The incidence of potato virus Y (PVY) infection was assessed in samples of potato tubers, cv. Record, taken from Scottish seed stocks and English ware crops grown from some of these seed stocks. PVY was readily detected by ELISA of tuber sprouts. PVY-infected tubers were found in 10 seed stocks of 84 tested. The mean level of virus infection was 0.23%, 0.76% and 0.56% in Super Elite, Elite and AA stocks respectively. In 46 commercial ware crops grown from some of these seed stocks, a substantial proportion of the harvested tubers in all but one of the crops were infected with PVY, the mean percentage of infected tubers was 58.5%. Ware crops grown from seven seed stocks in which PVY had been detected (mean 6.2% infection in seed) contained a mean of 70% infected tubers, compared with 56% infection in crops grown from 39 stocks in which PVY was not detected in the seed tubers. The predominant PVY strain detected in the ware crops was the veinal necrosis strain (PVYvn).  相似文献

Overwintering Colorado potato beetles (Leptinotarsa decemlineata (Say)) were concentrated primarily within woody borders, and mortality was lower in borders than in potato fields. After overwintering, only 15–44% of live beetles were in the potato fields. In experiments with small plots, colonization of fields from woody borders was reduced ∼60% by a trap crop, either treated with adulticide or with beetles collected daily. Such trap crops, or simply pitfall traps to prevent colonization from woody borders, could significantly reduce early-season adult numbers and subsequent larval populations. However, success is dependent on the local densities of overwintered beetles, and the prevalence of arrestment behavior in the case of trap crops.  相似文献

Sweet potato virus disease (SPVD) was common (25–30% average incidences), and farmers recognised it as an important disease, in sweet potato crops in southern Mpigi, Masaka and Rakai Districts in Uganda, but SPVD was rare in Soroti and Tororo Districts. Whiteflies, which are the vector of sweet potato chlorotic stunt crinivirus (SPCSV) a component cause of SPVD, were correspondingly common on sweet potato crops in Mpigi and rare on crops in Tororo. However, aphids, which are the vectors of sweet potato feathery mottle potyvirus (SPFMV), the other component cause of SPVD, were not found colonising sweet potato crops, and itinerant alate aphids may be the means of transmission. Different sweet potato cultivars were predominant in the different districts surveyed and four local cultivars obtained from Kanoni in S. Mpigi, where whiteflies and SPVD were common, were more resistant to SPVD than four cultivars from Busia in Tororo District, where whiteflies and SPVD were rare. However, nationally released cultivars were even more resistant than the local cultivars from Kanoni. Yield results and interviews with farmers indicated that farmers in S. Mpigi were making compromises in their choice of cultivars to grow, some key factors being SPVD susceptibility, and the yield, taste, and marketability, duration of harvest and in-ground storability of the storage roots. These compromises need to be included in an assessment of yield losses attributable to SPVD.  相似文献

Black dot and other fungal blemishing diseases were assessed on tubers from crops in eastern England in 1987 and 1988, and in 1989 and 1990 from crops throughout Great Britain. Black dot was found on tubers in crops from all areas of the country, and was most common in those from eastern and southern England. The disease was scarce on most Scottish crops, but occasionally some were severely affected. In 1988 there was no consistent relationship between the amounts of disease on the seed and subsequent ware crop. On average, the disease was more prevalent in irrigated crops and where the interval between successive potato crops was short. In 1990 treating seed tubers with fungicide had no effect on subsequent levels of disease. Few crops were unaffected by silver scurf, and it was most common in crops from southern and eastern England. In the national survey common scab was the most prevalent disease, black scurf was less common and skin spot and powdery scab uncommon except in some Scottish crops.  相似文献

For rapid and simultaneous detection of transgenic elements in genetically modified (GM) food crops, we explored DNA array technology. Forty-four oligonucleotide 23-to 31-mers were selected to use in an array on the basis of melting temperature and sequence specificity. Selected oligonucleotides consisted of DNA fragments corresponding to structural and regulatory elements and selectable markers used in developing transgenic crops, such as potato. Other oligonucleotides represented endogenous genes from potato to serve as positive controls and from heterologous crops, such as soybean and canola, to serve as negative controls. Amino-terminated oligonucleotides were hand-spotted on activated nylon membrane with a commercial spotting device. Target DNA was isolated from foliage of transgenic and nontransgenic crops, including potato, and labeled with digoxigenin-dUTP by random priming following restriction digestion to reduce DNA fragment size. Hybridization signals were visualized by an alkaline phosphatase anti-DIG-Fab conjugate and the chemiluminescent substrate, CDP-star. We detected the presence or absence of transgenic elements in transgenic and nontransgenic potato samples. Preliminary studies demonstrated that more specific and sensitive hybridization signals were generated from an oligonucleotide probe array than from a PCR product array. We anticipate that oligonucleotide probe arrays will be useful for regulatory monitoring of transgenic events.  相似文献

