The proteins of Hordeum chilense grain were resolved into 25 major components by two-dimensional electrophoresis. Their solubilities in aqueous alcohol solutions were determined to distinguish prolamin storage proteins from metabolic and structural proteins. The prolamins were divided into two groups, based on the presence or absence of intermolecular disulfide bonds determined by gel-filtration chromatography. Using an incomplete set of Chinese Spring wheat-H. chilense disomic addition lines, the structural genes of 21 of the 26 most dominant seed proteins were assigned to chromosomes. The great majority of the prolamin genes, including those coding for a high molecular weight (HMW) prolamin subunit, was present on chromosome 1Hch. However, a small number of prolamin genes also occurred on chromosomes 5Hch and 7Hch. A minor protein, probably belonging to the nonstorage group of proteins, is coded by genes on 5Hch. Various ditelosomic addition lines and ditelosomic and disomic substitution lines for chromosome 7Hch were also analyzed by electrophoresis. This technique revealed that the genes for three major prolamins occur on the arm of chromosome 7Hch and that a gene for a minor protein, also thought to be a prolamin, occurs on the arm. These results are discussed in relation to the evolution of prolamin genes in the Triticeae.  相似文献

Nine mutant lines lacking glutelin subunits were selected from M2 seeds of about 10000 M1 plants mutagenized with gamma rays or EMS and from 1400 mutant lines selected originally for morphological characters. There were three types of mutants, one line lacking the largest subunit among four minor bands of glutelin acidic subunits (Type 1), five lines lacking the second largest subunit band (Type 2), and three lines lacking the third largest subunit band (Type 3). Mutants lacking the smallest subunit band were not found. Type 1 lacked 2 of the 10 spots of glutelin acidic subunits separated by two-dimensional electrophoresis and 1 of the 11 spots of the 57-kDa glutelin precursor. Type 2 lacked 2 spots of acidic subunits and 1 spot of the 57-kDa glutelin precursor, and had low amounts of 1 of the 8 spots of glutelin basic subunits. Type 3 mutants lacked each of 1 spot of the acidic subunits and glutelin precursor and had low amount of 1 spot of the basic subunits. Genetic analysis of the mutated genes showed that these mutant characters were controlled by single recessive genes named glu-1, glu-2, and glu-3, respectively. Mutated genes of different lines of the same type were found to be at the same locus. RFLP analysis of F2 plants between the mutant lines and cv `Kasalath' indicated that glu-1 is on chromosome 2, glu-2 on chromosome 10, and glu-3 on chromosome 1. These mutant genes were combined by crossing, and a line lacking the 3 minor bands of the glutelin acidic subunits was developed. Two bands of putative Drosophila chorion mRNA, E3 and E4, have been shown to hybridize in situ near 7E11 (Spradling and Mahowald, 1979) within a region known to contain a gene, ocelliless, which may be involved in chorion production (Johnson and King, 1974). We have investigated the synthesis of “chorion mRNAs” and chorion proteins in flies carrying this mutation. A reduction of labeling of both E3 and E4 was observed in stage 12 egg chambers from homozygous ocelliless females. In addition, they produce mature oocytes which contain greatly reduced amounts of several major chorion proteins, including those normally produced in stage 12, c36 and c38. To investigate whether the reduction was due to a direct effect of the mutation, the genes for these proteins were mapped. Recombination analysis using electrophoretic variants of c36 and c38 showed that both proteins are coded on the X chromosome at a site between crossveinless and vermillion. Further mapping with the deficiency chromosomes Df(1)KA14 and Df(1)RA2 narrowed the region containing the structural genes to a 16 band region between 7E10 and 8A4. The ocelliless gene, as well as the site of in situ hybridization, are located within this same interval. In normal ovarian follicles, both c36 and c38 are produced in equal amounts and with the same developmental specificity (Waring and Mahowald, 1979). In the mutant, both are reduced to a similar extent. In oc+/oc heterozygotes, both the c36 and c38 genes on the mutant chromosome produce much less product than the corresponding genes on the oc+ chromosome. The cis-acting nature of the ocelliless mutation suggests that it may disrupt sequences involved in controlling the expression of the structural information for these proteins.  相似文献

