Alleged primitive feathers or protofeathers in the theropod dinosaur Sinosauropteryx have potentially profound implications concerning feather morphogenesis, evolution offlight, dinosaur physiology and perhaps even the origin of birds, yet their existence has never been adequately documented. We report on a new specimen of Sinosauropteryx which shows that the integumental structures proposed as protofeathers are the remains of structural fibres that provide toughness. The preservation in the proximal tail area reveals an architecture of closely associated bands offibres parallel to the tail's long axis, which originate from the skin. In adjacent more exposed areas, the fibres are short, fragmented and disorganized. Fibres preserved dorsal to the neck and back and in the distal part of the tail are the remains of a stiffening system of a frill, peripheral to the body and extending from the head to the tip of the tail. These findings are confirmed in the holotype Sinosauropteryx and NIGP 127587. The fibres show a striking similarity to the structure and levels of organization of dermal collagen. The proposal that these fibres are protofeathers is dismissed.  相似文献

The ultrastructure of dermal fibres of a 200Myr thunniform ichthyosaur, Ichthyosaurus, specifically the 67nm axial repeat D-banding of the fibrils, which characterizes collagen, is presented for the first time by means of scanning electron microscopy (SEM) analysis. The fragment of material investigated is part of previously described fossilized skin comprising an architecture of layers of oppositely oriented fibre bundles. The wider implication, as indicated by the extraordinary quality of preservation, is the robustness of the collagen molecule at the ultrastructural level, which presumably contributed to its survival during the initial processes of decomposition prior to mineralization. Investigation of the elemental composition of the sample by SEM-energy dispersive X-ray spectroscopy indicates that calcite and phosphate played important roles in the rapid mineralization and fine replication of the collagen fibres and fibrils. The exceedingly small sample used in the investigation and high level of information achieved indicate the potential for minimal damage to prized museum specimens; for example, ultrastructural investigations by SEM may be used to help resolve highly contentious questions, for example, 'protofeathers' in the Chinese dinosaurs.  相似文献

The cuticle of Gordius panigettensis (Sciacchitano, 1955) was studied by scanning electron microscopy (SEM), transmission electron microscopy (TEM) and atomic force microscopy (AFM). The cuticle is composed of 30-50 compact layers. The number of the layers is higher in the central part of the animal's body and decreases at the extremities. Each layer is composed of parallel tightly packed fibres approximately 640 nm in diameter and of indefinite length. The fibres run strictly parallel within each layer, while in adjoining layers they run at a variable angle from 45 degrees in the central body to 90 degrees in the extremities. Each fibre shows a barely detectable filamentous inner structure and is enveloped in a thin highly regular net formed by hexagonal meshes. Our results suggested that these fibres should be proteinaceous although non-collagenous. Thinner radial fibres run among the large fibres and across all the layers and span the whole thickness of the cuticle from the epithelial layer located deep underneath the large fibres up to the epicuticle on the external surface of the animal.  相似文献

Specimens of abdomen skin, comprising alternate areas of striae albae and healthy skin, were removed during surgical lipectomy from multiparous and obese women between the ages of 24 and 53 years. A flattening and thinning of the striae albae surface and the almost complete disappearance of dermal papillae was observed in paraffin and thin sections. The papillary dermis was found to be almost completely replaced by straight bundles of collagen fibres running parallel to the skin surface. Immunofluorescence data revealed in these bundles high positivity for type I collagen. The underlying reticular dermis was also found to contain large densely packed bundles of collagen fibres running parallel to the skin surface. Both papillary and reticular dermis collagen fibres were mainly arranged orthogonally to the main axis of the stria. Furthermore, the density of the collagen fibre bundles and the diameter of the collagen fibrils was found to be greater than that of the clinically healthy skin. A larger number of elastic fibres, which presented an abnormal ultrastructural appearance, were visible in pathological papillary and reticular dermis.  相似文献

