The phytoplankton primary production was studied in different types of six soft-water, weakly-mineralized karst lakes: Kschara, Sankhar, Yukhor, Poridovo, Svetlen’koye, and Bol’shiye Garavy (Vladimir oblast). The following aspects were studied: seasonal dynamics, vertical distribution of the photosynthetic intensity, relative algal photosynthetic activity, the relationships between production processes and water transparency, BOD5, and the concentrations of total forms of nitrogen and phosphorus. It was revealed that the maximal photosynthesis rate depends more on the total nitrogen than on the total phosphorus. A close correlation was found between the maximal photosynthesis rate and BOD5.  相似文献

We compared phytoplankton and phytobenthos pigment strategies in 17 shallow lakes and ponds from northern Canada and Alaska, sampled during mid to late summer. Benthic chlorophyll a concentrations (8–261 mg m−2) greatly exceeded those of the phytoplankton (0.008–1.4 mg m−2) in all sites. Cyanobacteria dominated the phytobenthos, while green algae and fucoxanthin-groups characterized the plankton. Both communities had higher photoprotection in cold, UV-transparent, high latitude waters. Phytoplankton had higher concentrations of photoprotective carotenoids per unit chlorophyll a than the phytobenthos. The planktonic photoprotective pigments were positively correlated with UV-penetration, and inversely correlated with temperature and coloured dissolved organic matter. A partial redundancy analysis showed that the benthic pigments were related to latitude, area and temperature. The UV-screening compound scytonemin occurred in high concentrations in the phytobenthos and was inversely related to temperature, while benthic carotenoids per unit chlorophyll a showed much lower variability among sites. These differing pigment strategies imply divergent responses to environmental change between the phytobenthos and phytoplankton in high latitude lakes.  相似文献

Concentrations of phytoplankton photosynthetic pigments were measured at the Biya head water (in the immediate vicinity of Lake Teletskoe) every 10 days from July 1998 through December 2001. Comparison of phytoplankton pigment characteristics at the river’s head waters and at the surface of the lake’s pelagic zone for the ice-free period shows that the monitoring data give a clear picture of the formation and functioning of the lake phytoplankton. Analysis of seasonal fluctuations of pigment characteristics and their proportions revealed characteristic stages in the development of lake algal cenoses. We have demonstrated similarities and differences in the seasonal dynamics of the phytoplankton between Lake Teletskoe, a deep oligotrophic lake in the south of West Siberia, as compared with the general pattern observed in temperate lakes. According to the concentration of chlorophyll a, the trophic status of the lake’s pelagic zone and the Biya head water is ultraoligotrophic-oligotrophic. Yellow:green pigment ratio ranks Lake Teletskoe among “carotenoid lakes“. Low coefficients of linear correlation between chlorophyll a, water level and biogenic matter concentration shows that the relationships analyzed are far more complex than has been inferred from the combined effect of abiotic and biotic factors.  相似文献

We studied micro-benthic copepods and cladocerans in the bottom substrates of 12 lakes in the Bol’shezemel’skaya tundra in North-East European Russia during the summer. About 30 species of Cladocera and 28 species and subspecies of Copepoda were found. The majority of the micro-crustaceans are palaearctic or northern palaearctic. But the harpacticoid fauna of the western part of the studied region had specific European features, and three species of the harpacticoid fauna in the eastern part of the region were Siberian. It is therefore a boundary territory for two zoogeographical regions, the arctic and subarctic zones. Acidophilic (Arcticocamptus arcticus (Lilljeborg 1902)) and halophilic (Microsetella norvegica (Boeck 1864)) harpacticoid species were found in the lakes of the eastern part of Bol’shezemel’skaya tundra. On basis of their way of locomotion we distinguished five ecological groups and on basis of their feeding habits four ecological groups of micro-crustaceans. Groups with a similar way of locomotion were likely distributed over the studied water bodies. Habitat (inshore versus offshore), substrate particle size and substrate hardness were of paramount importance for the species distribution.  相似文献

