In this paper data are presented on cows receiving no food but having access to all of the water which they wished. The yield and composition of the milk were determined at various times during the periods of starvation. The composition of the milk showed changes which were progressive in the sense that they followed a definite course. They were characterized by a marked lowering in the amount of milk produced, by an increase in the total solids (chiefly an increase in the percentage of fat and ash, with a slight increase in proteins), and by a pronounced decrease in the lactose. The decrease in lactose corresponded with a decrease in the dextrose content of the blood, thereby supporting the conclusion that the lactose of milk has as its precursor dextrose of the blood. All the changes in milk composition during starvation can be directly related to the simultaneous changes in the blood. The following companion paper on insulin and phloridzin as they affect milk secretion further develops these hypotheses.  相似文献

The results herein presented furnish exact critical evidence for one more stage in milk secretion. Cows producing up to 30 pounds of milk at one milking are shown to have the lactose equivalent of all this milk in the udder when milking commences. The average excess of lactose found in the udder after subtracting the amount necessary for the contained milk is equal to 2.1 pounds. This represents the milk retained in the udder when the cow is believed to be dry. These conclusions are further supported by the fact that no sugar is found in the udder in the quiescent state. The study of the total composition of the udder as fat, ash, nitrogen, and lactose, and of the contained milk shows that there is a large excess of fat, ash, and nitrogen in proportion to that necessary for milk formation. The excess of udder lactose over the milk lactose is much less. The lactose would therefore appear to be formed from some element in the blood, probably dextrose, only as needed for the formation of milk. The composition of the dry udder is quite different in certain respects from that of the actively secreting gland. It builds up a fat reserve of a quite different Reichert-Meissl number from that of butter-fat. It has no sugar, its ash content is reduced, and the nitrogen content is like that of the secreting gland.  相似文献

Rates of secretion of milk constituents (fat, protein, lactose, Na+ and K+) in the lactating goat were measured under normal circumstances and after injections of ouabain. In all experiments a close association was noted in the secretion rates for protein, lactose and K+. Under the influence of ouabain, the concentration of Na+ in the milk tended to rise and that of K+ to fall. The rate of milk fat secretion varied independently from the rates for the other constituents. It is reasonably assumed that the principal mechanism of milk protein secretion is by emptying of Golgi vesicles through the plasma membrane. The close correlation in rates for protein, lactose and K+ supports the contention that all three are assembled in Golgi vesicles and secreted by the same mechanism.  相似文献

Two lactating cows were given tritiated hay containing organically bound tritium (OBT) only for about 4 weeks. Tritium activity was determined in milk fat, casein, lactose, milk water, and whole milk. In one cow, milk was sampled for approximately 450 days, covering two lactation periods. At steady state, specific tritium activities in casein, lactose, and milk water were 58, 10, and 11%, respectively, of those in milk fat. Some OBT was converted into THO during catabolism and entered the body water pool. This 3H source accounted for nearly 40% of tritium in lactose, but in casein and milk fat about 97% of tritium was derived from ingested OBT. Comparison of the specific activity of milk constituents with the specific activity of ingested hay showed the following values: 0.84 for milk fat, 0.49 for casein, 0.05 for lactose, 0.10 for milk water. About one-half of the tritium transferred to milk was found in organic milk constituents and the other half in milk water. Decrease of tritium activity with time could be represented by three components with different half-lives for the organic milk constituents. Those for milk fat and casein were quite similar, with a slow component of nearly 3 months.  相似文献

1. Progressive changes in the composition of milk from rats has been studied from day 0 to 20 of lactation and for 3 days following separation of the dams and pups at day 20 post partum. 2. The changes in concentration of Na, K and lactose suggested that secretion both prepartum and following weaning occurred by a paracellular mechanism whereas a transcellular pathway existed during established lactation. 3. The concentration of total protein and casein increased gradually throughout lactation. In contrast, the concentration of serum albumin increased and transferrin decreased markedly during early lactation. The fat content of milk declined 3-fold within 5 days of birth but the concentration of Ca, Mg and inorganic P increased. The concentration of each of these milk constituents remained constant during established lactation. 4. Following weaning the pronounced decline in lactose, K and inorganic P was negatively correlated with an increase in all other milk constituents except fat. 5. Rats fed a low energy diet produced milk with a lower fat content but with an unaltered concentration of protein and carbohydrate. The growth rate of these litters was similar for the first 5 days of lactation when compared to litters from dams fed a high energy diet. The growth rate of litters thereafter and following weaning was greater for rats fed a high energy diet.  相似文献

