Summary The fine structure of the epidermis in Haplognathia simplex, Haplognathia rosea, Pterognathia meixneri (Filospermoidea) and Gnathostomula paradoxa (Bursovaginoidea) has been investigated. The epidermis in the filospermoidean species is uniform, consisting of epidermal cells with a single locomotory cilium. The structure and development, including ciliogenesis, of these epidermal cells are described. In G. paradoxa additional epidermal elements have been found: mucous cells with a presumably apocrine secretion modus are scattered in a strip-like arrangement within the epidermis. Their deverlopment is separate from epidermal cells with locomotory function. Two further types of glandular cells with either a single cilium or a diplosome are located ventrally. The cytochemical nature of the secretory contents of the unicellular glands—the mucous cells and the sacciform granulated cells in the epidermis of Monopterus cuchia has been described in order to understand their physiological significance.
The mucous cells secrete mainly the acid mucopolysaccharides and mucoproteins in addition to small amounts of phospholipids. The secretory contents of the sacciform granulated cells are proteinaceous in nature and give strong reactions for elastin, tryptophan and tryosine. High amounts of cholesterol and small amounts of phospholipids and acid mucopolysaccharides are also identified in these sacciform granulated cells.
Phospholipids, mucoproteins and elastin could well increase the viscosity of mucopolysaccharides retaining the slimy secretions on the surface for relatively longer periods. The retention of mucopolysaccharides and elastin on the surface for longer periods that can bind large amounts of water has been correlated to keep the skin moist, facilitating the cutaneous respiration and preventing the fish from desiccation when it is on land. The role of elastin in protecting the fish from various chemical attacks and mechanical stresses is also discussed.
Tryptophan secreted by sacciform granulated cells may have the immunological role facilitating the antibody transfer across the epidermis.  相似文献

Methyl methacrylate vascular corrosion replicas were used to examine the macrocirculation in the head region and the microcirculation of respiratory vessels in the air-breathing swamp eel Monopterus cuchia. Fixed respiratory tissue was also examined by SEM to verify capillary orientation. The respiratory and systemic circulations are only partially separated, presumably resulting in supply of mixed oxygenated and venous blood to the tissues. A long ventral aorta gives rise directly to the coronary and hypobranchial arteries. Two large shunt vessels connect the ventral aorta to the dorsal aorta, whereas the remaining ventral aortic flow goes to the respiratory islets and gills. Only two pairs of vestigial gill arches remain, equivalent to the second and third arches, yet five pairs of aortic arches were identified. Most aortic arches supply the respiratory islets. Respiratory islet capillaries are tightly coiled spirals with only a fraction of their total length in contact with the respiratory epithelium. Valve-like endothelial cells delimit the capillary spirals and are unlike endothelial cells in other vertebrates. The gills are highly modified in that the lamellae are reduced to a single-channel capillary with a characteristic three-dimensional zig-zag pathway. There are no arterio-arterial lamellar shunts, although the afferent branchial artery supplying the gill arches also supplies respiratory islets distally. A modified interlamellar filamental vasculature is present in gill tissue but absent or greatly reduced in the respiratory islets. The macro- and micro-circulatory systems of M. cuchia have been considerably modified presumably to accommodate aerial respiration. Some of these modifications involve retention of primitive vessel types, whereas others, especially in the microcirculation, incorporate new architectural designs some of whose functions are not readily apparent.  相似文献

The structure of the air-breathing organs ofMonopterus (=Amphipnous)cuchia has been studied by using light, scanning and transmission electron microscopy and the morphological basis for buccopharyngeal respiration, aerial as well as aquatic has been established. Respiratory islets are well distributed over the surface of the buccopharynx, hypopharynx and branchial arches extending deep into the gill clefts but occupy only the anterior two-thirds of the air sacs, the remaining posterior one-third part seems to be non-respiratory in function and may serve as a reservoir for residual air. Arterioles penetrate deep into the epithelial region of air sacs and buccopharynx in spiral-like fashion to form the characteristic vascular papillae of the respiratory islets. In juvenile fish new respiratory islets develop in the non-vascular part of the air sac in between large older islets as sprout-like structures. The respiratory area, capillary loading, thickness of air-blood tissue barrier, and the diffusing capacity of the respiratory membrane of a 200 g fish were found to be 20 cm2, 2.72 cm3/m2, 0.72 × 10?4 cm, and 0.00165 ml O2/min/mmHg/kg, respectively.  相似文献

