The peroxiredoxins (Prxs) constitute a very large and highly conserved family of thiol-based peroxidases that has been discovered only very recently. We consider here these enzymes through the angle of their discovery, and of some features of their molecular and physiological functions, focusing on complex phenotypes of the gene mutations of the 2-Cys Prxs subtype in yeast. As scavengers of the low levels of H2O2 and as H2O2 receptors and transducers, 2-Cys Prxs have been highly instrumental to understand the biological impact of H2O2, and in particular its signaling function. 2-Cys Prxs can also become potent chaperone holdases, and unveiling the in vivo relevance of this function, which is still not established, should further increase our knowledge of the biological impact and toxicity of H2O2. The diverse molecular functions of 2-Cys Prx explain the often-hard task of relating them to peroxiredoxin genes phenotypes, which underscores the pleiotropic physiological role of these enzymes and complex biologic impact of H2O2.  相似文献

The histochemical and cytochemical localization of abscisic acid (ABA)-induced H2O2 production in leaves of maize (Zea mays L.) plants were examined, using 3,3-diaminobenzidine (DAB) and CeCl3 staining, respectively, and the relationship between ABA-induced H2O2 production and ABA-induced subcellular activities of antioxidant enzymes was studied. H2O2 generated in response to ABA treatment was detected within 0.5 h in major veins of the leaves and maximized at about 2–4 h. In mesophyll and bundle sheath cells, ABA-induced H2O2 accumulation was observed only in apoplast, and the greatest accumulation occurred in the walls of mesophyll cells facing large intercellular spaces. Meanwhile, ABA treatment led to a significant increase in the activities of the leaf chloroplastic and cytosolic antioxidant enzymes superoxide dismutase (SOD), ascorbate peroxidase (APX) and glutathione reductase (GR), and pretreatment with the NADPH oxidase inhibitor diphenyleneiodonium (DPI), the O 2 scavenger Tiron and the H2O2 scavenger dimethylthiourea (DMTU) almost completely arrested the increase in the activities of these antioxidant enzymes. Our results indicate that the accumulation of apoplastic H2O2 is involved in the induction of the chloroplastic and cytosolic antioxidant enzymes. Moreover, an oxidative stress induced by paraquat (PQ), which generates O 2 and then H2O2 in chloroplasts, also up-regulated the activities of the chloroplastic and cytosolic antioxidant enzymes, and the up-regulation was blocked by the pretreatment with Tiron and DMTU. These data suggest that H2O2 produced at a specific cellular site could coordinate the activities of antioxidant enzymes in different subcellular compartments.  相似文献

Menadione-catalyzed H2O2 production by viable animal cells was proportional to the viable cell number, and H2O2 production decreased with increasing cytotoxic effects after the incubation of cells with cytotoxic compounds. The cytotoxic effects of food additives, pesticides, antibiotics, heavy metals, phytotoxins, mycotoxins, and marine toxins were estimated using the above test employingNIH/3T3 and Neuro-2a cells. Synergistic effects of the toxin mixture were observed and acute cytotoxicity detected 1 h after the incubation of cells with toxins. This menadione-catalyzed H2O2production assay is rapid and simple compared to other popular cytotoxicity tests such as the MTT reduction assay and Neutral red inclusion test, requiring4 h. The menadione-catalyzed H2O2 production assay is expected to be a useful food safety test for rapidly detecting toxic compounds having a basic cytotoxic effect on common animal cells. Oxidation of vanadyl sulfate by H2O2 involves multiple reactions at neutral pH conditions. The primary reaction was found to be oxidation of V(IV) to V(V) using 0.5 equivalent of H2O2, based on the loss of blue color and the visible spectrum. The loss of V(IV) and formation V(V) compounds were confirmed by ESR and51V-NMR spectra, respectively. In the presence of excess H2O2 (more than two equivalents), the V(V) was converted into diperoxovanadate, the major end-product of these reactions, identified by changes in absorbance in ultraviolet region and by the specific chemical shift in NMR spectrum. The stoichiometric studies on the H2O2 consumed in this reaction support the occurrence of reactions of two-electron oxidation followed by complexing two molecules of H2O2. Addition of a variety of compounds—Tris, ethanol, mannitol, benzoate, formate (hydroxyl radical quenching), histidine, imidazole (singlet oxygen quenching), and citrate—stimulated a secondary reaction of oxygen-consumption that also used V(IV) as the reducing source. This reaction requires concomitant oxidation of vanadyl by H2O2, favoured at low H2O2:V(IV) ratio. Another secondary reaction of oxygen release was found to occur during vanadyl oxidation by H2O2 in acidic medium in which the end-product was not diperoxovanadate but appears to be a mixture of VO 3 + (–546 ppm), VO3+ (–531 ppm) and VO 2 + (–512 ppm), as shown by the51V-NMR spectrum. This reaction also occurred in phosphate-buffered medium but only on second addition of vanadyl. The compounds that stimulated the oxygen-consumption reaction were found to inhibit the oxygen-release reaction. A combination of these reactions occur depending on the proportion of the reactants (vanadyl and H2O2), the pH of the medium and the presence of some compounds that affect the secondary reactions.  相似文献

