With the goal of developing a defined medium for the production of desiccation-tolerant blastospores of the bioinsecticidal fungus Paecilomyces fumosoroseus, we evaluated the impact of various media components such as amino acids, carbohydrates, trace metals and vitamins on hyphal growth and sporulation of P. fumosoroseus cultures and on the freeze-drying tolerance of blastospores produced under these conditions. A comparison of 13 amino acids as sole nitrogen sources showed that glutamate, aspartate, glycine and arginine supported biomass accumulations (12–16 mg ml−1) and blastospore yields (6–11 × 108 blastospores ml−1) comparable to our standard production medium which contains casamino acids as the nitrogen source. Using glutamate as the sole nitrogen source, tests with various carbohydrates showed that P. fumosoroseus grew best on glucose (18.8 mg biomass ml−1) but produced similar blastospore concentrations (7.3–11.0 × 108) when grown with glucose, glycerol, fructose or sucrose. P. fumosoroseus cultures grown in media with sodium citrate or galactose as the sole carbohydrate produced lower blastospore concentrations but more-desiccation-tolerant spores. Zinc was the only trace metal tested that was required for optimal growth and sporulation. In a defined medium with glutamate as the nitrogen source, vitamins were unnecessary for P. fumosoroseus growth or sporulation. When blastospores were freeze-dried in the absence of a suspension medium, residual glucose (>2.5% w/v) was required for enhanced spore survival. Thus, a defined medium containing basal salts, glucose, glutamate and zinc can be used to produce optimal concentrations of desiccation-tolerant blastospores of P. fumosoroseus. Dogs were trained to detect Eastern subterranean termites, Reticulitermes flavipes (Kollar), using the United States Customs method of scent detection dog training modified with a food reward. Dogs were tested with various numbers of Eastern subterranean termites placed in vented PVC containers. Trained dogs were 95.93% accurate in finding > or = 40 Eastern subterranean termite workers (positive indications) and incorrectly indicated the presence of termites in 2.69% of the containers without termites. Multiple species of termites [dark southern subterranean, R. virginicus (Banks); Formosan subterranean, Coptotermes formosanus Shiraki; powderpost, Cryptotermes cavifrons Banks; and southeastern drywood termites, Incisitermes snyderi (Light)], were similarly evaluated. Dogs trained to locate Eastern subterranean termites were also 100% accurate in finding dark southern subterranean termites, 98.89% accurate in finding Formosan subterranean termites, 97.33% accurate in finding powderpost termites, and 88.89% accurate in finding southeastern drywood termites. Dogs were able to discriminate live termites from non-termite material. Trained dogs' false response rate was 25.33% to Eastern subterranean termite-damaged wood, 6.67% to American cockroaches, Periplaneta americana (L.), and 2.67% to Florida carpenter ants, Camponotus floridanus Buckley.  相似文献

The production of fungal spores using on-site, non-sterile, portable fermentation equipment is technically constrained. Very little information is available on the production requirements, such as medium concentration, inoculum stabilization, required fermentation times, and maintenance of axenic growth. In this study, we developed a two-part, liquid concentrate of the production medium that remains stable and soluble at room temperature. We also examined inoculum stability and showed that freeze- or air-dried blastospore preparations were stable for 7 days after rehydration when stored at 4 °C. The use of a low-pH (pH 4), relatively rich complex medium provided a growth environment deleterious to bacterial growth yet conducive to rapid sporulation by Paecilomyces fumosoroseus. High concentrations of blastospores (7.9×108/ml) of P. fumosoroseus were produced in a 40-h fermentation with very low levels of bacterial contamination when the fermentor was charged with a blastospore production medium with a starting pH of 4 and inoculated with blastospore concentrations greater than 1×106 spores/ml. These studies demonstrate that the use of disinfected, portable fermentation equipment has potential for on-site production of high concentrations of blastospores of the bioinsecticidal fungus P. fumosoroseus.  相似文献

