A substance inducing teliospore production inPuccinia racondita f. sp.tritici was found in water and methanol extracts of wheat leaves with telia of the wheat leaf rust just before harvest time. Methanol (MeOH) and water extracts from uninfected wheat leaves also showed telia-inducing activity. However, the MeOH and water extracts from wheat leaves covered with telia showed much stronger activity than those from uninfected wheat leaves. We obtained a fraction (0.2 mg) showing activity at 2 ng/ml by purification of the water extract.  相似文献

The diurnal changes in leaf net photosynthetic rate (P N) and sugar and nitrogen contents in wheat [Triticum aestivum (L.) cv. HD 2285] and mungbean [Vigna radiata (L.) Wilczek cv. PS 16] were analysed under ambient, AC [350±25 µmol mol–1] and elevated, EC [600±50 µmol mol–1] CO2 concentrations. In both mungbean and wheat P N of AC- and EC-grown plants compared at the same CO2 concentration showed that P N was higher under EC. However, increased P N in EC-plants declined in the afternoon and approached P N of AC-plants. Depression in P N, however, was less in mungbean compared with the large depression in wheat. Greater down regulation of P N in wheat was associated with the accumulation of large amount of sugars and low nitrogen content in wheat leaves. Mungbean leaves accumulated mostly starch under EC and the difference in N content in AC- and EC-plants was relatively less than in wheat.  相似文献

The fate of the herbicide diphenamid was determined in cell suspensions of soybean [Glycine max (L.) Merr. ‘Wilkin’] at different stages of cell growth: early log phase (3 to 7 d), log phase (7 to 14 d), and stationary phase (14 to 18 d). [Carbonyl-14C]-diphenamid was added to the suspensions as an acetone solution. Neither diphenamid (2 to 3 μM) nor acetone (0.5% v/v) was phytotoxic. The 14C-labeled products were identified tentatively by thin layer chromatographic comparison with reference compounds. The major metabolic products formed were N-hydroxymethyl-N-methyl-2,2-diphenylacetamide, N-methyl-2,2-diphenylacetamide, 2,2-diphenylacetamide, and two polar metabolites (0.9 to 25% of the applied 14C activity) that appeared to be glucose conjugates; one an acidic glucoside. All metabolites were found in both the cell extract and the culture medium, except for the acidic glucoside, which was recovered in small amounts only from the cell extracts. These products were the same as those recovered from intact plants. Similar results were obtained from cell suspensions of different ages. The rate of metabolism by log phase cells was slightly less than the rate for either young or old cells. The results indicated that soybean cell suspensions can be used to obtain reliable information on the fate of agricultural chemicals in soybeans.  相似文献

Indole-3-acetic acid (IAA) labeled in its carboxyl group was metabolized by tobacco leaf discs (Nicotiana tabacum L. cv. Xanthi) into three metabolites, two of which were preliminarily characterized as a peptide and an ester-conjugated IAA. Reapplication of each of the three metabolites (at 10 M) resulted in a marked stimulation of ethylene production and decarboxylation by the leaf discs. Similarly, these three IAA metab olites could induce elongation of wheat coleoptile segments, which was accompanied by decarboxylation. Both the exogenously supplied esteric and peptidic IAA conjugates were converted by the leaf discs into the same metabolites as free IAA. (1-14C)IAA, applied to an isolated epidermis tissue, was completely metabolized to the esteric and peptidic IAA conjugates. This epidermis tissue showed much higher ethylene production rates and lower decarboxylation rates than did the whole leaf disc.The results suggest that the participation of IAA conjugates in the regulation of various physiological processes depends on the release of free IAA, which is obtained by enzymatic hydrolysis of the conjugates in the tissue. Net photosynthetic rate (P N) measured at the same CO2 concentration, the maximum in vivo carboxylation rate, and contents of ribulose-1,5-bisphosphate (RuBP) carboxylase/oxygenase (RuBPCO) and RuBPCO activase were significantly decreased, but the maximum in vivo electron transport rate and RuBP content had no significant change in CO2-enriched [EC, about 200 μmol mol−1 above the ambient CO2 concentration (AC)] wheat leaves compared with those in AC grown wheat leaves. Hence photosynthetic acclimation in wheat leaves to EC is largely due to RuBP carboxylation limitation.  相似文献

