AIMS: To test the suitability of the Agrobacterium tumefaciens-mediated transformation (AMT) method with Paecilomyces fumosoroseus, a fungal pathogen that causes diseases in a wide range of insects including whiteflies. METHODS AND RESULTS: Conidia of P. fumosoroseus were successfully transformed to hygromycin B resistance using the hph gene of Escherichia coli as the selectable marker. Transformation frequencies were 58.3 +/- 18.5, 98.3 +/- 24.8 and 169.7 +/- 35.5 (+/-SEM) transformants per 10(5), 10(6) and 10(7) target conidia respectively. After confirmation by PCR, transformants were subjected to Southern analysis, and the results revealed that 45% (four of nine) of the transformants contained single-copy integration of the T-DNA. CONCLUSIONS: In our AMT system, we efficiently transformed conidia of P. fumosoroseus. The employment of this method circumvents time-consuming protoplast preparation and allows the isolation of transformants containing single-copy integration of the T-DNA. SIGNIFICANCE AND IMPACT OF THE STUDY: Considering the efficiency of Ag. tumefaciens-mediated transformation, this method represents a useful tool for insertional mutagenesis to characterize genes that are important for the pathogenicity of P. fumosoroseus. 相似文献

Blastospores of the entomopathogenic fungus Paecilomyces fumosoroseus were formulated with 10% lactose/1% bovine serum albumin (BSA) or various compositions of Fantesk™, a starch-oil composite prepared by jet-cooking an aqueous dispersion of starch and oil. Storage stability studies with wet blastospore formulations showed that maximum blastospore survival was achieved during low-temperature storage at -20°C with lactose/BSA formulations or starch-oil formulations supplemented with sucrose, zein protein, and whole milk. Under conditions of wet storage at -20°C, the addition of whole milk to starch-oil formulations significantly improved blastospore stability while the addition of sucrose or zein protein had no effect. In freeze-drying studies, no significant differences were seen in blastospore desiccation tolerance or in stability during storage at either 4 or -20°C when blastospores of P. fumosoroseus were formulated with lactose/BSA or starch-oil formulations with sucrose, zein protein, and whole milk. Freeze-dried blastospore formulations stored at 4°C showed no loss in blastospore viability after 3 months storage and blastospore formulations stored at -20°C showed no loss in viability during the entire 12-month study. For freeze-dried, starch-oil formulations, sucrose was shown to improve blastospore survival during the freeze-drying process. The addition of whole milk to starch-oil formulations significantly improved the stability of freeze-dried blastospores stored at 4°C. Compared to unformulated blastospore suspensions that showed blastospore settling after 30 min, suspensions of blastospores formulated with lactose/BSA or starch-oil composites remained stable for up to 2 h after mixing. 相似文献

The entomopathogenic fungi, Isaria farinosa and Isaria fumosorosea were known as Paecilomyces farinosus and Paecilomyces fumosoroseus, for more than 30 years. Both fungi have a worldwide distribution and a relatively wide host range. While I. farinosa currently is of minor importance in research and as biocontrol agent, I. fumosorosea is regarded as a species complex, and various strains are successfully used for biocontrol of several pest insects, mainly whiteflies. During the past 40 years, numerous scientific papers on both fungal species covering various research areas were published. This is the first monographic review on the biology, ecology and use of I. farinosa and I. fumosorosea as biocontrol agents. In the following compilation, data on the identity, natural occurrence and geographical distribution, host range, production of metabolites, effect of abiotic and biotic factors (temperature, relative humidity, solar radiation, fungus, host plant) and on the use of these species in biocontrol experiments against pest insects, plant pathogens and nematodes in the laboratory and the field are summarised. Sections on their effects on non-target organisms and safety issues are also included. The aim of this review is to increase our knowledge on these entomopathogenic fungi. 相似文献

