Various combinations of intermorph, selfing and intramorph pollinations were carried out in Averrhoa carambola and the pollinated pistils were observed under fluorescence and scanning electron microscopes in time-course experiments. In both compatible and incompatible pollinations, similar behavior of pollen germination and penetration was observed in the first 4 h after pollination. In compatible intermorph pollination, pollen tubes were found at the base of the transmitting tract of the style at 8 h and 24 h after pollination in both the pin and thrum morphs. With thrum flowers, selfing resulted in pollen tubes being uniformly arrested at the junction between the stigmatic and stylar tissues. Penetration of pollen tubes into the upper portion of style was observed in thrum intramorph pollination and, when the second member was treated as the gynoecial tissue under the same pollination, penetration of tubes was further enhanced. Pin flowers, on selfing, resulted in pollen tube penetration farther down the style than was the case with thrum selfing. Intramorph pollination of pin morph behaved in a similar manner to selfing and was not affected by genotypes. Beside the stamen-style dimorphism, the receptive surface of the cob stigma was larger in pin than thrum flowers. While pin pollen was round, thrum pollen was oblong in shape with pin to thrum ratio on the polar axis being 1.2 and on the equatorial axis 0.8. The stigma of pin morph belonged to the dry type, while that of the thrum resembled the wet type.  相似文献

Distyly, floral polymorphism frequently associated with reciprocal herkogamy, self‐ and intramorph incompatibility and secondary dimorphism, constitutes an important sexual system in the Rubiaceae. Here we report an unusual kind of distyly associated with self‐ and/or intramorph compatibility in a perennial herb, Hedyotis acutangula. Floral morphology, ancillary dimorphisms and compatibility of the two morphs were studied. H. acutangula did not exhibit precise reciprocal herkogamy, but this did not affect the equality of floral morphs in the population, as usually found in distylous plants. Both pin and thrum pollen retained relatively high viability for 8 h. The pollen to ovule ratio was 72.5 in pin flowers and 54.4 in thrum flowers. Pistils of pin flowers remained receptive for longer than those of thrum flowers. No apparent difference in the germination rate of pin and thrum pollen grains was observed when cultured in vitro, although growth of thrum pollen tubes was much faster than that of pin pollen tubes. Artificial pollination revealed that pollen tube growth in legitimate intermorph crosses was faster than in either intramorph crosses or self‐pollination, suggesting the occurrence of cryptic self‐incompatibility in this species. Cryptic self‐incompatibility functioned differently in the two morphs, with pollen tube growth rates after legitimate and illegitimate pollination much more highly differentiated in pin flowers than in thrum flowers. No fruit was produced in emasculated netted flowers, suggesting the absence of apomixis. Our results indicate that H. acutangula is distylous, with a cryptic self‐incompatibility breeding system.  相似文献

Summary In Primula obconica, a species with a heteromorphic self-incompatibility system, the distinction between compatible and incompatible pollen tubes takes place on the stigma surface in thrum flowers, self tubes growing randomly over the papillar cells. No differences were seen between self and cross tube behaviour on the pin stigma surface, but self tubes were inhibited within the stigmatic tissue with differences in tube length evident after 24 h. The stigma surface bears a proteinaceous pellicle and binds the lectin Concanavalin A. Removal of the stigma removes the incompatibility barrier in mature gynoecia. Bud pollination shows that pollen tubes cannot grow in a normal manner on immature stigmas; the random growth of tubes over the stigma surface resembles that of mature thrum selfs. Fewer compatible tubes reach the style base of young gynoecia and smaller numbers of seeds are set than in mature flowers. Pin and thrum pollen grains germinate and grow in aqueous media, thrum tubes growing longer than pin. The presence of H3BO4 and CaCl2 in the growth medium promotes tube elongation and lengths equivalent to compatible styles can be obtained. The pollen grains have proteinaceous materials in their walls which diffuse out on moistening. Prolonged washing in aqueous media removes these materials but the incompatibility reaction remains unchanged. Thus the incompatibility reaction is between pollen tubes and stigmatic tissue and differs from the homomorphic, sporophytic system where pollen wall proteins elicit the incompatibility response.  相似文献

