Temperature-jump-induced absorption changes of a bromocresol purple in chloroplast suspensions in the dark were studied. After a rapid rise in temperature (less than 10 mus), a slow absorbance decrease of bromocresol purple (t1/2 approximately 0.2 s) following a fast absorbance decrease of chloroplasts and bromocresol purple (t1/2 less than 1 ms) was observed. The slow absorbance decrease corresponds to acidification of the suspending medium, indicating H+ efflux from chloroplasts after the temperature jump. Nigericin and gramicidin D suppressed the slow absorbance change completely in the presence of 10 mM KC1, while valinomycin did not affect it. The fast absorbance change was not affected by the above ionophores. 3-(3,4-dichlorophenyl)-1,1-dimethylurea also diminished the slow absorbance change.  相似文献

In intact, uncoupled type B chloroplasts from spinach, added ATP causes a slow light-induced decline (t12 ≈ 3 min) of chlorophyll a fluorescence at room temperature. Fluorescence spectra were recorded after fast cooling to 77 K and normalized with fluorescein as an internal standard. Related to the fluorescence quenching at room temperature, an increase in Photosystem (PS) I fluorescence (F735) and a decrease in PS II fluorescence (F695) were observed in the low-temperature spectra. The change in the F735F695 ratio was abolished by the presence of methyl viologen. Fluorescence induction at 77 K of chloroplasts frozen in the quenched state showed lowered variable (Fv) and initial (F0) fluorescence at 690 nm and an increase in F0 at 735 nm. The results are interpreted as indicating an ATP-dependent change of the initial distribution of excitation energy in favor of PS I, which is controlled by the redox state of the electron-transport chain and, according to current theories, is caused by phosphorylation of the light-harvesting complex.  相似文献

The influence of temperature on the rate of reduction of P-680+, the primary donor of Photosystem II, has been studied in the range 5–294 K, in chloroplasts and subchloroplasts particles. P-680 was oxidized by a short laser flash. Its oxidation state was followed by the absorption level at 820 nm, and its reduction attributed to two mechanisms: electron donation from electron donor D1 and electron return from the primary plastoquinone (back-reaction).Between 294 and approx. 200 K, the rate of the back-reaction, on a logarithmic scale, is a linear function of the reciprocal of the absolute temperature, corresponding to an activation energy between 3.3 and 3.7 kcal · mol?1, in all of the materials examined (chloroplasts treated at low pH or with Tris; particles prepared with digitonin). Between approx. 200 K and 5 K the rate of the back-reaction is temperature independent, with t12 = 1.6 ms. In untreated chloroplasts we measured a t12 of 1.7 ms for the back-reaction at 77 and 5 K.The rate of electron donation from the donor D1 has been measured in darkadapted Tris-treated chloroplasts, in the range 294–260 K. This rate is strongly affected by temperature. An activation energy of 11 kcal · mol?1 was determined for this reaction.In subchloroplast particles prepared with Triton X-100 the signals due to P-680 were contaminated by absorption changes due to the triplet state of chlorophyll a. This triplet state has been examined with pure chlorophyll a in Triton X-100. An Arrhenius plot of its rate of decay shows a temperature-dependent region (292–220 K) with an activation energy of 9 kcal · mol?1, and a temperature-independent region (below 200 K) with t12 = 1.1 ms.  相似文献

In order to test the question if a pool of lipophilic ions may exist in black lipid membranes which cannot be detected by electrical relaxation measurements we have performed simultaneously measurements of the optical absorption of a lipophilic ion. The absorbance of membrane-bound dipicrylamine at 410 nm was measured with a sensitive spectrophotometer which can detect absorbance changes ? 4 · 10?5. A minimal concentration of about 6 · 1011 dipicrylamine ions per cm2 of the membrane could be detected with this instrument. The dipicrylamine concentration in the membrane obtained with the optical method Ntopt is compared with the concentrations Ntel obtained from simultaneous electrical relaxation measurements. Ntopt and Ntel agreed at low dipicrylamine concentrations (10?8–10?7 M in the aqueous phase) and showed saturation at higher concentrations (up to 5 · 10?6 M). In the saturation range Ntopt was maximally four times higher than Ntel. The significance of this difference is discussed together with general aspects of the saturation phenomenon.  相似文献

