The changes of hydroxyproline (Hyp) content in cell wall after inoculation with Corynebacterium michiganense pv. insidiosum or Verticillium albo-atrum in 8 cultivars of alfalfa (Medicago sativa) were studied. In our experiments cultivars ‘Trek’ and ‘Vernal’– resistant, ‘Du Puits’ and ‘Vertus’– susceptible to C. michiganense;‘Vela’ and ‘Maris Kabul’– resistant to V. albo-atrum, ‘Sabilt’ and ‘Lahontan’– susceptible to V. albo-atrum were used. In resistant plants inoculated with C. michiganense none or a small increase of Hyp content in comparison with control plants was registered. On the other hand, inoculated susceptible plants showed markedly increase of Hyp content in comparison with the control plants. The differences in Hyp content between control and inoculated resistant and susceptible plants with V. albo-atrum were not markedly expressed in comparison with the alfalfa –C. michiganense pair. The changes had similar tendency both in C. michiganense and V. albo-atrum.alfalfa pairs.  相似文献

Comparative studies were made of the responses of resistant and of susceptible Gem tomato plants to infection by Verticillium albo-atrum. When inoculated through roots, there were striking differences in their responses. In susceptible plants, the foliar symptoms and amount of mycelium in the stem increased rapidly for some time. Then the mycelium started to disappear from the stem; this was accompanied by a check to the normal progress of symptoms, and by the formation of tyloses. In resistant plants, a limited invasion of the root and lower stem was accompanied by rapid and extensive tylosis. The mycelium soon disappeared from the stem and the plant then recovered from the initially mild symptoms. There was an inverse relationship between the amount of mycelium and the extent of tylosis in infected plants. The growth of susceptible plants was markedly reduced by infection. Total leaf area was much less because the newly produced leaves did not expand normally. The root system in infected plants was smaller because there were many fewer tertiary roots. In resistant plants infection stimulated growth. Tomato cuttings inoculated with conidia reacted similarly to root-inoculated plants. Hyphae grew well in the vascular system of susceptible cuttings whereas in resistant cuttings the pathogen started to grow but soon disappeared. Detached leaves of susceptible plants, inoculated through cut ends, wilted more than did leaves from resistant plants. It is suggested that resistance is mainly of the active type that develops after infection.  相似文献

The response of seedling roots of near-isogenic tomato varieties to infection by Verticillium albo-atrum or Fusarium oxysporum f. sp. lycopersici was investigated. Studies of the infection of seedling roots not artificially damaged indicated that there was an extra-vascular expression of resistance towards V. albo-atrum but not to F. oxysporum. Roots of resistant tomato seedlings infected by V. albo-atrum contained the fungus in the epidermis and outer cortex while susceptible roots became heavily colonised. Observations made by transmission electron microscopy showed that the fungus appeared to be abnormal in growth and appearance in the epidermal and cortical cells of resistant seedling roots but normal in susceptible roots. Two preformed antifungal terpenoids were detected in seedling roots in greater amounts in resistant that in susceptible varieties. The possible mechanisms of seedling root resistance to vascular wilts are discussed.  相似文献

In the present study, corn meal agar was found as the best medium for growth of Drechslera spicifera, a pathogen of water hyacinth. Small water hyacinth plants exhibited a smaller number of lesions/leaf when inoculated with D. spicifera compared with inoculated medium and large plants. However, percentage total diseased area/leaf was almost the same among the different plant sizes. The pathogen did not interfere with the vegetative plant growth, since, both infected and healthy water hyacinth plants of the same age showed similar percentages of new leaf growth. Leaf diffusates of water hyacinth plants significantly reduced conidiospore germination in D. spicifera.  相似文献

The Hyp content was studied in cell wall of alfalfa susceptible and resistant strains on the 3rd, the 7th and on the 14th day after inoculation with Verticillium albo-atrum or Corynebacterium michiganense pv. insidiosum. The changes of Hyp content after inoculation with both pathogens were markedly expressed in alfalfa roots. Resistant plants of R 337 strain responded to inoculation with V. albo-atrum or C. michiganense pv. insidiosum by the decrease of Hyp content mainly on the 3rd and on the 7th day. On the 14th day after inoculation Hyp content practically did not differ from that of the control. Susceptible plants of S 354 and S 321 srains responded to inoculation with wilt pathogens by the slight decrease of Hyp content at the 3rd day after inoculation. A significant increase of Hyp content was found on the 7th and mainly on the 14th day after inoculation in comparison with control plants. The cell wall Hyp content was also determined with 7 R-strains and 7 S-strains at 120 days after inoculation with both pathogens. In each R and S strain two categories of plants were used for chemical analyses: Wilt-free plants (0 to 1 classes) and diseased, wilted plants (2 to 6 classes). In the resistant alfalfa strains no differences in Hyp content between the wilt-free and diseased plants were found. In the susceptible alfalfa strains the Hyp content was significantly higher in roots of diseased plants comparing with the wilt-free ones. Only negligible changes in Hyp content were registered in the overground parts of all inoculated alfalfa strains.  相似文献

