Blossom-end rot (BER) in tomato fruit (Solanum lycopersicum) is believed to be a calcium (Ca(2+) ) deficiency disorder, but the mechanisms involved in its development are poorly understood. Our hypothesis is that high expression of pectin methylesterases (PMEs) increases Ca(2+) bound to the cell wall, subsequently decreasing Ca(2+) available for other cellular functions and thereby increasing fruit susceptibility to BER. The objectives of this study were to evaluate the effect of PME expression, and amount of esterified pectins and Ca(2+) bound to the cell wall on BER development in tomato fruit. Wild-type and PME-silenced tomato plants were grown in a greenhouse. At full bloom, flowers were pollinated and Ca(2+) was no longer provided to the plants to induce BER. Our results show that suppressing expression of PMEs in tomato fruit reduced the amount of Ca(2+) bound to the cell wall, and also reduced fruit susceptibility to BER. Both the wild-type and PME-silenced fruit had similar total tissue, cytosolic and vacuolar Ca(2+) concentrations, but wild-type fruit had lower water-soluble apoplastic Ca(2+) content and higher membrane leakage, one of the first symptoms of BER. Our results suggest that apoplastic water-soluble Ca(2+) concentration influences fruit susceptibility to Ca(2+) deficiency disorders.  相似文献

Although gibberellins (GAs) have been shown to induce development of the physiological disorder blossom-end rot (BER) in tomato fruit (Solanum lycopersicum), the mechanisms involved remain largely unexplored. BER is believed to result from calcium (Ca) deficiency, but the relationship between Ca content and BER incidence is not strong. Our objectives were to better understand how GAs and a GA biosynthesis inhibitor affect BER development in tomato fruit. Tomato plants of two BER-susceptible cultivars, ‘Ace 55 (Vf)’ and ‘AB2,’ were grown in a greenhouse environment and subjected to Ca-deficiency conditions. Plants were treated weekly during fruit growth and development with 300 mg L?1 GA4+7, 300 mg L?1 prohexadione-calcium (Apogee®, a GA biosynthesis inhibitor), or water beginning 1 day after flower pollination. GA4+7 treatment induced an increase in BER incidence in both cultivars up to 100%, whereas ‘Ace 55 (Vf)’ and ‘AB2’ plants treated with Apogee did not show BER incidence. The number of functional xylem vessels was higher in the placental and pericarp tissue of tomato fruit treated with Apogee at the early stages of fruit growth. Treatment with Apogee also increased fruit pericarp Ca concentration. GA4+7 treatment enhanced the expression of the putative CAX and Ca-ATPase genes, that code for proteins involved in Ca movement into storage organelles. The lowest water-soluble apoplastic Ca concentration and the highest membrane leakage values were observed in the pericarp of GA4+7-treated fruit. These results suggest that GAs consistently reduced fruit Ca uptake and water-soluble apoplastic Ca concentration, leading to leakier plasma membranes and an increase in BER development in fruit tissue of both tomato cultivars.  相似文献

The relative importance of growth rate and calcium concentration in sweet pepper fruits (Capsicum annuum L.) for the induction of blossom-end rot (BER) was investigated in (1) four pollination treatments in one cultivar, (2) four cultivars with the same fruit load and (3) three fruit load treatments in four cultivars. For fruits with the same pollination treatment those eventually developing BER had a higher initial fruit growth rate than those not developing BER. Within the same experiment both the growth rate of the young fruit and BER increased with the number of seeds. The Ca concentration of the pericarp in mature fruits was negatively related to both fruit size and BER incidence. Differences in levels of BER between different pollination experiments could not be explained solely by differences in growth rate of the young fruit, but related to different Ca concentrations in the mature fruits. In the spring, but not in the summer, cultivars more susceptible to BER had a larger final size but lower Ca concentration in the young fruit than the resistant ones. By lowering the fruit load in the summer both the final fruit size and the BER incidence increased, but the Ca concentrations of both proximal and distal pericarp in the young fruit of all cultivars were not consistently affected. Despite a correlation between growth rate and low Ca concentration in the fruit, the incidence of BER may only be predicted from separate effects of fruit growth and of Ca concentration of fruit. The data indicated that at a higher growth rate a higher Ca concentration is required to prevent the induction of BER. The usefulness of the total Ca concentration of the fruit for determining the critical Ca concentration in the induction of BER is discussed.Key words: Capiscum annuum L., sweet pepper, blossom-end rot, calcium, growth rate, pollination, fruit load.   相似文献

