Summary Microtubule (MT) distribution during the cell cycle, especially spindle organization, has been investigated using immunofluorescence light microscopy in cultured cells of two higher plant species, soybean (angiosperm) and black spruce (gymnosperm). In soybean, the prophase and metaphase spindles were different in morphology and structure. The prophase spindle covering the nucleus was barrel-shaped and MTs extended between poles. The metaphase spindle consisted mainly of short MT bundles on either side of the chromosome mass. During prometaphase, the polarity and shape of the prophase spindle disappeared, suggesting that the metaphase spindle is newly formed in prometaphase and not derived from the prophase spindle. A striking feature of MT organization in black spruce was sharply defined poles during prometaphase and anaphase. They were located close to the cell edge, suggesting that a structure in the cytoplasm or associated with the plasma membrane is responsible for their formation. In black spruce the metaphase spindle was long with pointed poles and MT fir tree structures. In contrast, the metaphase spindle of soybean was short with very broad poles and lacked MT fir trees. These results suggest that MT fir tree structure may not be necessary for a functional spindle.  相似文献

Summary Quinacrine, an acridine derivative, has previously been shown to disrupt lateral associations between non-kinetochore microtubules (nkMTs) of opposite polarity in PtK1 metaphase spindles such that the balance of spindle forces is significantly altered. We extended the analysis of the spatial relationship of spindle microtubules (MTs) in this study by using quinacrine to compare ATP-dependent requirements for early prometaphase centrosome separation and spindle formation. The route used for centrosome migration can take a variety of pathways in PtK1 cells, depending on the location of the centrosomes at the time of nuclear envelope breakdown. Following quinacrine treatment centrosome separation decresased by 1.9 to 14.0 m depending on the pathway utilized. However, birefringence of the centrosomal region increased approximately 50% after quinacrine treatment. Quinacrine-treated mid-prometaphase cells, where chromosome attachment to MTs had occurred, showed a decrease in spindle length of approximately 6.0 m with only a slight increase in astral birefringence. Computer-generated reconstructions of quinacrine-treated prometaphase cells were used to confirm changes in MT reorganization. Early-prometaphase cells showed more astral MTs (aMTs) of varied length while mid-prometaphase cells showed only a few short aMTs. Late prometaphase cells again showed a large number of aMTs. Our results suggest that: (1) quinacrine treatment affects centrosome separation, (2) recruitment of nkMTs by kinetochores is quinacrine-sensitive, and (3) development of the prometaphase spindle is dependent on quinacrine-sensitive lateral interactions between nkMTs of opposite polarity. Summary On exposure to high temperature of a temperature-sensitive RNA polymerase subunit (rpoC92) mutant of Escherichia coli, selective reduction was observed in the rate of synthesis of a group of proteins including RNA polymerase subunit. The finding that the synthesis of subunit but not subunit was specifically repressed in this mutant grown at non-permissive temperature indicates that the functionally intact RNA polymerase is required for the synthesis of subunits be coordinated. In addition, the assembly of newly synthesized RNA polymerase subunits was inefficient in this mutant at the steps where altered subunit was involved, and the unassembled enzyme subunits were rapidly and preferentially degraded. During recovery to non-restricted growth, the synthesis of both and subunits was transiently enhanced in parallel leading to recovery of the intracellular concentration of functional RNA polymerase.  相似文献

