The photochemical apparatus organization in the thylakoid membrane of Macrocystis pyrifera, the giant kelp, was investigated. Chloroplasts were isolated from surface and minus 20 meter blades. Photosynthetic electron-transport complex quantitation revealed ratios of photosystem (PS) II/cytochrome b6-f/PSI = 1.8:3.3:1.0 in surface and 2.2:2.3:1.0 in minus 20 meter blades. The apparent photosynthetic unit size of chloroplasts from minus 20 meter blades (chlorophyll/P700 = 1485:1) was about 45% larger than that of surface blades (chlorophyll/P700 = 1025:1). The larger photosynthetic unit size of minus 20 meter blades is attributed to the substantially lower intensity of sunlight reaching the minus 20 meter habitat. In different chloroplast preparations, the effective absorption cross section of PSI and PSII to 670 nanometer light (chlorophyll a) and 481 nanometer light (chlorophyll c and fucoxanthin) was investigated. The results showed larger functional antenna size for PSII (about 90%) and for PSI (about 50%) in minus 20 meter than in surface blades. Moreover, the efficiency of utilization of 481 nanometer light by Macrocystis chloroplasts was equal to that of 670 nanometer light. It is concluded that the chlorophyll c-fucoxanthin complex in brown algae enables the highly efficient utilization of blue-green wavelengths of the nearshore marine environment and contributes to the dominance of M. pyrifera in this habitat.  相似文献

Five γ-aminobutyric acid (GABA)-ρ subunits were cloned from a white perch retinal cDNA library and expressed in Xenopus oocytes. The deduced amino acid sequences indicated that all are highly homologous to the GABA-ρ subunits cloned from mammalian retinas; two clones (perch-ρ1A and perch-ρ1B) were in the ρ1 family, two (perch-ρ2A and perch-ρ2B) were in the ρ2 family, and one clone has been tentatively identified as a perch-ρ3 subunit. When expressed in Xenopus oocytes, all but one of the subunits (ρ3) formed functional homooligomeric receptors. However, the receptors expressed by each of the GABA-ρ subunits displayed unique response properties that distinguished one from the other. For example, receptors formed by perch-ρ1B subunits were more sensitive to GABA than the receptors formed by other GABA-ρ subunits, the dose–response curves for the various receptors revealed different Hill coefficients, and there were differences in the kinetics of the GABA-induced currents. In addition, the GABA-mediated current–voltage curve for ρ2 receptors was approximately linear, whereas the responses from ρ1 receptors showed outward rectification. A further division in the properties of the GABA-ρ subunits was revealed in their responses to imidazole-4-acetic acid (I4AA); the drug behaved as an antagonist on A-type ρ receptors and a partial agonist on the B-type ρ receptors. These results suggest that there is a large diversity of GABAc receptors in the vertebrate retina, probably formed by homooligomeric and heterooligomeric combinations of GABA ρ subunits, that exhibit different functional properties. © 1998 John Wiley & Sons, Inc. The saturating photon flux density (400 to 700 nanometers) for induction of flowering of the long day plant Anagallis arvensis L. was 1,900 micromoles per square meter per second (6,000 foot-candles) when an 8-hour daylength was extended to 24 hours by a single period of supplementary irradiation. The saturating photon flux density for photosynthetic CO2 uptake during the same single supplementary light period was lower, at about 1,000 to 650 micromoles per square meter per second (3,000 to 2,000 foot-candles).

The per cent flowering and mean number of floral buds per plant were significantly reduced when the light extension treatment was given in CO2-free air, and glucose (10 kilograms per cubic meter in water) relieved this effect. Glucose solution also significantly increased flowering of plants given supplementary light treatment in atmospheric air under a photon flux density of 80 micromoles per square meter per second. Increasing the CO2 concentration to 1.27 grams per cubic meter of CO2 in air during the supplementary light period did not increase flowering.

It is concluded that high photon flux densities promote flowering of Anagallis through both increased photosynthesis and the photomorphogenic action of high irradiance.