1. The ratios of stable carbon isotopes 13C/12C in milk constituents of Holstein dairy cattle were investigated by mass spectrometry. 2. Under physiological feeding conditions the natural abundance of 13C in lactose was greater than in milk fat. 3. Reduction of energy intake diminished the abundance of 13C in lactose resulting in values similar to those of fat. 4. It is suggested that by comparing the 13C/12C ratios in milk fat and lactose the metabolic energy state of cows may be rated.  相似文献

The ability of 8 pregnant ewes to maintain glucose homeostasis following an intravenous glucose load (glucose tolerance test) and during starvation in late pregnancy was studied. Following an intravenous load 2 out of 7 ewes tested showed an impaired insulin secretion during the first 10–15 min. An increase in plasma insulin concentration was found 50–60 min after the injection. The results of the effect of a starvation on a number of blood constituents are demonstrated. Two of the twin–pregnant ewes developed symptoms of pregnancy disease following starvation, these were the same ewes that showed an impaired insulin secretion after the glucose load. However, no difference was demonstrated in the clinical chemical parameters whether the experimental ewes developed pregnancy disease or not.  相似文献

An analysis of milk constituents during various stages of lactation in the southern elephant seal Mirounga leonina was carried out. Forty-six milk samples were taken from 30 females throughout lactation during 1985, 1987, 1990 and 1991 on Stranger Point, King George Island, South Shetland Islands. Total nitrogen (TN), non-protein nitrogen (NPN), sugar, fat, ash and water were measured, and from some of these data true protein and energy content were calculated. The results showed a high degree of variation in water and fat concentrations among samples at different stages of lactation. During the first 20 days the fat content of milk increased from about 12 to approximately 52%, while water content fell from 70 to 33%. The composition of milk changes rapidly during the first days post-partum. Protein, minerals and sugar appear to remain stable after the fourth day of lactation. Milk samples contain significant levels of sugars; thin layer chromatography indicates the presence of lactose and glucose together with other unidentified components. There is evidence of a striking change in composition of the milk in the later part of lactation; the progressive increase in the fat:water ratio is abruptly reversed just prior to weaning.  相似文献

Lactation plays a vital role in reproductive success, evolving with ecological adaptations of mammalian life histories. Knowledge of the lactation process in odontocetes is scarce and limited to a handful of species. We investigated the changes in milk composition across different lactation stages, including prepartum mammary secretion and early, mid‐ and late lactation of narrow‐ridged finless porpoises. Prepartum mammary secretion was greenish and characterized by high protein, low sugar, and negligible fat contents. In contrast, milk fat became predominant over protein and sugar contents throughout all lactation stages. At the early lactation stage, the contents of all milk constituents except water varied greatly. At the mid‐late lactation stage, the gross milk composition was relatively stable and was composed of, on average, 67.34% ± 4.13% water, 9.96% ± 0.75% protein, 21.40% ± 3.24% fat, and 1.72% ± 0.73% sugar. Our findings provide new insight into milk composition at different lactation stages in the narrow‐ridged finless porpoise.  相似文献

Early lactation parameters are difficult to estimate from commercial dairy records due to the small number of records available before the peak of production. A biological model of lactation was used with weekly milk records from a single Holstein herd to estimate these early lactation parameters and the secretion rate of milk from the average cell throughout lactation. A genetic analysis of the lactation curve parameters, calculated curve characteristics and secretion rate traits was undertaken. Early lactation traits were found to have little genetic variation and effectively zero heritability. Secretion rate traits for milk, protein, lactose and water were all moderately heritable and highly genetically correlated (>0.87) but fat secretion rate had lower genetic correlations with the other secretion rates. A similar pattern of correlations was seen between total lactation yield traits for fat, protein, lactose and water. The genetic correlations between the lactation curve traits and the secretion rate traits were calculated. Total milk yield, peak yield and maximum secretion potential were all highly correlated with milk, lactose and water secretion rates but less so with fat and protein secretion rates. In particular, fat secretion rate had a moderate to low genetic correlation with these lactation curve traits. Persistency of lactation was highly correlated with fat and protein secretion rates, more persistent lactations being associated with lower rates of secretion of these milk components. Similar levels of heritability were found, where trait genetic parameters were directly equivalent to those derived from the same dataset by random regression methods. However, by using a biological model of lactation to analyse lactation traits new insights into the biology of lactation are possible and ways to select cows on a range of lactation traits may be achieved.  相似文献