Numerous studies have demonstrated alternative regimes in shallow lake ecosystems around the world, with one state dominated by submerged macrophytes and the other by phytoplankton. However, the stability of each regime, and thresholds at which lakes shift to the alternative regime, are poorly known. We used a cross-sectional analysis of 72 shallow lakes located in prairie and parkland areas of Minnesota, USA, during 2005 and 2006 to assess the occurrence of alternative regimes and shifts between them. Cluster analysis revealed two distinct groups of lakes characterized not only by different macrophyte abundance and chlorophyll a levels but also by different total phosphorus–chlorophyll a relationships. Thirty-nine lakes were macrophyte- and 23 lakes phytoplankton-dominated in both years, whereas 10 sites shifted sharply between those regimes. We failed to detect a universal shifting threshold in terms of chlorophyll a or total phosphorus. However, 95% of the lakes with chlorophyll a concentrations less than 22 μg l−1 were in a clear-water regime, whereas 95% of the lakes with chlorophyll a higher than 31 μg l−1 were in a turbid regime. Total phosphorus less than 62 μg l−1 was an accurate predictor of lakes in a stable clear-water regime, whereas a large change in biomass of planktivores and benthivores between years was the only variable weakly related to regime shifts. Our results support the theoretical prediction that regime thresholds vary among lakes. We recommend that lake managers focus on improving resilience of clear regimes in shallow lakes by reducing nutrient loading, rather than attempting to identify and manage complex triggers of regime shifts. Seasonal nutrient enrichment experiments (short-term bioassays) were conducted in three Florida lakes of different trophic states to determine the effects of addition of various nutrient combinations upon chlorophyll a and phytoplankton standing crops. Nutrient enriched surface water samples with crustacean zooplankton removed were incubated in situ in clear polyethylene bags for 3 to 6 days. The 25 factorial design employed two levels (ambient and enriched) of each of five nutrients [NH4 +, PO inf4 sup3− , Fe -EDTA, SiO inf3 sup2− and a cation (Ca2+ or K+) or trace elements]. Ammonium produced significant increases in chlorophyll a and phytoplankton standing crops in all experiments. Phosphate produced similar results in the mesotrophic lake, but the eutrophic lakes had both positive and nonsignificant responses which varied seasonally between lakes. Iron increased chlorophyll a in most experiments but affected total phytoplankton standing crop only during the summer and fall. Silicon had negative effects in some experiments. Cations and trace elements produced marked differences between lakes for chlorophyll a, but total phytoplankton standing crop showed few significant responses. Synergistic responses to two- and three-factor interactions were observed in all lakes. Differences in the responses of phytoplankton taxonomic divisions to enrichment may be responsible for much of the between lake variation in chlorophyll a and total phytoplankton volume responses. Nutrient limitations in these lakes are discussed and related to limnological factors and predictive models.  相似文献

Recently, we found the presence of chlorophyll a, chlorophyll d′ and pheophytin a as minor pigments in the chlorophyll d-dominated cyanobacterium Acaryochloris marina. Chlorophyll a determinations were made on lakewater and algal samples by spectrophotometric, fluorometric and high performance liquid chromatographic methods. Acetone, methanol and ethanol solvents were evaluated for their ability to extract photosynthetic pigments from Scenedesmus sp. cultures. Routinely used methods overestimated the chlorophyll a concentrations present in the samples. Significant differences resulted when various standard equations were used to calculate chlorophyll a concentrations. Acetone did not quantitatively extract chlorophyll pigments, even after 24 h. Mechanical disruption was found to be important in assuring complete extraction of the chlorophyll pigments.  相似文献

1. Nutrient and chlorophyll a levels, and bacterial numbers of 84 glacial lakes in the Tatra Mountains (Slovakia and Poland, Central Europe) were determined to assess the impact of catchment vegetation and water acidity on lake trophic status. 2. Catchment vegetation was the crucial factor governing nutrient content of lakes. 3. Concentrations of organic carbon, organic nitrogen, and chlorophyll a, and bacterial numbers were tightly correlated with total phosphorus (TP) content. Their levels were the highest in forest lakes, then decreased in alpine lakes with decreasing amount of catchment vegetation and soil cover, and were the lowest in lakes situated in bare rocks. 4. The above pattern was further modified by lake water acidity. Concentrations of TP, organic carbon, and chlorophyll a were lower in alpine lakes with pH between 5 and 6 than in more or less acid alpine lakes. Zooplankton was absent in all alpine lakes with pH between 5 and 6. 5. Nitrate concentrations followed an inverse trend to TP; lowest values were in forest lakes, then increased with decreasing amount of catchment soils and vegetation. Within the lakes of the same type of catchment vegetation, nitrate concentrations were negatively correlated to TP. N‐saturation of catchment areas and lake primary production were dominant processes controlling nitrate levels in lakes and nitrate contribution to lake acidification.  相似文献

