The viability of axillary buds and the growth and potential yield of lateral shoots at nodes of red raspberry (Rubus idaeus) infected naturally by Didymella applanata or Botrytis cinerea were measured on excised nodes, decapitated nursery canes or on canes from fruiting plantations. In comparison with lesion-free nodes, buds at infected nodes were smaller and fewer of them were capable of growth when excised and ‘forced’, although the difference in growth between infected and uninfected nodes decreased during late winter. After February, those buds at infected nodes which were capable of forced growth did so as early and with a similar growth rate as those at lesion-free nodes. In April, 70% of buds at infected nodes were capable of growth compared with 94% of those at lesion-free nodes. When nursery canes of cv. Mailing Delight were decapitated above infected nodes the emergence of lateral shoots from the terminal infected node did not differ significantly from that at lesion-free nodes. On a range of farm sites in Scotland the emergence of shoots at infected nodes in the cropping region of canes was significantly poorer than from uninfected nodes but substantially better at infected nodes of cv. Glen Clova than at those of cvs Mailing Jewel and Mailing Orion. It is suggested that cv. Glen Clova is relatively tolerant of spur blight and cane botrytis. The length and potential yield of laterals which developed at infected nodes in the cropping region of canes in these three cultivars did not differ significantly from those at lesion-free nodes. In all tests there was no significant difference in growth at nodes infected by D. applanata and B. cinerea which may indicate a common mechanism for suppression of buds.  相似文献

The inheritance of resistance in red raspberry (Rubus idaeus) to yellow rust (Phragmidium rubi-idaei) was studied in a diallel cross among the cultivars Boyne, Meeker, Mailing Jewel, Glen Prosen and Glen Clova. The progenies and clonally propagated parents were exposed initially in a rust-infected plantation where the incidence and severity of infection was assessed at the telial stage. The following spring leaves on new canes of the same plants were inoculated with urediniospores in a glasshouse at 18 ± 2 °C. The latent period and number of uredinia per cm2 of abaxial leaf surface were determined 13 and 18 days after inoculation. Cv. Boyne developed only chlorotic flecking on inoculated leaves in the glasshouse and no sporulation was observed. It is postulated that this reaction was determined by a major gene which is designated Yr. Some of the other parents showed incomplete resistance of the slow rusting type, and segregants of Boyne selfed that were susceptible also showed low levels of rust, which suggests that Boyne carries both major- and minor-gene resistance. Diallel analyses of both the field and glasshouse data from the susceptible segregates indicated that the degree of incomplete resistance present was determined predominantly by additive gene action, though small but significant non-additive effects also occurred: cv. Meeker was the most resistant parent and contributed the most resistance to the progenies. The Meeker progenies also segregated for gene H, which determines cane hairiness and is known to be associated with resistance to other diseases. Segregants without hairs (genotypes hh) were found to be the more resistant to rust.  相似文献

Nodal lesions typical of natural infections of Didymella applanata were produced on young raspberry canes when wounded petioles were inoculated with mycelium. The buds which developed in the axils of these inoculated petioles were smaller and less likely to produce lateral shoots than those in the axils of delaminated or undamaged petioles, but only when the petioles were inoculated in the field before the end of July. This effect was more pronounced in cv. Mailing Jewel than in cv. Glen Clova which suggests that the latter is more tolerant of D. applanata. Sprays to control D. applanata in the cropping region of canes will probably be effective in Scotland only if applied before the end of July.  相似文献

The effects of cultivar, virulence of isolates of Leptosphaeria coniothyrium, cane maturation and wound healing were examined in a series of inoculation experiments carried out over a 3–yr period in three countries in an attempt to explain why cane blight has caused serious yield losses in machine-harvested red raspberries in Europe, but not in the Pacific Northwest of America. Three isolates of L. coniothyrium from Puyallup (USA), Dundee (UK) and Dossenheim (FRG) were pathogenic on the three test cultivars Willamette, Malling Jewel and Glen Isla in all the experiments. Isolates and cultivars differed for aggressiveness and susceptibility respectively but their ranking was dependent on the test conditions and the differences were small and unlikely to explain the differences in incidence and severity of cane blight in raspberry fields in Scotland and the Pacific Northwest. Conditions at Dossenheim were most favorable for lesion development. At all sites, canes inoculated in late summer produced shorter lesions than those inoculated earlier. A delay between wounding and inoculation produced shorter lesions than simultaneous wounding and inoculation.  相似文献