The necrotrophic pathogen Pectobacterium carotovorum ssp. carotovorum (Pcc) causes soft rot in a broad range of plant hosts. Approximately 60,000 independent seeds from Arabidopsis activation tagging lines were inoculated with Pcc and screened for resistant mutants. An Rpe1 (resistance protein to Pectobacterium 1) mutant, which had more resistance to Pcc than wild-type (WT) plants, was selected for further study. The T-DNA inserting locus in Rpe1 was located on the middle of chromosome V by flanking sequence analysis. Through expression analysis with several genes adjacent to the T-DNA tagging region, AtWRKY75 gene was highly up-regulated in the Rpe1 mutant compared to the WT plant. The up-regulation of AtWRKY75 gene was shown to be correlated on the induction of the PDF1.2, VSP1 and PR1 genes compared to the WT plant. AtWRKY75 over-expression lines exhibited reduced Pcc bacterial growth compared to WT. Taken together, our data suggest that AtWRKY75 should be a positive regulator in the JA- or SA-mediated defense signaling responses to Pcc.  相似文献

A method is described for unambiguous assignment of cloned genes to Aspergillus niger chromosomes by CHEF gel electrophoresis and Southern analysis. All of the eight linkage groups (LGs), with the exception of LG VII, have previously been assigned to specific chromosomal bands in the electrophoretic karyotype of A. niger. Using a LG VII-specific probe (nicB gene of A. niger) we have shown that LG VII corresponds to a chromosome of about 4.1 Mb. Furthermore, genetic localization of three unassigned genes (glaA, agIA and pepA) in strains in which these genes had been replaced by a selectable marker gene led to a revised karyotype for the chromosomes corresponding to LGs VIII and VI. The revised electrophoretic karyotype reveals only 5 distinct bands. The presence of three pairs of equally sized chromosomes precluded assignment of genes to one specific chromosome in the wild-type strain. However, unambiguous chromosome assignment of cloned genes using CHEF-Southern analysis was demonstrated using a set of A. niger strains with introduced chromosomal size variation. The availability of these tester strains obviates the need to isolate or construct mutant. strains for the purpose of chromosome assignment.  相似文献

The genetic properties of a recessive autosomal point mutant, "daughterless" (symbol: da), are described. They are: (1) da maps in the euchromatin of 2L at about position 39 on the genetic map and between 27D and 31E on the salivary map; (2) when homozygous in females, da causes the production of unisexual male progenies owing to sex differential zygote mortality in the egg stage; (3) da has no effect on the progeny of mutant males; (4) female zygotes die, while at least some male zygotes survive, irrespective of the number of Y chromosomes or the amount of X-chromosome heterochromatin carried by either the mutant female or her progeny; (5) homozygous da progeny of heterozygous parents also show sex differential survival favoring males indicating a da effect in development as well as in oogenesis, but with the developmental effect much less pronounced; (6) extra heterochromatin from either the X or Y chromosome in either the mother or her progeny can reduce the mortality caused by da in development (and, therefore, conceivably can also reduce the abnormality in oogenesis, although this is obscured by the severity of the maternal effect.)—From these results, it is suggested that (1) da regulates either the activity of structural genes in the sex chromosome heterochromatin or the activity or stability of their products, and (2) it is a defective product of sex chromosome heterochromatic genes that causes the abnormalities resulting in the observed mortality of heterozygous progeny of homozygous mutant mothers and of homozygous mutant progeny of heterozygous mothers.—The striking parallels with the properties of the gene, "abnormal oocyte" are noted as is the close linkage between the two loci. The possibility of a special sex-chromosome-heterochromatin-regulator region on chromosome 2 is considered.  相似文献

A cytological and genetic study of oogenesis in Drosophila melanogaster   总被引:5,自引:0,他引:5  
A newly derived collection of 98 autosomal recessive female-sterile (fs) lines induced by ethyl methanesulfonate (EMS) in Drosophila melanogaster has been studied genetically and cytologically. By the use of phenotype and complementation tests, the 98 fs lines were resolved into 19 fs genes on the 2nd chromosome and 17 fs genes on the 3rd chromosome. Nearly half of the fs lines turned out to be replicates of noninduced fs mutant loci that preexisted in the EMS-treated files.Systematizing these fs genes according to their phenotypic and morphological defects focuses attention on their relevance to five major aspects of oogenesis: (1) the developmental organization of the ovary, (2) the synthesis and deposition of yolk material, (3) the formation of the chorion, (4) the control of egg-laying, and (5) the construction of the internal milieu of the ovarian oocyte for normal embryogenesis.The results of this study demonstrate that a systematic collection and characterization of fs mutants can provide the genetic tools needed to study the complex interactions which proceed undetected during normal oogenesis.  相似文献

