A recognized invasive weed, Canada thistle ( Cirsium arvense L. Scop.) was grown at ambient and pre-ambient concentrations of atmospheric carbon dioxide [CO2] (373 and 287 μmol mol−1, respectively) at three levels of supplemental nitrogen (N) (3, 6 and 14.5 m M ) from seeding until flowering [77 days after sowing (DAS)]. The primary objective of the study was to determine if N supply limited the potential photosynthetic and growth response of this species to the increase in atmospheric [CO2] which occurred during the 20th century (i.e. approximately 290 to 370 μmol mol−1 CO2). Leaf photosynthesis increased both as a function of growth [CO2] and N supply during the first 46 DAS. Although by 46 DAS photosynthetic acclimation was observed relative to a common measurement CO2 concentration, there was no interaction with N supply. Both [CO2] and N increased biomass, relative growth rates and leaf area whereas root : shoot ratio was increased by CO2 and decreased by increasing N; however, N supply did not effect the relative response to [CO2] for any measured vegetative parameter up to 77 DAS. Due to the relative stimulation of shoot biomass, total above-ground N increased at elevated [CO2] for all levels of supplemental N, but nitrogen use efficiency (NUE) did not differ as a function of [CO2]. Overall, these data suggest that any potential response to increased atmospheric [CO2] in recent decades for this noxious weedy species was probably not limited by nitrogen supply.  相似文献

The activity of glutamine synthetase (GS) in mustard ( Sinapis alba L.) and Scots pine ( Pinus sylvestris L.) seedlings was used as an index to evaluate the capacity to cope with excessive ammonium supply. In these 2 species GS activity was differently affected by the application of nitrogen compounds (NH4+ or NO3). Mustard seedlings older than 5 days showed a considerable increase in GS activity after NH4+ or NO3 application. This response was independent of the energy flux, but GS activity in general was positively affected by light. Endogenous NH4+ did not accumulate greatly after nitrogen supply. In contrast, seedlings of Scots pine accumulated NH4+ in cotyledons and roots and showed no stimulation of GS activity after the application of ammonium. In addition, root growth was drastically reduced. Thus, the pine seedlings seem to have insufficient capacity to assimilate exogenously supplied ammonium. NO3, however, did not lead to any harmful effects.  相似文献

The effect of 700 μmol CO2 mol−1, 200 nmol ozone mol−1 and a combination of the two on carbon allocation was examined in Pinus halepensis co-cultured with Betula pendula in symbiosis with the ectomycorrhizal fungus Paxillus involutus . The results show that under low nutrient and ozone levels, elevated CO2 has no effect on the growth of B. pendula or P. halepensis seedlings nor on net carbon partitioning between plant parts. Elevated CO2 did not enhance the growth of the fungus in symbiosis with the birch. On the other hand, ozone had a strong negative effect on the growth of the birch, which corresponded with the significantly reduced growth rates of the fungus. Exposure to elevated CO2 did not ameliorate the negative effects of ozone on birch; in contrast, it acted as an additional stress factor. Neither ozone nor CO2 had significant effects on biomass accumulation in the pine seedlings. Ozone stimulated the spread of mycorrhizal infection from the birch seedlings to neighbouring pines and had no statistically significant effects on phosphoenolpyruvate carboxylase (PEPC) or ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activity in the pine needles or on PEPC activity in pine roots.  相似文献

A two-level multifactor experimental approach was used to compare seed germination and seedling performance of two Mediterranean tree species: the early successional Aleppo pine (Pinus halepensis Mill.) and the late successional holm oak (Quercus ilex L.). In a first experiment germination rate was evaluated under the combined effects of shade, nitrogen availability, and pine or holm oak leaf litter. In a second experiment we tested for the effects of shade, nutrient availability, and litter type on seedling survival, growth and biomass allocation. Holm oak showed higher germination rates under shaded than under unshaded conditions, while Aleppo pine showed no differences between shaded and unshaded conditions. Nitrogen availability and litter type had no significant effect on germination of either species. Both species showed increased RGR, but also higher mortality rates, when grown in an enriched nutrient environment. While Aleppo pine showed no differences in RGR and mortality rate under different shading levels, RGR decreased and mortality increased for holm oak in full light. Increased radiation decreased LAR, SLA and height:diameter ratio, and increased RWR and R/S in both species, although Aleppo pine showed more pronounced changes. Unlike Aleppo pine, holm oak responded to increased nutrient availability by decreasing R/S and increasing LAR. From these results, no seed-seedling conflicts were found in either species, but a trade-off does seem to exist for holm oak between biomass allocation traits deployed in response to increased nutrient availability and radiation. Aleppo pine outperformed holm oak under most environmental conditions tested and showed a wider regeneration niche.  相似文献

