Excised leaves of a C3-photosynthetic type, Hordeum vulgare,a C4-type, Panicum miliaceum, and an intermediate-type, Panicummilioides, were allowed to take up through their cut ends a1 mM solution of butyl hydroxybutynoate (BHB), an irreversibleinactivator of glycolate oxidase. After 30 to 60 min in BHB,extractable glycolate oxidase activity could not be detectedin the distal quarter of the leaf blades. Following this pretreatment,recovery of 14C-glycolate from 14CO2 incorporated in a 10 minperiod was nearly maximal for each of the three plant types.Labeled glycolate was 51% of the total 14CO2 incorporated forthe C3-species, 36% for the intermediate-species, and 27% forthe C4-species Increased labeling of glycolate was compensatedfor primarily by decreased labeling of the neutral and basicfractions for the C3 and intermediate-type species. In the C4-type,label decreased primarily in the neutral and insoluble fractions,but increased in the basic fraction. A lower rate of glycolatesynthesis is indicative of a lower rate of photorespirationand consistent with a lower O2/CO2 ratio present in the bundle-sheathcells of C4-plants. We conclude that both decreased glycolatesynthesis and the refixation of photorespiratory-released CO2are important in maintaining a lower rate of photorespirationin C4-plants compared to C3 plants. Intermediate glycolate synthesisin Panicum milioldes is consistent with its intermediate levelof O2 inhibition of photosynthesis and intermediate rate ofphotorespiration. Both malate and aspartate were decarboxylated at the 4-carbonposition by isolated bundle sheath strands of C4 plants butto different extents depending upon the species. In Digitariasanguinalis, an NADP-malic enzyme (NADP-ME) species, 100 µMoxalic acid blocked malate decarboxylation through NADP-ME withoutaffecting aspartate decarboxylation which apparently occursthrough NAD-ME. In several phosphoenolpyruvate carboxykinase(PEP-CK) type C4 species, 200 µM 3-mercaptopicolinic acid(3-MPA), an inhibitor of PEP-CK, specifically inhibited themalate decarboxylation and partially inhibited aspartate decarboxylation.The aspartate decarboxylation insensitive to 3-MPA may occurthrough NAD-ME. Neither inhibitor prevented C4 acid decarboxylationin bundle sheath cells of NAD-ME species. Patterns of initial photosynthetic CO2 incorporation were determinedfor some seagrasses and were related to activities of primarycarbon fixing enzymes, carbonic anhydrase activities, and 13Cvalues. According to the incorporation patterns, Cymodocea nodosa wasa C4 species while Thalassia hemprichli and Thalassodendronciliatum were C3 plants. Halophila stipulacea showed an unusualincorporation pattern which could be viewed as intermediatebetween typical C3 and C4 pathways. The activity ratios of ribulose-l,5-bisphosphate carboxylase (RUBPcase) to phosphoenolpyruvatecarboxylase (PEPcase) were about 3 for Thalassodendron ciliatumand 1 for Cymodocea nodosa and Halophila stipulacea. The lattervalue, which is intermediate to ratios found in terrestrialC3 and C4 plants, may correlate with the incorporation patternsfound for Halophila stipulacea. Since the C4 seagrass lackedthe Kranz anatomy, it may, in addition, point to a flexibleincorporation potential for these plants. The high 13C values found in these and other seagrasses didnot correlate with their photosynthetic pathways as in terrestrialplants. This discrepancy is probably due to a ‘closedsystem’ type of photosynthesis in which CO2 is efficientlyutilized. The C3 species which utilize CO2 enzymatically must convertexogenous HCO-3 to CO2 internally. Even though carbonic anhydraseactivities were very low, conversion rates seemed to be sufficientfor high rates of photosynthesis. Since enzymatic fixation ratesapproached photosynthetic rates even at CO2 saturation, thelimitation for these seagrasses to express their high photosyntheticpotential is most probably the HCO3 uptake system.  相似文献