Observations on the floral biology of Luculia gratissima (Rubiaceae)showed that this species is distylous with complementary positioningof anthers and stigmas in the two floral forms. Unusual featuresof distyly in this species include the larger size of the corolla,the stigma surface and the stigmatic papillae in the thrum flowerscompared to the pin ones. Stigmatic surfaces have similar secretionsbut they appear more copious in thrum than pin. The floral dimorphismwas accompanied by a very effective self-incompatibility systemand no seed was set on selfing. Seed number per capsule on crossingwas significantly greater in thrum flowers compared to pin.Incompatible pollen tubes were inhibited within 24 h at thebase of the stigma/top of the style in both morphs. Amputationof this region of the gynoecium removed the self-incompatibilityreaction in thrum but not pin flowers. Pollination with a mixtureof compatible and incompatible pollen and sequential pollinationwith self followed by cross pollen showed that there were interactionsbetween the two types of pollen tube. The presence of compatibletubes was found to cause the excessive swelling of the pollen-tubetip of the incompatible ones. The receptive surface of the stigma in distylous Linum grandiflorumand L. pubescens was studied by electron microscopy and cytochemicaltechniques. In both floral morphs a proteinaceous-lipoidal coatingis present on the papilla surface. In thrum stigmas the cuticleis highly irregular and pitted at the papilla tip. The cuticleis dislodged and torn at anthesis and an osmiophilic secretionproduct is released within a pectinaceous matrix. The secretionproduct stains for proteins and lipids and contributes to adhesionof pollen. In the larger pin papillae the cuticle is wavy, continuous,thicker than in thrum papillae and adjoins the cell wall. Inboth species the surface of the two types of pollen grains iscoated with lipids and protein. A similar behaviour of the male gametophyte is observed in incompatiblepollinations of L. pubescens, L. mucronatum and L. grandiflorum.In intramorph thrum pollinations pollen tubes are arrested withinthe stigma. In intramorph pin pollinations the majority of pollengrains fail to adhere to the stigma. Low permeability to waterin pin papillae, as determined by the neutral red test, maybe a factor preventing imbibition of the few adhering grains.Tubes of the few germinated grains are inhibited inside thestigma. On the part of the stigma, the difference in the major siteof inhibition in the two intramorph incompatible combinationsmay be accounted for by the dissimilar properties of the papillae,i.e. the occurrence of wet thrum stigmas and dry pin stigmas.Functionally, the unusual association of sporophytic incompatibilitywith wet thrum stigmas is attributed to retention of the secretorymaterial on individual papillae. Summary The stigmas of the heterostylous genusPrimula are of the dry type without a free-flowing surface secretion. The papillae of the stigma surface cells of the two morphs, in pin (stigma exserted) and thrum (stamens exserted), bear a thin proteinaceous surface pellicle, overlying a discontinuous cuticle. The vacuoles of the papillate cells contain tannins, and tannin cells extend in files through the stigma heads and form a loose sheath surrounding the pollen-tube transmitting tract in the styles. The cells of the transmitting tissue in the stigma heads have a normal complement of organelles, and abundant ribosomal endoplasmic reticulum. The intercellular spaces contain an internal secretion which reacts cytochemically for both carbohydrate and protein. The transmitting tract in the styles forms a central core surrounded by several vascular strands. The cells are elongated, and the intercellular spaces here also have a carbohydrate-protein content. In a compatible pollination, thrum pollen tubes enter the stigma by penetrating the cuticle at the tip or on the flank of the pin papilla. Pin tubes on the thrum stigma enter between adjacent papillae, penetrating the thin cuticle at the base. The tubes grow through the transmitting tracts in the intercellular material.  相似文献

In P. affinis, pin pollen is shorter on average than thrum pollen. Pins have more coronal hooks on stigmatic papillae than thrums, but gaps between papillae are relatively smaller for thrums. Pin stigmas receive more pollen than thrum stigmas. Thrum stigmas receive more (dissortive) pin pollen, but pin stigmas are assortively pollinated. Pollen only germinates when trapped below papilla coronas. On thrum stigmas, most trapped pollen is pin. Pollen germination is better on thrum stigmas than pin stigmas, and thrum stigmas show a close relationship between numbers of legitimate pollen grains, numbers of germinating grains, and numbers of pollen tubes in the style. There is no inhibition of illegitimate pollen germination. Illegitimate pollen tubes are inhibited in the style. Incompatibility operates by a combination of dissortive pollination, dissortive pollen trapping, and stylar pollen tube inhibition. All heteromorphic features differing between pins and thrums are implicated in the inhibition of within-morph fertilization in thrums.  相似文献