Although preference and utilization of clay have been studied in many higher termites, little attention has been paid to lower termites, especially subterranean termites. The Formosan subterranean termite, Coptotermes formosanus Shiraki, can modify its habitat by using clay to fill tree cavities. Here, the biological significance of clay on C. formosanus was investigated. Choice tests showed that significantly more termites aggre- gated in chambers where clay blocks were provided, regardless of colony group, observation period, or nutritional condition (fed or starved). No-choice tests showed that clay had no observable effect on survivorship, live or dry biomass, water content, and tunneling activity after 33-35 d. However, clay appeared to significantly decrease filter paper consumption (dry weight loss). Active particle (sand, paper, and clay) transport behavior was observed in both choice and no-choice tests. When present, clay was preferentially spread on the substrate, attached to the smooth surfaces of the containers, and used to line sand tunnels. Mechanisms and potential application of clay attraction are discussed.  相似文献

The efficacy and nonrepellency of indoxacarb (150 SC, 150 g [AI]/liter) and fipronil (Termidor SC, 9.1% [Al]) against field-collected eastern subterranean termite, Reticulitermes flavipes (Kollar), and the Formosan subterranean termite, Coptotermes formosanus Shiraki, were evaluated for mortality and penetration into treated soil in laboratory glass tube bioassays. Both insecticides were tested at five concentrations (0, 1, 10, 50, and 100 ppm) and two thicknesses (20 and 50 mm) of treated soil. Indoxacarb caused significantly greater mortality than controls at all treatment thicknesses of > or = 10 ppm, but not at 1 ppm. Concentration and treatment thickness of indoxacarb significantly affected termite mortality. Eastern subterranean termites were significantly more susceptible to indoxacarb than Formosan subterranean termites, but there were no intercolony differences in either species. Termites completely penetrated through all treatment thickness of indoxacarb-treated soil at all concentrations, except one of the six Formosan subterranean termite replicates of 50 mm at 50 ppm, when all termites were killed before tunneling through the treated soil. Fipronil resulted in significantly faster and greater termite mortality than indoxacarb at corresponding concentrations. Concentration and treatment thickness of fipronil also significantly affected termite mortality. There was no intercolony difference in susceptibility to either insecticide in either termite. Both termite species completely penetrated 20-mm treatments of all tested fipronil concentrations, as well as 50-mm soil treated with fipronil at < or = 10 ppm. At 50 and 100 ppm fipronil, termites tunneled only a mean of 87 +/- 0.21 and 47 +/- 0.18% deep into 50-mm treated soil, respectively, before death. Both insecticides demonstrated a delayed mode of activity and nonrepellency against the two termite species.  相似文献

Formulation matrices can play an important role in improving the storage survival and biocontrol efficacy of microorganisms used for the control of pest insects. In this study, liquid culture-produced blastospores of the entomopathogenic fungus Paecilomyces fumosoroseus were formulated with different inert and organic materials prior to air-drying. Paecilomyces fumosoroseus blastospores were produced in two different liquid media, a basal salts medium supplemented with Casamino acids and glucose (LM1) and a medium containing peptone of collagen and glucose (LM2). Blastospores produced in the two test media were formulated with various supports. The formulation supports were cornstarch, rice flour, talc powders, Mexican lime, calcined kaolin clay, and diatomaceous earth. Several of the supports were tested at different concentrations. The initial and long-term (after storage at 4 and 28 °C) survival of the formulated, air-dried blastospores were evaluated. Initial blastospore viabilities were affected by the formulation material and by the blastospore production medium. Medium composition, drying support and storage temperature had an impact on the long-term survival of the blastospores. Under the conditions of the study, LM1 produced higher concentrations of blastospores that not only survived drying better than blastospores produced in LM2 but also maintained viability longer during storage in the formulation supports tested. The nature of the drying supports was shown to have a significant impact on the storage stability of all blastospores, particularly those produced in LM1. Under the production, drying and storage conditions used in the study, calcined kaolin clay formulations stored at 4 °C had the best storage stability. In all formulations tested, spore survival over time was reduced for blastospore formulations stored at 28 °C rather than 4 °C.  相似文献

