The freetop of the fungiform papilla shows a sensorial area about 100 micron in diameter, surrounded by a ring of ciliated cells. Externally to the ciliated cells, i.e., in the lateral wall, numerous large goblet cells can be seen devoid of their mucous content. The sensorial area is composed by three types of cells: mucous, supporting, and neuroepithelial cells. Mucous cells form the most superficial layer, while the cell bodies of the other two are deep, and from them basal and apical processes arise. The above mentioned cells are connected by desmosomes preferentially located between the mucous and the supporting cells, rather than between the supporting and the neuroepithelial cells. The lateral wall of the papilla is made up of a multilayered epithelium that comprises two types of cells: the first type contains electron-dense granules and an abundant rough endoplasmic reticulum, the others are ciliated cells. In the connective axis of the papilla, numerous fenestrated capillaries with endothelial vesiculated cells and nerve fibers are found.  相似文献

In hydrophilic media of high refraction index the cilia of the epithelium of the tracheo-bronchial mucous membrane and the mucous film demonstrate birefringence positive to the longitudinal axis of the mucous membrane owing to the identical orientation the glycoprotein macromolecules and of the fatty acid chains of the ciliary membrane. By means of toluidine blue staining the mucous film and the ciliary zone can be studied selectively by polarization microscopy. The opposite optical character suggests that the dye molecules are perpendicularly connected to the long glycoprotein chains of the mucous, and there is an oriented connection perpendicular to the longitudinal axis of the cilium in its lipid membrane. In smears of bronchial secretion or mucous membrane scrapings after staining, with toluidine blue pH 7.0, the selective optical reaction of the cilia provides a possibility for the study of isolated desquamated ciliary cells. The reaction is given by the lipid membrane boundary of the cilia. In embedded, lipid-extracted sections, the cilia and the mucous film show a minimum of birefringence in the unstained state. After toluidine blue staining or aldehyde-bisulphite-toluidine blue (ABT) reaction the mucoid surface and the ciliary zone display an opposite optical reaction originating from the dehydrated mucoproteins adsorbed onto the surface of lipid-extracted cilia.  相似文献

This study has revealed that helical bacteria inhabiting the mucous membrane of the cecum of guinea pigs are localized in the parietal zone of the epithelium and can be detected as biological film consisting of many microcolonies. Helical bacteria are attached to the epithelium by insertion of one of the ends of an eukaryotic cell into the space between microvilli without damaging epithelial cells and their microvilli. Helical bacteria have been found to use the "anchor" type of attachment to the epithelium, which ensures the stability of their high population level in the biotope. These microorganisms appear on the mucous membrane of the epithelium, starting from day 15 of the life of guinea pigs. At the period of the transition of suckling guinea pigs to independent nourishment the population of helical bacteria is partially suppressed due to the appearance of bacillary and filamentous forms of bacteria, but later, after the adaptation of the animals to their diet, helical bacteria become normal resident microflora which forms biofilm covering large areas of the mucous membrane and the entrances of crypts of Lieberkühn.  相似文献

In 55 men and 60 women who had had not any pathology in their biliary-pancreato-duodenal system during life, microrelief of the mucous membrane in the major duodenal papilla has been studied by means of light and scanning electron microscopy. It is defined by variants of fusion of the common bile duct and the pancreatic duct in the papilla. Three main variants of the ducts fusion are defined and, respectively, three main varieties of the structure of the mucous membrane folds. When the ducts flow into the same orifice separately, the mucous membrane relief is even, parvigranular and there are no fold-valves. In the common major duodenal papilla, the architectonics of the mucous membrane resembles the valve that acts as a block-outlet mechanism.  相似文献

By means of the indirect immunoperoxidase method in deparaffinized slices a comparative investigation on distribution of the female milk lactoferrin (LF) in tissues of the mature person and in the fetus has been performed. LF is revealed in cells of the neutrophil line of the mature person and of the fetus and in the secretory epithelium of some organs of the mature person (in the mammary, submandibular and parotid salivary glands, in the bronchial mucous membrane glands in the fundal and pyloric glands of the stomach). In all the cases investigated LF is revealed in the cells producing serous secrete: in the cells of the serous terminal parts and in the serous semilunar mixed terminal parts of the salivary glands, in the serous cells of the bronchial glands and in the chief cells of the gastric mucous membrane glands. In the fetal secretory epithelium of the organs LF is not found. As LF is revealed in the secretory epithelium of the mature person and is not revealed in the corresponding epithelium of the fetus, it should be considered as a marker of the cells, that reach certain degree of differentiation.  相似文献