Summary Mitosis in living cells ofOedogonium observed by time-lapse, was blocked by cytochalasin D (CD; 25–100 g/ml). Normal prometaphase to anaphase takes 10–15 min; blockage of entry into anaphase by CD was reversible up to 2–2.5 h in CD and washout was followed within 10–20 min by normal anaphase and cytokinesis. After 3–6 h in CD, unseparated chromatids segregated randomly into two groups as the spindle slowly elongated considerably, becoming distorted and twisted. During this pseudoanaphase, chromatids sometimes split irregularly and this was stimulated by late washout of CD. CD affected chromosomal attachment to the spindle. If applied at prophase and prometaphase, spindle fibres entered the nucleus; chromosomes moved vigorously and irregularly. A few achieved metaphase only briefly. Treatment at metaphase caused chromosomes to irregularly release and after random movement, all slowly gathered at either pole. Upon removal of CD, chromosomes rapidly achieved metaphase and anaphase A and B soon followed. If CD took effect during anaphase, chromatids detaching from the spindle oscillated rapidly along it; anaphase and cytokinesis (phycoplast formation) were delayed as the cell attempted to correct for abnormal chromosomal behaviour. Summary Movement-induced visual orientation in flies depends largely upon predictable responses which establish simple optomotor balance or complex pseudo search in the appropriate visual environment. Less conspicuous course diverting spontaneous actions of the flies become important in pattern-induced visual orientation. The apparently stochastic spontaneous actions of the houseflyMusca domestica still allow powerful probabilistic predictions of orientation during stationary flight (Reichardt and Poggio 1981). The predominance of non-stochastic spontaneous actions such as body saccades, focussing and shift of visual attention, plasticity of response components etc. in the fruitflyDrosophila melanogaster (Heisenberg and Wolf 1979–1980) accounts for complementary behavioural options which reduce the relevance of probabilistic predictions of orientation in this fly.The conjecture of complementary options is based on a striking antagonism between orientation towards a visual object (fixation), and orientation in the opposite direction (anti-fixation), in the walking fly. Forced choice in a multiple-Y-maze quite definitely elicits fixation in the wild type, and antifixation in the optomotor blind mutantomb H31 (Fig. 3). However, these effects cannot be attributed to a continuous predominance of attraction in the wild type and repellence in the mutant. This is shown under comparable conditions of free choice in an arena: The flies of either strain alternate between fixation and anti-fixation of an inaccessible visual object (Fig. 4a), and keep running to and fro between two of these objects in Buridan's paradigm (Fig. 4b, c), even if the objects are not alike (Fig. 4d). The sequence of approach, retreat and transition may be repeated a few thousand times to the point of exhaustion (Fig. 5). The process resembles the recurrent alternation of ambiguous figures such as the Necker cube in human perception. The recurrent transition between competitive objects counteracts the accumulation of spontaneous preferences, and is likely to explain the apparent lack of pattern-discrimination under operant and non-operant conditions of continued free choice inDrosophila. The conspicuous dichotomy of fixation and anti-fixation in the same environment is, as yet, incompatible with the phenomenological theory of visually controlled orientation in larger flies.Abbreviation S.E.M. standard error of the mean  相似文献

Zusammenfassung Beim ThermometerhuhnLeipoa ocellata tragen die Partner eine Paares ein Rufduett vor. Der Anteil des besteht aus einer Rufreihe, die sich aus einer Folge von 2–7 identischen, zweisilbigen Rufen zusammensetzt. Das trägt einen einzelnen, obertonreichen und langgezogenen Ruf vor. Sowohl der Ruf des als auch die Rufreihe des wird in Serien vorgetragen. Innerhalb einer solcher Ruf- bzw. Rufreihenserie können mehrere Duette auftreten. Die Rufe sind jedoch nicht ausschließlich an das Duett gebunden. Die Variabilität im Aufbau des Duetts äußert sich im Zeitpunkt des Einsatzes des antwortenden Vogels, in der Anzahl der -Rufe während des Duetts und in der Anzahl der Einheiten, aus denen sich der Duettanteil des zusammensetzt. Das beginnt signifikant häufiger als das eine Serie, in der ein oder mehrere Duette vorkommen. Ebenso ist es häufiger der Initiator des ersten in dieser Serie liegenden Duetts. Das Duett dient wohl hauptsächlich zur Festigung des Zusammenhalts zwischen den Paarpartnern. Es erfüllt jedoch von seinen physikalischen Eigenschaften her auch die Bedingungen, die für ein territorial wirksames Signal gelten.