The effect of separate and combined activity of Paecilomyces fumosoroseus Wize (Brown and Smith) Trinidadian strain T11 and the parasitoid, Encarsia formosa Gahan, was assessed on populations of the greenhouse whitefly, Trialeurodes vaporariorum (Westwood), infesting Phaseolus vulgaris L. (French bean) and Pelargonium x domesticum (regal geranium) plants in replicate experiments. When infested bean and geranium plants were exposed to E. formosa for 2 days, and 4 days later sprayed with P. fumosoroseus blastospores, whitefly percent mortality was 99.5% and 75.5%, 94.6% and 59.4% for experiments 1 and 2, respectively. Treatment of infested bean plants with either E. formosa or P. fumosoroseus resulted in 87.8% and 78.7%, 73.1% and 97.0% whitefly mortality for experiments 1 and 2, respectively, while similar treatment of infested geranium plants resulted in 9.2% and 52.8%, 34.3% and 64.5% whitefly mortality for experiments 1 and 2, respectively. Our results support the use of E. formosa and P. fumosoroseus in combination in Experiment 1 for the treatment of whitefly infested P. vulgaris plants since a significant difference in mortality is observed than when either E. formosa or P. fumosoroseus is applied alone. However, in experiment 2, the combination treatment on P. vulgaris was no more effective than spraying P. fumosoroseus alone. On P. x domesticum plants, only P. fumosoroseus alone is needed for efficient control of the whitefly compared to the combination treatment. The relative timing of parasitoid oviposition and fungal infection are critical in determining the outcome of the interaction and are plant host dependent.  相似文献

Spore yields were measured for various fungal entomopathogens grown in six nutritionally different liquid media with low and high carbon concentrations (8 and 36 g l–1, respectively) at carbon-to-nitrogen (C:N) ratios of 10:1, 30:1 and 50:1. Six fungi were tested: two Beauveria bassiana strains, three Paecilomyces fumosoroseus strains and one Metarhizium anisopliae strain. Spore yields were examined after 2, 4 or 7 days growth. In general, highest spore yields were obtained in media containing 36 g/l and a C:N ratio of 10:1. After 4 days growth, highest spore yields were measured in the three Paecilomyces isolates (6.9–9.7 × 108 spores ml–1). Spore production by the B. bassiana isolates was variable with one isolate producing high spore yields (12.2 × 108 spores ml–1) after 7 days growth. The M. anisopliae isolate produced low spore concentrations under all conditions tested. Using a commercial production protocol, a comparison of spore yields for the coffee berry borer P. fumosoroseus and a commercial B. bassiana isolate showed that highest spore concentrations (7.2 × 108 spores ml–1) were obtained with the P. fumosoroseus isolate 2-days post-inoculation. The ability of the P. fumosoroseus strain isolated from the coffee berry borer to rapidly produce high concentrations of spores prompted further testing to determine the desiccation tolerance of these spores. Desiccation studies showed that ca. 80% of the liquid culture produced P. fumosoroseus spores survived the air-drying process. The virulence of freshly produced, air-dried and freeze-dried coffee berry borer P. fumosoroseus blastospores preparations were tested against silverleaf whiteflies (Bemisia argentifolii). While all preparations infected and killed B. argentifolii, fresh and air-dried preparations were significantly more effective. These results suggest that screening potential fungal biopesticides for amenability to liquid culture spore production can aid in the identification of commercially viable isolates. In this study, P. fumosoroseus was shown to possess the production and stabilization attributes required for commercial development.  相似文献

Enterobacter cloacae, one of the indigenous gut bacteria of the Formosan subterranean termite (Coptotermes formosanus), was genetically modified with a transposon Tn5 vector containing genes (tcdA1 and tcdB1) encoding orally insecticidal proteins from the entomopathogenic bacterium Photorhabdus luminescens subsp. laumondii TT01, a symbiont of the entomopathogenic nematode Heterorhabditis bacteriophora, for termite control. In the laboratory, termites were fed filter paper inoculated with the recombinant bacteria. The chromosomal expression of the introduced genes showed that there were insecticidal activities against termite workers and soldiers challenged with the transformed bacteria. After termites were fed recombinant bacteria, the termite mortality was 3.3% at day 5, and it increased from 8.7% at day 9 to 93.3% at day 29. All the dead termites contained the recombinant bacteria in their guts. Transfer of the recombinant bacteria occurred between donor workers (initially fed recombinant bacteria) and recipient workers (not fed). More than 20% of the recipient termites ingested recombinant bacteria within 2 h, and 73.3% of them had ingested recombinant bacteria after 12 h. The method described here provides a useful alternative for sustainable control of the Formosan subterranean termite (C. formosanus) and other social insects, such as the imported red fire ant (Solenopsis invicta).  相似文献