Studies of Ca uptake and distribution in relation to environmental variables were used to relate Ca status of tomato fruit to blossom-end rot (BER) incidence. Ca uptake was highly correlated with solar radiation and root temperature. The rate of Ca uptake decreased linearly with increasing salinity. High humidity reduced Ca import by the leaves but increased that by the fruit. While total plant dry weight was reduced more than fruit dry weight by salinity, total Ca uptake and the Ca content of the fruit were decreased similarly. Thus, the concentration of calcium in the fruit was substantially reduced by salinity. The distal half of the fruit contained less Ca than the proximal half. The lowest % Ca was found in the distal placenta and locular tissues, where BER first develops. The incidence of BER was often stimulated more by high salinity achieved with the addition of major nutrients than with NaCl. The cause of BER is usually an interaction between the effects of irradiance and ambient temperature on fruit growth and the effects of environmental stress on calcium uptake and distribution within the whole plant.  相似文献

Tomato plants (Lycopersicon esculentum Mill. var. DRK) were grown hydroponically to determine the effect of an uneven distribution of nutrients in the root zone on blossom end rot (BER) and Ca and K concentrations in the fruits. The plants were grown in rockwool with their root system divided into two portions. Each portion was irrigated with nutrient solutions with either the same or the different electrical conductivity (EC) in the range 0 to 6 dS m–1. Solutions with high EC supplied to both sides of the root system significantly increased the incidence of BER. However, when only water or a solution of low EC was supplied to one portion, BER was reduced by 80%. Fruit yields were significantly higher (P<0.01) for plants that received solutions of the uneven EC treatments (6/0 or 4.5/0 EC treatment). Plants supplied with solutions of uneven EC generally had higher leaf and fruit concentrations of Ca but lower concentrations of K than those supplied with solutions of high EC. There was no difference in Ca concentration at the distal end of young fruits of the uneven EC treatment but it was reduced in the high EC treatments. The concentration of K in the mature fruits of the uneven EC treatments was lower than that of the high EC treatments and higher or similar that of the 3/3 or 2.5/2.5 EC treatments (controls). A clear relationship was found between the incidence of BER and the exudation rate. High rate of xylem exudation was observed in the uneven EC treatments. Reduction of BER in the uneven EC treatments is most likely to be the effect of high exudation rate on Ca status in the young fruits. It was concluded that high EC of solution had positive effects on Ca concentration and incidence of BER provided that nutrient solution with low EC or water is supplied to the one portion of the root system.  相似文献

Calcium (Ca) uptake into fruit and leaves is dependent on xylemic water movement, and hence presumably driven by transpiration and growth. High leaf transpiration is thought to restrict Ca movement to low-transpiring tomato fruit, which may increase fruit susceptibility to the Ca-deficiency disorder, blossom end rot (BER). The objective of this study was to analyse the effect of reduced leaf transpiration in abscisic acid (ABA)-treated plants on fruit and leaf Ca uptake and BER development. Tomato cultivars Ace 55 (Vf) and AB2 were grown in a greenhouse environment under Ca-deficit conditions and plants were treated weekly after pollination with water (control) or 500 mg l(-1) ABA. BER incidence was completely prevented in the ABA-treated plants and reached values of 30-45% in the water-treated controls. ABA-treated plants had higher stem water potential, lower leaf stomatal conductance, and lower whole-plant water loss than water-treated plants. ABA treatment increased total tissue and apoplastic water-soluble Ca concentrations in the fruit, and decreased Ca concentrations in leaves. In ABA-treated plants, fruit had a higher number of Safranin-O-stained xylem vessels at early stages of growth and development. ABA treatment reduced the phloem/xylem ratio of fruit sap uptake. The results indicate that ABA prevents BER development by increasing fruit Ca uptake, possibly by a combination of whole-plant and fruit-specific mechanisms.  相似文献

Fruit size within a tomato (Lycopersicon esculentum Mill.) truss depends on both fruit position in the truss and the time of pollination among fruits. In the natural pollination sequence a difference of 5 days in the pollination of proximal and distal flowers results in significant final size differences between proximal and distal fruits. These final size differences were eliminated when all flowers were pollinated simultaneously. At anthesis proximal ovaries have higher cell numbers than distal ovaries but the cell division activity and cell enlargement in both positions was similar in the first 10 days of fruit growth. Simultaneous pollination resulted in lower cell numbers in proximal but higher cell numbers in distal fruits compared to control fruits.Hormone levels in different sized fruits were measured using radioimmunoassays. Cytokinin concentration during the cell division period indicated a possible role in the regulation of cell division. With other hormones no obvious correlations were found. The results are discussed in relation to factors determining final fruit size in tomato.  相似文献

