The form and localisation of Cu accumulation in the extraradical mycelium (ERM) of three arbuscular mycorrhizal fungi (AMF), isolated from the same polluted soil contaminated with the Cu and Arsenate, was studied. There were differences in the capacity of the ERM of the three AMF to sorb and accumulate Cu. Glomus caledonium BEG133 had a significantly lower Cu-sorption capacity than Glomus mosseae BEG132 and Glomus claroideum BEG134 isolated from the polluted soil as well as an isolate of G. mosseae BEG25 from a non-polluted soil. This was directly related to the cation exchange capacity (CEC) of the ERM of these fungi. Transmission Electron Microscopy (TEM) and Scanning Electron Microscopy (SEM) linked to an energy dispersive X-ray spectrometer (EDAX) gave more detailed information, showing that the ERM of AMF from the polluted soil was able to accumulate Cu in the mucilaginous outer hyphal wall zone, cell wall and inside the hyphal cytoplasm. The EDAX spectra showed that the accumulated Cu was mainly associated with Fe in the mucilaginous outer hyphal wall zone and in the cell wall. Cu was associated with traces of arsenate inside the cytoplasm of the ERM of Glomus mosseae BEG132 but this was not visible inside the ERM of Glomus caledonium BEG133 or Glomus claroideum BEG134. This work suggests that the ERM of AMF is able to sorb and accumulate Cu, but different tolerance mechanisms exist between the three AMF isolated from the same polluted soil providing further evidence for functional diversity within populations of AMF in soils. 相似文献

Interaction between arbuscular mycorrhizal fungus Glomus mosseae and plant growth promoting fungus Phoma sp. was studied for its effect on their root colonization and plant growth of cucumber. Two isolates of Phoma sp. (GS8-2 and GS8-3) were tested with G. mosseae. The percent root length colonized by G. mosseae was not adversely affected by the presence of Phoma isolates. In contrast, the root colonization of both isolates GS8-2 and GS8-3 in 4-week-old plants was significantly reduced (80.7% and 84.3%, respectively) by added G. mosseae. Inoculating plants with each Phoma isolate significantly increased the shoot dry weight. However, dual inoculation of each Phoma isolate with G. mosseae had no significant effect on growth enhancement. 相似文献

The arbuscular mycorrhizal fungi Glomus monosporum, G. vesiculiferum, G. deserticola, G. intraradices, G. mosseae, and two unidentified species were tested to determine their effect on plant growth and fruit production of tomato (Lycopersicon esculentum Mill.) cv. Trust inoculated with Fusarium oxysporum f. sp. radicis-lycopersici (FORL) under near-commercial greenhouse conditions. Inoculation with G. monosporum and G. mosseae significantly increased fruit yield and fruit number of tomato plants grown hydroponically in sawdust. Plant height and plant dry weight increased significantly when inoculated with G. monosporum and G. mosseae. Further, plants inoculated with G. monosporum and G. mosseae showed significantly lower FORL root infection than the untreated control plants. 相似文献

The 5.8 S subunit and flanking internal transcribed spacer (ITS) regions in nuclear ribosomal DNA (rDNA) from spores of Glomus mosseae FL156 and UK118 were amplified by polymerase chain reaction (PCR) using ITS1 and ITS4 as primers. The amplification product
from template DNA of UK118 was cloned and sequenced (569 bp); the amplified DNA from FL156 was sequenced directly (582 bp).
There was a 95% sequence similarity between DNAs amplified from the two isolates; in contrast, major dissimilarities with
partial sequences of seven other glomalean taxa were observed. Four oligonucleotide sequences unique to Glomus mosseae were identified as potential primers. Their specificity to Glomus mosseae was assessed by PCR amplification of genomic DNA from spores from 36 glomalean fungi: 13 isolates of Glomus mosseae, two Glomus monosporum, 10 other Glomus isolates, and 11 other glomalean taxa from each of four other genera. nonmycorrhizal controls. In addition, a 24-mer oligonucleotide, designated GMOS5, hybridized with Glomus mosseae and Glomus monosporum DNA amplified by PCR using primer pairs ITS1 : ITS4 and GMOS1 : GMOS2. Colony-blot assays showed that GMOS5 hybridized to
100% and 97% of E. coli pUC19 clones of amplification products from Glomus mosseae FL156 and UK118 DNA templates, respectively, indicating that nearly all clones contained an homologous sequence. GMOS5 was
used successfully to detect specifically Glomus mosseae in DNA extracted from colonized sudan grass (Sorghum sudanense L.) roots and amplified by PCR using the primer pair GMOS1 : GMOS2. The results confirm several previous indications that
Glomus mosseae and Glomus monosporum are indistinguishable taxonomic entities.