Counts of aphides infesting potato crops in north-west Derbyshire at altitudes between 340 and 1360 ft. were made during the summer of 1945. Mysus persicae was abundant on almost all crops, and peak figures on some reached over 1500 per 100 lower leaves. In many fields two peaks were noted, one towards the end of July and the second in mid-September, and in some fields there were still heavy infestations when the haulms died. The number of aphides varied considerably from field to field; sheltered fields were the least infested. In this area in 1945 there was no evidence to support the contention that the altitude and aspect of the field have any influence on the intensity of infestation. Tuber samples from six fields showed that there was a considerable spread of leaf roll.  相似文献

The natural defence system of plants often involves inhibitors of digestive enzymes of their pests. Modem and environmental-friendly methods try to increase this plant resistance by expressing heterologous protease inhibitors in crops. Here we report the effects of expressing a gene from desert locust (Schistocerca gregaria) encoding two serine protease inhibitors in potato on Colorado potato beetle (Leptinotarsa decemlineata) larvae. The gene encoding both peptides on a single chain was used for Agrobacterium-mediated transformation of potato plants. The presence of the active inhibitor protein in the leaves was verified. The feeding bioassays in the laboratory showed that despite the low level of the peptide in leaves, CPB larvae on transgenic plants have grown slightly but significantly more slowly than those on control potato plants. The results support the notion that expression of multifunctional proteinase inhibitors of insect origin in plants might be a good strategy to improve insect resistance.  相似文献

The concept of using a range of Solanaceae potato clones as trap crops for potato cyst nematode (PCN) management was investigated. A series of field trials were undertaken from 1999 to 2002 that evaluated 10 clones of either wild Solanum potato species, breeder’s hybrid lines or commercial cultivars. All had high resistance to all known PCN pathotypes (both Globodera rostochiensis and Globodera pallida) and the ability to stimulate high levels of PCN hatch. Investigations showed potential for the development of some clones as a means of reducing high PCN field population levels and for use by organic potato producers.  相似文献

Field experiments were carried out in eastern Scotland in 1976-78 to test the ability of granular insecticides, applied to soil at planting, and of insecticide sprays applied to the foliage, to control aphids and spread of potato leafroll virus (PLRV) in potatoes. The three years provided contrasting opportunities for virus spread. In 1976, the main vector of PLRV, Myzus persicae, arrived in early June and multiplied rapidly in untreated plots, and PLRV spread extensively. In 1977, M. persicae arrived 4–6 wk later than in 1976 and most spread of PLRV, which was less than in 1976, occurred after the end of July. In 1978, few M. persicae were recorded but the potato aphid, Macrosiphum euphorbiae, arrived early and very large populations developed in untreated plots. However, little spread of PLRV occurred in 1978, supporting other evidence that M. euphorbiae is an inefficient vector of PLRV in field conditions. In each year, granular insecticides decreased PLRV spread to a quarter or less of that in control plots. Thiofanox gave somewhat better and longer-lasting control of aphid populations than disulfoton, especially of M. persicae, but did not give greater control of PLRV spread. Application of three (1976) or five (1977) sprays of demeton-S-methyl to plots treated with granular insecticides further improved the control of M. euphorbiae but had less or no effect on M. persicae, especially where organophosphorus resistant aphids (R1 strain) were found. These supplementary sprays of insecticide did not further improve the control of PLRV but, in 1978, four sprays of demephion or pirimicarb to plots not treated with granular insecticide decreased PLRV spread. These data, together with previous findings, indicate that the amount of virus spread depends on the date of arrival and rate of multiplication of M. persicae in relation to the timing and effectiveness of removal of PLRV sources in crops. It is concluded that in Scotland insecticide granules should be used routinely only in crops of the highest grade of seed potato. Their use for other grades need be considered only in years following mild winters, when aphids can be expected to enter crops earlier and in larger numbers.  相似文献

