Methodology for the ready analysis of abscisic acid (ABA) in plant tissues based upon application of high-performance liquid chromatography (hplc) has been developed. The method involves isolation of the acid fraction, preparation of the methyl esters with diazomethane, and hplc using a combination procedure of two columns: (1) reversed-phase C18, and (2) porous silica in the absorption mode. Only isocratic elution is required so the method is readily adaptable to laboratories having limited hplc capability. Measured recoveries are 70% and the use of an internal standard allows quantification of ABA levels to 1 ng/g of tissue with minimum absolute detectable levels of ABA of 20 ng. The method is illustrated by analysis of ABA concentration in potato tubers at various times postcutting.  相似文献

In a previous paper [(1976) Anal. Biochem.74, 282] we reported a procedure for the analysis of 25-hydroxycholecalciferol (25-OH-D3) in cow plasma or serum by high performance liquid chromatography. Since then, we obtained synthetic 25-hydroxyergocalciferol (25-OH-D3) and found that it had the same retention time as 25-OH-D3 and interfered with the 25-OH-D3 analysis. We also encountered a relatively small number of samples that did not give sufficiently clean chromatograms by our procedure. This communication details an improvement we have made in the old procedure to overcome these problems.  相似文献

A highly sensitive and selective method for determining a dihydropyridine calcium antagonist, manidipine, by liquid chromatography using column switching with electrochemical detection was developed. Manidipine in serum was extracted by a rapid and simple procedure based on C8 bonded-phase extraction and then silica extraction. Manidipine and nilvadipine as an internal standard were separated on a C8 bonded-phase HPLC column and detected by high conversion efficiency amperometric detection at +0.7 V. Manidipine and nilvadipine (I.S.) were separated from an endogenous interference peak in serum and concentrated on a pre-column (C18) by column switching using an isocratic mobile phase, and then the corresponding fractions were introduced to an analytical column with a C8 stationary phase. Determination of manidipine was possible over the concentration range 0.5–10 ng/ml: the limit of detection was 0.3 ng/ml. The recovery of manidipine added to serum was 93.1–98.4% with coefficients of variation of less than 7.1%. The method is applicable to drug level monitoring in the serum of healthy volunteers treated with manidipine and to the analysis of pharmacokinetics.  相似文献

A method is described for the measurement of prednisone and prednisolone in human plasma by high-performance liquid chromatography (hplc). An ether/dichloromethane extract (60:40, vv) of plasma is evaporated to dryness and chromatographed on a 250 × 4.5-mm Whatman Partisil column with uv detection at 239 nm. Prednisone, prednisolone cortisol, and the internal standard dexamethasone are satisfactorily resolved with the elution system of water-saturated dichloromethane/ethanol (95:4, vv). The hplc method can be used for plasma prednisolone concentrations as low as 25 ng/ml. The values correlate well with those obtained by a radioimmunoassay procedure for prednisolone.  相似文献

A simplified method for the determination of 25-hydroxy and 1α,25-dihydroxy metabolites of vitamins D2 and D3 in human plasma was developed. Plasma samples were deproteinizated and applied to a Bond Elut C18 OH cartridge to separate 25-hydroxyvitamin D (25-OH-D) and 1α-25-dihydroxyvitamin D [1,25(OH)2D] fractions. The 25-OH-D fraction was purified by a Bond Elut C18 cartridge and 25-OH-D2 and 25-OH-D3 were assayed by HPLC using a Zorbax SIL column. The 1,25(OH)2D fraction obtained above was subsequently applied to HPLC using a Zorbax SIL column to separate 1,25(OH)2D2 and 1,25(OH)2D3 fractions which were determined by a radioreceptor assay (RRA) using calf thymus receptor. The method was applied to nutritional studies.  相似文献

A rapid method is described for the identification and determination of amphetamine and methamphetamine in human urine samples by liquid chromatography with UV-Vis detection. The samples were transferred onto a C18 solid-phase extraction column and chromatographed on a Hypersil ODS RP C18, 5 μm (250 × 4 mm I.D.) with an acetonitrile-water elution gradient containing propylamine. Under these conditions, the amines are eluted with a short retention time. The procedure has been applied to the determination of amphetamine and methamphetamine in the range 0.3–4.0 μg/ml in spiked urine samples. The detection limits at 280 nm were 4 and 2 ng/ml for amphetamine and methamphetamine, respectively. The intra-day and inter-day precision and accuracy of the method were studied.  相似文献

