Extracts of cotyledons of Brassica napus plants (seed progenies of doubled haploid plants) were separated by electrophoresis on polyacrylamide gels and stained for acid phosphatase (ACP-E.C. 3.1.3.2.) and leucine aminopeptidase (LAP-E.C. 3.4.11.1.) enzymes to investigate the possibility of utilising isozymes as markers of homogeneity (purity) of plant populations. One zone of activity for acid phosphatase and two zones of activity for leucine aminopeptidase were identified on gels, some variation in isozyme patterns occurred in several androgenetic lines. This method is appropriate and consistent for testing the homogeneity of breeding lines-progenies of double haploid (D.H.) plants.  相似文献

Size variability in plant populations has been extensively studied andmuch of this inquiry has focused on the types of competition that are involvedin increasing or decreasing size inequality. It is often assumed thatcompetition for light is size-asymmetric, meaning that a plant canpotentially dominate a competitive relationship through shading if it is tallerthan its competitor. The light resources obtained by the taller plant are thusdisproportionate to its size. In contrast, competition for soil resources maybemore size-symmetric, the amount of soil nutrients obtained seems to be indirect proportion to a plant's size. Most studies examining belowgroundcompetition have used homogeneouslydistributed nutrient resources. Soil homogeneity could makesize-asymmetric belowground competition unlikely, but homogeneity is notoften found in nature. In this study I use a greenhouse pot experimentutilizingIpomoea tricolor to examine the hypothesis thatsize-asymmetric competition for soil nutrients may result when soilresource distribution is spatially heterogeneous. The results did not supportthe hypothesis of belowgroundsize-asymmetric competition. Differences between experimental treatmentsand controls were not statistically significant suggesting size-symmetriccompetition. Implications of these results are discussed.  相似文献

Estimating the fitness of line crosses has been a key element in studies of inbreeding depression, hybridization, and speciation. Fitness values are typically compared using differences in the arithmetic mean of a fitness component between types of crosses. One aspect of fitness that is often overlooked is variance in offspring fitness over time. In the majority of studies, ignoring this aspect of fitness is unavoidable because it is impossible to estimate variance in offspring fitness over long time periods. Here, I describe a method of estimating variance in offspring fitness by substituting spatial variation for temporal variation and provide an empirical example. The method is based on Levene's test of homogeneity of variances. It is implemented by quantifying differences in residual variation among cross types. In a previous study, I performed crosses between populations of the annual plant Diodia teres and quantified hybrid fitness. In this study, another component of isolation and heterosis was revealed when considering variance in offspring fitness. When taking into account variance in offspring fitness using geometric mean fitness as the measure of performance, hybrids between populations from different habitats showed less heterosis than when calculating fitness based on arithmetic mean. This study demonstrates that variance in offspring fitness can be an important aspect of fitness that should be measured more frequently.  相似文献

Different studies suggest some social calls could be used in fish identification if their specificity is unambiguously assessed. Sounds of different populations of piranhas Serrasalmus maculatus Kner, 1858 were recorded to determine their homogeneity between rivers inside a single basin (Araguari and Grande River, upper Paraná River basin) and between separated basins (Amazon and Paraná basins). All fish from the different populations produced sounds with similar acoustic features. Consequently, the populations were not discernible based on individual sound characteristics. This high homogeneity between sounds from different populations indicates their usefulness for conservation projects using passive acoustic monitoring in piranhas. Moreover, it supports the use of acoustic features as complementary key characteristics in taxonomic studies.  相似文献

Significant reduction of genetic diversity was observed in seed coat colour and electrosphoretic isozyme genotypes when infected plants were eliminated in genetically heterogeneous pea populations. Some accessions where genetic homogeneity was expected were apparently mixtures. Detection of latent infections by pea seedborne mosaic virus by the ELISA method was more effective on single-plant samples than on bulked samples of four to five plants, and from older plants rather than young seedlings. The implication of these findings in relation to a virus eradication programme in plant germplasm collections are discussed.  相似文献