Summary The distribution and ultrastructure of serotonin- and dopamine-immunoreactive (5-HTi and DAi) neurones have been investigated in the terminal ganglion of the cricket, Acheta domestica, using a pre-embedding chopper technique. Special attention has been paid to the immunoreactive structures in the neuropil. 5-HTi structures are extensively distributed and densely packed throughout the 5 neuromeres of the terminal ganglion and originate from several interneurones and efferent neurones. In contrast, DAi fibres are distributed sparsely although they extend to all neuromeres of the ganglion and originate from 6 interneurons only. For both 5-HTi and DAi neurones characteristic axonal projections and branching patterns can be distinguished. The 5-HTi axons exhibit rich varicose arborizations, whereas DAi neurones possess fewer varicosities in the neuropil. Electron microscopy shows that 5-HTi varicosities contain small ( 60 nm) and large ( 100 nm) agranular vesicles, and large ( 100 nm) granular vesicles, whereas in DAi varicosities small ( 60 nm) agranular and large ( 100 nm) granular vesicles are seen. Both 5-HTi and DAi varicosities form synaptic contacts. Summary The osmotic potentials of needles were compared from numerous trees that had been classified according to needle loss along an altitude profile. With the increasing degree of damage of the trees, the maximum and minimum values deviated more strongly above and below the common mean of all samples. The level of water content of the needles unequivocally reflected the vitality of the trees. Experiments covering a whole vegetation period were performed on a tree pair selected from a natural stand. They were designed to demonstrate differences in water balance between the healthy and damaged state of the trees. For the damaged tree, measurements of the water potentials of single needles showed a greater reduction of potential during the course of the day compared to the healthy tree. Recovery in the evenings was slower and often incomplete. The osmotic potentials of damaged and healthy shoots measured in individual needles also differed both predawn and especially after transpiration stress. Depending on weather and soil desiccation, the differences were more or less pronounced. In the damaged tree, the rises in potential after saturation of the twigs fell far short of the healthy tree. Reduction of water potential, osmotic potential and relative water content under comparable stress conditions suggest a reduced tolerance of drought by damaged trees. In the context of earlier experiments this result was ascribed to a lack of stomatal control, and long-term pollution effects were thus explained as a specific disturbance of hydroregulation.  相似文献

Summary Using immunocytochemistry (ICC), the number of immunoreaction products (IRPs) visualized for the peptide substance P (SP) appears reduced within the human spinal cord (also substantia nigra) taken at autopsy from cases diagnosed with Huntington's disease (HD) compared with the non-HD cases (Vacca 1983). The reductions of SR-IRPs become apparent in the HD specimens when primary anti-SP serum is applied to the tissue sections at supra-optimal dilutions; that is, dilutions greater than the optimal dilutions which visualize maximal numbers of SP-IRPs and concomitantly give maximal staining intensity. Curiously, the application of optimal dilutions to the HD and non-HD specimens visualizes equivalent numbers of SP-IRPs; therefore, the qualitative and quantitative differences between the specimens become masked. Applying supra-optimal dilutions of the anti-SP serum unmasks the difference, and also reveals different end-points for the immunostain deposited in each type of specimen. In both the HD and non-HD specimens, two sizes of SP-IRPs could be identified, large (3 m) and small (0.7 m). Presumably they mark two different categories of axons defined by caliber (e.g.; C-type and A ), or origin (e.g., sensory intrinsic, or supraspinal). Alternatively the large SP-IRPs label tangentially-cut or large axons and nerve terminals. In the present report, counts of the large and small SP-IRPs visualized in the HD and the non-HD specimens have been plotted against the serial dilutions (optimal, supra-optimal and endpoint) of the primary anti-SP serum used for ICC. The graphs which result, describe a titration curve characteristic for the large SP-IRPs in each specimen. Applying supra-optimal dilutions of the primary antibody, causes the numbers of large and small SP-IRPs to decrease differentially. The numerical decrease probably largely reflects the concentration of immunoreactive antigen in different anatomic or metabolic tissue compartments. Alternatively, or perhaps to a lesser degree, the small and large SP-IRPs represent cross-reacting components which have different staining end-points, and may have biological significance in HD. Criteria are given for the proper use of ICC, and have significance regarding the use of commercial kits especially for pathological diagnosis.  相似文献

Summary The birefrigence of protoplast during mitosis in staminal hair cells ofTradescantia virginica was investigatedin vivo. The following results were obtained:Under given conditions, no birefringence could be observed in the clear zone (prophase), the chromosome fibers of the spindle, and the chromosomes themselves.The phragmoplast exhibits relatively strong birefringence during the early telophase.The continuous fibers of the spindle are clearly visiblein vivo. These form a portion of the phragmoplast.Neither cell plate nor middle lamella shows any birefringence. The primary wall exhibits birefringence which increases rapidly during the later stages of telophase.