In order to study the effects of inorganic phosphate (P1) starvationon C4plants, 3-week-old maize plants (Zea maysL cv. Brulouis)were grown in a growth chamber on a nutrient solution withoutP1 over 22 d During the first 2 weeks, Pi-starved plants grewas well as control plants The Pi concentration in the planttissue decreased rapidly with time, which suggests that normalbiomass production can be maintained at the expense of internalP1 In addition, photosynthetic CO2 assimilation measured 4-6h after dawn was not affected, but the concentration of glucose,sucrose, and starch in leaves was much higher than in the controls14CO2 pulse-chase experiments earned out on the ninth day oftreatment showed that 14CO2 assimilation was perturbed duringthis initial period, resulting in a larger flow of carbon toboth starch and sucrose At the beginning of the third week ofP1 starvation (15 d after treatment) 14C incorporation intosucrose stayed high relative to controls but this was not thecase for starch At the end of the third week of P1-deficiency,shoot growth was considerably reduced and fresh weight was onlyone-third of that of the control plants. The P1 concentrationof both the leaf and root tissues was less than 1.0 µmolg–1 FW compared to 20-25µmol g1 FW in the controls.Photosynthetic CO2 assimilation was reduced and the leaf concentrationof sucrose and starch, which had begun to decrease after theend of the second week of P1 limitation, became lower than inthe controls. These results obtained on maize plants show thatphotosynthesis and carbon partitioning between sucrose and starchwere strongly affected by P1 deficiency, similar to C3 species. Soybeans were grown for three seasons in open-top field chambersto determine (1) whether elevated CO2 (360 versus 700 µmolmol–1) alleviates some of the yield loss due to pollutantO3, (2) whether the partial stomatal closure resulting fromchronic O3 exposure (charcoal-filtered air versus 1.5 ambientconcentrations) is a cause or result of decreased photosynthesis,and (3) possible implications of CO2/O3 interactions to climatechange studies using elevated CO2. Leaf conductance was reducedby elevated CO2, regardless of O3 level, or by exposure to O3alone. As.a result of these effects on conductance, high CO2reduced estimated midday O3 flux into the leaf by an averageof 50% in charcoal-filtered air and 35% in the high O3 treatment.However, while exposure to O3 reduced seed yields by 41% atambient CO2 levels, the yield reduction was completely amelioratedby elevated CO2. The threshold midday O3 flux for yield lossappears to be 20–30 nmol m–2 s–1 in this study.Although elevated CO2 increased total biomass production, itdid not increase seed yields. A/Ci curves show a large reductionin the stomatal limitation to photosynthesis due to elevatedCO2 but no effect of O3. These data demonstrate that (1) reducedconductance due to O3 is the result, and not the cause, of reducedphotosynthesis, (2) 700 µmol mol–1 CO2 can completelyameliorate yield losses due to O3 within the limits of theseexperiments, and (3) some reports of increased yields underelevated CO2 treatments may, at least in part, reflect the ameliorationof unrecognized suppression of yield by O3 or other stresses. Light-enhanced active pyruvate uptake into mesophyll chloroplastsof C4 plants was reported to be mimicked by either of the twotypes of cation jump: H+-jump in maize and phylogenically relatedspecies (H+-type) and Na+-jump in all the other C4 species tested(Na+-type) [Aoki, N., Ohnishi, J. and Kanai, R. (1992) PlantCell Physiol. 33: 805]. In this study, medium and stromal pH was monitored in the suspensionof C4 mesophyll chloroplasts. Medium alkalization lasting for5 to 10 seconds after pyruvate addition was detected by a pHelectrode and observed only in the light and only in mesophyllchloroplasts from H+-type species, Zea mays L. and Coix lacryma-jobiL., but not in those from Na+-type species Panicum miliaceumL., Setaria italica (L.) Beauv. and Panicum maximum Jacq. Theinitial rate of H+ consumption showed good correlation with[14C]pyruvate uptake measured by silicone oil filtering centrifugation,both being inhibited by N-ethylmaleimide and 7-chloro-4-nitrobenzo-2-oxa-l,3-diazole to the same degree. The ratio of the rate of H+ uptaketo that of pyruvate uptake was always about 1. Pyruvate-inducedacidification of the stroma was observed in maize mesophyllchloroplasts. The quantum yields of C3 and C4 plants from a number of genera and families as well as from ecologically diverse habitats were measured in normal air of 21% O2 and in 2% O2. At 30 C, the quantum yields