The observation that concanavalian A can inhibit milk secretion was evaluated in an in vitro system employing minced mammary gland or isolated alveoli from lactating rats. Release of milk constituents (casein, lactose and fat globules) into the medium in the presence and absence of concanavalin A was measured during 1 or 2 h incubations. The effect of concanavalin A on glucose uptake and CO2 production of the minced tissued was also studied. Concanavalin A suppressed release of milk components at a concentration as low as 80 μg/ml of medium. Respiration of minced mammary tissue in the presence of concanavalin A (100 μg/ml of medium) was essentially the same as that of the control. The data are evidence that concanavalin A acts directly on the mammary cell in suppressing milk secretion and that the effect is not due to cytotoxicity.  相似文献

Consumer demand for the major components of milk has changed considerably over the past 10 years. The trend in recent years is for reduced consumption of milk fat as a component of fluid milk and in dairy products such as butter, cheese and yoghurt. Matching production of milk components to demand improves biological efficiency and ultimately enhances the overall profitability of the dairy industry. Genetic selection of dairy cattle allows the dairy industry to respond to consumer demand in the long-term. However, it is not a satisfactory means of manipulating milk composition in the short- or medium-term. The potential of nutrition as a tool to alter the composition of milk — especially milk fat percentage — has not been fully exploited. Changes in the composition of the diet, in particular the forage to concentrate ratio, can result in changes in milk fat percentage in the range of 2.0 to 4.0%. The largest reduction in milk fat is achieved in mid- to late-lactation and this is also a time when cows are less susceptible to metabolic disorders. Although experiments designed to evaluate the long-term effects of feeding extreme fat-depressing diets have not been conducted there are sufficient examples of apparently healthy herds which produce milk with a fat content in the range of 2.5 to 3.0% to allow cautious optimism that low milk fat is not necessarily incompatible with ‘healthy’ cows.  相似文献

An experiment was carried out on 10 advance pregnant Murrah buffaloes to determine the role of hormones in milk secretion around parturition. Experimental animals were administered with a single injection of bromocryptine, @ 100 μg/kg BW, for 5 days before expected calving, whereas control group buffaloes were injected with placebo injections. Blood samples collected before parturition (-5,-4,-3,-2,-1 days), on day of parturition (day-0) and on day 1, 2, 3, 4, 5, 10 and 15 post partum were analyzed for growth hormone (GH), insulin like growth factor-I (IGF-I) and prolactin (PRL) by radioimmunassay methods. Milk samples were collected daily for 5 days and on day 10 and 15 after parturition. Milk fat, protein, lactose, citric acid, non-esterified fatty acids (NEFAs) and somatic cell counts (SCCs) were determined in milk samples. Bromocryptine treatment significantly (P < 0.01) decreased pre partum PRL and increased GH levels (P < 0.01) on day of parturition in experimental buffaloes without influencing plasma IGF-I level. Milk yield was significantly lower (P < 0.01) in experimental than in control group. Further, effect of bromocryptine on milk yield was only for a week. Milk yield increased (P < 0.01) gradually and was similar to control group on day 15 post partum. Bromocryptine treatment significantly increased milk SCC (P < 0.01) and protein content (P < 0.01) but there was no effect of treatment on fat, lactose, citric acid, glucose, milk and plasma NEFA concentration. It was concluded that prepartum suppression of PRL by bromocryptine impairs milk secretion temporarily in ensuing lactation. The significant rise in GH level before parturition and on day of parturition suggests a role of it in milk secretion of buffaloes.  相似文献