Species dynamics of fungi (filamentous fungi and yeasts) on apple leaves were studied within the framework of the theory of island biogeography by following immigration and extinction patterns on individual apple leaf islands over time. Total fungi were censused on unmanipulated leaves collected throughout two seasons; filamentous fungi only were monitored additionally for several weeks in one season on newly created, axenic, model (seedling) islands introduced to the orchard, and on surface-sterilized, preexisting leaves. Analyses based on both the natural and the surface-sterilized systems showed that an equilibrium in species number was reached and turnover in species composition occurred in both. Immigration and extinction events were strongly related to number of species present on each island. The balance between immigration and extinction implies that species number on leaves and real (oceanic) islands is determined by a common mechanism, and emphasizes the need to regard leaf microbial communities as dynamic.  相似文献

Chloroplast DNA restriction fragment length polymorphisms have been used to reconstruct the maternal phylogeny of all the known taxa in the small neotropical legume genusLeucaena. Three major plastome clades were recognized, but these did not conform with relationships between the taxa proposed on other characters from morphology, cytology or hybridization. The maternal parentage of tetraploids within the genus has been proposed. Evidence for introgression was found between diploidL. diversifolia and tetraploidL. diversifolia. The implications of these results for the origin of the cultivated taxa are discussed.  相似文献

Activities and properties of some enzymes of gluconeogenesis, the malic enzyme, phosphoenolpyruvate carboxykinase, glucoso-6-phosphatase, alanine aminotransferase, and aspartate aminotransferase, were studied in turbellarians Phagocata sibirica and trematodes Eurytrema pancreaticum. It was shown that adaptation of trematodes to the parasitic mode of life affected essentially the biosynthetic processes and led to formation of different metabolic pathways.  相似文献

Summary Variable numbers of bivalents and sex chromosomes do not attach to the spindle when prophase or early prometaphase cranefly spermatocytes (2n=8) are treated with cytochalasin D or latrunculin. The unattached bivalents lie in the cytoplasm or at the spindle pole, and they do not delay onset of autosomal anaphase; sometimes they disjoin at the same time as the attached bivalents, so they respond to the global signals that initiate anaphase. Unattached sex chromosomes do not delay autosomal anaphase, either. Of various interpretations of these data, we think the best explanation is that the checkpoint system responds to physical rather than chemical cues; we think that the spindle is a tensegral structure, that chromosomes need to interact with the spindle in order to be recognised by the anaphase-onset checkpoint control, and that the physical interaction of chromosomes with spindle acts as a signalling network. Cytochalasin D and latrunculin treatments delay onset of sex chromosome anaphase (which normally occurs about 15 min after autosomal anaphase) and cause altered patterns of sex-chromosome segregation.  相似文献

Transformation ofEscherichia coli strains with the recombinant plasmid, prepared by shot gun cloning with pBR322 and containing the geneasd, the structural gene of aspartate--semialdehyde dehydrogenase, results in an increase in specific activity of 65-fold of the enzyme in crude extracts. Approximately 60 mg of pure enzyme may be obtained from 10 g of transformed cells (wet weight) in a simplified purification procedure. The molecular weight, amino acid composition, and kinetic properties of the enzyme appear to be the same as previously reported, and the first 36 amino acids of the N-terminal sequence have been determined.  相似文献