AIMS: This study was to illustrate the relationship between the thermotolerance and the contents of hydrophobin-like or formic-acid-extractable (FAE) proteins in aerial conidia of Beauveria bassiana and Paecilomyces fumosoroseus produced on rice-based substrate. METHODS AND RESULTS: Survival indices of 11 isolates were separately assessed as a ratio of the viability of conidia after 3-150 min thermal stress at 48 degrees C over that of unstressed conidia and fitted well to a survival model (r(2) >/= 0.97). For a given isolate, the fitted model generated an LT(50), the time for 50% viability loss under the stress. The LT(50)s of six B. bassiana isolates (10.1-61.9 min) and five P. fumosoroseus isolates (2.8-6.2 min) were correlated (r(2) = 0.81) with FAE protein contents (6.9-23.4 microg mg(-1)). The survival indices of a fixed B. bassiana isolate after 45-min thermal stress at 48 degrees C were also correlated to the FAE protein contents from conidia produced on glucose-, sucrose-, or starch-based substrate (0.79 = r(2) = 0.86). Three bands of 12.0, 15.0 and 17.5 kDa were recognized from SDS-PAGE profiles of protein extracts from different isolates or from the fixed isolate grown on the three carbon sources, but the 12 kDa was less likely related to the thermotolerance. CONCLUSIONS: Approximately 80% of variability in conidial thermotolerance was attributed to either 15.0- or 17.5-kDa FAE protein or both. SIGNIFICANCE AND IMPACT OF THE STUDY: The relationship between conidial thermotolerance and FAE protein contents implicates a new way to search for fungal biocontrol agents with more thermotolerance and environmental persistence. 相似文献

Selected allelochemicals that protect plants from invasion by plant pathogenic fungi were investigated for their activity
against the entomopathogenic fungus, Paecilomyces fumosoroseus. The alkaloids tomatine, solanine, and camptothecin; the furanocoumarin,
xanthotoxin; and the phenolic, tannic acid were tested for their effects on germination of conidia and blastospores and growth
of mycelia. The LC50 values (corresponding to 50% inhibition of germination) for tomatine, solanine, camptothecin, xanthotoxin
and tannic acid were 51.6, 95.9, 55.9, 83.0 and 72.8 mg/l respectively. When blastospores were placed on media containing
a concentration of the individual allelochemicals that inhibit germination in approximately 50% of conidia, all but blastospores
on tomatine had significantly less germination than did aerial conidia. Growth rates of mycelia were slowest in the camptothecin
medium, followed by those of tomatine and xanthotoxin and were not significantly different from controls in the media containing
solanine and tannic acid. A multitude of biotic and abiotic factors are responsible for specificity and degree of pathogenicity
of entomopathogens. The effect of crop plant chemistry on the efficacy of entomopathogens should be quantified further in
order to maximize their potential when used concomitantly with resistant plant varieties.

Abstract: An isolate of Paecilomyces fumosoroseus was obtained from Coptotermes formosanus collected in Hong Kong, and a commercially available isolate of Metarhizium anisopliae , were both tested against C. formosanus shipped live from China. Survivorship of termites treated with a suspension of 5 × 105 M. anisopliae conidia/ml and kept alone declined more rapidly than for those treated at the same concentration of P. fumosoroseus conidia. At a 5 × 106 conidia/ml concentration, no significant differences in terms of termite survivorship were observed between the two fungal species. However, among termites kept in groups of 10 after treatment, those sprayed with P. fumosoroseus conidia at either 5 × 105 or 5 × 106 conidia/ml had significantly lower survivorship than those sprayed with M. anisopliae conidia. All the cadavers of termites treated with P. fumosoroseus and kept alone sporulated and among grouped termites 29% of the cadavers sporulated. By comparison, 53% of the cadavers of termites treated with M. anisopliae and kept alone sporulated, and only 4% of the cadavers of treated termites kept in groups sporulated. 相似文献

