Summary Transport of indolyl-3-acetic acid (IAA) was studied during the phototropic responses of intact shoots and detached coleoptiles of Zea mays L. and Avena sativa L. The use of a high specific activity [5-3H]IAA and glass micropipettes enabled asymmetric application of the IAA to be made to individual coleoptiles with minimal tissue damage.A unilateral stimulus of 2.59×10-11 einstein cm-2 of blue light, probably in the dose range of the first positive phototropic response, caused significant net lateral movement of radioactivity from [5-3H]IAA away from the illuminated side of intact shoots and detached coleoptile apices of both Avena and Zea. The magnitude of the net lateral movement was 15.3% in Zea seedlings and 12.3% in Avena seedlings. Chromatographic analyses indicated that the movement of radioactivity reflected that of IAA. A phototropic stimulus of 1.24×10-7 einstein cm-2, which was probably in the second positive dose range, caused significant lateral movement of radioactivity in intact shoots and detached coleoptiles of Zea but not of Avena.In intact Zea seedlings, neither phototropic dosage affected the longitudinal transport of IAA. In intact Avena seedlings, first positive stimulation inhibited longitudinal transport only when the IAA was applied to the illuminated side of the coleoptile, but second positive stimulation inhibited basipetal movement of IAA regardless of the side of application.Exposing the intact seedlings to red light before phototropic stimulation abolished lateral transport after a first positive stimulus in Zea and in Avena.Phototropic stimulation can thus induce a lateral transport of IAA towards the shaded side of the coleoptiles of both Zea and Avena seedlings, and can affect longitudinal movement of IAA in the coleoptile of Avena. However, since phototropic curvature was observed under certain conditions in the absence of either of these effects, the extent to which they are involved in the induction of asymmetric growth in a stimulated coleoptile has yet to be resolved.  相似文献

Summary Movement of IAA was studied in excised coleoptile apices and whole seedlings of Zea mays L. and Avena sativa L. during geotropic stimulation. A micropipette technique permitted the application of [5-3H]IAA at predetermined points on the coleoptiles with minimal tissue damage.When [5-3H]IAA was applied to the upper side of a horizontal excised Zea coleoptile, about 60% of the recoverable radioactivity had moved into the lower half after 2 h. In contrast, when application was made to the lower side of a horizontal excised coleoptile, only 4% of the radioactivity migrated to the upper half. There was, thus, a net downward movement of 56%. Similar patterns of distribution were found for radioactivity in both the tissue and the basal receiver blocks. In horizontal shoot tissues of intact Zea seedlings a net downward movement of about 30% of the recoverable radioactivity occurred after 1 h of geotropic stimulation. Comparable experiments with Avena indicated a net downward movement of 6–12% in excised apices of coleoptiles and in the intact shoot. In both Zea and Avena chromatographic analyses of tissue and receiver blocks indicated that the movement of radioactivity reflected that of IAA.In Zea coleoptiles, the lateral migration of radioactivity after 2 h was 3 to 4 times greater in the apical tissues than in the basal tissues. A significant net downward movement of radioactivity was detected after 10 min of geotropic stimulation in the extreme apex of Zea coleoptiles but not in the more basal regions.These experiments show that downward lateral transport of IAA occurs in intact shoots of Zea and Avena seedlings upon geotropic stimulation. Lateral transport of IAA had previously been demonstrated only in sub-apical segments of Zea coleoptiles.  相似文献

The vertical elongation of normal roots of Lupinus seedlings proceeds at constant rate over periods of 4 to 5 hours. The decapitation of a root stops its elongation for a variable length of time, followed by a period of renewed elongation at a rate lower than that of the normal root. The tipping of the decapitated root with a tip of a coleoptile of Zea induces a decrease in the rate of elongation of the root. The same effect can be obtained with the diffusate from tips of coleoptile of Avena and to a lesser extent with diffusate of root tips. The reduction in the rate of elongation of the root determined by diffusate from the lower half of the tip of a coleoptile placed horizontally is more pronounced than the inhibition elicited by the diffusate of the upper half of the same tip. Various experiments with the diffusate of tips support the idea that under the conditions used the growth-promoting substance of the coleoptile tip or root tip inhibits the elongation of the decapitated root.  相似文献