The organization of the microtubule (Mt) cytoskeleton during mitosis and cytokinesis of the generative cell (GC) in Ornithogalum virens L. (bicellular pollen type, chromosome number, n = 3) from prophase to telophase/sperm formation was investigated by localization of -tubulin immunofluorescence using a conventional fluorescence microscope and a confocal laser scanning microscope. Chromosomes were visualized with DNA-binding fluorochrome dyes (ethidium bromide and 46-diamino-2-phenyl-indole). The GC of O. virens is characterized by G2/M transition within the pollen grain and not in the pollen tube as occurs in the majority of species with bicellular pollen. It was found that prophase in the GC starts before anthesis and prometaphase takes place after 10 min of pollen germination. The prophase Mts are organized into three prominent bundles, located near the generative nucleus. The number of these Mt bundles is the same as the number of GC chromosomes, a relation which has not previously been considered in other species. The most evident feature in the prophase/ prometaphase transition of O. virens GC is a direct rapid rearrangement of Mt bundles into a network which appears to interact with kinetochores and form a typical prometaphase Mt organization. The metaphase chromosomes are arranged into a conventional equatorial plate, and not in tandem as is thought to be characteristic of GC metaphase. The metaphase spindle consists of kinetochore fibres and a few interzonal fibres which form dispersed poles. Anaphase is characterized by a significant elongation of the mitotic spindle concomitant with the extension of the distance between the opposite poles. At anaphase the diffuse poles converge. Cytokinesis is realized by cell plate formation in the equatorial plane of the GC. The formation of kinetochore (chromosomal) and continuous fibers, and the behavior of the nuclear envelope (NE) was described in studies combining light and electron microscopy. Microtubules (MTs) push and pull the NE which becomes progressively weaker before breaking. It breaks to a certain extent due to mechanical pressure. Clear zone MTs penetrate into the nuclear area as dense bundles and form continuous fibers. These MTs also attach to some kinetochores during this process. Some kinetochore fibers seem to be formed by the kinetochores themselves which are also responsible for further development and changes of kinetochore fibers. Formation of kinetochore fibers is asynchronous for different chromosomes and even for two sister kinetochores. Often temporary faulty connections between different kinetochores or the polar regions are formed which usually break in later stages. This results in movements of chromosomes toward the poles and across the spindle during prometaphase. The NE, whose fine structure has been described, breaks into small pieces which often persist to the next mitosis. Old pieces of NE are utilized in the formation of new NE at telophase. Several problems concerning the mechanism of chromosome movements, visibility of the NE, etc., have also been discussed.  相似文献

Summary Amphicarpum purshii Kunth is an annual amphicarpic grass of recently disturbed sandy areas in the Coastal Plain of eastern North America, producing small aerial and larger subterranean seeds. At study sites on the eastern edge of the Pine Barrens of New Jersey, the temporal aspects of reproductive allocation were investigated through biweekly whole-plant field harvests of 25 plants each throughout the 1980 growing season, and the survivorship of plants arising from aerial and subterranean seeds was monitored in a total of 10 high and 10 low density quadrats at two sites differeing in soil moisture. Plants arising from subterranean propagules began to allocate early (July 16) considerable energy to subterranean reproduction (ca 40% of whole-plant dry weight by the last half of August); allocation to aerial culms and flowers did not begin until mid-August and never exceeded 3% on a population basis. Plants arising from aerial propagules (aerial plants) produced only subterranean flowers, and these flowers first appeared 2 wk later than they did on subterranean plants. Survivorship was greater for the subterranean seedlings at both the dry and wet sites and at low and high densities, and aerial plants showed significantly less total growth and seed production than subterranean plants. The pessimistic strategy (early allocation of energy to large subterranean propagules) in the annual Amphicarpum has its selective basis in the relative vigor, survival, and timing and amount of reproduction of the two types of seedlings, and appears comparable to the allocation strategies of at least eight other amphicarpic annuals in at least five other families of plants.  相似文献

In legumes, ENOD40 expression is increased upon interaction of plants with rhizobia. Little is known of the expression pattern of ENOD40 during other stages of the plant life cycle. Studies of ENOD40 expression in non-legume development may give an indication of the function of the gene. To investigate the ENOD40 expression pattern during plant development, a fusion between the -glucuronidase (GUS) reporter gene and 150 bp of the 5 untranslated region plus 3,000 bp of 5 untranscribed tomato ENOD40 sequence was constructed and introduced into Lycopersicon esculentum Miller.