Fibre-type differentiation of the lateral musculature has been studied in Sparus aurata (L.) and Dicentrarchus labrax (L.) during larval development. Histochemical and ultrastructural techniques show two presumptive muscle layers and two germinative zones of presumptive myoblasts. At hatching, myotomal muscle consists of a monolayer of thin undifferentiated cells near the skin (first germinative zone) overlying another mono-layer of small diameter fibres extending hypaxially and epaxially away from the transverse septum. Below this, there is a much thicker, deep layer of fibres, generally large in diameter and polygonal in shape. The presumptive myoblasts are located between these two layers of fibres in the second germinative zone. Initially, the superficial and deep muscle fibres show high and low myosin ATPase activity, respectively. Both layers grow by generating new fibres from the two mentioned germinative zones. At the end of larval life, the superficial layer changes its histochemical profile from high to low myosin ATPase activity and, at the same time, intermediate or pink muscle fibres can be observed by oxidative activity (the NADH-TR reaction). Morphometric analysis shows a significant increase in mean fibre diameter during successive ages, as shown by the Student's t-test (hypertrophic growth). Skewness and kurtosis values of fibre diameters point to the generation of a new fibre population from the germinative zones (hyperplastic growth).  相似文献

The aim of this study was to describe the histological structure of the skin of greater rhea (Rhea americana), a ratite bird native to South America. Skin samples were taken from three regions of the trunk (alar, dorsal and pelvic) in 14 specimens which ages ranged from 7 days to adulthood. Serial sections were obtained and subjected to different staining procedures (haematoxylin and eosin, orcein, Masson's trichrome and Gomori), and a morphometric analysis was carried out on stained slides. In general, both epidermis and dermis showed increased thickness of its layers with age. Some differences between regions can be detected both in epidermis and in dermis; for example in adults and 7‐day‐old birds, the stratum corneum of the alar region was thicker than of the dorsal region. In general, the skin of greater rhea was similar to that described in ratites and other birds (a thin epidermis compared to dermis, dermis with scarce elastic fibres, a slender and vascularized stratum superficiale, collagen fibres arranged in three directions). The scarcity of elastic fibres and the general cross‐weaved arrangement of the collagen fibres in the dermis of the adult greater rhea provide strength and flexibility to the dermis, two important features in leather industry.  相似文献

Epithelial cells destined to form the hair fibre begin to differentiate while still in the hair follicle bulb. The fibre cells continue to differentiate as they migrate out of the bulb and up the follicle towards the skin surface. The anatomy of the hair follicle and the different cell types observed within the follicle are briefly reviewed. A theoretical scheme for cell differentiation, capable of producing all the observed mature cell types, is presented. A major component of the scheme is a reaction-diffusion system of morphogens similar to that originally proposed by Turing (1952). The mathematical solution of the equations defining the reaction-diffusion system within the follicle bulb is discussed. The sequence of patterns in the spatial distribution of the morphogens expected in the hair follicle bulb is calculated and found to be in good agreement with the sequence of patterns of orthocortical and paracortical cells in the fibre cross section as the diameter of the fibre increases. The spatial patterns of the morphogens are also compared with the shape of the fibre cross section. It is concluded that a reaction-diffusion system may play a major role in the morphogenesis of hair fibres.  相似文献

Abstract:  An exceptionally preserved herd of the ceratopsian dinosaur Psittacosaurus was recovered from the Lujiatun Beds of the Lower Cretaceous Yixian Formation, Liaoning Province, north-east China. This specimen provides the earliest direct evidence of post-nestling gregarious behaviour in this important dinosaur clade. The sediment in which the skeletons occur represents a lahar (volcanic mudflow), indicating that the six individuals present were entombed simultaneously during a catastrophic mass mortality event. Reconstruction of the herd age profile shows it was composed of offspring from at least two clutches, revealing that group fidelity in these dinosaurs extended beyond time spent together in the nest. This demonstrates that sociality appeared much earlier in ceratopsian evolution than is generally acknowledged and that such behaviour preceded the evolution of the elaborate horns and frills seen in derived ceratopsids.  相似文献