Formulation matrices can play an important role in improving the storage survival and biocontrol efficacy of microorganisms used for the control of pest insects. In this study, liquid culture-produced blastospores of the entomopathogenic fungus Paecilomyces fumosoroseus were formulated with different inert and organic materials prior to air-drying. Paecilomyces fumosoroseus blastospores were produced in two different liquid media, a basal salts medium supplemented with Casamino acids and glucose (LM1) and a medium containing peptone of collagen and glucose (LM2). Blastospores produced in the two test media were formulated with various supports. The formulation supports were cornstarch, rice flour, talc powders, Mexican lime, calcined kaolin clay, and diatomaceous earth. Several of the supports were tested at different concentrations. The initial and long-term (after storage at 4 and 28 °C) survival of the formulated, air-dried blastospores were evaluated. Initial blastospore viabilities were affected by the formulation material and by the blastospore production medium. Medium composition, drying support and storage temperature had an impact on the long-term survival of the blastospores. Under the conditions of the study, LM1 produced higher concentrations of blastospores that not only survived drying better than blastospores produced in LM2 but also maintained viability longer during storage in the formulation supports tested. The nature of the drying supports was shown to have a significant impact on the storage stability of all blastospores, particularly those produced in LM1. Under the production, drying and storage conditions used in the study, calcined kaolin clay formulations stored at 4 °C had the best storage stability. In all formulations tested, spore survival over time was reduced for blastospore formulations stored at 28 °C rather than 4 °C. 相似文献

The entomopathogenic Hyphomycete Paecilomyces fumosoroseus was grown in five different liquid media, which have been developed
for mass production of Beauveria spp. or P. fumosoroseus. Production was followed for 96 h by measuring both biomass and concentration
of propagules. Maximal biomass was obtained with two media, Jackson and Catroux media (40–60 mg ml-1 suspension produced after
42 h incubation), where the exponential phase of growth began earlier than in the other media. While high concentrations of
propagules (1.4–5.5 × 108 propagules ml-1) were produced in three media (Jackson, Paris, and Catroux media) after 48–72 h
incubation, production of propagules was lower in the two other media, containing maltose as carbon source (Goral and Kondryatiev
media) with 0.4–3.7 × 107 propagules ml-1 after 96 h incubation. P. fumosoroseus produced oblong blastospores in the three
most productive media and conidiospore-like (ovoid to subspherical) propagules in the two other media. Infection potential
of produced propagules was tested on the silverleaf whitefly ( Bemisia argentifolii). Whiteflies were sprayed as 2nd instars
with P. fumosoroseus propagules produced in the five liquid media (1.9 × 103 and 1.9 × 104 propagules cm-2). All the media
produced propagules that were infectious for larvae. With the lower dosage, mortality rates were significantly lower with
propagules produced in one of the two least productive media (57%) (in the Kondryatiev medium) compared to those obtained
with the three most productive media (>90%). However, when whiteflies were treated with the higher dosage, mortality rates
(91–99%) between media were not significantly different.

Abstract: The detrimental photic effects of natural solar radiation on the conidial persistence of the entomopathogenic hyphomycete Paecilomyces fumosoroseus were investigated by exposing quiescent conidia either to total solar radiation or to its diffuse component. A given amount of UVB diffuse radiation was found to be as detrimental, and sometimes twice as detrimental, as the same amount of total solar radiation. The variability in quantity and spectral distribution of the diffuse component of UVB solar radiation reaching the earth's surface, observed through spectral measurements, may be responsible for the difference in biological effects. 相似文献

AIMS: In the present study, two different optimization techniques were used to determine the suitable operating parameters for exo-biopolymer production in submerged mycelial cultures of two entomopathogenic fungi Paecilomyces japonica and Paecilomyces tenuipes. METHODS AND RESULTS: First, the rotating simplex method, a nonstatistical optimization technique, was employed to obtain the best combination of physical parameters (viz. pH, agitation intensity, aeration rate) for maximum exo-biopolymer production by P. japonica in a batch bioreactor. The optimal combination was determined to be a pH of 8.06, an aeration of 3 vvm, without any impeller agitation, producing a 17-time increase in exopolymer production (34.5 g l(-1)) when compared with that achieved in unoptimized flask cultures. Second, the uniform design method, a statistical optimization technique, was employed to determine the best operating parameters for submerged culture of P. tenuipes. The optimal combination for mycelial growth was determined to be a pH of 4.88, an aeration of 2 vvm and an agitation of 350 rpm, while a pH of 4, an aeration of 2 vvm and an agitation of 150 rpm was best for exo-biopolymer production. CONCLUSIONS: The exo-biopolymer production in P. japonica optimized by the rotating simplex method was strikingly improved (max. 34.5 g l(-1)), and the exo-biopolymer production in P. tenuipes optimized by the uniform design method was also significantly increased (max. 3.4 g l(-1)). SIGNIFICANCE AND IMPACT OF THE STUDY: The successful application of these two different optimization techniques in this study implies that these methods are worthy of applying to other fermentation systems for the production of bioactive mycelial biomass and exo-biopolymers in liquid culture of higher fungi. 相似文献