Seven banana cultivars (Musa acuminata, AAA group) were inoculated with two species of vesicular arbuscular mycorrhizal (VAM) fungi (Glomus mosseae and Glomus macrocarpum) in a greenhouse experiment. Inoculated plants had generally greater shoot dry weight and shoot phosphorus concentrations compared to the noninoculated plants. A great variation in dependency on mycorrhizal colonization was observed among the banana cultivars. Cv. Williams showed the highest relative mycorrhizal dependency (RMD) and cv. Poyo the lowest. For all the cultivars studied, inoculation with G. macrocarpum resulted in the highest RMD values. Both root dry weight and root hair length or density of the noninoculated plants were inverserly correlated with the RMD values of cultivars. 相似文献

丛枝菌根(arbuscular mycorrhizal,AM)真菌在退化生态系统恢复与重建实践中具有重要作用。采用盆栽模拟方法,重点分析不同土壤磷条件下小马鞍羊蹄甲(Bauhinia faberi)幼苗接种AM真菌后,幼苗的形态、生物量积累、菌根侵染率和菌根效应(mycorrhizal growth response, MGR)在一个生长季内的动态变化。结果表明,Glomus mosseae和 Glomus coronatum能较好地侵染幼苗,两种AM真菌显著地增加幼苗根系、叶片数和生物量;接种AM真菌显著影响幼苗的生物量分配,而土壤磷对幼苗的生物量分配影响不明显,AM真菌和土壤磷对幼苗生长的交互作用显著;G. mosseae是小马鞍羊蹄甲的优势AM菌,其接种的幼苗根长、叶片数、生物量、侵染率和菌根效应都显著高于G. coronatum处理的幼苗;菌根效应显著,接种AM真菌能有缓解土壤磷素缺乏的限制作用,且随着苗龄增大促生作用表现更为明显。不同AM菌种对小马鞍羊蹄甲幼苗生长的促生作用表现出的差异,提示在多元资源限制的干旱贫瘠环境中进行生物修复须为目标恢复物种筛选出高效的优势AM真菌。 相似文献

Six species of arbuscular mycorrhizal (AM) fungi (Glomus aggregatum, G. clarum, G. etunicatum, G. intraradices, G. mosseae and G. versiforme) were evaluated, in three greenhouse experiments, for their effects on reproduction of the root-lesion nematode, Pratylenchus penetrans, and growth of Ottawa 3 apple rootstock. Glomus mosseae increased total dry weights of nematode-inoculated and non-inoculated rootstock in all three greenhouse experiments, and G. intraradices increased dry weights in two of three greenhouse experiments. Plants inoculated with G. mosseae generally supported fewer P. penetrans per gram of root than plants inoculated with other AM fungi, but did not differ significantly from the controls in any greenhouse experiment. Colonization of roots by AM fungi was reduced by P. penetrans at initial inoculum densities greater than 250 nematodes/L soil. In field trials, preplant inoculation with either G. intraradices or G. mosseae increased rootstock growth and leaf concentrations of P, Mg, Zn and Cu in fumigated plots but not in non-fumigated plots, indicating that colonization by native AM fungi in non-fumigated plots may have been sufficient for adequate nutrient acquisition. The abundance of vesicles and arbuscules was greater in roots of plants inoculated with AM fungi before planting than in roots of non-inoculated plants, in both fumigated and non-fumigated plots. P. penetrans per gram of root and per 50 ml soil were significantly lower for G. mosseae- inoculated plants than for non-inoculated plants in fumigated soil but not in non-fumigated soil. 相似文献