Abstract

The receptor protein tyrosine phosphatase T PTPρ is the most frequently mutated tyrosine phosphatase in human cancer. PTPρ mediates homophilic cell-cell aggregation. In its extracellular region, PTPρ has cell adhesion molecule–like motifs, including a MAM domain, an immunoglobulin domain, and four fibronectin type III (FNIII) repeats. Tumor-derived mutations have been identified in all of these extracellular domains. Previously, the authors determined that tumor-derived mutations in the MAM and immunoglobulin domains of PTPρ reduce homophilic cell-cell aggregation. In this paper, the authors describe experiments in which the contribution of the FNIII repeats to PTPρ-mediated cell-cell adhesion was evaluated. The results demonstrate that deletion of the FNIII repeats of PTPρ result in defective cell-cell aggregation. Furthermore, all of the tumor-derived mutations in the FNIII repeats of PTPρ also disrupt cell-cell aggregation. These results further support the hypothesis that mutational inactivation of PTPρ may lead to cancer progression by disrupting cell-cell adhesion.  相似文献

—The kinetics of the uptake of various compounds into the crude mitochondrial fraction (P2-fraction) of the pigeon optic tectum were studied. Aspartate, GABA, glutamate, glycine, proline and choline were taken up by high and low affinity systems. Only low affinity uptake was found for leucine and dopamine. The uptake for taurine was not saturable. The uptakes for all of the above compounds were temperature and sodium dependent. The high affinity system was more effected by sodium withdrawal than the low affinity system. Continuous sucrose gradients from 0.4 to 1.5 m were run with incubated P2 fractions. Particle-bound radioactivity sedimented in a density range of 1.0 to 1.2 m -sucrose. Additional experiments with glutamate showed that its uptake is competitively inhibited by aspartate. The addition of a 1000-fold excess of glutamate to a P2 fraction incubated at a concentration of 10–6m -glutamate led to a massive decrease of particle-bound radioactivity, suggesting a coupled action of uptake and efflux.  相似文献

The successful performance of a rapid profiling light absorption instrument in making vertically detailed measurements of gelbstoff (dissolved yellow material) in a mesotrophic water supply reservoir is demonstrated. In situ measurements of gelbstoff spectra (g λ; m? 1) for the photosynthetically active radiation wavelength (λ) interval (400–700 nm), and values of g 440, matched closely values obtained through traditional laboratory spectrophotometric analyses. The advantages of the high vertical resolution capability of the in situ absorption instrument (~10 measurements per meter of depth) was demonstrated through the depiction of the vertical details (subsurface peak about 2.25 m thick) of the enrichment in gelbstoff that occurred following a major runoff event, associated with the entry of inflow(s) as a plunging interflow. Values of g 440 increased at the depth of the subsurface peak by nearly 100% as a result of this event. The detailed vertical resolution of gelbstoff is valuable to managers of water supply lakes and reservoirs to support selection of intake depths to avoid delivery of water with high levels of color, and to researchers investigating the origins, fate and transport of this material.  相似文献

The ammonia-oxidizing chemoautotrophic bacterium Nitrosomonas europaea possesses prominant succinate-reducing activity of succinyl-Coenzyme A synthetase (SCS, EC 6.2.1.5). SCS was purified as an electrophoretically homogeneous protein from Nitrosomonas europaea strain ATCC 25978 about 275-fold, with a 3.9% activity yield. The molecular mass of the native enzyme was estimated to be about 130 kDa by gel filtration, whereas SDS-PAGE gave two protein bands with Mr values of 29 (α) and 36 kDa (β). The isoelectric point of the enzyme was 5.3. The apparent Km values of the enzyme for ATP, succinate and CoA were 0.4 mM, 5 mM and 0.1 mM, respectively. The pH and temperature optima of the SCS were about 5.0 and 55°C, respectively. The SCS was stable in the pH range of 8.0–10.0 and up to 70°C. The enzyme was thermostable; 50% of the enzyme activity was retained at 90–100°C for 10 min. The SCS was activated by Mg2+ at 1.0–100 mM, but inhibited by Cu2+ (0.1 mM) and SDS (1.0 mM). The enzyme utilized ATP as the preferred substrate.  相似文献

Filling of the gastric lumen of rats with 1.0 M NaCl solution (5 ml) for 10 min under urethane anesthesia caused an increase in the gastric fluid concentrations of prostaglandin (PG) E2, 13, 14-dihydro-15-keto-PGE2 and 6-keto-PGF as determined by radioimmunoassay. PGE2 was the major PG generated. The levels of PGE2 in the gastric fluid were increased dose-dependently after filling the lumen with 0.3, 0.5, 0.7 or 1.0 M NaCl solutions. The pH of the gastric fluid increased similarly after 0.5 to 1.0 M NaCl solutions. Indomethacin (10 mg/kg, i.p.) suppressed the PGE2 increase caused by 1.0 M NaCl solution, but did not prevent the increase of the pH of the gastric fluid induced by intragastric 1.0 M NaCl. Infusion of tetragastrin (62.5 μg/kg/hr, i.v., for 10 min) caused a marked increase of acid secretion without modifying intragastic concentration of PGE2. The acid secretion due to tetragastrin was completely inhibited after intragastric administration of 1.0 M NaCl solution, while indomethacin restored the tetragastrin-induced acid secretion, with prevention of a rise of intragastric PGE2 levels. These observations suggest that 1.0 M NaCl solutions suppress basal intragastric acid through a mechanism which is independent of prostaglandins. In contrast, the suppression of tetragastrin-induced acid secretion by intragastric 1.0 M NaCl solution appears to be mediated through a release of prostaglandins  相似文献