The distribution of callose plugs and pollen tubes was investigated following inter- and intramorph crosses of Amsinckia grandiflora (Boraginaceae), a distylous species possessing cryptic self-incompatibility. Callose plug distribution provided a good indication of the distribution of pollen tubes. Compared to intramorph crosses, many more callose plugs and pollen tubes were found in basal stylar regions following intermorph crosses, indicating that differential pollen tube growth is a likely cause of cryptic self-incompatibility. The incompatibility response differed for the floral morphs: in the pin (long-styled) morph pollen tubes were most likely to cease growth in the midstylar region, while inhibition was more likely to occur in the upper stylar region of the thrum (short-styled) morph. There was no evidence of stigmatic inhibition of pollen tubes for either morph, although the incompatibility response in the Boraginaceae is normally located in the stigmatic region.  相似文献

Observations on two natural and one artificial distylous populations ofPrimula veris in Britain revealed very high proportions of intramorph pollen on pin stigmas and moderate proportions of intermorph pollen on thrum stigmas. The average ratio of pin: thrum pollen production per flower was 2.4:1. Large amounts of pollen remained in dehisced anthers, perhaps reflecting low levels of pollinator activity. Population size and density had no clear effect on pollen flow patterns, which are asymmetrical for the two morphs.  相似文献

二型花柱的维持机制和自然选择压力多年来一直是生态学和进化学研究领域的热点之一。通过实验室栽培和野外观察统计相结合的方法,对安徽羽叶报春两种花型(长柱花和短柱花)的形态特征、花粉活力、柱头可授性、花粉/胚珠比、自然授粉及结籽能力、自交亲和性等繁育特性进行了比较研究。结果表明:长、短柱花的花冠直径和裂片宽无明显差异,而花冠筒、雌蕊和雄蕊高、花粉数目及大小、P/O比均有显著差异。在自然条件下,长柱花所接受的总花粉数要明显高于短柱花的总花粉数,但所接受的异型花花粉数和平均每果结籽数两者无显著差异。长柱花和短柱花的花粉和柱头活力相似,均能在较长时间内维持较高活力,仅在开花末期显著下降。两种花型的花在自花授粉、同型异花授粉、异型花授粉条件下均能结籽,但异型花授粉的结籽数均明显高于自花授粉和同型异花授粉结籽数。在长柱花各种授粉方式中,花粉萌发率无明显差异,但异型花花粉管的生长速度明显比同型异花花粉和自花花粉的快,而在短柱花柱头上表现为异型花授粉的萌发率最高,但只要萌发后在花柱中的生长速度无显明差异。此外,综合上述结果,对二种花型花部综合征的维持机制及自然选择压力进行了探讨。  相似文献

Hand pollinations experiments with distylous Amsinckia douglasiana A. DC. (Boraginaceae) revealed that legitimate (intermorph) crosses produce more seed than self-pollinations and that self-pollinations yield more seed than illegitimate (intramorph) crosses. We tested the occurrence of cryptic incompatibility in both style-length morphs by pollinating them with a mixture of legitimate and illegitimate pollen, each homozygous for a different electrophoretically distinguishable allele of phosphoglucose isomerase. The success of intramorph pollen was greatly reduced in mixture; only about 4% of the offspring of both pins and thrums were sired by illegitimate pollen. One pin and one thrum had sufficiently high seed set that the results cannot be attributed to selective abortion of embryos. In those individuals, discrimination against illegitimate pollen must have occurred on the stigma or in the style. Based on our findings, it appears that illegitimate matings are rare in natural populations. The progeny of legitimate crosses was biased toward pins, which suggests some regulation of morph ratios independent of the style-length locus. Prezygotic discrimination against the thrum genotype or selective abortion of embryos with the thrum genotype are both possible, but selective mortality of seedlings with the thrum genotype cannot be ruled out for some families.  相似文献