Alterations in the milk constituents throughout the lactation cycle have been studied in 20 Karan-Fries cross-bred cows (Holstein Fresian x Tharparkar) divided into two groups, viz. elite (n = 10) and non-elite (n = 10). About 100 ml of composite milk samples (from all the quarters) were collected through hand milking in sterile tubes from the day of calving at 15-day interval till 300 days of lactation cycle. Different milk constituents (viz. fat, protein, lactose, Solid not fat) were estimated by automatic milk analyzer. The daily milk yield varied significantly (p < 0.01) between group and different days of lactation cycle. Milk fat percentage of milk did not differ significantly between groups and different days of lactation cycle. Milk protein and lactose percentage did not differ significantly between groups but differed significantly (p < 0.001) between days. Solid not fat (SNF) percentage of milk was significantly (p < 0.01) higher in elite cows compared to non-elite cows and also varied significantly (p < 0.05) during different days of lactation cycle. In both the group of cows, milk protein, fat, and SNF percentage was highest and lactose percentage was lowest up to 1st week of lactation cycle and rapidly declined thereafter due to the transition of the colostrum into milk. After 2nd week of lactation, all the milk constituents under study were almost remained unaltered till the end of the cycle. The above stated investigation not only substantiates the already existing information of lactation stage-specific alteration in milk constituents but also depict the exact point of transition of these constituents during the lactation cycle so the nutritional and managemental interventions could be carried out in proper time.  相似文献

Milk reduces post-meal glycemia when consumed either before or within an ad libitum meal. The objective of this study was to compare the effect of each of the macronutrient components and their combination with whole milk on postprandial glycemia, glucoregulatory and gastrointestinal hormones and gastric emptying in healthy young men. In a randomized, crossover study, 12 males consumed beverages (500ml) of whole milk (3.25% M.F.) (control), a simulated milk beverage based on milk macronutrients, complete milk protein (16g), lactose (24g) or milk fat (16g). Whole and simulated milk was similar in lowering postprandial glycemia and slowing gastric emptying while increasing insulin, C-peptide, peptide tyrosine tyrosine (PYY) and cholecystokinin (CCK), but simulated milk resulted in higher (41%) glucagon-like peptide-1 (GLP-1) and lower (43%) ghrelin areas under the curve (AUC) than whole milk (P=.01 and P=.04, respectively). Whole and simulated milk lowered glucose (P=.0005) more than predicted by the sum of AUCs for their components. Adjusted for energy content, milks produced lower glucose and hormone responses than predicted from the sum of their components. The effect of protein/kcal on the AUCs was higher than fat/kcal for insulin, C-peptide, insulin secretion rate, GLP-1, CCK and paracetamol (P<.0001), but similar to lactose except for CCK and paracetamol, which were lower. The response in PYY and ghrelin was similar per unit of energy for each macronutrient. In conclusion, milk lowers postprandial glycemia by both insulin and insulin-independent mechanisms arising from interactions among its macronutrient components and energy content.  相似文献

1. The incorporation of radiolabelled plasma glucose into mammary lactose was used to measure the rate of lactose synthesis in lightly anaesthetized lactating rats. 2. Lactose synthesis showed a diurnal variation with a minimum at 18:00h 3. Food withdrawal for 6h did not affect lactose synthesis in the early morning but greatly decreased it in the afternoon or evening. 4. Plasma glucose, milk sugars and total galactosyltransferase activity (EC 2.4.1.22) did not show the above changes. 5. Measurements of plasma insulin, which varies diurnally, and experiments with injected insulin suggested that variations of insulin within the physiological range do not account for the changes in lactose synthesis described.  相似文献

