Summary Crude protein extracts from single seeds of nondomesticated Mexican bean accessions were analysed by SDS polyacrylamide gel electrophoresis for variability in phaseolin protein. Six new phaseolin types; M1, M2, M3, M4, M5, M6, which contained polypeptides within the same range of molecular weights (51,000 to 45,000 daltons) as occur in the S, T and C phaseolin types of cultivated beans were identified. No T and C types were found among the non-domesticated Mexican accessions, and the S type occurred in less than 7% of the seeds screened. Genetic analyses of F2 progenies from crosses between Sanilac (S), and five of the M types showed that each M phaseolin phenotype was allelic to the S type and expressed codominantly.  相似文献

Summary The self-condensation of 2(3)-O-glycyl esters of adenosine, adenosine-5-(O-methylphosphate) and P1, P2-diadenosine-5-pyrophosphate in 6.2 mM solutions at pH 8.0 and -5°C in the presence of 12.5 mM poly(U) yields approximately 3 times as much diketopiperazine as reactions without poly(U). As the concentration of 2(3)-O-(glycyl)-P1, P2-diadenosine-5-pyrophosphate is decreased from 6.2 mM to 1.5 mM the yield of diketopiperazine in the presence of poly(U) decreases slightly from 6.6% to 5.2%, whereas, in the absence of poly(U) the yield of diketopiperazine decreases substantially from 2.4% to 0.75%. The enhanced yield of diketopiperazine that is attributed to the template action of poly(U) is temperature dependent and is observed only at temperatures below 10°C (5°C to -5°C) for 6.2 mM 2(3)-O-(glycyl)-adenosine-5-(O-methylphosphate) and below 23°C (15°C to -5°C) for 6.2 mM 2(3)-O-(glycyl)-P1, P2-diadenosine-5-pyrophosphate. The absence of a template effect at high temperatures is attributed to the melting of the organized helices. The hydrolysis half-lives at pH 8.0 and -5°C of 2(3)-O-(glycyl)-adenosine, 2(3)-O-(glycyl)-adenosine-5-(O-methylphosphate), 2(3)-O-(glycyl)-P1, P2-diadenosine-5-pyrophosphate, and 5-O-(glycyl)-adenosine in the presence of poly(U) are substantially larger than their half-lives in the absence of poly(U). Studies of molecular mechanisms of chaperone-like activity of -crystallin became an active field of research over last years. However, fine interactions between -crystallin and the damaged protein and their complex organization remain largely uncovered. Complexation between - and L-crystallins was studied during thermal denaturation of L-crystallin at 60°C using small-angle X-ray scattering (SAXS), light scattering, gel-permeation chromatography, and electrophoresis. A mixed solution of - and L-crystallins at concentrations about 10 mg/ml incubated at 60°C was found to contain their soluble complexes with a mean radius of gyration 14 nm, mean molecular mass 4 MDa and maximal size over 40 nm. In pure L-crystallin solution, no complexes were observed at 60°C. In SAXS studies, transitions in the -crystallin quaternary structure at 60°C were shown to occur and result in doubling of the molecular weight. This suggests that during the temperature-induced denaturation of L-crystallin it binds with modified -crystallin or, alternatively, L-crystallin complexation and -crystallin modifications are concurrent. Estimates of the -L-crystallin complex size and relative contents of - and -L-crystallins in the complex suggest that several -crystallin molecules are involved in complex formation.  相似文献

