In southern Udmurtia, where occur both Dermacentor reticulatus and Ixodes persulcatus, in the years of high abundance of small mammals feeding periods of larvae and nymphs of these ticks coincide completely. The duration of simultaneous parasitism of preimago of the above ixodid species on animals is of epidemiological significance as far as within this period the infection of young stages of D. reticulatus with the virus of tick-borne encephalitis occurs. This is confirmed by the isolation of I strain of the virus from D. reticulatus collected at the state of engorged nymphs on hosts common with I. persulcatus and moulted in the laboratory.  相似文献

Sexual transmission of tick-borne encephalitis virus from infected ixodid males to noninfected females is shown: in Ixodes persulcatus in 50% (6 of 12) and in Hyalomma anatolicum in 6.2% (1 of 16). The sexual transmission of tick-borne encephalitis virus is shown to provide a transmissible transfer of the virus into eggs in ixodid ticks. Electron microscope studies of the sexual system of ixodid males infected with the virus have revealed numerous morphologically mature virus particles in lumens of endoplasmic reticulum, in vacuoles of Golgi complex of spermatocytes and in association with tubular elements of spermatids.  相似文献

Rickettsiae, obligate intracellular Gram-negative bacteria, responsible for mild to severe diseases in humans are associated with arthropod vectors. Dermacentor marginatus and Dermacentor reticulatus are known vectors of Rickettsia slovaca and Rickettsia raoultii distributed across Europe. A total of 794 D. marginatus, D. reticulatus and Ixodes ricinus adult ticks were collected from the vegetation, removed from horses, sheep, goats and dogs in Slovakia. The DNA of Rickettsia sp. was found in 229 ticks by PCR amplifying parts of gltA, ompA and sca4 genes. Next analyses of Rickettsia-positive samples by PCR-RFLP and/or sequencing showed D. reticulatus ticks were more infected with R. raoultii and D. marginatus were more infected with R. slovaca. The prevalence of R. raoultii was 8.1-8.6% and 22.3-27% in D. marginatus and D. reticulatus, respectively. The prevalence of R. slovaca was 20.6-24.3% in D. marginatus and 1.7-3.4% in D. reticulatus. Intracellular growth of R. raoultii isolate from D. marginatus tick was evaluated by rOmpA-based quantitative SybrGreen PCR assay. The highest point of multiplication was recorded on the 7th and 8th day postinfection in Vero and L929 cells, respectively. R. raoultii was transmitted during feeding of R. raoultii-positive ticks to guinea pigs and subsequently rickettsial infection was recorded in all organs, the highest infection was in spleen, liver and heart. Our study describes the detection and isolation of tick-borne pathogens R. raoultii and R. slovaca, show that they are spread in Slovakia and highlight their risk for humans.  相似文献

To investigate the relationships between tick-borne encephalitis (TBE) virus and the bacterial spirochaete Borrelia burgdorferi sensu lato in vectors with mixed infections, unfed adult Ixodes persulcatus ticks were collected by flagging from vegetation in southern-taiga forests of the Pre-Urals region of Russia where both infections circulate sympatrically. Prevalences of TBE and Borrelia infections in a total of 4234 ticks were compared over 5 years. No significant differences were revealed between the prevalence of Borrelia infection in ticks with and without TBE virus (29.4+/-7.8% vs 23+/-3.6%), or between the prevalence of TBE virus infection in ticks with and without Borrelia (24.0+/-6.6% vs 18.4+/-3.4%). In ticks with mixed infection (40/689 = 5.8%), concentrations of TBE virus and Borrelia were not significantly correlated with one another. Field observations showed parallel trends in the prevalence of these pathogens in tick populations from year to year (1993-1997) indicating that, in I. persulcatus with mixed infection, Borrelia and TBE virus do not seem to interfere with each other and are apparently not involved in any antagonistic relationships.  相似文献

Background  

Large regions of central and eastern Europe are recognized as areas where tick-borne encephalitis virus (TBEV) is endemic, including countries neighbouring Denmark. It is therefore timely and relevant to determine if TBEV infections occur in Denmark. This study investigates the presence of antibodies against TBEV in a cross-section of the Danish canine population to assess the level of exposure to TBEV and possibly identify TBEV microfoci in Denmark.  相似文献

An attempt of chacteristization of tick-borne encephalitis virus strains, isolated in Poland in 1955--1970, was based on the determination and analysis of neurovirulence for white mice at intracerbral and subcutaneous virus inoculation, neuroinvasion, antigenic properties, cytopathogenic abilities, susceptibility to nonspecific hemagglutination inhibitors, susceptibility to temperature of 50degree and to 2M urea. Strains isolated in different regions of the country showed certain differentiation in pathogenic and antigenic properties as well in susceptibility to nonspecific inhibitors.  相似文献