Two independent isolates of Saccharomyces cerevisiae lacking hexokinase activity (EC 2.7.1.1) are described. Both mutant strains grow on glucose but are unable to grow on fructose, and contain two mutant genes hxk1 and hxk2 each. The mutations are recessive and noncomplementing. Genetic analysis suggests that these two unlinked genes hxk1 and hxk2 determine, independently of each other, the synthesis of hexokinase isozymes P1 and P2, respectively. hxk1 is located on chromosome VIR distal to met10, and hxk2 is on chromosome IIIR distal to MAL2. Of four hexokinase-positive spontaneous reversions, one is very tightly linked to hxk1 and the other three to the hxk2 locus. The reverted enzymes are considerably more thermolabile than the respective wild-type enzymes, and in one case show altered immunological properties. Data are presented which suggest that the hxk1 and hxk2 mutations are missense mutations in the structural genes of hexokinase P1 and hexokinase P2, respectively. These are presumably the only enzymes that allow S. cerevisiae to grow on fructose.  相似文献

A novel yellow-green leaf mutant yellow-green leaf-1 (ygl-1) was isolated in self-pollinated progenies from the cross of maize inbred lines Ye478 and Yuanwu02. The mutant spontaneously showed yellow-green character throughout the lifespan. Meanwhile, the mutant reduced contents of chlorophyll and Car, arrested chloroplast development and lowered the capacity of photosynthesis compared with the wild-type Lx7226. Genetic analysis revealed that the mutant phenotype was controlled by a recessive nuclear gene. The ygl-1 locus was initially mapped to an interval of about 0.86 Mb in bin 1.01 on the short arm of chromosome 1 using 231 yellow-green leaf individuals of an F2 segregating population from ygl-1/Lx7226. Utilizing four new polymorphic SSR markers, the ygl-1 locus was narrowed down to a region of about 48 kb using 2930 and 2247 individuals of F2 and F3 mapping populations, respectively. Among the three predicted genes annotated within this 48 kb region, GRMZM2G007441, which was predicted to encode a cpSRP43 protein, had a 1-bp nucleotide deletion in the coding region of ygl-1 resulting in a frame shift mutation. Semi-quantitative RT-PCR analysis revealed that YGL-1 was constitutively expressed in all tested tissues and its expression level was not significantly affected in the ygl-1 mutant from early to mature stages, while light intensity regulated its expression both in the ygl-1 mutant and wild type seedlings. Furthermore, the mRNA levels of some genes involved in chloroplast development were affected in the six-week old ygl-1 plants. These findings suggested that YGL-1 plays an important role in chloroplast development of maize.  相似文献

The extracellular proteases ofAspergillus nidulans are known to be regulated by carbon, nitrogen and sulphur metabolite repression. In this study, a mutant with reduced levels of extracellular protease was isolated by screening for loss of halo production on milk plates. Genetic analysis of the mutant showed that it contains a single, recessive mutation, in a gene which we have designatedxprE, located on chromosome VI. ThexprE1 mutation affected the production of extracellular proteases in response to carbon, nitrogen and, to a lesser extent, sulphur limitation. Three reversion mutations,xprF1, xprF2 andxprG1, which suppressxprE1, were characterised. BothxprF andxprG map to chromosome VII but the two genes are unlinked. ThexprF1, xprF2 andxprG1 mutants showed high levels of milk-clearing activity on medium containing milk as a carbon source but reduced growth on a number of nitrogen sources. Evidence is presented that thexprE1 andxprG1 mutations alter expression of more than one protease and affect levels of alkaline protease gene mRNA.  相似文献

The storage protein polymers in the endosperm, stabilised by disulphide bonds, determine a number of processing qualities of wheat dough. The enzyme protein disulphide isomerase (PDI), involved in the formation of disulphide bonds, is strongly suggested to play a role in the formation of wheat storage protein bodies. Reports of the rice mutant esp2 exhibiting aberrant storage protein deposition in conjunction with a lack of PDI expression provided strong indications of a direct role for PDI in storage protein deposition. The potential significance of wheat PDI prompted the present studies into exploring any orthology between wheat PDI genes and rice PDI and esp2 loci. By designing allele-specific (AS)-polymerase chain reaction (PCR) markers, two of the three wheat PDI genes could be genetically mapped to group 4 chromosomes and showed close association with GERMIN genes. Physical mapping led to localisation of wheat PDI genes to chromosomal “bins” on the proximal section of chromosome 4AL and distal sections of 4BS and 4DS. Identification of the putative PDI gene of rice and its comparison to the esp2 locus revealed that they were present at similar positions on the short arm of chromosome 11. Analysis of a large section of the PDI-containing section of rice chromosome 11S revealed a number of putative orthologues from The Institute for Genomic Research Triticum aestivum Gene Index database, of which five had been mapped, each localising to group 4 chromosomes, many in good agreement with our mapping results. The results strongly suggest a close linkage between the esp2 marker and the PDI gene of rice and an orthology between the PDI loci of rice and wheat and predict quantitative-trait loci involved in storage protein deposition at the PDI loci.  相似文献