The deleterious effects of H2O2 on the electron transport chain of yeast mitochondria and on mitochondrial lipid peroxidation were evaluated. Exposure to H2O2 resulted in inhibition of the oxygen consumption in the uncoupled and phosphorylating states to 69% and 65%, respectively. The effect of H2O2 on the respiratory rate was associated with an inhibition of succinate-ubiquinone and succinate-DCIP oxidoreductase activities. Inhibitory effect of H2O2 on respiratory complexes was almost completely recovered by β-mercaptoethanol treatment. H2O2 treatment resulted in full resistance to QO site inhibitor myxothiazol and thus it is suggested that the quinol oxidase site (QO) of complex III is the target for H2O2. H2O2 did not modify basal levels of lipid peroxidation in yeast mitochondria. However, H2O2 addition to rat brain and liver mitochondria induced an increase in lipid peroxidation. These results are discussed in terms of the known physiological differences between mammalian and yeast mitochondria.  相似文献

In many legume nodules, the H2 produced as a byproduct of N2 fixation diffuses out of the nodule and is consumed by the soil. To study the fate of this H2 in soil, a H2 treatment system was developed that provided a 300 cm3 sample of a soil:silica sand (2:1) mixture with a H2 exposure rate (147 nmol H2 cm–3hr–1) similar to that calculated exist in soils located within 1–4 cm of nodules (30–254 nmol H2 cm–3hr–1). After 3 weeks of H2 pretreatment, the treated soils had a Km and Vmax for H2 uptake (1028 ppm and 836 nmol cm–3 hr–1, respectively) much greater than that of control, air-treated soil (40.2 ppm and 4.35 nmol cm–3 hr–1, respectively). In the H2 treated soils, O2, CO2 and H2 exchange rates were measured simultaneously in the presence of various pH2. With increasing pH2, a 5-fold increase was observed in O2 uptake, and CO2 evolution declined such that net CO2 fixation was observed in treatments of 680 ppm H2 or more. At the H2 exposure rate used to pretreat the soil, 60% of the electrons from H2 were passed to O2, and 40% were used to support CO2 fixation. The effect of H2 on the energy and C metabolism of soil may account for the well-known effect of legumes in promoting soil C deposition.  相似文献

A new procedure for fluorescent detection of intracellular H2O2 in cells transiently expressing the catalyst Horseradish Peroxidase (HRP) is setup and validated. More specific reaction with HRP largely amplifies oxidation of the redox probes used (2′,7′-dichlorodihydrofluorescein and dihydrorhodamine). Expression of HRP does not affect cell viability. The procedure reveals MAO activity, a primary intracellular H2O2 source, in monolayers of intact transfected cells. The probes oxidation rate responds specifically to the MAO activation/inhibition. Their oxidation by MAO-derived H2O2 is sensitive to intracellular H2O2 competitors: it decreases when H2O2 is removed by pyruvate and it increases when the GSH-dependent removal systems are impaired. Specific response was also measured after addition of extracellular H2O2. Oxidation of the fluorescent probes following reaction of H2O2 with endogenous HRP overcomes most criticisms in their use for intracellular H2O2 detection. The method can be applied for direct determination in plate reader and is proposed to detect H2O2 generation in physio-pathological cell models.  相似文献