Abstract

Pseudomonas spp. strains capable of inducing systemic resistance were applied to sugarcane by sett treatment followed by soil applications in the field. Later the fungal pathogen Colletotrichum falcatum causing red rot disease of sugarcane was inoculated in the treated canes and its colonization was assessed by ELISA at different nodal positions above the point of inoculation. Studies with three cvs showed a significant variation in pathogen colonization only in disease susceptible cv CoC 671 and not with cvs Co 8021 and BO 91, moderately susceptible and moderately resistant to the disease, respectively. In further studies when pathogen colonization was assessed on the entire stalks of cv CoC 671, the pathogen titre was significantly reduced from three nodes upwards in the treated canes. In the upper nodes no pathogen colonization was noticed in bacteria-treated canes, whereas in the control all the nodes recorded higher titre for pathogen infection. Incorporation of chitin in the talc formulation caused further reduction in fungal colonization in the stalks.  相似文献

A single population of container-grown red raspberry plants cv. Mailing Jewel was divided and grown in two environments prior to wound inoculating young canes with Didymella applanata in late July. The plants were then sub-divided and held in one of three contrasting post-inoculation environments. Various assessments were made to examine the effect on spur blight of the different environmental treatments. Inoculation of internodes showed that D. applanta produced longer lesions and more pseudothecia on canes grown in spring in an unheated glasshouse than on canes grown outside, but post-inoculation conditions had little effect on these aspects of disease. Petiole inoculations showed that dwarfing of axillary buds in the first season and lateral shoot failure in the year after infection were most serious on canes grown outside in spring but held at relatively high temperatures in August and September after inoculation. The results indicate that spur blight may have the potential to cause higher yield losses in areas with warmer spring and autumn conditions than those usually experienced in north-east Scotland.  相似文献

A method was developed for the inoculation of flowers of cut roses with conidia of Botrytis cinerea. Flower buds were inoculated by spraying of conidial supensions that were ultrasonicated for 10 s. The differences in susceptibility between 8 rose cvs to infections of isolate Bc-33 and the differences in pathogenicity between 14 5. cinerea isolates to cv. ‘Sonia’ were evaluated. Isolates obtained from rose flowers caused higher infection rates than those obtamed from various other hosts. The CVS ‘Madelon’, ‘Melody’ and ‘Sonia’ were found to be highly susceptible, whereas the cvs ‘Caramboie’, ‘Gabriella’, ‘Pasadena’ and ‘Rubinette’ were only slightly susceptible. The lower disease severity in the less susceptible cvs was based on a retardation of the growth of mfection hyphae in the petals, which may have been due to partial resistance. The formation of symptoms is effectuated already by the initial stages in the infection process. Thus, the apparent existing partial resistance to infections of B. cinerea cannot prevent the loss of ornamental value.  相似文献

Mycelial inoculation of canes with Botrytis cinerea proved a useful method for assessing Rubus material for resistance. When canes were inoculated in summer resistant genotypes developed relatively small lesions which produced few small sclerotia in the following spring. The size of lesions in autumn generally provided the best discrimination between genotypes. Very strong resistance was found in several species, of which Rubus pileatus and R. occidentalis are the most useful for breeding; hybrids of raspberry with these species or with R. crataegifolius also had strong resistance. The resistance shown by a derivative of red raspberry cv. Chief was less strong, as was the resistance conferred by gene H, which determines cane pubescence.  相似文献

The effectiveness of resistance to the aphid Amphorophora rubi in restricting the spread of aphid-borne viruses was assessed in a field experiment using six genotypes of red raspberry. In one block of the experiment, the genotypes alternated with rows of virus-infected Mailing Jewel raspberry, and in the other they alternated with virus-free Mailing Jewel. During 4 years, the numbers of A. rubi and the amount of 52V virus spread in the two blocks were similar, suggesting that this virus was mostly introduced from outside the plots. Lloyd George and Mailing Jewel raspberry became heavily infested with A. rubi and were rapidly infected with raspberry leaf mottle, raspberry leaf spot and 52V viruses. Glen Clova and Norfolk Giant raspberry, which contain minor genes for resistance to A. rubi, were infested with fewer A. rubi and virus spread more slowly in these cultivars. A. rubi were rare on Mailing Orion and an East Mailing raspberry selection (888/49) which have genes A1 and A10 respectively for resistance to A.rubi, and these plants remained largely free of virus. The role of minor and major gene resistance to A. rubi in restricting virus spread is discussed. A few Macrosiphum euphorbiae and Myzus ornatus were recorded on several of the raspberry genotypes.  相似文献