  • 1.1. The contents of vitamin D3, 25-hydroxyvitamin D3 (25-OH-D3) and 7-dehydrocholesterol (7-DHC) in 22 kinds of fish liver samples were determined by a high-performance liquid chromatographic (HPLC) method.
  • 2.2. Vitamin D3 was detected in all fish liver samples, but its contents varied from 84 to 264,000 ng/g wet tissue. The liver of fish belonging to Carangidae and Scombridae contained large amounts of the vitamin and therefore we deduced that vitamin D3 levels in liver might have some relations with taxonomical positions of fishes.
  • 3.3. 25-OH-D3 was detected in 7 out of 22 kinds of fish liver samples, while 7-DHC was in 14 out of 22. The contents of the two sterols were generally much lower than those of vitamin D3 and there was no special relationship between the contents of the sterols and the vitamin.

A high-performance liquid chromatographic (hplc) procedure is described for the determination of pyridoxal 5′-phosphate (PLP) in animal tissues. The procedure is based on extraction with perchloric acid and treatment with semicarbazide to form PLP-semicarbazone. This derivative is quantitatively determined using hplc with an octylsilica column, an acidic phosphate mobile phase buffer, and fluorometric detection. The validity of the method was confirmed by inspection of the fluorescence spectra of the PLP-semicarbazone hplc peaks of semicarbazide-treated standards and tissue extracts. Preparative chromatography for extract purification is not required because of the hplc efficiency and detection specificity. This method provides a simple technique for the rapid, direct assay of PLP in animal tissues.  相似文献

The aim of this study was to describe an high-performance liquid chromatographic assay for the simultaneous determination of two HIV protease inhibitors, saquinavir and ritonavir, in human serum. The method involved extraction of ritonavir and saquinavir from serum with the aid of solid-phase extraction C18 cartridges followed by high-performance liquid chromatography with a C8 column and ultraviolet detection set at a wavelength of 240 nm. The assay was linear and has been validated over the concentrations range of 0.5–32 μg/ml for ritonavir and 0.075–4.8 μg/ml for saquinavir, from 600 μl serum extracted. In future, the assay will be used to support human population pharmacokinetic studies, and therapeutic drug monitoring for ritonavir and saquinavir.  相似文献

A high-performance liquid chromatographic screening method (HPLC) is described for the determination of seven selective serotonin reuptake inhibitors (SSRIs) (fluvoxamine, milnacipran, paroxetine, sertraline, fluoxetine, citalopram, venlafaxine) and for three pharmacologically active N-demethylated metabolites (desmethylcitalopram, didesmethylcitalopram and norfluoxetine). A tricyclic antidepressant, clomipramine, was used as an internal standard. The method consists of liquid extraction of serum after alcalinisation at pH 9.50, followed by chromatography on a Beckman C18 reversed-phase column. Compounds were detected at 200.4 nm. The standard curves were linear over a working range of 50–1000 ng/ml for fluvoxamine, 15–1000 ng/ml for fluoxetine, 25–500 ng/ml for norfluoxetine, 50–500 ng/ml for sertraline, 20–500 ng/ml for paroxetine, 25–550 ng/ml for citalopram, 25–750 ng/ml for desmethylcitalopram, 25–800 ng/ml for didesmethylcitalopram, 25–650 ng/ml for milnacipran, and 25–500 ng/ml for venlafaxine. The quantitation limits of the method were 15 ng/ml for fluoxetine, 20 ng/ml for paroxetine, 25 ng/ml for venlafaxine, norfluoxetine and citalopram, and its metabolites, 40 ng/ml for sertraline and 50 ng/ml for fluvoxamine. No interferences were noted with this sensitive and specific method which can be used for therapeutic drug monitoring.  相似文献

