Evaluating corticosterone (CORT) responses to stress in free-living vertebrates requires knowing the unstressed titers prior to capture. Based upon laboratory data, the assumption has been that samples collected in less than 3 min of capture will reflect these unstressed concentrations. This assumption was tested for six species using samples collected from 945 individuals at 0-6 min after capture. Samples were from five avian species trapped at multiple times of year and one reptilian species, comprising a total of 14 different data sets for comparisons. For seven of 14 data sets, including five species, there was no significant increase in corticosterone titers within 3 min of capture. In six of the 14 data sets, corticosterone titers increased significantly after 2 min, and in one data set, the increase started at 1.5 min. In all seven of the cases showing an increase before 3 min, however, corticosterone titers from the time of increase to 3 min were significantly lower than titers after 30 min of restraint stress. These results indicate a high degree of confidence for these species that samples collected in less than 2 min reflect unstressed (baseline) concentrations, and that samples collected from 2-3 min also will likely reflect baseline concentrations but at worst are near baseline.  相似文献

Several free-living avian species have recently been shown to seasonally modulate corticosterone release in response to capture and restraint. We examined possible mechanisms underlying seasonal adrenocortical modulation in snow buntings (Plectrophenax nivalis), a species that breeds and molts (the energetically costly replacement of feathers) in the Alaskan Arctic. Snow buntings dramatically reduced baseline and maximal corticosterone titers during molt compared to the breeding season. This effect is not explained by changes in either corticosterone binding protein capacity or the overall condition of the bird (assessed by weight and fat storage). Although the adrenal's capacity to secrete corticosterone is reduced during molt, adrenal insensitivity does not fully explain reduced maximal output since exogenous adrenocorticotropic-hormone enhanced corticosterone release during both seasons. In contrast, no exogenous adrenocorticotropic hormone releasing factor (corticotrophin-releasing factor, arginine vasotocin or mesotocin) enhanced corticosterone secretion during molt. This suggests that the pituitary's endogenous adrenocorticotropic secretion was maximal in response to capture and handling, making the pituitary an important site regulating corticosterone levels. Taken together, these results indicate that seasonal modulation of corticosterone release in this species is controlled at both the adrenal and pituitary glands. Accepted: 16 February 1998  相似文献

We measured plasma levels of testosterone, corticosterone, and cortisol in free-living male yellow-pine chipmunks to demonstrate the patterns of seasonal variation and to assess the effects of capture and handling on hormone levels. We achieved the latter by modifying our standard trapping technique (blood samples collected within 1–3 h of capture) to obtain blood samples that allowed measurement of hormone levels within 3 min of capture (basal) and again 30 min later. By alternating the modified and standard trapping techniques over 7 months of the active season we demonstrated that seasonal patterns of variation in steroid hormone levels can be accurately described with the simpler, standard trapping technique. Basal and 30-min post-capture testosterone levels were high during mating and dropped to a persistently low level thereafter. Conversely, both cortisol and corticosterone were at their seasonal low during mating and climbed to peak levels in June following reproduction. Plasma glucocorticosteroid levels increased during the 30 min after capture and handling at all times of the active season, and these elevated levels were similar to the levels obtained by standard trapping. Testosterone levels during the mating period also increased in response to capture and handling. The contrasting patterns of seasonal variation in glucocorticosteroid and testosterone levels and the changes induced by capture and handling suggest that when testosterone concentration is high, adrenocortical activity is suppressed. Fecal hormone assays provide a powerful tool for noninvasive monitoring of endocrine status in wild animals. In this study we validated a protocol for extracting and measuring glucocorticoids in free-living and captive Belding's ground squirrels (Spermophilus beldingi). We first compared two commonly used extraction protocols to determine which performed better with commercially available antibodies. We next verified the preferred extraction method by correlating circulating and fecal glucocorticoid measures from a group of individuals over time. For this comparison, we used both a cortisol and a corticosterone antibody to determine which had greater affinity to the fecal metabolites. Cortisol was the primary circulating glucocorticoid, but both hormones were present in well above detectable concentrations in the blood, which does not occur in other sciurids. In addition, the cortisol antibody showed greater binding with the fecal extracts than did the corticosterone antibody. Finally, we used adrenocorticotropic hormone and dexamethasone challenges to demonstrate that changes in adrenal functioning are reflected in changing fecal corticoid levels. These results suggest that our extraction protocol provides a fast, reliable assay of stress hormones in free-living ground squirrels without the confounding influence of short-term rises in glucocorticoid concentrations caused by handling and restraint stress and that it can facilitate ecological and evolutionary studies of stress in wild species.  相似文献