In this article we report the characterization of the energy transfer process in the reconstituted isoforms of the plant light-harvesting complex II. Homotrimers of recombinant Lhcb1 and Lhcb2 and monomers of Lhcb3 were compared to native trimeric complexes. We used low-intensity femtosecond transient absorption (TA) and time-resolved fluorescence measurements at 77 K and at room temperature, respectively, to excite the complexes selectively in the chlorophyll b absorption band at 650 nm with 80 fs pulses and on the high-energy side of the chlorophyll a absorption band at 662 nm with 180 fs pulses. The subsequent kinetics was probed at 30–35 different wavelengths in the region from 635 to 700 nm. The rate constants for energy transfer were very similar, indicating that structurally the three isoforms are highly homologous and that probably none of them play a more significant role in light-harvesting and energy transfer. No signature has been found in the transient absorption measurements at 77 K for Lhcb3 which might suggest that this protein acts as a relative energy sink of the excitations in heterotrimers of Lhcb1/Lhcb2/Lhcb3. Minor differences in the amplitudes of some of the rate constants and in the absorption and fluorescence properties of some pigments were observed, which are ascribed to slight variations in the environment surrounding some of the chromophores depending on the isoform. The decay of the fluorescence was also similar for the three isoforms and multi-exponential, characterized by two major components in the ns regime and a minor one in the ps regime. In agreement with previous transient absorption measurements on native LHC II complexes, Chl b → Chl a energy transfer exhibited very fast channels but at the same time a slow component (ps). The Chls absorbing at around 660 nm exhibited both fast energy transfer which we ascribe to transfer from ‘red’ Chl b towards ‘red’ Chl a and slow transfer from ‘blue’ Chl a towards ‘red’ Chl a. The results are discussed in the context of the new available atomic models for LHC II.  相似文献

Summary High Performance Liquid Chromatography analysis of algal pigments from inter- and subtidal (deep and shallow) sediments from the Kerguelen Islands showed clear differences in the pigment composition at the different stations. High concentrations of chlorophyll c and fucoxanthin were present at all locations, indicating significant diatom densities, chlorophyll b was detected at all sites. At one station the other green algal pigments were also present; here green algae contributed more to chlorophyll a concentrations than diatoms, as estimated by using pigment ratios and microscopic observations. At another location chlorophyll b was associated with a high concentration of diadinoxanthin, indicating an abundance of euglenoids. This indicates that chemotaxonomy can be powerful tool in microphytobenthos studies since enumeration of living cells are difficult as many algae are attached to sediment particles (epipsammic algae). Ways of diagenesis, carotenoid degradation and the role of grazing are briefly mentioned. Phaeophorbide a-like pigments were the most significant chlorophyll a degradation products, with concentrations up to 110 g · g–1 dry weight sediment, i.e. 10 times the chlorophyll a concentration. Some taxonomic estimations, based on pigments ratios, and their limits, are discussed.  相似文献

Budgets for chlorophyll a and selected degradation products were constructed during summer stratification in 1984 and 1985 in two lakes. In Paul Lake, the reference ecosystem, pigment sedimentation showed no significant interannual differences. In Tuesday Lake, fish manipulations in May 1985 changed the zooplankton from an assemblage of Bosmina and small cyclopoids to one of large cladoceran grazers. Sedimentation of pigments, especially the grazing indicator pheophorbide a, increased significantly as the grazer assemblage changed. Mean grazer size was positively related to pheophorbide deposition rate in both lakes. Results of this whole-lake experiment indicate that major changes in lake food webs alter pigment deposition rates.  相似文献

Peridinin–chlorophyll–protein (PCP), containing differently absorbing chlorophyll derivatives, are good models with which to study energy transfer among monomeric chlorophylls (Chls) by both bulk and single-molecule spectroscopy. They can be obtained by reconstituting the N-terminal domain of the protein (N-PCP) with peridinin and chlorophyll mixtures. Upon dimerization of these “half-mers”, homo- and heterochlorophyllous complexes are generated, that correspond structurally to monomeric protomers of native PCP from Amphidinium carterae. Heterochlorophyllous complexes contain two different Chls in the two halves of the complete structure. Here, we report reconstitution of N-PCP with binary mixtures of Chl a, Chl b, and [3-acetyl]-Chl a. The ratios of the pigments were varied in the reconstitution mixture, and relative binding constants were determined from quantification of these pigments in the reconstituted PCPs. We find higher affinities for both Chl b and [3-acetyl]-Chl a than for the native pigment, Chl a.  相似文献