Summary A comparative study was undertaken on the innervation of mucous and granular glands in frog skin. Results obtained by the Falck-Hillarp fluorescence technique and cholinesterase staining indicated that both types of glands receive exclusively adrenergic innervation. Electron microscopy was used to investigate the innervation pattern at the ultrastructural level. The distribution of nerve terminals was found to differ in the two types of glands. In the mucous gland, terminals were found at a distance of about 0.5 m from the basement membrane but never within the gland parenchyma. In the granular gland, the terminals were located between smooth muscle cells and also in direct contact with the secretory epithelium but never outside the basement membrane.This work was carried out in part at King Gustaf V:s Forskningsinstitut, Stockholm, and was supported by a grant from Karolinska Institutet  相似文献

Summary The distribution of an antigenic material specific for the cervicovaginal epithelium (CVA) was studied in the vaginal epithelium of the adult mouse with immunofluorescence and immunoferritin techniques. The antigen localization has been correlated to the fine structure of the vaginal epithelium in various states of functional activity. The antigen distribution in adult ovariectomized mice and in ovariectomized mice treated with estradiol was compared with that in normal cycling mice. CVA was found to be associated with the exterior of the cell membrane of vaginal epithelium cells, being part of the glycocalyx.Two cell types, mucous or keratinized, are derived from the germinative cell layer of the vaginal epithelium, depending on the hormonal environment. Mucous cells with morphological features that characterize cells about to cornify have been demonstrated. Fluorescence as well as ferritin particles, indicating the presence of antigen-antibody complexes, were seen within the mucous droplets of the mucous cells. The CVA production is apparently connected with vaginal mucus formation. The CVA distribution in the adult vaginal epithelium is discussed in relation to the distribution demonstrated earlier in the cervicovaginal epithelium of neonatal mice.This investigation was supported by grants from the Norwegian Cancer Society (Landsforeningen mot Kreft) and the Norwegian Research Council.  相似文献

Scanning electron microscopy shows that lingual papillae occur all over the dorsal surface of the tongue of the freshwater turtle, Geoclemys reevesii. The surface of each papilla is composed of compactly distributed hemispherical bulges, each composed of a single cell. Microvilli are widely distributed over the surface of cells. Histological examination reveals that the connective tissue penetrates deep into the center of papillae and that the epithelium is stratified columnar. Under the transmission electron microscope, the cells of the basal and the deep intermediate layers of the epithelium appear rounded. A large nucleus lies in the central area of each cell. The cytoplasm contains mitochondria, endoplasmic reticulum and free ribosomes. The cell membrane form numerous processes. The shallow intermediate layer contains two types of cell. The cytoplasm of the first has numerous fine granules, in addition to mitochondria, ribosomes, and endoplasmic reticulum. The other type of cell contains highly electron-dense granules. The surface layer shows two cell types. One type consists of typical mucous cells. The other type of cell contains fine, electron-lucent granules. The latter cells lie on the free-surface side, covering the mucous cells, and have microvilli on their free surfaces.  相似文献

Endometrium obtained during menses from 46 healthy women in reproductive age was investigated morphologically and cytospectrophotometrically in order to solve the problem on the source of the cells reepithelizing the uterine mucous membrane after desquamation. It was stated that desquamation takes place not in the whole functional layer of endometrium, some mucous fragments, covered with persisting luminal epithelium, are always preserved. During endometrial regeneration the cells of the luminal and glandular epithelia and those of endometrial stroma are predominantly diploid. The amount of premitotic cells in population is so small that they cannot secure any intensive cellular proliferation. Mitogenesis in endometrium is stimulated only after a complete restoration of the epithelial layer. It is suggested that persisting luminal epithelium is the source of cells for reepithelization; they migrate towards endometrial "wounds" and repair defects in the uterine mucosa during the regeneration phase.  相似文献

A small number of epithelial cells which combine features of two cell types were observed in the descending colon and pyloric stomach of the mouse. In the descending colon, where the base of the crypts is mainly composed of poorly differentiated "vacuolated" cells, a few of these cells contain, besides the characteristic "vacuoles," mucous globules identical to those in mucous cells or, less frequently, dense granules such as are found in entero-endocrine cells. Because there is evidence that the poorly differentiated vacuolated cells give rise to the other cells of the epithelium, those which also contain mucous globules or dense granules are likely to be differentiating into mucous cells or entero-endocrine cells respectively. In the pyloric stomach, where the glands are mainly composed of mucous cells, some of which are poorly differentiated, a few of the latter exhibit, besides the mucous globules, entero-endocrine type granules or features of caveolated cells. It is likely that the poorly differentiated mucous cells give rise to the other gland cells; and, therefore, those mucous-containing cells which also display dense granules or caveolated cell features are taken to be differentiating into entero-endocrine or caveolated cells respectively. Most of the cells containing two kinds of secretory materials are believed to be stem cells which initially contain a few vacuoles (colon) or mucous globules (pylorus) but are differentiating into a cell containing a different type of secretion. Rare observations of two kinds of secretory materials in a mature cell suggest that the transitional period may be prolonged, perhaps indefinitely.  相似文献