Light inhibition of primary root elongation in Zea mays seedlings was found to be an almost instantaneous response and the inhibitory effect was shown to persist for at least six hours following a short exposure to white light. Removal of the root cap completely removed the inhibitory effect of light. The apical region of the root appeared to be mainly responsible for perceiving light and initiating the production of factors involved in the inhibition of elongation. Extraction of the roots of light-exposed Zea seedlings revealed the presence of the carotenoid pigments, violaxanthin, neoxanthin and lutein, together with an unidentified non-acidic growth-inhibitory compound. The movement of kinetin has been investigated in roots of Zeaseedlings, grown in the dark, using kinetin-8-14C. Segments (10 and 11 mm) with and without an apex were used andthe following results obtained: The transport of kinetin is polarized basipetally. Kinetin-8-14C is transported in roots without vascular tissue,but the amount transported is always lower than in roots withvascular tissue. We explored the different mechanisms developed by naturally regenerated seedlings of Quercus ilex L. (Holm oak) under Mediterranean conditions compared to container-seedlings commonly used in plantations. We examined the differences in root architecture (including topology and morphology) and shoot parameters. The results showed that there are many differences in the architecture of the root system as well as in the shoot morphology between the two types of seedlings. The naturally regenerated seedlings were smaller with regard to most of the shoot and root parameters, but they developed a longer taproot, only first order lateral roots, and presented a more herringbone-like root system compared to the container seedlings. Conversely, all types of container seedlings, were larger and had a more extended root system with many orders of lateral roots, while their taproot length was restricted within the container’s depth. The quotient log (α)/ log (μ) for all seedlings, showed a tendency to decrease with plant size. A strict herringbone root system with an elongated taproot may be the optimal root architecture for Quercus ilex L. seedlings in order to survive under Mediterranean conditions.  相似文献

The effect of osmotic stress on wheat (Triticum aestivum L.) mitochondrial activity and phospholipid composition was investigated. Preliminary growth measurements showed that osmotic stress (−0.25 or −0.5 megapascal external water potential) inhibited the rate of shoot dry matter accumulation while root dry matter accumulation was less sensitive. We have determined that differences in sensitivity to osmotic stress existed between tissues at the mitochondrial level. Mitochondria isolated from roots or shoots of stressed seedlings showed respiratory control and ADP/O ratios similar to control seedlings which indicates that stressed mitochondria were well coupled. However, under passive swelling conditions in a KCl reaction mixture, the rate and extent of valinomycin-induced swelling of shoot mitochondria were increased by osmotic stress while root mitochondria were largely unaffected. Active ion transport studies showed efflux transport by stressed-shoot mitochondria to be partially inhibited since mitochondrial contraction required the addition of N-ethylmaleimide or nigericin. Efflux ion transport by root mitochondria was not inhibited by osmotic stress which indicates that stress-induced changes in ion transport were largely limited to shoot mitochondria. Characterization of mitochondrial fatty acid and phospholipid composition showed an increase in the percentage of phosphatidylcholine in stressed shoot mitochondria compared to the control. Mitochondrial fatty acid composition was not markedly altered by stress. No significant changes in either the phospholipid or fatty acid composition of stressed root mitochondria were observed. Hence, these results suggest that a tissue-specific response to osmotic stress exists at the mitochondrial level.  相似文献