Summary The ultrastructure and vascular supply of the different fibre types in the lateral muscles of the sturgeon Acipenser stellatus were studied by light- and electron microscopy and morphometry. Three fibre types form separate layers without intermingling. The red fibres are superficial, the white fibres deep and the intermediate fibres between them. From morphometric analyses, the mitochondrial volume fraction in red fibres is 30%, in intermediate fibres 3.7% and in white fibres 0.7%. Z lines are most fuzzy in the red fibres. Triads of the sarcotubular system are always situated at the level of the Z discs. In red fibres the three elements are arranged in a series along the myofibrils, whereas in white fibres they are arranged transversely and in the intermediate fibres they are aligned obliquely. The number of capillaries surrounding each fibre is 2.3, 0.9 and 0.2 for the red, intermediate and white fibres, respectively. In red fibres 16% of the surface is directly covered by capillaries. The corresponding percentages for intermediate and white fibres are 5 and 1, respectively. Per unit volume of the fibre, the directly vascularised fibre surface in red fibres is about ten times larger than that of white fibres.The degree of vascularisation of the fibre types is directly related to the volume fraction of mitochondria, and thus to their aerobic capacities.  相似文献

The nerves and nerve endings in the skin of tropical cattle were studied using histological and histochemical techniques. Many nerve trunks and fibres were present in the reticular and papillary dermis in both hairy and non-hairy skin sites. In non-hairy skin locations such as the muzzle and lower lip, encapsulated endings akin to Krause and Ruffini end bulbs, which arise from myelinated nerve trunks situated lower down the dermis were observed at the upper papillary layer level. Some fibre trunks seen at this level extended upwards to terminate within dermal papillae as bulb-shaped longitudinally lamellated Pacinian-type endings, while other onion-shaped lamellated nerve structures were located either within dermal papillae or near the dermo-epidermal area. Intraepidermal free-ending nerve fibres, appearing non-myelinated were observed in areas with thick epidermis. Intraepidermal free-ending nerve fibres, appearing non-myelinated were observed in areas with thick epidermis. On hairy skin sites, however, organized nerve endings or intraepidermal nerve endings were not readily identifiable.  相似文献

The origin of birds and avian flight from within the archosaurian radiation has been among the most contentious issues in paleobiology. Although there is general agreement that birds are related to theropod dinosaurs at some level, debate centers on whether birds are derived directly from highly derived theropods, the current dogma, or from an earlier common ancestor lacking suites of derived anatomical characters. Recent discoveries from the Early Cretaceous of China have highlighted the debate, with claims of the discovery of all stages of feather evolution and ancestral birds (theropod dinosaurs), although the deposits are at least 25 million years younger than those containing the earliest known bird Archaeopteryx. In the first part of the study we examine the fossil evidence relating to alleged feather progenitors, commonly referred to as protofeathers, in these putative ancestors of birds. Our findings show no evidence for the existence of protofeathers and consequently no evidence in support of the follicular theory of the morphogenesis of the feather. Rather, based on histological studies of the integument of modern reptiles, which show complex patterns of the collagen fibers of the dermis, we conclude that "protofeathers" are probably the remains of collagenous fiber "meshworks" that reinforced the dinosaur integument. These "meshworks" of the skin frequently formed aberrant patterns resembling feathers as a consequence of decomposition. Our findings also draw support from new paleontological evidence. We describe integumental structures, very similar to "protofeathers," preserved within the rib area of a Psittacosaurus specimen from Nanjing, China, an ornithopod dinosaur unconnected with the ancestry of birds. These integumental structures show a strong resemblance to the collagenous fiber systems in the dermis of many animals. We also report the presence of scales in the forearm of the theropod ornithomimid (bird mimic) dinosaur, Pelecanimimus, from Spain. In the second part of the study we examine evidence relating to the most critical character thought to link birds to derived theropods, a tridactyl hand composed of digits 1-2-3. We maintain the evidence supports interpretation of bird wing digit identity as 2,3,4, which appears different from that in theropod dinosaurs. The phylogenetic significance of Chinese microraptors is also discussed, with respect to bird origins and flight origins. We suggest that a possible solution to the disparate data is that Aves plus bird-like maniraptoran theropods (e.g., microraptors and others) may be a separate clade, distinctive from the main lineage of Theropoda, a remnant of the early avian radiation, exhibiting all stages of flight and flightlessness.  相似文献