Temperature dependencies were established for the egg-to-mummy and mummy-to-adult phases, for mummy mortality, and for parasitism of Aphidius ervi Haliday, Aphidius rhopalosiphi De Stefani-Perez, and Praon volucre (Haliday) (Hymenoptera, Aphidiidae), three parasitoids of Sitobion avenae (Fabricius) (Homoptera, Aphididae), at 8°C, 12°C, 16°C, 20°C, and 25°C on winter wheat (cv. Haven). A physiological model described temperature-dependent development over the full temperature range, whereas a linear model was fitted for data above 8°C and used to estimate the lower temperature thresholds and day-degrees (° D) required for development. The thresholds for A. ervi were 2.2°C for egg-mummy development and 6.6°C for mummy-adult development, those for A. rhopalosiphi were 4.5°C and 7.2°C, and those for P. volucre were 3.8°C and 5.5°C. The time to develop into mummies and adults differed significantly between the three species: A. ervi development into mummies required an average of 159 ° D, while development into adults took an average of 73 ° D. The corresponding average times required for A. rhopalosiphi and P. volucre to develop mummies were 124° D and 126° D, while their development into adults required an average of 70° D and 150° D, respectively. Mummy mortality was 25–35% at 8°C and less at the higher temperatures tested, but began to increase again at 25°C, showing a quadratic relationship between mortality and temperature. Parasitization was very low or, in the case of P. volucre, absent up to 12°C and thereafter increased with increasing temperature. The relationship between parasitization, recorded as percent aphids mummified, and temperature was linear at the temperatures tested and depended on species. A. ervisuperparasitized 11.1% aphids at 20°C and 16.6% aphids at 25°C, whereas superparasitism was low in A. rhopalosiphi and absent in P. volucre. From 16°C to 25°C the P. volucre sex ratio increased. For A. ervi and A. rhopalosiphi there was no trend with temperature, but at 20°C and 25°C it was close to even. Field data for 1996 and 1997 allowed for a comparison of actual and expected emergence of overwintering mummies. In both years, parasitoids were predicted to have emerged from overwintering mummies well in advance of the onset of aphid infestation, and more than a month earlier than the first parasitized aphids were found in winter wheat. Observations from trap plants in other crops supported the predictions of the models. Other factors that can affect biological control by cereal aphid parasitoids are discussed.  相似文献

The three-dimensional structure in aqueous solution of native huwentoxin-I, a neurotoxin from the venom of the spider Selenocosmia huwena, has been determined from two-dimensional 1H NMR data recorded at 500 and 600 MHz. Structural constraints consisting of interproton distances inferred from NOEs and dihedral angles from spin–spin coupling constants were used as input for distance geometry calculation with the program XPLOR 3.1. The best 10 structures have NOE violations <0.3 Å, dihedral violations <2°, and pairwise root-mean-square differences of 1.08 (±0.20) Å over backbone atoms (N, C;, C). The molecule adopts a compact structure consisting of a small triple-stranded antiparallel -sheet and five -turns. A small hydrophobic patch consisting of Phe 6, Trp 28, and Trp 31 is located on one side of the molecule. All six lysine residues are distributed on the molecular surface. The three disulfidc bridges are buried within the molecule. The structure contains an inhibitor cystine knot motif which is adopted by several other small proteins, such as -conotoxin, agatoxin IVA, and gurmarin.  相似文献

Zusammenfassung In Temperaturkabinen wird die Fortpflanzungspotenz von Myzus persicae (Sulzer) (Herkunft Groß-Lüsewitz) bei Dauertemperaturen von 15 bis 30° auf Kohlrüben (Brassica napus subspec. rapifera) untersucht.Eine Populationsanalyse nach Birch (1948) (intrinsic rate of increase) ergab den höchsten Wachstumsfaktor bei Dauertemperaturen zwischen 20 und 23°.Dauertemperaturen >25° führten zu einer starken Minderung der. Fortpflanzungspotenz. 30° ist die obere Grenze der Fortpflanzung der untersuchten Myzus persicae-Population.
The reproductive potential of the peach-potato aphid (origin Gross-Lüsewitz) was studied at temperatures between 15° and 30° in constant temperature chambers. They were cultivated on Swede (Brassica napus spp. rapifera) which stood in Knop's nutrient under gauze cloches in petri dishes. The production of juvenile larvae and the mortality of the mothers was measured daily. The total of all larvae (including those which were dropped) and the total of larvae on the leaf were separately enumerated. The larvae on the leaf were designated as viable larvae. A population analysis using Birch's method showed a maximum value for the growth factor k (difference between birth and mortality rates) of 23° for the total of all larvae, and of 20° for the viable larvae (Fig. 6). The daily relative growth-ratio was at the same temperatures respectively 1.36 and 1.34 (Table IV). Optimum development of M. persicae on swedes occurs thus between 20° and 23°. The percentages of viable larvae which add to the net production of total larvae are 53, 61, 30, and 24 (Table III) at temperatures of 15, 20, 25, and 30° respectively. The average length of a generation was 18.5 days at 15° and less than 13 days at 28 to 30° (Fig. 5). The multiplication rate per generation was 38 at 15°, 48 at 20°, but only 5.5 at 30° (Fig. 4). The time of development from first-stage larva to adult was 12.5 days at 15°, 5 days at 28° and 6 days at 30° (Table VII). The upper limit, where a weak multiplication was still possible, was at 30°. It is concluded that in regions where such limiting temperatures occur during some part of the day, the temperature can be the major regulating factor of the insect populations.