The main objective of the study was to compare the effects of short-duration pH treatments on root hydraulic properties in trembling aspen (Populus tremuloides) seedlings that were either inoculated with the ectomycorrhizal fungus Hebeloma crustuliniforme or remained non-inoculated (control). Inoculated and non-inoculated plants were exposed in solution culture to the root zone pH ranging from 4 to 9 and their root hydraulic conductivity was examined using the hydrostatic method and after subjecting the plants to treatments with 100 ??M HgCl2 (aquaporin blocker) and 0.02% trisodium 3-hydroxy-5,8,10-pyrenetrisulfonic acid (apoplastic transport tracer). In a separate experiment, pure cultures of H. crustuliniforme were also grown on a slid medium with the pH ranging from 4 to 9 to determine their pH growth optimum and changes in medium pH over time in the presence and absence of 8 mM NH4NO3. When grown in pure culture, H. crustuliniforme demonstrated maximum growth at pH 7?C8 and was capable of modifying the pH of its growth media, especially in the presence of NH4NO3. The plants that were inoculated with H. crustuliniforme had a maximum root hydraulic conductivity at pH 7. At this pH, root hydraulic conductivity was significantly higher compared with non-inoculated plants and showed greater sensitivity of root water transport to pH changes relative to non-inoculated seedlings. Relative apoplastic flux was largely unaffected by pH in inoculated seedlings. Fungal inoculation modified the response of root hydraulic conductivity to pH. The increased root hydraulic conductivity in inoculated seedlings was likely due to an increase in aquaporin-mediated cell-to-cell water transport, particularly at the higher pH. A possible role of fungal aquaporins in the root hydraulic conductivity responses of mycorrhizzal plants should be examined.  相似文献

Oxamyl was coated on carrot (Daucus carota L. cv. Spartan Fancy-80) and tomato (Lycopersicon esculentum Mill. cv. Glamour) seeds with a polymer sticker for the control of Meloidogyne hapla. The sticker diluted in water 1:1 delayed carrot seedling emergence. Oxamyl at 40 mg/ml in a 1:5 dilution of sticker lowered the rate of carrot seedling emergence until day 13 and plant growth until day 28. Oxamyl at 20 or 40 mg/ml in a 1:5 dilution of sticker on carrot seeds planted in M. hapla-infested muck soil resulted in fewer galled tap roots and fewer galls per root system 4 weeks after planting. Tap root lengths were greater than those of the control. Tomato seedling emergence was delayed and top and root weights were reduced, relative to the control, at 25 days by the sticker diluted 1:1 to 1:3. Oxamyl at 20 or 40 mg/ml in a 1:5 diluted sticker delayed tomato seedling emergence. Top weights of tomato seedlings from seeds coated with 20 mg/ml of oxamyl in a 1:5 diluted sticker planted in a silt loam were greater than control top weights at 4 and 6 weeks. Root weights were greater than those of the control only at 4 weeks. There were fewer galls per gram of root on seedlings from oxamyl-coated seeds and fewer juveniles per pot of soil, relative to the controls, only at 4 weeks.  相似文献

The quantities of endogenous indol-3yl-acetic acid (IAA) in endosperms and scutella of 6-day-old maize seedlings (Zea mays L. cv Giant White Horsetooth) were determined by a fluorimetric method. Endosperms were found to contain 33.4 nanograms IAA per plant, and scutella 7.5 nanograms IAA per plant. [5-3H]IAA applied to endosperms of 6-day-old seedlings moved into the roots and radioactivity accumulated at the apex of the primary root within 8 hours. Two to 7-day-old seedlings were treated simultaneously with [5-3H]IAA in the endosperm and [2-14C] IAA on the shoot apex. The patterns of transport into the root were found to change during ontogeny: in successively older plants, transport from the shoot into the roots increased relative to transport from the endosperm into the roots. The auxin required for the growth of maize roots could, therefore, partially be contributed by the shoot and endosperm. Ontogenetic changes in the relative importance of these two supplies could be of significance for the integration of growth and development between shoot and root.  相似文献