Summary Ultrastructural and histochemical studies performed on the skeletal elements of the tail fins of six representative species of teleosts enabled the following observations to be made. The electron microscopic pattern of amorphous substance deposition, and the diameter of the collagen fibrils, in lepidotrichia closely resemble those which are typical of cartilage. In addition, lepidotrichia contain chondroitin sulfate AC as the only sulfated glycosaminoglycan, and this glycosaminoglycan shows high levels of interaction with collagen, both features being characteristic of cartilage. Furthermore, the histochemical data presented in this paper suggest that not all of the glycosaminoglycans present in lepidotrichia are bound to protein cores to form proteoglycans. Each actinotrichium consists of a single ultrastructural entity of remarkable width and, thus, is not composed of a bundle of discretely separated collagen fibrils but rather of hyperpolymerized collagen molecules. This aspect differs from the arrangement pattern of all the other interstitial collagens, suggesting that actinotrichia may contain a new type of collagen.To whom offprint requests should be sent  相似文献

The fine structure of clear cells located within the frog epidermis was investigated. Characteristic features of clear cells were, the 'flask-like' shape, the sparse amount of filaments resulting in a clear cytoplasmic appearance, abundant mitochondria, numerous small vesicles, and a system of convoluted apical membranes in cells adjacent to horny cells. These features were suggestive of a secretory function for the clear cell. A comparison of clear cells with fully differentiated epidermal cells (functioning in protection) was made, in an attempt to discover how structural differences between the two cells were related to the basic functions of the cells.  相似文献

Summary The annual changes of the spermatic duct ofSalaria pavo and the characteristics of the spermatic ducts of spawningAidablennius sphynx, Lipophrys adriaticus, L. dalmatinus, andParablennius incognitus are described by fine structural and histochemical methods. The spermatic duct ofS. pavo has an inactive phase during the interspawning period, a proliferation phase during the prespawning period, a phase of high secretory activity during the spawning period and a regression phase in the postspawning period. During spawning the spermatic duct ofS. pavo is involved in the secretion of glycogen, lipids, and sulfomucins and has similar fine structural and histochemical characteristics as the spermatic ducts ofA. sphynx, L. adriaticus, L. dalmatinus, andP. incognitus. The spermatic ducts of bleniid fish function (1) in stabilizing and continuation of the final differentiation processes of spermatozoa, (2) in storage of mature spermatozoa, (3) in nutrition of germ cells by glycogen, and (4) in the production of sulfomucins regarded as important for the mode of fertilization. Possible meanings of the varying size of the spermatic ducts in different species of blennies are discussed.  相似文献

A morphological study by light and electron microscopy on the lateral line system of the urodele amphibian Pleurodeles waltii demonstrates the presence of sensory organs other than neuromasts in the head. From their morphology, they have been called ampullary organs. The ampullary organs occur in the bottom of a groove and consist of three different types of cells: sensory, supporting and mantle cells. Histochemical analysis indicates that the last two are secretory cells, probably involved in the production of the material filling the ampulla and the groove.  相似文献

The fine structure of the integument in several species of Pogonophora has been examined by electron microscopy. The cuticle over the main body is composed of several layers of orthogonally arranged fibres embedded in an amorphous matrix. It is regularly traversed by microvilli from underlying epidermal cells. Toothed bristles of the annuli and setae of the anchor are composed of closely packed fibrous cylinders wrapped in a cortical material. In fine structure the cuticle, setae, toothed bristles (or setae) and setal sacs forming the setae closely resemble the corresponding structures in annelids. The cuticle is maximally thick over the forepart (protosome + mesosome) ; it is very thin and non-fibrous over the surface of the metameric papillae and over extensive areas of post-metameric trunk. The possibilities of a collagenous nature of the cuticle fibres and their mode of secretion by the epidermal cells are discussed. The organization of various cell-types forming the epidermis over the entire animal is examined. Possible functions of these cell-types are discussed. Notable amongst these are 'possible zymogen cells' and some absorptive cells. The intriguing question of nutrition in these gut-less tubiculous animals is re-examined in the light of present observations.  相似文献

The spermathecal accessory gland of female Tenebrio molitor is examined by histochemicai and electron microscopical techniques. Immediately after ecdysis of the female, neither Golgi regions nor the endoplasmic reticulum of the secretory cells are well developed. In two days' time, the cytoplasm is rich in rough endoplasmic reticulum and the Golgi areas are expanded. Membrane-bound droplets of secretion move from the Golgi zone to a central cavity, formed by the invaginated plasma membrane of this cell. As the secretion accumulates this cavity swells until the fourth day after ecdysis when the females first mate. An efferent cuticular ductule, ensheathed in a ductulecarrying cell, carries the product to the main axial duct of the tubular gland. By histochemical criteria, the product is a glycoprotein.  相似文献

The histochemical observations performed by the authors on the stomach wall of Donax trunculus are not wholly in agreement with the results of workers on other bivalves. The ultrastructure of the gastric shield and underlying cells was therefore, studied. It was confirmed that the epithelial cells contain glycogen granules at the base. In addition two distanct P.A.S. positive inclusions were identified: lysosomal residual bodies and neutral mucopolysaccharidic inclusions. The gastric shield is composed largely of long narrow microvilli embedded in a chitinous glycocalyx but is nevertheless easily removable. Hence, there is no causal relationship between abundance of microvilli and the possibility of removing the shield.  相似文献

