The activities of catalase, peroxidase, indoleacetic acid (IAA) oxidase and peroxide levels in cucumber plants during and after chilling were determined. During 96 hours at 5 C and 85% relative humidity, catalase activity declined, IAA oxidase activity increased, and peroxide concentrations increased. Peroxidase activity was not affected by chilling. When chilled plants were returned to 25 C to recover, enzyme activities and peroxide concentration were restored to their prechilling levels. The increase in peroxide and IAA oxidase activity may inactivate or destroy IAA and thus retard growth.  相似文献

Peroxidase was extracted from the apple (Delicious) fruit and the apple stalk during the various stages of ripening. Activities of peroxidase and IAA oxidase of fruits are different.The activities of peroxidase and IAA oxidase in the apple stalk are much higher than that in the apple fruits. Gel electrophoresis revealed that the number of isozymic bands of peroxidase in the stalk (7–9 bands) were more than that in the fruit (6 bands). It is inferred that peroxidase in apple stalk might play a more important role in IAA and some other inhibitors oxidized before then went into the fruits.  相似文献

Vinca rosea L. crown-gall tumor callus tissue cultures treated with N-benzyl-N methyl propargylamine (pargyline) exhibited a decrease in the level of endogenous indoleacetic acid from 0.42 μg/mg of protein to less than 0.30 μg/mg of protein. A simultaneous decrease in the specific activity of mitochondrial amine oxidase from 3000 units to less than 250 units at 1.0 μM, 0.01 mM, 0.1 mM and 1.0 mM pargyline, suggested a relationship between amine oxidase function and indoleacetic acid synthesis. Tryptamine incorporation into indoleacetic acid was also decreased at these concentrations. Pargyline inhibited tumor callus growth significantly (based on fresh weight measurements) at the highest concentration, 1.0mM. These data support the hypothesis of a coordinate metabolic system linking mitochondrial amine oxidase activity and indole acetic acid synthesis. Inhibitory action of pargyline on the enzyme is reflected in reduced indoleacetic acid levels and, ultimately, in reduced callus growth rates.  相似文献

对光叶楮扦插生根过程中吲哚乙酸氧化酶(IAAO)、多酚氧化酶(PPO)、过氧化物酶(POD)3种酶进行了动态跟踪分析。结果表明:IAAO活性在扦插初期逐渐上升,第10d上升到高峰,之后下降再上升,第30d达到新高峰,然后迅速下降;前25d POD活性变化规律与IAAO相似,但30d以后活性一直上升;PPO活性在扦插前期缓慢上升,第20d上升到了最高点,此后变化不大。还研究了IAAO、PPO、POD与不定根的发生和发展关系,认为光叶楮扦插生根可分为愈伤组织形成期、根诱导期和根的伸长期3个阶段,愈伤组织形成期3种酶活性都呈上升趋势,根诱导期IAAO和POD的活性达到高峰;而根伸长期IAAO和POD活性下降,PPO活性上升。  相似文献

Aspen, Hami melon, soybean and tobacco calli were incubated in Miller's solid medium supplemented with IAA 4 mg/L and kinetin 0.5 mg/L. Activities of IAA-oxidase and peroxidase were determined at 0,5,10,15 and 25 days after incubation. The activities of IAA oxidase and peroxidase of Hami melon callus were found to be the highest and the tobacco was the lowest among the four different kinds of calli, both enzymes showed their peak value in 10 days after incubation. There were no change in pattern of peroxidase isoenzyme among the four kinds of calli during the incubation, but the activities of IAA-oxidase and its isoenzyme of Hami melon at bands A6, A7 and A8 were 3 to 17 folds higher than that of corresponding isoenzyme of other three kinds of calli.  相似文献

IAA oxidase/peroxidase active fractions were separated using concanavalin A-sepharose into glycoprotein and non-glycoprotein fractions. No IAA oxidase peak was separable from peroxidase activity. The development of these fractions were followed in pea roots over a four day period. Initially the nonglycosylated IAA oxidase/peroxidase was the dominant fraction with the IAA oxidase/peroxidase ratio remaining close to unity. The glycosylated fraction developed later with IAA oxidase the dominant activity. Differential carbohydration of IAA oxidase/peroxidase fractions may determine the intracellular sites of activity of this molecular species.  相似文献