The species status of Dermacentor marginatus and Dermacentor reticulatus was evaluated by scanning electron microscope (SEM) examination of adult ticks, cross-breeding experiments and molecular biological analysis of eggs derived from transspecific pairings. The SEM investigations including the morphometric quantification of phenotypic features resulted in an unequivocal differentiation of adult D. marginatus and D. reticulatus ticks. The cross-breeding experiments demonstrated that irrespective of whether female ticks of both species were applied with con- or transspecific male ticks or without males to sheep, they engorged and laid eggs. The larvae, however, developed only in eggs which originated from conspecific matings. A nested polymerase chain reaction (PCR) of the second internal transcribed spacer (ITS2) using the DNA of eggs from transspecific pairings and sequencing of the PCR products revealed two different genotypes. The genotypes of eggs originating from D. marginatus and D. reticulatus females of these pairings differed. However, the eggs deposited by D. marginatus always possessed the same two genotypes as did the eggs produced by D. reticulatus. These results argue for a strict reproductive isolation of D. marginatus and D. reticulatus and, therefore, for a separate species status.  相似文献

Abstract. A total of 924 questing Dermacentor reticulatus (Fabricius), 504 Ixodes ricinus (L.), sixty Haemaphysalis concinna Koch and 718 mosquitoes (Aedes spp.) were examined in a floodplain forest ecosystem during the 1994-95 outbreak of tularaemia in South Moravia, Czech Republic. Francisella tularensis was not isolated from H.concinna ticks or Aedes spp. mosquitoes, whereas twenty-one isolates were recovered from the other haematophagous arthropods. Dermacentor reticulatus revealed a significantly higher infection rate (2.6%) than I.ricinus (0.2%). This tick species acts as principal vector for tularaemia in the enzootic focus. Monitoring of D.reticulatus for F.tularensis thus seems to be a very efficient approach in the surveillance of tularaemia in the flood-plain forest ecosystems of Europe.  相似文献

The decisive effect of physiological age of ticks on the infection of their saliva was revealed by means of parenteral infection of D. marginatus females with tick-borne encephalitis virus. The virus was not found in the saliva of young individuals of instar II. Maximum number of cases of saliva infection was recorded in mature ticks of instar III (50%). It was established that the infection of saliva did not depend on the amount of virus in the tick's body. Less susceptibility to the virus of young individuals, associated probably with the way of infection, was noted. Under light microscopy there were observed no significant age differences in salivary glands of ticks of instars II and III. The infection of saliva of young individuals is, apparently, opposed by the barrier of fat body on the way of virus to salivary glands, depending on the age of ticks. The barrier is supposed to have an influence under natural transphase infection of ticks.  相似文献

Females and males of ixodid ticks (Ixodes and Dermacentor) infected parenterally with the tick-borne encephalitis virus at the adult or nymphal phase and containing the virus in the saliva are able to transmit the agent in the first minutes after the bite of the sensitive animal host. Preservation of the cement conus produced by saliva in the animal skin enhances the infection intensity because the quantity of the virus in the conus is comparable with that in the tick's body. The virus quantity in the fluid saliva, excreted by I. persulcatus females and measured during different periods of bloodsucking (at least during the first three days), increases 10 to 100 times in comparison with a comparable volume of hungry ones.  相似文献

We present the data of 12-year survey (1989-2001) of the red vole population in southeastern West Siberia, including estimation of host relative numbers, abundance of immature taiga ticks, and percentage of animals with antigemagglutitnins against tick-borne encephalitis (TBE) virus. We discuss the role of demographic groups of voles as tick's hosts and their participation in the maintenance of TBE causative agent population. The estimation of spontaneous TBE infection rate in summer as well as in winter and early spring seasons, which have been made using a set of molecular-biological, serological and virological methods, demonstrates that a high proportion of red voles maintain non-pathogenic TBE causative agent over a long time, presumably, in the form of persistent infection.  相似文献

The ticks Ixodes persulcatus and I. pavlovskyi collected from people visited gardens and suburban forests have been examined by the IFA methods on the presence of tick-borne encephalitis virus (TEV). It was established, that the most part of ticks collected from humans belongs to I. persulcatus, despite the fact that I. pavlovskyi dominates on the territory of the city and its suburbs. TEV infection was registered more often in fed ticks, in comparison with those without signs of preceding feeding. Infected specimens constituted 9.4% of 53 I. persulcatus individuals examined. In I. pavlovskyi 3.7% of 27 examined specimens were infected. In fed ticks (41 specimens of I. persulcatus and 20 I. pavlovskvi) the rates of infected specimens were higher and constituted 48.8 and 35%, respectively. In suburban forests, as well as in the city, I. persulcatus was found to attack humans more often than I. pavlovskyi. TEV infection was recorded in 12.7% of 220 unfed I. persulcatus specimens examined and in 41.6% of 142 fed individuals. In I. pavlovskyi, 6.6% of unfed and 25% of fed specimens was infected (33 unfed and 24 fed specimens were examined). Thus, in all territories examined, percent of infected specimens was significantly lower in I. pavlovskyi, and in fed ticks of both species, TEV was recorded more often.  相似文献