Effective control of raspberry spur blight (Didymella applanata) and increased crop yields in the following season were obtained in a plantation of cv. Mailing Jewel with a programme of four or five sprays commencing when the new canes first appeared. Treatments were applied at 14-day intervals and continued until c. 2 wk before harvest. Thiram (0.16% a.i), benomyl (0.025% a.i.), dichlofluanid (0.10% a.i.) and captan (0.10% a.i.) gave good control of the disease but as benomyl and dichlofluanid were also effective in controlling powdery mildew (Sphaero-theca macularis) and Botrytis cinerea on the berries these fungicides appear to be the most suitable.  相似文献

The effects of various cane removal treatments on the epidemiology of Phragmidium rubi-idaei were studied in four experiments in infected raspberry plantations in Wales and Scotland. In 1981, removal of 15 cm high young canes either by cutting or by spraying dinoseb-in-oil reduced infection of a second flush of canes (replacement canes) compared to that of the first flush of canes on untreated plots of cv. Mailing Delight in Wales and of cv. Glen Clova in Scotland. In 1982, removal of 20 cm high young canes reduced infection of cv. Mailing Delight in Wales but in Scotland only removal of 60 cm high canes by cutting reduced infection of replacement canes. Removal of all fruiting canes and old cane stubs reduced rust infection of young canes, but removal of successive flushes of replacement canes did not reduce infection of lateral shoots on the fruiting canes. Dinoseb-in-oil applied to the bases of fruiting canes before emergence of young canes had no effect on infection of young canes in Wales or Scotland. The production of basidiospores was inhibited in vitro by dinoseb-in-oil at concentrations over 1.0 μg a.i./ml.  相似文献

Spread of Septoria nodorum from inoculated point sources was examined in pure stands and mixtures of two spring wheat cultivars Kolibri and Maris Butler. Gradients in disease were observed soon after inoculation; the presence of the more resistant cultivar (Maris Butler) in the mixtures retarded the outward spread of disease compared with the susceptible pure stand (Kolibri). Regression analysis suggested that gradients in incidence were influenced by nearness to the source whereas gradients in severity were not. Spread of disease was also examined in pure stands and mixtures for the host/pathogen combinations, winter wheat (cvs Maris Huntsman and Maris Ranger)/S. nodorum and winter barley (cvs Maris Otter and Hoppel)/Rhynchosporium secalis. In contrast to the spring wheat experiment, no gradients were observed; explanations for the uniform distribution of disease were (a) extensive spread prior to the period of assessment, (b) no physical barrier to dispersal due to the juvenile growth stage of the crop and (c) exhaustion of the point-source.  相似文献

Two complementary experiments were conducted in a controlled environment to elucidate the interactions between the fungus Sclerotinia trifoliorum Erikss. and red clover (Trifolium pratense L.). In one of these studies, two hardened diploid red clover cultivars (cvs) were inoculated with 20 isolates of S. trifoliorum of various geographic origins. In the other study, 20 red clover cvs, diploid or tetraploid, including late and medium‐late flowering types, were inoculated with two isolates of the fungus. Prior to inoculation, some plants were hardened by subjecting them to a low temperature and light treatment mimicking autumn conditions. Late flowering cvs were found more resistant than medium‐late ones. Isolates collected in the northern region, where late cvs are grown, were significantly more aggressive than isolates from southern locations, where medium‐late cvs are more prevalent. Such an adaptation has not previously been reported for this fungus. This is the first report concerning flowering type and resistance in red clover. Tetraploids were generally not more resistant than diploids. A hardening procedure for red clover plants was found to be a prerequisite for detecting the differences in disease resistance.  相似文献