Zusammenfassung In der Neurohypophyse fetaler und neugeborener Ratten entsteht die Mehrzahl der synaptischen Bläschen aus Erweiterungen der Neurotubuli. Ferner können pinocytotische Bläschen als synaptische Vesikel imponieren. Die Bläschenbildung aus Membranen von Elementargranula (vgl. Herlant, 1967) tritt dagegen in den Hintergrund. Ein Auftreten von Vesikeln im Innern von Elementargranula wurde nicht beobachtet.
The origin of synaptic vesicles in neurosecretory axons
Summary In the neurohypophysis of fetal and newborn rats the majority of synaptic vesicles originate from dilatations of neurotubuli. Moreover, pinocytotic invaginations give rise to synaptic vesicles. Evaginations of elementary granule membranes, as described by Herlant (1967), are seldom to be found and do not seem to play an important role in the formation of synaptic vesicles. The occurrence of vesicles within elementary granules was not observed.

A new computer program, HYPER, has been developed for automated analysis of protein dihedral angle values and CH2 stereospecific assignments from NMR data. HYPER uses a hierarchical grid-search algorithm to determine allowed values of , , and 1 dihedral angles and CH2 stereospecific assignments based on a set of NMR-derived distance and/or scalar-coupling constraints. Dihedral-angle constraints are valuable for restricting conformational space and improving convergence in three-dimensional structure calculations. HYPER computes the set of , , and 1dihedral angles and CH2 stereospecific assignments that are consistent with up to nine intraresidue and sequential distance bounds, two pairs of relative distance bounds, thirteen homo- and heteronuclear scalar coupling bounds, and two pairs of relative scalar coupling constant bounds. The program is designed to be very flexible, and provides for simple user modification of Karplus equations and standard polypeptide geometries, allowing it to accommodate recent and future improved calibrations of Karplus curves. The C code has been optimized to execute rapidly (0.3–1.5 CPU-sec residue–1 using a 5° grid) on Silicon Graphics R8000, R10000 and Intel Pentium CPUs, making it useful for interactive evaluation of inconsistent experimental constraints. The HYPER program has been tested for internal consistency and reliability using both simulated and real protein NMR data sets.  相似文献