Ibuprofen has previously, after ingestion by man, been demonstrated to yield four major phase I metabolites, which are excreted in the urine partly as glucuronic acid conjugates. However, in previous investigations the quantitative determinations of the conjugates were performed by indirect methods. The purpose of the present investigation was to develop a high-performance liquid chromatographic (HPLC) system for the simultaneous determination of the major phase I and II metabolites of ibuprofen in biological fluids. The separation was performed using bare silica dynamically modified with N-cetyl-N,N,N-trimethylammonium hydroxide ions contained in the mobile phase. The separation of the metabolites of ibuprofen is greatly improved with this system compared to other published reversed-phase HPLC systems intended for the same purpose. The method developed makes it possible to simultaneously determine the intact glucuronic acid conjugates of ibuprofen as well as its phase I metabolites in human urine. In a study involving four healthy volunteers, a total recovery in urine of the dose given was found to be 58–86% within 8 h. This may be compared to an average of 67% earlier reported in the literature.  相似文献

Wheat (T. durum cvs. HD 4502 and B 449, T. aestivum cvs. Kalyansona and Kundan) and sunflower (Helianthus annuus L. cv. Morden) were grown under atmospheric (360±10 cm3 m–3, AC) and elevated CO2 (650±50 cm3 m–3, EC) concentration in open top chambers for entire period of growth and development till maturity. Leaf net photosynthetic rate (P N) of EC-grown plants of wheat measured at EC was significantly decreased in comparison with AC-plants of wheat measured at EC. Sunflower, however, showed no significant depression in P N in EC-plants. There was a decrease in ribulose-1,5-bisphosphate carboxylase (RuBPC) activity, its activation state and amount in EC-plants of wheat, whereas no significant decrease was observed in sunflower. The above different acclimation to EC in wheat and sunflower was related with saccharide constituents accumulated in the leaves. Under EC, sunflower accumulated in the leaves more starch, whereas wheat accumulated more sugars.  相似文献

It has been shown that intensive elongation of wheat coleoptile sections is correlated with indoleacetic acid-1-14C metabolism. In tissues actively growing the disappearance of indoleacetic acid-1-14C and formation of conjugates and metabolites (indoleacetamide, indoleacetyl-β-D-glucose, indoleacetyl aspartic acid) is twice as high as in sections where growth has been stopped by physical constraint (plaster) or in tissue which did not elongate by an unknown reason.  相似文献

Three filamentous fungi were examined for the ability to biotransform phenanthrene to oxidative (phase I) and conjugative (phase II) metabolites. Phenanthrene metabolites were purified by high-performance liquid chromatography (HPLC) and identified by UV/visible absorption, mass, and1H NMR spectra.Aspergillus niger ATCC 6275,Syncephalastrum racemosum UT-70, andCunninghamella elegans ATCC 9245 initially transformed [9-14C]phenanthrene to produce metabolites at the 9,10-, 1,2-, and 3,4- positions. Subsequently, sulfate conjugates of phase I metabolites were formed byA. niger, S. racemosum, andC. elegans. Minor glucuronide conjugates of 9-phenanthrol and phenanthrenetrans-9,10-dihydrodiol were formed byS. racemosum andA. niger, respectively. In addition,C. elegans produced the glucose conjugates 1-phenanthryl -d-glucopyranoside and 2-hydroxy-1-phenanthryl -d-glucopyranoside, a novel metabolite. [9-14C]Phenanthrene metabolites were not detected in organic extracts from biotransformation experiments with the yeasts,Candida lipolytica 37-1,Candida tropicalis ATCC 32113, andCandida maltosa R-42.  相似文献