BACKGROUND AND AIMS: The anatomy of bamboo culms and the multilayered structure of fibre cell walls are known to be the main determinant factors for its physical and mechanical properties. Studies on the bamboo cell wall have focussed mainly on fully elongated and mature fibres. The main aim of this study was to describe the ultrastructure of primary and secondary cell walls in culm tissues of Dendrocalamus asper at different stages of development. METHODS: The development of fibre and parenchyma tissues was classified into four stages based on light microscopy observations made in tissues from juvenile plants. The stages were used as a basis for transmission electron microscopy study on the ultrastructure of the cell wall during the process of primary and early secondary cell wall formation. Macerations and phloroglucinol-HCl staining were employed to investigate fibre cell elongation and fibre cell wall lignification, respectively. KEY RESULTS: The observations indicated that the primary wall is formed by the deposition of two distinct layers during the elongation of the internode and that secondary wall synthesis may begin before the complete cessation of internode and fibre elongation. Elongation was followed by a maturation phase characterized by the deposition of multiple secondary wall layers, which varied in number according to the cell type, location in the culm tissue and stage of shoot development. Lignification of fibre cell walls started at the period prior to the cessation of internode elongation. CONCLUSIONS: The structure of the primary cell wall was comprised of two layers. The fibre secondary cell wall began to be laid down while the cells were still undergoing some elongation, suggesting that it may act to cause the slow-down and eventual cessation of cell elongation.  相似文献

Tracks and trackways of theropod dinosaurs (Grallator footprints) are abundant in the Late Triassic lake sediments of East Greenland. For this study we selected a rather diffuse theropod track preserved on the upper surface of a red heterolithic mudrock, and a better preserved track seen on the upper surface of a greyish mudrock. In order to examine undertracks and other subsurface deformation structures, both slabs were sectioned vertically at closely-spaced intervals, perpendicular to the length of the axis of the impression of digit III. Each section was subsequently polished and internal structures revealed. The digit impressions of both tracks were associated with well-defined undertracks which were cut by deep and narrow claw imprints at the distal end of the digit impressions. Marginal ridges at the tracking surfaces were typically associated with subsurface marginal folds. The marginal ridges were asymmetrically developed suggesting an outward movement of the proximal part of the foot, probably during the kick-off; this is in contrast to what is observed in tracks from Lower Jurassic theropods. The study shows that cross-sections through dinosaur tracks display large structural variation and it is suggested that some disturbed layers in continental deposits could be the result of trampling by vertebrates.  相似文献

The behaviour of fossil organisms can typically be inferred only indirectly, but rare fossil finds can provide surprising insights. Here, we report from the Eocene Messel Pit Fossil Site between Darmstadt and Frankfurt, Germany numerous pairs of the fossil carettochelyid turtle Allaeochelys crassesculpta that represent for the first time among fossil vertebrates couples that perished during copulation. Females of this taxon can be distinguished from males by their relatively shorter tails and development of plastral kinesis. The preservation of mating pairs has important taphonomic implications for the Messel Pit Fossil Site, as it is unlikely that the turtles would mate in poisonous surface waters. Instead, the turtles initiated copulation in habitable surface waters, but perished when their skin absorbed poisons while sinking into toxic layers. The mating pairs from Messel are therefore more consistent with a stratified, volcanic maar lake with inhabitable surface waters and a deadly abyss.  相似文献

