Low temperature 31P and 15N NMR spectroscopy was used to investigate the species forming in the organic layer following the extraction of uranium from nitric acid solutions with the synergistic mixture, TBP-DEHPA. It was found that the complex formed during extraction was a mixed nitrato complex UO2(NO3)A·TBP. Although the existence of the complex UO2A2·χTBP may be demonstrated in model systems, the complex does not form under actual extraction conditions.  相似文献

为了探究铀对藻类生长及光合作用的影响,筛选新的基于光合作用的水体铀污染生态风险评价指标,本试验采用不同浓度铀(0、0.5、1、5、10、20mg U·L-1)分别处理普通小球藻(Cholorella vulgaris)和黄龙普通小球藻两种来自不同生境的微藻,在处理后的第3、5、7、10、14d进行相对生长速率、光合放氧速率、叶绿素含量和叶绿素荧光动力学参数等指标的测定。结果表明:(1)0.5mg·L-1低浓度铀处理显著促进两种小球藻的生长和光合作用效率,表现为两种微藻的相对生长速率、光合放氧速率、光系统II最大光化学量子产量Fv/Fm、实际光化学量子产量Y(Ⅱ)、相对电子传递速率rETR等叶绿素荧光参数等指标均显著高于对照, 而5-20 mg·L-1高浓度铀处理则显著抑制两种小球藻的生长和光合作用;(2)黄龙普通小球藻比普通小球藻对铀处理更敏感,在1mg·L-1处理浓度下生长与光合作用就受到显著抑制,可以用来作为水体铀污染生物监测的指示生物;(3)回归分析表明,不同浓度铀处理下,叶绿素荧光参数Y(II)和rETR的响应速度快于相对生长速率、光合放氧速率、叶绿素含量和Fv/Fm等指标的变化,可以作为水体铀污染生态风险评价的敏感指标。  相似文献

The uranium uptake ability of silk fibroin was investigated. High ability to uptake uranium from nonsaline water containing 2.500 mg of uranium was observed with the silk fibroin tested. The uranium uptake was very rapid and was dependent on pH, uranium concentration, temperature, and retention time. Almost all uranium taken up is easily eluted with 1 mol/L CH3COONH4. This biomatrix, therefore appears to have potential for use in a commercial process for uranium recovery from uranium-containing waste water.  相似文献

Trametes versicolor and Agaricus augustus, with a maximum tolerable concentration (MTC) of 80 μg ml−1 tribromophenol (TBP), were selected to evaluate TBP biodegradation capacity. These fungi were capable of decreased TBP concentrations and A. augustus was also capable of biotransforming TBP to tribromoanisole (TBA). Peroxidase and laccase activities were observed in the T. versicolor supernatant but not in that of A. augustus. These tolerance levels could be due to either lignolytic enzymes that degrade TBP or the ability of the fungi to biotransform TBP to tribromoanisole, respectively. The sustained ability of T. versicolor to degrade TBP (total of 40 μg ml−1) in the presence of an additional carbon source suggests that it may have potential applications in the degradation of forestry industry waste.  相似文献

A procedure was developed for real-time measurement of the interaction between an archaeal TATA-binding protein (TBP) with stress-gene promoters from the archaeon Methanosarcina mazeii using surface plasmon resonance (SPR), the BIACORE 3000 equipment, and the SA (streptavidin) Sensor Chip. Measurements were based on the SPR optical phenomenon, which resulted in light extinction when TBP bound a promoter. This process, detected as a change in a particular angle, was recorded in a sensorgram. The BIA-evaluation program allowed the calculation of the equilibrium constant (K(A)) of the interaction of M. mazeii TBP with the promoters of the stress genes grpE, hsp70(dnaK), and hsp40(dnaJ) (0.47, 0.26, and 1.21x10(7)M(-1), respectively) and, for comparison, with the promoter of a non-heat-shock gene, orf16 (0.08x10(7)M(-1)). The association rate (k(a)) of the non-heat-shock gene orf16 was 0.4x10(4)M(-1)s(-1) and those for the stress genes, grpE, hsp70(dnaK), and hsp40(dnaJ) were higher: 2.8, 1.5, and 3.5x10(4)M(-1)s(-1), respectively. The new procedure will allow a comparative analysis of different TPBs and promoters (wild type and mutants) under physiologic and stress conditions, and a correlation of TBP binding parameters with constitutive and stress-induced gene expression.  相似文献

Theileria annulata is a tick-borne intracellular protozoan parasite that causes tropical theileriosis, a fatal bovine lymphoproliferative disease. The parasite predominantly invades bovine B lymphocytes and macrophages and induces host cell transformation by a mechanism that is not fully comprehended. Analysis of signaling pathways by quantitative real-time PCR (qPCR) could be a highly efficient means to understand this transformation mechanism. However, accurate analysis of qPCR data relies on selection of appropriate reference genes for normalization, yet few papers on T. annulata contain evidence of reference gene validation. We therefore used the geNorm and NormFinder programs to evaluate the stability of 5 candidate reference genes; 18S rRNA, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), ACTB (β-actin), PRKG1 (protein kinase cGMP-dependent, type I) and TATA box binding protein (TBP). The results showed that 18S rRNA was the reference gene most stably expressed in bovine PBMCs transformed and non-transformed with T. annulata, followed by GAPDH and TBP. While 18S rRNA and GAPDH were the best combination, these 2 genes were chosen as references to study signaling pathways involved in the transformation mechanism of T. annulata.  相似文献

