Rooted cuttings of Kalanchoë blossfeldiana cv. Feuer Bluteand K. crenatum failed to show a net dark CO2 fixation whenraised in dilute nutrient solution. Dark CO2 fixation (CAM)in these plants was initiated either by increasing the soluteconcentration or lowering the water potential of the nutrientsolution by addition of mannitol (0.11 M and 0.25 M) and carbowax4000 (0.16 M and 0.3 M). Initiation was also brought about byspraying the leaves with B-9 (N,dimethylamino-succinamicacid,300mg1–1) or by addition of CCC (2 chloroethyl trimethylammonium chloride, 300 or 750 mg1–1) to the nutrient medium.Failure of CAM in dilute solution was suggested to be due tolack of accumulation of photosynthates in the leaves. Waterstress and growth retardants brought about reduction of monilizationand/or translocation thereby leading to accumulation of assimilatesin the leaves and to initiation of dark CO2 fixation.  相似文献

Dark fixation of CO2 by leaf disks or whole leaves taken fromplants of variety ‘Feuer Blute’ was measured using14CO2. Results indicate that dark fixation by leaf disks isindependent of photoperiodic induction of the plant, but isquantitatively related to the amount of light, over a fairlywide range, to which the leaf is exposed in the single precedinglight period.  相似文献

Cratoneuron filicinum, a drought-sensitive moss, and Tortularuralis, a drought-tolerant moss, fix CO2 non-autotrophicallyat a rate of about 1.2 and 2.2 µmol h–1 g–1dry wt. respectively. During drying, T. ruralis fixes CO2 atan undiminished rate until the tissue loses about 60% of theinitial fresh weight. Thereafter, CO2 fixation rapidly declinesto zero. Dark CO2 fixation by C.filicinum declines steadilyduring the dehydration period. On rehydration, dark CO2 fixationis resumed immediately in T. ruralis but not in C.filicinum.When dried T. ruralis is equilibrated with an atmosphere ofnearly 100% relative humidity, its weight increases to about40% of the original fresh weight and dark CO2 fixation resumesat a rate about 60% of the fresh moss. In C.filicinum thereis only a small increase in weight and little CO2 fixation inthe dark. The non-autotrophically fixed carbon, in both mossesstudied, is incorporated into amino acids (more than 60% ofthe total, mainly into aspartate, alanine and glutamate) andorganic acids (less than 40% of the total, mainly into malate).It is suggested that on rehydration immediate availability ofNADPH, known to be produced by transhydrogenation from NADHduring dark CO2 fixation, may be an important factor in therepair of drought-induced cellular damage by reductive biosynthesisof membrane components and other cellular constituents. Tomato plants were grown in solution culture in a controlledenvironment at 20 ?C with a 12 h photoperiod of 400 µmolquanta m–2 s–1 PAR with either normal ambient CO2,approximately 340 vpm, or with 1000 vpm CO2. The short- andlong-term effects of CO2 enrichment on photosynthesis were determinedtogether with the levels of ribulose-1, 5-bisphosphate carboxylase(RuBPco) E.C. 4.1.1.39 [EC] protein and activity throughout leafdevelopment of the unshaded 5th leaf above the cotyledons. Thehigh CO2 concentration during growth did not appreciably affectthe rate of leaf expansion or final leaf area but did increasethe fresh weight per unit area of leaf. With short-term CO2enrichment, i.e. only during the photosynthesis measurements,the light-saturated photosynthetic rate (Pmax) of young leavesdid not increase while those reaching full expansion more thandoubled their net rate of CO2 fixation. However, with longerterm CO2 enrichment, i.e. growing the crop in high CO2, theplants did not maintain this photosynthetic gain. While theCO2 concentration during growth did not affect the peak in Pmaxmeasured in 300 vpm CO2 or Pmax in 1000 vpm CO2, RuBPco proteinor its activity, the subsequent ontogenetic decline in theseparameters was greatly accelerated by the high CO2 treatment.Compared with plants grown in normal ambient CO2 the high CO2grown leaves, when almost fully expanded, contained only approximatelyhalf as much RuBPco protein and Pmax in 300 vpm CO2 and Pmaxin1000 vpm CO2 were similarly reduced. The loss of RuBPco proteinmay be a major factor associated with the accelerated fall inPmax since it was