Several applications in tissue engineering require transplantation of cells embedded in appropriate biomaterial scaffolds. Such structures may consist of 3D non-woven fibrous materials whereas little is known about the impact of mesh size, pore architecture and fibre morphology on cellular behavior. In this study, we have developed polyvinylidene fluoride (PVDF) non-woven scaffolds with round, trilobal, or snowflake fibre cross section and different fibre crimp patterns (10, 16, or 28 needles per inch). Human mesenchymal stromal cells (MSCs) from adipose tissue were seeded in parallel on these scaffolds and their growth was compared. Initial cell adhesion during the seeding procedure was higher on non-wovens with round fibres than on those with snowflake or trilobal cross sections. All PVDF non-woven fabrics facilitated cell growth over a time course of 15 days. Interestingly, proliferation was significantly higher on non-wovens with round or trilobal fibres as compared to those with snowflake profile. Furthermore, proliferation increased in a wider, less dense network. Scanning electron microscopy (SEM) revealed that the MSCs aligned along the fibres and formed cellular layers spanning over the pores. 3D PVDF non-woven scaffolds support growth of MSCs, however fibre morphology and mesh size are relevant: proliferation is enhanced by round fibre cross sections and in rather wide-meshed scaffolds.  相似文献

采用光学显微镜、扫描电镜和透射电镜,对松江鲈鱼(Trachidermus fasciatus)成体皮肤的显微和亚显微结构进行了观察。结果表明,松江鲈鱼体表不同部位皮肤的厚薄不一,但基本结构相似。皮肤由表皮和真皮层构成。松江鲈鱼的皮肤裸露无鳞,表皮层较薄,由约4~8层细胞构成,主要由复层上皮细胞和黏液细胞及基底细胞组成。表层细胞呈扁平、多边形,细胞之间主要靠桥粒紧密连接,连接处形成增厚的边缘嵴状突起。表皮细胞游离面向内凹陷,表面形成指纹状微嵴。黏液细胞呈圆形或卵圆形,散布在上皮细胞之间。黏液细胞内的黏原颗粒具有椭圆颗粒状、均匀致密的块状和疏松丝状3种不同形态。真皮通过基膜与表皮相连,由稀疏层和致密层构成。真皮结缔组织在腹部较厚而在其他部位较薄。表皮与真皮连接处有色素层,头部、背部、尾柄和体侧皮肤色素细胞分布多,色素层明显,而腹部和颏部皮肤缺少色素。松江鲈鱼黄河群体真皮层中有角质棘状突起,而滦河群体则无。头部、体侧和尾柄处皮肤上还分布有侧线孔和表面神经丘等感觉器官。  相似文献

We have previously reported that the fragility of skin, tendon and bone from the oim mouse is related to a significant reduction in the intermolecular cross-linking. The oim mutation is unlikely to affect the efficacy of the lysyl oxidase, suggesting that the defect is in the molecule and fibre. We have therefore investigated the integrity of both the oim collagen molecules and the fibre by differential scanning calorimetry.The denaturation temperature of the oim molecule in solution and the fibre from tail tendon were found to be higher than the wild-type by 2.6deg.C and 1.9deg.C, respectively. With the loss of the alpha2 chain, the hydroxyproline content of the homotrimer is higher than the heterotrimer, which may account for the increase.There is a small decrease in the enthalpy of the oim fibres but it is not significant, suggesting that the amount of disorder of the triple-helical molecules and of the fibres is small and involves only a small part of the total bond energy holding the helical structure together. The difference in denaturation temperature of the skin collagen molecules (t(m)) and fibres (t(d)) is significantly lower for the oim tissues, 19.9deg.C against 23.1deg.C, indicating reduced molecular interactions and hence packing of the molecules in the fibre. Computation of the volume fraction of the water revealed that the interaxial separation of the oim fibres was indeed greater, increasing from 19.6A to 21.0A. This difference of 1.4A, equivalent to a C-C bond, would certainly decrease the ability of the telopeptide aldehyde to interact with the epsilon -amino group from an adjacent molecule and form a cross-link. We suggest, therefore, that the reduction of the cross-linking is due to increased water content of the fibre rather than a distortion of the molecular structure.The higher hydrophobicity of the alpha2 chain appears to play a role in the stabilisation of heterotrimeric type I collagen, possibly by increasing the hydrophobic interactions between the heterotrimeric molecules, thereby reducing the water content and increasing the binding of the molecules in the fibre.  相似文献