Adequate methods to measure the rate of mitochondrial oxygen radical generation are needed since oxygen radicals are involved in many pathologies. A fluorometric method appropriate to measure the rate of generation of H2O2 in intact mitochondria is described. Just after isolation of functional mitochondria from fresh tissues, rates of generation of H2O2 are kinetically measured by fluorometry in the presence of homovanillic acid and horseradish peroxidase. The method is specific for H2O2 and is sensitive enough to assay mitochondrial H2O2 generation in the presence of respiratory substrate without inhibitors of the respiratory chain. Simultaneous measurement of mitochondrial oxygen consumption allows calculation of the free radical leak: the percentage of electrons out of sequence which reduce oxygen to oxygen radicals along the mitochondrial respiratory chain instead of reducing oxygen to water at the terminal cytochrome oxidase. The method shows instantaneous response to H2O2. This makes it appropriate to study the quick effects of different inhibitors and modulators on the rate of mitochondrial oxygen radical production. Its application to the localization of the sites where caloric restriction decreases mitochondrial oxygen radical generation in heart mitochondria is described.  相似文献

SB203580 is a specific inhibitor of p38 mitogen-activated protein (MAP) kinase and has been widely used to investigate the physiological roles of p38 in animal and yeast cells. Here by using an epidermal strip bioassay, laser-scanning confocal microscopy and whole-cell patch clamp analysis, we assess the effects of pyridinyl imidazoles-like SB203580 on the H2O2 signaling in guard cells of Vicia faba L. The results indicated that SB203580 blocks H2O2- or ABA-induced stomatal closure, ABA-induced H2O2 generation, and decrease in K+ fluxing across plasma membrane of Vicia guard cells by application of ABA and H2O2, whereas its analog SB202474 had no effect on these events. Thus, these results suggest that activation of p38-like MAP kinase modulates guard cell ROS signaling in response to stress.  相似文献

We have shown earlier a requirement for Ca2+ and calmodulin (CaM) in the H2O2-induced activation of extracellular signal-regulated kinases 1 and 2 (ERK1/2) and protein kinase B (PKB), key mediators of growth-promoting, proliferative, and hypertrophic responses in vascular smooth muscle cells (VSMC). Because the effect of CaM is mediated through CaM-dependent protein kinase II (CaMKII), we have investigated here the potential role of CaMKII in H2O2-induced ERK1/2 and PKB phosphorylation by using pharmacological inhibitors of CaM and CaMKII, a CaMKII inhibitor peptide, and siRNA knockdown strategies for CaMKIIα. Calmidazolium and W-7, antagonists of CaM, as well as KN-93, a specific inhibitor of CaMKII, attenuated H2O2-induced responses of ERK1/2 and PKB phosphorylation in a dose-dependent fashion. Similar to H2O2, calmidazolium and KN-93 also exhibited an inhibitory effect on glucose/glucose oxidase-induced phosphorylation of ERK1/2 and PKB in these cells. Transfection of VSMC with CaMKII autoinhibitory peptide corresponding to the autoinhibitory domain (aa 281–309) of CaMKII and with siRNA of CaMKIIα attenuated the H2O2-induced phosphorylation of ERK1/2 and PKB. In addition, calmidazolium and KN-93 blocked H2O2-induced Pyk2 and insulin-like growth factor-1 receptor (IGF-1R) phosphorylation. Moreover, treatment of VSMC with CaMKIIα siRNA abolished the H2O2-induced IGF-1R phosphorylation. H2O2 treatment also induced Thr286 phosphorylation of CaMKII, which was inhibited by both calmidazolium and KN-93. These results demonstrate that CaMKII plays a critical upstream role in mediating the effects of H2O2 on ERK1/2, PKB, and IGF-1R phosphorylation.  相似文献

Enzymatic microelectrode biosensors have been widely used to measure extracellular signaling in real-time. Most of their use has been limited to brain slices and neuronal cell cultures. Recently, this technology has been applied to the whole organs. Advances in sensor design have made  possible the measuring of cell signaling in blood-perfused in vivo kidneys. The present protocols list the steps needed to measure ATP and H2O2 signaling in the rat kidney interstitium. Two separate sensor designs are used for the ex vivo and in vivo protocols. Both types of sensor are coated with a thin enzymatic biolayer on top of a permselectivity layer to give fast responding, sensitive and selective biosensors. The permselectivity layer protects the signal from the interferents in biological tissue, and the enzymatic layer utilizes the sequential catalytic reaction of glycerol kinase and glycerol-3-phosphate oxidase in the presence of ATP to produce H2O2. The set of sensors used for the ex vivo studies further detected analyte by oxidation of H2O2 on a platinum/iridium (Pt-Ir) wire electrode. The sensors for the in vivo studies are instead based on the reduction of H2O2 on a mediator coated gold electrode designed for blood-perfused tissue. Final concentration changes are detected by real-time amperometry followed by calibration to known concentrations of analyte. Additionally, the specificity of the amperometric signal can be confirmed by the addition of enzymes such as catalase and apyrase that break down H2O2 and ATP correspondingly. These sensors also rely heavily on accurate calibrations before and after each experiment. The following two protocols establish the study of real-time detection of ATP and H2O2 in kidney tissues, and can be further modified to extend the described method for use in other biological preparations or whole organs.  相似文献

