an important environmental stimulus in determining plant growth and development. We used factorial combinations of two photoperiods (16H, 12H) and three temperature levels (11, 18 and 25 °C) to study the temperature responses of 19 wheat cultivars with established genetic relationships. Temperature produced more significant effects on plant development than photoperiod, with strong genotypic components. Wheat genotypes with PPD‐D1 photoperiod sensitive allele were sensitive to temperature; their development was delayed by higher temperature, which intensified under non‐inductive conditions. The effect of temperature on plant development was not proportional; it influenced the stem elongation to the largest extent, and warmer temperature lengthened the lag phase between the detection of first node and the beginning of intensive stem elongation. The gene expression patterns of VRN1, VRN2 and PPD1 were also significantly modified by temperature, while VRN3 was more chronologically regulated. The associations between VRN1 and VRN3 gene expression with early apex development were significant in all treatments but were only significant for later plant developmental phases under optimal conditions (16H and 18 °C). Under 16H, the magnitude of the transient peak expression of VRN2 observed at 18 and 25 °C associated with the later developmental phases.  相似文献

Seedlings of Stylosanthes guianensis var. guianensis cv. Cook and of two selections of S. guianensis var. pauciflora (CIAT 1280 and CIAT 1062) were grown at day/night temperatures of 20°/25°, 30°/25°, and 35°/30°C in a naturally-lit glasshouse at latitude 27°30'S. Sowings were made in decreasing daylength at 30-day intervals from 22 January to 21 May. Cv. Cook and the CIAT 1280 selection did not flower fully if sown after 22 January and the CIAT 1062 selection did not flower if sown after 22 March. This is interpreted as a long-short day flowering response. Usually, flowers were initiated earlier and at a lower node at 25?/20°C than at the warmer temperatures. At 25?/20°C the first flowers to appear were produced predominantly at the terminal apex of the main stem in cv. Cook and the CIAT 1062 selection, but not in the CIAT 1280 selection. At the two warmer temperatures first flowers were more commonly produced at the terminal apex of primary, secondary and tertiary branches. There were more inflorescences per plant on earlier than later sown plants when measured 21 days from appearance of the first flower and the most inflorescences were produced by cv. Cook at 25?/20°C, by selection CIAT 1062 at 30°/25°C, and by CIAT 1280 at 35°/30°C.  相似文献

Warming ocean temperatures have been linked to kelp forest declines worldwide, and elevated temperatures can act synergistically with other local stressors to exacerbate kelp loss. The bull kelp Nereocystis luetkeana is the primary canopy-forming kelp species in the Salish Sea, where it is declining in areas with elevated summer water temperatures and low nutrient concentrations. To determine the interactive effects of these two stressors on microscopic stages of N. luetkeana, we cultured gametophytes and microscopic sporophytes from seven different Salish Sea populations across seven different temperatures (10–22°C) and two nitrogen concentrations. The thermal tolerance of microscopic gametophytes and sporophytes was similar across populations, and high temperatures were more stressful than low nitrogen levels. Additional nitrogen did not improve gametophyte or sporophyte survival at high temperatures. Gametophyte densities were highest between 10 and 16°C and declined sharply at 18°C, and temperatures of 20 and 22°C were lethal. The window for successful sporophyte production was narrower, peaking at 10–14°C. Across all populations, the warmest temperature at which sporophytes were produced was 16 or 18°C, but sporophyte densities were 78% lower at 16°C and 95% lower at 18°C compared to cooler temperatures. In the field, bottom temperatures revealed that the thermal limits of gametophyte growth (18°C) and sporophyte production (16–18°C) were reached during the summer at multiple sites. Prolonged exposure of bull kelp gametophytes to temperatures of 16°C and above could limit reproduction, and therefore recruitment, of adult kelp sporophytes.  相似文献