In flowering plants, penetration of the pollen tube through stigma, style, and transmitting tract is essential for delivery of sperm nuclei to the egg cells embedded deeply within female tissues. Despite its importance in plant reproduction, little is known about the underlying molecular mechanisms that regulate the navigation of the pollen tube through the stigma, style, and transmitting tract. Here, we report the identification and characterization of an Arabidopsis thaliana gene, VANGUARD1 (VGD1) that encodes a pectin methylesterase (PME)-homologous protein of 595 amino acids and is required for enhancing the growth of pollen tubes in the style and transmitting tract tissues. VGD1 was expressed specifically in pollen grain and the pollen tube. The VGD1 protein was distributed throughout the pollen grain and pollen tube, including the plasma membrane and cell wall. Functional interruption of VGD1 reduced PME activity in the pollen to 82% of the wild type and greatly retarded the growth of the pollen tube in the style and transmitting tract, resulting in a significant reduction of male fertility. In addition, the vgd1 pollen tubes were unstable and burst more frequently when germinated and grown on in vitro culture medium, compared with wild-type pollen tubes. Our study suggests that the VGD1 product is required for growth of the pollen tube, possibly via modifying the cell wall and enhancing the interaction of the pollen tube with the female style and transmitting tract tissues.  相似文献

Summary For this work we have used various microscopical methods (LM, SEM, and TEM) to study pollen tube growth and interaction with the transmitting tisse inStrelitzia reginae, which has an open style. By the use of SEM it was possible to trace the exact route of the pollen tubes in the ovary of this plant and demonstrate that they exclusively follow the outlines of the transmitting tissue. The average rate of pollen tube growth through the style was 1.8 mm h–1. The most significant effect of the pollination was a thickening of the distal wall of the subepithelial cells in the style. A secretion covers the stigma and the ovarian transmitting tissue and fills the stylar canal. This exudate contains lipids, polysaccharides, and proteins.  相似文献

 We examined the influence of pollen competitive environment on pollen performance in Mirabilis jalapa. We used the number of pollen grains and the number of pollen tubes per pistil as measures of pollen competition. Pollen germination, pollen tube penetration into the style, and pollen tube growth rates were used as measures of pollen performance. All three measures of pollen performance were affected by the competitive environment. Pollen germination was greatest at intermediate pollen load sizes. The percentage of germinated pollen grains that penetrated the stigma and grew into the style decreased with pollen load size. Pollen tube growth rate in the style was greater and more variable with larger numbers of pollen tubes in the style. Controlling for the degree of selection at the stigma indicated that pollen-pollen or pollen-style interactions were the likely causes of increased growth rates. We studied a population of the distylousPalicourea padifolia (Rubiaceae) in a cloud forest remnant near Xalapa City, Veracruz, México to explore possible asymmetries between floral morphs in the attractiveness to pollinators, seed dispersers, nectar robbers, floral parasites, and herbivores. We first assessed heterostyly and reciprocal herkogamy by measuring floral attributes such as corolla length (buds and open flowers), style and anther heights, stigma and stamen lengths and the distance between the anther tip to the stigma lobe. We then estimated floral and fruit attributes such as flower size, anther height, number and size of pollen grains, fruit size, seed size, nectar production, and flower and fruit standing crops to assess differences between floral morphs in attracting and effectively using mutualistic pollinators and seed dispersers. Also, floral parasitism and nectar robbing were assessed in this study as a measure of flower attractiveness to antagonists. The system seems to conform well to classical heterostyly (e.g. reciprocal stamen/style lengths, pollen and anther dimorphism, intramorph incompatibility) yet, there were several tantalizing differences observed between pin and thrum morphs. Thrum flowers have longer corollas and larger but fewer pollen grains than pin flowers. Both morphs produced the same total number of inflorescences, developed the same number of buds, and opened the same number of flowers per inflorescence during the flowering season. Nectar production and sugar concentration were similar between floral morphs but the reward was not offered symmetrically to floral visitors throughout the day. Nectar concentration was higher in pin flowers in the afternoon. The numbers of developing, fully developed, and ripe fruits were the same between floral morphs, however, fruits and seeds were larger than those of thrums. The incidence of fly larvae was higher among thrum flowers and damage by nectar robbing was the same between floral morphs. Fruit abortion patterns of flowers manually pollinated suggest intra-morph sterility (self and intramorph incompatibility). There were no differences between morphs in fruit and seed set per flower following legitimate pollination although thrums were more leaky than the pins (intramorph compatibility).  相似文献

Removal of substances loosely associated with the wall of Lilium longiflorum pollen did not affect in vitro germination and tube growth, tube growth within compatible styles, or production of viable seeds. Nor was growth of pollen tubes within incompatible styles enhanced by prior removal of the loosely bound materials. Hence, these materials appear not to be recognition factors in the gametophytic self-incompatibility system of Lilium and to have no role in pollen germination. Heat treatment of selected portions of lily pistils prior to pollination indicated that the incompatibility factor(s) was located in the lower half of the style, and that the stigma plays no role in incompatibility.  相似文献