Use of proper application methods and formulations of termiticides are im- portant to reduce their negative impact to the environment. In this study, we conducted laboratory experiments to determine the effect of localized treatments with commercial dust and liquid formulations of fipronil against Formosan subterranean termites, Coptoter- mes formosanus Shiraki. The test arena consisted of a specially designed 16-chambered structure with a center chamber connected to 5 foraging chambers that themselves were connected to 10 additional foraging chambers. One peripheral chamber received a liquid or dust treatment and termites were released in the center chamber. Results showed that 〉 91% of the termites were dead within the 9-d test period despite the localized treatment of only 1 foraging chamber. Termites that were still alive after 9 d were transferred to an untreated dish and held for 10 more days. The majority of those termites were dead and the rest were moribund on day 19. Regardless of the specific dish treated, both formulations of fipronil were found to be highly efficacious. Termites did not exhibit repellency to either formulation. Our results suggest that localized (or spot) treatment with either commer- cially available dust or liquid formulations of fipronil can be a viable option for control of a termite infestation where complete soil drenching is not desirable.  相似文献

Experiments were conducted to examine competitive interactions between the Formosan subterranean termite, Coptotermes formosanus Shiraki (FST), and the eastern subterranean termite, Reticulitermes flavipes (Kollar) (EST), using groups of termites with different worker:soldier proportions. Experiments were conducted using three connected test chambers: an FST chamber, an unoccupied center chamber, and an EST chamber. When groups of FST were comprised of 20% soldiers versus 2% EST soldiers, only 8% of center chambers were occupied exclusively by EST. When groups of FST were comprised of 10% soldiers versus 1% EST soldiers, 44% of center chambers were occupied exclusively by EST. When the only food source was located in the center chamber, 60% of center chambers were occupied by both species. FST did not completely displace EST in any of these experiments.  相似文献

The Formosan subterranean termites, Coptotermes formosanus, were incubated in soils collected from various locations on the island of Hawaii. More than 70% of the termites died within 2 weeks in seven of nine soil samples collected from Kamuela. In control soil termites remained alive after 4 weeks. Entomophthora coronata, Metarrhizium anisopliae, Aspergillus flavus, and an unidentified bacterium were isolated from 60 to 100% of dead termites incubated in the pernicious soils. E. coronata and M. anisopliae were pathogenic to C. formosanus. The innocuous soil from Hilo became pernicious to C. formosanus after it was inoculated with E. coronata or M. anisopliae.  相似文献

Levee breaches because of Hurricane Katrina in 2005 inundated 80% of the city of New Orleans, LA. Formosan subterranean termites were observed actively foraging within in-ground monitoring stations within months after this period of flooding. It was unknown if the activity could be attributed to preexisting colonies that survived inundation or to other colonies surviving flooding by being located at higher elevations readily invading these territories. Genotypic profiles of 17 termite colonies collected from eight inundated locations before flooding were compared with termite colonies after flooding from the same locations to determine Formosan subterranean termite survival after sustained flooding. Results indicate that 14 colonies were able to survive inundation for extended periods.  相似文献

The impact of growing cultures of Paecilomyces fumosoroseus in liquid media containing four combinations of glucose and casamino acids (8 g l–1 or 80 g l–1 glucose, 1.32 g l–1 or 13.2 g l–1 casamino acids) was evaluated, based on blastospore production, germination rate, viability after freeze-drying and short-term storage stability. When blastospores were produced using a high casamino acid concentration, blastospore yields and germination rates were significantly higher (13.2–18.5×107 blastospores ml–1, 50–60% germination after 4 h), compared to cultures grown in media containing lower casamino acid concentrations (0.4–2.3×107 blastospores ml–1, 10–20% germination after 4 h). Chemical analyses of blastospore composition showed that accelerated blastospore germination may be related to increased proteinaceous reserves rather than to glycogen or lipid accumulation. Tolerance to freeze-drying by blastospores suspended in spent medium was enhanced by a high initial casamino acid concentration in the culture medium (75% survival) and by the residual glucose concentrations in the spent medium. Under the conditions of this study, the storage stability of blastospores of P. fumosoroseus was unaffected by the nutritional condition in which they were produced.  相似文献