In response to a variety of unpredictable conditions, birds secrete the steroid hormone corticosterone, which has numerous effects on physiology and behavior. A standardized protocol of handling and restraint has been demonstrated to elicit a robust corticosterone response in many species of birds. In contrast, comparatively little is known about the effects of capture technique on corticosterone secretion in wild birds. Setting up multiple live traps checked at regular intervals allows field researchers to capture many birds in a short period of time. However, one potential drawback of this technique is that birds may spend unknown lengths of time in traps prior to sampling. Many birds appear to remain calm and/or feed during this period, potentially leading researchers to assume that corticosterone secretion is unaffected by trapping. We assessed the impact of being left in traps for up to 30 minutes on baseline corticosterone and subsequent corticosterone responses to restraint in non‐breeding and breeding house sparrows Passer domesticus. Traps were baited with seed, and birds were either removed immediately after entry (controls), or left in the trap undisturbed for 15 or 30 min. Upon removal, birds were subjected to a standardized handling/restraint protocol in which blood samples were collected within 3 min, and again at regular intervals for 60 min. Analysis of blood samples revealed that both non‐breeding and breeding sparrows that were held in the traps had significantly higher baseline corticosterone than controls, and showed no further increase in corticosterone secretion in response to handling. However, corticosterone responses to trapping differed seasonally. Our study indicates that although birds did not exhibit prolonged escape behavior while trapped, entry into a walk‐in trap initiated a robust stress response. Taken together with data from a previous study, our data suggest that ornithologists should consider species‐specific and stage‐specific effects of trapping on physiology when designing field studies.  相似文献

We use museum and other collection records to document large and extraordinarily rapid changes in the ranges and relative abundance of nine species of mammals in the northern Great Lakes region (white-footed mice, woodland deer mice, southern red-backed voles, woodland jumping mice, eastern chipmunks, least chipmunks, southern flying squirrels, northern flying squirrels, common opossums). These species reach either the southern or the northern limit of their distributions in this region. Changes consistently reflect increases in species of primarily southern distribution (white-footed mice, eastern chipmunks, southern flying squirrels, common opossums) and declines by northern species (woodland deer mice, southern red-backed voles, woodland jumping mice, least chipmunks, northern flying squirrels). White-footed mice and southern flying squirrels have extended their ranges over 225 km since 1980, and at particularly well-studied sites in Michigan's Upper Peninsula, small mammal assemblages have shifted from numerical domination by northern species to domination by southern species. Repeated resampling at some sites suggests that southern species are replacing northern ones rather than simply being added to the fauna. Observed changes are consistent with predictions from climatic warming but not with predictions based on recovery from logging or changes in human populations. Because of the abundance of these focal species (the eight rodent species make up 96.5% of capture records of all forest-dwelling rodents in the region and 70% of capture records of all forest-dwelling small mammals) and the dominating ecological roles they play, these changes substantially affect the composition and structure of forest communities. They also provide an unusually clear example of change that is likely to be the result of climatic warming in communities that are experienced by large numbers of people.  相似文献

Information on the widespread function of the suprachiasmatic (SCN) pacemaker has increased dramatically in the past decade for laboratory rodents and for humans. Interest is now also growing in the adaptive value of SCN pacemakers in wild species of free-living mammals in natural pristine habitat. The squirrel family, Sciuridae, is highly diverse in North America. Radiation into many specialized groups has evolved in response to the temporal and spatial characteristics of specific habitats. The sciurids thus offer possibilities for comparison of SCN function for closely related species from very different habitats. Results from field and laboratory investigations concerning the ecological significance of the SCN are reported here for three ground squirrel species. The semi-fossorial antelope squirrels of the American southwest deserts were arrhythmic in above ground activities after deletion of the SCN pacemaker. In a desert enclosure, predation rates rose dramatically for lesioned animals in comparison to intact controls. The semi-arborial chipmunks were prone to capture by mustelid burrow predators such as weasels after SCN-deletion, even though the chipmunks did not exhibit any above ground activity at night. The telltale cue for the predator was apparently arrhythmic restlessness within the complex home burrow. The semi-fossorial golden-mantled squirrels of the Cascade Mountains of western United States rely on profound, long-lasting hibernation for over-wintering. Periods of torpor alternate with brief euthermic arousal episodes lasting less than 24 h throughout the 5- to 8-month winter phase. Deletion of the SCN in golden-mantled squirrels resulted in aberrations of hibernation arousal patterns that were metabolically expensive and put individuals at risk of starvation immediately after emergence in spring.  相似文献