Nine lakes in northern Wisconsin were sampled from February through September 1996, and HPLC analysis of water column pigments was carried out on epilimnetic seston. Pigment distributions were evaluated throughout the water column during summer in Crystal Lake and Little Rock Lake. The purpose of our study was to investigate the use of phytopigments as markers of the main taxonomic groups of algae. As a first approach, multiple regression of marker pigments against chlorophyll a (chl a) was used to derive the best linear combination of the main xanthophylls (peridinin, fucoxanthin, alloxanthin, lutein, and zeaxanthin). A significant regression equation (r2= 0.98) was obtained for epilimnion data. The good fit indicates that the chl a:xanthophyll ratios were fairly constant in the epilimnion of the nine lakes over time. Chlorophyll a recalculated from the main xanthophylls in each sample showed good agreement with measured chl a in epilimnetic waters. A second approach used the CHEMTAX program to analyze the same data set. CHEMTAX provided estimates of chl a biomass for all algal classes and allowed distinction between diatoms and chrysophytes, and between chlorophytes and euglenophytes. These results showed a reasonably good agreement with biomass estimates from microscope counts, despite uncertainties associated with differences in sampling procedure. Changes of pigment ratios over time in the epilimnetic waters were also investigated, as well as differences between surface and deep samples of Little Rock Lake and Crystal Lake. We found evidence that changes in the ratio of photoprotective pigments to chl a occurred as a response to changes in light climate. Changes were also observed for certain light‐harvesting pigments. The comparison between multiple regression and CHEMTAX analyses for inferring chl a biomass from concentrations of marker pigments highlighted the need to take account of variations in pigment ratio, as well as the need to acquire additional data on the pigment composition of planktonic algae.  相似文献

Benthic microbial mat communities were sampled from 20 lakes, ponds and streams of the McMurdo Sound region, Antarctica. At least five distinct assemblages could be differentiated by their cyanobacterial species composition, pigment content and vertical structure. The most widely occurring freshwater communities were dominated by thin-trichome (0·5–3 µm) oscillatoriacean species that formed benthic films up to several millimetres thick. ‘Lift-off mats’ produced mucilaginous mats 1–5 cm thick at the surface and edge of certain ponds. Another group of oscillatoriacean communities was characteristic of hypersaline pond environments; these communities were dominated by species with thicker trichomes such as Oscillatoria priestleyi. Black mucilaginous layers of Nostoc commune were widely distributed in aquatic and semi-aquatic habitats. Dark brown sheath pigmentation was also characteristic of less cohesive mats and crusts dominated by Pleurocapsa, Gloeocapsa and Calothrix. High performance liquid chromatography analysis of the lipophilic pigments showed that the upper region of most of the Antarctic mats was enriched in sheath pigments (scytonemin) and/or certain carotenoids such as myxoxanthophyll and canthaxanthin. Most of the chlorophyll a (Chla), as well as phycocyanin, β-carotene and echinenone, was located in the lower strata of the mat profiles. In many of these communities most of the photosynthetic biomass occurred in a ‘deep Chla maximum’ that was well protected from short-wavelength radiation by the surface layer of light-screening pigments.  相似文献

We investigated the shade adaptation of a seasonally well-developed ice algal community in thin sea ice at Saroma-Ko Lagoon, Hokkaido, Japan on 3–4 March 2006 and 4–5 March 2007, by examining photosynthetic pigment concentrations, the chlorophyll a-specific light-absorption coefficient (a ph *), and the light-saturation index (E k ). The high proportions of photosynthetic pigments, including chlorophyll a, fucoxanthin, and chlorophyll c, and the low values of a ph *(440) and a ph *(675) suggested that the lagoon’s ice algal community was shade-adapted. The high ratio of E k to total photosynthetically active radiation (PAR) in the ice algal habitat suggested that the degree of shade adaptation is weak. Scaling of E k to total PAR could be extended to studies of the degree of photoadaptive succession of ice algal communities in the Northern Hemisphere. The degree of shade adaptation of ice algal communities in the Northern Hemisphere might be related to ice thickness, regardless of latitude.  相似文献