Hydrogen peroxide (H2O2), an oxidizing agent, has been widely used as a disinfectant. Recently, because of its reactive properties, H2O2 has also been used as a tooth bleaching agent in dental care. This is a cause for concern because of adverse biological effects on the soft and hard tissues of the oral environment. To investigate the influence of H2O2 on odontoblasts, the cells producing dentin in the pulp, we assessed cellular viability, generation of reactive oxygen species (ROS), alkaline phosphatase (ALP) activity, and nodule formation of an odontoblastic cell line (MDPC-23) after treatment with H2O2, and compared those with the effects on preosteoblastic MC3T3-E1 cells. Cytotoxic effects of H2O2 began to appear at 0.3 mmol/L in both MDPC-23 and MC3T3-E1 cells. At that concentration, the accumulation of intracellular ROS was confirmed by a fluorescent probe, DCFH-DA. Although more ROS were detected in MDPC-23, the increasing pattern and rate are similar between the two cells. When the cells were treated with H2O2 at concentrations below 0.3 mmol/L, MDPC-23 displayed a significant increase in ALP activity and mineralized bone matrix, while MC3T3-E1 cells showed adverse effects of H2O2. It is known that ROS are generally harmful by-products of aerobic life and represent the primary cause of aging and numerous diseases. These data, however, suggest that ROS can induce in vitro cell differentiation, and that they play a more complex role in cell physiology than simply causing oxidative damage.  相似文献

Abstract

The inhibitory or activating effect of H2O2 on large conductance calcium and voltage-dependent potassium (BKCa) channels has been reported. However, the mechanism by which this occurs is unclear. In this paper, BKCa channels encoded by mouse Slo were expressed in HEK 293 cells and BKCa channel activity was measured by electrophysiology. The results showed that H2O2 inhibited BKCa channel activity in inside-out patches but enhanced BKCa channel activity in cell-attached patches. The inhibition by H2O2 in inside-out patches may be due to oxidative modification of cysteine residues in BKCa channels or other membrane proteins that regulate BKCa channel function. PI3K/AKT signaling modulates the H2O2-induced BKCa channel activation in cell-attached patches. BKCa channels and PI3K signaling pathway were involved in H2O2-induced vasodilation and H2O2-induced vasodilation by PI3K pathway was mainly due to modulation of BKCa channel activity.  相似文献

In Streptomyces coelicolor A3(2), as the content of palmitoleic acid increased with decreasing growth temperature, H2O2 resistance decreased. Production of thiobarbituric acid-reactive substances upon H2O2 treatment was increased by supplementing unsaturated fatty acids. Therefore, the content of palmitoleic acid is a determining factor for the survival of Streptomyces coelicolor A3(2) subjected to H2O2 stress.  相似文献

Sesquiterpenes have attracted much interest with respect to their protective effect against oxidative damage that may be the cause of many diseases including several neurodegenerative disorders and cancer. Our previous unpublished work suggested that cyclosativene (CSV), a tetracyclic sesquiterpene, has antioxidant and anticarcinogenic features. However, little is known about the effects of CSV on oxidative stress induced neurotoxicity. We used hydrogen peroxide (H2O2) exposure for 6 h to model oxidative stress. Therefore, this experimental design allowed us to explore the neuroprotective potential of CSV in H2O2-induced toxicity in new-born rat cerebral cortex cell cultures for the first time. For this aim, MTT and lactate dehydrogenase release assays were carried out to evaluate cytotoxicity. Total antioxidant capacity (TAC) and total oxidative stress (TOS) parameters were used to evaluate oxidative changes. In addition to determining of 8-hydroxy-2-deoxyguanosine (8-OH-dG) levels, the single cell gel electrophoresis (or Comet assay) was also performed for measuring the resistance of neuronal DNA to H2O2-induced challenge. Our results showed that survival and TAC levels of the cells decreased, while TOS, 8-OH-dG levels and the mean values of the total scores of cells showing DNA damage (Comet assay) increased in the H2O2 alone treated cultures. But pre-treatment of CSV suppressed the cytotoxicity, genotoxicity and oxidative stress which were increased by H2O2. On the basis of these observations, it is suggested that CSV as a natural product with an antioxidant capacity in mitigating oxidative injuries in the field of neurodegenerative disorders.  相似文献