A microarray carrying 5,648 probes of Medicago truncatula root-expressed genes was screened in order to identify those that are specifically regulated by the arbuscular mycorrhizal
(AM) fungus Gigaspora rosea, by Pi fertilisation or by the phytohormones abscisic acid and jasmonic acid. Amongst the identified genes, 21% showed a common
induction and 31% a common repression between roots fertilised with Pi or inoculated with the AM fungus G. rosea, while there was no obvious overlap in the expression patterns between mycorrhizal and phytohormone-treated roots. Expression
patterns were further studied by comparing the results with published data obtained from roots colonised by the AM fungi Glomus mosseae and Glomus intraradices, but only very few genes were identified as being commonly regulated by all three AM fungi. Analysis of Pi concentrations in plants colonised by either of the three AM fungi revealed that this could be due to the higher Pi levels in plants inoculated by G. rosea compared with the other two fungi, explaining that numerous genes are commonly regulated by the interaction with G. rosea and by phosphate. Differential gene expression in roots inoculated with the three AM fungi was further studied by expression
analyses of six genes from the phosphate transporter gene family in M. truncatula. While MtPT4 was induced by all three fungi, the other five genes showed different degrees of repression mirroring the functional differences
in phosphate nutrition by G. rosea, G. mosseae and G. intraradices.

Cucumber plants were treated with plant growth promoting fungi (PGPF), Phoma sp. (isolates GS8-2 and GS8-3) and Penicillium simplicissimum (isolate GP17-2) with or without the arbuscular mycorrhizal fungus (AMF) Glomus mosseae. Induction of systemic resistance in cucumber against the anthracnose disease caused by Colletotrichum
orbiculare was tested to evaluate the nature of the interaction between the PGPF and AMF. Root colonizing ability of each fungal species as influenced by their interaction was also evaluated. Plant roots were pre-inoculated with each PGPF isolate and/or G. mosseae for four weeks and leaves were then challenge inoculated with the pathogen C. orbiculare. Plants treated with each PGPF isolate showed considerable protection against the disease, but the treatment of G. mosseae had no significant effect on disease development. However, combined inoculation of Phoma GS8-2 or GS8-3 with G. mosseae reduced the level of disease protection induced by single inoculation of each Phoma isolate. In contrast, the high levels of protection induced by the P. simplicissimum GP17-2 were not altered by combining it with G. mosseae. Root colonization of both Phoma sp. isolates was also suppressed by the presence of the G. mosseae, but such an effect was not found on the population development of P. simplicissimum. Three arbuscular mycorrhizal (AM) fungi (Glomus mosseae, Glomus claroideum, and Glomus intraradices) were compared for their root colonizing ability and activity in the root of Astragalus sinicus L. under salt-stressed soil conditions. Mycorrhizal formation, activity of fungal succinate dehydrogenase, and alkaline phosphatase,
as well as plant biomass, were evaluated after 7 weeks of plant growth. Increasing the concentration of NaCl in soil generally
decreased the dry weight of shoots and roots. Inoculation with AM fungi significantly alleviated inhibitory effect of salt
stress. G. intraradices was the most efficient AM fungus compared with the other two fungi in terms of root colonization and enzyme activity. Nested
PCR revealed that in root system of plants inoculated with a mix of the three AM fungi and grown under salt stress, the majority
of mycorrhizal root fragments were colonized by one or two AM fungi, and some roots were colonized by all the three. Compared
to inoculation alone, the frequency of G. mosseae in roots increased in the presence of the other two fungal species and highest level of NaCl, suggesting a synergistic interaction