Spinach chloroplasts were dark adapted and then submitted to a sequence of short saturating flashes. The resulting absorption changes in the near ultraviolet were analyzed and attributed to the donor and acceptor sides of Photosystem II. Our results provide a spectroscopic support to some current models of these parts of the photosynthetic electron transport.In Tris-treated chloroplasts (supplied with artificial donors) the absorption changes are largely due to the acceptor side. After each flash the signal decays with a fast phase (t12 = 1.2 ms at 9 °C) leaving a stationary level (on a 100-ms time scale). The fast phase has a small amplitude after odd-numbered flashes, whereas the stationary level behaves in a complementary fashion. The non-decaying signal is attributed mostly to the reduced secondary acceptor (A2?) and the fast phase to the simultaneous reoxidation of A2? and of the reduced primary acceptor (A1?). The effect of 3-(3,4-dichlorophenyl)-1, 1-dimethylurea and of redox mediators (ascorbate, ferricyanide) also support this assignment. A fraction of A2 is shown to be reduced in dark-adapted chloroplasts, as proposed by Velthuys and Amesz (Biochim. Biophys. Acta (1974) 333, 85–94). The difference spectra support the view that A1? and A2? are plastoquinone radical anions. There are also some absorption changes that we cannot identify.In untreated chloroplasts a non-decaying absorption change (“slow phase”) occurs with a 4-flash periodicity. It is attributed to the transitions among the S states associated with the O2-evolving complex. A fast phase (t12 = 1.2 ms) in the decay following the first two flashes behaves like in Tris-treated chloroplasts, so that the assignment is tentatively the same. After the third flash, however, the magnitude of this fast phase is too large according to the hypothesis, so that there may be some contribution from the donor side. The fast phases become slower at lower pH (5.5 instead of 7.6), although there is no evidence for a protonation A1? or A2?.  相似文献

(1) The polymorphic phase behaviour of aqueous dispersions of various synthetic phosphatidylethanolamines, both singly and in mixtures, has been investigated by 31P-NMR. (2) 14:014:0 PE remains in the lamellar phase up to 90°C. 18:1t18:1t PE exhibits a lamellar to hexagonal (HII) transition between 60°C and 63°C. For 18:1c18:1c PE, the lamellar to hexagonal (HII) transition occurs between 7 and 12°C, whereas for 18:2c18:2c PE, the hexagonal (HII) phase is the preferred structure above ?15°C. (3) Mixtures of 18:1c18:1c PE and 18:1t18:1t PE exhibit near-ideal miscibility behaviour. For mixtures of 18:1c18:1c PE and 14:014:0 PE there is evidence of fluid-solid immiscibility at temperatures below the gel-liquid crystalline transition temperature of the 14:014:0 PE component. Mixtures of 18:2c18:2c PE and 18:1t18:1t PE exhibit complex phase behaviour involving limited fluid-solid immiscibility at low temperatures and formation of a phase allowing isotropic motional averaging at higher temperatures. (4) 31P-NMR provides a graphic method for investigating the miscibility properties of mixed PE systems.  相似文献

Isolation and characterization of isocitrate lyase of castor endosperm   总被引:1,自引:0,他引:1  
Isocitrate lyase (threo-DS-isocitrate glyoxylate-lyase, EC 4.1.3.1) has been purified to homogeneity from castor endosperm. The enzyme is a tetrameric protein (molecular weight about 140,000; gel filtration) made up of apparently identical monomers (subunit molecular weight about 35,000; gel electrophoresis in the presence of sodium dodecyl sulfate). Thermal inactivation of purified enzyme at 40 and 45 °C shows a fast and a slow phase, each accounting for half of the intitial activity, consistent with the equation: At = A02 · e?k1t + A02 · e?k2t, where A0 and At are activities at time zero at t, and k1 and k2 are first-order rate constants for the fast and slow phases, respectively. The enzyme shows optimum activity at pH 7.2–7.3. Effect of [S]on enzyme activity at different pH values (6.0–7.5) suggests that the proton behaves formally as an “uncompetitive inhibitor.” A basic group of the enzyme (site) is protonated in this pH range in the presence of substrate only, with a pKa equal to 6.9. Successive dialysis against EDTA and phosphate buffer, pH 7.0, at 0 °C gives an enzymatically inactive protein. This protein shows kinetics of thermal inactivation identical to the untreated (native) enzyme. Full activity is restored on adding Mg2+ (5.0 mm) to a solution of this protein. Addition of Ba2+ or Mn2+ brings about partial recovery. Other metal ions are not effective.  相似文献

