The leakage of solutes from foliar tissue is utilized as a dynamic measure of apparent changes in membrane integrity in response to desiccation. It is found that rehydrating leaf discs of cowpea (Vigna sinensis [L.] Endl.) show increasing leakiness in proportion to the extent of prior desiccation, whereas Selaginella lepidophylla Spring., a resurrection plant, does not. The elevated leakage rate of cowpea after desiccation recovers with time, and the passage of time in the stressed condition results in reduced subsequent leakiness. These characteristics are interpreted as suggesting that the leakage of solute reflects the condition of cellular membranes, and that desiccation stress leads to lesions in the membranes. The kinetics of solute leakage is suggested as a simple means of following changes in membrane lesions and associated features of membrane repair and hardening.  相似文献

Leaf blades of different ages from a salt-tolerant wheat x Lophopyrum elongatum (Host) A. Love (syn. Agropyron elongatum Host) amphiploid and its salt-sensitive wheat parent (Triticum aestivum L.cv Chinese Spring) were compared for their ionic relations, organic solute accumulation, and sap osmotic potential ([pi]sap). The plants were grown for 18 d in nonsaline (1.25 mM Na+) and salinized (200 mM NaCl) nutrient solutions. The response of leaf blades to NaCl salinity depended greatly on their age or position on the main stem. Na and proline levels were highest in the oldest leaf blade and progressively lower in younger ones. Glycine betaine and asparagine levels were highest in the youngest blade. The [pi]sap was similar for corresponding leaf blades of both genotypes, but contributions of various solutes to the difference in [pi]sap between blades from control and 200 mM NaCl treatments differed greatly. The NaCl-induced decline in [pi]sap of the youngest leaf blade of Chinese Spring was predominately due to the accumulation of Na and to a lesser extent asparagine; in the amphiploid, it was due to a combination of glycine betaine, K, Na, and asparagine. Proline contributed little in the youngest blade of either genotype. In the older blades Na was the major solute contributing to the decline in [pi]sap. Thus, the maintenance of low Na and high K levels and the accumulation of glycine betaine in the young leaf tissues contributed to the NaCl tolerance of the amphiploid. Solutes distribute differentially between leaf tissues and cells. The present study tested the hypothesis that certain solutes are supplied preferentially to the epidermis in the transpiration stream, by-passing mesophyll cells along bundle sheath extensions. Using energy dispersive X-ray analysis of extracted cell sap, the distribution of solutes was studied in the emerged zone (transpiring) and the elongation zone (non-transpiring) of the developing leaf three of barley (Hordeum vulgare L.). The basic distribution of Cl, K, P and Ca between epidermis and bulk tissue, and between cells within the epidermis, was similar in the two leaf regions. However, in the emerged zone differences in solute concentrations between tissues and cells were greater. A local reduction in transpiration rate along the emerged portion of the blade specifically prevented Ca from accumulating to high levels in epidermal cells close to stomata. It is concluded that differences in solute concentrations between epidermal cells and other leaf tissues can be established in the absence of transpiration, but that they require transpiration for their full expression. Ion chromatographic methods determined organic acids and mainnutrient minerals in the apoplastic solution from leaves ofseveral Fagaceae (Quercus ilex L., Quercus cerris L., Quercusvirgiliana (Ten.) Ten, and Fagus sylvatica L.). The anions oforganic acids found in high amounts (250 to 650 µM) werequinate, malate, and oxalate. Lactate, pyruvate, formate andacetate were detected in relatively low amounts with concentrationsbetween 20 and 200 µM. The total concentration of organicacids in the apoplastic sap ranged between 1.5 and 2 mM. Thetotal concentration of