The influences of photosynthetically active radiation (PAR) and water status on nocturnal Crassulacean acid metabolism (CAM) were quantitatively examined for a widely cultivated cactus, Opuntia ficus-indica (L.) Miller. When the total daily PAR was maintained at 10 moles photons per square meter per day but the instantaneous PAR level varied, the rate of nocturnal H+ accumulation (tissue acidification) became 90% saturated near 700 micromoles per square meter per second, a PAR level typical for similar light saturation of C3 photosynthesis. The total nocturnal H+ accumulation and CO2 uptake reached 90% of maximum for a total daily PAR of about 22 moles per square meter per day. Light compensation occurred near 0 moles per square meter per day for nocturnal H+ accumulation and 4 moles per square meter per day for CO2 uptake. Above a total daily PAR of 36 moles per square meter per day or for an instantaneous PAR of 1150 micromoles per square meter per second for more than 6 hours, the nocturnal H+ accumulation actually decreased. This inhibition, which occurred at PAR levels just above those occurring in the field, was accompanied by a substantial decrease in chlorophyll content over a 1-week period.

A minimum ratio of H+ accumulated to CO2 taken up of 2.5 averaged over the night occurred for a total daily PAR of 31 moles per square meter per day under wet conditions. About 2 to 6 hours into the night under such conditions, a minimum H+-to-CO2 ratio of 2.0 was observed. Under progressively drier conditions, both nocturnal H+ accumulation and CO2 uptake decreased, but the H+-to-CO2 ratio increased. A ratio of two H+ per CO2 is consistent with the H+ production accompanying the conversion of starch to malic acid, and it apparently occurs for O. ficus-indica when CAM CO2 uptake is strongly favored over respiratory activity.

Effects of retinoic acid (RA) on prostaglandin E2 (PGE2) and cyclic AMP (cAMP) concentrations were investigated in high density, micromass cultures of mesenchymal cells derived from chick limb buds. Exposure of cells during the initial 24 h of culture to RA concentrations between 0.05–1.0 μg/ml inhibited chondrogenesis in a dose-dependent manner with 1.0 μg/ml totally inhibiting cartilage formation. Concentrations of PGE2 and cAMP increased during the prechondrogenic period in control cells in a closely related way and remained elevated throughout the six-day period examined. Addition of RA (0.05 and 0.5 μg/ml) did not significantly alter cAMP concentrations at any time point, but significantly elevated PGE2 levels relative to control cells in six-day cultures in a concentration-dependent manner. Addition of dibutyryl cAMP enhanced chondrogenesis in control cells between days 3 and 4, but failed to alter the inhibitory effect of RA on chondrogenesis. The results indicate that while PGE2 and cAMP are important signals in cartilage differentiation, the inhibitory effects of RA on this process are mediated through some other mechanism.  相似文献

O-Nitratedchitosan (d.s. 1.5–1.8 for NO2 was N-acylated with carboxylic anhydride to afford products (d.s. 1.4–1.8 for NO2 in 78–100% yield, and N-acylchitosan (d.s. 1.0 for N-acyl was nitrated with fuming nitric acid-acetic anhydride to afford products (d.s. 1.7–2.0 for NO2 in 90–100% yield. N-Benzoylchitosan was nitrated with fuming nitric acid-acetic anhydride to afford a product of d.s. 3.0 for NO2. All the products were soluble in methanesulphonic acid. The nitrated products containing N-fatty acyl groups (> C2) were soluble in N, N-dimethylformamide and dimethyl sulphoxide, but the nitrated products containing N-fatty acyl groups (> C6) were insoluble or swelled. O-Nitrated chitosan (d.s. 1.5–1.8 for NO2) had ignition points 150–1562C, but the nitrated products containing N-acyl groups had no ignition point, and decomposed at over 2402C.  相似文献