Paspalum simplex is a grass distributed throughout the phytogeographic Chaco region in South America from which sexual diploid and apomictic tetraploid races have been reported. We analysed native populations to determine their homogeneity of ploidy level, and the relationship between geographic distribution, ploidy levels, and reproductive systems. The ploidy level was established for 379 plants from 32 wild populations. Tetraploidy and apomixis constitute the most common combination for this species all over the Chaco region. Apomictic hexaploid plants were found associated with 4x populations. Diploids were confined to a small sector of the region. One sexual triploid plant arose from seed harvested in a pure 2x population, and one apomictic 3x plant was found in a mixed 2x-4x population. The results suggest that P. simplex is a core agamic complex characteristic of the Chaco region from which other apomictic polyploid species of the subgenus Anachyris could have evolved. This article integrates studies on the genetic variation of T. urticae populations and the interspecific variation of several tetranychid species. It aims at obtaining insights into the roles of the historical, geographical and ecological factors in the partitioning of variation of species. Two types of molecular markers were used to determine whether the patterns of genetic variation in mites inhabiting different host plants can shed light on the existence of host plant associations. The ribosomal sequences of the second internal transcribed spacer (ITS2), which evolves through concerted evolution, are good indicators of long-term isolation between populations and reveal exceptional homogeneity in a worldwide sampling of T. urticae. The mitochondrial cytochrome oxidase I (COI) sequences do not disclose old divergences related to host plant in this mite but rather suggest recent geographic colonization patterns of the species. Allozyme variation on a fine scale gives some evidence of host associations in the case of citrus trees. However, if any divergence of mites related to this host plant exists, it probably prevails in local populations only and it should not be old enough to be revealed by a phylogenetic analysis of mitochondrial COI sequences. The phyletic constraint in the evolution of feeding specificity in the family Tetra-nychidae is investigated based on a phylogenetic analysis of mitochondrial sequences. The results provide some support for the hypothesis that an evolutionary trend towards polyphagy has occurred in the family. Overall, it seems that the major characteristic of T. urticae is its high colonization potential. Polyphagy has enhanced its successful spread and may have led to connectivity between populations worldwide. © Rapid Science Ltd. 1998  相似文献

Inbreeding and inbreeding depression are important issues in the biology and conservation of natural plant and animal populations, primarily when subpopulation sizes are reduced due to habitat fragmentation. In this study, we propose a method for estimating inbreeding depression in progenies of natural plant populations, combining the estimation of the fixation index by codominant markers with the experimental evaluation of quantitative traits. Our technique estimates apparent inbreeding depression in structured natural populations using the linear regression of phenotypic means on the inbreeding coefficients estimated with codominant markers. This method was applied to data from 112 maternal progenies of 10 natural subpopulations of Eugenia dysenterica DC, a fruiting tree species from the Brazilian savanna (Cerrado). The results show that the proposed method was efficient at detecting the presence of inbreeding depression for seedling emergence and initial growth traits in the species. This corroborates the importance of maintaining high levels of heterozygosity for in situ conservation or genetic restoration of natural populations.  相似文献