The surface coat, ciliary process, and microvilli of the lamprey neuromast were examined with electron microscopy after tannic acid prefixation and lectin histochemistry. The neuromast was found to exist in the form of a dermal mound with a furrow in the middle. On the bottom of the furrow, the hair cell was characterized by a kinocilium and 15–20 stereocilia, arranged along the longitudinal axis of the furrow. Spanning structures were demonstrated between the kinocilium and stereocilia as well as between stereocilia. The surface coat, enhanced by tannic acid prefixation, was particularly rich over the surface of the supporting cell; by contrast, it was thin over the hair cell. Some lectins (PNA, GS-I, SBA, WGA) showed affinity to the surface coat of the supporting cell as well as the hair cell, and the others (RCA-I, MPA, ConA) showed affinity only to the supporting cell. These differences in the structure and affinities of the surface coat suggest an extracellular milieu highly specialized for the hair cell in this particular form of the mechanoreceptor.  相似文献

The ultra-structure of the epidermis of the mudskipper,Periophthalmus modestus, was examined by both light and transmission electron microscopies. The epidermis is exceptionally not well endowed with mucous or granular cells. Filament-containing cells occur in three distinct layers of the surface, middle and basal epidermis. The surface layer is further subdivided into two layers, an outermost and less superficial one. Two different cell types were identified in the epidermis. Type I cells are fiat cells in a single stratum. Type II cells are enormous cells, characterized by having a large vacuole in the cytoplasm. The outermost layer is composed of a free surface of Type I cells and numerous microridges covered with a fuzzy, fibrillar substance. The “fuzz” forms a cuticule-like structure, but keratinization as found in terrestrial animals does not occur. The superficial layer contains Type I cells and intraepithelial blood capillaries. When Type I cells become senescent, numerous intercellular spaces are formed in the plasma membranes of adjacent cells, with the senescent cells finally falling off. Just beneath these cells, however, young cells of Type I are always found. The blood capillaries are usually reinforced with young Type I cells. A large volume of oxygen may be absorbed through the skin using the blood capillary network. The middle layer contains several strata of Type II cells. The special corky structure of these cells seems to play an important role in thermal insulation and protection against ultraviolet light in relation to life out of water. However, by comparison with terrestrial animals, the histological design of the epidermis of this goby appears incomplete, so as to reduce desiccation on land, owing to the epidermis lacking a keratinized stratum. The differentiation of the epidermis seems to be an adaptation for a terrestrial habit in this species.  相似文献

Summary The organization of the wall of epidermal cells in the petiole of species of Apium, Eryngium, Rumex, and Abutilon as well as that of the epidermis of Avena coleoptile has been investigated. The outer and inner tangential walls consist of layers in which the cellulose microfibrils are oriented alternately parallel or transverse to the longitudinal cell axis. This organization resembles that previously described for collenchyma cell walls (Wardrop, 1969; Chafe, 1970). On the radial (anticlinal) walls the orientation of the microfibrils is transverse and these appear continuous with the layers of transverse orientation of the outer and inner tangential walls. Variation in thickness of the outer tangential, and radial, and inner tangential walls appears to result from the variation in thickness of those layers in which the microfibrils have a longitudinal orientation. The extent to which these observations can interpreted in terms of some type of modified multi-net growth is discussed.  相似文献

Summary In species of Apium, Eryngium and Humulus, the cuticular membrane of the petiole could be resolved into two parts, of which the inner one appeared amorphous and after staining appeared to be penetrated by an electron-dense reticulum, whereas the outer layer showed a lamellate structure consisting of electron-dense and electron-transparent plates, 50–80 Å in thickness. These layers are considered to correspond with the cuticular layer and the cuticle proper, respectively. In species of Abutilon and Rumex the cuticle proper did not exhibit the lamellate structure. In the leaves of Eryngium the outer lamellated structure was present in the cuticle of both young and mature leaves. Both the lamellate and non-lamellate types of the cuticle proper increased in thickness with age of the specimen. The results are discussed in relation to earlier investigations.  相似文献

The fine structure of a cell type in the epidermis of brown trout, Salmo trutta L., is described. Its cytological features are compared with those of other sacciform cells reported in several species of teleost fish. This cell type also has a highly electron-dense cytoplasm with numerous Golgi systems and extensive rough endoplasmic reticulum. However, surrounding the large central vacuole there are no peripheral vesicles—'bubbles'—which are characteristic features in non-salmonid teleosts. Instead of these, in the vacuole there are cytoplasmic intrusions of circular cross-sections forming a mesh in the cortical zone. Inside these intrusions there are granules which are interpreted as ribosomes trapped by fusion of the vacuole and reticulum membranes. The way in which the secretion is released into the lumen of the central vacuole is discussed. It is suggested that this proteinaceous material does not pass through the Golgi system, but flows directly from endoplasmic cisternae to the vacuole.  相似文献