In glasshouse experiments, barley seedlings were inoculated with barley yellow mosaic virus (BaYMV) either mechanically or by using zoospores or cystosori of a viruliferous isolate of the vector, Polymyxa graminis, maintained on barley in sand culture. Experiments using mechanical inoculation showed that seedlings became more resistant with age. Consistent cultivar differences were obtained: cvs Maris Otter and Halcyon were very susceptible and cv. Athene seemed immune. Symptoms developed more rapidly at 23 than at 17 or 11 oC. After vector inoculation, symptoms developed more slowly than after mechanical inoculation but cultivar ranking was similar. Cultivars did not differ in susceptibility to the vector, as measured by zoospore production on their roots. Spring barley cultivars supported the growth of the vector which remained viruliferous and some showed symptoms although, in the field, symptoms do not appear on spring-sown crops.  相似文献

When released to commerce in 1981, the red raspberry cv. Glen Moy was reported to be immune to the Scottish type isolate of raspberry bushy dwarf virus (RBDV-D200). Field observations of this cultivar in localities where RBDV was prevalent tended to support this claim of its resistance, but in the past 6–10 yr, RBDV infection has been reported in this cultivar in Australasia, USA and in several commercial crops in England. Therefore, experiments were made to investigate the reason(s) for this apparent anomaly using RBDV-infected material, putatively of cv. Glen Moy, from two locations in southern England and one each from Australia, New Zealand (NZ) and the USA. Genetic fingerprinting of genomic DNA from samples of these five RBDV-infected raspberry sources confirmed their identity as cv. Glen Moy. Comparisons of some serological and genomic properties of the five Glen Moy RBDV isolates indicated that, whilst they shared many properties with previously well characterised isolates of this virus, they were distinguishable from them. Characterisation of the isolate from NZ maintained in raspberry showed that it did not have a Rubus host range characteristic of resistance-breaking (RB) isolates, indicating that for this location, and probably also for those of Australia and the USA, RB isolates were not the cause of infection in cv. Glen Moy. When virus-tested plants of cv. Glen Moy and 45 progeny seedlings from the cross between cv. Glen Moy and the RBDV-susceptible cv. Autumn Bliss were graft inoculated with RBDV-D200, all grafted plants became infected indicating that cv. Glen Moy does not contain the RBDV resistance gene, Bu. Possible reasons for the previously reported resistance of cv. Glen Moy to RBDV are discussed.  相似文献

The interaction of two selected isolates of Botrytis cinerea with bean suspension cells and bean leaf discs was compared in relation to levels of reactive oxygen intermediates (ROI). Isolate B 1.7 was arrested by a hypersensitive‐like necrosis of bean leaf tissue. According to its inability to spread and produce conidia on the bean leaf tissue it was classified as non‐aggressive. The second isolate induced a fast expanding light brownish necrosis of the leaf tissue. It was able to produce conidia on bean leaf discs and was classified as aggressive. The generation of superoxide was followed biochemically in inoculated bean cell suspensions. Both isolates induced a similar early superoxide peak approximately 18‐h post inoculation (hpi). While the non‐aggressive isolate induced a much stronger secondary superoxide burst at 33 hpi, the level of superoxide of suspension cells inoculated with the aggressive isolate was below the control level. This is the first report on the occurrence of a biphasic oxidative burst in plant cells induced by a fungal pathogen. Such a suppression of superoxide generation was also observed in bean leaf discs inoculated with the aggressive isolate. An oxidative burst‐suppressing agent was extracted from inoculated cell culture medium and determined as 2‐methyl‐succinate (2‐MS) by GC/MS analysis. The compound was detected approximately 20 hpi in the aggressive fungus–plant interaction. 2‐MS was able to suppress the hypersensitive response‐like necrosis on leaf discs as well as the second superoxide burst in suspension cells when inoculated with the non‐aggressive isolate. The early superoxide burst at 18 hpi was not affected. The results confirm the important role of enhanced production of ROI in plant resistance reactions, also for a necrotrophlike B. cinerea.  相似文献