This article suggests a new sequential injection analysis chemiluminescence (SIA‐CL) strategy for monitoring the caffeine (CAF) content in soft and energy drinks using the catalytic activities of different nano‐metal oxides. The present study describes three different SIA‐CL systems (luminol–ferricyanide (III) coupled with Fe2O3 or ZnO nanoparticles (NPs), and luminol–H2O2 coupled with CuONPs. All experimental conditions were optimized and the linear concentration ranges of pure CAF were evaluated using the calibration graphs. The selectivity of the developed SIA‐CL systems was studied under the influence of various interfering species that may be present in soft or energy drinks such as sodium ions, sucrose, glucose, sodium benzoate, sodium citrate, riboflavin, niacin, citric, phosphoric and ascorbic acids. International Council for Harmonization (ICH) guidelines were obeyed for the validation of the suggested CL methods. The developed SIA‐CL systems displayed linear relationships over the concentration ranges 1.0–350, 5.0–400 and 10.0–400 μg ml?1 with Fe2O3 NPs, ZnO NPs and CuO NPs, respectively. The recorded lower limits of detection and quantification were 0.7, 2.7 and 7.8 μg ml?1, and 1.0, 5.0 and 10.0 μg ml?1 for the previously mentioned SIA‐CL systems. The results revealed high selectivity for CAF determination and were in good agreement with those obtained by other reported methods.  相似文献

Phosphopentomutase (PPM) catalyzes the interconversion of α-d-(deoxy)-ribose 1-phosphate and α-d-(deoxy)-ribose 5-phosphate. We developed a coupled or uncoupled enzymatic assay with an enzyme nucleoside phosphorylase for determining PPM activities on d-ribose 5-phosphate at a broad temperature range from 30 to 90 °C. This assay not only is simple and highly sensitive but also does not require any costly special instrument. Via this technology, an open reading frame TM0167 from a thermophilic bacterium Thermotoga maritima putatively encoding PPM was cloned. The recombinant PPM was overexpressed in Escherichia coli Rosetta. This enzyme has the highest activity at 90 °C. MnCl2 (0.1 mM) and 50 μM α-d-glucose 1,6-bisphosphate are cofactors. The kinetic parameters of Km and kcat are 1.2 mM and 185 s−1 at 90 °C, respectively. The enzyme has a half-life time of up to 156 min at 90 °C. This enzyme is the most active and thermostable PPM reported to date.  相似文献

ObjectiveTo investigate nutritive microvascular function in young nonobese females with polycystic ovary syndrome (PCOS) and to correlate microvascular reactivity with sex steroids, inflammatory markers, and metabolic variables.MethodsFourteen nonobese females with PCOS (24.6 ± 2.7 years, body mass index [BMI] 23.7 ± 3.1 kg/ m2) and 13 age- and BMI-matched controls (22.8 ± 2.3 years, 22.5 ± 3.4kg/m2) underwent anthropometric, hormonal, and microvascular evaluations. The main outcome measures were capillary density, red blood cell velocity (RBCV) at resting and peak during postocclusive reactive hyperemia (RBCVmax), and time taken to reach RBCVmax (TRBCVmax).ResultsSubjects with PCOS had lower RBCV and higher TRBCVmax compared to controls, respectively (0.237 [0.220-0.324] vs. 0.362 [0.297-0.382] mm/s, F < .01) and (5 [5-6] vs. 4 [3-5] s, P < .05]. The free androgen index (FAI) and sex hormone-binding globulin (SHBG) level were different between groups. FAI correlated to RBCVmax (ρ = -0.49, P < .05) and to TRBCVmax (ρ = 0.41, P < .05). SHBG correlated with RBCVmax (ρ = 0.52, P < .01) while estradiol (E2) levels correlated with RBCV (ρ = 0.80, P < .001) and RBCVmax (ρ = 0.46, P < .05).ConclusionMicrovascular dysfunction characterized by reduced RBCVmaxand prolonged TRBCVmaxwas present in young, nonobese PCOS subjects. Microbial transglutaminase (MTG) has been widely used in the food and pharmaceuticals industries. In this study, MTG was purified using affinity precipitation with an affinity polymer (PMMDN-T), which was synthesized using a pH-responsive polymer (PMMDN) coupled with L-thyroxin as an affinity ligand. Interactions between MTG and PMMDN-T were investigated using turbidimetric titration, zeta potential measurements, and low-field nuclear magnetic resonance (LF-NMR). We found different behaviors, architectures, and phase states of pH-dependent interactions between MTG and PMMDN-T interactions. Binding energetics between MTG and PMMDN-T were determined by isothermal titration calorimetry (ITC). The isoelectric point (pI) of the affinity polymer was 4.65 and was recovered with 96.7% efficiency after recycling the polymer three times. The optimal adsorption condition was 0.02 mol/L phosphate buffer (pH 6.0) with 1.0 mol/L NaCl at 30.0°C and a ligand density of 50.0 μmol/g. The maximum elution recoveries of total MTG were 98.44% (protein) with 92.19% (activity) using 0.02 mol/L pH 10.0 Gly-NaOH as the eluent.  相似文献