The biotransformation of [2-14C](±)9, 10-dihydrojasmonic acid (DJA) was studied in excised shoots of 6-day-old barley seedlings after 72 h. From the ethyl acetate extract, some minor metabolites were isolated and purified by DEAE-Sephadex A-25 chromatography, thin-layer chromatography (TLC), C18-cartridges, and high-performance liquid chromatography (HPLC). The structural identification of these metabolites was performed by gas chromatography-mass spectrometry (GC-MS), circular dichroism (CD), and amino acid analysis, and the following amino acid conjugates were found:N-[(–)9,10-dihydrojasmonoyl]valine,N-[(–)9,10-dihydrojasmonoyl]isoleucine,N-[9,10-dihydrojasmonoyl]leucine,N-[11-hydroxy-9,10-dihydrojasmonoyl]valine,N-[11-hydroxy-9,10-dihydrojasmonoyl]isoleucine,N-[12-hydroxy-9,10-dihydrojasmonoyl]isoleucine; and the cucurbic acid-related compoundsN-{[3-hydroxy-2(4-hydroxypentyl)-cyclopent-1-yl]-acetyl}isoleucine andN-{[3-hydroxy-2(5-hydroxypentyl)-cyclopent-1-yl]-acetyl}isoleucine. The results suggest conjugation with isoleucine and valine, as well as preferential hydroxylation at position C-11 or hydrogenation at position C-6, as being important steps in the metabolism of (±)DJA in barley shoots.  相似文献

We found that extracts from the leaves of medicinal comfrey and cowparsnip strongly inhibit the germination of Erysiphe graminisconidia and uredospores of Puccinia graminis. Spraying wheat seedlings with these extracts, in contrast to the irrigation of soil, markedly diminished infection in plants with powdery mildew. Antifungal activity in vitroand protective activity (when plants were sprayed) correlated with the level of phenolic compounds in these extracts. Experiments with healthy plants have demonstrated that the photosynthetic apparatus of wheat plants is stimulated by extracts. Spraying seedlings with the extracts resulted in an increased rate of O2evolution calculated per unit of chlorophyll, an increase in the ratio (FM– FT)/FTin the experiments that recorded slow fluorescence induction, an increase in the relative light intensity of band A, and a decrease of relative intensity of band Cin experiments with thermoluminescence of wheat leaves. These results provide evidence that the protective activity of comfrey and cowparsnip extracts is associated with their action on the pathogenic fungus and with the activation of natural defense reactions of the host plant.  相似文献

Hydrolysis, extraction, and radioimmunoassay techniques for the estimation of excreted testosterone metabolites in the urine of saddle-back tamarins have been validated and are described. The steroids measured with the testosterone antiserum used are mostly present as glucuronides and sulfates. Immunoreactivity in high-performance liquid chromatography (HPLC) fractions of urinary extracts and in a standard mixture of cortisol, testosterone, and dihydrotestosterone (DHT) were compared. The fractions with the same retention data as testosterone accounted for the major part of the immunoreactivity. Several other immunoreactive compounds of unknown identity were present in low concentrations. These results suggest that testosterone conjugates are the major steroid metabolites measured with this method in Saguinus fuscicollis. Urinary testosterone levels of castrated males were much lower than those of intact males. Testosterone treatment of castrated males resulted in a temporary superphysiological increase in the levels of urinary testosterone and in an individually variable increase in the levels of the minor immunoreactive compounds. These results suggest that estimation of testosterone metabolite levels in urine is a valid method for the assessment of testicular activity in Saguinus fuscicollis.  相似文献