inorganic cations (K+, Mg2+, NH4+, Ca2+,Na+) and anions (C1, NO3, SO2–4 and PO3–4)in the apoplastic sap varied between 5 and 10 mM, and 0.35 and1.8 mM, respectively. We conclude that the concentration oforganic acid ions in the leaf apoplast depends mainly on theexchange with the leaf cells and is influenced by the electrochemicalgradient between the symplast and the apoplast in relation tothe water potential of the leaf. The determination of formateand acetate in the apoplastic compartment of leaves lend weightto the argument that the production of these acids by treesis a important emission source to the atmosphere. Two methods for measuring the turgor pressures of cells in discsof storage tissue of red beet (Beta vulgaris L.) were compared,and a centrifugation method for extracting sap from frozen andthawed tissue was evaluated. Turgor pressures were measureddirectly using a pressure probe, or indirectly using a vapourpressure osmometer. With the latter, discs were placed directlyin the osmometer chamber and turgor was calculated as the differencein osmotic pressure before and after freezing and thawing. Turgorin freshly cut discs, measured with the pressure probe, wasbetween 0-012 MPa and 0.118 MPa with a mean ±s.d. of0.092±;0.032 MPa (n = 24). That measured with the osmometervaried between 0.08 MPa and 0.12 MPa with a mean ±s.d.of 0.09±0.10 MPa (n = 54). After vacuum infiltrationof discs with distilled water, the turgor measured with thepressure probe increased to 1.05–1.12 MPa. Turgor measuredwith the osmometer also increased after vacuum infiltrationbut was, on average, 12% lower than that measured with the pressureprobe. Overall, the results suggest that for routine measurements,the osmometer can provide reasonable estimates of the turgorof cells in beet discs. This is because a number of factorsthat, potentially, could interfere with this method have onlya small effect in this tissue. None of the measured turgorsis indicative of that occurring in intact storage roots becauseboth excision and vacuum infiltration of discs alter the concentrationsof solutes in the extracellular space. The osmotic pressureof sap extracted by centrifugation from frozen and thawed discswas not significantly different from that measured by placingfrozen and thawed discs directly in the osmometer. Hydroponic-grown seedlings of aspen (Populus tremuloides Michx.) were used to investigate how low root temperatures (5°C) affect stomatal conductance and water relations. An isohydric manner of the stomatal behaviour was found with the seedlings when their roots were subjected to the low temperature. Stomatal conductance rapidly and dramatically reduced in response to the low root temperature, while the xylem water potential did not significantly alter. Under the low root temperature, pH value of the xylem sap increased from 6.15 to 6.72 within the initial 4 h, while abscisic acid (ABA) concentration increased by the eighth hour of treatment. K+ concentration of the xylem sap significantly decreased within the 8th h and then reversed by the 24th h. The ion change was accompanied by a decrease and then an increase in the electrical conductivity, and an increase and then a decrease in the osmotic potential. The tempo of physiological responses to the low root temperature suggests that the rapid pH change of the xylem sap was the initial factor which triggered stomatal closure in low temperature-treated seedlings, and that the role of the more slowly accumulating ABA was likely to reinforce the stomatal closure. Xylem sap from the seedlings subjected low root temperature affected stomatal aperture on leaf discs when they were floated on the sap solution. The stomatal aperture correlated (P = 0.006) with the changed pattern of [K+] in the sap while the range of pH or ABA found in the xylem sap did not influence stomatal aperture of leaf discs in solution. The effect of xylem sap on stomatal aperture on leaf discs was different from on stomatal conductance in the intact seedlings. Comparison was made with previous study with the soil-grown seedlings.  相似文献