Coptotermes formosanus Shiraki worker termites were sampled from 20 locations in the City Park area of New Orleans, LA. The termites were subsequently assayed to determine their susceptibility to cypermethrin, chlordane and chlorpyrifos, and detoxication enzyme activity. Cypermethrin was most toxic against Formosan subterranean termite workers, chlorpyrifos exhibited intermediate toxicity and chlordane was least toxic. A comparison of insecticide susceptibility between the most and least tolerant colonies revealed 1.9-, 1.7- and 1.8-fold differences in susceptibility for cypermethrin, chlorpyrifos and chlordane, respectively. As with the bioassay data, although significant differences were noted, a great deal of overlap was observed among the colonies for total cytochrome P450 content (difference of 2.2-fold between high and low value) aldrin epoxidation (3.6-fold) and cytosolic esterase (3.9-fold) activity. No significant differences were observed among the colonies for methoxyresorufin O-demethylase or glutathione S-transferase activity. Conversely, microsomal esterase activity varied greatly; a 38-fold difference was observed between the most (Cf1776) and least (Cf1387) active colonies. However, no significant correlation was observed between insecticide susceptibility and microsomal esterase activity. In fact, no significant correlations were observed between any of the enzyme activities measured and insecticide susceptibility. These results are discussed in the context of insecticide selection and future control effectiveness.  相似文献

Invasive ant species have general diet and nest requirements, which facilitate their establishment in novel habitats and their dominance over many native ants. The Asian needle ant, Pachycondyla chinensis, native throughout Australasia was introduced to the southeastern US where it has become established in woodland habitats, nests in close proximity to and consumes subterranean termites (Rhinotermitidae). P. chinensis do not occur in habitats lacking Rhinotermitidae. We suggest that subterranean termites are critical for P. chinensis success in new habitats. We demonstrate that P. chinensis is a general termite feeder, retrieving Reticulitermes virginicus five times more often than other potential prey near P. chinensis colonies. Odors produced by R. virginicus workers, as well as other potential prey, attract P. chinensis. Furthermore, P. chinensis occupy R. virginicus nests in the lab and field and display behaviors that facilitate capture of R. virginicus workers and soldiers. Termites are an abundant, high quality, renewable food supply, in many ways similar to the hemipteran honeydew exploited by most other invasive ant species. We conclude that the behavior of P. chinensis in the presence of termites increases their competitive abilities in natural areas where they have been introduced.  相似文献

Cuticular hydrocarbon patterns of the Formosan subterranean termite, Coptotermes formosanus Shiraki, were similar among colonies from the same geographical location. Hydrocarbon patterns of Florida colonies were easily distinguished from those of Hawaii colonies by using canonical discriminant analysis. Groups of termites from the same colony did not fight one another when placed in an arena. Intercolonial aggression was not recorded among C. formosanuspopulations from Florida but three colonies from Hawaii fought with the other Hawaiian and three Florida colonies. Of the 12 colonies (six each from Florida and Hawaii) tested, 3 Florida colonies did not direct or receive aggression from any other colony. Cuticular hydrocarbon patterns were not correlated with agonistic behavior.  相似文献

Strains of the entomopathogens Beauveria bassiana (Balsamo) Vuillemin and Metarhizium anisopliae (Metschnikoff) Sorokin were screened for effectiveness against the Formosan subterranean termite, Coptotermes formosanus (Shiraki). Biological control methods are being considered to augment current integrated pest management schemes that involve chemical baits and nonrepellent termiticides. In this study Formosan subterranean termites selected from four colonies in the field were exposed in the laboratory to three strains of Beauveria bassiana and two strains of Metarhizium anisopliae . The exposed termites were then allowed to mingle with an equal number of unexposed termites from the same nest and mortality of the combined group was determined. One week after exposure, B. bassiana strains, ATCC 26037 and ATCC 90519, had caused significantly higher mortality than the two M. anisopliae strains, ESC 1 and II B and the remaining strain of B. bassiana , ATCC 90518. The LT50 measurement for strains 26037 and 90519 was comparable at 1.8 and 2.0 days, respectively. Strains ESC 1 and 90518 caused 50% mortality at 6.5 and 11.0 days, respectively. Termites exposed to strain II B and the control group of termites did not reach 50% mortality during the 21 day experimental period. Exposure of termites to all fungal strains resulted in mortality in excess of that seen in the control groups of unexposed termites.  相似文献
