The relationship of GA to apical dominance in Coleus was examinedby substituting 1 % IAA, in lanolin, for the shoot apex of CCC-treated,control and GA-treated plants containing, theoretically, hyponormal,normal and hypernormal GA levels, respectively. The greatestinhibition of lateral bud growth was obtained in the treatmentcombining 1 % IAA and 100 ppm GA, suggesting that GA may beimportant in the apical dominance of Coleus. CCC inhibited main axis growth, reduced the level of endogenousGA and caused a marked release of lateral buds from apical dominance. The significant stimulation of lateral bud growth by CCC couldnot be ascribed to reduced endogenous GA since it was not reversedby exogenous GA, or by GA plus IAA, whereas 100 ppm GA overcamethe inhibition of main axis growth by CCC. It was also shownthat the CCC stimulation was not a result of compensatory growth,that is, enhanced lateral bud growth resulting from reducedapical bud growth. The CCC effect on lateral buds was interpretedas involving a system independent of auxin and GA or else apossible immobilization of auxin in addition to inhibition ofGA biosynthesis. The adventitious bud forming ability of Sitka spruce (Picea sitchensis (Bong.) Carr.) buds in vitro was found to be dependent upon the age of tree from which the explants were taken. Bud formation declined exponentially with increasing tree age when 1.0 and 10 μM 6-benzylaminopurine (BA) were used to induce adventitious buds. When less BA was used (0.1 μM) bud production was much lower with all ages of tree and no mathematical relationship between declining bud production and tree age was found. By a tree age of 6 years bud-forming ability had declined severely. Even the few buds that developed on older tree tissues failed to elongate into shoots, became necrotic and eventually died, indicating that adventitious bud induction in this species is not a rejuvenative process. Callusing of bud explants also declined with increasing tree age when 0.1 μM BA was used whilst very little callusing occurred at the higher cytokinin concentrations (1.0 and 10 μM BA). Tissue necrosis in vitro increased with tree age, whilst the ability of BA to retard necrosis declined with increasing tree age. Buds from epicormic shoots, formed on the lower trunks of 20-year-old trees when these were exposed to light, were not significantly better at forming adventitious buds in vitro than buds taken from the lower branches of the crown.  相似文献

In the tropical tree Tabebuia rosea, seedlings form an erect unbranched stem with rhythmic growth. Three distinct and predictable geometrical stages are then recognized in crown development. Sympodial branching begins with the arrest of the terminal bud of the trunk and symmetric outgrowth of a pair of subtending lateral buds. During an intermediate phase, branching becomes asymmetric at about Order 5. At each sympodial bifurcation there is differentiation between vigorous, relatively straight main branches (leaders) and less vigorous laterals forming regular pseudomonopodial branch complexes, which collectively constitute the cup-shaped crown. Finally, dormant lateral buds in the lowest bifurcation of the trunk are released and reiterate the original crown form. Ultimately an erect, apparently monopodial tree is formed by a set of superimposed cup-shaped crowns. Crown development of Tabebuia is unique because it involves predictable ontogenic changes in branching patterns. Crowns of open-grown Tabebuia consist of relatively few, wide branch tiers, crowns of forest grown trees are tall and narrow. Analysis of the adaptive geometry of wide vs. narrow crowns through computer simulation illustrates the precise cost of mechanical support for terminal leaf rosettes at successive developmental phases and suggests that tall, erect, narrow, and multi-tiered crowns are more efficient than wide open crowns.  相似文献

The purine analogs 8-azaadenine, 8-azaguanine, 4-aminopyrazolo [3,4-d] pyrimidine, 2-fluoroadenine and 6-methylpurine inhibited bud burst in non-dormant vegetative axillary bud explants obtained from apple (Malus domestica Borkh.) seedlings. The reversal of inhibition was attempted by incorporating increasing concentrations of benzyladenine (BA) in the culture medium. The inhibition caused by 8-azaadenine and 8-azaguanine was reduced especially when BA was added to the media at equimolar ratios along with the inhibitors. The inhibition due to 4-aminopyrazolo [3,4-d] pyridine was reduced only with equimolar BA, BA was ineffective or only minimally effective in reducing inhibition due to 2-fluoroadenine and 6-methypurine. Short pretreatment with 8-azaadenine and 8-azaguinine failed to cause any inhibition when explants were subsequently transferred to BA -containing medium. The effectiveness of 2-fluoroadenine and 6-methylpurine was maintained or only slightly reduced, indicating their long-term inhibitory action.  相似文献