The role of H2O2 in abscisic acid (ABA)-induced NH4+ accumulation in rice leaves was investigated. ABA treatment resulted in an accumulation of NH4+ in rice leaves, which was preceded by a decrease in the activity of glutamine synthetase (GS) and an increase in the specific activities of protease and phenylalanine ammonia-lyase (PAL). GS, PAL, and protease seem to be the enzymes responsible for the accumulation of NH4+ in ABA-treated rice leaves. Dimethylthiourea (DMTU), a chemical trap for H2O2, was observed to be effective in inhibiting ABA-induced accumulation of NH4+ in rice leaves. Inhibitors of NADPH oxidase, diphenyleneiodonium chloride (DPI) and imidazole (IMD), and nitric oxide donor (N-tert-butyl-α-phenylnitrone, PBN), which have previously been shown to prevent ABA-induced increase in H2O2 contents in rice leaves, inhibited ABA-induced increase in the content of NH4+. Similarly, the changes of enzymes responsible for NH4+ accumulation induced by ABA were observed to be inhibited by DMTU, DPI, IMD, and PBN. Exogenous application of H2O2 was found to increase NH4+ content, decrease GS activity, and increase protease and PAL-specific activities in rice leaves. Our results suggest that H2O2 is involved in ABA-induced NH4+ accumulation in rice leaves.  相似文献

The arachidonate cascade is important for the generation of reactive species (RS), and cyclooxygenase (COX) is a key enzyme of this cascade. Tissues of 24-month-old rat lung showed a 2-fold increase in RS, malondialdehyde and thromboxane B2 than those of 6-month-old rat. We found that the effects of 50 µM H2O2 and 200 µM t-butylhydroperoxide (t-BHP) specify on COX activity, and that their effects increased cytosolic COX activity in a concentration-dependent manner (1–50 µM) in 24-month-old rat. Our results suggested that COX activators such as t-BHP and H2O2, which are located in cytosol, are essential for the activation of COX in aged lung.  相似文献

ABSTRACT

Antioxidant enzymes are essential proteins that maintain cell proliferation potential by protecting against oxidative stress. They are present in many organisms including harmful algal bloom (HAB) species. We previously identified the antioxidant enzyme 2-Cys peroxiredoxin (PRX) in the raphidophyte Chattonella marina. This enzyme specifically decomposes a hydrogen peroxide (H2O2). PRX is the only antioxidant enzyme so far identified in C. marina. This study used mRNA-seq, using Trinity assemble and blastx for annotation, to identify a further five antioxidant enzymes from C. marina: Cu Zn superoxide dismutase (Cu/Zn-SOD), glutathione peroxidase (GPX), catalase (CAT), ascorbate peroxidase (APX) and thioredoxin (TRX). In the gene expression analysis of six enzymes (Cu/Zn-SOD, GPX, CAT, APX, TRX and PRX) using light-acclimated (100 μmol photons m?2 s?1) C. marina cells, only PRX gene expression levels were significantly increased by strong light irradiation (1000 μmol photons m?2 s?1). H2O2 concentration and scavenging activity were also increased and significantly positively correlated with PRX gene expression levels. In dark-acclimated cells, expression levels of all antioxidant enzymes except APX were significantly increased by light irradiation (100 μmol photons m?2 s?1). Expression decreased the following day, with the exception of PRX expression. With the exception of CAT, gene expression of antioxidant enzymes was not significantly induced by artificial H2O2 treatment, although average gene expression levels were slightly increased in some enzymes. Thus, we suggest that light is the main trigger of gene expression, but the resultant oxidative stress is also a possible factor affecting the gene expression of antioxidant enzymes in C. marina.  相似文献