close to that predicted from the amount andkinetics of RuBPco assuming RuBP saturation. In the oldest leavesexamined grown in high CO2 additional factors may be limitingphotosynthesis since RuBPco kinetics marginally overestimatedPmax in 300 vpm CO2 and the initial slope of photosynthesisin response to intercellular CO2 was also less than expectedfrom the extractable RuBPco. The CO2 compensation point at 25 °C and 250 µEinsteinsm–2 s–1 wasmeasured for 27 bryo-phyte species, andwas found to be in the range of 45–160 µl CO2 I–1air. Under the same conditions Zea mays gave a value of 11 µlI–1 and Horde um vulgare 76 µI–1. The rate of loss of photosyntheticallyfixed 14CO2 in the light and dark in six bryophytes (three mosses,two leafy liverworts, one thalloid liverwort) was determinedin CO2-free air and 100% O2. The rate of 14CO2 evolution inthe light was less than that in the dark in CL2-free air, butin 100% O2 the rate in the light increased, so that in all butthe leafy liverworts it was greater than that in the dark. Raisingthe temperature tended to increase the rate of 14CO2 evolutioninto CO2-free air both in the light and dark, so that the light/dark(L/D) ratio did not greatly vary. The lower rate of loss of14CO2 in the light compared tothe dark could be due to partialinhibition of ‘dark respiration’ reactions in thelight, a low rate of glycolate synthesis and oxidation, or partialreassimilation of the 14CO2 produced, or a combination of someor all of these factors.  相似文献

The effects of exposure of up to 2 h with sulphur dioxide ona range of plant species was observed by measuring changes inthe rate of net photosynthesis under closely controlled environmentalconditions. Ryegrass, Lolium perenne ‘S23’ was thespecies most sensitive to SO2; significant inhibition was detectedat 200 nl l–1. Fumigations at 300 nl l–1 also inhibitedphotosynthesis in field bean (Vicia faba cv. ‘Three FoldWhite’ and ‘Blaze’) and in barley (Hordeumvulgare cv. ‘Sonja’). No effect was detected inwheat (Triticum aestivum cv. ‘Virtue’) at concentrationsup to 600 nl l–1 SO2, or in oil-seed rape (Brassica napuscv. ‘Rafal’) except at 800 nl l–1 SO2). Recoverycommenced immediately after the fumigation was terminated andwas complete within 2 h when inhibition had not exceeded 20%during the SO2 treatment. The influence of salinity in the growing media on ribulose-1,5-bisphosphate (RuBP) carboxylase and on CO2 fixation by intactsugar beet (Beta vulgaris) leaves was investigated. RuBP carboxylase activity was mostly stimulated in young leavesafter exposure of plants for 1 week to 180 mM NaCl in the nutrientsolution. This stimulation was more effective at the higherNaHCO2 concentrations in the reaction medium. Salinity also enhanced CO2 fixation in intact leaves mostlyat rate-limiting light intensities. A 60 per cent stimulationin CO2 fixation rate was obtained by salinity under 450 µEm–2 s–1. At quantum flux densities of 150 µEm–2 s–1 (400–700 nm) this stimulation was280 per cent. Under high light intensities no stimulation bysalinity was found. In contrast, water stress achieved by directleaf desiccation or by polyethylene glycol inhibited enzymeactivity up to fourfold at –1.2 MPa. When Chlorella oulgaris ll h cells grown in air containing 4%CO2 (high-CO2 cells) were given low concentrations of14CO2 (<150ppm), the initial rate of photosynthetic 14CO2 fixation wasvery low and linear 14CO2 fixation was observed after an inductionperiod which lasted for ca. 45 min. No such induction period was observed when high-CO2 cells weregiven high concentrations of 14CO2 (10,000 ppm) or when IOW-CO2cells were given either low or high concentrations of 14CO2,supporting the observations by Briggs and Whittingham (l). However,irrespective of CO2 concentrations during growth and of 14CO2concentrations during the experiments, most of the 14C was incorporatedinto phosphate esters during the initial periods of photosynthetic14CO2 fixation. Intracellular accumulation of inorganic carbon (Ci) and itsfixation in photosynthesis were investigated using siliconeoil layer filtering centrifugation technique with the cellsof Chlorella vulgaris 11h grown under ordinary air. Both CO2and HCO3 were transported into the cells from the reactionmedium and accumulated in the cells, but the rate of transportwas much faster for the former than the latter. 