The rate of net CO2 assimilation (A), the stomatal (gs) and residual (gr) conductances to CO2, the intercellular CO2 concentration, the CO2 compensation points at 21% O221) and at 2% O22), and the amounts of dry matter, nitrogen, and carbohydrates were determined, from anthesis through grain filling, in the flag leaf blade and sheath of spring wheat (Triticum aestivum L. cv Kolibri). The nitrogen content and the rate of net CO2 assimilation declined slowly until the onset of senescence in both organs, about 3 weeks after anthesis. During senescence the reduction of A in both organs was not primarily caused by a decrease in gs; the main factor is the decrease in gr. From values of Γ21 and Γ2 it is suggested that the rate of respiration in the light contributing to the CO2 compensation point is higher in sheaths than in blades irrespective of the O2 level considered. The role of sheaths storing and later transporting assimilates to the developing grains seems to be more important for shoot yield than that of sheaths functioning as photosynthetic organs after the onset of senescence occurs. It is suggested that accumulation of carbohydrates in leaves might somehow trigger senescence in the flag leaf blade and sheath simultaneously.  相似文献

Constructs containing the cDNAs encoding the primary leaf catalase in Nicotiana or subunit 1 of cottonseed (Gossypium hirsutum) catalase were introduced in the sense and antisense orientation into the Nicotiana tabacum genome. The N. tabacum leaf cDNA specifically overexpressed CAT-1, the high catalytic form, activity. Antisense constructs reduced leaf catalase specific activities from 0.20 to 0.75 times those of wild type (WT), and overexpression constructs increased catalase specific activities from 1.25 to more than 2.0 times those of WT. The NADH-hydroxypyruvate reductase specific activity in transgenic plants was similar to that in WT. The effect of antisense constructs on photorespiration was studied in transgenic plants by measuring the CO2 compensation point (Γ) at a leaf temperature of 38°C. A significant linear increase was observed in Γ with decreasing catalase (at 50% lower catalase activity Γ increased 39%). There was a significant temperature-dependent linear decrease in Γ in transgenic leaves with elevated catalase compared with WT leaves (at 50% higher catalase Γ decreased 17%). At 29°C, Γ also decreased with increasing catalase in transgenic leaves compared with WT leaves, but the trend was not statistically significant. Rates of dark respiration were the same in WT and transgenic leaves. Thus, photorespiratory losses of CO2 were significantly reduced with increasing catalase activities at 38°C, indicating that the stoichiometry of photorespiratory CO2 formation per glycolate oxidized normally increases at higher temperatures because of enhanced peroxidation.  相似文献

The effect of photosynthetic photon flux density (PPFD) on photosynthetic response (A) to CO2 partial pressures between 35 pascals and CO2 compensation point (Γ) was investigated, especially below PPFD saturation. Spinacia oleracea cv `Atlanta,' Glycine max cv `Clark,' and Arbutus unedo were studied in detail. The initial slope of the photosynthetic response to CO2 (A/C[Γ]) was constant above a PPFD of about 500 to 600 micromoles per square meter per second for all three species; but declined rapidly with PPFD below this critical level. For Γ there was also a critical PPFD (approximately 200 micromoles per square meter per second for S. oleracea and G. max; 100 for A. unedo) above which Γ was essentially constant, but below which Γ increased with decreasing PPFD. All three species showed a dependence of A/C(Γ) on PPFD at low PPFD. Simulated photosynthetic responses obtained with a biochemically based model of whole-leaf photosynthesis were similar to measured responses.  相似文献