Epithelium of the duodenal mucous membrane has been studied in frog, rat and man by means of electron microscopy. Together with typical endocrinocytes, incretory cells of "mixed" type are revealed in the epithelium. They are classified into two types: 1. exo-endocrinic containing in their cytoplasm simultaneously: a) mucous and EC-granules, b) mucous and S-granules, c) granules specific for Paneth's cells and S-granules, d) granules specific for Paneth's cells and L-granules; 2. endocrinocytes with incretory granules of various types: with D and I, with D and EC, with D and L granules. Various ultramicroscopic organization of the "mixed" exo-endocrinic cells is demonstrated. It can be considered as various stages of their differentiation. In cytoplasm of the "mixed" cells of the second type (endocrinic) a constant presence of D-granules is revealed, that is, evidently, characteristic, within the limits of the gastro-entero-pancreatic system, for the cells with two types of the endocrinic granules.  相似文献

The occurrence of foeveola palatina was studied in 264 patients. The distance of the foveola palatina from the papilla incisiva and palatinal raphe was measured. Light and electron microscopic examinations were carried out on specimens obtained from cadavers. It was found that ducts of functioning mucous glands open into the foveolar lumen lined by stratified epithelium.  相似文献

The papillae basilares of three species of turtles and four species of snakes were studied by SEM. The papillae of turtle are relatively large among reptiles and are characterized by a long, horizontal middle section resting on wide basilar membrane. Both terminal ends of the papilla extend onto the surrounding limbus in the form of a forked or "T" -shaped end or as a curved, "hook"- like processes. Details vary with the species. In the three species of turtles studied, there were between 1,100 and 1,400 hair cells on a papilla. The tectorial membrane covering the horizontal portion of the papilla is heavy in appearance and tightly attached to the kinocilial bulbs. The terminal ends of the papilla are covered by a thin gelatinous material. In addition, mat-like tectorial network covers the supporting cells and extends from the microvilli of the supporting cells to the overlying tectorial membrane. All hair cells are unidirectionally and abneurally oriented. The supporting cell surfaces form a large part of the papilla and, thus, hair cell density is low. The papillae of the two boid snake species studied are moderately long among snakes and contain a moderate number of hair cells (574 in Epicrates and 710-780 in Constrictor). Papillar form is elongate, avoid, or canoe-shaped. The tectorial membrane may be either highly fenestrated or moderately dense and covers all but a few of the terminal hair cells. A tectorial-like mat covers all but a few of the terminal hair cells. Most hair cells are unidirectionally and abneurally oriented. A few terminal cells in boids may show reverse orientation. Hair cell density is similar to that of turtles.  相似文献

The Chinese giant salamander belongs to an old lineage of salamanders and endangered species. Many studies of breeding and disease regarding this amphibian had been implemented. However, the studies on the ultrastructure of this amphibian are rare. In this work, we provide a histological and ultra-structural investigation on posterior esophagus of Chinese giant salamander. The sections of amphibian esophagus were stained by hematoxylin & eosin (H&E). Moreover, the esophageal epithelium was observed by transmission electron microscopy (TEM). The results showed that esophageal epithelium was a single layer epithelium, which consisted of mucous cells and columnar cells. The esophageal glands were present in submucosa. The columnar cells were ciliated. According to the diverging ultrastructure of mucous vesicles, three types of mucous cells could be identified in the esophageal mucosa: i) electron-lucent vesicles mucous cell (ELV-MC); ii) electron-dense vesicles mucous cell (EDV-MC); and iii) mixed vesicles mucous cell (MV-MC).Key words: Amphibian, esophagus, ultrastructure, mucous cells, heterogeneous vesicles  相似文献