Little is known about baseline concentrations of adrenal hormones and hormonal responses to stress in sea ducks, although significant population declines documented in several species suggest that sea ducks are exposed to increased levels of environmental stress. Such declines have been observed in geographically distinct harlequin duck populations. We performed an adrenocorticotropic hormone (ACTH) challenge to evaluate adrenal function and characterize corticosterone concentrations in captive harlequin ducks and investigated the effects of capture, surgery, and short term confinement on corticosterone concentrations in wild harlequin ducks. Harlequin ducks responded to the ACTH challenge with an average three-fold increase in serum corticosterone concentration approximately 90 min post injection, and a four- to five-fold increase in fecal glucocorticoid concentration 2 to 4 h post injection. Serum corticosterone concentrations in wild harlequin ducks increased within min of capture and elevated levels were found for several hours post capture, indicating that surgery and confinement maintain elevated corticosterone concentrations in this species. Mean corticosterone concentrations in wild harlequin ducks held in temporary captivity were similar to the maximum response levels during the ACTH challenge in captive birds. However, large variation among individuals was observed in responses of wild birds, and we found additional evidence suggesting that corticosterone responses varied between hatch year and after hatch year birds.  相似文献

Corticosterone, a glucocorticoid secreted during stress responses, has a range of actions that help birds respond to stressors. Although effects of corticosterone treatment have been described in several avian species, the impacts of defined increases in plasma corticosterone on early development and on corticosterone stress responses are little known. These issues were addressed by providing quail with different doses of corticosterone in drinking water from days 8 to 38 post-hatch. The corticosterone dose consumed by each bird during treatment days 15-30 was calculated by measuring water intake. The corticosterone dose was inversely, but weakly, correlated with weights of the bursa, thymus, spleen, liver, testes, oviduct, muscle, and body, and positively correlated with peritoneal fat deposition. When birds were divided into groups based on their corticosterone intake, weights of the spleen, thymus, bursa, muscle, testes, and oviduct were significantly reduced in birds receiving the highest doses; with the exception of muscle, similar reductions were also observed in birds receiving medium doses, and thymic growth was inhibited in birds receiving low doses. The acute corticosterone stress response was measured by handling birds for 15 min. Plasma corticosterone was transiently increased at 15 min in control birds in response to the handling stressor. Some birds consuming low doses of corticosterone had corticosterone responses similar to control birds. Initial corticosterone concentrations were elevated in birds consuming higher doses of corticosterone. Plasma corticosterone in these birds decreased from 0 to 15 min, then increased from 15 to 30 min. The initial decrease could be due to corticosterone clearance, whilst the increase could indicate that the birds had a greater response than control birds to isolation as a stressor. Corticosterone treatment may have reduced the strength of corticosterone negative feedback within the hypothalamo-pituitary-adrenal axis. The results indicate that individuals and organs differ in their sensitivity to corticosterone. Moreover, elevated plasma corticosterone may disrupt the acute corticosterone stress response, and may thus reduce the ability of birds to cope with stressors.  相似文献

Brown lemmings reach much higher densities than collared lemmings near Barrow, Alaska, and captures from 19 summers of snap trapping confirm previous reports that brown lemmings prefer lower, wetter habitats than do collared lemmings. Data also support the hypothesis that brown lemmings concentrate in higher habitats during early summer when melt water floods lower habitats, then shift to lower habitats where preferred food is more available as the waters recede. This pattern appears similar to seasonal shifts in habitat use reported for Norwegian lemmings. Two hypotheses were not supported by our data: (1) Unlike Norwegian lemmings, brown lemmings did not expand their use of suboptimal habitats at higher population densities. Rather, absolute densities changed in concert so that the relative densities among habitats remained unchanged. (2) Preferential use of polygon troughs during winter, as indicated by patterns of winter grazing, was not simply a function of snow depth. Instead it appeared to be related to shoot density of preferred foods. Nearly all patterns of habitat use seemed to be linked to food availability. Other factors, such as protection from predators by vegetative cover in summer and increased insulation from deeper snow in winter, did not appear to influence the distribution of lemmings as strongly.  相似文献