Callus cultures derived from leaf segments of chrysanthemum cultivar ‘Snow Ball’ which was susceptible to Septoria obesa were successfully used for in vitro selection for resistance to this pathogenic fungus. Resistant cell lines were selected by culturing callus on growth medium containing various concentrations of S. obesa filtrate. Resistant calluses obtained after two cycles (30 d each cycle) of selection were used for plant regeneration. About 30% of the plants regenerated from the resistant calluses and 70–80% of the plants raised from cuttings had acquired considerable resistance against the pathogen in the field. No phenotypic variation was observed in the selected regenerates.  相似文献

Somaclonal variation in disease reaction type to infection by the vascular wilt pathogen Verticillium albo-atrum Reinke & Berth was assessed in a population of lucerne plants regenerated from callus lines obtained from a susceptible cultivar. Disease severity in the regenerant population was reduced by comparison with parental controls. Seed progeny and plants recovered via a second tissue culture cycle reverted to mainly susceptible reaction types. In a further experiment a low molecular weight toxic fraction from culture filtrates of the fungus was incorporated into the callus medium prior to regeneration. Toxin treatment reduced the regenerative capacity of callus, and there was little evidence for a higher frequency of wilt resistant plants in populations selected at low toxin concentrations. The results suggest that somaclonal variation as an alternative breeding strategy for disease resistance in lucerne offers no advantages over conventional recurrent selection.  相似文献

Although the isolation of Verticillium albo-atrum and V. dahliae from soil and dried moribund stems following infection of a potato crop proved extremely difficult, both fungi were equally capable of overwintering in these substrates and of inducing disease in a subsequently planted susceptible crop. In the absence of a susceptible crop some weed species became colonized. The two species, however, appeared to differ in their capacity for survival both beneath a monocotyledonous crop and within the potato tubers. Colonization of the roots of wheat, barley, oats, rye and maize was observed with V. dahliae but not with V. albo-atrum. The latter appeared to be capable of prolonged survival in the tubers, whereas V. dahliae did not remain viable in storage over winter. Consequently only tubers infected with V. albo-atrum produced infected plants. The presence of the fungi within the tubers affected neither dormancy nor the initial development of the sprouts. Some correlation was noted between tuber size, the percentage of tubers infected, the distribution of V. albo-atrum within the tubers and the development of disease in plants subsequently grown from these tubers.  相似文献

The germination of conidia of Peronospora farinosa f. sp. betae, collected from sugar beet and suspended in deionized water, was inhibited by dilution with 10% solutions of glycerol, glucose or sucrose and with sap from sugar-beet leaves. Germination was stimulated by diluting with deionized water but not with tap water or biological saline. Substances that diffused from excised buds of sugar-beet plants into deionized water also stimulated germination of conidia but diffusates from leaves did not. This may partly explain why buds are more susceptible to downy mildew than leaves in sugar beet. Germination of conidia was apparently stimulated more by diffusates from buds of seedlings than by those from buds of older plants; this may help to explain why sugar-beet seedlings are more susceptible to downy mildew than older plants. Diffusates from plants of four sugar-beet stocks, that differed from each other in susceptibility to downy mildew, had very similar effects on germination of P. farinosa conidia. Stimulation of spore germination on the surfaces of buds and leaves did not seem, therefore, to be an important factor in determining resistance or susceptibility to downy mildew in these stocks.  相似文献

As the cowpea rust fungus penetrates the wall of a cowpea epidermal cell, resistant and susceptible plants exhibit different ultrastructural and cytochemical changes within the epidermal protoplast. To examine plant gene expression at this stage of infection, cytoplasm was extracted from individual inoculated or uninoculated epidermal cells before the fungal penetration peg reached the cell lumen. Initial differential colony hybridization screening of an expressed sequence tag library constructed from globally amplified cDNAs generated from the inoculated resistant cells resulted in 80 clones (out of 835) with a differential hybridization pattern. Further slot-blot screening and screening of the amplified cDNAs generated from inoculated or uninoculated, resistant or susceptible cells revealed 28 separate genes, mostly with no matching sequences in the databases, that were up-regulated in response to the growth of the fungus through the wall of resistant or susceptible cells. Five genes, including those coding for beta- and alpha-tubulin, were found to be down-regulated specifically in inoculated, susceptible cells, and five were specifically up-regulated in inoculated, resistant cells, including a PR-10 homolog and a phenylalanine ammonia-lyase gene. Probing the amplified cDNAs from each cell type for the expression of cell death-related genes revealed that an LLS1 homolog (vuLLS1), cloned from cowpea, was up-regulated by infection in both resistant and susceptible cells and that a homolog of HSR203J was differentially up-regulated in resistant cells. These data show that changes in gene expression predicting the subsequent expression of susceptibility or hypersensitive resistance to fungal infection occur prior to the fungus entering the cell lumen.  相似文献