Chloroplast-targeted overexpression of an Fe superoxide dismutase (SOD) from Arabidopsis thaliana resulted in substantially increased foliar SOD activities. Ascorbate peroxidase, glutathione reductase, and monodehydroascorbate reductase activities were similar in the leaves from all of the lines, but dehydroascorbate reductase activity was increased in the leaves of the FeSOD transformants relative to untransformed controls. Foliar H2O2, ascorbate, and glutathione contents were comparable in all lines of plants. Irradiance-dependent changes in net CO2 assimilation and chlorophyll a fluorescence quenching parameters were similar in all lines both in air (21% O2) and at low (1%) O2. CO2-response curves for photosynthesis showed similar net CO2-exchange characteristics in all lines. In contrast, values of photochemical quenching declined in leaves from untransformed controls at intercellular CO2 (Ci) values below 200 μL L−1 but remained constant with decreasing Ci in leaves of FeSOD transformants. When the O2 concentration was decreased from 21 to 1%, the effect of FeSOD overexpression on photochemical quenching at limiting Ci was abolished. At high light (1000 μmol m−2 s−1) a progressive decrease in the ratio of variable (Fv) to maximal (Fm) fluorescence was observed with decreasing temperature. At 6oC the high-light-induced decrease in the Fv/Fm ratio was partially prevented by low O2 but values were comparable in all lines. Methyl viologen caused decreased Fv/Fm ratios, but this was less marked in the FeSOD transformants than in the untransformed controls. These observations suggest that the rate of superoxide dismutation limits flux through the Mehler-peroxidase cycle in certain conditions.  相似文献

The effect of leaf temperature on the post-illumination burst of CO2 (PIB) in 15 day-old primary bean leaves (Phaseolus vulgaris L.) was studied by means of infrared gas analysis in a closed gas exchange system. The amplitude and kinetic of PIB was different with its own characteristic course for different temperature steps. The temperature optimum for the PIB at 21% O2 near the carbon dioxide compensation concentration Λ (150 mg m-3) was 33 °C while that for net photosynthetic rate (P N) at 21% O2 and 600 mg m-3 CO2 was 24.5 °C. The PIB was observed to 12…33 s after the darkening of leaves. No PIB was observed at 2% O2. The applicability of PIB as an estimate of photorespiration rateR p has been proved by comparing it with extrapolation of the relationship betweenP n and CO2 concentration to zero.  相似文献

Effect of phosphorus deficiency on photosynthetic and respiratory CO2 exchanges were analysed in primary leaves of 2-week-old bean (Phaseolus vulgaris L. cv. Golden Saxa) plants under non-photorespiratory (2 % O2) and photorespiratory (21 % O2) conditions. Low P decreased maximum net photosynthetic rate (PNmax) and increased the time necessary to reach it. In the leaves of P-deficient plants the relative decrease of PNmax at 2 % O2 was larger than at 21 % O2. The results suggested the influence of photorespiration in the cellular turnover of phosphates.  相似文献

The relative composition of stable carbon isotopes, δ13C, was determined in flag leaves and grain of spring wheat (Triticum aestivum L. cv Albis) grown in open-top field fumigation chambers and exposed to different O3 levels during the growing season. The aim of the study was to establish exposure-response relationships for the radiation-weighted seasonal mean O3 concentration and δ13C (relative deviation of the 13C/12C ratio) values of the two plant parts. Samples were collected at harvest in 1986, 1987, and 1988. With increasing O3 concentration, δ13C values increased (became less negative) proportionally. Year to year δ13C differences at equivalent O3 concentrations were small. The shift in δ13C caused by O3 was more pronounced in grain than in leaves. According to models of 13C discrimination in C3 plants, these results indicate increasing limitation of photosynthesis by CO2 diffusion relative to limitation by carboxylation with increasing O3 exposure. This conclusion is not in agreement with results from gas exchange analysis. Water use efficiency in green flag leaves tended to decrease with increasing O3, indicating a dominating effect of O3 on CO2 carboxylation.  相似文献

The rate of net CO2 assimilation of mature wheat (Triticum aestivum L.) leaves in ambient air (21% O2, 340 microbars CO2) declined with time of illumination at temperatures lower than 25°C, but not at higher temperatures, and the rate of decline increased when maintained in air with higher CO2 concentration (700-825 microbars). In this latter case, the decline in the rate of net CO2 assimilation also occurred at high temperatures. Stomatal conductance also declined with time in some cases and stomata became more sensitive to CO2, but this was not the primary cause of the decrease in CO2 assimilation because internal partial pressure of CO2 remained constant. Treatments which reduced the rate of translocation (e.g. lower temperatures, chilling the base of the leaf) produced a marked decline in CO2 assimilation of leaves in atmospheric and high CO2 concentrations. The decreased net CO2 assimilation was correlated with carbohydrate accumulation in each case, suggesting end product inhibition of photosynthesis. Analysis of CO2 assimilation in high carbohydrate leaves as a function of intercellular CO2 partial pressure showed reduction in the upper part of the curve. The initial slope of this curve, however, was not affected. Photosynthetic rates in the upper part of this curve generally recovered after a short period in darkness in which carbohydrates were removed from the leaf. The stimulation of net CO2 assimilation by 2% O2 (Warburg effect), and the apparent quantum yield, decreased after several hours of light.  相似文献