Chromosomal rearrangements involving the EVI1 proto-oncogene are a recurrent finding in myeloid leukemias and are indicative of a poor prognosis. Rearrangements of the EVI1 locus are often associated with monosomy 7 or cytogenetic detectable deletions of part of 7q. As EVI1 overexpression alone is not sufficient to induce leukemia, loss of a 7q tumour suppressor gene might be a required cooperating event. To test this hypothesis, we performed high-resolution array comparative genomic hybridization analysis of twelve EVI1 overexpressing patients and three EVI1 deregulated cell lines to search for 7q submicroscopic deletions. This analysis lead to the delineation of two critical regions, one of 0.39 Mb on 7q35 containing the CNTNAP2 gene and one of 1.33 Mb on chromosome bands 7q35–q36 comprising nine genes in EVI1 deregulated cell lines. These findings open the way to further studies aimed at identifying the culprit EVI1 implicated tumour suppressor genes on 7q.  相似文献

The locations of the larval serum protein one (LSP-1) α, β and γ genes were determined in Drosophila melanogaster and in 14 other species of Drosophila by in situ hybridization to polytene chromosomes. The LSP-1 α gene mapped to bands 11B on the X chromosome, the LSP-1 β gene mapped to bands 21D-E on chromosome 2L, and the LSP-1 γ gene mapped to band 61A in all the melanogaster subgroup species. In eight other species, both the LSP-1 α and β genes mapped to one site on Muller's element E which corresponds to chromosome 3R of D. melanogaster. No hybridization of LSP-1 γ was detected in these eight species. Restriction enzyme digestion and analysis of genomic DNA by filter transfer hybridization confirmed the presence of LSP-1 α-like and β-like genes in seven of these species. These results are discussed with respect to conservation of the chromosomal elements in the genus Drosophila.  相似文献

The 5S RNA genes of Drosophila melanogaster in either an isogenic wild-type or a multiply inverted (SM1) chromosome 2 increase their multiplicity when opposite a deficiency for the 5S gene site. This is analogous to the compensation phenomenon previously described for the 18S and 28S ribosomal RNA genes of the X chromosome nucleolus organizer region. Molecular hybridization of 5S RNA to DNA containing various doses of the 56F1-9 region of chromosome 2 demonstrates that most, if not all, of the 5S genes reside in or near this region. Also, a deficiency missing approximately one-half of the wild-type number of 5S genes was isolated and genetically localized. This mutant has a phenotype like that of bobbed, a mutant known to be partially deficient in 18S and 28S ribosomal RNA genes. Finally, we report the existence of a chromosomal rearrangement which splits the second chromosome into two segments, each containing 5S DNA.  相似文献

Region 98EF-100F in chromosome 3 is interesting for genetic analysis because it contains a number of genes of developmental importance. Although there are no preexisting simple deficiency stocks, this region is amenable to genetic manipulation using other types of rearrangements. In the present investigation we obtained deficiencies by combining the terminal deficiencies formed by segregation of Y;3 translocations with a series of duplications of the tip of 3R, both from Y;3 translocations with different breakpoints and from 3;1 duplications in which the 3R tip is carried as a second arm on the X chromosome. Analysis of such synthetic deficiencies reveals five haplo-abnormal loci in the 98A-100F interval. These include a haplolethal site, a newly described Minute and three previously reported Minute mutations. The newly discovered Minute has been designated M(3)99D and is localized cytologically to bands 99D1-9. The three previously reported Minute loci in the region have been localized more precisely: M(3)1 to bands 99B5-9, M(3)f to bands 99E4-F1 and M(3)g to region 100C-F. In addition, we have been able to obtain synthetic deficiencies uncovering all of the intervals from 99B5 to 100B. These deficiencies will be useful for future genetic and molecular analyses of the genes that map within the right tip of chromosome 3.  相似文献