of C3 plants averaged 0.0524 ± 0.0014 mol CO2/absorbed einstein and 0.0733 ± 0.0008 mol CO2/absorbed einstein under 21 and 2% O2. At 30 C, the quantum yields of C4 plants averaged 0.0534 ± 0.0009 mol CO2/absorbed einstein and 0.0538 ± 0.0011 mol CO2/absorbed einstein under 21 and 2% O2. At 21% O2, the quantum yield of a C3 plant is shown to be strongly dependent on both the intercellular CO2 concentration and leaf temperature. The quantum yield of a C4 plant, which is independent of the intercellular CO2 concentration, is shown to be independent of leaf temperature over the ranges measured. The changes in the quantum yields of C3 plants are due to changes in the O2 inhibition. The evolutionary significance of the CO2 dependence of the quantum yield in C3 plants and the ecological significance of the temperature effects on the quantum yields of C3 and C4 plants are discussed.  相似文献

Cyclopenin (C17H14O3N2) and cyclopenol (C17H14O4N2), isolatedfrom an abberent strain of Penicillium cyclopium (NRRL 6233),significantly inhibited the growth of etiolated wheat (Triticumaestivum) coleoptile segments. The former inhibited at 10–3and 10–4 M, the latter at 10–3 M. Cyclopenin producedmalformation of the first set of trifoliate leaves in bean (Phaseolusvulgaris) at 10–2 M and necrosis and stunting in corn(Zea mays) at 10–2 M. Cyclopenol induced no apparent effectsin bean or corn plants. Neither compound changed the growthor morphology of tobacco (Nicotiana tabacum) plants. Cyclopenininduced intoxication, prostration and ataxia in day-old chicksat 500 mg/kg, but they recovered within 18 hours. Cyclopenolwas inactive against chicks when dosed at levels up to 500 mg/kg. Non-dormant small cocklebur seeds (Xanthium pennsylvanicum Wallr.)are potentiated to germinate, if they are subjected to anaerobiccondition for certain time periods after being sufficientlypre-soaked under aerobic conditions. This is termed "anaerobicinduction" of seed germination. Such induction was slightlyinhibited by CO2 applied during anaerobiosis, but markedly promotedby C2H4 Thus, C2H4 can exert its action even in anaerobiosis,but does not enhance the fermentative CO2 evolution. No actualanaerobic induction occurred when over 1? O2 was present, evenif C2H4 had been applied. Two common tallgrass prairie species, Andropogon gerardii, thedominant C4 grass in this North American grassland, and Salviapitcheri, a C3 forb, were exposed to ambient and elevated (twiceambient) CO2 within open-top chambers throughout the 1993 growingseason. After full canopy development, stomatal density on abaxialand adaxial surfaces, guard cell length and specific leaf mass(SLM; mg cm-2) were determined for plants in the chambers aswell as in adjacent unchambered plots. Record high rainfallamounts during the 1993 growing season minimized water stressin these plants (leaf xylem pressure potential was usually >-1·5 MPa in A. gerardii) and also minimized differencesin water status among treatments. In A. gerardii, stomatal densitywas significantly higher (190 ± 7 mm-2; mean ±s.e.) in plants grown outside of the chambers compared to plantsthat developed inside the ambient CO2 chambers (161 ±5 mm-2). Thus, there was a significant 'chamber effect' on stomataldensity. At elevated levels of CO2, stomatal density was evenlower (P < 0·05; 121 ± 5 mm-2). Most stomatawere on abaxial leaf surfaces in this grass, but the ratio ofadaxial to abaxial stomatal density was greater at elevatedlevels of CO2. In S. pitcheri, stomatal density was also significantlylower when plants were grown in the open-top chambers (235 ±10 mm-2 outside vs. 140 ± 6 mm-2 in the ambient CO2 chamber).However, stomatal density was greater at elevated CO2 (218 ±12 mm-2) compared to plants from the ambient CO2 chamber. Theratio of stomata on adaxial vs. abaxial surfaces did not varysignificantly in this herb. Guard cell lengths were not significantlyaffected by growth in the chambers or by elevated CO2 for eitherspecies. Growth within the chambers resulted in lower SLM inS. pitcheri, but CO2 concentration had no effect. In A. gerardii,SLM was lower at elevated CO2. Maize seedlings were grown in pots either with or without preconditionedseeds of the parasitic weed, Striga hermonthica. After between4 and 8 weeks, net photosynthesis