A Paecilomyces fumosoroseus strain was mutagenized by u.v. Among 200 colonies, one mutant (M84), showed a large and stable chitin hydrolysis-halo. Glucose consumption and biomass production were similar for M84 and the parental strain. Chitinase was inducible by chitin and repressed by glucose in both strains but, when they were grown on minimal medium plus colloidal chitin as sole carbon source, the parental and M84 strains yielded 198 and 690 mol N-acetylglucosamine, respectively. This results indicate that the mutant strain synthesized a chitinase with a higher activity. Bioassays against Bemisia tabaci nymph, showed that M84 incited a 2-fold higher incidence of disease compared to the parental strain. 相似文献

Persistence of conidia of the entomopathogenic fungus Paecilomyces fumosoroseus exposed to artificial and solar radiation at a constant temperature was studied by monitoring the ability to germinate and to form colonies (colony - forming units , CFUs) . The photic effect of radiation on each of these variables was modelled by a decreasing function of UVB irradiation ( in J m 2) . Germination ability was represented by a logistic function and viability (log CFU) by an infinitely decreasing function . Experiments carried out under artificial conditions , at three different UVB irradiances ( from 0 . 3 to 1 . 6 W m 2) , similar to those observed in nature , confirmed the adequacy of the predictor variable and of the functions chosen for describing these data . The proposed models appeared to be irradiance independent . Under solar radiation , the models were able to describe data collected on three different summer days in France (48 o 51 N , 2 o 06 E) . However , it took a greater amount of solar UVB radiation to produce the same effect as that achieved indoors . This could be explained by differences in radiation spectra . For each model , one set of parameters was sufficient for representing all three sets of data: this constitutes an initial validation of the models proposed . 相似文献

With the goal of developing a defined medium for the production of desiccation-tolerant blastospores of the bioinsecticidal
fungus Paecilomyces fumosoroseus, we evaluated the impact of various media components such as amino acids, carbohydrates, trace metals and vitamins on hyphal
growth and sporulation of P. fumosoroseus cultures and on the freeze-drying tolerance of blastospores produced under these conditions. A comparison of 13 amino acids
as sole nitrogen sources showed that glutamate, aspartate, glycine and arginine supported biomass accumulations (12–16 mg ml−1) and blastospore yields (6–11 × 108 blastospores ml−1) comparable to our standard production medium which contains casamino acids as the nitrogen source. Using glutamate as the
sole nitrogen source, tests with various carbohydrates showed that P. fumosoroseus grew best on glucose (18.8 mg biomass ml−1) but produced similar blastospore concentrations (7.3–11.0 × 108) when grown with glucose, glycerol, fructose or sucrose. P. fumosoroseus cultures grown in media with sodium citrate or galactose as the sole carbohydrate produced lower blastospore concentrations
but more-desiccation-tolerant spores. Zinc was the only trace metal tested that was required for optimal growth and sporulation.
In a defined medium with glutamate as the nitrogen source, vitamins were unnecessary for P. fumosoroseus growth or sporulation. When blastospores were freeze-dried in the absence of a suspension medium, residual glucose (>2.5%
w/v) was required for enhanced spore survival. Thus, a defined medium containing basal salts, glucose, glutamate and zinc
can be used to produce optimal concentrations of desiccation-tolerant blastospores of P. fumosoroseus.