Senescence of Grand Rapids leaf lettuce was greatly reduced at 3 storage temperatures by post-harvest treatment with N,N-dimethylaminosuccinamic acid (Alar) and 2-chloroethyltrimethylammonium chloride (CCC), but not with 6N-benzyladenine (BA). Conversely, Alar and CCC were inactive on broccoli while BA was markedly effective. The deterioration and discoloration of mushrooms was inhibited by Alar with no effect observed from BA or CCC.  相似文献

This paper deals with apical dominance using a dicotylar model obtained after decapitation of pea seedlings with two shoots — one dominant and the other inhibited. When the dominant shoot was decapitated the inhibited one is released from inhibition and after 24 to 72 h begins to grow. However, the levels of trans-zeatin and production of ethylene increase within 4 and 6 hours respectively after release from inhibition, and within an interval of 72 h the levels of both phytohormones begin gradually to decrease. This indicates that also in this model, the release from apical dominance is associated with an increase in the level of cytokinin zeatin and, thereafter, also with an increased production of ethylene. If indolyl-3-acetic acid (IAA) is applied on the decapitated main stem after decapitation of the dominant shoot, the growth of the initially inhibited one is very strongly retarded; if, however, IAA is applied on the decapitated dominant shoot, this inhibition is significantly weaker. This means that the inhibiting effect of IAA on the inhibited shoot originates to a greater degree from the main stem rather than from the dominant shoot. The effect of benzyladenine (BA) is transferred equally from the decapitated main stem and from the decapitated dominant shoot because the initially inhibited shoot begins to grow as well as also other shoots from serial cotyledonary buds.  相似文献

The variation for embryo production in anther ofSolanum phureja was examined as a function of maximum greenhouse temperature prior to bud harvest and innate responsiveness among anthers within a bud. Four clones ofS. phuyreja were grown in a greenhouse under a 16-h photoperiod. The temperature was monitored continuously. Buds (60 per day on 10 days) were collected and the anthers cultured in two groups of five flasks (30 anthers per flask). In the first group, each flask contained the 30 anthers from six buds; in the second group, each flask contained one anther from each of 30 buds. Significantly smaller coefficients of variation were observed for the second group, suggesting that variation for embryogenic capcity among buds was greater than that among anthers within a bud. Variation in embryo yield as a function of greenhouse temperature was examined by stepwise regression analysis. Embryogenic capacity of one clone was adversely affected by high temperatures (31–37°C) that occurred two and seven days before bud harvest. However, similarly high temperatures appeared to enhance the androgenic response of another clone. Conversion of anther-derived embryos over three subcultures to fresh regeneration medium was examined as a function of anther donor or clone, cold pretreatment of embryos, and morphological classification of embryos. Only clonal origin significantly affected conversion rate which ranged from 12.5% to 46.0%.   1. It is often suggested to release oaks (Quercus robur) from competition, to ensure their survival and boost their conservational value. However, few studies have explored how long-lasting this effect is and how it affects saproxylic beetles.
  2. Ten years after cutting, we investigated effects of different release cutting levels (high, medium, and no release) around 140-year-old retained oaks in a commercial forest with Norway spruce (Picea abies).
  3. We evaluated oak vitality using crown and dead wood measurements. Saproxylic beetles were caught in window traps, identified to species level, and grouped according to their association with oak and/or Norway spruce.
  4. Released oaks had more light, higher temperatures, greater crown growth, and less dead wood in the crown compared to the no-release control.
  5. After 10 years we still found a higher abundance of oak-associated beetles and higher overall species richness of saproxylic beetles in the released oaks. Beetle species composition significantly differed between released oaks and control.
  6. We suggest avoiding planting trees beneath retained oaks when regenerating conifer forests and to proceed with conservation management during subsequent thinning by removing regeneration under the oak crowns. We emphasize the benefits of monitoring the retained oaks to maintain tree vitality, habitat quality, and insolation.

With applied to the petioles of detached Begonia x cheimantha leaves before planting, Gibberellic acid (GA3) inhibited the formation of adventitious buds and roots ill an apparently irreversible manner. Bud formation was entirely suppressed by 10?6M and higher concentrations and a significant inhibition was still present at 10?9M the lowest concentration tested. Root formation was not affected by GA3 below 10?7M and was possible even at 10?4 M GA3. Petiole elongation was stimulated by GA3 with an optimum at 10?5M. GA3 also blocked the action of 6-benzyiamino-purine (BAP) and 1-naphthaleneacetic acid (NAA), compounds which are potent stimulators of bud and root formation, respectively. When applied simultaneously with GA3 they were, at their optimal concentrations, devoid of any effect in counteracting or reversing the gibberellin-induced inhibitions. Abscisic acid and the growth retardants CCC and Phosfon also were unable to restore bud and root formation. In leaves initially treated with water or 10?5M BAP, endogenous bud and root formation as well as BAP-induced bud formation were entirety suppressed when 10?5M GA3 was applied 8 days after the initial treatments. Even when delayed for 14 days GA3 treatment inhibited BAP-induced bud formation, while treatment after 21 days bad little effect on bud and root formation. Development of pre-existing, visible bud primordia was not inhibited by GA3. BAP and NAA competitively inhibited the action of GA3 in petiole extension growth. The results are discussed in relation to results obtained in other plant systems. It is suggested that GA3 acts by blocking of the organized cell divisions initiating the formation of bud and root primordia.  相似文献