in the leaves of maize plantsinfected with Striga decreased compared to leaves of uninfectedcontrol plants. The activities of four enzymes of photosyntheticmetabolism were, however, little affected by infection. A pulse-chaseexperiment using 14CO2 showed that C4 acids were the main earlyproducts of assimilation even when the rate of photosynthesiswas much decreased by infection, but more radio-activity appearedin glycine and serine than in leaves of healthy maize plants.Leaves of infected maize required longer to reach a steady rateof photosynthesis upon enclosure in a leaf chamber than leavesof uninfected plants after similar treatment. Electron microscopy of transverse sections of the leaves ofinfected maize indicated that the cell walls in the bundle sheathand vascular tissue were less robust than in leaves of healthyplants. The results suggest that infection with Striga causesan increase in the permeability of cell walls in the bundlesheath, leakage of CO2 from the bundle sheath cells and decreasedeffectiveness of C4 photosynthesis in host leaves. The photosynthetic characteristics of Eleocharis baldwinii (Torr.)Chapman, an amphibious leafless plant in the Cyperaceae, wereinvestigated in both the terrestrial form and the submergedform of the plant. Anatomical observation of the culm, whichis the photosynthetic organ in this plant, revealed that theterrestrial form has the Kranz type of anatomy, whereas thesubmerged form has an inner structure that is similar to thatof submerged aquatic plants, with a reduction in both the numberand the size of bundle sheath cells and vascular bundles andrelatively well developed mesophyll cells. In 14C-pulse 12C-chaseexperiments with the terrestrial form, 80% of the total fixed14C was incorporated into C4 dicarboxylic acids after a 10-spulse. The radioactivity in the C4 acids decreased rapidly,while that in sucrose increased to 36% during a 120-s chase.In the submerged form, 64% and 30% of the total fixed 14C wasincorporated into C4 acids and phosphate esters, respectively,after a 10-s pulse. The radioactivity of these compounds decreasedrelatively slowly during a 120-s chase. The specific activitieson a chlorophyll basis of C4 photosynthetic enzymes that areinvolved in the NAD-ME subtype were high in the terrestrialform, while they were intermediate between those of C3 and C4plants in the submerged form. The activity of ribulose 1,5-bisphosphatecarboxylase was 1.5 times higher in the submerged form thanin the terrestrial form. By contrast, the activity of carbonicanhydrase exhibited the reverse tendency. Western blot analysisof soluble proteins extracted from the mesophyll cells and thebundle sheath strands of the terrestrial form demonstrated thatribulose 1,5-bisphosphate carboxylase/oxygenase protein waspresent in the mesophyll cells as well as in the bundle sheathcells, with a higher level in the latter, although phosphoenolpyruvatecarboxylase and pyruvate, Pi dikinase proteins were restrictedto the mesophyll cells. In the submerged form, diurnal fluctuationsin levels of malate were observed with significant fixationof CO2 at night. However, the diurnal changes of malate weresmaller than those reported for CAM plants. These data indicatethat the terrestrial form of Eleocharis baldwinii fixes atmosphericCO2 essentially via the C4 pathway, while the submerged formfixes inorganic carbon via a complex metabolic system that resemblesan intermediate between C3 and C4 metabolism in associationwith a CAM-like profile. Pascopyrum smithii (C3) andBouteloua gracilis (C4) are importantforage grasses native to the Colorado shortgrass steppe. Thisstudy investigated photosynthetic responses of these grassesto long-term CO2enrichment and temperature in relation to leafnonstructural carbohydrate (TNC) and [N]. Glasshouse-grown seedlingswere transferred to growth chambers and grown for 49 d at twoCO2concentrations (380 and 750 µmol mol-1) at 20 and 35°C, and two additional temperatures (25 and 30 °C) at750 µmol mol-1CO2. Leaf CO2exchange rate (CER) was measuredat a plant's respective growth temperature and at two CO2concentrationsof approx. 