A new technique is presented that utilizes relative velocity vectors between articulating surfaces to characterize internal/external rotation of the tibio-femoral joint during dynamic loading. Precise tibio-femoral motion was determined by tracking the movement of implanted tantalum beads in high-speed biplane X-rays. Three-dimensional, subject-specific CT reconstructions of the femur and tibia, consisting of triangular mesh elements, were positioned in each analyzed frame. The minimum distance between subchondral bone surfaces was recorded for each mesh element comprising each bone surface, and the relative velocity between these opposing closest surface elements was determined in each frame. Internal/external rotation was visualized by superimposing tangential relative velocity vectors onto bone surfaces at each instant. Rotation about medial and lateral compartments was quantified by calculating the angle between these tangential relative vectors within each compartment. Results acquired from 68 test sessions involving 23 dogs indicated a consistent pattern of sequential rotation about the lateral condyle (approximately 60 ms after paw strike) followed by rotation about the medial condyle (approximately 100 ms after paw strike). These results imply that axial knee rotation follows a repeatable pattern within and among subjects. This pattern involves rotation about both the lateral and medial compartments. The technique described can be easily applied to study human knee internal/external rotation during a variety of activities. This information may be useful to define normal and pathologic conditions, to confirm post-surgical restoration of knee mechanics, and to design more realistic prosthetic devices. Furthermore, analysis of joint arthrokinematics, such as those described, may identify changes in joint mechanics associated with joint degeneration.  相似文献

Mechanical dissociation of neurons from the central nervous system has the advantage that presynaptic boutons remain attached to the isolated neuron of interest. This allows for examination of synaptic transmission under conditions where the extracellular and postsynaptic intracellular environments can be well controlled. A vibration-based technique without the use of proteases, known as vibrodissociation, is the most popular technique for mechanical isolation. A micropipette, with the tip fire-polished to the shape of a small ball, is placed into a brain slice made from a P1-P21 rodent. The micropipette is vibrated parallel to the slice surface and lowered through the slice thickness resulting in the liberation of isolated neurons. The isolated neurons are ready for study within a few minutes of vibrodissociation. This technique has advantages over the use of primary neuronal cultures, brain slices and enzymatically isolated neurons including: rapid production of viable, relatively mature neurons suitable for electrophysiological and imaging studies; superior control of the extracellular environment free from the influence of neighboring cells; suitability for well-controlled pharmacological experiments using rapid drug application and total cell superfusion; and improved space-clamp in whole-cell recordings relative to neurons in slice or cell culture preparations. This preparation can be used to examine synaptic physiology, pharmacology, modulation and plasticity. Real-time imaging of both pre- and postsynaptic elements in the living cells and boutons is also possible using vibrodissociated neurons. Characterization of the molecular constituents of pre- and postsynaptic elements can also be achieved with immunological and imaging-based approaches.  相似文献

Developing an efficient parallel application is not an easy task, and achieving a good performance requires a thorough understanding of the program’s behavior. Careful performance analysis and optimization are crucial. To help developers or users of these applications to analyze the program’s behavior, it is necessary to provide them with an abstraction of the application performance. In this paper, we propose a dynamic performance abstraction technique, which enables the automated discovery of causal execution paths, composed of communication and computational activities, in MPI parallel programs. This approach enables autonomous and low-overhead execution monitoring that generates performance knowledge about application behavior for the purpose of online performance diagnosis. Our performance abstraction technique reflects an application behavior and is made up of elements correlated with high-level program structures, such as loops and communication operations. Moreover, it characterizes all elements with statistical execution profiles. We have evaluated our approach on a variety of scientific parallel applications. In all scenarios, our online performance abstraction technique proved effective for low-overhead capturing of the program’s behavior and facilitated performance understanding.  相似文献

Severe thermal injuries to the external ear often lead to extensive loss of peripheral structures such as helix and lobule but frequently spare the more central parts of the ear, even though they may be grossly deformed by scar contracture. The use of spared conchal structures as a transposition flap in combination with remodeling of the residual auricle and release of surrounding scar when indicated has been a useful technique in the reconstruction of a frequently occurring type of postburn ear deformity. Twenty-four ears have been reconstructed in 18 patients over the past 5 years using a conchal transposition flap. The residual concha with its overlying skin can be transposed superiorly, based on a very narrow pedicle in the area of the crus helicis. The raw central area remaining is then resurfaced with a split-thickness skin graft. This technique maximally utilizes the unique remaining auricular elements and can provide a satisfactory reconstruction in selected patients without resorting to more complex and extensive procedures. There have been no significant complications in this series, and patient acceptance of the results has been excellent.  相似文献