This study compares the potential of natural charcoal from Scots pine ( Pinus sylvestris L.) and activated carbon to improve germination under the hypothesis that natural charcoal adsorbs phytotoxins produced by dwarf-shrubs, but due to it's chemical properties to a lesser extent than activated carbon. Activated carbon has been used in many bioassays as an adsorbate to clean aqueous solutions.
We used aqueous extracts from young leaves of Calluna vulgaris (L.) Hull and Vaccinium myrtillus (L.) as phytotoxin sources in two different concentrations (10 and 14 gr. of dried leaves in 100 ml distilled water). Germination of pine seeds was prevented by the higher concentration of both species, while the lower ones did not show significantly reduced germination. Both ericaceous species showed a very similar potential to prevent germination of Scots pine seeds.
Supplemented carbon (activated carbon, powdered or granulated pine charcoal) restored germination in strong extracts. Adding activated carbon resulted in germination of almost 100%. With pine charcoals added, lower germination percentages were observed. The charcoal powder was more effective (60% for C. vulgaris ; 28% for V. myrtillus ) than the charcoal granulate (30% and 16%, respectively) in restoring germination.
Chemical and surface analysis of the three carbon supplements revealed that activated carbon had by far the biggest active surface area (641 m2 g−1), and thus many more cavities to bind phytotoxins than natural charcoal (total surface area of 142 m2 g−1).
We conclude, that charcoal produced by forest fires can have a positive effect on seed germination, but to a much lesser extent than activated carbon. Previous studies, which used activated carbon as an equivalent for charcoal, overestimated the effect of charcoal on germination.  相似文献

Changes in the patterns of gene expression were examined during loblolly pine ( Pinus taeda L.) seed stratification, germination, and post-germinative growth. In both the megagametophyte and the embryo, DNA contents remained relatively constant at all stages examined. RNA contents, however, increased in both tissues following seed germination, particularly in the embryo where a 7-fold increase in the RNA content was observed 5 days after germination. Poly(A)+ RNA, extracted from megagametophytes and embryos, was translated in vitro in a rabbit reticulocyte lysate cell-free system. Analysis of [35S]-methionine-labelled translation products by two-dimensional electrophoresis/fluorography indicated that there were changes in the populations of mRNAs during all developmental stages examined. In both the megagametophyte and the embryo several distinct mRNA populations, including one constitutively present at all stages examined, were identified. One mRNA population, present in the mature seed, decreased during seed stratification. Another population, not present in the mature seed, rose during the period of stratification that coincided with an increase in seed germinability. A third population, which appeared during seed germination, increased steadily during post-germinative growth. Besides these similarities, specific differences between megagametophyte and embryo were noted. For example, one mRNA population, which was present in the megagametophyte of the mature seed and remained constant during the stratification period, disappeared immediately following seed germination. In the embryo, one set of messages was germination specific. In total, these results show that mRNA populations change in a temporal fashion that is consistent with the patterns of de novo protein synthesis known to occur in loblolly pine during the same developmental periods.  相似文献

Aims:  The major objective of this study was to determine the effects of low levels of Escherichia coli O157:H7 contamination on plant by monitoring the survival of the pathogen on the rhizosphere and leaf surfaces of lettuce during the growth process.
Methods and Results:  Real-time PCR and plate counts were used to quantify the survival of E. coli O157:H7 in the rhizosphere and leaf surfaces after planting. Real-time PCR assays were designed to amplify the stx 1, stx 2 and the eae genes of E. coli O157:H7. The detection limit for E. coli O157:H7 quantification by real-time PCR was 2·4 × 103 CFU g−1 of starting DNA in rhizosphere and phyllosphere samples and about 102 CFU g−1 by plate count. The time for pathogens to reach detection limits on the leaf surface by plate counts was 7 days after planting in comparison with 21 days in the rhizosphere. However, real-time PCR continued to detect stx 1, stx 2 and the eae genes throughout the experimental period.
Conclusion:  Escherichia coli O157:H7 survived throughout the growth period as was determined by real-time PCR and by subsequent enrichment and immunomagnetic separation of edible part of plants.
Significance and impact of the Study:  The potential presence of human pathogens in vegetables grown in soils contaminated with E. coli O157:H7 is a serious problem to our national food supply as the pathogen may survive on the leaf surface as they come in contact with contaminated soil during germination.  相似文献