Wheat (Triticum aestivum L. cv. HD 2329 and DL 1266-5) and sunflower (Helianthus annuus L. cv. MSFH 17 and MRSF 1754) plants were grown in field under atmospheric (360±10 cm3 m−3, AC) and elevated (650±50 cm3 m−3, EC) CO2 concentrations in open top chambers for entire period of growth and development till maturity. Net photosynthetic rate (P N) of wheat cvs. when compared at the same internal CO2 concentration (C i), by generating P N/C i curves, showed lower P N in EC plants than in AC ones. EC-grown wheat cultivars also showed a lesser response to irradiance than AC plants. In sunflower cultivars, P N/C i curves and irradiance response curves were not significantly different in AC and EC plants. CO2 and irradiance responses of photosynthesis, therefore, further revealed a down-regulation of P N in wheat but not so in sunflower under long-term CO2 enrichment. Wheat cvs. accumulated in leaves mostly sugars, whereas sunflower accumulated mainly starch. This further strengthened the view that accumulation of excess assimilates in the leaves under EC as starch is not inhibitory to P N.  相似文献

Requirements for mitigation of the continued increase in greenhouse gas (GHG ) emissions are much needed for the North China Plain (NCP ). We conducted a meta‐analysis of 76 published studies of 24 sites in the NCP to examine the effects of natural conditions and farming practices on GHG emissions in that region. We found that N2O was the main component of the area‐scaled total GHG balance, and the CH 4 contribution was <5%. Precipitation, temperature, soil pH , and texture had no significant impacts on annual GHG emissions, because of limited variation of these factors in the NCP . The N2O emissions increased exponentially with mineral fertilizer N application rate, with =  0.2389e0.0058x for wheat season and =  0.365e0.0071x for maize season. Emission factors were estimated at 0.37% for wheat and 0.90% for maize at conventional fertilizer N application rates. The agronomic optimal N rates (241 and 185 kg N ha?1 for wheat and maize, respectively) exhibited great potential for reducing N2O emissions, by 0.39 (29%) and 1.71 (56%) kg N2O‐N ha?1 season?1 for the wheat and maize seasons, respectively. Mixed application of organic manure with reduced mineral fertilizer N could reduce annual N2O emissions by 16% relative to mineral N application alone while maintaining a high crop yield. Compared with conventional tillage, no‐tillage significantly reduced N2O emissions by ~30% in the wheat season, whereas it increased those emissions by ~10% in the maize season. This may have resulted from the lower soil temperature in winter and increased soil moisture in summer under no‐tillage practice. Straw incorporation significantly increased annual N2O emissions, by 26% relative to straw removal. Our analysis indicates that these farming practices could be further tested to mitigate GHG emission and maintain high crop yields in the NCP .  相似文献

Summary Two experiments were performed to examine the effects of inoculation of field grown wheat with various Azospirillum strains. In the first experiment the soil was sterilized with methyl bromide to reduce the Azospirillum population and15N labelled fertilizer was added to all treatments. Two strains ofAzospirillum brasilense isolated from surface sterilized wheat roots and theA. brasilense type strain Sp7 all produced similar increases in grain yield and N content. From the15N and acetylene reduction data it was apparent that these increases were not due to N2 fixation. In the second experiment performed in the same (unsterilized) soil, twoA. brasilense strains (Sp245, Sp246) and oneA. amazonense strain (Am YTr), all isolated from wheat roots, produced responses of dry matter and N content while the response to the strain Sp7 was much smaller. These data confirm earlier results which indicate that if natural Azospirillum populations in the soil are high (the normal situation under Brazilian conditions), strains which are isolated from wheat roots are better able to produce inoculation responses than strains isolated from other sources. The inoculation of a nitrate reductase negative mutant of the strain Sp245 produced only a very small inoculation response in wheat. This suggests that the much greater inoculation response of the original strain was not due to N2 fixation but to an increased nitrate assimilation due to the nitrate reductase activity of the bacteria in the roots. Mycobacterium sp. 7E1B1W and seven other mycobacterial strains known to degrade hydrocarbons were investigated to determine their ability to metabolize the piperazine ring, a substructure found in many drugs. Cultures were grown at 30°C in tryptic soy broth and dosed with 3.1 mM N-phenylpiperazine hydrochloride; samples were removed at intervals and extracted with ethyl acetate. Two metabolites were purified from each of the extracts by high-performance liquid chromatography; they were identified by mass spectrometry and 1H nuclear magnetic resonance spectroscopy as N-(2-anilinoethyl)acetamide and N-acetyl-N′-phenylpiperazine. The results show that mycobacteria have the ability to acetylate piperazine rings and cleave carbon-nitrogen bonds.  相似文献