The rate of dark CO2 efflux from mature wheat (Triticum aestivum cv Gabo) leaves at the end of the night is less than that found after a period of photosynthesis. After photosynthesis, the dark CO2 efflux shows complex dependence on time and temperature. For about 30 minutes after darkening, CO2 efflux includes a large component which can be abolished by transferring illuminated leaves to 3% O2 and 330 microbar CO2 before darkening. After 30 minutes of darkness, a relatively steady rate of CO2 efflux was obtained. The temperature dependence of steady-state dark CO2 efflux at the end of the night differs from that after a period of photosynthesis. The higher rate of dark CO2 efflux following photosynthesis is correlated with accumulated net CO2 assimilation and with an increase in several carbohydrate fractions in the leaf. It is also correlated with an increase in the CO2 compensation point in 21% O2, and an increase in the light compensation point. The interactions between CO2 efflux from carbohydrate oxidation and photorespiration are discussed. It is concluded that the rate of CO2 efflux by respiration is comparable in darkened and illuminated wheat leaves.  相似文献

The growth kinetics of spinach plants (Spinacia oleracea L. cv Savoy) grown at 5°C or 16°C were determined to allow us to compare leaf tissues of the same developmental stage rather than chronological age. The second leaf pairs reached full expansion at a plant age of 32 and 92 days for the 16°C and 5°C plants, respectively. Growth at 5°C resulted in an increased leaf area, dry weight, dry weight per area, and leaf thickness. Despite these changes, pigment content and composition, room temperature in vivo fluorescence, and apparent quantum yield and light-saturated rates of CO2 exchange or O2 evolution were not affected by the growth temperature. Furthermore, 5°C expanded leaves were found to be more resistant to photoinhibition at 5°C than were 16°C expanded leaves. Thus, it is concluded that spinach grown at low temperature is not stressed. However, shifting spinach leaves from 5°C to 16°C or from 16°C to 5°C for 12 days after full leaf expansion had occurred resulted in a 20 to 25% reduction in apparent quantum yields and 50 to 60% reduction in light saturated rates of both CO2 exchange and O2 evolution. This was not accompanied by a change in the pigment content or composition or in the room temperature in vivo fluorescence. It appears that leaf aging during the temperature shift period can account for the reduction in photosynthesis. Comparison of cold-hardened and non-hardened winter rye (Secale cereale L. cv Muskateer) with spinach by in vivo fluorescence indicated that rye is more sensitive to both short term and longer duration temperature shifts than is spinach. Thus, susceptibility to an abrupt temperature shift appears to be species dependent.  相似文献

The action spectrum of apparent photosynthesis for attached radish (Raphanus sativus L. var. Early Scarlet Globe) and corn (Zea mays L. var. Pride V.) leaves was measured at 300 μl/l CO2 and both 21% and 2% O2. The spectra were measured at light intensities where apparent photosynthesis was proportional to intensity. For radish, a high compensation point plant, oxygen had an inhibiting effect on photosynthesis at all wavelengths from 402 to 694 mμ. If a constant rate of photosynthesis at 21% O2 for the different wavelengths was chosen, then the percent increase in net CO2 fixation at 2% O2 was constant. For corn, a low compensation point plant, no inhibitory effect of oxygen concentration from 2% to 21% O2 was found over the visible spectrum. The CO2 compensation point for light intensities greater than the light compensation point was found to be constant and independent of wavelength for both radish and corn leaves. For radish, the lowering of the oxygen concentration from 21% to 2% at these intensities was found to reduce the CO2 compensation point by the same amount for the wavelengths studied.  相似文献