Verticillium dahliae but not V. albo-atrum Berth was isolated from eight out of twenty-one stone fruit orchards surveyed for Verticillium wilt disease in western New York. Wilt incidence was related to the cultivation of tomato or legumes as previous or inter-crop with stone fruit trees. A limited cross species inoculation using isolates of V. albo-atrum and V. dahliae from woody and herbaceous plants showed that peach and cherry were susceptible to both species. The effect of V. dahliae on growth of cherry seedlings in the presence of Tylenchorynchus claytoni, Pratylenchus penetrans and Meloidogyne hapla was compared. P. penetrans and M. hapla produced more severe growth reduction than T. claytoni. The adverse effect of Verticillium on the growth of cherry seedlings was greater acting together with any one of the three nematodes than acting alone. V. dahliae was shown to be capable of passage through earthworms without loss of infectivity.  相似文献

Verticillium albo-atrum was apparently eradicated from soil in which dicotyledonous plants were suppressed. In one field experiment V. albo-atrum was not recovered after 4 years of weed-free grass cover; in the same period soil infectivity reduced to very low levels under two bare fallow treatments. Where weeds developed after single annual cultivations soil infectivity declined from the initially high level but finally increased. In three observation trials in which hop plants were used to detect V. albo-atrum after grass-cover treatments, soil infectivity was very low after 2 years and apparently nil after 3–5 years. Grass cover effectively stabilizes infested soil; it may be used for limiting spread from primary disease outbreaks, and also as a short-term treatment to reduce the infectivity of soil to be re-planted with resistant varieties. The frequent incidence of V. dahliae hop infections in one trial indicated that this fungus was not amenable to control by grass cover treatment and hence may be ecologically distinct from V. albo-atrum.  相似文献

 The effect of different genotypes of the ectomycorrhizal fungus Hebeloma cylindrosporum on in vitro rooting of micropropagated cuttings of Prunus avium and P. cerasus was studied in an attempt to determine whether ectomycorrhizal fungi could enhance in vitro adventitious root formation in plants which form arbuscular endomycorrhizas. The rooting percentage of P. avium cuttings was approximately 16% in the absence of hormonal treatment; it increased up to 30% in the presence of 5.7 μM IAA which was the most favourable auxin concentration. The rooting percentage of cuttings cultivated in the absence of IAA was enhanced by all the studied strains of H. cylindrosporum. It ranged from 50 to 60% with the IAA-overproducing mutant D 111 or the wild-type dikaryon D1, to 100% in the presence of the mutants 331 or D 117. The cuttings of P. cerasus showed a higher rooting ability than those of P. avium since approximately 40% of them were able to root in the absence of hormonal treatment. Except for the mutant D117, their rooting percentage was not significantly improved by H. cylindrosporum. Fungal inoculation also affected the survival of cuttings at acclimatization: 50% of the uninoculated P. avium cuttings survived whereas the survival percentage of inoculated cuttings ranged from 30 to 100% depending on the fungal genotype. With P. cerasus, the percentage of survival of uninoculated cuttings ranged from 85 to 100% and fungi either did not significantly improve it or lowered it. At acclimatization fungal hyphae could be observed in close contact with adventitious roots, but they did not establish mycorrhizal association. The shoot height of P. avium plantlets obtained from inoculated cuttings was not significantly different from that of plantlets originating from uninoculated ones. By contrast, fungal inoculation generally depressed the growth of acclimatized P. cerasus plantlets. The possibility of using ectomycorrhizal fungi as a tool to enhance rooting of micropropagated cuttings of plants which do not form ectomycorrhizas is discussed. Meloidogyne hapla-resistant plants grown from cuttings and inoculated with M. hapla larvae were free of galls. However, 35 to 48% of the seedling intercross progeny of resistant genotypes that were inoculated in the germinated seed stage were galled. There was an inverse relationship between the age of plants grown from seed and the percentage of plants galled by M. hapla; the older the plants at inoculation, the greater the percentage of gall-free plants. The per cent of galled plants was significantly reduced when galled roots were removed and plants reinoculated. Reproduction of M. hapla on galled progeny of resistant plants was significantly less than that on susceptible plants. There were no differences in nematode reproduction on galled progeny of resistant plants, regardless of age at time of inoculation. An in,ease in inoculum levels from 100 to 10,000 M. hapla larvae did not affect resistance or susceptility. There was a direct correlation between galling of inoculated seedlings of resistant progeny and temperature; inoculated 8-week-old cuttings of resistant plants were galled only at 32 C.  相似文献