Both axillary buds belonging to the cotyledons (cotyledonary buds) start to grow on decapitated pea seedlings, but one of them (the dominant shoot) prevails in growth over the other (the inhibited shoot). If the dominant' cotyledonary shoot is removed, the inhibited shoot is released from inhibition and starts to grow. This release from inhibition of the inhibited cotyledonary shoot is accompanied within two hours from the removal of the dominant cotyledonary shoot by a marked increase in the level of endogenous cytokinin-like substances and by a decrease in the level of endogenous IAA. By contrast, a significant increase in IAA level and a decreasing trend in the level of cytokinin-like substances occur in the originally inhibited cotyledonary shoot between hour 4 and hour 48 after the release from inhibition of the inhibited cotyledonary shoot. The level of gibberellin-like substances in the cotyledonary shoot released from inhibition steadily increases from the beginning of the release.  相似文献

The effects of various concentrations of several auxins and cytokinins separately and in combination on epiphyllous budding of Bryophyllum calycinum were investigated, using a marginal leaf strip method. The number of buds released per leaf in 10−6 m benzyladenine (BA) was 2.8 times greater than those in water, and 1.6 times greater in 10−5 m isopentyladenine (IPA). BA, IPA and kinetin were antagonistic to indoleacetic acid (IAA) or naphthaleneacetic acid (NAA) when used in combination treatments. The IAA oxidase co-factor p-coumaric acid (PCA) increased the number of epiphyllous buds to twice that of the water controls. This stimulatory effect of PCA on bud release suggests that an increase in IAA oxidase activity may be one of the significant changes that precedes the release of epiphyllous buds.  相似文献

Using our improved method for culturing 11-day mouse forelimb buds in vitro, we have investigated the effects of a local application of all-trans-retinoic acid (RA) on growth, cartilaginous differentiation and skeletal patterning in the mammalian limb bud. Carrier implants of catgut impregnated with DMSO or various doses of RA in DMSO were inserted at the apex of the buds in the proximo-distal axis just beneath the apical ectodermal ridge. After 6 days of culture, cartilaginous skeletons were stained and explants were processed for morphological analysis and quantitative study using computerized optical image analysis. Buds treated with low doses of RA exhibited stimulated growth and chondrogenesis. Moreover, hypertrophied and fused metacarpals were seen within explants treated with the lowest dose. High doses strongly inhibited growth and skeletal morphogenesis. An intermediate dose sustained cartilaginous differentiation at the same level as low doses, but concomitantly disturbed the skeletal pattern. These results are discussed considering reported RA effects on other experimental systems including avian limb bud as an in vivo model or cell cultures as an in vitro simplified model.  相似文献

Filaments of Fuchsia hybrida cv “Brilliant” double in length within 24 hr after bud opening. Filament growth characterized by fresh wt increase and cell elongation was significantly inhibited in vitro by l-aminocyclopropane-l-carboxylic acid (ACC) but was not promoted by any growth regulator tested. Ions of Co2+ blocked the inhibitive effects of ACC in vitro suggesting that ethylene produced from ACC is the growth inhibiting substance. Ethylene levels surrounding the filaments within the closed bud decreased during development, and premature opening of the sepals which released the ethylene into the atmosphere resulted in rapid filament growth. The ACC levels were found to be much higher in the anthers than the filaments. This suggests that ethylene produced from floral organs other than filaments regulates filament elongation in Fuchsia. This is the first report of filament growth which cannot be promoted by application of growth regulators but which is inhibited by ethylene.  相似文献

The effect of a range of commercially available C16 and C18 fatty acids, on the release of cutinase by Ascochyta pisi, was studied. When juniperic acid was used as sole carbon source, cutinase activity was released in the culture medium, and was markedly enhanced by post-treatment of A. pisi cultures with acetone used as enzyme extractor. Without acetone, less cutinase was naturally released in culture medium containing juniperic acid at 0.5 %, than at 0.0l % or 0.05 %. Upon post-treatment with acetone, the same level of cutinase was released with all three concentrations, thus suggesting that the enzyme was induced, but not completely released in the presence of 0.5 % juniperic acid. When ricinelaidic or ricinoleic acids were supplemented at 0.5 % to cutin in the culture medium, they strongly inhibited the release of cutinase, even with acetone post-treatment. Comparable inhibition by ricinoleic acid was also observed when juniperic acid was used as cutinase inducer, thus suggesting that not only the release, but also the production of cutinase were inhibited.  相似文献