The soft tissue preserved in the holotype (IVPP V12705) of Jeholopterus ningchengensis from the Daohugou Bed (Late Jurassic or Early Cretaceous) of China is described in detail. The plagiopatagium can be divided into the distal, comparatively more rigid actinopagatium and a proximal, more tensile tenopatagium. The actinopatagium extends from the wing finger to the articulation between the humerus and the forearm, and shows the presence of at least three layers containing actinofibrils. In each layer, the actinofibrils are parallel to subparallel, but this direction diverges from layer to layer. When distinct layers of actinofibrils are superimposed (owing to taphonomic compression), a reticular pattern is generated. The presence of layers with differently oriented actinofibrils is widespread in this pterosaur. A well-developed integumental covering formed by fibres (here named pycnofibres) that are thicker than the actinofibrils is present. Ungual sheaths that extend the length of the pedal and manual claws of this taxon are also observed. Although the understanding of the mechanical properties of the wing membrane is hampered by the lack of knowledge regarding the composition of the actinofibrils, the configuration observed in Jeholopterus might have allowed subtle changes in the membrane tension during flight, resulting in more control of flight movements and the organization of the wing membrane when the animal was at rest.  相似文献

The free-living marine nematode Theristus (Penzancia) anoxybioticus n. sp. is described from specimens collected in muddy sediment at 10-12 m water depth in the northern Kattegat, Denmark, where the benthic environment is influenced by methane seepages. Mean body length of the male is 1,121 μm and of the female 1,159 μm. Theristus (Penzancia) anoxybioticus n. sp. has one crown of 10 cephalic setae and a clavate tail tip without setae. The three caudal gland cells are prominent. The intestinal lumen is hexaradiate in cross section and the lining is devoid of microvilli. Reproductive adults have so far only been found in the uppermost centimeter of sediment, and their presence is restricted to April and May. Juveniles are found in deep anoxic sediment layers during other months of the year.  相似文献

The evolution of the locomotor apparatus in vertebrates is marked by major reorganizations in trunk's musculature. The hypothesized functions of mammalian back muscles in the literature are discussed under consideration of the distribution and proportion of oxidative, type‐I‐fibres, oxidative‐glycolytic, type‐IIa‐fibres and glycolytic, type‐IIb‐fibres in paravertebral muscles of a small mammal. The fibre type distribution was examined from a complete series of histological sections maintaining topographical relationships between the muscles as well as within the muscle, in order to establish the overall distribution pattern. The deep and short muscles showed the highest percentage of oxidative fibres. The larger, superficial paravertebral muscles contained the highest percentage of glycolytic fibres. Two muscles were intermediate in their proportion of fibre types. All epaxial muscles together can be interpreted as an antigravity muscle–complex counteracting enduringly against the rebound tendency caused by gravitation, comparable with antigravity muscles in limbs. A gradient from deep to superficial, or a clear regionalization of oxidative muscle fibres in central deep regions around a large intramuscular tendon was found in the m. spinalis and the m. quadratus lumborum, respectively. Concepts of the function of human back muscles as those of A. Bergmark (1989: Acta Orthop. Scand. 230 , 1) or S.G.T. Gibbons & M.J. Comerford (2001: Orthop. Division Rev. March/April, 21) were exposed to be more general within mammals. Functional specializations of different muscles and muscle parts are discussed under the consideration of evolutionary reorganization of the paravertebral musculature in tetrapods. Along the cranio‐caudal axis, the percentage of oxidative fibres was decreased in caudal direction within the same muscles, whereas the proportion of glycolytic fibres was increased. Therefore, classifications of muscles as ‘glycolytic’ or ‘oxidative’ based on biopsies or analyses of single cross‐sections may result in wrong interpretations. Changes in the proportions of the fibre type distribution pattern were mostly due to oxidative and glycolytic fibre types, whereas the percentage of oxidative‐glycolytic fibres had only minor influence. A significant positive correlation between the cross‐sectional area of the single fibre and its percentage in the area investigated were observed for oxidative fibres, whereby the size was positive correlated to the proportion of the oxidative fibres.  相似文献