When leaf discs of a C4 species, Alternanthera pungens (L.) H.B. and K. or Amaranthus hypochondriacus L., were preincubated in 7.5 m M NH4Cl, the pH of the cell sap increased by nearly 0.3 unit, while the activity of phosphoenolpyruvate carboxylase (PEPC) about doubled compared to the cell sap from control leaf discs (preincubated in 5 m M Tricine‐KOH, pH 8.5). The sensitivity of PEPC to L ‐malate (a feedback inhibitor) decreased marginally as a result of cytosolic alkalization. The pH of the cell sap and PEPC activity decreased by nearly 0.4 unit and 50%, respectively, when leaf discs were incubated in weak organic acids such as propionic, butyric or salicylic acid. Thus, our results demonstrate a marked modulation in vivo of cell sap pH and PEPC activity in leaf discs from C4 plants by external alkalizing or acidifying reagents. The rise in PEPC activity due to alkalization of leaf discs was not sensitive to cycloheximide, implying that cytosolic protein synthesis was not involved in the activation of PEPC. Despite the marked increase in the PEPC activity due to the base‐loading of leaf discs, the change in malate sensitivity of the enzyme was only marginal, indicating that there was no significant increase in the extent of PEPC‐phosphorylation. Besides the physiological significance, the technique of acid/ base‐loading may be an important tool for studying the regulation of PEPC in leaf discs of C4 species, since the activity of PEPC could be enhanced apparently without phosphorylation of the enzyme.  相似文献

The mechanisms regulating stomatal response following exposure to low (5°C) soil temperature were investigated in aspen ( Populus tremuloides Michx.) seedlings. Low soil temperature reduced stomatal conductance within 4 h, but did not alter shoot xylem pressure potential within 24 h. The xylem sap composition was altered and its pH increased from 6.5 to 7.1 within the initial 4 h of the low temperature treatment. However, the increase in abscisic acid (ABA) concentration in xylem sap was observed later, after 8 h of treatment. These changes were accompanied by a reduction in the electrical conductivity and an increase in the osmotic potential of the xylem sap. The timing of physiological responses to low soil temperature suggests that the rapid pH change of the xylem sap and accompanying changes in ion concentration were the initial factors which triggered stomatal closure in low temperature-treated seedlings, and that the role of the more slowly accumulating ABA was likely to reinforce the stomatal closure. When leaf discs were exposed to xylem sap extracted from low soil temperature-treated plants, stomatal aperture was negatively correlated with ABA and positively correlated with K+ concentrations of the xylem sap. The stomatal opening in the leaf discs linearly increased in response to exogenous KCl concentrations when K+ concentrations were in the similar range to those detected in the xylem sap. The lowest concentration of exogenous ABA to induce stomatal closure was several-fold higher compared with the concentration present in the xylem sap.  相似文献

This study examined the potential role of restricted phloem export, or import of substances from the roots in the leaf growth response to root hypoxia. In addition, the effects of root hypoxia on abscisic acid (ABA) and zeatin riboside (ZR) levels were measured and their effects on in vitro growth determined. Imposition of root hypoxia in the dark when transpirational water flux was minimal delayed the reduction in leaf growth until the following light period. Restriction of phloem transport by stem girdling did not eliminate the hypoxia-induced reduction in leaf growth. In vitro growth of leaf discs was inhibited in the presence of xylem sap collected from hypoxic roots, and also by millimolar ABA. Disc growth was promoted by sap from aerated roots and by 0.1 micromolar ZR. The flux of both ABA and ZR was reduced in xylem sap from hypoxic roots. Leaf ABA transiently increased twofold after 24 hours of hypoxia exposure but there were no changes in leaf cytokinin levels.  相似文献

The transport of sugars and amino acids into the mycelium of Erysiphe pisi DC. was investigated using two different systems, intact leaf discs and mycelial suspensions. Of the sugars tested, glucose was preferentially taken up by both uninfected and mildew-infected leaf discs, whereas glutamine was taken up by both tissues at a higher rate than lysine or aspartic acid. Leaf discs from infected tissue had a greater uptake capacity than those from healthy tissue for both sugars and amino acids. The uptake of glucose was inhibited more markedly than that of sucrose and fructose by 10 μ m carbonyl cyanide m -chlorophenylhydrazone (CCCP), 1 m m N -ethylmaleimide (NEM), 1 m m diethyl pyrocarbonate (DEPC) and 1 m m phenylglyoxal, whereas 1 m m PCMBS ( p -chloro-mercuribenzenesulphonic acid) inhibited sucrose uptake to the greatest extent. Uptake of glutamine, lysine and aspartic acid was inhibited similarly by CCCP (80%), NEM (20%), DEPC (70%) and PCMBS (60%). Additionally, leaf discs were used to determine which solutes could be taken up from leaf tissue by the fungus. The uptake of sugars into the mycelium was greater than that of amino acids.
Suspensions of powdery mildew mycelium accumulated glucose at about three times the rate of sucrose or fructose, and the amino acid glutamine was taken up at three times the rate of lysine or aspartic acid. Spores separated from the suspension had a low uptake capacity.
When the reducing sugar concentration of leaf apoplastic fluid was estimated, leaves infected by powdery mildew had much higher amounts in the apoplast, whereas the activity of acid invertase also appeared to be higher in apoplastic fluids from infected leaves. When apoplastic fluid samples were run on SDS gels, an invertase antibody detected two bands in samples from infected tissues that were not found in the uninfected samples.  相似文献