Apoplastic pH and ionic conditions exert strong influence on cell wall metabolism of many plant tissues; however, the nature of the apoplastic environment of ripening fruit has been the subject of relatively few studies. In this report, a pressure-bomb technique was used to extract apoplastic fluid from tomato fruit ( Lycopersicon esculentum Mill.) pericarp at several developmental stages. pH and the levels of K+, Na+, Ca2+, Mg2+, Cl and P were determined and compared with the values for the bulk pericarp and locule tissues. The pH of the apoplastic fluid from pericarp tissue decreased from 6.7 in immature and mature-green fruits to 4.4 in fully-ripe fruit. During the same period, the K+ concentration increased from 13 to 37 m M . The levels of Na+ and divalent cations did not change, whereas the anions P and Cl increased in ripe fruit. Ca2+ levels remained relatively constant during ripening at 4–5 m M , concentrations that effectively limit pectin solubilization. The electrical conductivity of the apoplastic liquid increased 3-fold during ripening, whereas osmotically active solutes increased 2-fold. Pressure-treated fruit retained the capacity to ripen. The decline in apoplastic pH and increase in ionic strength during tomato fruit ripening may regulate the activity of cell wall hydrolases. The potential role of apoplastic changes in fruit ripening and softening is discussed.  相似文献

It has been reported that PG is a key enzyme related to the tomato fruit ripening. In this study tomato fruits were harvested at the mature-green stage and stored at room temperature. The cell ultrastructure of pericarp tissue was observed at different ripening stages, and the effects of treatments with ethylene and calcium on PG activity and fruit ripening were examined. The object of this study is to elucidate the role of PG in regulation of tomato fruit ripening by ethylene and calcium. PG activity, was undetectable at mature-green stage, but it rose rapidly as fruif ripening. The rise in PG activity was coincided with the dechnmg of fruit firmness during ripening of tomato fruits. The observation of cell ultrastructure showed that the most of grana in chloroplast were lost and the mitochondrial cristae decreased as fruit ripening. Striking changes of cell wall structure was most noted, beginning with dissolution of the middle lamella and eventual disruption of primary cell wall. A similar pattern of changes of cell wall and chloroplast have been observed in pericarp tissue treated with PG extract. In fruits treated with calcium and other divalent metal ions atmature-green stage, the lycopene content and PG activity decreased dramatically. Ethylene application enhanced the formation of lycopene and PG activity. The inhibition of Ca2+ on PG ac ivity was removed by ethylene. Based on the above results, it was demonstrated that PG played a major role in ripening of tomato fruits, and suggested that the regulation of fruit ripening by ethylene and Ca2+ was all mediated by PG. PG induced the hydrolysis of cell wall and released the other hydrolytic enzymes, then effected the ripening processes follow up.  相似文献

The cessation of tomato fruit growth has been associated with the appearance of three 'wall-bound' peroxidase isozymes in the skin of tomato fruit. However, the presence of these isozymes in the ionically eluted 'wall-bound' fraction may be an artefact of either non-specific binding of symplastic peroxidase to the cell wall, or isozymes bound to membranes included in the 'wall-bound' fraction. Therefore, subcellular localization of peroxidase in both immature and mature tomato fruit skins was studied. Immature fruits showed intense peroxidase activity associated with the tonoplast and pro-vacuolar membranes, but little or no activity associated with the cell wall. However, the presence of peroxidase activity within the cell wall of mature green fruits was confirmed. Furthermore, peroxidase activity was also observed associated with the plasma membrane and large vesicles allied to the plasma membrane. While cross-linking in cell wall components was previously assumed to be the mechanism by which peroxidase might control fruit growth, the incorporation of 'lignin-like' phenolics may also play a part. Isoelectric focusing (IEF) of both symplastic and apoplastic peroxidase extracted from immature and mature tomato fruit skin showed that all peroxidase isozymes present were highly anionic. In this current study, histochemical techniques are used to demonstrate a developmental increase in 'lignin-like' phenolics within the sub-cuticular cell walls of the fruit skin. The localization of peroxidase within tomato fruit skin is discussed in relation to its potential role in the regulation of tomato fruit growth.  相似文献