Stomatal conductance ( g s) and photosynthetic rate ( A ) were measured in young beech ( Fagus sylvatica ), chestnut ( Castanea sativa ) and oak ( Quercus robur ) growing in ambient or CO2-enriched air. In oak, g s was consistently reduced in elevated CO2. However, in beech and chestnut, the stomata of trees growing in elevated CO2 failed to close normally in response to increased leaf-to-air vapour pressure deficit (LAVPD). Consequently, while g s was reduced in elevated CO2 on days with low LAVPD, on warm sunny days (with correspondingly high LAVPD) g s was unchanged or even slightly higher in elevated CO2. Furthermore, during drought, g s of beech and chestnut was unresponsive to [CO2], over a wide range of ambient LAVPD, whereas in oak g s was reduced by an average of 50% in elevated CO2. Stimulation of A by elevated CO2 in beech and chestnut was restricted to days with high irradiance, and was greatest in beech during drought. Hence, most of the additional carbon gain in elevated CO2 was made at the expense of water economy, at precisely those times (drought, high evaporative demand) when water conservation was most important. Such effects could have serious consequences for drought tolerance, growth and, ultimately, survival as atmospheric [CO2] increases.  相似文献

Aims:  To investigate the infection biology of Colletotrichum sublineolum (isolate CP2126) and defence responses in leaves of resistant (SC146), intermediately resistant (SC326) and susceptible (BTx623) sorghum genotypes.
Methods and Results:  Infection biology and defence responses were studied quantitatively by light microscopy, H2O2 accumulation by DAB staining and HRGP accumulation by immunological methods. Inhibition of conidial germination and appressorium formation may represent prepenetration defence responses on the leaf surface. Inducible defence responses in the resistant genotypes included decreases in formation of appressoria as well as accumulation of H2O2, HRGPs and phytoalexins. Concomitant with these inducible responses, fungal growth was stopped during or just after penetration in genotypes SC146 and SC326. High levels of H2O2 accumulating at late infection stages (5 days after inoculation) in the susceptible genotype BTx623 correlated with necrosis and tissue degeneration.
Conclusions:  The early accumulation of H2O2 and HRGPs indicates roles in defence whereas the late accumulation in genotype BTx623 correlated with successful pathogenesis.
Significance and Impact of the Study:  The fact that there is a significant correlation between induced accumulation of H2O2, papilla formation and cell wall cross-linking, as evidenced by HRGP accumulation, and cessation of pathogen growth in resistant genotypes may help exploit host resistance in sorghum.  相似文献

The growth rates of naturally sympatric juvenile pink Oncorhynchus gorbuscha and sockeye Oncorhynchus nerka salmon were compared in a common lacustrine environment in south‐west Alsaka, an unusual opportunity given the normal disparity in freshwater residence time of these two species. Fork length ( L F) frequency distributions of juvenile pink salmon caught in the lake during the summer in 1991 and 1999–2003 indicated a growth rate of 0·54 mm day−1, 54% greater than the estimated growth rate of juvenile sockeye salmon sampled from 1958 to 2003 (0·35 mm day−1). Examination of daily growth rings on otoliths indicated that pink salmon in Lake Aleknagik grew an average of 1·34 mm day−1 in 2003 but sockeye salmon grew only 0·63 mm day−1(average specific growth rates were 3·0 and 1·8% day−1, respectively). Pink salmon increased from c . 32 mm L F and 0·2 g at emergence to 78 mm L F and 3·0 g within 3–4 weeks. After experiencing these rapid growth rates, the pink salmon appeared to leave the lake by late July in most years. The diets of pink and sockeye salmon in the littoral zone of the lake were very similar; >80% of the stomach contents consisted of adult and pupal insects and the remainder was zooplankton. This high degree of diet overlap suggested that the observed differences in growth rate were not attributable to variation in prey composition.  相似文献

Cell cycle activity was studied in apical and axillary buds of Norway maple ( Acer platanoides L.), apple ( Malus ' M9 ') , pedunculate oak ( Quercus robur L.), Scots pine ( Pinus sylvestris L.) and rose ( Rosa corymbifera 'Laxa') during dormancy induction and release. Flow cytometric analyses revealed that in dormant buds, cells mainly were quiescent at the G0/G1 phase, while in non-dormant buds, a significantly higher frequency of G2 cells was found in all species. In western blots accumulation of 55 kDa beta -tubulin was found in active growing plant material, whereas in dormant buds the accumulation was much lower or below detection level. It was observed for all species that during dormancy induction the amount of beta -tubulin decreased, while during dormancy release a fast accumulation of beta -tubulin occurred. The dynamics of the beta -tubulin accumulation reflected the dormancy status of tree buds of the five species studied suggesting that the beta -tubulin level might be useful as a marker for the dormancy status in buds of temperate woody species.  相似文献