14C-fixationfrom the total transported Ci was much more efficient when CO2was added in the external medium than when HCO3 was added.This indicates that CO2 and HCO3 were not converted tothe common compound in the cells during the initial period ofphotosynthesis. Accumulation of Ci into the cells was much lesssusceptible to low temperature than its fixation. Accumulationof Ci was also observed in the dark. Ethoxyzolamide, an inhibitorof carbonic anhydrase (CA), inhibited the fixation of accumulatedCO2 in the cells, suggesting that CA enhanced the supply ofCO2 to the reaction site of ribulose bisphosphate carboxylasein the stroma. Rates of CO2 and HCC3 fixation in cells of various Chlorellaspecies in suspension were compared from the amounts of 14Cfixed during the 5 s after the injection of a solution containingonly 14CO2 or H14CO3. Results indicated that irrespectiveof the CO2 concentration during growth, Chlorella vulgaris 11h and C. miniata mainly utilized CO2, whereas C. vulgaris C-3,C. sp. K. and C. ellipsoidea took up HCO3 in additionto CO2. Cells of C. pyrenoidosa that had been grown with 1.5%CO2 (high-CO2 cells) mainly utilized CO2, whereas those grownwith air (low-CO2 cells) utilized HCO3 in addition toCO2. Cells that utilized HCO3 had carbonic anhydrase(CA) on their surfaces. The effects of Diamox and CA on the rates of CO2 and HCO3fixation are in accord with the inference that HCO3 wasutilized after conversion to CO2 via the CA located on the cellsurface. CA was found in both the soluble and insoluble fractions;the CA on the cell surface was insoluble. Independent of the modes of utilization, the apparent Km (NaHCO3)for photosynthesis was much lower in low-CO2 cells than in high-CO2ones. The distribution of radioactivity in Chlorella during dark 14CO2fixation was investigated either (a) in normal cells with andwithout added ammonium chloride, or (b) in nitrogen-starvedcells supplied with intermediates of the Krebs-Henseleit ureacycle. In the control experiments almost all the activity was presentin compounds of or associated with, the tricarboxylic acid cycle. The amino-acids citrulline and arginine became radioactive onlyin the presence of ammonia or ornithine where initially theycomprised 40–60 per cent. of the total activity, reactionsof the Krebs–Henseleit urea cycle being implicated intheir formation. No evidence could be found for a complete ureacycle. Unidentified compounds deriving their radioactivity fromthe C4 carbon of citrulline and/or arginine were detected andformed up to 40 per cent. of the total 14CO2 incorporated after25 min.  相似文献

Inorganic carbon transport during photosynthesis of cyanobacteriumAnabaena variabilis grown under ordinary air was investigatedby supplying 14CO2 or H14CO3 solution to three differentstrains. Both CO2 and HCO3 were accumulated within thealgal cells. In the cell suspension from which dissolved inorganiccarbon had been depleted by pre-illumination, CO2 was transportedand accumulated faster than HCO3. When the concentrationof HCO3 injected into the cell suspension of A. variabilisM3 was 25 times as high as that of CO2 (the expected ratio atequilibrium at pH 7.8), the initial rates of fixation of bothinorganic carbon species were practically the same. On the otherhand, when 14CO2 or H14CO3 was added under steady statephotosynthetic conditions, both carbon species were transportedat similar rates. The ratio of fixed to transported carbon measuredafter the initial 5 s was only 23–27% regardless of thecarbon species supplied. Mass spectrometry has been used to investigate the transportof CO2 in the freshwater diatom Navicula pelliculosa. The timecourseof CO2 formation in the dark after addition of 100 mmol m–3dissolved inorganic carbon (DIC) to cell suspensions showedthat no external carbonic anhydrase (CA) was present in thesecells. Upon illumination, cells pre-incubated at pH 75 with100 mmol m–3 DIC, removed almost all free CO2 from themedium at an initial rate of 285 µmol CO2 mg–1Chl h–1. Equilibrium between HCO3 and CO2 in themedium occurred rapidly upon addition of bovine CA, showingthat CO2 depletion resulted from a selective uptake of CO2 ratherthan an uptake of all inorganic