A complete titration of phosphatidic acid bilayer membranes was possible for the first time by the introduction of a new anaologue, 1,2-dihexadecyl-sn-glycerol-3-phosphoric acid, which has the advantage of a high chemical stability at extreme pH values. The synthesis of this phosphatidic acid is described and the phase transition behaviour in aqueous dispersions is compared with that of three ester phosphatidic acids; 1,2-dimyristoyl-sn-glycerol-3-phosphoric acid, 1,3-dimyristoylglycerol-2-phosphoric acid and 1,2-dipalmitoyl-sn-glycerol-3-phosphoric acid.The phase transition temperatures (Tt) of aqueous phosphatidic acid dispersions at different degrees of dissociation were measured using fluorescence spectroscopy and 90° light scattering. The Tt values are comparable to the melting points of the solid phosphatidic acids in the fully protonated states, but large differences exist for the charged states.The Tt vs. pH diagrams of the four phosphatidic acids are quite similar and of a characteristic shape. Increasing ionisation results in a maximum value for the transition temperatures at pH 3.5 (pK1). The regions between the first and the second pK of the phosphatidic acids are characterised by only small variations in the transition temperatures (extended plateau) in spite of the large changes occurring in the surface charge of the membranes. The slope of the plateau is very shallow with increasing ionisation. A further decrease in the H+ concentration results in an abrupt change of the transition temperature. A comparative study is made, at 15 °C, of flash-induced absorption changes around 820 nm (attributed to the primary donors of Photosystems I and II) and 705 nm (Photosystem I only), in normal chloroplasts and in chloroplasts where O2 evolution was inhibited by low pH or by Tris-treatment.At pH 7.5, with untreated chloroplasts, the absorption changes around 820 nm are shown to be due to P-700 alone. Any contribution of the primary donor of Photosystem II should be in times shorter than 60 μs.When chloroplasts are inhibited at the donor side of Photosystem II by low pH, an additional absorption change at 820 nm appears with an amplitude which, at pH 4.0, is slightly higher than the signal due to oxidized P-700. This additional signal is attributed to the primary donor of Photosystem II. It decays (t12 about 180 μs) mainly by back reaction with the primary acceptor and partly by reduction by another electron donor. Acid-washed chloroplasts resuspended at pH 7.5 still present the signal due to Photosystem II (t12 about 120 μs). This shows that the acid inhibition of the first secondary donor of Photosystem II is irreversible.In Tris-treated chloroplasts, absorption changes at 820 nm due to the primary donor of Photosystem II are also observed, but to a lesser extent and only after some charge accumulation at the donor side. Amiloride in nM to μM concentrations stimulates the short circuit current (Isc) of the toad urinary bladder by as much as 120% when applied in conjunction with apical Ca2+ and a divalent cation chelator. A significant decrease in transepithelial resistance (Rt) is observed simultaneously. This response is spontaneously reversible and its amplitude is dependent upon apical sodium concentrations. The stimulated Isc persisted when acetazolamide (1 mM) was introduced, HPO42? substituted for HCO3? or SO42? replaced Cl?. Consequently, the increase in Isc is not due to the change of Cl?, H+ or HCO3? flux. This behavior in a ‘tight’ epithelium may be related to the mechanism controlling apical sodium permeability.  相似文献

Chloroplasts which were rapidly isolated from illuminated leaves showed activity of ATP hydrolysis at a level much higher than that of the dark control. Under the high-intensity illumination or under repetitive flash excitation, the activated chloroplasts synthesized more ATP than those with a low ATP hydrolysis activity. Δ\?gmH+ formed under repetitive flashes was smaller in the activated chloroplasts than in the inactive chloroplasts. The inhibition of ATP yield per flash by valinomycin or nigericin in the presence of K+ was stronger in the inactive chloroplasts than in the activated chloroplast. ATP synthesis in the activated chloroplasts seems to have a lower Δ\?gmH+ threshold.  相似文献

The effects of the prostaglandins PGE1 and PGE2 on the deformability of the human erythrocyte were studied using spin-labeled erythrocytes. Two magnetic resonance parameters were measured: (1) The orientation relaxation time, t12, for the erythrocyte, and (2) the order parameter, S, for a fatty acid spin label bound to the membrane. Prostaglandins PGE1 and PGE2 exhibited opposite effects on both t12 and S. PGE2 made the cell less deformable (increases of t12 and S) and PGE1 made the erythrocyte more deformable (decrease of t12 and S).  相似文献