The biological control potential of Pochonia chlamydosporia against root-knot ( Meloidogyne spp.) and cyst-forming ( Heterodera and Globodera spp.) nematodes is widely appreciated. In spite of this, little has been undertaken to determine the compatibility of this fungus with modern fungicides. A series of experiments were undertaken to investigate the effect of azoxystrobin on P. chlamydosporia . Initial Petri dish experiments found a significant reduction in the growth of P. chlamydosporia . EC50 values were calculated at 7 and 14 mg L−1, respectively, for hyphal growth and chlamydospore germination. A microcosm experiment based on counts of the colony-forming units of the re-isolated fungus showed a reduction in EC50 values for azoxystrobin in soil after 12, 20 and 40 days incubation (1.25, 1.00 and 3.00 mg kg−1 soil, respectively). There was evidence the fungus recovered over time in response to azoxystrobin application. This was also demonstrated in a glasshouse experiment where EC50 values of 1.32 and 4.4 mg kg−1 soil were obtained for 35 and 49 days after planting (DAP), respectively.  相似文献

Yeast exo-β-1,3-glucanase (EXG1) was evaluated as an inhibitory agent of Colletotrichum lupini and Botrytis cinerea. Extracts obtained from yeast transformed with the exg1 gene, expressing high levels of EXG1 activity, or control untransformed yeast cultures that lacked EXG1 activity, were added to different starting concentrations of C. lupini fungal spore suspensions (2.5 × 103 to 80 × 103 spores per flask), and mycelial dry weight was measured after 5 days. Inhibition of C. lupini mycelial growth by EXG1 compared with control extracts ranged from 41 to 20% when added to starting fungal spore concentrations of 2.5 × 103 to 80 × 103, respectively. EXG1 activity in the extracts from the transformed yeast remained high over the 5-day incubation period. Addition of the EXG1 extract after C. lupini spore germination resulted in lower inhibition, indicating that the EXG1 targets the β-glucan in the cell walls of the fungal spores at an early stage of germination. Furthermore, the yeast EXG1 extracts were also shown to inhibit Botrytis cinerea spore germination and growth. Thus, the use of the yeast exg1 gene for protection of crops, such as lupin and pear in transgenic strategies against C. lupini and B. cinerea , respectively, could be considered.  相似文献

The food habits and habitat use of pine marten ( Martes martes L.) on the Balearic Island of Mincorca were studied from March-August 1990. In a 28.2 km2 area, a series of hiking trails/forest roads were used to collect pine marten faeces bi-monthly and study te species' differential use of habitats. A total of 28 different food items were identified in 723 faeces. Small mammals were the most important food overall, constituting 34% of the volume During March-April, small mammals were the principal food consumed (63% of volume), followed by birds (19%). In May-June, birds were the main food (40%), then small mammals. Plant material and insects were the most important foods in July-August, both made up 68% of the pine marten diet. The abundance of certain foods and the degree of difficulty in obtaining them appear to be important factors that govern pine marten food selection in Minorca. Habitat use was studied along 12 routes totalling 32.6 km. Pine marten showed preference in March-April for Coastal Shrubland habitat, while during May-June they were found most in the Upland Pine type. During July-August, the Upland Pine and Open Pine habitats were most frequented by pine marten. According to bimonthly food habits data, habitat use seems largely determined by the abundance and availability of their prey items in the available habitat types. Martens may concentrate hunting efforts in areas different from when they are not hunting. The Minorcan pine marten differs from continental populations in its wide use of shrublands, in addition to forests.  相似文献

Long-term ammonia exposure of turbot: effects on plasma parameters   总被引:2,自引:0,他引:2  
Turbot juveniles were exposed to four ammonia concentrations [0·17 (L), 0·34 (M), 0·73 (MH) and 0·88 (H) mg l−1 NH3-N] for different exposure durations (28 days minimum to 84 days). Their physiological status and growth performances were compared to a control group [0·004 (C) mg l−1 NH3-N]. No growth was observed in the H group, and by day 57, mass increase in the MH group was only 15% of that in group C. During the first month growth in the L group was similar to that in control group while it was lower (33%) in the M group; afterwards the L and M groups had a similar growth (half that of controls). Accumulation of total ammonia nitrogen (TA-N) in plasma was dependent on ambient ammonia concentrations. Plasma urea levels in ammonia-exposed fish were lower, similar or greater than in controls (depending on ammonia concentration or exposure duration). Osmolarity, Cl and Na+ plasma concentrations were stable in the L and M groups. The increases in Na+, Cl, K+ and total Ca concentrations observed by the end of the experiment in the H and MH groups suggest that fish failed to adapt. There was an initial rise in plasma cortisol in all ammonia-exposed groups followed by a return to basal level (1·7–4 ng ml−1) in the L and M groups. In group MH, plasma cortisol peaked at 42 ng ml−1 by day 14, and after a decline at c . 1 month (14 ng ml−1), it rose again.  相似文献