carbon species. However, photosyntheticO2 evolution rate remained constant after CO2 had been depletedfrom the medium indicating that photosynthesis is sustainedprimarily by active HCO3 uptake. Treatment of cells with2-iodoacetamide (83 mol m–3) completely inhibited CO2fixation but had little effect on CO2 transport since initialrates of CO2 depletion were about 81% that of untreated cells.Transfer of iodoacetamide-treated cells to the dark caused arapid increase in the CO2 concentration in the medium largelydue to the efflux of the unfixed intracellular DIC pool whichwas found to be about 194 times the concentration of that inthe external medium. These results indicate that Navicula pelliculosaactively takes up molecular CO2 against a concentration gradientby a process distinct from HCO3 transport. Measuring the Canopy Net Photosynthesis of Glasshouse Crops   总被引:3,自引:0,他引:3  
A null balance method is described for measuring net photosynthesisof mature canopies of cucumber and other protected crops overperiods of 10 min in a single-span glasshouse (c. 9m x 18m inarea). Accuracy of control of the CO2 concentration in the greenhouseatmosphere is within ±10 vpm of the normal ambient level(c. 350 vpm). The amounts of CO2 used in canopy net photosynthesisare measured with linear mass flowmeters accurate to within±0.80g. The total errors incurred in measuring canopynet photosynthesis at an ambient CO2 level are estimated tobe of the order of ± 1·2% in bright light (350W m–2, PAR)and ±3·6% in dull light (100W m–2, PAR). Measurements of the rates of net photosynthesis of a maturecanopy of a cucumber crop were made at near-ambient CO2 concentrationsover a range (0–350 W m–2) of natural light fluxdensities. A model of light absorption and photosynthesis applicableto row crops was used to obtain a net photosynthesis versuslight response curve for the cucumber crop. At a light fluxdensity of 350 W m–2 the fitted value of canopy net photosynthesiswas 2.65 mg CO2 m–2s–1 (equivalent to over 95 kgCO2 ha–1h–1). The results are discussed in relationto the need for CO2 supplements to avoid depletion in both ventilatedand unventilated glasshouses during late spring and summer. The rate of net CO2 fixation in Lemna gibba L. was decreasedto 50% by 100–150 min incubation in the presence of 0•5mol m–3 L-methionine-D,L-Sulphoximine (MSO), an irreversibleinhibitor of glutasnine synthetase (GS). The pattern of inhibitionwas similar in both 21% O2 and 2% O2. The inhibition was accompaniedby increased intracellular levels. Incubation with 10 mol m–3 under the same conditions, but without MSO, resulted in even higher levels but the rate of CO2 fixation was unaffected. Additions of glutamine, glutamate, glycine or serine delayedthe MSO-induced inhibition of CO2 fixation. The same amino acidsdelayed the inactivation of GS by MSO. Thus inhibition of CO2 fixation by MSO in Lemna is neither causedby elevated levels nor closely related to photorespiration. Fully light-intercepting simulated swards of S24 perennial ryegrasswere exposed to contrasting environmental conditions in a growthroom for 4 days. Half experienced 20 h days of 120 Wm–2(400–700. nm) and 5 °C, and came to have a WSC (watersoluble carbohydrate) content of 235 mg g–1 and half 4h days of 20 Wm–2 and 25 °C leading to a WSC of 25mg g–1. Their rates of CO2 efflux were monitored at anumber of temperatures during an 8 h dark period; half experiencedincreasing (5–30 °C) and half decreasing (30–5°C) temperatures. The ‘high’ WSC swards hadrespiration rates of 3.7 mg CO2 g–1 (d. wt) h–1at 15 °C, and the ‘low’ swards 0.8 mg CO2 g–1h–1. The order in which the temperatures were experiencedwas immaterial. Even the ‘low’ WSC swards showedno evidence of a respiratory decline during the dark periodthat could be attributed to substrate shortage. The relationshipbetween temperature and CO2 efflux was best represented by logisticcurves. Even so, a Q10 of 2 fitted the data reasonably well,at least up to 20 °C, and has practical advantages wheninterpolating estimated between measured values of respirationin the construction of a carbon balance sheet. Etiolated Avena sativa L. coleoptile sections were used to determinethe influence of C2H4 on in vivo and in vitro rates of CO2 fixation,and