脂多糖诱导的肿瘤坏死因子(Lipopolysaccharide-induced TNF-alpha factor,LITAF)是一类重要的炎症细胞因子,在先天性免疫系统中发挥重要的介质作用。文章根据虾夷扇贝LITAF基因EST序列,应用RACE技术克隆了虾夷扇贝LITAF全长cDNA,对序列及编码的氨基酸进行生物信息学分析。结果显示,该基因cDNA全长1 551 bp,其5′非编码区包含76 bp,3′非编码区包含1 001 bp;开放阅读框(ORF)为474 bp,编码157个氨基酸,氨基酸序列中存在一个保守的LITAF结构域;理论分子量16.99 kDa,等电点为6.24。LITAF基因序列为3 698 bp,由3个外显子和两个内含子组成。利用实时荧光定量PCR技术分析LITAF在虾夷扇贝不同组织、不同胚胎发育阶段以及鳗弧菌(Vibrio anguillarum)刺激后各时间段的表达情况。结果表明:LITAF基因在所检测的6个成体组织中均有表达,其中肾脏的表达量最高;胚胎发育的7个时期中,担轮幼体时期表达量最高;菌刺激36 h实验组与对照组的表达量差异大。LITAF基因是LITAF家族的一员,推测LITAF基因参与虾夷扇贝的先天性免疫反应。  相似文献

The retaining endo-1,3-β-d-glucanase (LV) with molecular mass of 36 kDa was purified to homogeneity from the crystalline styles of scallop Mizuhopecten yessoensis. The purified enzyme catalyzed hydrolysis of laminaran as endo-enzyme forming glucose, laminaribiose and higher oligosaccharides as products (Km  600 μg/mL). The 1,3-β-d-glucanase effectively catalyzed transglycosylation reaction that is typical of endo-enzymes too. Optima of pH and temperature were at 4.5 and 45 °C, respectively. cDNA encoding the endo-1,3-β-d-glucanase was cloned by PCR-based methods. It contained an open reading frame that encoded 339-amino acids protein. The predicted endo-1,3-β-d-glucanase amino acid sequence included a characteristic domain of the glycosyl hydrolases family 16 and revealed closest homology with 1,3-β-d-glucanases from bivalve Pseudocardium sachalinensis, sea urchin Strongylocentrotus purpuratus and invertebrates lipopolysaccharide and β-1,3-glucan-binding proteins.The fold of the LV was more closely related to κ-carrageenase, agarase and 1,3;1,4-β-d-glucanase from glycosyl hydrolases family 16. Homology model of the endo-1,3-β-d-glucanase from M. yessoensis was obtained with MOE on the base of the crystal structure of κ-carrageenase from P. carrageonovora as template. Putative three-dimensional structures of the LV complexes with substrate laminarihexaose or glucanase inhibitor halistanol sulfate showed that the binding sites of the halistanol sulfate and laminarihexaose are located in the enzyme catalytic site and overlapped.  相似文献

A normalized cDNA library was constructed from the adductor muscle of M. yessoensis and acquired 4595 high quality expressed sequence tags (ESTs). After clustering and assembly of the ESTs, 3061 unigenes containing 654 contigs and 2407 singletons were identified. The contig length ranged from 266 bp to 2364 bp and the average length of these contigs was 544 bp. Blastx nonredundant protein database analysis showed that 1522 unigenes had significant homology to known genes (E value ≤ 10? 5). By comparing to Clusters of Orthologous Groups (COG) categories, 460 unigenes were annotated (E value ≤ 10? 10). Using Kyoto Encyclopedia of Genes and Genomes (KEGG), 345 of 3061 unigenes were assigned into 103 pathways (E value ≤ 10? 5). For InterProScan searches, 1237 unigenes were annotated containing 727 different types of protein domains. 941 of the 1237 unigenes were annotated for Gene Ontology (GO) classification using Uniprot2GO associations in any category (biological, cellular, and molecular). By sequences comparability and analysis of Blastx NCBI nonredundant protein database and KEGG, 66 unigenes were identified that may be involved in genetic information processing based on the known knowledge. The study provides a material basis as useful information for the genomic analysis of shellfish.  相似文献

Ten novel microsatellite loci were isolated from giant scallop Mizuhopecten yessoensis, and the polymorphisms were examined to estimate genetic variability. The genetic variabilities varied depending on the locus. The number of alleles ranged from two to 17, and the observed and expected heterozygosities ranged from 0.17 to 0.99 and 0.33 to 0.90, respectively. Six loci showed significant Hardy–Weinberg disequilibrium. The high variabilities revealed in this study suggest that microsatellites should prove useful for various genetic investigations.  相似文献

Composition of neurons, their structure and neuromediatory specialization in the Japanese scallop ganglia have been studied by means of morphological, morphometrical and histochemical methods. Three groups of neurons, differing in their histophysiological characteristics have been defined. Large neurons are cholinergic ones. A well developed system of smooth membranes and large amount of cytosomes are specific for them. Middle size and small neurons of the I type contain a granular endoplasmic reticulum, elementary neurosecretory peptide granules and are considered as peptidergic. Small neurons of the II type possess increased nuclear-plasmic relations, moderately developed endoplasmic reticulum, positively react to biogenic amines and according to their mediatory specialization are monoaminergic.  相似文献

