(1) H+/electron acceptor ratios have been determined with the oxidant pulse method for cells of denitrifying Paracoccus denitrificans oxidizing endogenous substrates during reduction of O2, NO?2 or N2O. Under optimal H+-translocation conditions, the ratios H+O, H+N2O, H+NO?2 for reduction to N2 and H+NO?2 for reduction to N2O were 6.0–6.3, 4.02, 5.79 and 3.37, respectively. (2) With ascorbate/N,N,N′,N′-tetramethyl-p-phenylenediamine as exogenous substrate, addition of NO?2 or N2O to an anaerobic cell suspension resulted in rapid alkalinization of the outer bulk medium. H+N2O, H+NO?2 for reduction to N2 and H+NO?2 for reduction to N2O were ?0.84, ?2.33 and ?1.90, respectively. (3) The H+oxidant ratios, mentioned in item 2, were not altered in the presence of valinomycinK+ and the triphenylmethylphosphonium cation. (4) A simplified scheme of electron transport to O2, NO?2 and N2O is presented which shows a periplasmic orientation of the nitrite reductase as well as the nitrous oxide reductase. Electrons destined for NO?2, N2O or O2 pass two H+-translocating sites. The H+electron acceptor ratios predicted by this scheme are in good agreement with the experimental values.  相似文献

2-(Methylamino)pyridine reacts with RuCl2(CO)3 to give a carbamoyl complex, [Ru(C(O)N(CH3)(C5H4N)Cl(CO)2], which yields with pyridine (py) and acetylacetone (Hacac), respectively, [Ru(C(O)N(CH3)C5H4N)Cl(CO)2(py)] and [Ru(C(O)N(CH3)C5H4N)(CO)2(acac)]. These complexes are characterized spectroscopically. The amino group of the ligand is carbonylated and the resulted carbamoyl ligand is chelating through a pyridine ring-N and a carbamoyl-C atom. 2-Aminopyridine and 2-aminopyrimidine react similarly with RuCl2(CO)3 to give the corresponding carbamoyl complexes.  相似文献

Respiration (O), ammonium (NH4), phosphate (PO4), total nitrogen (NT) and phosphorus (PT) excretions were measured on mixed zooplankton during 3-, 6-, 9-, 12-, 21-, and 24-h incubation periods at 20–23 C. The excretion rates of PO4, NT. and PT decrease during a 21-h period, while rates of respiration and excretion of NH{IN4} are constant. The percentage of inorganic nitrogen excreted increases regularly from 3 h (30–40% of total nitrogen) to 21 h (70–80%) and it could be either due to a bacterial activity which was measured or to a decrease with time of organic nitrogen excreted because of starvation. ONT, OPO4, OPT, and NH4PO4 ratios increase during the first 9 h of incubation; the percentage of inorganic phosphorus excreted is higher at the very beginning and then remains constant from 6 to 24 h. ONH4 and NTPT ratios are constant during a 24-h term, which makes them useful metabolic indexes.  相似文献

5-hydroxylysine, an analogue of glutamate and lysine, causes NH4+ production by N2-fixing A. cylindrica; it also reversibly inhibits GS activity in vitro but has no effect on alanine dehydrogenase or GOGAT. On adding 5-hydroxylysine intracellular pools of glutamine, glutamate and aspartate decrease; those of alanine and serine increase. 5-hydroxylysine alleviates the inhibitory effect of NH4+ on heterocyst production and C2H2 reduction and in NH4+-grown cultures results in heterocyst synthesis and in C2H2 reduction. The data suggest that the GS-GOGAT pathway is the sole route of importance in primary NH4+ assimilation in A. cylindrica, that NH4+ alone does not inhibit nitrogenase and heterocyst production, and that GS and/or a product is involved in regulating the production of both.  相似文献