In this study, it is aimed to asses the association of arbuscular mycorrhizal (AM) fungi within colonised rhizosphere of Gramineae family members through a survey by using nested- polymerase chain reaction method in Van province (Turkey). From 24 agro-ecological fields, a total of 82 samples belonging to Gramineae family were tested by molecular methods. The presence of Glomus intraradices and Glomus mosseae was ascertained in 10 plants belonging to eight different species by using fungus specific primers. Screening of arbuscular mycorrhizal fungi for the revegetation of eroded red soils in subtropical China 总被引:5,自引:0,他引:5
Some acidic red soils in hilly regions of subtropical China were degraded as a result of slope erosion following the removal of natural vegetation, primarily for fuel. Revegetation is important for the recovery of the degraded ecosystem, but plant growth is limited by the low fertility of eroded sites. One factor contributing to the low fertility may be low inoculum density of arbuscular mycorrhizal (AM) fungi. Compared to red soils under natural vegetation or in agricultural production, substrates on eroded sites had significantly lower AM fungal propagule densities. Thus, the management and/or application of AM fungi may increase plant growth and accelerate revegetation. Thirteen species of AM fungi were identified in red soils by spore morphology. Scutellospora heterogama, Glomus manihotis, Gigaspora margarita, Glomus aggregatum and Acaulospora laevis were among the most common according to spore numbers. Pot cultures were used to isolate and propagate 14 isolates of AM fungi indigenous to red soil. The effectiveness of each fungus in promotion of growth of mungbean was evaluated in red soil. For comparison, three isolates from northern China, known to be highly effective in neutral soils, and two isolates from Australia, known to be from acidic soil were used. Effectiveness was positively related to root infection (r
2 = 0.601). For two of these isolates, Glomus caledonium (isolated from northern China) and Glomus manihotis (an isolate indigenous to red soil), the applied P concentration giving the highest infection and response to infection was approximately 17.5 mg P kg–1 soil. In field experiments in which this concentration of P was applied, the five most effective isolates were tested on mungbean. The Glomus caledonium isolate from northern China was the most effective, followed by the indigenous Glomus manihotis isolate. The Glomus caledonium isolate was also shown to be effective on Lespedeza formosa, which is commonly used in revegetation efforts. We conclude that inoculation of plants with selected isolates of AM fungi may aid in revegetation efforts on eroded red soils in subtropical China. 相似文献

Although members of the Oxalidaceae family have been described as host plants of vesicular-arbuscular mycorrhizal fungi, Oxalis pes-caprae did not become colonized by Glomus mosseae. Extracts of Ox. pes-caprae root inhibited the germination of G. mosseae spores. Analysis of the community of arbuscular mycorrhizal (AM) fungi in roots of Fragaria vesca growing in a heavy metal contaminated site was carried out on a Zn waste site near Chrzanow (southern Poland). The waste
substratum was characterized by high contents of Pb, Zn, Cd, Cu and As, and by low levels of N, P and organic matter. Spores
of Glomales were isolated by wet sieving and DNA was isolated from individual spores. Nested polymerase chain reaction (PCR)
with taxon-specific primers was used to identify the species Glomus mosseae, Glomus intraradices and Glomus claroideum. Spores of other fungi were morphologically characterized and new taxon-discriminating molecular probes were developed for
two of them (Glomus sp. HM-CL4 and HM-CL5) based on variations in the large ribosomal subunit (25S rDNA). High sequence similarities were found
between Glomus sp. HM-CL4 and Glomus gerdemanii, and between Glomus sp. HM-CL5 and Glomus occultum. The designed primers were used to characterize the population of AM fungi colonizing the roots of F. vesca collected from the Zn waste site. The analysis, carried out on roots stained with trypan blue, showed that the most effective
colonizer was closely related to G. gerdemannii. G. claroideum and the G. occultum-like fungus were slightly less common whilst frequencies of G. intraradices and G. mosseae in roots were much lower. The analysis of mycorrhiza stained with rhodizoniate to localize heavy metal accumulation showed
that the stain does not influence the PCR reaction. Seventy percent of the root samples containing positively stained fungal
hyphae were found to be colonized by G. mosseae. The data obtained demonstrate the usefulness of nested PCR for studies carried out in polluted areas. It will enable selection
of AM fungi which are able to colonize plant roots under heavy metal stress conditions, as well as the identification of fungi
showing high in situ accumulation of potentially toxic elements.

Summary The influence of vesicular-arbuscular mycorrhizal symbiosis on cytokinin activity in Citrus jambhiri Lush, seedlings was investigated. C. jambhiri inoculated with cultures of Glomus caledonium (Nicol. and Gerd.), G. epigaeum (Dan. and Trappe), G. etunicatum (Becker and Gerd.), G. fasciculatum Thaxt. (Gerd, and Trappe) or G. mosseae (Nicol and Gerd.) was grown from seed for 105 days in a glasshouse. Cytokinin activity in roots and leaves of seedlings was analyzed using high-performance liquid chromatography, mass spectrometry and a bioassay. Seedling leaf tissue had greater cytokinin activity than root tissue. Zeatin, zeatin riboside, and their dihydro- and glucoside derivatives were isolated from leaves of 105-day-old seedlings inoculated with G. fasciculatum and G. mosseae. Cytokinin activity in roots and leaves was associated with differences in seedling total dry weight and vesicular-arbuscular mycorrhizal colonization. The ribose moiety and the saturated side chain apparently influence cytokinin transport and physiological activity in Citrus seedlings. 相似文献