Micro-finite element (FE) analysis is a well established technique for the evaluation of the elastic properties of trabecular bone, but is limited in its application due to the large number of elements that it requires to represent the complex internal structure of the bone. In this paper, we present an alternative FE approach that makes use of a recently developed 3D-Line Skeleton Graph Analysis (LSGA) technique to represent the complex internal structure of trabecular bone as a network of simple straight beam elements in which the beams are assigned geometrical properties of the trabeculae that they represent. Since an enormous reduction of cputime can be obtained with this beam modeling approach, ranging from approximately 1,200 to 3,600 for the problems investigated here, we think that the FE modeling technique that we introduced could potentially constitute an interesting alternative for the evaluation of the elastic mechanical properties of trabecular bone.  相似文献

We propose an excitation technique for observing single and two photon excitation in those molecules for which such transitions are forbidden by the selection rules. This is possible by the application of an external electric field that perturbs the molecular orbitals, thereby resulting in a significant shift of energy levels. Such a shift of energy levels may bring those levels in resonance with the radiation field which is normally forbidden by selection rules. Further, parity of the these states may significantly improve the emission process. The external electric field results in the mixing of excited (short lifetime) and metastable states (long lifetime), thus reducing the lifetime of metastable (or near metastable) states. This may provide an effective channel for allowing transition from the metastable states. An application of electric field may result in the excitation of poorly excitable biomolecules. This excitation technique may find applications in single- and multi-photon fluorescence microscopy, bioimaging and optical devices.  相似文献

MOTIVATION: A plugin for the Java-based PathVisio pathway editor has been developed to help users draw diagrams of bioregulatory networks according to the Molecular Interaction Map (MIM) notation. Together with the core PathVisio application, this plugin presents a simple to use and cross-platform application for the construction of complex MIM diagrams with the ability to annotate diagram elements with comments, literature references and links to external databases. This tool extends the capabilities of the PathVisio pathway editor by providing both MIM-specific glyphs and support for a MIM-specific markup language file format for exchange with other MIM-compatible tools and diagram validation. AVAILABILITY: The PathVisio-MIM plugin is freely available and works with versions of PathVisio 2.0.11 and later on Windows, Mac OS X and Linux. The microbeam synchrotron X-ray fluorescence (MSXRF) technique is powerful tool for determining elemental composition and spatial distribution of elements within materials on the microscopic level. Therefore, application of this technique seems to be helpful for investigation of the role of trace elements in neurodegenerative processes. As a preliminary study the MSXRF technique was applied for elemental microanalysis in cryo-sections of substantia nigra tissue for cases of Parkinson's disease (PD) and the control. Two-dimensional maps of elemental distribution showed significant topographic and quantitative differences between the pathological and control tissues. Quite interesting is significant increased accumulation of Ca, Cu, Fe, Fe, and Zn that was observed in PD neuron perikaryal parts.  相似文献

The skills that mountain gorillas use to deal with the stings, tiny hooks, and spines protecting common plant leaves in their diet were examined for variation within and between animals. Many elements of uni- and bimanual performance were identified, often involving delicate precision and coordination, and varying idiosyncratically, erch individual having a different set of preferred elements. Many of these elements are functionally equivalent, and all but one weaned animals showed full processing capability; the history of the one exception suggests that early experience with the task may be important. Gorillas' idiosyncrasy in manual skill elements is entirely consistent with trial-and-error learning at this level. By contrast, each individual uses very few techniques (structured sequences of elements) for most processing, and these techniques are the same across the population. Where animals deviate from this generalization, they largely employ the simpler technique normally used for undefended leaves. Lateralization increases from start to finish, consistent with a logical structure in which each stage has a laterality bias and each stage is sequentially dependent on the last. Variations from their commonest, techniques occur in all animals (on average, about nine variant techniques were recorded from each animal). The repertoire of techniques increases significantly with age, whereas the repertoire of elements does not. This points to an initial reliance on a single logical structuring that is well established by weaning (about 3.5 years), with subsequent development of the ability to vary the technique used so as to take advantage of variations in the environment. Standardization of logical organization, despite variability between different animals in individual elements and behavioral laterality, suggests that the logical ordering of elements and the interrelationships of processing stages is copied by program-level imitation. © 1993 Wiley-Liss, Inc.  相似文献

丛枝菌根(arbuscular mycorrhizal,AM)真菌是生态系统中重要的土壤微生物之一。AM真菌菌丝体网络是由AM真菌菌丝体在土壤生态系统中连接两株或两株以上植物根系所形成的菌丝体网络。随着菌根学研究的深入,如何直观的揭示AM真菌的生态学功能已经成为相关领域关注的热点问题。研究发现,利用同位素示踪技术可以开展AM真菌与宿主植物对土壤矿质营养的吸收、转运等方面的研究,以及菌丝体网络对不同宿主植物之间营养物质的分配研究和AM真菌在生态系统生态学中的功能研究。基于此,为了阐明同位素示踪技术在AM真菌研究中的价值,围绕菌根学最新研究进展,系统回顾了利用同位素示踪技术探究AM共生体对不同元素吸收和转运的机制、同位素示踪技术在AM真菌菌丝体网络研究中的价值和利用同位素示踪技术研究AM真菌在生态系统中的功能,为AM真菌生态学功能的研究提供理论基础,并对本领域未来的研究方向和应用前景进行展望。  相似文献