In his study of a kin selection model for the evolution of workers' behavior in an incipiently social insect, Craig (1979) found that the haplodiploid mode of sex determination, combined with a female-biased sex ratio, cannot accelerate an evolutionary trend toward eusociality. This seems to contradict to Hamilton's (1964) theory. It is my intention to prove that Craig's result is due to the dependence of relative reproductive success in each sex on the sex ratio of a population. The oviposition of unfertilized eggs by workers is indispensable, in primitive stages in the evolution of eusociality, for maintaining the relative reproductive success of a female. Assuming that workers control the sex ratios, we can distinguish the following three evolutionary phases in accordance with the ratio N2N10 (the ratio of the number of the queen's second brood to that of the progeny laid by workers derived from the same nest). During Phase 1 in which N2N10 < 12, the reproductive successes of a male and a female are equal, and Hamilton's rule holds. In Phase 2 in which 12 <N2N10 < 32, the queen produces females, and workers oviposit males. As N2N10 increases, the sex ratio in the whole population becomes female-biased and relative reproductive success of a male increases. In Phase 3 in which N2/N10 >32, the queen lays some male eggs in addition to female eggs. The sociality threshold (BC)crit the minimum value of the benefit/cost ratio leading to the evolution of altruism, is 23on Phase 1. It rises threefold as N2/N10 increases in Phase 2, and, in Phase 3, it is twice as high as that in a diploid species. The anatomical and physiological characteristics of workers must have been so developed that they are efficient in helping activities after the beginning of eusociality. The evolutionary process before the beginning of helping behavior is also discussed. The egg laying by unmated females, which stay their mother's nest, seems to have been an important preadaptation for the evolution of eusociality in Hymenoptera.  相似文献

N-Phenylhydroxylamine is oxidized in aqueous phosphate buffer to nitrosobenzene, nitrobenzene, and azoxybenzene. Degradation is O2 dependent and shows general catalysis by H2PO4? (k1 = 2.3 M?2 sec?1) and PO4?3 (k2 = 2.3 × 105M?2 sec?1) or kinetically equivalent terms. (1) Na+ currents and Na+-current fluctuations were measured in myelinated frog nerve fibres at 15°C during 7.7 ms depolarizations to V = 40, 60 and 80 mV. (2) The conductance γ of a single Na+ channel and the number N0 of channels per node were calculated from ensemble average values of the mean Na+ current and the variance of Na+-current fluctuations. (3) For a hyperpolarizing holding potential of VH = ?28 mV the mean values of the channel conductance and number were γ = 9.8 pS and N0 = 74 000. (4) After changing the holding potential to the resting potential (VH = 0) the conductance γ increased by a factor of 1.37 whereas the number N0 decreased by a factor of 0.60. (5) Addition of 8 nM tetrodotoxin at a holding potential of VH = ?28 mV increased γ by a factor of 1.55 and reduced N0 by a factor of 0.25. (6) The increase of the channel conductance at reduced channel numbers suggests negative cooperativity between Na+ channels in the nodal membrane.  相似文献

The action of xanthine oxidase upon acetaldehyde or xanthine at pH 10.2 has been shown to be accompanied by substantial accumulation of O2? during the first few minutes of the reaction. H2O2 decreases this accumulation of O2? presumably because of the Haber-Weiss reaction (H2O2+O2?OH?+OH+O2) and very small amounts of superoxide dismutase eliminate it. This accumulation of O2? was demonstrated in terms of a burst of reduction of cytochrome c, seen when the latter compound was added after aerobic preincubation of xanthine oxidase with its substrate. The kinetic peculiarities of the luminescence seen in the presence of luminol, which previously led to the proposal of H2O4?, can now be satisfactorily explained entirely on the basis of known radical intermediates.  相似文献

Infrared spectra of N2O in a variety of solvents and in the brain of a dog under typical conditions of halothane-N2O anesthesia have been determined. The appearance or disappearance of N2O in the brain was readily followed as N2O was administered or withdrawn. The sites in brain were of two major types; one, with ν3 = 2229.8 ± 0.4 cm?1 and Δν12 = 13.0 ± 0.6 cm?1, is rather like the polar site in water and the other, with ν3 = 2216.8 ± 0.8 cm?1 and Δν12 = 9.6 ± 1.0 cm?1, is non-polar and is probably associated with membrane lipid. The significant variations in the antisymmetric stretch (ν3) of N2O as the polarity and other properties of the medium (solvent) vary make possible the characterization of in tissue sites occupied by this anesthetic.  相似文献