An optimized method for the determination of flecainide in serum is presented. Extraction using a solid-phase C18 column and chromatography on a stabilized fluorocarbon-bonded silica gel column effectively separate flecainide from an internal standard (a positional isomer of flecainide). The HPLC apparatus and conditions were as follows: analytical column, Fluofix 120N; sample solvent, 20 μl; column temperature, 40°C; detector, Shimadzu RF-5000 fluorescence spectrophotometer (excitation wavelength=300 nm, emission wavelength=370 nm); mobile phase, 0.06% phosphoric acid containing 0.1% tetra-n-butyl ammonium bromide–acetonitrile (75:25, v/v); flow-rate, 1.0 ml/min. The standard curves for flecainide were linear in the concentration range examined (10–2000 ng/ml). The regression equation was y=0.08+0.0078x (r=0.9998). The minimum detectable amount of flecainide was approximately 5 ng/ml. In the within-day study, the precision coefficients of variation were 2.66, 2.18, 2.54, 2.72, 2.88, 2.24, and 3.29% for the 10, 50, 100, 200, 500, 1000, and 1500 ng/ml standards, respectively. The absolute recovery rates of flecainide at each concentrations were 94–100%. The method described provides analytical sensitivity, specificity and reproducibility suitable for both biomedical research and therapeutic drug monitoring.  相似文献

A HPLC method was developed for the determination of the metabolites of coumarin and 7-hydroxycoumarin in plasma and serum. Separation was based on gradient elution of 7-hydroxycoumarin glucuronide, 7-hydroxycoumarin, coumarin and finally 4-hydroxycoumarin (which is used as an internal standard). Standards, prepared in plasma or serum, and samples were treated with trichloroacetic acid, mixed and centrifuged. The supernatant was removed and analyzed by reversed-phase high-performance liquid chromatography on a C18 column. The limit of detection was 50 ng/ml for 7-hydroxycoumarin and 200 ng/ml for coumarin and 7-hydroxycoumarin glucuronide. The linear range was 0.5–100 μg/ml for each of the analytes. The percentage relative standard deviation about the mean measured concentrations were all below 10%. There was no statistical difference between the standard curves prepared in plasma or serum. The method developed was applied to the determination of each of the three compounds in serum, after the administration of 7-hydroxycoumarin, and in plasma after the administration of coumarin. The concentrations of total 7-hydroxycoumarin in the serum samples were also determined by another HPLC method and the results were compared. There was no statistical difference between the results determined.  相似文献

A method for the quantitative analysis of indomethacin and salicylic acid in blood serum and urine by high-performance liquid chromatography is described. A C18-bonded silica was used as the stationary phase and mixtures of ethanol, n-butanol and aqueous buffer as the mobile phase. Before injection the serum is deproteinized and extracted in one step.The recovery of the extraction was found to be 88% and 77% for indomethacin and salicylic acid, respectively. The relative standard deviations of the analysis for 0.5 μg indomethacin and 5 μg salicylic acid per ml serum were 3.6% and 3.2%, respectively. The detection limits for indomethacin and salicylic acid were 2 ng. This corresponds for both substances to 0.1 μg/ml serum for an injection volume of 100 μl.The method enables simultaneous determination of possibly formed metabolites. A number of concurrently administered drugs do not interfere with the analysis. The interactive effects of co-medication of indomethacin and salicylic acid on the serum concentration of indomethacin is demonstrated by measuring the pharmacokinetic curves.  相似文献

A rapid, reproducible high-performance liquid chromatographic method for the determination of secnidazole, 5-nitroimidazole class of antiprotozoals from blood is described. Metronidazole was used as an internal standard. A simple extraction step with dichloromethane was done before chromatography on a C18 column with the wavelength fixed at 276 nm on the UV detector. Blood levels up to 500 ng/ml have been measured with good precision in the healthy volunteers after 1 g of secnidazole was administered. The present described method can readily be utilized for routine pharmacokinetic studies.  相似文献