Summary With the use of the unlabelled antibody peroxidase-antiperoxidase complex (PAP) technique at the electron microscopic level, it was shown that both the internal and the external regions of the median eminence of the frog contain separate vasotocinergic and mesotocinergic nerve fibres. This observation confirms the results of previous immunocytochemical studies at the light microscopic level. The mean size of the neurohypophysial hormone-containing granules in the external region of the median eminence is significantly smaller than that of the neurohypophysial hormone-containing granules in the internal region of the median eminence. Patients with breast cancer are often treated with radiation to the breast (or chest wall) and draining regional lymph nodes. This is typically performed with a three-field technique in which an anterior supraclavicular field is matched to opposed tangent fields. A single isocenter technique is not always possible. Several techniques have been described to create a perfect match using a conventional simulator. We describe and test a simple, fast and accurate technique to estimate the couch and collimator angles required for a perfect geometric match using 3-D treatment planning software. This method requires no mathematical formulae and is verifiable relative to patient anatomy.An external skin contour is created on the axial slice at the match line and displayed with a 3-D representation. Using a beam's eye view (BEV) of a tangent field, small couch and collimator rotation adjustments are made sequentially until the contour edges are superimposed. The virtual external contour technique was easy to use, gave verification of the match in the BEV and yielded estimates of couch and collimator rotations very close to those calculated using published formulae.  相似文献

Developmental processes in cells require a series of complex steps. Often only a single master regulator activates genes in these different steps. This poses several challenges: some targets need to be ordered temporally, while co-functional targets may need to be synchronized in both time and expression level. Here we study in single cells the dynamic activation patterns of early meiosis genes in budding yeast, targets of the meiosis master regulator Ime1. We quantify the individual roles of the promoter and protein levels in expression pattern control, as well as the roles of individual promoter elements. We find a consistent expression pattern difference between a non-cofunctional pair of genes, and a highly synchronized activation of a co-functional pair. We show that dynamic control leading to these patterns is distributed between promoter, gene and external regions. Through specific reciprocal changes to the promoters of pairs of genes, we show that different genes can use different promoter elements to reach near identical activation patterns.  相似文献

The application of liquid crystal (LC) materials has undergone a modern-day renaissance from its classical use in electronics industry as display devices to new-fangled techniques for optically detecting biological and chemical analytes. This review article deals with the emergence of LC materials as invaluable material for their use as label-free sensing elements in the development of optical, electro-optical and electrochemical biosensors. The property of LC molecules to change their orientation on perturbation by any external stimuli or on interaction with bioanalytes or chemical species has been utilized by many researches for the fabrication of high sensitive LC-biosensors. In this review article we categorized LC-biosensor based on biomolecular reaction mechanism viz. enzymatic, nucleotides and immunoreaction in conjunction with operating principle at different LC interface namely LC-solid, LC-aqueous and LC-droplets. Based on bimolecular reaction mechanism, the application of LC has been delineated with recent progress made in designing of LC-interface for the detection of bio and chemical analytes of proteins, virus, bacteria, clinically relevant compounds, heavy metal ions and environmental pollutants. The review briefly describes the experimental set-ups, sensitivity, specificity, limit of detection and linear range of various viable and conspicuous LC-based biosensor platforms with associated advantages and disadvantages therein.  相似文献

植物修复是利用植物的吸收和代谢功能将环境介质中的污染物进行分解、富集和稳定的过程。由于污染物的理化特性和环境行为不同,加之植物新陈代谢的各异,污染物植物修复的作用机理也不尽相同。与其它修复方法相比,植物修复技术投入低、效果好和纯自然,并对土壤功能的维护和土壤生物恢复有积极的作用。本文综述了近年来国外有关植物修复技术在污染治理领域中的研究进展,详细论述了推广应用污染物植物修复过程中应注意的几个技术关键;并就植物修复技术中存在的问题进行了讨论。总之,污染物植物修复技术的发展目前还不完善,就其更为详细具体的作用机理和商业推广的研究还需要做大量的工作。  相似文献

In the study of active transport it is important to distinguish between oxygen consumption sustaining transepithelial transport and that responsible for other tissue functions (basal metabolism). Since amiloride blocks transepithelial active sodium transport and the associated oxygen consumption in the frog skin and toad bladder, we and others have employed this agent to evaluate the rate of basal metabolism. This technique has recently been criticized in a report that amiloride (and ouabain) increased oxygen consumption when no sodium was available for transport. We have been unable to corroborate these observations. With magnesium-Ringer as external bathing solutions, amiloride and ouabain failed to stimulate oxygen consumption. With sodium-Ringer as external bathing solution amiloride reduced oxygen consumption about 30%, to a level indistinguishable from that found on external substitution of magnesium-Ringer for sodium-Ringer. We conclude that the use of amiloride permits evaluation of the rate of basal metabolism with acceptable accuracy; a possible slight depressant effect of ouabain on basal metabolism remains to be investigated.  相似文献