Blossom-end rot (BER) is a physiopathy that affects tomato fruits causing disorganisation, cell breakage and darkening of the tissues. In this study we describe a tomato fruit protein extraction protocol that includes polyvinyl polypyrrolidone, ascorbic acid and protease inhibitors to promote depletion of phenolics and to avoid protein degradation. The temperature-induced phase separation of plant extracts with nonionic detergent Triton X-114 favours the solubilisation of partially-hydrophobic species in the low-detergent upper phase, making them suitable for further analysis using two-dimensional gel electrophoresis. The analysis of two-dimensional images revealed differences in number and expression levels of several proteins from the control and BER-affected tomato fruits. Although the appearance of BER in tomato is primarily attributed to a lack of calcium supply to fruits, very little is known about the molecular and biochemical mechanisms involved. The identification of differential proteins from affected fruits with matrix-assisted laser desorption/ionisation-time of flight and peptide mass fingerprinting analysis revealed the induction of proteins participating in antioxidant processes (ascorbate-glutathione cycle) and the pentose phosphate pathway. We suggest that these two biochemical pathways, acting as reactive oxygen species scavengers in BER-affected fruits, restrain the spread of the blackening to the whole fruit.  相似文献

Summary. Blossom-end rot (BER) of tomato (Lycopersicon esculentum) fruits is considered to be a physiological disorder caused by calcium deficiency. We attempted to clarify the localization of calcium in the pericarp cells and the ultrastructural changes during the development of BER. Calcium precipitates were observed as electron-dense deposits by an antimonate precipitation method. Some calcium precipitates were localized in the cytosol, nucleus, plastids, and vacuoles at an early developmental stage of normal fruits. Calcium precipitates were increased markedly on the plasma membrane during the rapid-fruit-growth stage compared with their level at the early stage. Cell collapse occurred in the water-soaked region at the rapid-fruit-growth stage in BER fruits. There were no visible calcium precipitates on the traces of plasma membrane near the cell wall of the collapsed cells. The amount of calcium precipitates on plasma membranes near collapsed cells was smaller than that in the cells of normal fruits and normal parts of BER fruits, and the amount on cells near collapsed cells was small. The amount of calcium precipitates on the plasma membranes increased as the distance from collapsed cells increased. On the other hand, calcium precipitates were visible normally in the cytosol, organelles, and vacuoles and even traces of them in collapsed cells. The distribution pattern of the calcium precipitates on the plasma membrane was thus considerably different between normal and BER fruits. Two cultivars of tomato with contrasting response to elevated temperature were compared: sensitive-Roma and tolerant-Robin. Experiments were done on fruit explants and on rooted cuttings with small fruits. In both cases45Ca was poorly transported to the fruits. Nevertheless in fruit explants elevated temperature (40°C) increased45Ca import into the fruits in both cultivars. In the compared cuttings, treated or not treated with growth regulators and at various temperatures, the greatest differences were observed in the amount of45Ca transported to the fruits. Sensitive Roma cuttings scantily supplied their fruits with45Ca both under optimal temperature and heat stress. In plants previously treated with NOA+GA3 high temperature increased45Ca transport to the fruits. Robin cuttings inversely responded to heat stress by transporting a much higher portion of45Ca to the fruits, both in control and NOA+GA3 cuttings.The diversity of45Ca distribution during elevated temperature in cuttings, but not in fruit explants of both cultivars seems to be connected with an ability to control calcium supply to fruit or at least to prevent its decrease; this mechanism is perhaps located outside the cluster.  相似文献

By using immunolocalization and differential extraction methods we show that only apoplastic invertase, but not vacuolar invertase, was present in the mature, sucrose-accumulating L. hirsutum pericarp. In contrast, in the hexose-accumulating L. esculentum fruit, both the apoplastic and vacuolar invertase activities and protein content increase in the mature fruit. Quantitative expression studies of the soluble invertase gene (TIV1) and the apoplastic invertase genes (LINs) showed that only TIV1 gene expression could account for the species and developmental differences of both soluble and insoluble enzyme activity of the pericarp. The expression of the LIN genes encoding for apoplastic tomato invertases was unrelated to the differences in bound enzyme activity and could not account for the rise in bound invertase activity in the mature L. esculentum fruit. Evidence is presented that the bound invertase activity of tomato fruit is also the TIV1 gene product. The presence of apoplastic invertase in the mature sucrose-accumulating L. hirsutum fruit suggests a hydrolysis-resynthesis mechanism of sucrose uptake. In order to test this hypothesis, we studied short- and long-term uptakes of asymmetrically labelled 3H-fructosyl-sucrose accompanied by compartmental analysis of the sugars in attached whole fruits of L. hirsutum and L. esculentum. The results indicate that hydrolysis-resynthesis is slow in the sucrose-accumulating fruit but is not an integral part of an uptake and compartmentation mechanism.  相似文献