The effects of salinity on growth, stomatal conductance, photosynthetic capacity, and carbon isotope discrimination (Δ) of Gossypium hirsutum L. and Phaseolus vulgaris L. were evaluated. Plants were grown at different NaCl concentrations from 10 days old until mature reproductive structures were formed. Plant growth and leaf area development were strongly reduced by salinity, in both cotton and bean. Stomatal conductance also was reduced by salinity. The Δ always declined with increasing external salinity concentration, indicating that stomatal limitation of photosynthesis was increased. In cotton plant dry matter, Δ correlated with the ratio of intercellular to atmospheric CO2 partial pressures (pl/pa) calculated by gas exchange. This correlation was not clear in bean plants, although Δ showed a more pronounced salt induced decline in bean than in cotton. Possible effects of heterogeneity of stomatal aperture and consequent overestimation of pl as determined from gas exchange could explain these results. Significant differences of Δ between leaf and seed material were observed in cotton and bean. This suggests different patterns of carbon allocation between leaves and seeds. The photon yield of O2 evolution determined at rate-limiting photosynthetic photon flux density was insensitive to salinity in both species analyzed. The light- and CO2-saturated rate of CO2 uptake and O2 evolution showed a salt induced decline in both species. Possible explanations of this observation are discussed. O2 hypersensitivity was observed in salt stressed cotton plants. These results clearly demonstrate that the effect of salinity on assimilation rate was mostly due to the reduction of stomatal conductance, and that calculation of pl may be overestimated in salt stressed plants, because of heterogeneity of stomatal aperture over the leaf surface.  相似文献

The net photosynthetic rate (P N), the sample room CO2 concentration (CO2S) and the intercellular CO2 concentration (C i) in response to PAR, of C3 (wheat and bean) and C4 (maize and three-colored amaranth) plants were measured. Results showed that photorespiration (R p) of wheat and bean could not occur at 2 % O2. At 2 % O2 and 0 μmol mol?1 CO2, P N can be used to estimate the rate of mitochondrial respiration in the light (R d). The R d decreased with increasing PAR, and ranged between 3.20 and 2.09 μmol CO2 m?2 s?1 in wheat. The trend was similar for bean (between 2.95 and 1.70 μmol CO2 m?2 s?1), maize (between 2.27 and 0.62 μmol CO2 m?2 s?1) and three-colored amaranth (between 1.37 and 0.49 μmol CO2 m?2 s?1). The widely observed phenomenon of R d being lower than R n can be attributed to refixation, rather than light inhibition. For all plants tested, CO2 recovery rates increased with increasing light intensity from 32 to 55 % (wheat), 29 to 59 % (bean), 54 to 87 % (maize) and 72 to 90 % (three-colored amaranth) at 50 and 2,000 μmol m?2 s?1, respectively.  相似文献