Abstract: Based on surveys of species of museum pest insects commonly found in Finland, Norway, Denmark and Sweden, six species were selected for a study of the effect of exposure to anoxic treatment on the larval stage. An oxygen level of 0.3% (the rest, nitrogen) was applied and lethal exposure times were determined. Anthrenus museorum L. was the most susceptible species (LT99 = 32.2 h), while Attagenus woodroffei Halst. & Green and Attagenus smirnovi Zhantiev (LT99 = 88.1 h) were the most tolerant species. The LT99 value of At. woodroffei was not calculated, as only 50% of the larvae were killed by the treatment. The results indicate that large intraspecific variation is present in these two Attagenus species. Larvae of Anthrenus verbasci (L.) (LT99 = 43.9 h), Reesa vespulae (Milliron) (LT99 = 53.6 h), and Trogoderma angustum (Solier) (LT99 = 57.2 h) showed intermediate tolerance.  相似文献

Abstract. 1. The acceptability of sucrose solutions to Formica lugubris Zett. in the field was tested by offering droplets of known concentrations to ants tending aphids.
2. A probit model fitted to the data allowed the computation of the Median Effective Concentration (ED50) and its confidence limits.
3. In spring and early summer ED50 was about 0.15 M sucrose, but the ED50 rose in 1 week at the end of June from 0.2 M to 0.9 M. The change was simultaneous on pine and birch trees. The ED50 fell gradually to about 0.4 M in early November.
4. The increase in ED50 is related to the production of the summer apterae in the aphids tended, Cinara pini (L.) on pines and Symydobius oblongus (von Heyden) on birch. This increases the 'quality' of food resource so that low sucrose concentrations can no longer compete for ant attendance.
5. The increase in ED50 may limit the attendance of wood-ants at the nectaries of bracken, Pteridium aquilinum (L.) Kuhn. and at colonies of some other aphid species which are attended only in early summer.  相似文献

The effects of medium, isolate, temperature, light and pH on the formation and germination of Septoria tritici secondary conidia were tested. Of the six media tested, the malt–yeast extract agar was the best and generated 1.82 × 109 conidia/plate. The ten isolates tested showed different ability of conidia production. Darkness significantly reduced conidial formation and enhanced the transition of intermediates. The conidial germination and germ tube growth was strongly inhibited at 30°C. The suitable pH for conidial budding in malt–yeast broth (MYB) was between 5 and 9. At pH 2, 10 and 11, almost no new conidia were formed. The number of conidia reached 1.27 × 108 conidia/ml after 7 days in MYB, significantly more than that in potato dextrose broth, wheat leaf extract and H2O.  相似文献

Aims:  To evaluate the impact of modified atmosphere packaging on in vitro growth of Aspergillus carbonarius and Aspergillus niger , and possible effects on ochratoxin A (OTA) biosynthesis.
Methods and Results:  Ochratoxigenic isolates belonging to the species A. carbonarius and A. niger were grown on a synthetic grapejuice medium (SNM) and packaged in combinations of controlled O2 (1% and 5%) and CO2 levels (0% and 15%), and in air as a control. Colony diameters were recorded every 3 days up to 21 days, and OTA was analysed after 7, 14 and 21 days. The greatest reductions in mycelial growth rate were observed at 1% O2 followed by 1% O2/15% CO2, whereas 5% O2 stimulated the growth of all isolates. OTA production by A. carbonarius and A. niger isolates was minimized at 1% O2/15% CO2 and 1% O2, respectively, after 7 days of incubation. Maximal OTA accumulation after 7 days was observed for all isolates in the control pack and at 5% O2.
Conclusions:  Of the atmospheres tested, only 1% O2 combined with 15% CO2 consistently reduced fungal growth and OTA synthesis by A. carbonarius and A. niger .
Significance and Impact of the Study:  Storage under modified atmospheres is unlikely to be suitable as the sole method for OTA minimization and grape preservation; other inhibitory factors are necessary.  相似文献