Following the molecular characterisation of functional disease resistance genes in recent years, methods to track and verify the integrity of multiple genes in varieties are needed for crop improvement through resistance stacking. Diagnostic resistance gene enrichment sequencing (dRenSeq) enables the high‐confidence identification and complete sequence validation of known functional resistance genes in crops. As demonstrated for tetraploid potato varieties, the methodology is more robust and cost‐effective in monitoring resistances than whole‐genome sequencing and can be used to appraise (trans) gene integrity efficiently. All currently known NB‐LRRs effective against viruses, nematodes and the late blight pathogen Phytophthora infestans can be tracked with dRenSeq in potato and hitherto unknown polymorphisms have been identified. The methodology provides a means to improve the speed and efficiency of future disease resistance breeding in crops by directing parental and progeny selection towards effective combinations of resistance genes.  相似文献

Summary Mitochondrial DNA variation was detected in potato plants (protoclones) regenerated from leaf mesophyll protoplasts. Two forms of variation were evident; (1) DNA sequence alterations within the high molecular weight mitochondrial chromosome and (2) the appearance of an additional low molecular weight mitochondrial DNA species. Variation in chloroplast DNA was not detected. The data suggests that protocloning can introduce molecular diversity into mitochondrial genomes and thereby assist in overcoming the cytoplasmic genetic uniformity prevalent in most major crops.  相似文献

In tobacco plants infected with mild strains of virus X , severe strains may arise as mutations which multiply locally.
Several strains of virus X gradually lost infectivity for potato on continued culture in other hosts such as tobacco.
Strains that are poorly infective or invasive for potato may take considerable time to move into the growing shoot from the tuber in detectable amounts.

Abstract 1 The attractiveness of pitfall traps baited with a synthetic host volatile attractant to colonizing adult Colorado potato beetle, Leptinotarsa decemlineata (Say) was evaluated in a field setting. 2 Significantly more postdiapause, colonizing adult L. decemlineata were captured in baited than unbaited pitfall traps. 3 The potential for this synthetic kairomone to enhance the efficacy of trap cropping as a management tool was evaluated by comparing conventionally managed plots with like‐sized plots bordered by either attractant‐treated trap crop or untreated trap crop. 4 More postdiapause, colonizing adults, egg masses and small larvae were present in attractant‐treated trap crops than in untreated trap crops. 5 There were no significant differences in egg mass and small larvae densities between plots bordered by attractant‐treated trap crops and conventionally managed plots, but there were significantly fewer large larvae and adult beetles in conventionally managed plots. 6 Plant canopy area of conventionally managed plots was significantly greater than in plots bordered by either type of trap crop. 7 Yields for conventionally managed plots and plots bordered by attractant‐treated trap crops did not differ, and less insecticide (44%) was applied to plots bordered by attractant‐treated trap crops.  相似文献

A study is presented of the Hospital de la Sangre account books in Seville at the Archivo Hispalense for the period 1546 to 1601, to verify purchases of potatoes (Solanum tuberosum) during that period. Potatoes were bought regularly in the Seville market from 1580 onwards, with the first record appearing in 1573 thus agreeing with Salaman’s conclusion that potatoes became established in Spain by about 1570. Purchases were confined almost entirely to December and January each year, lending weight to the hypothesis that these were short day adaptedS. tuberosum ssp.andigena, actually grown in Spain and forming tubers in the short days at the end of the year. If potatoes had been imported for direct consumption from South America shipping records indicate that they could have arrived in Seville at all times of the year. A listing of other fruits and vegetables bought in the Seville market shows a wide range of mostly mediterranean crops with some of Near Eastern origin and certain spices imported through Lisbon from the Ear East. Surprisingly, very few New World crops are mentioned.  相似文献

57 native potato tuber samples collected from different potato growing region of Iran, planted on single rows in Karaj College experimental station. Plant samples of each single row plus 9.25 Fresh foliage samples collected from fields under new introduced cultivars were tested for potato virus (PVM) infection during growing season. Also 78 weeds and field crops belonging to Solonacae and Leguminosae from or neighboring to potato field were tested. Results indicated that PVM was not found on any plant other than potatoes. PVM was detected on 16 samples of 57 old vars, Virus was not seen in any samples collected from fields under new varieties. Results show that PVM is limiting in this crop. PVM detecting is difficult using assay hosts. Best test plants were French bean var Red kidney, Showing pinpoint necrotic LL, also Datura metel and Nicotiana debneyi are useful for virus detection showing chlorotic local lesion. Also microprecipition and gel diffusion test can be used for virus detection but Elisa was the best method. PVM infected plant showed 11-19.5 percent yield decrease in 3 cultivars tested.  相似文献