Leaf, stem, and root extracts of near-isogenic tomato plantscv. Craigella, resistant and susceptible to Verticillium albo-atrum,showed constitutive 1,3-ß-glucanase activity whichincreased following inoculation with the pathogen. Partiallypurified enzyme extracts were obtained by dialysing a 30–80%ammonium sulphate fraction of the tissue brei. The enzyme hadpH and temperature optima of 5?5 and 44 ?C respectively, withhigh activity between 50 and 60 ?C. The response to laminarinconcentration was linear between 1?2 and 7?5 mg ml–1.Root inoculation of susceptible plants with 106 propagules ml–1V. albo-atrum led to a umform 300 per cent increase in all steminternodes except the terminal one, which was 500 per cent ofthe controls. No spatial relationship of enzyme activity tothe localization of fungus within the stem was apparent. Petioles,leaves, and roots of susceptible infected plants similarly showedan increase in activity but less than that in stems. Changedlevels of stern enzyme activity at different times after inoculationwere associated with reductions in the number of vessels containinghyphae. Extracts of plants of the resistant isoline showed increasedglucanase activity over controls, but this was substantiallylower than that in susceptible plants and was associated withthe greatly reduced mycelial colonization in resistant plants. It is concluded that single gene resistance in tomato to Verticilliumis not associated with innately higher levels of 1,3-ß-glucanasein healthy plants. The increased activity in infected plantsis proportional to the overall quantity of pathogen in the plantor of pathogenic metabolites.  相似文献

Systemic acquired resistance (SAR) can be induced in plants by incompatible pathogens, pathogen derived extracts, or certain chemicals as benzothiadiazole (BTH). The aim of this work was to compare changes in peroxidase and chitinase activities, enzymes considered as PR-proteins, caused by BTH and the pathogen Plasmopara halstedii. Hypocotyls from susceptible and resistant BTH-treated sunflower seedlings showed increased peroxidase and chitinase activities. Inoculation with P. halstedii increased chitinase and peroxidase activities in inoculated hypocotyls from susceptible but not from resistant sunflower seedlings.  相似文献

In glasshouse tests, sap from plants infected with 15 different isolates of tomato spotted wilt tospovirus (TSWV) from three Australian states was inoculated to nine genotypes of tomato carrying TSWV resistance gene Sw-5 or one of its alleles. A further two resistant tomato genotypes were inoculated with four isolates each. The normal response in resistant genotypes was development of necrotic local lesions in inoculated leaves without systemic invasion, but 22/752 plants also developed systemic reactions in addition to local hypersensitive ones. Using cultures from two of these systemically infected plants and following four cycles of subculture in TSWV resistant tomato plants, two isolates were obtained that gave susceptible type systemic reactions but no necrotic spots in inoculated leaves of resistant tomatoes. When these two isolates, DaWA-1d and ToTAS-1d, were maintained by repeated subculture for 10 successive cycles in Nicotiana glutinosa or a susceptible tomato genotype, they still induced susceptible type systemic reactions when inoculated to resistant tomato plants. They were therefore stable resistance breaking isolates as regards overcoming gene Sw-5. When resistance-breaking isolate DaWA-1ld multiplied together with original isolate DaWA-l in susceptible tomato, it was fully competitive with the original isolate. However, when DaWA-ld and ToTAS-ld were inoculated to TSWV resistant Lycopersicon peruvianum lines PI 128660R and PI 128660S and to TSWV resistant Capsicum chinense lines PI 152225, PI 159236 and AVRDC CO0943, they failed to overcome the resistance, producing only necrotic local lesions without systemic infection. Thus, although the ease of selection, stability and competitive ability of resistance breaking isolates of TSWV is cause for concern, L. peruvianum and C. chinense lines are available which are effective against them. The effectiveness of the resistance to TSWV in nine tomato genotypes was examined in a field experiment. Spread was substantial in the susceptible control genotype infecting 42% of plants. Resistance was ineffective in cv. Bronze Rebel, 26% of plants developing infection. In contrast, it held up well in the other eight resistant genotypes with only 1–3 or no plants of each becoming infected. Accumulated numbers of Thrips tabaci, Frankliniella occidentalis and F. schultzei were closely correlated with TSWV spread.  相似文献