The magnitude of the proton motive force (p) and its constituents, the electrical () and chemical potential (-ZpH), were established for chemostat cultures of a protease-producing, relaxed (rel ) variant and a not protease-producing, stringent (rel +) variant of an industrial strain ofBacillus licheniformis (respectively referred to as the A- and the B-type). For both types, an inverse relation of p with the specific growth rate was found. The calculated intracellular pH (pHin) was not constant but inversely related to . This change in pHin might be related to regulatory functions of metabolism but a regulatory role for pHin itself could not be envisaged. Measurement of the adenylate energy charge (EC) showed a direct relation with for glucose-limited chemostat cultures; in nitrogen-limited chemostat cultures, the EC showed an approximately constant value at low and an increased value at higher . For both limitations, the ATP/ADP ratio was directly related to .The phosphorylation potential (G'p) was invariant with . From the values for G'p and p, a variable H+/ATP-stoichiometry was inferred: H+/ATP=1.83+0.52µ, so that at a given H+/O-ratio of four (4), the apparent P/O-ratio (inferred from regression analysis) showed a decline of 2.16 to 1.87 for =0 to max (we discuss how more than half of this decline will be independent of any change in internal cell-volume). We propose that the constancy of G'p and the decrease in the efficiency of energy-conservation (P/O-value) with increasing are a way in which the cells try to cope with an apparent less than perfect coordination between anabolism and catabolism to keep up the highest possible with a minimum loss of growth-efficiency. A method is described whereby arrays of samples ofClupea pallasi eggs may be stored during their preparation. The high fertilization potential retained by the eggs during short-term storage allows them to be fertilized synchronously when sample preparation is complete. A variation of the dry method of storage retained maximum fertilization potential (80–85%) of the eggs for about 1 hr, and of milt dilution (18 with 17 S sea water), about 7 hr. Following dry storage, eggs fertilized in salinities of 0–45 showed maximum rates of fertilization in salinities of 10–20, and fertilization rates 50% in salinities of 4.5–42. Salinities of fertilization influenced egg diameter, median hatching time, and larval length at hatching in egg samples transferred 21/2 hr after fertilization to an incubation salinity of 17 at 7°C. Fertilization rates were higher (90–95%) for eggs stored in 17 S at 7°C prior to fertilization. Under such wet storage conditions, maximum fertilization pontential was retained for about 2 hr. Highest fertilization rates (95–96%) were obtained for eggs stored and fertilized in salinities of 12–15. For the species and the area of origin considered (British Columbia), wet storage of eggs should result in maximum fertilization when the eggs are stored at 4°C for a period not greater than 2 hr prior to fertilization in the 12–15 S storage medium.Prepared under the auspices of the Canadian-German Scientific and Technical Cooperation Agreement.  相似文献

Summary Flash illumination alters the voltage across bilayer lipid membranes in the presence of certain cyanine dyes. The waveforms of the photo-voltage vary systematically with dye structure and imposed transmembrane voltage. Experimental results are reported for 27 positively charged cyanine dyes, primarily oxazole derivatives, using lecithin/oxidized cholesterol bilayer membranes and 10-mm sodium chloride solutions. Several dyes do not induce any photo-voltages. Examples are 3,3 diethyl 9 ethyl 2,2 oxacarbocyanine iodide, 3,3 diethyl 2 oxa 2 thiacyanine iodide, and 3,3 dimethyl 2,2 indocarbocyanine iodide. Several dyes, when added to one side of the membranes, induce monophasic waveforms. Examples are 3,3 dimethyl 2,2 oxacarbocyanine chloride, and 3,4,3,4 tetramethyl 2,2 oxazalinocarbocyanine iodide. Other dyes induce a photo-voltage only if transmembrane voltages are imposed. These waveforms are biphasic with some dyes (3,3 diethyl 2,2 oxacarbocyanine iodide, for example) and monophasic with other dyes (3,3 dibutyl 2,2 oxacarbocyanine iodide, for example).The photo-voltage waveforms are explained by models that consider the movement of charged dye molecules within the membrane, following optical excitation. The dye movements are probably induced through charge rearrangements in the dye associated with long-lived triplet states, isomerization, or through excimer formation. These results provide information on the location and orientation of the dye molecules within bilayer membranes. The variations which occur in the waveforms with applied voltage indicate that these membranes are fluid in the direction perpendicular to the membrane plane.  相似文献

A simple one variable caricature for oscillating and excitable reaction-diffusion systems is introduced. It is shown that as a parameter, , varies the system dynamics change from oscillatory ( > 0) to excitable ( < 0) and the frequency of the oscillation vanishes as for 0. When such dynamics are coupled by continuous diffusion in a ring geometry (1-space dimension), propagating wave trains may be found. On an infinite ring excitable devices lead to unique solitary waves which are analogous to pulse waves. A solvable example is presented, illustrating properties of dispersion, excitability, and waves. Finally it is shown that the caricature arises in a natural way from more general excitable/oscillatory systems.  相似文献