Summary To clarify that the presence of Ri T-DNA genes are not prerequisite for the light-induced bud formation in horseradish (Armoracia rusticana) hairy roots, leaf and root segments of nontransformed horseradish plants were used as explants. Bud formation from nontransformed tissues was observed in hormone-free medium under 16 h daylight conditions, but not under continuous darkness. To investigate the effects of growth regulators on bud formation, leaf and root explants were treated with auxin (1-naphthaleneacetic acid; NAA) and / or cytokinin (6-benzyl-aminopurine; BA). The most effective treatment in the dark to stimulate bud formation was BA at 1 mg·1-1. Root applications of 0.1 M 6-benzyladenine (BA) and 10.0 M indole-3-butyric acid (IBA) enhanced or inhibited, respectively, root bud growth in hydroponically grown Canada thistle [Cirsium arvense (L.) Scop.]. Translocation of14C-glyphosate [N-(phosphonomethyl)glycine] into roots was positively correlated with this growth. Foliar applications of ethephon or chlorfluorenol also enhanced root bud growth, but glyphosate translocation was only weakly correlated with such growth in soil-grown Canada thistle. At glyphosate rates above 0.56 kg/ha, root bud growth was not stimulated by plant growth regulators (PGRs) and basipetal translocation was not enhanced. Paradoxically, ethephon and chlorflurenol restrained root bud growth in the field since thistle control steadily improved during the 3 years following treatment.  相似文献

Bud dormancy in woody plants is released by long-term exposure to non-freezing chilling temperatures, whereas freezing temperatures have been considered to have little or no effect. However, the present results demonstrate that short-term exposure to freezing can release bud dormancy in Betula pubescens (Ehrh.) and B. pendula (Roth). Short-term freezing during the dormancy induction phase improved the release of bud dormancy only if an adequate level of dormancy had been reached. In fully dormant or chilled plants both the percentage and the speed of bud-burst increased, the more so the lower the temperature. Our results rule out the possibility that endogenous abscisic acid could be directly involved in the physiological control of bud dormancy release. The fast, easily applicable method presented here for bud dormancy release could further investigations into the biochemical and biophysical background to the process. The mechanisms of bud dormancy release and its relationship to cold acclimation are discussed in the light of these results, as also are the implications of the findings for modelling of bud dormancy.  相似文献

Because light conditions in the forest understory are highly heterogeneous, photosynthetic acclimation to spatially variable irradiance within a crown is important for crown‐level carbon assimilation. The effect of variation in irradiance within the crown on leaf nitrogen content and photosynthetic rate was examined for pinnate compound leaves in saplings of Cedrela sinensis, a pioneer deciduous tree. Five shading treatments, in which 0, 25, 50, 75 and 100% of leaves were shaded, were established by artificial heavy shading using shade screen umbrellas with 25% transmittance. Although the nitrogen content of leaves was constant regardless of shading treatment, ribulose 1·5‐bisphosphate carboxylase/oxygenase (Rubisco) content and light‐saturated photosynthetic capacity were lower in shade leaves within partially shaded crowns than within fully shaded crowns. Shade leaves within partially shaded crowns contained higher amount of amino acids. Most shade leaves died in partially shaded crowns, whereas more than half of shade leaves survived in totally shaded crowns. Assumptions on photosynthetic acclimation to local light conditions cannot explain why shade leaves have different photosynthetic capacities and survival rates in between partially and totally shaded crowns. Irradiance heterogeneity within the crown causes a distinct variation in photosynthetic activity between sun and shaded leaves within the crown.  相似文献

Callus cultures of Haplopappus gracilis, Nicotiana tabacum and Allium cepa var. proliferum were in varying degrees inhibited by blue to near-UV light obtained from fluorescent tubes. The inhibition was considerably reduced for Haplopappus cultures by 6-(3-methyl-2-buten-1-ylamino)-purine (2 iP) and (2-chloro-ethyl)-trimethylammonium chloride (CCC) in combination. Even separately these compounds stimulated growth in blue but not in white light. A high concentration of 2 iP reduced the inhibiting effects of near-UV on tobacco tissue cultures, and a synergism was observed between 2 iP and CCC in respect to shoot formation in blue light. Allium callus was not significantly affected by CCC. It was also observed that the concentration of indol-3-acetic acid (IAA) was more important for the growth of tobacco cultures in blue than in white light. It is believed that the light-inhibition of growth is partly due to a photoinactivation of IAA and that 2 iP and CCC might be active through processes controlling the levels of IAA and other growth hormones.  相似文献