Kinetic studies on the RNase T1-catalyzed transesterification of 12 dinucleoside monophosphates, Np1N2 (N1 = A, C, and U; N2 = A, C, G, and U) at pH 5, 25 °C, and 0.2 m ionic strength, revealed that the catalytic efficiency (kcatKm) for GpN substrates (H. L. Osterman, and F. G. Walz, Jr., 1978, Biochemistry, 17, 4142) was ~106-fold greater than corresponding ApNs and at least 108-fold greater than corresponding CpNs and UpNs. The catalytic activity with ApN substrates survives phenol extraction which indicates (along with other criteria) that it is intrinsic to RNase T1 and is not due to trace contamination by other nucleases. Circumstantial evidence is presented which suggests that homologous GpN and ApN substrates bind productively at different sites on the enzyme. The results of steady-state kinetic studies of RNase T1 with IpNs (N = C and U) were compared with those for GpNs and indicated that the primary effect of the guanine 2-NH2 group is to enhance substrate binding at the primary recognition site by ~2.6 kcal/mol. Values of (kcatKm) showed the order NpC > NpU (N = A, G, and I) which evidences the existence of a subsite for the leaving nucleoside group that prefers cytidine: interactions at this subsite are reflected in kcat rather than Km.  相似文献

In order to test the question if a pool of lipophilic ions may exist in black lipid membranes which cannot be detected by electrical relaxation measurements we have performed simultaneously measurements of the optical absorption of a lipophilic ion. The absorbance of membrane-bound dipicrylamine at 410 nm was measured with a sensitive spectrophotometer which can detect absorbance changes ? 4 · 10?5. A minimal concentration of about 6 · 1011 dipicrylamine ions per cm2 of the membrane could be detected with this instrument. The dipicrylamine concentration in the membrane obtained with the optical method Ntopt is compared with the concentrations Ntel obtained from simultaneous electrical relaxation measurements. Ntopt and Ntel agreed at low dipicrylamine concentrations (10?8–10?7 M in the aqueous phase) and showed saturation at higher concentrations (up to 5 · 10?6 M). In the saturation range Ntopt was maximally four times higher than Ntel. The significance of this difference is discussed together with general aspects of the saturation phenomenon.  相似文献

The kinetics of bisulfite addition to 5-fluorouracil were studied as a function of increasing concentrations of potential general acids. Values of kobsd[SO3=] measured at 25°C and ionic strength 1.0 M increased linearly and then became invariant with increasing concentrations of either HSO3? or (OHCH2CH2)2N+C(CH2OH)3 HCl (BisTris+HCl). A small kinetic hydrogen-deuterium isotope effect (kHSkDS = 1.10) was observed for the general acid catalysed portion of the addition reaction. The kinetics of bisulfite elimination from 5-fluoro-5,6-dihydrouracil-6-sulfonate were studied in ethanolamine buffers. As previously observed with 1,3-dimethyl-5,6-dihydrouracil-6-sulfonate, this reaction is subject to general base catalysis and exhibits a large kinetic hydrogen-deuterium isotope effect (k2H2Ok2D2O = 3.8). The kinetic results for the addition reaction are consistent with a multistep reaction pathway involving the initial formation of an oxyanion sulfite addition intermediate (II) which subsequently adds a proton and undergoes tautomerization to yield the final 5-fluoro-5,6-dihydrouracil-6-sulfonate product. Thus the elimination of bisulfite from 5-fluoro-5,6-dihydrouracil-6-sulfonate probably proceeds by an ElcB mechanism which involves, at relatively low concentrations of general base, rate determining general base catalyzed proton abstraction from carbon 5 to yield intermediate II followed by the rapid elimination of sulfite to yield 5-fluorouracil. These results may be related to both the enzymatically catalyzed dehalogenation of bromoand iodouracil and the methylation of deoxyuridylate by thymidylate synthetase.  相似文献