A thermocouple psychrometer method, previously described foruse in determining tissue and sap water potentials, has beenadapted for determining matric potentials. Matric values ranging from approximately zero to –10 x105 Pa were observed with wilting wheat leaves. Mean valuesof total tissue water potential and of its solute (osmotic),matric, and cell wall pressure components were –16.6,–18.3, –30 and +4.7 Pa x 105, respectively. Matric potentials are often ignored in investigations of plantwater relations. This practice is shown to result in underestimationof wall pressures (in this case by a mean of 64 per cent) andsometimes in spuriously negative values.  相似文献

Beetroot storage tissue that had been aged in an aerated solutionwas particularly suited for studies of solute losses duringanoxia;retention of betacyanin being a good indicator of tonoplastintegrity. During anoxia, loss of K+ was nearly always greater than thatof Na+ while Cl loss was intermediate. Supply of glucoseduringageing increased the tolerance of beetroot tissue to anoxia.In these tolerant tissues, there were three phases of soluteloss.During the first phase, losses of K+ and amino acids wererapid, presumably due to membrane depolarization from –156to –95 mV. In contrast, losses of Na+ and Cl wereslow. During the second phase, K+ loss had decreased to a lowrate, while losses of Na+ and Cl+ remained slow. Furthermore,the membrane potential remained at –95 to –90mV,which was consistent with the diffusion potential estimatedfrom the modified Goldman equation. In the third and final phase,loss of K+ Na+ Cl+,sugars, and amino acids began to increase,soon followed by loss of betacyanin. Tissues that had lost their betacyanin during anoxia were irreversiblyinjured, as shown by rapid uptake of Evans Blue and afailureto take up K+ , Na+ and Cl+ during re–aeration. In contrast,tissues which had retained their betacyanin did not take upEvansBlue, but took up substantial amounts of K+ , Na+ , and Clafter re–aeration. We have been examining the importance of the root system on shoot growth and development using a developmentally disabled hybrid of the common bean Phaseolus vulgaris L. Parental cultivars (P. Vulgaris cv. Redkloud of Mesoamerican origin, and P. vulgaris cv. Batt of Andean origin) grow normally, but crosses produce F1 hybrids exhibiting hybrid weakness associated with reduced root and shoot growth. In this study, applications of benzylaminopurine (BAP) to roots of F1 hybrids increased the number of root tips and leaves. Reciprocal grafting was used to study the effects of the root system on shoots. Grafting of roots of the Mesoamerican cultivar onto shoots of F1 hybrids increased the cytokinin concentrations in leaves of F1 hybrids and removed the characteristics associated with hybrid weakness. To determine whether factors in the xylem sap enhanced leaf growth, leaf discs were incubated on sap collected from Mesoamerican and Andean cultivars. Sap from Mesoamerican plants enhanced the growth of leaf discs excised from F1 hybrids more than sap collected from Andean cultivars. Estimates of the transport of zeatin riboside (ZR)–type cytokinins from roots of F1 hybrids indicated that transport out of hybrid roots was reduced compared with those transported out of Mesoamerican or Andean roots. Results suggest that ZR-type cytokinins are involved in hormonal integration between roots and shoots of P. vulgaris and that one of the barriers to hybridization between Andean and Mesoamerican landraces is related to hormone transport. Ethylene production from leaf discs of dwarf bean (Phaseolausvulgaris L.) was less than 02 nl g–1 h–1 at 5 Cbut rapidly increased tenfold on transfer to 25 C. The lowethylene production at 5 C and the potential for overshootproduction on transfer to 25C were not associated with accumulationof the ethylene synthesis intermediate 1-aminocyclopropane-1-carboxylicacid (ACC). Addition of exogenous ACC to leaf discs incubatedat 5C increased ethylene production, while similarly incubatedleaf discs did not synthesize increasing amounts of endogenousACC until they were transferred to 25 C. The basis for theovershoot in ethylene production when leafdiscs were transferredfrom 5 to 25 C appears to reside in changes to the pathwayleading to the synthesis of ACC or an earlier intermediate inthe pathway of ethylene biosynthesis. Summary Young seedlings of English Oak, Quercus robur L., and Silver Birch, Betula verrucosa Ehrl., were subjected to a number of consecutive periods during which water was withheld. During one 14-day period leaf-and soil-water potentials and leaf- and root-solute potentials of two groups of plants were sampled at noon of each day. One group of plants was watered every day while water was withheld from the other group. Solute accumulation in roots and leaves of oak seedlings subjected to water stress resulted in maintenance of turgor and high leaf conductance as the soil dried. In birch seedlings turgor was only maintained by stomatal closure at high soil water potential.Fourteen consecutive water stress cycles greatly reduced the growth of birch seedlings but had little effect on oak seedlings other than to alter root morphology. Water stress treatment resulted in the production of long thin roots in this plant. Stomatal behaviour in oak and birch seedlings during the 14-week stress period was consistent with observed changes in leaf water and solute potentials. Daily solute accumulation in oak leaves was presumably responsible for the maintenance of plant growth as water potentials fell.  相似文献