The luminal epithelium of the oviduct (magnum) of laying quails is composed of ciliated cells and mucous cells. Ciliogenesis was observed in some of the mucous cells. Both centrioles of the diplosome migrate to the top of the cell, and one of them induces the formation of a rudimentary cilium. In some of the other cells, that are filled with mucous granules, the formation of basal bodies by an acentriolar pathway was observed. In these cells, numerous, dense fibrous masses are associated with the forming face of the Golgi apparatus. In the Golgi zone, generative complexes composed of a deuterosome and some forming procentrioles were found. Cilia develop from completed basal bodies. During ciliogenesis, the Golgi apparatus is disorganized, and generally the production of mucous granules is arrested. The nucleus is also modified: it becomes larger and the chromatin is dispersed. It is assumed that mucous cells are able to be transformed into ciliated cells in the oviduct of laying quails.  相似文献

The conjunctiva of the domestic ruminants is covered by several kinds of epithelia which intergrade smoothly. There is a stratified nonkeratinized squamous epithelium which covers the greater part of the conjunctival mucous membrane. There is a stratified cuboidal, stratified mixed and occasionally a stratified goblet cell epithelium, localized mainly in the area of the fornix conjunctivae. In the area of the "serrated crest" of the nictitating membrane as well as the transition between conjunctiva and cornea, but also in the other conjunctival regions, the propria mucosae and the epithelium contains a few or numerous migrating lymphoid cells.  相似文献

The development of the bronchial and alveolar epithelium was observed in rabbits from the 15th day post conception until the time of birth with the scanning electron microscope. In the pseudoglandular phase, primitive bronchi proliferate in the mesenchyme. The epithelial cells are not differentiated and have single cilia. After retraction of these single cilia cell differentiation begins. Flat cells densely populated with cytopodia can be recognized on the 22nd day, ciliated cells on the 23rd day post conception. Both are located in the bronchi near the hilus. In the canalicular phase of development, the differentiation of the mucoid cells and the Clara-cells begins. The interstitial connective tissue develops more and more capillaries. The alveolar phase begins around the 26th day p. c. The lung capillaries reach the alveolar epithelial cells and arrange themselves directly beneath the epithelial basement membrane. This "alveolarization" of the lung tissue starts in the centre of the lung lobules and proceeds to the periphery. After the 26th day post conception the alveolar epithelial cells retract their single cilium and at the same time become type I or type II pneumocytes. The undifferentiated entodermal stem cell of the alveolar epithelium is the pneumoblast.  相似文献

Superficial cells of the oral mucosal epithelium in the carp and the cytoskeleton of the epithelial cells are examined by scanning and transmission electron microscopy. Microridges are formed on the surface of the epithelium. Epithelial cells contain two types of vesicles: mucous secretory vesicles and coated vesicles. Most of the mucous vesicles are situated in the center of the cell near the Golgi apparatus. In freeze-fracture replicas, intramembranous particles are abundant in the membranes of the secretory vesicles but rare in the apical plasma membrane. Coated vesicles are situated in the apical and subapical cytoplasm. A great number of thick filaments, considered to be keratin filaments, run randomly throughout the cell to form a meshwork. Thick filaments, which are sparse in the central cytoplasm, are connected to the membranes of the secretory vesicles and other membranous organelles. A layer of closely packed thin filaments, considered to be actin filaments, is found just beneath the apical plasma membrane. Microtubules also occur in the apical cytoplasm and run almost parallel to the cell surface. Both kinds of vesicles are connected to the thin and thick filaments. Their functional significance in the regulation of membrane at the free surface is discussed.  相似文献

Trefoil factor 2 (TFF2) is mucin associated peptide that has a mucosal barrier function in addition to participating in repair and healing. We examined the localization of TFF2 and gastric mucins in gastric mucous cells, the surface mucous gel layer (SMGL) adherent to normal gastric mucosa, and in the mucoid cap covering gastric erosions. Carnoy’s solution, or formalin/picric acid-fixed paraffin embedded materials from resected stomachs and formalin-fixed paraffin embedded gastric biopsy materials were used. Sections were immunostained for the TFF2 and histochemically stained for gastric mucins. In addition, thick sectioned gastric mucosa fixed in Carnoy’s solution were stained with FITC-labeled GSA-II lectin specific for gland mucous cell mucin and examined for three-dimensional images of the SMGL using a confocal laser scanning microscope. The TFF2 and gland mucous cell mucin were found intermixed together in the gastric gland mucous cells, in the SMGL in laminated layers, and in the mucoid cap. A laminated arrangement of continuous sheets of gland mucous cell mucin in the SMGL was demonstrated in the three-dimensional images. Co-localization of the TFF2 with gland mucous cell mucin suggests a physical interaction between the TFF2 and gland mucous cell mucin. The TFF2 trapped in the adherent mucins may be responsible for mucosal defense, healing, and repair.  相似文献