The homogeneity of the microbial community structure of a sediment landfill was examined by a culture-independent method and compared with physico-chemical parameters, i.e. organic matter, CaCO3 content, pH, and texture. Total genomic DNA was extracted from samples derived from different places and depths. After amplification with two different primer sets of partial bacterial 16S rRNA genes, the products were separated by denaturing gradient gel electrophoresis (DGGE). The DGGE fingerprints of different sediment samples taken in regular patterns at the same depth were similar, which indicates a spatial homogeneity in the numerically dominant bacterial populations in a landfill over 10,000 m2 in size. In a vertical column of approx. 10 m, only some differences in a few bands of the bacterial community structure were observed between samples taken from different depths. This DNA homogeneity coincided with a similar homogeneity of the physico-chemical parameters in the landfill at this site. Nevertheless, the DGGE technique revealed small differences in less prominent bacteria and was capable of separating the upper and lower samples of one column into two clusters. It therefore seems more sensitive than the physico-chemical approach for characterising the homogeneity of an environmental habitat. The control region of mitochondrial DNA has been widely studied in various human populations. This paper reports sequence data for hypervariable segments 1 and 2 of the control region from a population from southern Tuscany (Italy). The results confirm the high variability of the control region, with 43 different haplotypes in 49 individuals sampled. The comparison of this set of data with other European populations allows the reconstruction of the population history of Tuscany. Independent approaches, such as the estimation of haplotype diversity, mean pairwise differences, genetic distances and discriminant analysis, place the Tuscan sample in an intermediate position between sequences from culturally or geographically isolated regions of Europe (Sardinia, the Basque Country, Britain) and those from the Middle East. In spite of the remarkable genetic homogeneity in Europe, a degree of variability is shown by local European populations and homogeneity increases with the relative isolation of the population. The pattern of mitochondrial variation in Tuscany indicates the persistence of an ancient European component subsequently enriched by migrational waves, possibly from the Middle East. © 1996 Wiley-Liss, Inc.  相似文献

In plant species that are pollinated by a variety of animal species, spatial and temporal variability in the taxonomic composition of those pollinators may cause spatial and temporal variability in selection on floral traits. While temporal variation in pollinator composition has been widely reported, spatial variability, particularly on a local scale, has been studied less frequently; nevertheless, available evidence suggests that local spatial variability may be a widespread feature of plant-pollinator interactions. In addition, the causes of this spatial variability are poorly known. This study was undertaken to quantify variability in the taxonomic composition of pollinators visiting local populations of black mustard, Brassica nigra, and to determine some of the causes of this variability. Simultaneous observations were conducted in seven pairs of black mustard populations. The members of each pair were in close spatial proximity but differed in number of plants. Larger plant populations were visited by significantly greater numbers of honey bees and significantly fewer small bees than small populations on a per-plant basis. There was also a trend toward greater syrphid fly visitation in small populations. The increased numbers of honey bees at large plant populations is probably due to their ability to recruit long distances to the most rewarding plant populations. The lower number of small bees at large plant populations may be due to competition from honey bees and/or spreading a constant number of small bees over the larger number of plants in large populations.This paper is dedicated to the memory of George Eickwort  相似文献

This study is intended to inform the search for a biocontrol solution to stem the spread of Rubus niveus, a morphologically-diverse and invasive Rubus species which has become an aggressive invader in areas of the world outside its native range. The fragile biodiversity of islands can be especially threatened by this plant – exemplars of this are the Galápagos archipelago and Hawaii. This report focuses on the morphological and molecular work carried out to establish the homogeneity or intra-specific variation of R. niveus populations within the Galápagos Islands, as well as their kinship with plants of the same name found in Asia. The premise of this study is that genetic closeness between these populations offers increased probability that pathogens of R. niveus in its native range will also provide effective control in the target environment. Increased knowledge of the genetic make-up of R. niveus in both its native and naturalised locations will also help to assess the feasibility of using organisms which have been found to be effective against other Rubus species, but not hitherto considered for R. niveus itself.  相似文献

Background

Studies of natural animal populations reveal widespread evidence for the diffusion of novel behaviour patterns, and for intra- and inter-population variation in behaviour. However, claims that these are manifestations of animal ‘culture’ remain controversial because alternative explanations to social learning remain difficult to refute. This inability to identify social learning in social settings has also contributed to the failure to test evolutionary hypotheses concerning the social learning strategies that animals deploy.

Methodology/Principal Findings

We present a solution to this problem, in the form of a new means of identifying social learning in animal populations. The method is based on the well-established premise of social learning research, that - when ecological and genetic differences are accounted for - social learning will generate greater homogeneity in behaviour between animals than expected in its absence. Our procedure compares the observed level of homogeneity to a sampling distribution generated utilizing randomization and other procedures, allowing claims of social learning to be evaluated according to consensual standards. We illustrate the method on data from groups of monkeys provided with novel two-option extractive foraging tasks, demonstrating that social learning can indeed be distinguished from unlearned processes and asocial learning, and revealing that the monkeys only employed social learning for the more difficult tasks. The method is further validated against published datasets and through simulation, and exhibits higher statistical power than conventional inferential statistics.