The concentration of particles of black raspberry necrosis virus (BRNV), which is normally extremely low in herbaceous plants, increased about 1000-fold when Nicotiana clevelandii plants were inoculated with a mixture of BRNV and an unrelated virus, solanum nodiflorum mottle (SNMV). In sap from N. clevelandii infected with the mixed culture, BRNV infectivity survived dilution to 10?4 but not 10?5, and storage for 6 but not 8 days at 20 oC, for 6 but usually not 10 days at 4 oC and for more than 13 days at – 15 oC. When plants were inoculated with the mixed culture, BRNV induced typical symptoms in several Chenopodium species and infected several previously unreported hosts. Purified preparations of particles of the mixed culture contained only a small proportion of BRNV particles, which sedimented in sucrose density gradients as two components, one, probably non-infective, of c. 505, and the other, infective, of 120-130S. An antiserum prepared to purified particles of the mixed culture was cross-absorbed with SNMV particles and used in indirect ELISA to detect BRNV in herbaceous plants infected with the mixed culture, and also in a wide range of Rubus species, cultivars and hybrids infected naturally, by grafting or by inoculation with the aphid Amphorophora idaei. The reliability of ELISA for detecting BRNV in raspberry leaves depended on the cultivar and time of year. Some cultivars, such as Glen Clova, had low concentrations of BRNV, which was detected reliably only in late spring/early summer, whereas other cultivars, such as Lloyd George and Mailing Enterprise, had greater BRNV concentrations. In small-scale surveys in eastern Scotland, BRNV was detected by ELISA in many raspberry cvs, including some that contain major gene resistance to the vector, A. idaei; in five of nine raspberry stocks entered for the Standard grade certificate but in none of five stocks entered for the Stock Cane certificate; and in 40% of wild raspberry and 14% of wild bramble plants growing near commercial raspberry crops. The significance of these findings for the control of BRNV is discussed.  相似文献

In studies on the infection of potato cultivars with different field isolates of Phoma exigua var. foveata, cultivar × isolate interactions, although sufficiently large to influence the relative pathogenicities of isolates on different cultivars, were not substantial enough to affect cultivar ranking order. Cultivar rank was markedly influenced by pathogen isolate only when both field and culture collection isolates were compared. This suggests that the complications of cultivar × isolate interactions can be avoided in cultivar screening tests by the use of recent field isolates of high pathogenicity. It was found that cultivar was considerably more important than isolate in determining gangrene lesion size. The need to consider both lesion diameter and depth when estimating rot size following point inoculation of different cultivars was confirmed.  相似文献

When raspberry canes were inoculated in summer with a mycelial inoculum of Didymella applanata resistant genotypes developed relatively small lesions which produced few fruiting bodies in the following spring. Scores of the frequency of these fruiting bodies provided the best discrimination between genotypes. Very strong resistance was found in R. pileatus, R. occidentalis and R. coreanus and in hybrids of these species with red raspberry. Hybrids with R. crataegifolius were less resistant. Resistance was also found in Malling 1473/35, a derivative of red raspberry cv. Chief, and in red raspberry segregates with gene H which determines cane pubescence. Both the expression of resistance and the ranking of resistance sources for their resistance was similar for D. applanata to that reported for Botrytis cinerea.  相似文献

Levels and components of resistance to Amphorophora idaei in raspberry cultivars containing different A. idaei resistance genes were studied under infestation tunnel, glasshouse and laboratory conditions. Each test consistently ranked raspberry cultivars, with increasing levels of resistance in the order, non-resistant (cv. Mailing Jewel), resistant cultivars containing minor genes, major gene A1, gene A10 (red raspberry) and gene A10 (black raspberry) respectively. Resistance was expressed in three different ways; decreased alatae settling and feeding, decreased apterae settling and decreased aphid fecundity and rate of nymphal development. Following exposure to a large population of alatae, significantly fewer aphids settled on A1, A10 and to a lesser extent, minor gene-containing cultivars, compared to cv. Mailing Jewel. More alatae settled on the top than the bottom zone of non-resistant, minor gene resistant and two of four A1-containing cultivars; alatae settling was low on all zones of A10-containing raspberries. Aphid fecundity and nymph development patterns on different cultivars and resistance classes were similar to those found for alatae settling. After 7 days reproduction more than 30% of the nymphs developed to third or fourth instar on cv. Mailing Jewel, whilst on minor gene and major gene-containing cultivars the total number of nymphs and the proportion of later instars decreased. On resistant cultivars the nymphs were found mainly on the middle and bottom leaf zones, compared to cv. Mailing Jewel. A rapid (48 h) screening test using floating leaflets was developed and, on the basis of apterae settling, ranked cultivars reliably; it was particularly effective in distinguishing moderate and strong resistance to strain 1 A. idaei. Gene A1-containing cultivars bred in England were much more resistant to strain 1 A. idaei than those bred in Scotland. Possible reasons for this difference are discussed, in relation to modified screening procedures and to the control of the viruses transmitted by this aphid vector.  相似文献