Abstract. The effects of temperature on the development of early stages and the thermopreference of nymphs and adults were analysed in the haematophagous bug Triatoma brasiliensis Neiva, 1911 (Hemiptera, Reduviidae). Egg hatching, mortality of nymphs, feeding and moulting success of the early stages of T. brasiliensis were all affected by temperature. While high rates of egg hatching were observed between 25 and 27 °C, no hatching occurred at 12, 19 and 38 °C. The mortality of first‐instar nymphs was highest at 38 °C, at which no insects survived after 10 days of exposure. Feeding success was only affected at the lowest temperature (12 °C). No ecdysis was observed in the groups exposed to 12, 19 and 21 °C. Recently fed fourth‐instar nymphs preferred to stay at a temperature of approximately 30 °C. The preferred temperature began to decline gradually to approximately 27 °C during ecdysis, reaching 26 °C at 30 days after ecdysis. After a second blood meal, the insects' preferred temperature was again approximately 30 °C. The thermopreference pattern of females was similar to that of nymphs. Nymphs and females showed a daily fluctuation in their preferred temperature, moving towards higher values at the beginning of the dark phase, and choosing lower ones after this time interval, at which they remained until the end of the light phase. The females laid their eggs in all sectors of the arena, although the largest numbers of eggs were found between 28 and 32 °C.  相似文献

Soybean [Glycine max (L.) Merr] plants were exposed to three temperature regimens during seed development to investigate the effect of temperature on the expression of eight defense-related genes and the accumulation of two fungal pathogens in inoculated seeds. In seeds prior to inoculation, either a day/night warm (34/26°C) or a cool temperature (22/18°C) relative to normal (26/22°C) resulted in altered patterns of gene expression including substantially lower expression of PR1, PR3 and PR10. After seed inoculation with Cercospora kikuchii, pathogen accumulation was lowest in seeds produced at 22/18°C in which of all defense genes, MMP2 was uniquely most highly induced. For seeds inoculated with Diaporthe phaseolorum, pathogen accumulation was lowest in seeds produced at 34/26°C in which of all defense genes, PR10 was uniquely most highly induced. Our detached seed assays clearly demonstrated that the temperature regimens we applied during seed development produced significant changes in seed defense-related gene expression both pre- and post inoculation and our findings support the hypothesis that global climate change may alter plant–pathogen interactions and thereby potentially crop productivity.  相似文献

We analysed the relative effects of food availability and temperature on rates of growth and development of a predatory planktonic water mite, Piona exigua. Growth in length of mites fed Daphnia, Ceriodaphnia and Chydorus was analysed by Gompertz or von Bertalanffy curves; these curves were compared by parallel curve analysis. Growth rates of nymphs and adult female mites increased with temperature; the duration of the imagochrysalis stage decreased. Females grown at 10 °C were smaller at final size than females grown at 15 °C, 18 °C or 22 °C. Females reared at food levels of 15 or 30 prey l−1 grew more slowly and were smaller than those provided with 60 or 120 prey l−1. Nymphs grew more slowly when Daphnia were the only prey, than when smaller prey were available. Food level did not affect nymph growth at 10 °C or 15 °C, but growth at 18 °C or 22 °C may have been slowed at the lowest food levels. Synergistic effects of temperature and food level on nymph growth were apparent only from analysis of growth curves and not from stage duration data.  相似文献

Blastospores of three strains of Metarhizium anisopliae were stored in 18 liquids at 4°C, 20°C and 35°C for 18 weeks, 12 weeks or 9 days respectively. Viability was quantified by determination of their germination. In bioassays the virulence of stored blastospores was studied using adults and third instars of Locusta migratoria migratorioides (R. & F.) and compared to those of freshly produced blastospores and conidia. Generally, there was great variability in the viability of blastospores, depending on the fungal strain and the liquids used. Blastospores survived best at 4°C in 10% hydroxyethyl starch; for example, germination of M. anisopliae strain 97 still amounted to more than 80% after storage for 18 weeks. Other suitable liquids were deionized water, 25% Ringer's solution and 1% sodium alginate. The viability of blastospores stored at 20°C was considerably shorter than at 4°C. During storage for 12 weeks at 20°C the best protective liquids for M. anisopliae strain 97 were 25% Ringer's solution (43% germination), deionized water (23%) and 10% hydroxyethyl starch (23%). At 35°C, 45% of M. anisopliae strain 97 blastospores still germinated after storage for 7 days in 25% glycerol. The bioassays revealed that the virulence of blastospores after storage was comparable to that of fresh ones and even better than that of fresh conidia. In general, the LT50 was about 4–6 days at an alternating day/night temperature of 28/20°C.  相似文献