Internal motions of d-ribose selectively 2H-labeled at the 2 position were measured using solid state 2H NMR experiments. A sample of d-ribose-2 -d was prepared in a hydrated, non-crystalline state to eliminate effects of crystal-packing. Between temperatures of –74 and –60°C the C2–H2 bond was observed to undergo two kinds of motions which were similar to those of C2–H2/H2 found previously in crystalline deoxythymidine (Hiyama et al. (1989) J. Am. Chem. Soc., 111, 8609–8613): (1) Nanosecond motion of small angular displacement with an apparent activation energy of 3.6 ± 0.7 kcal mol–1, and (2) millisecond to microsecond motion of large amplitude with an apparent activation energy 4 kcal mol–1. At –74°C, the slow, large-amplitude motion was best characterized as a two-site jump with a correlation time on the millisecond time scale, whereas at –60°C it was diffusive on the microsecond time scale. The slow, large-amplitude motions of the C2–H2 bond are most likely from interconversions between C2-endo and C3-endo by way of the O4-endo conformation, whereas the fast, small-amplitude motions are probably librations of the C2–H2 bond within the C2-endo and C3-endo potential energy minima.  相似文献

Summary One hundred and twenty-two varieties, lines and wild accessions of Lycopersicon were screened under three different regimes during the autumn/winter season of 1982–83 and 1983–84 for resistance to tomato leaf curl virus (TLCV). L. hirsutum f. glabratum (B6013) and L. hirsutum f. typicum (A1904) proved to be highly resistant to TLCV in all three environments. Various accessions of L. peruvianum were also highly resistant. L. pimpinellifolium (A1921) exhibited no TLCV symptoms within 90 days. Of the cultivated varieties, Acc 99 exhibited the minimim score for susceptibility; AC 142, Collection No. 2, Kalyanpur Angurlata and HS 101 had a low rating for virus incidence. The inheritance of resistance was studied in the interspecific crosses between a TLCV resistant line of L. pimpinellifolium (A1921) and five (HS 101, HS 102, HS 110, Pusa Ruby and Punjab Chhuhara) susceptible cultivars of L. esculentum. Parents, F1, F2 and backcross progenies were artificially inoculated with local strains of TLCV using vector the viruliferious whitefly, Bemisia tabaci (Genn.). Data indicated that the resistance of L. pimpinellifolium (A 1921) is monogenic and incompletely dominant over susceptibility.  相似文献

Summary Respiration of an undescribed species of soil nematode of the genus Chiloplacus from the Canadian High Arctic was measured at 2°, 5°, 10°, 15°, 20° and 25°C. The corresponding metabolic rates were 0.2697×10-3 l, 0.3406×10-3 l, 0.8408×10-3 l, 0.8539×10-3 l, 1.8420×10-3 l and 2.9360×10-3 l O2 ind-1 h-1, respectively, for a nematode of 1.0 g dry weight. The relationship between respiration and dry weight for Chiloplacus sp. at 10°C is described by the function log R=-3.0693+0.8844 log W. Q10 values for the 2°–5°, 5°–10°, 10°–15°, 15°–20° and 20°–25°C temperature intervals were 2.18, 6.09, 1.03, 4.65 and 2.54, respectively. Chiloplacus sp. showed raised metabolic rates at low tempetatures compared with species from warmer environments. Metabolic rates of representative samples of the soil, nematode fauna (dominated by individuals of the genus Plectus) from the same location were 0.1593×10-3 l, 0.3603×10-3 l and 0.5332×10-3 l O2 ind-1 h-1 at 5°, 10° and 15°C for an average nematode of 0.4297 g dry weight.  相似文献

The solid-state conformation of the first N-protected ornithine derivative has been established by X-ray analysis. The hydrochloride of N-(9-fluorenyl)methoxycarbonyl-L-ornithine crystallises as diethyl ether solvate. The backbone (0, , , 1) torsion angles are (174.9°, –84.0°, 145.9°, –171.0°). The conformation of the urethane amide bond is trans. The ornithine aliphatic side chain adopts preferred fully extended conformation which is stabilised by the hydrogen bonding of the –NH3 + group to the diethyl ether molecule, carboxyl group and Cl- anions.  相似文献