Quercetin inhibited a dog kidney (Na+ + K+)-ATPase preparation without affecting Km for ATP or K0.5 for cation activators, attributable to the slowly-reversible nature of its inhibition. Dimethyl sulfoxide, a selector of E2 enzyme conformations, blocked this inhibition, while the K+-phosphatase activity was at least as sensitive to quercetin as the (Na+ + K+)-ATPase activity, all consistent with quercetin favoring E1 conformations of the enzyme. Oligomycin, a rapidly-reversible inhibitor, decreased the Km for ATP and the K0.5 for cation activators, and its inhibition was also diminished by dimethyl sulfoxide. Although oligomycin did not inhibit the K+-phosphatase activity under standard assay conditions, a reaction presumably catalyzed by E2 conformations, its effects are nevertheless accommodated by a quantitative model for that reaction depicting oligomycin as favoring E1 conformations. The model also accounts quantitatively for effects of both dimethyl sulfoxide and oligomycin on Vmax, Km for substrate, and K0.5 for K+, as well as for stimulation of phosphatase activity by both these reagents at low K+ but high Na+ concentrations.  相似文献

Respiration-driven proton translocation has been studied with the oxidant pulse method for cells of denitrifying Paracoccus denitrificans oxidizing H2 during reduction of O2, NO?3, NO?2 or N2O. A simplified scheme of anaerobic electron transport and associated proton translocation is shown that is consistent with the measured H+oxidant ratios. Furthermore, the kinetics and energetics of NO?3 uptake in whole cells of P. denitrificans were studied. For this purpose, we measured H2 consumption or N2O production after addition of NO?3 to a cell suspension, which indirectly gave information about uptake (and reduction) of NO?3. It was found that a lag phase in H2 consumption or N2O production appeared whenever the membrane potential was dissipated by addition of thiocyanate, carbonyl cyanide m-chlorophenylhydrazone or triphenyl-methylphosphonium bromide. However, these lag phases were not observed when NO?2 was present at the moment of introduction of NO?3. On the basis of these findings we conclude that there are two uptake systems for NO?3. One system is dependent on the proton-motive force and is probably used for initiation of NO?3 uptake. The other is an NO?3NO?2 antiport and its function is to take over NO?3 uptake from the first system.  相似文献

Laser flash photolysis techniques have yielded rate constants for physical and reactive quenching modes of O2(1Δg) by nicotine, nicotinamide adenine dinucleotide (oxidized and reduced forms) and the reduced forms of nicotinamide mononucleotide, nicotinamide adenine dinucleotide phosphate and nicotinamide hypoxanthine dinucleotide. In the case of the last four named compounds, kinetic spectroscopy furnished evidence for one-electron transfers to O2(1Δg). Specifically, production of O?2 was demonstrated unequivocally by reaction with 1,4-benzoquinone. Quantitative determinations revealed the extent of reactive quenching to be near 60% in each case.  相似文献

The effects of inorganic cations, n-hexanol, saccharose and 2H2O on the electrophoretic mobility and ζ-potential of membrane vesicles from nerve myelin were measured and the results compared with the corresponding effects of the same reagents on the transition voltage, VTr, of the nerve axon membrane. Different cation concentrations and 2H2O affect both potentials, the ζ-potential and VTr, in a parallel way. Saccharose and n-hexanol, however, shift VTr but leave the electrophoretic mobility of the myelin vesicles unchanged. These results suggest that VTr shifts are not necessarily linked to changes in the membrane surface charge density but may also be caused by an interaction between the reagent and non-polar groups of the membrane interior.  相似文献

The stoichiometry of free NADPH oxidation in phenobarbital induced rabbit liver microsomes was measured by means of registering the rates of NADPH, H+ and O2 consumption and O2? and H2O2 production. ΔO2?:ΔH2O2 ratio is approximately I indicating that about half H2O2 results from O2? dismutation, the second half being formed directly. ΔNADPH:ΔH2O2 and ΔO2:ΔH2O2 ratios exceed I and therefore another product of the reaction is water. The fact that the ratio (ΔNADPH-ΔH2O2):(ΔO2-ΔH2O2) is 2 allows one to consider direct 4-electron O2 reduction as the major way of water formation rather than endogenous substrate hydroxylation.  相似文献