The metabolism of the herbicide, diclofop-methyl (methyl-2-[4-(2′,4′-dichlorophenoxy)phenoxy]propanoate, in cell suspensions of resistant diploid wheat (Triticum monococcum L.) was determined 1, 8, and 24 h after treatment with 14C-diclofop-methyl. The 14C-labeled products were identified by thin layer chromatographic comparisons to appropriate standards. Eight hours after treatment with 5 μM diclofop-methyl in 0.8% acetone (neither of which were toxic to the cell suspensions) 87.2% (84.0% methanol soluble, 3.2% methanol insoluble) of the total 14C recovered (90.4%) was in the cells and 12.8% was in the medium. Major metabolites found in methanol extracts of the cells were diclofop (2-[4-(2′,4′-dichlorophenoxy)phenoxylpropionic acid), diclofop hydroxylated at an undetermined position on the 2,4-dichlorophenyl ring (ring-OH diclofop), and conjugates of ring-OH diclofop. Acid hydrolysis of the conjugated metabolite(s) yielded ring-OH diclofop and diclofop. Twenty-four hours after treatment 70–75% of the total 14C recovered was present as conjugated metabolites. With the exception of ring-OH diclofop, all metabolites present in the cells were also recovered from the medium. A metabolite found in low concentrations in the medium that yielded diclofop upon hydrolysis was identified as an ester conjugate. Toxic concentrations of diclofop-methyl (10 and 20μM) had no effect on the metabolism of the herbicide, although the rate of uptake was slower than for cells treated with 5 μM herbicide. The products of diclofop-methyl metabolism in cell suspensions of T. monococcum were compared to previous data from T. aestivum intact plant metabolism of diclofop-methyl.  相似文献

1. Four new metabolites of phenacetin in the urine of the rat are described; these are (i) N-acetyl-S-ethylcysteine, (ii) quinol, (iii) acetamide and (iv) probably N-acetyl-S-2-(4-ethoxyacetanilido)cysteine S-oxide. 2. Metabolites (i), (iii) and (iv) were characterized and estimated by g.l.c., by t.l.c., by paper chromatography, by chemical reactions or by radioactive techniques after administration to rats of [ethyl-14C]phenacetin and [acetyl-3H]phenacetin; metabolite (ii), which was excreted mainly as conjugates of sulphuric acid and glucosiduronic acid, was measured by paper chromatography and characteristic colour reactions after enzymic and chemical hydrolysis of the conjugates. 3. Small amounts of azoxy-4-[ethyl-14C]ethoxybenzene and an unknown metabolite were also found in the urine of rats after administration of [ethyl-14C]phenacetin. 4. The likely mechanisms and some biological implications of these metabolic reactions are discussed.  相似文献

Partitioning of current photosynthates towards primary metabolites and its simultaneous incorporation in leaf alkaloids was investigated in developing leaves of medicinally important Catharanthus roseus. Of the total 14CO2 assimilated, the leaves at positions 1–6 fixed 8, 22, 25, 19, 13, and 8 %, respectively, and stem 3 %. Leaf fresh mass, chlorophyll content, and CO2 exchange rate increased up to the third leaf. The total alkaloid content was highest in young actively growing leaves, which declined with age. Total 14C fixed and its content in ethanol soluble fraction increased up to the third leaf and then declined. The 14C content in primary metabolites such as sugars and organic acids was also highest in the 3rd leaf. The utilization of 14C assimilates into alkaloids was maximum in youngest leaf which declined with leaf age. Hence the capacity to synthesize alkaloids was highest in young growing leaves and metabolites from photosynthetic pathway were most efficiently utilized and incorporated into alkaloid biosynthetic pathway by young growing leaves.  相似文献