Dahlia mosaic virus (DMV) infected twenty-five of the eighty-five plant species from four of eighteen families inoculated, but only dahlias were found naturally infected. DMV infected fourteen members of the Solanaceae, Amaranthaceae and Chenopodiaceae, and eleven of twenty-nine Compositae. Verbesina encelioides was the best plant for diagnosis, assay and source of virus. Systemically infected hosts contained ovoid intracellular inclusions 2–5–10 μm in diameter which were shown by electron microscopy to consist of a finely granular, vacuolated matrix containing numerous virus particles. V. encelioides sap was sometimes infective after dilution to 1/2000 but not 1/3000, after heating for 10 min to 75 °C but not 80 °C, and after 4 days at 18 °C or 32 days at 2 °C. Sap from infected dahlia, Zinnia elegans or Ageratum houstonianum rapidly became non-infective, but extracts made with 0·05 M sodium thioglycollate or 0·03 M sodium diethyldithiocarbamate remained infective for 24–48 h at 18 °C. Some purified preparations remained infective for up to 3 years at 2 °C. DMV was best purified from V. encelioides by one or more cycles of differential centrifugation, followed by density-gradient centrifugation and further concentration. Composition, molarity, and pH of the extracting buffer had little effect on yield of virus. Best yields were obtained from extracts stored with 8-5% (v/v) n-butanol at 2 °C for 10–14 days. Purified preparations were infective at dilutions up to 1/5000, had ultraviolet absorption spectra typical of a nucleoprotein (Å 260/280 = 1·47), probably contained DNA, and had a single sedimenting component having isometric particles c. 50 nm in diameter with a sedimentation coefficient of 254 S. The cryptogram of DMV is (D)/*:*/(16):S/S:S/Ap. DMV is serologically closely related to cauliflower mosaic virus, but the viruses are distinct pathogens. The two viruses have similar properties, size, shape and other characteristics, and together with at least three others form a small but apparently homogeneous group of aphid-borne viruses.  相似文献

Abstract

Cheese production is projected to reach 20 million metric tons by 2020, of which 33% is being produced using calf rennet (EC 3.4.23.4). There is shortage of calf rennet, and use of plant and microbial rennets, hydrolyze milk proteins non-specifically resulting in low curd yields. This study reports fungal enzymes obtained from cost effective medium, with minimal down streaming, whose activity is comparable with calf and Mucor rennet. Of the fifteen fungi that were screened, Mucor thermohyalospora (MTCC 1384) and Rhizopus azygosporus (MTCC 10195) exhibited the highest milk-clotting activity (MCA) of 18,383?±?486?U/ml and 16,373?± 558?U/ml, respectively. Optimization exhibited a 33% increase in enzyme production (30?g wheat bran containing 6% defatted soy meal at 30?°C, pH 7) for M. thermohyalospora. The enzyme was active from pH 5–10 and temperature 45–55?°C. Rhizopus azygosporus exhibited 31% increase in enzyme production (30?g wheat bran containing 4% defatted soy meal at 30?°C, pH 6) and the enzyme was active from pH 6–9 at 50?°C. Curd yields prepared from fungal enzyme extract decreased (5–9%), when compared with calf rennet and Mucor rennet. To study a possible adaptation of the symbiosis between white clover (Trifolium repens L.) and Rhizobium leguminosarum biovar trifolii with regard to light and temperature at northern latitudes, local seed populations of white clover and isolates of R. leguminosarum biovar trifolii from 3 different latitudes in Norway, 58°48'N, 67°20'N and 69°22'N, were used. The commercial cultivar Undrom was used as a reference plant. The experiments were done at 18 and 9°C under controlled conditions in a phytotron during the natural growing season at 69° 39'N. Growth of the plants was evaluated by number and size of leaves, dry matter production and total N-content. At 18°C the white clover plants were harvested twice while at 9°C there was only one growth period. The results from first harvest at 18°C and total growth at 9°C, showed that white clover populations from northern Norway had a lower growth potential than the population from the south and cv. Undrom. This difference was not apparent in the second growth period at 18°C. Growth of the plants from seeds to first harvest was enhanced by mineral nitrogen compared to plants dependent on Rhizobium only. However, after a second growth period dry weight and total nitrogen content of the plants with nitrogen fixation were comparable to the plants receiving mineral nitrogen. Statistical analysis showed that the most important factor for the variation in dry matter production was the plant population. Within the populations at 9°C and at first harvest at 18°C, there were no significant differences in dry matter production with different Rhizobium inoculum. In the second growth period at 18°C, different inoculum gave significantly different amount of dry matter within a population. The results showed a significant interaction between plant population and Rhizobium inoculum, and the results indicated that plants from the north gave higher yield when nodulated by Rhizobium from the north than from the south.  相似文献