Cellulysin-induced ethylene production in tobacco (Nicotiana tabacum L.) leaf discs was enhanced several-fold by prior exposure of the leaf tissue to ethylene. This enhancement in the response of the tissue to Cellulysin increased rapidly during 4 and 8 hours of pretreatment with ethylene and resulted from greater conversion of methionine to ethylene. On treatment with Cellulysin, the content of 1-aminocyclopropane-1-carboxylic acid (ACC) in leaf discs not pretreated with ethylene markedly increased while that of the ethylene-pretreated tissue was only slightly higher than in the tissue incubated in the absence of Cellulysin. Ethylene-treated tissue, however, converted ACC to ethylene at a faster rate than air controls. These data indicate that ethylene stimulates Cellulysin-induced ethylene production by stimulating the conversion of ACC to ethylene. Data are also presented on a possible relation of this phenomenon to ethylene produced by the tobacco leaf upon interaction with its pathogen, Alternaria alternata.  相似文献

Exogenously supplied indole-3-acetic acid (IAA) stimulated ethylene production in tobacco (Nicotiana glauca) leaf discs but not in those of sugar beet (Beta vulgaris L.). The stimulatory effect of IAA in tobacco was relatively small during the first 24 hours of incubation but became greater during the next 24 hours. It was found that leaf discs of these two species metabolized [1-14C]IAA quite differently. The rate of decarboxylation in sugar beet discs was much higher than in tobacco. The latter contained much less free IAA but a markedly higher level of IAA conjugates. The major conjugate in the sugar beet extracts was indole-3-acetylaspartic acid, whereas tobacco extracts contained mainly three polar IAA conjugates which were not found in the sugar beet extracts. The accumulation of the unidentified conjugates corresponded with the rise of ethylene production in the tobacco leaf discs. Reapplication of all the extracted IAA conjugates resulted in a great stimulation of ethylene production by tobacco leaf discs which was accompanied by decarboxylation of the IAA conjugates. The results suggest that in tobacco IAA-treated leaf discs the IAA conjugates could stimulate ethylene production by a slow release of free IAA. The inability of the exogenously supplied IAA to stimulate ethylene production in the sugar beet leaf discs was not due to a deficiency of free IAA within the tissue but rather to the lack of responsiveness of this tissue to IAA, probably because of an autoinhibitory mechanism existing in the sugar beet leaf discs.  相似文献