Iron is an important factor in algal blooms because it is involved in cyanobacterial pigment biosynthesis and therefore has the ability to influence the pigment status of algal cells. This role in pigment biosynthesis offers the opportunity for rapid monitoring of iron availability to cyanobacteria through spectral reflectance characterization. In the present study, the freshwater cyanobacterium Microcystis viridis was cultured with different levels of iron. Cell density, cellular content of iron and photosynthetic pigments, and spectral reflectivity of M. viridis were determined daily during the course of the culture experiment. The results showed that at the lowest iron concentration (0.01 μM) the growth of M. viridis was seriously limited, and the maximal cell density was only approximately 6.4% of the density observed with an iron concentration of 18 μM. Iron availability dramatically affected chlorophyll a, carotenoid and phycocyanin content, with the greatest impact on chlorophyll a. The iron‐induced changes in content and ratios of pigments were detectable through spectral reflectance. Eleven spectral indices previously developed for the estimation of concentrations and/or ratios of pigments and a newly proposed chlorophyll a/phycocyanin index were found to be suitable for generating sensitive regression models between cellular iron content and spectral parameters. The comprehensive application of key sensitive spectral indices and regression equations should help to support monitoring and diagnosis of iron availability to cyanobacteria via remote sensing.  相似文献

An HPLC analysis of the summer phytoplankton assemblage in Lake Baikal   总被引:5,自引:0,他引:5  
1. The enormous size and spatial heterogeneity of Lake Baikal require rapid methods for large sample sets. We therefore tested the applicability of a novel, high‐performance liquid chromatography (HPLC)‐based, combination of methods for analysing phytoplankton. In July 2001, samples were collected in a transect across the lake at various depths down to 30 m. Phytoplankton (>3 μm) and autotrophic picoplankton (APP) were counted under light and epifluorescence microscopes, respectively. Pigments were analysed with HPLC. 2. The pigment data allowed the contributions of the dominant phytoplankton groups to the total chlorophyll a (Chl a) in the lake to be estimated by multiple linear regression and by the CHEMTAX matrix factorisation program. Three marker pigments, fucoxanthin, lutein and zeaxanthin, were shown to be useful indicators of the abundance and spatial distribution of certain phytoplankton groups. The relative contributions of the various phytoplankton groups to the total Chl a in the lake determined using these marker pigments were similar, but not identical, to those determined by cell counts. 3. Pigment analyses of isolated strains from Lake Baikal and some European lakes confirmed that phycoerythrin‐containing Cyanobacteria with very high amounts of zeaxanthin were responsible for the low Chl a/zeaxanthin ratios of the water samples. A picoplanktonic species of Eustigmatophyceae was isolated from the lake. Its high violaxanthin content, responsible for very low Chl a/violaxanthin ratios of some water samples, can be used to estimate the contribution of this group to total Chl a.  相似文献

The lipophilic photosynthetic pigments in Limnothrix redekei, Planktothrix agardhii (cyanobacteria), Stephanodiscus minutulus, Synedra acus (diatoms), Scenedesmus acuminatus, and Scenedesmus armatus (chlorophycean) all isolated from an eutrophic lake were quantitatively determined by HPLC. The algae were grown semi-continuously under nutrient sufficient conditions at 20°C at a 12/12 h light/dark cycle with constant irradiance or with simulated natural light fluctuations as well as at a 6/18 h light/dark cycle with constant irradiance, all at the same daily light exposure. The zeaxanthin and the myxoxanthophyll contents of cyanobacteria were not influenced by fluctuating light, a short photoperiod or a different sampling time. The chlorophyll b/a ratio, the lutein/chlorophyll a ratio, and the neoxanthin content of chlorophycean as well as the chlorophyll c/a and the fucoxanthin/chlorophyll a ratio of diatoms were only slightly influenced by these factors. Therefore in some cases marker pigment contents and in other cases marker pigment/chlorophyll a ratios may be more useful for quantifying the relative importance of different taxonomic groups in natural phytoplankton. Simulated natural light fluctuations or the length of the photoperiod only slightly influenced the pigment content or the marker pigment/chlorophyll a ratio.  相似文献