Strips of tissue containing the germ aleurone layer were removedfrom dry, harvest-ripe grains of barley (Hordeum vulgare L.)and incubated in buffered solutions of phenolic compounds, withand without the addition of hydrogen peroxide. Peroxidase ando-diphenol oxidase activity were found in the material releasedinto the incubation medium, and in the cytoplasm of the germaleurone cells. Peroxidase activity was located in the cellwalls and appeared to be high in the region where the germ aleuronecovering the embryonic axis merges into that which adheres tothe scutellum i.e. the region in which a row of germ aleuronecells becomes lignified following germination. Monophenol oxidaseactivity was not detected in the released enzymes or in theintact tissue. Although hydroquinone was oxidized in the cytoplasmof the germ aleurone tissue, unequivocal evidence of the presenceof laccase was not obtained. Seselin, a natural coumarin derivative isolated from citrus roots, inhibited radicle growth in seedlings of cucumber (Cucumis sativa), lettuce (Lactuca sativum), radish (Raphanus sativus), and wheat (Triticum aestivum) grown in the dark. Coumarin similarly inhibited radicle growth of cucumber seedlings. Growth retardation of the cucumber radicles was accompanied by an increased activity of peroxidase and indole-3-acetic acid oxidase. Both compounds antagonized indole-3-acetic acid-induced growth of wheat coleoptiles, whereas coumarin was much less effective than seselin in antagonizing gibberellic acid-induced release of reducing sugars from barley endosperm. It is suggested that seselin plays an important role in the regulation of root growth, and that it is the indole-3-acetic acid oxidase cofactor previously detected in citrus roots.  相似文献

Growth and peroxidase activity of roots and stems of lentil seedlings were compared after treatment with Amo-1618, alone or in combination with gibberellic acid (GA) at varying concentrations. The peroxidase enhancement in Amo-1618 treated stems could not be attributed to a decrease in the gibberellin content since GA alone had no effect on this enzyme. In other experiments, AMO, at low concentrations, was able to induce α-amylase production in barley aleurone layers; the lag period needed for this induction, was longer than for GA. These facts seem to indicate that some growth retardants might act at least in some cases by mechanisms other than inhibition of gibberellin biosynthesis and reversal of GA action.  相似文献

A study was conducted to determine the activity of the 3-methylindole (3MI)-forming enzyme in Lactobacillus sp. strain 11201. Cells were incubated anaerobically with 17 different indolic and aromatic compounds. Indoleacetic acid (IAA), 5-hydroxyindoleacetic acid, 5-methoxy-3-indoleacetic acid, indole-3-pyruvate, or indole-3-propionic acid induced 3MI-forming activity. The highest total enzyme activity induced by IAA was observed in cells incubated with an initial concentration of 1.14 mM IAA. Peak activity of the 3MI-forming enzyme occurred 4 h after bacteria were incubated with either 0.114 or 1.14 mM IAA. Enzyme activity peaked earlier (2 h) and disappeared more rapidly at 5.7 mM IAA than at other concentrations of IAA. The effects of IAA and 3MI on the growth of Lactobacillus sp. strain 11201 and formation of 3MI from IAA also were determined. Bacterial growth and 3MI formation from IAA were reduced in medium containing exogenous 3MI. The growth depression observed in medium containing 5.7 mM IAA appears to be due to the toxicity of 3MI rather than IAA. The formation of 3MI in this ruminal Lactobacillus sp. is mediated by an inducible enzyme, and as 3MI accumulates, bacterial growth and rates of 3MI formation from IAA are reduced.  相似文献

The effect of IAA on growth, dinitrogen fixation, and heterocystsfrequency of Anabaena PCC 7119 and Nodularia sp. have been investigated.Concentrations of IAA ranging from 10–10 to 10–4M did not change the growth of Anabaena PCC 7119. Concentrationshigher than 10–4 M were inhibitory. Similar results werefound in Nodularia sp. although in this case the inhibitoryeffect appeared with 10–5M of IAA.