Glutamine synthetase and asparagine synthetase systems with reactions involving lysis of ATP to ADP and Pi or AMP and PPi are usually assayed by discontinuous sampling and analysis or by coupled enzymic systems. Experimental results confirm theoretical predictions that such reactions may be continuously and directly monitored by pH stat devices. Sample volumes of 0.5–1.0 ml and buret volumes of 0.05–0.25 ml, with ATP levels near 1 mm can be used routinely. The number of enzyme reactions involving ATP to which this technique can be applied is quite large.  相似文献

IntroductionIn chronic hepatitis C virus (HCV) infection there is increased iron absorption leading to iron overload, a fact that may promote ferritin synthesis. Theoretically, increased ferritin should promote ongoing liver fibrosis but disparate results have been described.ObjectiveWe analyze the behavior of iron metabolism- related variables, comparing them with fibrosis and inflammatory activity in liver biopsy in HCV infected patients.Patients and MethodsWe analyzed among 90 HCV patients subjected to liver biopsy prior to antiviral treatment the relationships of serum levels of iron, ferritin, transferrin, transferrin saturation index (TSI) and total iron binding capacity (TIBC) with liver fibrosis and histological severity, assessed by Metavir-f, Metavir-a and Knodell indices, as well as with liver function, and also compared the aforementioned iron metabolism- related variables with 34 controls.ResultsPatients showed higher values of sideremia (T = 2.04; p = 0.044) and transferrin (T = 2.29; p = 0.004) compared with controls; but not ferritin, that was significantly higher among the 33 patients who also consumed alcohol (Z = 2.05; p = 0.041). Most patients showed a well preserved liver function (86 cases, Child A). Patients with Child B or C showed higher ferritin levels (Z = 2.68; p = 0.007) and TSI (Z = 2.41; p = 0.016), but lower transferrin and TIBC (Z = 3.25; p = 0.001) than Child A patients. Transferrin and TIBC were directly related to albumin (ρ = 0.24; p = 0.026), whereas bilirubin showed direct relationships with iron (ρ = 0.25; p = 0.016), TSI (ρ = 0.39; p < 0.001) and ferritin (ρ = 0.36; p < 0.001). Both ferritin (ρ = −0.22; p = 0.04) and TSI (ρ = −0.25; p = 0.016) were related to platelet count. No relationships were observed between iron variables and Knodell index, but serum iron, serum transferrin, and TSI were directly related to Metavir-f score (ρ = 0.28; p = 0.009, ρ = 0.22; p = 0.044, and ρ = 0.22; p = 0.044, in this order).ConclusionAlterations of iron related variables are relatively subtle in our series of 90 well compensated HCV patients. Serum ferritin was not related to liver fibrosis and increases only when alcoholism co-exists with HCV infection.  相似文献

The interaction of a mammalian cell culture broth with two commercially available adsorbents for the use in expanded bed adsorption (EBA) has been studied. A cation exchange resin (Streamline SP) and an affinity adsorbent (Streamline rProtein A) were compared with regard to adsorption of hybridoma cells during sample application as well as potential cell damage. The results showed that hybridoma cells interact significantly with an expanded bed of cation exchange adsorbents but not with the Protein A adsorbent. After application of 17–20 sedimented bed volumes a saturation of the Streamline SP resin with cells was noted. With both adsorbents no measurable cell damage was found and IgG1 was recovered in approximately 95% yield. The capacity for IgG1 adsorption at 3% breakthrough was 2.7 mg IgG1/ml Streamline rProtein A at a constant fluid velocity of 380 cm/h and 1.0 mg IgGl/ml Streamline SP at 215–240 cm/h fluid velocity.  相似文献

Genipin gel dosimeters are hydrogels infused with a radiation-sensitive material which yield dosimetric information in three dimensions (3D). The effect of inorganic salts and glucose on the visible absorption dose–response, melting points and mass density of genipin gel dosimeters has been experimentally evaluated using 6-MV LINAC photons. As a result, the addition of glucose with optimum concentration of 10% (w/w) was found to improve the thermal stability of the genipin gel and increase its melting point (Tm) by 6 °C accompanied by a slight decrease of dose–response. Furthermore, glucose helps to adjust the gel mass density to obtain the desired tissue-equivalent properties. A drop of Tm was observed when salts were used as additives. As the salt concentration increased, gel Tm decreased. The mass density and melting point of the genipin gel could be adjusted using different amounts of glucose that improved the genipin gel suitability for 3D dose measurements without introducing additional toxicity to the final gel.  相似文献