The longitudinal relaxation rate (1T1p) of water protons was studied in solutions of Mn(II)-concanavalin A at a number of frequencies. These relaxation rates were lowered in the presence of a variety of saccharides which have affinities for concanavalin A which range over two orders of magnitude. A good correlation was found in which saccharides which bind tightly have the greatest effect and saccharides which bind weakly or not at all have little effect on the 1T1p values. The temperature dependence of the proton relaxation rates showed that the lowering of these rates in the presence of saccharides was most likely due to a change in the exchange rate of solvent interacting with protein-bound Mn(II), 1Tm.An analysis of the temperature and frequency dependence of the 1T1p and 1T2p (transverse) solvent proton relaxation rates resulted in evaluation of a number of parameters for solvent water molecules interacting in the first coordination sphere of Mn(II) bound to concanavalin A. The ratio of the number of water molecules (q) to the Mn(II)-proton distance (r) obtained from a computer fit of the data over a limited temperature range is in accord with the findings of Koenig et al. ((1973) Proc. Nat. Acad. Sci.70, 475) and Meirovitch and Kalb ((1973) Biochim. Biophys. Acta303, 258). However, our studies of 1T1p and 1T2p of water over a more extensive temperature range are best fit with the following conclusions: at low temperatures (<20 °C), the data are consistent with an outer-sphere relaxation process. At higher temperatures (> 30 °C), the water molecule in the inner coordination sphere of the bound Mn(II) begins exchanging more rapidly and contributes to the relaxation processes (1T1p and 1T2p). The relaxation time of protons in the inner coordination shell, T1M, contributes over the entire temperature range and produces a frequency dependence in the relaxivity data from 6 to 100 MHz since the contributions to the correlation times are in the range 10?9-10?8 sec.  相似文献

The relative effectiveness of oxidizing (.OH, H2O2), ambivalent (O2?) and reducing free radicals (e? and CO2?) in causing damage to membranes and membrane-bound glyceraldehyde-3-phosphate dehydrogenase of resealed erythrocyte ghosts has been determined. The rates of damage to membranebound glyceraldehyde-3-phosphate dehydrogenase (R(enz)) were measured and the rates of damage to membranes (R(mb)) were assessed by measuring changes in permeability of the resealed ghosts to the relatively low molecular weight substrates of glyceraldehyde-3-phosphate dehydrogenase. Each radical was selectively isolated from the mixture produced during gamma-irradiation, using appropriate mixtures of scavengers such as catalase, superoxide dismutase and formate. .OH, O2? and H2 O2 were approximately equally effective in inactivating membrane-bound glyceraldehyde-3-phosphate dehydrogenase, while e? and CO2? were the least effective. R(enz) values of O2? and H2O2 were 10-times and of .OH 15-times that of e?. R(mb) values were quite similar for e? and H2O2 (about twice that of O2?), while that of .OH was 3-times that of O2?. Hence, with respect to R(mb): .OH >e? = H2O2 >O2? , and with respect to R(enz): .OH >O2? = H2O2 >e?. The difference between the effectiveness of the most damaging and the least damaging free radicals was more than 10-fold greater in damage to the enzyme than to the membranes. Comparison between H2O2 added as a chemical reagent and H2O2 formed by irradiation showed that membranes and membrane-bound glyceraldehyde-3-phosphate dehydrogenase were relatively inert to reagent H2O2 but markedly susceptible to the latter.  相似文献

The mean fixation index within subpopulations (FIS) has been defined as F̄IS = ∑wiFISior asF̂IS = ∑wipiqiFISi∑wipiqi. The latter definition is preferred because it can be obtained from the two other fixation indices, FST and FIT and because it is unaffected by the mean gene frequency. The expected frequency of heterozygotes in small subpopulations of dioecious organisms will exceed Hardy-Weinberg expectations and this can be measured by F̂IS. In an isolated subpopulation of constant variance effective size N, F̂IS rapidly tends to 1 − 4N2(N − 1 + [N2 + 1]12)2. In the Island model of population structure, F̂IS is approximately −(1 − m)Nwhere m is the immigration rate.When a sample is drawn from a natural population, the observed FIS will depend upon the genetic structure of the population. The values of FIS expected in three different types of population structure are discussed.  相似文献