A method is given for the determination of carminomycin (CMM) and a major metabolite carminomycinol (CMMOH) in serum from cancer patients after intravenous administration of carminomycin as the free drug.CMM and CMMOH are extracted from serum with chloroform, the extract evaporated and the residue dissolved in methanol. High-performance liquid chromatography analysis utilized a C18 μBondapak reversed-phase column eluted with 0.1 mol/l acetate buffer (pH 4) — acetonitrile (60:40, v/v) with fluorescence detection. The assay is linear, reproducible, and precise with a limit of detection of 2 ng/ml. Representative serum levels of CMM and CMMOH in a cancer patient are presented.  相似文献

A simplified high-pressure liquid chromatographic method for determination of furose-mide in plasma and urine has been developed using a fluorometric detector directly coupled to the column effluent. The method includes an ether extraction from acidified biologic samples. The mobile phase used for chromatography on a reversed-phase column (C18 hydro-carbon permanently bonded to silica particles) is sufficiently acidic to induce fluorescence of furosemide. The methylester of furosemide is employed as an internal standard. The sensitivity is 0.1 and 0.25 μg per ml plasma and urine, respectively. The applicability to pharmacokinetic studies of furosemide is shown.  相似文献

A routine method for the determination of a novel class III antidysrhythmic agent, 1-(4-methane-sulphonamidophenoxy)-2-[N-(4-methanesulponamidophenethyl)-N-methylamino]ethane, in human urine has been developed. The method involves solvent extraction followed by high-performance liquid chromatography on an unmodified silica column with ultraviolet detection. Despite a low recovery of drug through the three-stage extraction procedure a reliable assay with high precision (coefficient of variation less than 6%) and a limit of determination of 2.5 ng/ml was achieved. The method has been applied to the analysis of samples following single oral and intravenous doses of 1–12.5 μg/kg of the drug to human volunteers.  相似文献

A high-performance liquid chromatography (hplc) solvent system was developed for use with C18 bonded packings that effected the chromatographic resolution of monoaromatic hydroxamic acids in complex mixtures. The success of this hplc system resulted from the incorporation of an aliphatic hydroxamic acid additive into the elution solvent. This additive, desferal mesylate, suppressed chemisorption effects between the stationary phase of the chromatographic packing and the aromatic hydroxamic acids being analyzed. The detectability limit for acetyl-derived aromatic hydroxamic acids was about 10 ng, while that for glycolyl-derived aromatic hydroxamic acids was about 5 ng. This level of sensitivity was sufficiently low to allow for the direct detection of these components in enzymatic reaction mixutres. By manipulation of the percentage methanol in the aqueous solvent the resolution of either type of hydroxamic acid from other related metabolites was readily accomplished.  相似文献

A rapid clean-up procedure based on ion-pair solid-phase extraction (SPE) for the high-performance liquid chromatographic (HPLC) determination of spectinomycin in swine, calf and chicken plasma at a limit of detection of 50 ng/ml is described. After dilution with water and adjustment of the pH to approximately 5.6, the plasma is applied to a high-hydrophobic C18 SPE column treated with sodium dioctylsulphosuccinate. Spectinomycin is eluted with methanol and derivatized with 2-naphthalene sulphonyl chloride prior to chromatography. The HPLC set-up consists of a dual-column system using two Chromspher silica columns and dichloromethane—acetonitrile—ethyl acetate—acetic acid, in different ratios, as mobile phases. Detection is performed at 250 nm. Quantification is carried out using external standards prepared in blank cleaned plasma. Mean recoveries were 83 ± 3% (n = 5), 93 ± 6% (n = 5) and 92 ± 6% (n = 6) for swine, calf and chicken plasma, respectively, at the 0.1 μg/ml level.  相似文献

A specific and selective high-performance liquid chromatography (HPLC) technique that requires few manipulations, and is readily adaptable to analysis for a large series of samples, has been developed for the determination of the concentration of the anticancer drug doxorubicin (DXR) in rat serum and tissues. The biological samples were efficiently deproteinised and resolved from a reversed-phase nucleosil C18 column with fluorescence detection. The validation study of the proposed method was successfully carried out in an assay range of between 5 and 5000 ng/ml and was subsequently implemented in a pharmacokinetic study of DXR in Wistar rats that were treated by intravenous administration of the drug.  相似文献