An isotope-dilution method has been developed for the assay of free indole-3-acetic acid and ester indole-3-acetic acid as measured by indole-3-acetic acid liberated by mild alkaline hydrolysis. Application of this method to seedlings of Avena sativa and Zea mays indicates the upper limit of free indole-3-acetic acid in Avena to be about 16 μg per kg and in Zea, about 24 μg. The amount of 1 n alkali-labile indole-3-acetic acid in Zea is about 330 μg per kg and there is very little 1 n alkali-labile IAA in Avena. A chemical characterization of the indole-3-acetic acid of Avena and a confirmation of the chemical characterization of the indole-3-acetic acid of Zea is presented.  相似文献

The rate of auxin transport in sunflower hypocotyls (Helianthus annuus L. cv `Russian mammoth') or corn coleoptiles (Zea mays L. cv `WF9 × 38') was less in seedlings grown in Ca-deficient medium than in controls. The rate of IAA transport depended on the concentration of Ca in the root medium up to 1 millimolar. Further increases in auxin transport were observed when the isolated segments were incubated in medium containing up to 30 millimolar Ca. We suggest that the rate of auxin transport in plant tissue is dependent on the pool of ionic Ca in the extracellular space.

Segments from Ca-deficient seedlings exhibited a high specific requirement for Ca2+ in auxin transport. Magnesium, strontium, and several other divalent cations tested for their ability to replace Ca2+ in restoring auxin transport showed no effect; partial replacement by lanthanum was observed.

Auxin transport, or auxin flux through the segment, which is the result of IAA secretion by individual cells, was reduced in the low Ca2+ segments due both to lowered velocity and to reduced capacity of transport. The requirement for Ca2+ in the secretion of auxin is believed to be equivalent to the phenomenon observed in animal cell secretion, where the influx of Ca2+ serves as a link between an external stimulus and the secretion response.

To investigate the possible transport of polyamines and their precursor amino acids, 14C-labeled putrescine, spermidine, arginine, or lysine were injected into cotyledons of 4-day etiolated pea (Pisum sativum L. cv Alaska) seedlings. After 4 hours the shoot, root, and cotyledons were homogenized and the extracted, dansylated polyamines separated by thin-layer chromatography. Little radioactivity was transported from the cotyledons when [14C]putrescine or [14C]spermidine were injected and of the radioactivity in the axis, none could be recovered as polyamines. Injection of [14C]arginine or [14C]lysine, on the other hand, led to a significant transport of radioactivity into the axis, of which a large fraction was present in the form of the diamines, putrescine or cadaverine, respectively. These results indicate that polyamines in the growing regions of etiolated pea seedlings probably arise from transport and conversion of amino acid precursors.  相似文献

The effect of bilateral irradiation with white light (1000 Meter Candle Sec) on the basipetal transport of auxin has been investigated. Illumination of either the intact shoot or the excised coleoptile tip of the Zea seedling, decreased the amount of diffusible auxin obtained from the tip, and decreased Avena curvature response to unilaterally applied indoleacetic acid. Irradiation of the intact Zea seedling did not affect the absorption of 14C-labeled indoleacetic acid from an agar block subsequently placed on the decapitated coleoptile. However, light caused a significant decrease in the amount of labeled auxin basipetally transported, without affecting materially the velocity of that transport. These and other observations are interpreted as support for the hypothesis that the primary hormonal phenomenon in first-positive phototropism is a light-induced impairment in the basipetal transport of auxin.  相似文献

Radiolabeled oxindole-3-acetic acid was metabolized by roots, shoots, and caryopses of dark grown Zea mays seedlings to 2,3-dihydro-7-hydroxy-2-oxo-1H indole-3-acetic acid-7′-O-β-d-glycopyranoside with the simpler name of 7-hydroxyoxindole-3-acetic acid-glucoside. This compound was also formed from labeled indole-3-acetic acid supplied to intact seedlings and root segments. The glucoside of 7-hydroxyoxindole-3-acetic acid was also isolated as an endogenous compound in the caryopses and shoots of 4-day-old seedlings. It accumulates to a level of 4.8 nanomoles per plant in the kernel, more than 10 times the amount of oxindole-3-acetic acid. In the shoot it is present at levels comparable to that of oxindole-3-acetic acid and indole-3-acetic acid (62 picomoles per shoot). We conclude that 7-hydroxyoxindole-3-acetic acid-glucoside is a natural metabolite of indole-3-acetic acid in Z. mays seedlings. From the data presented in this paper and in previous work, we propose the following route as the principal catabolic pathway for indole-3-acetic acid in Zea seedlings: Indole-3-acetic acid → Oxindole-3-acetic acid → 7-Hydroxyoxindole-3-acetic acid → 7-Hydroxyoxindole-3-acetic acid-glucoside.  相似文献