海金沙草总黄酮提取工艺的响应面优化   总被引:2,自引:1,他引:1  
为了提高海金沙草总黄酮提取效率,运用了Plackett-Burman试验设计、爬陡坡试验和Box-Behnken设计对提取工艺进行了响应面优化试验。利用Plackett-Burman对影响总黄酮提取的诸多相关因素进行了评价并成功筛选出主效应因子,即提取时间、提取温度和乙醇浓度。在Plackett-Burman设计基础上,根据主效应因子作用大小与方向进行了爬陡坡试验。最后用Box-Behnken响应面技术优化了总黄酮提取工艺并建立了关键影响产量的二次多项式数学模型,解模型逆矩阵得最优解(优化方案),即提取温度X1=45.45℃,乙醇体积分数X2=47.1%、提取时间X3=84.8 min。模型预测结果为0.433 mg.L-1,验证试验结果为0.428±0.004 mg.L-1(n=3)。  相似文献

Bioleaching of uranium was carried out with Turamdih ore sample procured from Uranium Corporation of India Limited, Jaduguda. The bacterial strain that was used in the leaching experiments was isolated from the Jaduguda mine water sample. Efficiency of bioleaching was studied by varying parameters like pulp density and initial ferrous concentration as source of energy. It is observed that the efficiency of bioleaching was 49% at 10% pulp density (w/v) and initial pH 2.0. Addition of external has no effect on efficiency of bioleaching showing domination of direct leaching mechanism over indirect.  相似文献

为获得聚-β-羟基丁酸(PHB)提取的最佳工艺,以氯仿浓度,提取时间,和提取温度为影响因子,在单因素实验结果的基础上,应用Box-Benhnken中心组合方法进行三因素三水平的实验设计,以PHB提取量为响应值,运用响应面法(RSM)对提取条件进行进一步的优化。结果表明:在氯仿浓度69.7%,提取温度59.7℃,提取时间3.0 h的条件下,PHB提取量达到最大值4.430 g/L。  相似文献

Summary A process for concentrating uranium from contaminated soils in which the uranium is first extracted with bicarbonate and then the extracted uranium is precipitated with U(VI)-reducing microorganisms was evaluated for a variety of uranuum-contaminated soils. Bicarbonate (100 mM) extracted 20–94% of the uranium that was extracted with nitric acid. The U(VI)-reducing microorganism,Desulfovibrio desulfuricans reduced the U(VI) to U(IV) in the bicarbonate extracts. In some instances unidentified dissolved extracted components, presumably organics, gave the extract a yellow color and inhibited U(VI) reduction and/or the precipitation of U(IV). Removal of the dissolved yellow material with the addition of hydrogen peroxide alleviated this inhibition. These results demonstrate that bicarbonate extraction of uranium from soil followed by microbial U(VI) reduction might be an effective mechanism for concentrating uranium from some contaminated soils.  相似文献

A technique based on physical maceration of root tissue was developed to extract vermiform and swollen stages of Meloidogyne incognita and Rotylenchulus reniformis. Experiments conducted on soybean and tomato evaluated the efficiency of method (stir, grind), NaOC1 concentration (0%, 0.5%), and duration (lx, 2x) on extraction of nematodes and eggs from 60-day-old populations. Root-associated populations of R. reniformis were considerably lower than those of M. incognita, so development of the method focused on the latter. Grinding liberated more nematodes than stirring, but the reverse was true for egg extraction. Among grinding treatments, a duration of 10 seconds in 0.5% NaOCl provided the most efficient extraction of nematodes and eggs. Among stirring treatments, a duration of 10 minutes in 0.5% NaOCl provided the most efficient extraction of eggs. These techniques were compared on soybean roots 30 days older than those on which the procedures were first evaluated, with consistent results.  相似文献

干花豆( Fordia cauliflora)的主要有效成分为黄酮类、生物碱、有机酸等,具有益智、抗衰老、抗炎等作用。目前的研究主要集中在化学成分及药理活性等方面,对总黄酮成分提取工艺优化报道较少。该研究以新鲜干花豆为材料,以总黄酮提取量为评价指标,在提取温度、料液比、乙醇浓度和提取时间单因素实验基础上,采用响应面法优化了干花豆总黄酮提取工艺,同时测定了总黄酮对1,1-二苯基苦基苯肼自由基( DPPH.)和羟基自由基(.OH)清除能力。结果表明:干花豆总黄酮提取最佳工艺条件为提取温度78℃、料液比为1∶30(g.mL-1)、乙醇浓度71%和提取时间为187 min。在此条件下,总黄酮得率预测值为10.61 mg.g-1,实际为10.53 mg.g-1,理论值与预测值的相对误差为0.76%;干花豆总黄酮对DPPH和OH自由基清除能力IC50值分别为14.09和78.43μg.mL-1,弱于Vc(8.11和67.95μg.mL-1)。该提取工艺稳定合理,准确可靠,是提取干花豆总黄酮的可行方法。该研究结果为干花豆中总黄酮成分的进一步开发利用奠定了基础。  相似文献