异型花柱是一种受遗传因素控制的花型多态性现象,其适应意义在于提高不同花型间传粉精确性,从而促进异交。为检验二型花柱植物滇丁香(Luculia pinceana)是如何促进异交并确定有效传粉者的类型,实验统计自然种群中长、短柱型的植株数量,测量花部特征,统计自然种群植株柱头上的型间花粉落置的数量和比例,比较型内、型间花粉的萌发率以及花粉管长度,比较不同人工授粉处理结实情况,观察传粉昆虫类型并测量虫体特征。结果表明(1)滇丁香自然居群中长、短柱型植株的数量没有显著性差异。长柱型的筒深、筒直径、开口直径、雄蕊长和柱头长均显著小于短柱型,而长柱型的叶片长和宽、雌蕊长、花药体积、花粉粒体积、柱头厚均显著大于短柱型。(2)自然种群植株柱头上型间花粉所占比例显著低于型内花粉的比例。(3)型间授粉的花粉萌发率以及花粉管长度均大于型内授粉和混合授粉。(4)型间授粉的结实率显著高于型内授粉的结实率,型内授粉具有一定的结实。(5)夜行性长喙条背天蛾(Cechenena lineosa)是滇丁香的有效传粉者,其吻长和滇丁香的花筒深相适应。本研究表明二型花柱植物滇丁香主要是通过与其花部特征相匹配的长喙天蛾实现有效的传粉,通过促进型间花粉的萌发和花粉管的伸长而有利于型间授粉结实,提高传粉精确性。  相似文献

General patterns of floral morphology and incompatibility mechanisms have been described for many distylous plants. The absence of these patterns in typically distylous groups, as observed especially in tropical environments, is interpreted as atypical distyly, or as a new reproductive strategy derived from it. Data are presented here on the morphological and compatibility relations between floral morphs of four Psychotria dimorphic species in the Atlantic rain forest in SE Brazil: Psychotria jasminoides, P. birotula, P. mapourioides, and P. pubigera. When significant differences were found, floral parts were larger in thrum flowers. Results of controlled crosses showed that most incompatible pollen tubes were arrested in the stigma, and only in a low proportion in the upper parts of the style. We conclude that, at the study site, the majority and most important morphological and mating features of typical distyly seem to be conserved in P. jasminoides and P. mapourioides, which presented reciprocal herkogamy, self and intramorph incompatibility, and a balanced morph ratio in the population. Typical distyly in P. birotula is supported by floral morphology, pollen tube data and morph ratio and, in P. pubigera, only by floral morphology and pollen tube data.  相似文献

Hedyotis caerulea, a spring flowering herb widely distributed in eastern North America, has distylous flowers that differ by a number of morphological and physiological traits. The presence of a strong incompatibility system is indicated by the fact that intramorph crosses or self-pollinations produced little or no seed and intermorph crosses produced copious seed. An unusual homostyle was located that had most floral characters intermediate between pin and thrum flowers, although its pollen size was that of pin. The homostyle was only moderately self-compatible, and its pollen did not behave as pin pollen on thrum stigmas. It appears that, despite their intermediate position and morphology, the homostyle stigmas were fundamentally pin. Although the exceptional rarity of the homostyle suggests that it is an inadaptive failure, workers should watch for its further occurrence since additional study of it may provide insights into the genetic and physiological basis of heterostyly in theRubiaceae.  相似文献

I hand sectioned styles of Nicotiana glauca at intervals along their length and counted the number of pollen tubes in each section using fluorescence microscopy. Evidence of density dependent growth was found for three stages of pollen growth. Pollen germination on the stigma increased with increasing pollen population size. Pollen tube penetration in the stigma was unaffected by increasing density from low to moderate levels but was reduced at high densities. Pollen tube penetration in the style was enhanced by increasing density. This enhanced growth in the style was apparently confounded by interference among pollen tubes growing at high densities. In particular, the area of tissue able to support pollen tube growth decreases from the stigma into the lower style, which could cause overcrowding of pollen tubes growing at high densities. Enhanced pollen tube penetration with increasing density combined with interference among pollen tubes growing at high densities resulted in greater mean pollen tube lengths for populations with moderate densities. The shift from density independent growth in the stigma to positively density dependent growth in the style may represent a shift from autotrophic to heterotrophic growth stages of pollen.  相似文献