Plants respond to low nutrient availability by modifying root morphology and root system topology. Root responses to nitrogen (N) and phosphorus (P) limitation may affect plant capacity to withstand water stress. But studies on the effect of nutrient availability on plant ability to uptake and transport water are scarce. In this study, we assess the effect of nitrogen and phosphorus limitation on root morphology and root system topology in Pistacia lentiscus L seedlings, a common Mediterranean shrub, and relate these changes to hydraulic conductivity of the whole root system. Nitrogen and phosphorus deprivation had no effect on root biomass, but root systems were more branched in nutrient limited seedlings. Total root length was higher in seedlings subjected to phosphorus deprivation. Root hydraulic conductance decreased in nutrient-deprived seedlings, and was related to the number of root junctions but not to other architectural traits. Our study shows that changes in nutrient availability affect seedling water use by modifying root architecture. Changes in nutrient availability should be taken into account when evaluating seedling response to drought.  相似文献

The absorption of K+ by excised roots of barley (Hordeum vulgare L. cv California Mariout) has been systematically compared with that of entire, undisturbed seedlings. Some experiments have also been done with wheat (Triticum aestivum L.) and an amphiploid obtained from a cross between it and salt-tolerant tall wheatgrass (Lophopyrum elongatum Host Löve [syn. Agropyron elongatum Host]). For all three genotypes, the rate of K+ absorption measured in a 20-min period was identical for entire 8-d-old seedlings and their excised roots within the experimental error. Manipulation gentler than root excision, viz. careful transfer of seedlings from one experimental solution to another, was also without effect on the rate of K+ absorption. Absorption of K+ measured by assay of its 86Rb label in the tissue was identical with that measured by K+ depletion of the experimental solutions assayed chemically. For the plant materials and conditions of these experiments, the excised root technique for studying ion transport into roots is validated. The advantages of the technique, and findings differing from the present ones, are discussed.  相似文献

Sugar beet (Beta vulgaris L. cv. Monogerm C.S.F. 1971) seeds sown into Vineland fine sandy loam, infested with 15,500 H. schachtii juveniles/pot, showed little growth during an 11-week test in the greenhouse. Seedlings transplanted at 2, 4, and 6 weeks of age had 32, 30, and 31% less top weight and 71, 68, and 59% less root weight, respectively, compared to controls grown in nematode-free soil. Nematode reproduction in both direct-seeded and transplanted sugar beets was limited and related to root weight. Shoot/root ratios were increased by the nematodes in all nematode-infected beets compared to those grown in soil without nematodes. In contrast to seeding or transplanting sugar beets into nematode-infested Vineland fine sandy loam, an inoculation of Beverly fine sandy loam supporting 0 (seeds), 2-, 4-, and 6-week-old sugar beet seedlings with 7,400 juveniles/pot, followed by 11 weeks of growth in the growth-room, resulted in top weight losses of only 13, 3, 18, and 15% and losses in root weight of 44, 38, 36, and 38%, respectively. Nematode reproduction was high and all shoot/root ratios were increased by the nematode compared to the noninoculated controls. These experiments have shown that sugar beets sown into nematode-infested soil are damaged much more heavily by H. schachtii juveniles than seeds inoculated with the nematode immediately following sowing. Results indicate that an increase in tolerance of sugar beets to attack by H. schachtii does not occur beyond the first 2 weeks of growth and that transplanting damage lowers the tolerance of seedlings to nematode attack.  相似文献