The water content, the survival time at various relative humidities(r.h.) and the critical equilibrium activity of unfed adultDermacentor marginatus and D.reticulatus ticks were investigated at a constant temperature of20 °C. It was also examined whether these ticks use liquidwaterto compensate water loss. Both Dermacentor spp. showed nosignificant differences in water content in relation to body mass. The meanwater content of D. marginatus and D.reticulatus was 54.6% and 54.7%, respectively, in females and 56.3%and 57.0%, respectively, in males. The survival time of unfed adults prolongedwith decreasing saturation deficits. On average, males survived longer thanfemales and D. marginatus ticks survived mostly longerthanD. reticulatus ticks. The 50% mortality period rangedbetween 40 d at 33% r.h. and 420 d at 95% r.h. in D.marginatus, and between 43 d at 33 r.h. and 366 d at 95% r.h. inD. reticulatus. The critical equilibrium activity of unfedadults was estimated to be 0.84 for both species and was independent of sex.When dehydrated adult D. marginatus and D.reticulatus ticks were offered liquid water, only a few slightlygained weight while most further lost weight. Liquid water was not attractivefor dehydrated or non-dehydrated ticks and drinking was not observed. Aftersubmerging in water for 2 d, most of the dehydrated ticks had gained weight.  相似文献

Worldwide, ticks are important vectors of human and animal pathogens. Besides Lyme Borreliosis, a variety of other bacterial and protozoal tick-borne infections are of medical interest in Europe. In this study, 553 questing and feeding Ixodes ricinus (n = 327) and Dermacentor reticulatus ticks (n = 226) were analysed by PCR for Borrelia, Rickettsia, Anaplasma, Coxiella, Francisella and Babesia species. Overall, the pathogen prevalence in ticks was 30.6% for I. ricinus and 45.6% for D. reticulatus. The majority of infections were caused by members of the spotted-fever group rickettsiae (24.4%), 9.4% of ticks were positive for Borrelia burgdorferi sensu lato, with Borrelia afzelii being the most frequently detected species (40.4%). Pathogens with low prevalence rates in ticks were Anaplasma phagocytophilum (2.2%), Coxiella burnetii (0.9%), Francisella tularensis subspecies (0.7%), Bartonella henselae (0.7%), Babesia microti (0.5%) and Babesia venatorum (0.4%). On a regional level, hotspots of pathogens were identified for A. phagocytophilum (12.5–17.2%), F. tularensis ssp. (5.5%) and C. burnetii (9.1%), suggesting established zoonotic cycles of these pathogens at least at these sites. Our survey revealed a high burden of tick-borne pathogens in questing and feeding I. ricinus and D. reticulatus ticks collected in different regions in Belarus, indicating a potential risk for humans and animals. Identified hotspots of infected ticks should be included in future surveillance studies, especially when F. tularensis ssp. and C. burnetii are involved.  相似文献

The process of oviposition in D. reticulatus was observed and found to be a sequence of exactly coordinated, interlocking events independent of the phase of oviposition. The average period of oviposition in the investigated ticks was 31.6 days at 20 °C and 95% relative humidity. The number of eggs deposited on each day increased until reaching a maximum on the fifth day of oviposition and then decreased continuously. As a result, most of the eggs were deposited during the initial phase of oviposition. The total number of eggs was proportional to the ticks' weight replenishment. Egg-laying commenced with the lowering of the capitulum and the simultaneous spread of the pedipalps which were lowered to the body wall embracing the genital aperture on both sides. Immediately afterwards the cuticular sac of Gene's organ was pushed out and retracted several times. At the cuticular sac's maximum extension, the vestibulum vaginae prolapsed, forming the ovipositor as an extended tube which handed over an egg to the two horns of the cuticular sac after a brief, but intensive, contact with the cuticular sac. Then the vestibulum vaginae invaginated, the pedipalps closed, and the cuticular sac was retracted. Finally, the egg was transported onto the dorsal area of the tick by means of a vigorous rising of the capitulum. During the course of oviposition most of the events, especially the period of egg embracement by the cuticular sac, were prolonged, as was the total time for laying one egg. Similarly, the intervals between successive egg-laying processes increased continuously.The number of eggs deposited was not dependent on the functional ability of Gene's organ, as shown by similar numbers of deposited eggs from ticks with and without mechanical blocking of the cuticular sac. But the participation of the organ in the process of oviposition proved to be a prerequisite for the viability of the eggs. Larvae developed and hatched only from those eggs which were deposited from ticks with an undisturbed Gene's organ. In comparison, eggs without contact to the cuticular sac of Gene's organ dried up and shrivelled immediately after being deposited and did not hatch. Consequently, it strongly suggests, together with the results from other studies, that Gene's organ covers the eggs with a secretion that prevents the loss of water.  相似文献