The effect of sap sucrose concentration on fruit expansion is poorly understood. We investigated the effects of sucrose concentration on growth of tomato fruits, detached from the plant after the cell multiplication phase. A semi-open device was built to grow detached fruits, where the fruit pedicel was immersed in a nutrient solution which was automatically replaced each day. About 22 d after anthesis, tomato fruits were harvested, disinfected in alcohol, and placed in the device with a sterile nutrient solution containing 5% sucrose. After about 1 wk, the sucrose concentration was either increased to 15% or maintained at 5%. The fruit expansion rate decreased when the sucrose concentration was increased, and after about 1 wk, these fruits weighed less than those grown at 5% sucrose. Although all the fruits in the device grew much less than those maintained on the plant, a high sap sucrose concentration applied to the excised fruits after the cell multiplication phase inhibited the expansion of tomato fruits. The device and method developed here will be useful for future studies of fruit growth.  相似文献

The softening of fleshy fruits, such as tomato (Solanum lycopersicum), during ripening is generally reported to result principally from disassembly of the primary cell wall and middle lamella. However, unsuccessful attempts to prolong fruit firmness by suppressing the expression of a range of wall-modifying proteins in transgenic tomato fruits do not support such a simple model. 'Delayed Fruit Deterioration' (DFD) is a previously unreported tomato cultivar that provides a unique opportunity to assess the contribution of wall metabolism to fruit firmness, since DFD fruits exhibit minimal softening but undergo otherwise normal ripening, unlike all known nonsoftening tomato mutants reported to date. Wall disassembly, reduced intercellular adhesion, and the expression of genes associated with wall degradation were similar in DFD fruit and those of the normally softening 'Ailsa Craig'. However, ripening DFD fruit showed minimal transpirational water loss and substantially elevated cellular turgor. This allowed an evaluation of the relative contribution and timing of wall disassembly and water loss to fruit softening, which suggested that both processes have a critical influence. Biochemical and biomechanical analyses identified several unusual features of DFD cuticles and the data indicate that, as with wall metabolism, changes in cuticle composition and architecture are an integral and regulated part of the ripening program. A model is proposed in which the cuticle affects the softening of intact tomato fruit both directly, by providing a physical support, and indirectly, by regulating water status.  相似文献

The changes taking place during fruit development in the concentration of the 3 polyamine fractions, i.e. free, perchloric acid-soluble conjugates and perchloric acid-insoluble bound polyamines, were analyzed in tomato fruits ( Lycopersicon esculentum Mill, cv. F121) induced to set by either pollination or auxin application. Before the onset of cell division, total polyamines were 50% higher in auxin-treated fruits than in pollinated ones, most of the polyamines being found as perchloric acid-soluble conjugates in both fruit set treatments. At the onset the level of polyamines in both fruit types was 100 times higher than during cell expansion or ripening. The highest polyamine found during cell division was perchloric acid-soluble conjugated spermidine in both fruit set treatments. After cell division, polyamine concentration was similar in both fruit set treatments. The concentration of polyamines in the jelly was similar in pollinated and auxin-induced fruits during cell expansion but not during ripening. At the onset of ripening there was an increase of one order of magnitude in the concentration of perchloric acid-insoluble bound putrescine in the jelly of pollinated fruits. Polyamines were more than 5-fold higher in unpollinated ovaries than in fruits induced to set by either pollination or auxins. It is suggested that pollinated and parthenocarpic fruits differ in their polyamine metabolism during the initial stages of development, but not after cell division. It is also suggested that polyamines undergo rapid turnover during cell division. Perchloric acid-insoluble bound putrescine might play a role in seed formation in tomatoes.  相似文献

Calcium is an essential plant nutrient. It is required for various structural roles in the cell wall and membranes, it is a counter-cation for inorganic and organic anions in the vacuole, and the cytosolic Ca2+ concentration ([Ca2+]cyt) is an obligate intracellular messenger coordinating responses to numerous developmental cues and environmental challenges. This article provides an overview of the nutritional requirements of different plants for Ca, and how this impacts on natural flora and the Ca content of crops. It also reviews recent work on (a) the mechanisms of Ca2+ transport across cellular membranes, (b) understanding the origins and specificity of [Ca2+]cyt signals and (c) characterizing the cellular [Ca2+]cyt-sensors (such as calmodulin, calcineurin B-like proteins and calcium-dependent protein kinases) that allow plant cells to respond appropriately to [Ca2+]cyt signals.  相似文献