Poplar cuttings of a resistant clone, Populus ‘Grandis’, and susceptible clones, Populus nigra ‘Italica’ and Populus ‘Robusta’, were infected with the pathogenic fungus Dothichiza populea alone, or with the pathogen and one of five strains of epiphytes antagonistic towards it (in vitro), isolated from poplar bark. The extent of injury was examined for 28 days after infection by determining the length of necrotic patches and their area as expressed in per cent of the total area of a cutting or the area of necrotic injuries caused by the pathogen alone.All the poplar cuttings of both the resistant and susceptible clones became diseased when infected with the pathogen alone. Surprisingly enough, however, the least affected clone was the susceptible P. ‘Robusta’, in which necrotic injuries covered 28% of the total area, as against 40% and 70% in the resistant P. ‘Grandis’ and the susceptible P. nigra ‘Italica’, respectively.When the cuttings were infected simultaneously with Dothichiza populea and its antagonistic epiphytes, the diseased area in the resistant clone diminished by as much as two-thirds, and in the susceptible P. nigra ‘Italica’, by one-third in comparison with the area affected by the pathogen alone. In turn, in the susceptible P. ‘Robusta’ the introduction of three out of five epiphytes stimulated the growth of the pathogenic fungus producing on average a double increase in the necrotic area. The differences in the response of the pathogen to the presence of epiphytes recorded in the susceptible clones indicate a marked influence of the plant on the nature of interactions between its epiphytic microflora and the pathogen.  相似文献

Alfalfa plants were regenerated from callus cultures of three source plants that differed in resistance to anthracnose, caused by Colletotrichum trifolii. All regenerant plants were evaluated for variation in resistance to disease caused by races 1 and 2 of the pathogen. Of eighty-two plants that were regenerated and evaluated, no plants responded differently to inoculation with race 1 of C. trifolii, but two plants (2.4%) differed in resistance when inoculated with race 2. The source plant of these regenerants was resistant to races 1 and 2 of the pathogen but the regenerants were resistant to race 1 and susceptible to race 2. No variants to race 1 were detected. The susceptible response of the variant plants to race 2 was confirmed by cytological analysis and was consistent with the response of nonregenerant susceptible plants. These plants represent a near-isogenic plant model for studying the molecular biology of resistance and susceptibility to anthracnose of alfalfa.  相似文献

In glasshouse tests, infective sap from plants infected with 17 different isolates of Tomato spotted wilt virus (TSWV) from four Australian states was inoculated to three Capsicum chinense accessions (PI 152225, PI 159236 and C00943) carrying single genes that confer hypersensitive resistance to TSWV. The normal response to inoculation was development of necrotic (hypersensitive) local lesions in inoculated leaves without systemic invasion, but 3/1386 infected plants also developed systemic susceptible reactions in addition to hypersensitive ones. Similarly when two isolates were inoculated to C. chinense backcross progeny plants, 1/72 developed systemic susceptible reactions in addition to localised hypersensitive ones. Using cultures from the four plants with susceptible reactions and following three to five further cycles of serial subculture in TSWV‐resistant C. chinense plants, four isolates were obtained that gave systemic susceptible type reactions in the three TSWV‐resistant accessions, and in TSWV‐resistant cultivated pepper (C. annuum). When three of these isolates were inoculated to tomato (Lycopersicon esculentum) breeding lines with single gene resistance to TSWV, resistance was not overcome. Similarly, none of the four isolates overcame partial resistance to TSWV in Lactuca virosa. When TSWV isolates were inoculated to tomato breeding lines carrying partial resistance from L. chilense, systemic infection developed which was sometimes followed by ‘recovery’. After four successive cycles of serial passage in susceptible cultivated pepper of a mixed culture of a resistance‐breaking isolate with the non resistance‐breaking isolate from which it came, the resistance‐breaking isolate remained competitive as both were still found. However, when the same resistance‐ breaking isolate was cultured alone, evidence of partial reversion to wild‐type behaviour was eventually obtained after five but not four cycles of long term serial subculture in susceptible pepper, as by then the culture had become a mixture of both types of strain. This work suggests that resistance‐breaking strains of TSWV that overcome single gene hypersensitive resistance in pepper are relatively stable. The findings have important implications for situations where resistant pepper cultivars are deployed widely in the field without taking other control measures as part of an integrated TSWV management strategy.  相似文献