Cabbage (Brassica oleracea, var. capitata, cv. Hercules) seedlings were inoculated with vesicular-arbuscular mycorrhizal (VAM) fungi Glomus fasciculatum, G. aggregatum, and G. mosseae. Differential efficiency in mycorrhizal colonization and the specificity of fungal symbiont to stimulate the growth and nutrient uptake of the host were observed. In addition, there was an increase in phenol, protein, reducing sugar contents, and peroxidase activity in the VAM inoculated seedlings. Diversity and infectivity of arbuscular mycorrhizal fungi in agricultural soils of the Sichuan Province of mainland China 总被引:1,自引:0,他引:1
Knowledge about the presence and diversity of arbuscular mycorrhizal fungi (AMF) in a specific area is an essential first
step for utilizing these fungi in any application. The community composition of AMF in intensively managed agricultural soil
in the Sichuan Province of southwest China currently is unknown. In one set of samples, AMF were trapped in pot cultures from
40 fields growing legumes in the Panxi region, southeast Sichuan. In a second set of samples, the MPN method with four-fold
dilutions and maize as host was used to estimate infective propagules in soil from another 50 agricultural sites throughout
the province. Soil types were heterogeneous and were classified as purple, yellow, paddy and red. Crops at each site were
either maize, wheat or sweet orange. From this set of soil, AMF spores were also extracted and identified. Including all ninety
soils, thirty glomeromycotan species in Glomus (20 species), Acaulospora (four species), Scutellospora (three species), Ambispora (one species), Archaeospora (one species) and Paraglomus (one species) were identified. Yellow, red and purple soils yielded similar numbers of AMF species, while AMF species diversity
was clearly lower in paddy soil. In trap culture soils, the most frequent species were Glomus aggregatum or Glomus intraradices, Glomus claroideum and Glomus etunicatum. The species Acaulospora capsicula, Acaulospora delicata, G. aggregatum (or intraradices), G. claroideum, Glomus epigaeum, G. etunicatum, Glomus luteum, Glomus monosporum, Glomus mosseae and Glomus proliferum were successfully cultured as single-species pot cultures in Plantago lanceolata. The three most frequent species in field soils were G. mosseae, Glomus caledonium and Glomus constrictum. MPN values varied between 17 and 3334 propagules 100 g soil−1 among the fifty field sites sampled. Regression analysis, including host&soil, log(P) and pH as explanatory variables explained
59% of the variation in log(MPN). The highest MPN estimates were found in purple soil cropped with maize and citrus, 324 and
278 propagules 100 g soil−1, respectively. The lowest MPN value, 54 propagules 100 g soil−1, was measured in wheat in purple and yellow soil. Despite intensive agricultural management that can include often repeated
tillage, our examination of 90 agricultural sites revealed that soils of the Sichuan region have moderate to high numbers
of infective AMF propagules as well as a high AMF species diversity. This opens possibilities for further studies and utilization
of AMF in agriculture and horticulture in the Sichuan province, People’s Republic of China. 相似文献

We studied the role of modification in root exudation induced by colonization with Glomus intraradices and Glomus mosseae in the growth of Phytophthora nicotianae in tomato roots. Plants were grown in a compartmentalized plant growth system and were either inoculated with the AM fungi
or received exudates from mycorrhizal plants, with the corresponding controls. Three weeks after planting, the plants were
inoculated or not with P. nicotianae growing from an adjacent compartment. At harvest, P. nicotianae biomass was significantly reduced in roots colonized with G. intraradices or G. mosseae in comparison to non-colonized roots. Conversely, pathogen biomass was similar in non-colonized roots supplied with exudates
collected from mycorrhizal or non-mycorrhizal roots, or with water. We cannot rule out that a mycorrhiza-mediated modification
in root exudation may take place, but our results did not support that a change in pathogen chemotactic responses to host
root exudates may be involved in the inhibition of P. nicotianae. 相似文献