Paecilomyces fumosoroseus (Wize) Brown & Smith is under development as a mycoinsecticide for control of the Russian wheat aphid, Diuraphis noxia Kurdjumov. Interactions with other natural enemies within the agro-ecosystem, such as the coccinellid Hippodamia convergens Guerin, require evaluation before its potential can be realized. In laboratory bioassays H. convergens adults were sprayed with suspensions of P. fumosoroseus conidia at different concentrations (including potential field rates) and mortality assessed. Although a proportion of coccinellids succumbed to infection (the greatest proportion was 22% when the ladybirds had suffered stress) it is unlikely that they would be at risk from infection as a direct result of a spray application unless there were prolonged periods of high humidity or the coccinellids were stressed. When provided with uninfected or P. fumosoroseus -infected D. noxia cadavers as prey, coccinellids consumed more uninfected aphids. The predators never consumed aphid cadavers from which the fungus was erupting or sporulating. Hippodamia convergens is, therefore, unlikely to be a significant intraguild predator of P. fumosoroseus . Predators contaminated with conidia of P. fumosoroseus using different methods (sprayed coccinellids, coccinellids foraging on sprayed aphids, and those foraging in the presence of sporulating D. noxia cadavers) were able to transfer conidia to healthy D. noxia populations and initiate infection in a proportion of those aphids. The proportion of aphids becoming infected was greatest when the coccinellids became contaminated when foraging amongst sporulating cadavers. The nematophagous fungi Paecilomyces lilacinus and Pochonia chlamydosporia have been extensively studied as biological control agents for plant-parasitic nematodes. This study describes the formulation of alginate pellets containing mycelia of these fungi and also describes the effect of storage conditions on shelf-life of the pellets. The shelf-lives of P. lilacinus and P. chlamydosporia, which were measured monthly for 6 months, were significantly improved at low temperatures and low water activity (a w) values (<0.33). Vacuum did not affect the viability of the formulated P. lilacinus but increased the viability of P. chlamydosporia. Carbon dioxide reduced the activity of P. lilacinus as compared to ambient air but increased the activity of P. chlamydosporia. Nitrogen, however, significantly improved the viability of both fungi. The optimal parameters of each factor for our formulation of P. lilacinus and P. chlamydosporia included a temperature range of 4 to ?20°C, a w=0.12, and a nitrogen-filled atmosphere. 相似文献

Several entomopathogenic fungi produce toxins that could be used as bioinsecticides in integrated pest management programs.
Paecilomyces fumosoroseus is currently used for the biological control of the whiteflies Bemisia tabaci and B. argentifolii. Supernatants from submerged batch culture, where the fungus produced abundant dispersed mycelium, conidia and blastospores,
were toxic to the whitefly nymphs. The most abundant metabolite was purified by HPLC and identified by mass spectrometry and
NMR as dipicolinic acid. Both the dipicolinic acid produced by the fungus and the chemically synthesized compound had insecticidal
activity against third-instar nymphs of the insect. Dipicolinic acid was toxic to the whitefly nymphs in bioassays involving
topical applications. In submerged culture, the specific growth rate of P. fumosoroseus was 0.054 h−1, the specific glucose consumption rate was 0.1195 g g−1 h−1 and the specific dipicolinic acid production rate was 0.00012 g g−1 h−1. Dipicolinic acid was detected after 24 h when the fungus started growing; and dipicolinic acid production was directly correlated
with fungal growth. Nevertheless, the yield was low and the maximal concentration was only 0.041 g l−1. The maximal concentrations of conidia and blastospores (per milliliter) were 1.4×108 and 7×107, respectively. 相似文献

In shake flask and fermentor studies, various media components and culture inocula were tested to improve P. fumosoroseus spore production rates, yield and stability. To evaluate inoculum potential and inoculum scale-up for fermentor studies, conidia and liquid culture-produced spores of various strains of P. fumosoroseus were compared as inoculum. Inoculation of liquid cultures with blastospores at concentrations of at least 1×106 spores mL-1 resulted in the rapid production of high concentrations of blastospores (∼1×109 spores mL-1, 48 h fermentation time) for all strains tested. The rapid germination rate of blastospores (90% after 6 h incubation) compared to conidia (>90% after 16 h incubation) and the use of higher inoculum rates reduced the fermentation time from 96 to 48 h for maximal spore yields. A comparison of various complex nitrogen sources showed that liquid media supplemented with acid hydrolyzed casein or yeast extract supported the production of high concentrations of blastospores that were significantly more desiccation-tolerant (79-82% survival after drying) when compared to blastospores produced in media supplemented with other nitrogen sources (12-50% survival after drying). For rapid spore production, requirements for trace metals and vitamin supplementation were dependent on the type of hydrolyzed casein used in the medium. Fermentor studies with two strains of P. fumosoroseus showed that high concentrations (1.3-1.8×109 spores mL-1) of desiccation-tolerant blastospores could be produced in 48-h fermentations. These studies have demonstrated that the infective spores of various strains of the fungal bioinsecticide Paecilomyces fumosoroseus can be rapidly produced using deep-tank, liquid culture fermentation techniques. 相似文献