Conclusions/Significance

The method is potentially a significant technological development, which could prove of considerable value in assessing the validity of claims for culturally transmitted behaviour in animal groups. It will also be of value in enabling investigation of the social learning strategies deployed in captive and natural animal populations.  相似文献

Serum samples from two populations of Catalonia, Spain, 208 from Olot (Gerona) and 209 from Tortosa (Tarragona), were typed for G1m (1, 2, 3, 17), G3m (5, 10, 11, 13, 14, 15, 16, 26), and Km (1). The Gm patterns of the Catalonian populations are characterized by the presence of four haplotypes, Gm 1,17;21,26 Gm 1,2,17;21,26 Gm 1,3;5,10,11,13,14,26 and Gm 3;5,10,11,13,14,26. The homogeneity for haplotype Gm 1,17;21,26 among our data and other European populations suggests the existence of an isofrequency line which starts from the Mediterranean zone of Iberian Peninsula and continues through the northwestern part of Europe. From this line a decreasing cline towards the south can be observed. For the haplotype Gm 1,2;17,21,26, affinities are observed between Catalonian populations and other populations from central Europe. This confirms the existence of a gradient towards low values from NW to SE. The presence of the typical Mongoloid haplotype Gm 1,3;5,10,11,13,14,26 is discussed in this paper. No significant differences in the frequencies of the Km1 allele were observed among the European populations.  相似文献

Thlaspi caerulescens is known to hyperaccumulate high quantities of Cd with Cd concentrations up to 3000 mg kg(-1) in some populations from south of France. However, within these populations, the Cd concentrations can vary widely from plant to plant in a way that appears to be not entirely due to variations in soil Cd. The aim of this work was to investigate the variability in the Cd uptake ability of individual plants within a population and among seedlings grown from seeds from a single plant. Ten populations of T. caerulescens plants were selected from four locations (V: Viviez; SF: Saint Félix-de-Pallières; LB: Le Bleymard; CMA: Col du Mas de l'Air) depending of the extent and soil homogeneity of the site. One population from CMA consisted of the progeny of a single maternal plant. Hundred plants of each population were grown for three months in the same homogeneous and lightly Cd-polluted soil (about 20 mg total Cd kg(-1) dry soil). Cadmium uptake behavior of the plants was monitored by labeling the soil with 109Cd. To allow partial plant destruction, radioanalysis was performed on the largest leaf of each plant as an indicator of the total Cd concentration in plant shoots. Results showed significant differences in biomass production and Cd uptake by T. caerulescens between sites and between populations within sites. We observed a wide intra-population variation in biomass, Cd concentration and total Cd uptake. For these properties, 1 to 5 percents of the plants in each population varied by more than a factor of two from the mean values. The mean Cd uptake by the single-plant population from CMA was more than 40% higher than for the population at large. T. caerulescens would respond to traditional selection methods, which would significantly improve the phytoextraction of Cd.  相似文献

QTL mapping experiments in plant breeding may involve multiple populations or pedigrees that are related through their ancestors. These known relationships have often been ignored for the sake of statistical analysis, despite their potential increase in power of mapping. We describe here a Bayesian method for QTL mapping in complex plant populations and reported the results from its application to a (previously analysed) potato data set. This Bayesian method was originally developed for human genetics data, and we have proved that it is useful for complex plant populations as well, based on a sensitivity analysis that was performed here. The method accommodates robustness to complex structures in pedigree data, full flexibility in the estimation of the number of QTL across multiple chromosomes, thereby accounting for uncertainties in the transmission of QTL and marker alleles due to incomplete marker information, and the simultaneous inclusion of non-genetic factors affecting the quantitative trait.  相似文献