The effect of leaf temperature on stomatal conductance and net CO2 uptake was studied on French bean (Phaseolus vulgaris L.) using either dehydrated attached leaves (25–40% water deficit) or cut leaves supplied with 10–4 M abscisic acid (ABA) solution to the transpiration stream. Decreasing leaf temperature caused stomatal opening and increased net CO2 uptake (which was close to zero at around 25° C) to a level identical to that of control leaves (without water deficit) at around 15° C. (i) The ABA effect on stomatal closure was modulated by temperature and, presumably, ABA is at least partly responsible for stomatal closure of french bean submitted to a drought stress. (ii) For leaf temperatures lower than 15° C, net CO2 uptake was no longer limited by water deficit even on very dehydrated leaves. This shows that dehydrated leaves retain a substantial part of their photosynthetic capacity which can be revealed at normal CO2 concentrations when stomata open at low temperature. In contrast to leaves fed with ABA, decreasing the O2 concentration from 21% to 1% O2 did not increase either the rate of net CO2 uptake or the thermal optimum for photosynthesis of dehydrated leaves. The quantum yield of PSII electron flow (measured by F/Fm) was lower in 1% O2 than in 21% O2 for each leaf pretreatment given (non-dehydrated leaves, dehydrated leaves, and leaves fed with ABA) even within a temperature range in which leaf photosynthesis at normal CO2 concentration was the same in these two O2 concentrations. Potato plants (Solanum tuberosum L. cv. Bintje) were grown to maturity in open-top chambers under three carbon dioxide (CO2; ambient and 24 h d−1 seasonal mean concentrations of 550 and 680 μmol mol−1) and two ozone levels (O3; ambient and an 8 h d−1 seasonal mean of 50 nmol mol−1). Chlorophyll content, photosynthetic characteristics, and stomatal responses were determined to test the hypothesis that elevated atmospheric CO2 may alleviate the damaging influence of O3 by reducing uptake by the leaves. Elevated O3 had no detectable effect on photosynthetic characteristics, leaf conductance, or chlorophyll content, but did reduce SPAD values for leaf 15, the youngest leaf examined. Elevated CO2 also reduced SPAD values for leaf 15, but not for older leaves; destructive analysis confirmed that chlorophyll content was decreased. Leaf conductance was generally reduced by elevated CO2, and declined with time in the youngest leaves examined, as did assimilation rate (A). A generally increased under elevated CO2, particularly in the older leaves during the latter stages of the season, thereby increasing instantaneous transpiration efficiency. Exposure to elevated CO2 and/or O3 had no detectable effect on dark-adapted fluorescence, although the values decreased with time. Analysis of the relationships between assimilation rate and intercellular CO2 concentration and photosynthetically active photon flux density showed there was initially little treatment effect on CO2-saturated assimilation rates for leaf 15. However, the values for plants grown under 550 μmol mol−1 CO2 were subsequently greater than in the ambient and 680 μmol mol−1 treatments, although the beneficial influence of the former treatment declined sharply towards the end of the season. Light-saturated assimilation was consistently greater under elevated CO2, but decreased with time in all treatments. The values decreased sharply when leaves grown under elevated CO2 were measured under ambient CO2, but increased when leaves grown under ambient CO2 were examined under elevated CO2. The results obtained indicate that, although elevated CO2 initially increased assimilation and growth, these beneficial effects were not necessarily sustained to maturity as a result of photosynthetic acclimation and the induction of earlier senescence.  相似文献

The extent and occurrence of water stress-induced “patchy” CO2 uptake across the surface of leaves was evaluated in a number of plant species. Leaves, while still attached to a plant, were illuminated and exposed to air containing [14C]CO2 before autoradiographs were developed. Plant water deficits that caused leaf water potential depression to −1.1 megapascals during a 4-day period did result in heterogenous CO2 assimilation patterns in bean (Phaseolus vulgaris). However, when the same level of stress was imposed more gradually (during 17 days), no patchy stomatal closure was evident. The patchy CO2 assimilation pattern that occurs when bean plants are subjected to a rapidly imposed stress could induce artifacts in gas exchange studies such that an effect of stress on chloroplast metabolism is incorrectly deduced. This problem was characterized by examining the relationship between photosynthesis and internal [CO2] in stressed bean leaves. When extent of heterogenous CO2 uptake was estimated and accounted for, there appeared to be little difference in this relationship between control and stressed leaves. Subjecting spinach (Spinacea oleracea) plants to stress (leaf water potential depression to −1.5 megapascals) did not appear to cause patchy stomatal closure. Wheat (Triticum aestivum) plants also showed homogenous CO2 assimilation patterns when stressed to a leaf water potential of −2.6 megapascals. It was concluded that water stress-induced patchy stomatal closure can occur to an extent that could influence the analysis of gas exchange studies. However, this phenomenon was not found to be a general response. Not all stress regimens will induce patchiness; nor will all plant species demonstrate this response to water deficits.  相似文献

Excised green leaves of mung bean (Phaseolus aureus L. var. Mungo) were used to determine the effect of light on the rate of endogenous respiration via the tricarboxylic acid cycle. Illumination with white light at an intensity of 0.043 gram calories cm−2min−1 (approximately 8600 lux) of visible radiation (400-700 nm) gave a rate of apparent photosynthesis, measured as net CO2 uptake, of 21 mg CO2 dm−2hr−1 which was about 11-fold greater than the rate of dark respiration. The feeding of 14CO2 or 14C-labeled acids of the tricarboxylic acid cycle in the dark for 2 hours was established as a suitable method for labeling mitochondrial pools of cycle intermediates.  相似文献