Mineral oil-based emulsifiable preparations of Beauveria bassiana (Bb) and Paecilomyces fumosoroseus (Pfr) conidia were separately applied alone or together with low rates of imidacloprid 10% WP at 4.7% (Im 1), 14.0% (Im 2), and 23.3% (Im 3) of its recommended application rate, respectively, against the greenhouse whitefly, Trialeurodes vaporariorum, on lettuce grown in the greenhouse. Besides eight fungal treatments, the three low application rates of imidacloprid in the oil-based carrier and a blank control (CK) were also included as treatments of the trials conducted in 2002 and 2003. For the 12 treatments of each trial with three replicates, 1,000-fold aqueous dilutions were sprayed twice on transplanted lettuce at a 15-day interval at a rate of ∼1.43×1013 conidia ha-1 for each fungal treatment or at one of the low rates of imidacloprid using a backpack hand-operated hydraulic sprayer. Based on whitefly densities, mortalities, relative efficacies and percent density declines estimated from whitefly counts made at 5-day intervals, all B. bassiana and P. fumosoroseus sprays were highly effective against T. vaporariorum compared to CK and Im 1-3 in both trials. In trial 1, the estimates of whitefly density decline and relative efficacy ranged from 44 and 72% (Bb) to 79 and 90% (Pfr+Im 2-3) on day 10 after the first spray and exceeded 94% for all fungal treatments 10 days after the second spray. Similar trends in whitefly control were also achieved in the corresponding treatments of trial 2. A more desirable and faster control resulted from fungal sprays containing more imidacloprid, but none of the three low imidacoprid rates alone suppressed the whitefly population more effectively than any fungal treatment despite a varying degree of efficacy. Moreover, P. fumosoroseus tended to be more effective against T. vaporariorum than B. bassiana when applied alone or together with the same low rate of imidacloprid but the difference in whitefly control eventually achieved was not significant between the two fungal agents. Thus, the emulsifiable preparations of both fungal agents can be considered as alternatives for whitefly management and for a slightly faster control can be applied together with imidacloprid at around 15% of its common application rate. An economic balance between the efficacy of whitefly control and the ease and cost of conidial production is needed when both fungi are incorporated into whitefly management. 相似文献

Hyphomycete Paecilomyces fumosoroseus that is well known as saprophytic and entomopatogenic fungus was investigated for its mycoparasitism on the cucumber powdery mildew pathogen. Mycoparasitism was documented by using standard bioassay and SEM. Effects of mycoparasitism were evaluated in three types of experiments. Paecilomyces fumosoroseus was applied in the form of graded suspensions into a colony of powdery mildew on a leaf segment. Interaction between both fungi was observed as the percentage of colonized area vs. experimental time. In the second experiment, young cucumber plants were sprayed with a suspension of Paecilomyces fumosoroseus 24 h before inoculation of Sphaerotheca fuliginea. Pre-treatment with P. fumosoroseus reduced development and spreading of powdery mildew infection significantly 15 days post-inoculation in contrast to pre-treatments with sulfur fungicide and distilled water. The development of pure culture powdery mildew under determined experimental conditions was observed and compared with treated variants. In the third experiment, mildewed plants were treated with a suspension of P. fumosoroseus. The control treatments with sulfur fungicide and distilled water were tested. Effects of P. fumosoroseus on the dispersion of powdery mildew during a 21-day period were observed.P. fumosoroseus suppressed the development and spread of cucumber powdery mildew significantly during the time of the experiment. The mechanical and physical damages and disruptions of vegetative and fruiting structures of powdery mildew were recorded under light microscopy and S.E.M.Results were concluded in pursuance to differences between the natural behaviour and development of S. fuliginea on cucumber plants treated with P. fumosoroseus and non-treated plants. 相似文献