380 and 700 µmol mol-1. Long-term CO2enrichmentstimulated CER in both species, although the response was greaterin the C3,P. smithii . Doubling the [CO2] from 380 to 750 µmolmol-1stimulated CER ofP. smithii slightly more in plants grownand measured at 30 °C compared to plants grown at 20, 25or 35 °C. CO2-enriched plants sometimes exhibited lowerCER when compared to ambient-grown controls measured at thesame [CO2], indicating photosynthetic acclimation to CO2growthregime. InP. smithii , such reductions in CER were associatedwith increases in TNC and specific leaf mass, reductions inleaf [N] and, in one instance, a reduction in leaf conductancecompared to controls. InB. gracilis , photosynthetic acclimationwas observed more often, but significant changes in leaf metabolitelevels from growth at different [CO2] were generally less evident.Temperatures considered optimal for growth (C3: 20 °C; C4:35 °C) sometimes led to CO2-induced accumulations of TNCin both species, with starch accumulating in the leaves of bothspecies, and fructans accumulating only inP. smithii. Photosynthesisof both species is likely to be enhanced in future CO2-enrichedand warmer environments, although responses will sometimes beattenuated by acclimation. Lehnherr, B. M?chler, F. and N?sberger, J. 1985. Effect of CO2concentration during growth on a CO2 concentrating mechanismin white clover as predicted from differential 14CO2/12CO2 uptake.-J. exp. Bot. 36: 1835-1841. White clover was grown at 20 and100 Pa p(CO2). The CO2 response of net photosynthesis and differentialuptake of 14CO2 and 12CO2 by leaves were measured at varioustemperatures and at various O2 and CO2 partial pressures andcompared with predictions from ribulose bisphosphate carboxylase/oxygenasekinetics. Discrepancies between the observed gas exchange characteristicsfor the leaves and those predicted from the enzyme kineticswere interpreted as being due to a CO2 concentrating mechanism.Plants grown at 20 Pa p(CO2) showed a higher affinity for CO2than plants grown at 100 Pa p(CO2) when measured at 10 ?C. Nodifference in affinity was found at 30 ?C. The postulated CO2concentrating effect was greater in plants grown at low CO2than in plants grown at high CO2 concentration and occurredonly at low temperature and low CO2 partial pressure. It issuggested that plants grown at the lower CO2 partial pressurehave a higher affinity for CO2 due to a more efficient CO2 concentratingsystem than plants grown at the higher CO2 partial pressure. Panicum hians and Panicum milioides were found to have characteristicsintermediate to those of C3 and C4 species with respect to CO2compensation point, percentage inhibition of photosynthesisby O2 at various O2/CO2 solubility ratios, and water use efficiency.C4 species have a higher carboxylation efficiency than eitherthe intermediate or C3 species. During photosynthesis, evenunder 2.5% O2, C4 species have a higher affinity for intercellularCO2 (Km 1.6 µM) apparently due to the initial carboxylationthrough PEP carboxylase. Under low O2 the intermediate and C3species had a similar affinity for intercellular CO2 duringphotosynthesis (Km 5–7 µM) consistent with carboxylationof atmospheric CO2 through RuDP carboxylase. There were considerablevariation in photosynthesis/unit leaf area at saturating CO2levels in the species examined which in part is due to differencesin RuDP carboxylase /unit leaf area. The highest rates of photosynthesis/unitleaf area under CO2-saturating conditions were with the C3 specieswhich had a correspondingly high level of RuDP carboxylase/unitleaf area. Sunflower plants were grown under controlled environmental conditionswith either 0 or 10 mol m–3 phosphate (Pi). From steady-statemeasurements of gas exchange and chlorophyll fluorescence madeon intact leaves, the in vivo CO2/O2 specificity factor (invivo Ksp) of ribulose 1,5-Aisphosphate carboxylase-oxygenase(Rubisco) was determined following two methods based on modelsof C3 photosynthesis by Brooks and Farquhar (1985) and Peterson(1989). The two methods gave in vivo Ksp values for controlsunflower leaves which were similar to published values forhigher plants. Extreme Pi deficiency decreased in vivo Ksp,in sunflower leaves compared to adequate Pi. This suggests thatPi deficiency affected photorespiration less than photosynthesis.The decrease in in vivo Ksp may be due to a real change in theenzyme kinetics favouring oxygenation more than