The distribution of solutes in the various cells of sugar beet (Beta vulgaris L.) source leaves, petioles, and sink leaves was studied in tissue prepared by freeze-substitution. The differences in degree of cryoprotection indicated that sieve elements and companion cells of the source leaf, petiole, and sink leaf contain a high concentration of solute. The osmotic pressure of various types of cells was measured by observing incipient plasmolysis in freeze-substituted tissues equilibrated with a series of mannitol solutions prior to rapid freezing. Analysis of source leaf tissue revealed osmotic pressure values of 13 bars for the mesophyll and 30 bars for the sieve elements and companion cells. The osmotic pressure of the mesophyll of sink leaves was somewhat higher.  相似文献

Lowland dry forests are unique in Hawaii for their high diversity of tree species compared with wet forests. We characterized spatial and temporal partitioning of soil water resources among seven indigenous and one invasive dry forest species to determine whether the degree of partitioning was consistent with the relatively high species richness in these forests. Patterns of water utilization were inferred from stable hydrogen isotope ratios (δD) of soil and xylem water, zones of soil water depletion, plant water status, leaf phenology, and spatial patterns of species distribution. Soil water δD values ranged from –20‰ near the surface to –48‰ at 130 cm depth. Metrosideros polymorpha, an evergreen species, and Reynoldsia sandwicensis, a drought-deciduous species, had xylem sap δD values of about –52‰, and appeared to obtain their water largely from deeper soil layers. The remaining six species had xylem δD values ranging from –33 to –42‰, and apparently obtained water from shallower soil layers. Xylem water δD values were negatively correlated with minimum annual leaf water potential and positively correlated with leaf solute content, an integrated measure of leaf water deficit. Seasonal patterns of leaf production ranged from dry season deciduous at one extreme to evergreen with near constant leaf expansion rates at the other. Species tapping water more actively from deeper soil layers tended to exhibit larger seasonality of leaf production than species relying on shallower soil water sources. Individuals of Myoporum sandwicense were more spatially isolated than would be expected by chance. Even though this species apparently extracted water primarily from shallow soil layers, as indicated by its xylem δD values, its nearly constant growth rates across all seasons may have been the result of a larger volume of soil water available per individual. The two dominant species, Diospyros sandwicensis and Nestegis sandwicensis, exhibited low leaf water potentials during the dry season and apparently drew water mostly from the upper portion of the soil profile, which may have allowed them to exploit light precipitation events more effectively than the more deeply rooted species. Character displacement in spatial and temporal patterns of soil water uptake was consistent with the relatively high diversity of woody species in Hawaiian dry forests. The mechanism of iron (Fe) uptake from the leaf apoplast into leaf mesophyll cells was studied to evaluate the putative Fe inactivation as a possible cause of Fe deficiency chlorosis. For this purpose, sunflower (Helianthus annuus L.) and faba bean plants (Vicia faba L.) were precultured with varied Fe and bicarbonate (HCO 3 - ) supply in nutrient solution. After 2–3 weeks preculture, FeIII reduction and 59Fe uptake by leaf discs were measured in solutions with Fe supplied as citrate or synthetic chelates in darkness. The data clearly indicate that FeIII reduction is a prerequisite for Fe uptake into leaf cells and that the Fe nutritional status of plants does not affect either process. In addition, varied supply of Fe and HCO 3 - to the root medium during preculture had no effect on pH of the xylem sap and leaf apoplastic fluid. A varied pH of the incubation solution had no significant effect on FeIII reduction and Fe uptake by leaf discs in the physiologically relevant pH range of 5.0–6.0 as measured in the apoplastic leaf fluid. It is concluded that Fe inactivation in the leaf apoplast is not a primary cause of Fe deficiency chlorosis induced by bicarbonate.