to measure the influence of various permutations of C2H4,CO2, and malate on growth. Whereas 1 mM malate or 320 µI-1 CO2 stimulated growth by approximately 100 per cent, inhibitionof growth by 10-8 µ I-1 C2H4 was substantial only in thepresence of malate or CO2 The increase in growth rate in responseto these two agents was eliminated by the simultaneous applicationof C2H4. The in vivo rate of dark [14C]bicarbonate fixationand in vitro enzymic assays of fixation were not measurablyinhibited by C2H4. These results are discussed in the lightof evidence which indicates that CO2-stimulated growth is mediatedby dark fixation. The data do not support the view that C2H4inhibition of growth results from an inhibition of fixation,but suggests that C2H4 may inhibit some step in the processby which malate stimulates growth.  相似文献

Activities of photosynthetic and photorespiratory enzymes viz.,ribulose bisphosphate carboxylase, phosphoenol pyruvate carboxylaseand glycolate oxidase from jute (Corchorus olitorius L.; cv.JRO 632) leaves were compared with those from maize (C4) andsunflower (C3) leaves. The photosynthetic CO2 fixation products,the release of 14CO2 in light and dark following photosynthesisin 14CO2, chlorophyll a: b ratio, gross leaf photosyntheticrate and dry matter production rate were also studied. The carbon balance of shade-grown Ananas comosus was investigatedwith regard to nitrogen supply and responses to high PAR. Netdark CO2 uptake was reduced from 61.2 to 38.5 mmol CO2 m–2in N limited (–N) plants grown under low PAR (60 µmolm–2 s–1) and apparent photon yield declined from0.066 to 0.034 (mol 02.mol–1 photon), although photosyntheticcapacities (measured under 5% CO2) were similar. Following transferfor 7 d to high PAR (600. µmol m–2 s–1), netCO2 uptake at night increased by 14% in +N plants, and daytimephotosynthetic capacity was higher, with a maximum value of7.8 µmol m–2 s–1. The magnitude of dark CO2 fixation during CAM was measured asdawn—dusk variations in leaf-sap titratable acidity (H+)and as the proportion of malic and citric acids. The contributionfrom re-fixation of respiratory CO2 recycling (measured as thedifference between net CO2 uptake and malic acid accumulation)varied with growth conditions, although it was generally lower(30%) than reported for other bromeliads. Assuming a stoichiometryof 2H+: malate and 3H+: citrate, there was a good agreementbetween titratable protons and enzymatically determined organicacids. The accumulation of citric acid was related to nitrogensupply and PAR regime, increasing from 7.0 mol m–3 (+Nplants) to 18 mol m–3 (–N plants) when plants weretransferred to high PAR; malate: citrate ratios decreased from13.1 to 2.5 under these conditions. Under the low PAR regime, leaf-sap osmotic pressure increasedat night in proportion to malic acid accumulation. However,following the transfer to high PAR for 7 d, there was a muchgreater depletion of soluble sugars at night which correspondedto a decrease in leaf-sap osmotic pressure. Although a rolefor citric acid in CAM has not been properly defined, it appearsthat the accepted stoichiometry for CAM in terms of gas exchange,titratable acidity, malic acid and osmotic pressure may nothold for plants which accumulate citric acid. Chlorella cells incubated in the dark longer than 12 hr showedpronounced blue light-induced 14CO2 fixation into aspartate,glutamate, malate and fumarate (blue light effect), whereasthose kept under continuous light showed only a slight bluelight effect, if any. 2) During dark incubation of Chlorellacells, phosphoenolpyruvate carboxylase activity and the capacityfor dark 14CO2 fixation decreased significantly, whereas ribulose-1,5-diphosphatecarboxylase activity and the capacity for photosynthetic 14CO2fixation (measured under illumination of white light at a highlight intensity) did not decrease. 3) In cells preincubatedin the dark, intracellular levels of phosphoenolpyruvate and3-phosphoglycerate determined during illumination with bluelight were practically equal to levels determined during illuminationwith red light. 4) The blue light effect was not observed incells incubated widi chloramphenicol, indicating that blue light-inducedprotein synthesis is involved in the mechanism of the effect.