Summary The development of common scab on potato crops growing in slightly acid to neutral soils has been shown to be greatly reduced by application of manganese sulphate to the soil at planting time. A general improvement in health and appearance of tubers was noted and increased yields of marketable tubers were obtained. The possibility that environmental factors known to influence the development of scab are only important in so far as they affect the solubility of soil manganese is discussed.  相似文献

In 1982 – 88, potatoes were grown in 2-, 4- and 6-course rotations with spring barley on a field infested with Globodera rostochiensis. Severity of stem canker and black scurf increased with increasing frequency of previous potato crops, and seed tuber treatment with tolclofos-methyl became less effective in controlling diseases. This suggested that previous crops had increased the amounts of soil-borne inoculum of Rhizoctonia solani. Oxamyl soil treatment increased stem canker in one year and decreased black scurf in four years. Seed tuber treatment with imazalil or prochloraz decreased stem base infection by Polyscytalum pustulans and skin spot and silver scurf on tubers. Black dot was prevalent on tubers in all years and was not affected by seed tuber treatment or previous cropping. Oxamyl increased black dot and common scab in five years and decreased % tuber dry matter in six years. Cysts of G. rostochiensis were found attached to Désirée but not to Maris Piper tubers in August. At harvest tubers of both cultivars were affected by superficial pitting and its severity was related to soil populations of G. rostochiensis at planting. This damage was controlled by oxamyl. It is suggested that the pitting developed from holes made in the tuber skin at larval invasion. In 1989, Désirée seed tubers and healthy mini tubers were planted in all plots and severity of stem canker and black scurf increased with increasing proximity of previous potato crops and with the number of previous crops. Black dot on stems and tubers was not affected by previous cropping but was much less severe in a plot that had not grown potatoes during the seven years of the experiment. The severity of common scab generally decreased as the number of preceding potato crops increased.  相似文献

Black scurf and stem canker of the potato was investigated in field trials on infected soil at Warburton, Cheshire, to study the effect of planting clean and contaminated seed, and the relation between the dates of planting and lifting and the contamination of the crop tubers.
Black scurf was prevalent on crops grown from clean seed but was more severe on crops raised from contaminated seed. Under conditions favourable for the disease the yields from clean and contaminated seed were satisfactory and were not significantly different. Young shoots of plants from contaminated seed were severely attacked and tuber formation was checked; these results were not obtained on clean seed plots.
More black scurf occurred on late-dug crops; on early-dug plots the disease, although almost absent in 1941, was prevalent in 1942. Late planting failed to reduce appreciably the amount of black scurf.
Variety trials included Arran Banner, Kerr's Pink (two vigorous varieties), King Edward and Majestic (two less vigorous varieties). All were heavily infected and each recovered well from an attack on the young shoots.
Inoculation of the seed at planting time did not affect the results; in all trials, misses and wilted shoots caused by Corticium were rare and there was no relation between the yield and the amount of black scurf on the crop tubers.
The results indicate that the disease causes little, if any, loss under farm conditions.  相似文献

The flight take-off frequency of adult Colorado potato beetles, Leptinotarsa decemlineata (Say), from potato plants, Solanum tuberosum L. 'Red Pontiac' at the bloom stage of development was 2.2-2.5-fold that of Colorado potato beetle from plants at the vegetative stage. Tests were conducted in a flight chamber over a period of 3 h. Prefeeding Colorado potato beetles for 48 h on potato plants at the bloom or at the vegetative stage before placing them into the flight chamber resulted in the same significantly higher flight take-off frequency from potato plants at the bloom stage than from plants at the vegetative stage. These results demonstrate that the factor in potato plants in bloom that stimulates the flight take-off of the Colorado potato beetle is independent of the feeding history of the beetles and begins acting only when the beetles are in the presence of the plant. According to these results, the dispersal of adult Colorado potato beetles from potato fields in bloom to younger potato fields with plants at the vegetative stage, previously reported in the literature, is at least partly explained by the effect of plant phenology on the frequency of flight take-off. Results confirm the value of planting potato fields of similar phenology over as wide an area as possible to reduce Colorado potato beetle dispersal between fields. Results also imply that staggering the planting dates of conventional potato refuge areas near Colorado potato beetle transgenic or conventionally resistant potato fields is a sound management practice, because it promotes the movement of wild beetles over to the adjacent younger resistant crops.  相似文献