Summary The ultrastructure of spermatogenesis is described for the first time in an aspidogastrean. The zone of differentiation which is usually formed during digenean spermiogenesis was not observed inMulticotyle purvisi. Instead, spermatid components are assembled within the common cytoplasmic mass before the outgrowth of spermatids. Microtubules, mitochondrion, nucleus and axonemes including their basal body regions, migrate from the cytoplasm into the spermatid which is pinched off at the level of the arching membrane. An unusual, complex structure of the basal body region is described. Intercentriolar bodies and striated rootlets are left behind and quickly disappear from the residual cytoplasm. Summary An alkalophilic bacterium producing high amounts of the cell-associated -mannosidase and extracellular -mannanase was isolated from soil. The isolate (AM-001) that grew well in alkaline pH media was identified as a strain of Bacillus sp. The optimal cultivation temperature for enzyme production was 31° C for -mannosidase and 37° C for -mannanase with the optimum production medium composed of 1% konjac powder, 0.2% yeast extract, 2% Polypepton, 0.1% K2HPO4, 0.02% MgSO4 · 7H2O and 0.5% Na2CO3. The relation between the quantum yield of oxygen evolution of open photosystem II reactions centers (p), calculated according to Weis and Berry (1987), and non-photochemical quenching of chlorophyll fluorescence of plants grown at 19°C and 7°C was measured at 19°C and 7°C. The relation was linear when measured at 19°C, but when measured at 7°C a deviation from linearity was observed at high values of non-photochemical quenching. In plants grown at 7°C this deviation occurred at higher values of non-photochemical quenching than in plants grown at 19°C. The deviations at high light intensity and low temperature are ascribed to an increase in an inhibition-related, non-photochemical quenching component (qI).The relation between the quantum yield of excitation capture of open photosystem II reaction centers (exe), calculated according to Genty et al. (1989), and non-photochemical quenching of chlorophyll fluorescence was found to be non-linear and was neither influenced by growth temperature nor by measuring temperature.At high PFD the efficiency of overall steady state electron transport measured by oxygen-evolution, correlated well with the product of q N and the efficiency of excitation capture (exe) but it deviated at low PFD. Changes in body temperature (Tb) and feeding were characterized in unrestrained rats following the micro-injection into the anterior hypothalamic preoptic area (AH/POA) of macrophage inflammatory protein-1 (MIP-1), MIP-1 or MIP-1. After the rats recovered from the stereotaxic implantation of a single guide tube placed in the AH/POA, either one of the MIP-1 compounds or control CSF was micro-injected in a volume of 1.0 l into this area. Changes in body temperature (Tb) and food and water intakes were monitored throughout each experiment. When micro-injected into the AH/POA in a dose of 28 or 280 pg, doublet MIP-1 and MIP-1 evoked a monophasic fever which increased above baseline to a mean maximum of 2.17±0.14°C and 2.1±0.24°C, respectively. MIP-1 micro-injected similarly evoked a biphasic fever, with the Tb declining transiently at the 30 min point 0.4°C lower than the congruent rises in Tb evoked by doublet MIP-1 or MIP-1. The secondary rise in Tb induced by MIP-1 had a latency of 1.5–2.0 hrs and reached a maximum of 1.56±0.16°C. Although all three cytokines significantly attentuated the rats' mean intake of food during the 24 hr interval after their micro-injection into the AH/POA, doublet MIP-1 exerted the most potent anorexic effect in comparison to that of the saline control rats. However, neither body weight nor intake of water was altered significantly by the three cytokines. These results demonstrate a functional distinction between the febrile actions of MIP-1 and MIP-1 on cells of the AH/POA. It is envisaged that MIP-1 underlies the initial phase of a pyrogen-induced fever, whereas MIP-1 could mediate the secondary phase of a biphasic fever. Clearly, the localized elevation of either MIP-1 or MIP-1 in the AH/POA perturbs the neuronal mechanism underlying the set-point for body temperature. Thus, both MIP-1 and MIP-1 could play a functional role in the pathological events occurring in neurons of the AH/POA in response to an endotoxin or other pyrogen challenge.  相似文献