Age-related variation in the absolute and relative weights of the adductor has been studied for 8 years in six samples of the Japanese scallop (Mizuhopecten yessoensis Jay) of different generations from a shellfish farm in Alekseev Bight, Sea of Japan. The test character has proved to vary widely within the samples, with variation in shell size being relatively low. It has been shown that the dependence of adductor weight on the age of the scallop is not absolute: equally aged samples from different generations may differ significantly in this character. It is assumed that this effect is due to the influence of environmental factors on the development of scallops at early stages, which is especially strong under conditions of dense spat planting in the course of cultivation.  相似文献

【目的】研究苜蓿夜蛾Heliothis viriplaca保幼激素酯酶(Juvenile hormone esterase,JHE)的功能和作用机理。【方法】本试验提取苜蓿夜蛾的总RNA,并利用RT-PCR和RACE技术,扩增得到苜蓿夜蛾保幼激素酯酶基因的全长c DNA序列,命名为Hvjhe(Gen Bank登录号:JQ901384)。【结果】该基因含有3 106 bp,包括一个1 746 bp的开放阅读框,编码一个含581个氨基酸的多肽,分子量约为63.9 ku,多肽的等电点为5.11,且氨基酸序列具含有5个保幼激素酯酶特有的氨基酸保守模块。推导的氨基酸序列与烟芽夜蛾Heliothis virescens和棉铃虫Helicoverpa armigera中获得的保幼激素酯酶相似性较高,分别达到83%和82%。荧光定量PCR结果表明,该基因在苜蓿夜蛾不同发育时期和不同组织中都有m RNA水平的特异性表达,且在预蛹期表达量相对较高,取食期、蛹期期和成虫期表达量相对较低;在中肠和脂肪体内的表达量相对于其它组织较高。该基因在大肠杆菌Escherich coli表达系统中进行了诱导表达,经SDS-PAGE和Western blot检测结果表明,表达出与预测的蛋白分子量相符的融合蛋白。【结论】研究结果表明本试验获得了苜蓿夜蛾中一个新的保幼激素酯酶基因的c DNA序列。  相似文献

虾夷扇贝(Patinopecten yessoensis)5个群体的遗传多样性   总被引:21,自引:0,他引:21  
虾夷扇贝为20世纪80年代初从日本引入我国并逐渐开展养殖的双壳贝类,目前已在我国北方地区大面积养殖。实验采用微卫星分子遗传标记技术对大连獐子岛底播增殖放流群体(CC)、黄海北部海区采集的野生群体(HQ)、日本青森养殖群体(JX)、俄罗斯远东日本海沿岸养殖群体(RX)及大连大长山岛养殖上壳白化群体(ZB)等5个虾夷扇贝群体的遗传多样性进行研究。其中HQ群体为本课题组2005年在黄海北部采集的野生群体,本研究筛选出一个该群体的特异性遗传标记。用8个微卫星位点进行扩增,共获得45个等位基因,每个位点的等位基因数处于3—9之间,大小为100—340bp,平均有效等位基因数为3.1535,基因型数为3—21个,PIC(PolymorphismInformationContent)值处于0.0322-0.5944之间。5个群体的平均观测杂合度分别为0.3292、0.3048、0.3167、0.2708、0.3042,平均期望杂合度分别为0.4595、0.4002、0.3838、0.3620、0.3885,群体间的多态性差异不显著。根据群体间遗传相似性系数、遗传距离及UPGMA聚类分析发现,CC和HQ群体亲缘关系最近,JX和RX群体的亲缘关系较近,ZB群体与JX和RX群体的亲缘关系较近。通过Hardy—Weinberg平衡及F-检验发现,5个群体都不同程度的偏离平衡,表明各群体基因频率和基因型频率的稳定性较低,且5个群体均处于不同程度的杂合子缺失状态,群体间的遗传分化程度较高,但遗传变异主要来自群体内的个体间。  相似文献

利用RT-PCR和RACE方法,获得了棉铃虫Helicoverpa armigera酚氧化酶原(prophenoloxidase,PPO)基因一个亚型cDNA的完整序列。该序列全长2 405 bp,含有一个2 097 bp的开放阅读框,编码一个由698个氨基酸残基组成的蛋白质。推导的氨基酸序列与其他鳞翅目昆虫PPO2基因相应氨基酸序列有较高的同源性(76%~80%),同时该序列具有铜离子结合位点等PPO基因所具有的典型特征。组织特异性表达分析表明,该基因在棉铃虫血细胞、体壁和中肠中均有表达。  相似文献

Heat shock protein 90 (HSP90) is a highly conserved molecular chaperone contributing to the folding, maintenance of structural integrity and proper regulation of a subset of cytosolic proteins. The full-length cDNA of Zhikong scallop Chlamys farreri HSP90 (designated CfHSP90) was cloned by EST and rapid RACE techniques. It was of 2710 bp, including an open reading frame (ORF) of 2181 bp encoding a polypeptide of 726 amino acids with all the five HSP90 family signatures. BLAST analysis revealed that the CfHSP90 gene shared high similarity with other known HSP90 genes. Fluorescent real-time quantitative RT-PCR was used to examine the expression pattern of CfHSP90 mRNA in haemocytes of scallops exposed to Cd2+, Pb2+ and Cu2+ for 10 and 20 days, respectively. All the three heavy metals could induce CfHSP90 expression. There was a clear dose-dependent expression pattern of CfHSP90 after heavy metals exposure for 10 days or 20 days. Different concentrations of the same metal resulted in different effects on CfHSP90 expression. The results indicated that CfHSP90 responded to various heavy metal stresses with a dose-dependent expression pattern as well as exposure time effect, and could be used as a molecular biomarker in a heavy metal polluted environment.  相似文献