The topologies of phylogenetic trees characterized by a high level of intraspecific divergence between the phylogenetic DNA groups (clades) are often explained in terms of the theory of Pleistocene refugia. To elucidate the issue of the adaptive role of intraspecific divergence, the changes in the physicochemical properties of amino acids in the course ofcladogenesis (MM01 model of the TreeSAAP 3.2 package) were analyzed in this work using as an example the nucleotide sequences of the cytochrome b gene in some species of northern animals (lemmings, redbacked voles, chipmunks, flying squirrels, ermines). It was shown that the process of intraspecific divergence was rarely accompanied by radical amino acid substitutions in cytochrome b caused by adaptation (directional selection). In connection with this, the hypothesis is discussed according to which the adaptive variants formed in the species at the peak of cold were lost with climatic warming due to the drift or selection against individuals adapted to cold.  相似文献

Rising temperatures and changes in the precipitation regime will have a strong impact on the quality of the snow cover in the Arctic. A snow cover of good quality protecting lemmings from cold temperatures and predators is thought to be an important factor for maintaining the cyclic dynamic of their populations in the tundra. We examined if the characteristics of annual fluctuations (amplitude and shape of phases) in brown lemming (Lemmus trimucronatus) density could be determined by snow depth, snow density, sub-nivean temperature and persistence of snow. Using an 18-year time series of brown lemming abundance on Bylot Island in the Canadian Arctic, we tested if snow variables could explain the residual variation between the observed lemming density and the one predicted by models where cyclicity had been accounted for. Our analysis provides support for the hypothesis that snow cover can affect the amplitude and possibly also the periodicity of lemming population cycles in the High Arctic. Summer abundance of brown lemmings was higher following winters with a deep snow cover and a low-density snow pack near the ground but was unaffected by the date of establishment or melting and duration of the snow cover. Two snow variables showed a temporal trend; mean winter snow depth tended to increase and date of establishment of the hiemal threshold occurred earlier over time. These temporal trends, which should be favourable to lemmings, may explain why healthy population cycles have apparently been maintained at our study site contrary to other Arctic sites.  相似文献

Postnatal hippocampal neurogenesis in wild mammals may play an essential role in spatial memory. We compared two species that differ in their reliance on memory to locate stored food. Yellow-pine chipmunks use a single cache to store winter food; eastern gray squirrels use multiple storage sites. Gray squirrels had three times the density of proliferating cells in the dentate gyrus (determined by Ki-67 immunostaining) than that found in chipmunks, but similar density of young neurons (determined by doublecortin immunostaining). Three explanations may account for these results. First, the larger population of young cells in squirrels may increase the flexibility of the spatial memory system by providing a larger pool of cells from which new neurons can be recruited. Second, squirrels may have a more rapid cell turnover rate. Third, many young cells in the squirrels may mature into glia rather than neurons. The densities of young neurons were higher in juveniles than in adults of both species. The relationship between adult age and cell density was more complex than that has been found in captive populations. In adult squirrels, the density of proliferating cells decreased exponentially with age, whereas in adult chipmunks the density of young neurons decreased exponentially with age.  相似文献

Acute glucocorticoid elevations can be adaptations to short-term stressors. The breeding season hypothesis predicts reduced glucocorticoid responsiveness to acute stressors in populations or species with short breeding seasons. The striped plateau lizard (Sceloporus virgatus) has a short breeding season in Arizona. We measured plasma corticosterone and total androgen levels (dihydrotestosterone and testosterone) following one of the four stress-handling treatments (0, 10, 60, or 180 min). In both sexes, longer handling stress yielded higher corticosterone; females had higher corticosterone than males at all time points. Androgens did not vary with handling duration, in either sex. Combining treatments, plasma androgens correlated positively with corticosterone (CORT) in females but not in males; plasma CORT and body mass residuals were negatively correlated in both sexes, suggesting lizards in poor body condition and/or not investing heavily in reproduction (follicle mass) have higher acute corticosterone. Total plasma androgens and body mass residuals were positively associated in males, but showed no association in females. The maximal CORT elevation after handling stress in this single-clutching species was of comparable magnitude to responses in related multi-clutching lizard species with longer breeding seasons. Using data from studies of multiple populations of three Sceloporus species, we found no relationship between the relative magnitude of the CORT increase and either latitude or elevation, two variables in the literature correlated with duration of the breeding season, and only weak relationships with geographic elevation and actual (not relative) stress-elevated CORT values in this multi-population comparison.  相似文献