Observations of nonuniform photosynthesis across leaves cast doubt on internal CO2 partial pressures (pi) calculated on the assumption of uniformity and can lead to incorrect conclusions about the stomatal control of photosynthesis. The problem can be avoided by measuring pi directly because the assumptions of uniformity are not necessary. We therefore developed a method that allowed pi to be measured continuously in situ for days at a time under growth conditions and used it to investigate intact leaves of sunflower (Helianthus annuus L.), soybean (Glycine max L. Merr.), and bush bean (Phaseolus vulgaris L.) subjected to high or low leaf water potentials (ψw) or high concentrations of abscisic acid (ABA). The leaves maintained a relatively constant differential (Δp) between ambient CO2 and measured pi throughout the light period when water was supplied. When water was withheld, ψw decreased and the stomata began to close, but measured pi increased until the leaf reached a ψw of −1.76 (bush bean), −2.12 (sunflower) or −3.10 (soybean) megapascals, at which point Δp = 0. The increasing pi indicated that stomata did not inhibit CO2 uptake and a Δp of zero indicated that CO2 uptake became zero despite the high availability of CO2 inside the leaf. In contrast, when sunflower leaves at high ψw were treated with ABA, pi did not increase and instead decreased rapidly and steadily for up to 8 hours even as ψw increased, as expected if ABA treatment primarily affected stomatal conductance. The accumulating CO2 at low ψw and contrasting response to ABA indicates that photosynthetic biochemistry limited photosynthesis at low ψw but not at high ABA.  相似文献

Two naturally occurring species of the genus Alternanthera, namely A. ficoides and A. tenella, were identified as C3-C4 intermediates based on leaf anatomy, photosynthetic CO2 compensation point (Γ), O2 response of г, light intensity response of г, and the activities of key enzymes of photosynthesis. A. ficoides and A. tenella exhibited a less distinct Kranz-like leaf anatomy with substantial accumulation of starch both in mesophyll and bundle sheath cells. Photosynthetic CO2 compensation points of these two intermediate species at 29°C were much lower than in C3 plants and ranged from 18 to 22 microliters per liter. Although A. ficoides and A. tenella exhibited similar intermediacy in г, the apparent photorespiratory component of O2 inhibition in A. ficoides is lower than in A. tenella. The г progressively decreases from 35 microliters per liter at lowest light intensity to 18 microliters per liter at highest light intensity in A. tenella. It was, however, constant in A. ficoides at 20 to 25 microliters per liter between light intensities measured. The rates of net photosynthesis at 21% O2 and 29°C by A. ficoides and A. tenella were 25 to 28 milligrams CO2 per square decimeter per hour which are intermediate between values obtained for Tridax procumbens and A. pungens, C3 and C4 species, respectively. The activities of key enzymes of C4 photosynthesis, phosphoenolpyruvate carboxylase, pyruvate Pi dikinase, NAD malic enzyme, NADP malic enzyme and phosphoenolpyruvate carboxykinase in the two intermediates, A. ficoides and A. tenella are very low or insignificant. Results indicated that the relatively low apparent photorespiratory component in these two species is presumably the basis for the C3-C4 intermediate photosynthesis.  相似文献

When leaflets of bean and leaves of other species of C3 plants are illuminated in the absence of CO2 and at low O2 partial pressure, the capacity for CO2 assimilation at saturating light and its efficiency at low light intensities are inhibited. This photoinhibition is dependent on leaflet age and period of illumination. In young leaflets and following short exposure to these photoinhibitory conditions, some recovery of CO2 assimilation capacity is observed immediately after treatment. Following substantial (70 to 80%) photoinhibition of CO2 assimilation, recovery in fully expanded leaflets is observed only after 48 hours in normal air. The photoinhibition is largely prevented by providing CO2 at partial pressures equivalent to the CO2 compensation point, or by >210 millibars O2 which permits internal CO2 production by photorespiration. If leaflets are illuminated in 60 microbars CO2 and 210 millibars O2 (the CO2 compensation point in air), no photoinhibition is observed. Electron transport processes and fluorescence emission associated with photosystem II are inhibited in chloroplast thylakoids isolated from leaflets after illumination in zero CO2 and 10 millibars O2. These studies support the hypothesis that CO2 recycling through photorespiration is one means of effectively dissipating excess photochemical energy when CO2 supply to illuminated leaves is limited.  相似文献