In this study we have analyzed the effects of variations in the concentrations of oxygen and of blood sugar on light sensitivity; i.e. dark adaptation. The experiments were carried out in an air-conditioned light-proof chamber where the concentrations of oxygen could be changed by dilution with nitrogen or by inhaling oxygen from a cylinder. The blood sugar was lowered by the injection of insulin and raised by the ingestion of glucose. The dark adaptation curves were plotted from data secured with an apparatus built according to specifications outlined by Hecht and Shlaer. During each experiment, observations were first made in normal air with the subject under basal conditions followed by one, and in most instances two, periods under the desired experimental conditions involving either anoxia or hyper- or hypoglycemia or variations in both the oxygen tension and blood sugar at the same time. 1. Dark adaptation curves were plotted (threshold against time) in normal air and compared with those obtained while inhaling lowered concentrations of oxygen. A decrease in sensitivity was observed with lowered oxygen tensions. Both the rod and cone portions of the curves were influenced in a similar way. These effects were counteracted by inhaling oxygen, the final rod thresholds returning to about the level of the normal base line in air or even below it within 2 to 3 minutes. The impairment was greatest for those with a poorer tolerance for low O2. Both the inter- and intra-individual variability in thresholds increased significantly at the highest altitude. 2. In a second series of tests control curves were obtained in normal air. Then while each subject remained dark adapted, the concentrations of oxygen were gradually decreased. The regeneration of visual purple was apparently complete during the 40 minutes of dark adaptation, yet in each case the thresholds continued to rise in direct proportion to the degree of anoxia. The inhalation of oxygen from a cylinder quickly counteracted the effects for the thresholds returned to the original control level within 2 to 3 minutes. 3. In experiments where the blood sugar was raised by the ingestion of glucose in normal air, no significant changes in the thresholds were observed except when the blood sugar was rapidly falling toward the end of the glucose tolerance tests. However, when glucose was ingested at the end of an experiment in low oxygen, while the subject remained dark adapted, the effects of the anoxia were largely counteracted within 6 to 8 minutes. 4. The influence of low blood sugar on light sensitivity was then studied by injecting insulin. The thresholds were raised as soon as the effects of the insulin produced a fall in the blood sugar. When the subjects inhaled oxygen the thresholds were lowered. Then when the oxygen was withdrawn so that the subject was breathing normal air, the thresholds rose again within 1 to 2 minutes. Finally, if the blood sugar was raised by ingesting glucose, the average threshold fell to the original control level or even below it. 5. The combined effects of low oxygen and low blood sugar on light sensitivity were studied in one subject (W. F.). These effects appeared to be greater than when a similar degree of anoxia or hypoglycemia was brought about separately. 6. In a series of experiments on ten subjects the dark adaptation curves were obtained both in the basal state and after a normal breakfast. In nine of the ten subjects, the food increased the sensitivity of the subjects to light. 7. The experiments reported above lend support to the hypothesis that both anoxia and hypoglycemia produce their effects on light sensitivity in essentially the same way; namely, by slowing the oxidative processes. Consequently the effects of anoxia may be ameliorated by giving glucose and the effects of hypoglycemia by inhaling oxygen. In our opinion, the changes may be attributed directly to the effects on the nervous tissue of the visual mechanism and the brain rather than on the photochemical processes of the retina.  相似文献

Milk yield and composition of major milk constituents were measured in captive, nursing reindeer. Registration of milk production was performed during two successive lactations (2001 and 2002). The milk yield was significantly affected by week of lactation (P<0.001) and by individual (P<0.001). The lactation curve had an asymmetrical peak 3 weeks postpartum and the milk yield at peak lactation was 983 g/day (range 595-1239). The length of lactation varied from 24 to 26 weeks and average total milk production was 99.5 kg. From peak lactation the milk production decreased linearly (P<0.001) until milk production was terminated. Mean values for content of major milk constituents were 15.5% fat, 9.9% protein and 2.5% lactose. The content of fat and protein increased markedly with the lactation stage (P<0.001), while lactose showed a slight decrease (P<0.001). The milk composition was significantly affected by stage of lactation (P<0.001). There was a marginally significant decrease in protein:fat ratio (P=0.06) as protein was substituted by fat with stage of lactation. The caloric value of the milk averaged 8.7 kJ/g and increased significantly with the stage of lactation (P<0.001). The overall increase in milk gross energy content during lactation was 67.6%. The energy output averaged 7996 kJ/day at peak lactation and decreased significantly during the course of lactation (P=0.002).  相似文献

Lactose synthesis and fatty acid synthesis in intact lactating-rat mammary gland were measured simultaneously by incorporation of [U-14C]glucose and of both [U-14C]glucose and 3H2O respectively. Both processes were almost abolished by overnight starvation. Self-re-feeding caused recovery of lipogenesis to 100% of normal by 2 h and to 170% by 5 h. Lactose synthesis recovered to 80% of normal by 5 h. Food intubated to starved rats caused partial recovery in 3 h, standard diet favouring lactose synthesis and sugars favouring lipogenesis. Casein and starch were ineffective. Olive oil intubated to fed rats suppressed lipogenesis greatly and lactose synthesis slightly. Paraffin oil or water partly mimicked these effects. Adrenaline (subcutaneous) decreased lipogenesis from glucose, whereas insulin (subcutaneous) caused hypoglycaemia associated with loss of lactose synthesis but unchanged fatty acid synthesis. Streptozotocin and 2-bromo-alpha-ergocryptine (CB-154) impaired lipogenesis but not lactose synthesis. The results are interpreted in terms of competition for intracellular glucose by biosynthetic pathways for lactose and fat, and the possible implications for variations in milk composition are discussed.  相似文献