Stress and reproduction are generally thought to work in opposition to one another. This is often manifested as reciprocal relationships between glucocorticoid stress hormones and sex steroid hormones. However, seasonal differences in how animals respond to stressors have been described in extreme environments. We tested the hypothesis that garter snakes, Thamnophis sirtalis, with limited reproductive opportunities will suppress their hormonal stress response during the breeding season relative to conspecifics with an extended breeding season. The red-sided garter snake, T.s. parietalis, of Manitoba, Canada, has a brief breeding season during which males displayed no change in either plasma levels of testosterone or corticosterone, which were both elevated above basal levels, in response to capture stress. During the summer, capture stress resulted in increased plasma corticosterone and decreased testosterone. During the fall, when mating can also occur, males exhibited a significant decrease in testosterone but no increase in corticosterone in response to capture stress. The red-spotted garter snake, T.s. concinnus, of western Oregon, has an extended breeding season during which males displayed a stress response of increased plasma corticosterone and decreased testosterone levels. The corticosterone response to capture stress was similar during the spring, summer, and fall. In contrast, the testosterone response was suppressed during the summer and fall when gametogenesis was occurring. These data suggest that male garter snakes, in both populations, seasonally adapt their stress response but for different reasons and by potentially different mechanisms. J. Exp. Identifying and understanding why traits make species vulnerable to changing climatic conditions remain central problems in evolutionary and applied ecology. We used spring snow cover duration as a proxy for phenological timing of wetland ecosystems, and examined how snow cover duration during spring and during the entire snow season affected population dynamics of duck species breeding in the western boreal forest of North America, 1973–2007. We predicted that population level responses would differ among duck species, such that late‐nesting species with reduced flexibility in their timing of breeding, i.e. scaup (Aythya spp.) and scoter (Melanitta spp.), would be more strongly affected by changing snow cover conditions relative to species better able to adjust timing of breeding to seasonal phenology, i.e. mallard (Anas platyrhynchos) and American wigeon (Anas americana). Population growth rates of scaup and scoter were positively linked to spring snow cover duration; after accounting for effects of density dependence, larger populations resulted after springs with long snow cover duration than after springs with short snow cover duration. In contrast, population growth rates of mallard and wigeon were either negatively or only weakly associated with snow cover duration. Duck population models were then incorporated with snow cover duration derived from climate model simulations under the A2 emission scenario, and these predictions suggested that late‐nesting duck species will experience the most severe population declines. Results are consistent with a hypothesis that the gradual climatic warming observed in the western boreal forest of North America has contributed to and may continue to exacerbate population declines of scaup and scoter.  相似文献

It is common knowledge that winter temperatures influence the life history of small mammals. Cold temperatures necessitate increased energy requirements for survival, and recent studies indicate that snow cover can have both negative and positive influences. With each new observation, we develop a more comprehensive understanding of the mechanisms that influence small mammal populations. Here we report on our recent study on Japanese field vole Microtus montebelli, which reaches its peak in population during the early spring and its low during the autumn. To understand the population dynamics of these voles, we conducted a capture-mark-recapture survey, then estimated the seasonal abundance, recruit, capture probabilities, and survival probabilities using the Bayesian hierarchical model. We also analyzed the impact of mammalian generalist predator visits on the survival probabilities. Our data indicates that the early spring peak in population is due to intensive winter breeding and the highest survival probabilities during the periods of deep snow cover. When snow cover reaches a certain depth, the circumstances can combine to raise survival probability and favor breeding. During the breeding season in May and June, on the contrary, the survival probability reached its lowest, resulting in a decrease in population despite breeding. The low survival probability between spring and autumn could be attributed to the impact of generalist predators, and low vegetation may have amplified the effect. In summary, the deep snow cover and generalist predators were considered to be the key factors shaping this unique population dynamics in this orchard area.  相似文献

Mixed hardwood forests of the northeast United States support a guild of granivorous/omnivorous rodents including gray squirrels (Sciurus carolinensis), eastern chipmunks (Tamias striatus), and white-footed mice (Peromyscus leucopus). These species coincide geographically, co-occur locally, and consume similar food resources. Despite their idiosyncratic responses to landscape and patch variables, patch occupancy models suggest that competition may influence their respective distributions and abundances, and accordingly their influence on the rest of the forest community. Experimental studies, however, are wanting. We present the result of a large-scale experiment in which we removed white-footed mice or gray squirrels from small, isolated forest fragments in Dutchess County, New York, and added these mammals to other fragments in order to alter the abundance of these two species. We then used mark–recapture analyses to quantify the population-level and individual-level effects on resident mice, squirrels, and chipmunks. Overall, we found little evidence of competition. There were essentially no within-season numerical responses to changes in the abundance of putative competitors. Moreover, while individual-level responses (apparent survival and capture probability) did vary with competitor densities in some models, these effects were often better explained by site-specific parameters and were restricted to few of the 19 sites we studied. With only weak or nonexistent competition among these three common rodent species, we expect their patterns of habitat occupancy and population dynamics to be largely independent of one another.  相似文献