The risetime of EPR signal IIvf (S IIvf) has been measured in oxygen-evolving Photosystem II particles from spinach chloroplasts at pH 6.0. The EPR signal shows an instrument-limited rise upon induction (t12 ? 3 μs). These data are consistent with a model where the species Z responsible for S IIvf is the immediate electron donor to P-680+ in spinach chloroplasts. Iron uptake by rat reticulocytes is blocked by 20 mM NH4Cl, while 125I-diferric transferrin (Tf) uptake is relatively unaffected. At pH 5.0 both apo- and diferric Tf bind with high affinity; at pH 7.4 diferric Tf binds avidly, but apoTf binds very poorly. The dissociation rate (4°C) of diferric Tf is extraordinarily slow at pH 5.0 (extrapolated t12 = 32 hrs) and faster at pH 7.4 (t12 = 101 min). At pH 5.0 apoTf also dissociates slowly (t12 = 205 min), but at pH 7.4 apoTf exhibits a much faster dissociation rate (t12 = 62 min). 20 mM NH4Cl slows the release of Tf from cells at 37°C, but the rate of externalization of ligand is unaffected. Ligand dissociation at 37° involves both externalization of receptor-ligand complexes and receptor-ligand separation; the NH4Cl effect may result from an increased fraction of externalized Tf in the differric form which may dissociate more slowly. Receptor-mediated movement of Tf through acid intracellular compartments provides a mechanism to remove iron from Tf and for apoTf to remain receptor-bound for externalization to the cell surface and subsequent dissociation.  相似文献

The Km(app) of ADP for photophosphorylation in lettuce chloroplasts was measured both at various light intensities and in the presence of various uncoupler (nigericin + K+) concentrations. Lowering the light intensity results in both, a decrease in the rate of phosphorylation and a several fold decrease in the Km(app) of ADP for the reaction. However, when increasing concentrations of the uncoupler nigericin + K+ are employed, the rate of photophosphorylation is decreased but a several-fold increase in the Km(app) of ADP for the reaction is observed. The results are discussed in terms of the chemiosmotic hypothesis. It is suggested that these effects might indicate the existence of a mechanism controlling the rate of ATP formation which is different than the formation of the electrochemical gradient.  相似文献

Absorption changes (ΔA) at 820 nm, following laser flash excitation of spinach chloroplasts and Chlorella cells, were studied in order to obtain information on the reduction time of the photooxidized primary donor of Photosystem II at physiological temperatures.In the microsecond time range the difference spectrum of ΔA between 750 and 900 nm represents a peak at 820 nm, attributable to a radical-cation of chlorophyll a. In untreated dark-adapted material the signal can be attributed solely to P+?700; it decays in a polyphasic manner with half-times of 17 μs, 210 μs and over 1 ms. The oxidized primary donor of Photosystem II (P+II) is not detected with a time resolution of 3 μs. After treatment with 3–10 mM hydroxylamine, which inhibits the donor side of Photosystem II, P+II is observed and decays biphasically (a major phase with t12 = 20–40 μs, and a minor phase with t12 ? 200 μs), probably by reduction by an accessory electron donor.In the nanosecond range, which was made accessible by a new fast-response flash photometer operating at 820 nm, it was found the P+II is reduced with a half-time of 25–45 ns in untreated dark-adapted chloroplasts. It is assumed that the normal secondary electron donor is responsible for this fast reduction.  相似文献

Unidirectional fluxes of [14C]lactose by whole cells of Escherichia coli under highly energized and partially de-energized (in the presence of CN?) conditions are analyzed kinetically.When the cells are energized, the value for V influx is 0.45 ± 0.01 mM internal concentration increment/s and Kt is 0.26 ± 0.03 mM. At an external concentration of 0.61 mM the steady-state internal concentration is 0.25 M, reached after about 1h. The maximum steady-state concentration ratio is 2 · 103.The efflux process under these conditions is non-saturable, being linearly dependent upon internal concentration over the range 25–250 mM with a first-order rate constant of 8.8 ± 0.2 · 10?4 s?1.The transport in the presence of CN? is active, with a maximum concentration ratio (internal concentration/external concentration) of 104, and the uptake is mimicked by anoxia (< 70 ppm O2).The effects of CN? are to lower the V for influx and to change the efflux from a non-saturable to a saturable process with a value for Kt (60 mM) intermediate between that for energized efflux (> 250 mM) and influxe (0.3–0.6 mM), the latter value not changing appreciably.