carboxylationor due to an increase in the number of CO2 molecules releasedper oxygenation; in which case the observed decrease in thein vivo Ksp determined on intact leaves will not agree numericallywith the true Ksp of Rubisco determined in vitro using purifiedenzyme from the same leaf. We discuss the implications of therelatively large photorespiration in Pi-deficient sunflowerleaves with respect to the increased dissipation of photosyntheticelectrons and photorespiratory recycling of Pi in thechloroplaststroma. Although our results on in vivo Ksp suggested a relativelylarger photorespiratory potential in Pi-deficient than controlsunflower leaves, photosynthesis was insensitive to O2 in Pi-deficientleaves; the possible reasons for this phenomenon are discussed.Under extreme Pi deficiency, O2 sensitivity of photosynthesisis not a reflection of the in vivo photorespiratory rates. Determinationof in vivo Ksp of Rubisco is a useful approach to study thephotorespiratory potential of intact leaves. The effects of Na application on growth and nitrate reductaseactivity of seven C4 plant species, Zea mays, Echinochloa crus-galli,Panicum miliaceum, Panicum coloratum, Panicum dichotomiflorum,Panicum maximum and Chloris gayana were studied. Predicting responses of plant and global carbon balance to theincreasing concentration of carbon dioxide in the atmosphererequires an understanding of the response of plant respirationto carbon dioxide concentration ([CO2]). Direct effects of thecarbon dioxide concentration at which rates of respiration ofplant tissue are measured are quite variable and their effectsremain controversial. One possible source of variation in responsivenessis the energy status of the tissue, which could influence thecontrol coefficients of enzymes, such as cytochrome-c oxidase,whose activity is sensitive to [CO2]. In this study we comparedresponses of respiration rate to [CO2] over the range of 60to 1000 µmol mol-1in fully expanded leaves of four C3andfour C4herbaceous species. Responses were measured near themiddle of the normal 10 h dark period, and also after another24 h of darkness. On average, rates of respiration were reducedabout 70% by the prolonged dark period, and leaf dry mass perunit area decreased about 30%. In all species studied, the relativedecrease in respiration rate with increasing [CO2] was largerafter prolonged darkness. In the C3species, rates measured at1000 µmol mol-1CO2averaged 0.89 of those measured at 60µmol mol-1in the middle of the normal dark period, and0.70-times when measured after prolonged darkness. In the C4species,rates measured at 1000 µmol mol-1CO2averaged 0.79 of thoseat 60 µmol mol-1CO2in the middle of the normal dark period,and 0.51-times when measured after prolonged darkness. In threeof the C3species and one of the C4species, the decrease in theabsolute respiration rate between 60 and 1000 µmol mol-1CO2wasessentially the same in the middle of the normal night periodand after prolonged darkness. In the other species, the decreasein the absolute rate of respiration with increase in [CO2] wassubstantially less after prolonged darkness than in the middleof the normal night period. The CO2 compensation point at 25 °C and 250 µEinsteinsm–2 s–1 wasmeasured for 27 bryo-phyte species, andwas found to be in the range of 45–160 µl CO2 I–1air. Under the same conditions Zea mays gave a value of 11 µlI–1 and Horde um vulgare 76 µI–1. The rate of loss of photosyntheticallyfixed 14CO2 in the light and dark in six bryophytes (three mosses,two leafy liverworts, one thalloid liverwort) was determinedin CO2-free air and 100% O2. The rate of 14CO2 evolution inthe light was less than that in the dark in CL2-free air, butin 100% O2 the rate in the light increased, so that in all butthe leafy liverworts it was greater than that in the dark. Raisingthe temperature tended to increase the rate of 14CO2 evolutioninto CO2-free air both in the light and dark, so that the light/dark(L/D) ratio did not greatly vary. The lower rate of loss of14CO2 in the light compared tothe dark could be due to partialinhibition of ‘dark respiration’ reactions in thelight, a low rate of glycolate synthesis and oxidation, or partialreassimilation of the 14CO2 produced, or a combination of someor all of these factors.  相似文献
