The effect of photosynthetic photon flux density (PPFD) on carboxylationefficiency, estimated as the initial slope (IS) of net CO2 assimilationrate versus intercellular CO2 partial pressure response curve,as well as on ribulose-1, 5-bisphosphate carboxylase (Rubisco)activation was measured in Trifolium subterraneum L. leavesunder field conditions. The relationship between IS and PPFDfits a logarithmic curve. Rubisco activation accounts for theIS increase only up to a PPFD of 550 µmol photons m-2s-1. Further IS increase, between 550 and 1000 µmol photonsm-2 s-1, could be related to a higher ribulose fcwphosphate(RuBP) availability. The slow, but sustained IS increase above1000 µmol photons m-2 s-1 could be explained by the mesophyllCO2 diffusion barriers associated with the high chlorophylland protein content in field developed leaves. The nature of the lack of oxygen inhibition of C3-photosynthesisat low temperature was investigated in white clover (Trifoliumrepens L.). Detached leaves were brought to steady-state photosynthesisin air (34 Pa p(CO2), 21 kPa p(O2), balance N2) at temperaturesof 20°C and 8°C, respectively. Net photosynthesis, ribulose1,5-bisphosphate (RuBP) and ATP contents, and ribulose 1,5-bisphosphatecarboxylase/oxygenase (RuBPCO) activities were followed beforeand after changing to 2·0 kPa p(O2). At 20°C, lowering p(O2) increased net photosynthesis by37%. This increase corresponded closely with the increase expectedfrom the effect on the kinetic properties of RuBPCO. Conversely,at 8°C net photosynthesis rapidly decreased following adecrease in p(O2) and then increased again reaching a steady-statelevel which was only 7% higher than at 21 kPa p(O2). The steady-staterates of RuBP and associated ATP consumption were both estimatedto have decreased. ATP and RuBP contents decreased by 18% and33% respectively, immediately after the change in p(O2) suggestingthat RuBP regeneration was reduced at low p(O2) due to reducedphotophosphorylation. Subsequently, RuBP content increased again.Steady-state RuBP content at 2·0 kPa p(O2) was 24% higherthan at 21 kPa p(O2). RuBPCO activity decreased by 22%, indicatingcontrol of steady-state RuBP consumption by RuBPCO activity. It is suggested that lack of oxygen inhibition of photosynthesisat low temperature is due to decreased photophosphorylationat low temperature and low p(O2). This may be due to assimilateaccumulation within the chloroplasts. Decreased photophosphorylationseems to decrease RuBP synthesis and RuBPCO activity, possiblydue to an acidification of the chloroplast stroma. Bot. 38: 1937–1948. Associations between photosynthesis, the activation state ofRuBP carboxylase and the contents of photosynthetic intermediateswere compared in soybean and wheat leaves before and after exposureto photoinhibitory treatments in the presence of atmosphericO2. Exposing attached leaves to a supra-saturating irradiance(3 800 µmol quanta m– 2 s–1) for 2 h in CO2-freeair decreased carboxylation efficiency and the light-saturatedphotosynthetic rate in air by approximately 50%. Exposure tothe photoinhibitory treatment for periods in excess of 2 h didnot cause a further decrease of photosynthesis in soybean. Althoughphotosynthesis was reduced, the initial and total (fully-activated)activities of ribulose 1,5-bisphosphate carboxylase (RuBPCase)in leaf extracts were unaltered in each species by the photoinhibitorytreatment. This was true for leaves sampled under both air andat a rate-limiting intercellular CO2 partial pressure (Ci) of75 µPa Pa–1. The contents of ribulose l,5-bisphosphate(RuBP) and 3-phosphoglyceric acid (3-PGA) were reduced by thephotoinhibitory treatment in soybean leaves sampled in air andat a rate-limiting Ci, although the RuBP/3-PGA ratio was unaffected.The relative reduction of RuBP content in soybean leaves atrate-limiting C1 was similar to the corresponding reductionof carboxylation efficiency. For wheat,the relative reductionof RuBP content at rate-limiting Ci (–19%) caused by thephotoinhibitory treatment was considerably less than the correspondingdecrease of carboxylation efficiency (–49%).The RuBP/3-PGAratio of wheat was also increased significantly by the photoinhibitorytreatment The significance of these observations to the regulationof CO2-limited photosynthesis in leaves experiencing photoinhibitionunder atmospheric oxygen is discussed. Consideration is alsogiven to the previous contention that contemporary measurementsof initial activity in crude extracts may provide a spuriousindication of the amount of the enzyme-CO2-Mg2 + form of RuBPcarboxylase present in the leaf. Time-courses of 14CO2-fixation and of enzyme activities involvedin photorespiration and photosynthesis were determined duringthe life span of cotyledons from sunflower seedlings (Helianthusannuus L.). Glycolate formation in vivo was estimated from theresults of combined labelling and inhibitor experiments. NADPH-glyceraldehyde-3-phosphatedehydrogenase, NADPH-glyoxylate reductase and chlorophyll werewell correlated with the time-course of 14CO2-fixation (photosynthesis).There was, however, a considerable discrepancy between the developmentalsequence of photosynthesis and that of both ribulose-l,5-bisphosphatecarboxylase and glycolate oxidase. Furthermore, time-coursesof glycolate oxidase activity in vitro and of glycolate formationin vivo differed significantly. Therefore, the use of glycolateoxidase as a marker for the activity of photorespiration ingreening sunflower cotyledons may be questionable. Results from14CO2-labelling experiments with cotyledons treated with theglycolate oxidase inhibitor 2-hydroxy butynoic acid suggestthat glycolate formation relative to CO2-fixation is reducedin senescent cotyledons. Detached leaves of white clover (Trifolium repens L.) were keptfor 1 h under various conditions of temperature, oxygen concentrationand light intensity. Rates of photosynthesis were measured whereappropriate and then ribulosebisphosphate carboxylase oxygenase(RuBPCO) was extracted rapidly and its initial activity measuredimmediately. The extracted activity increased with increased intensity ofillumination of the leaves. Where leaves were pretreated atlow light intensity, the lower the temperature of the leavesthe higher the extracted activity of RuBPCO. At high light intensitytemperature did not affect the activity of subsequently extractedRuBPCO but the light intensity which was necessary for maximumactivity increased with temperature. Activity of RuBPCO fromleaves pretreated in the dark was least when CO2 was low andtemperature high. Leaves, pretreated at low temperatures andhigh light intensity in 20% O2, yielded higher activity in extractsthan leaves pretreated under similar conditions but in 2% O2.A relatively weak temperature response of photosynthesis atlow irradiances was associated with a decrease in extractableRuBPCO activity with increasing temperature. A strong temperaturedependence of the oxygen inhibition of photosynthesis was associatedwith lower extractable RuBPCO activity in leaves pretreatedat low oxygen concentration at low temperatures. With leavesfrom plants grown at low temperatures prior to treatment ofleaves, oxygen inhibition of photosynthesis was less temperaturedependent and activity of RuBPCO in extracts was not decreasedby low O2 at low temperatures. Differences in the activationof RuBPCO appear to influence photosynthesis and account foran absence of oxygen inhibition of photosynthesis at low temperaturesin plants grown in warm conditions. The effects of nitrate supply on the composition (cell numbers,protein and chlorophyll contents) of flag leaves of winter wheatgrown with two amounts of N fertilizer and of spring wheat grownin the glasshouse under controlled nitrate supply are describedand related to photosynthesis. Nitrogen deficiency decreasedthe size of leaves, mainly by reducing cell number and, to asmaller extent, by decreasing cell volume. Protein content perunit leaf area, per cell and per unit cell volume was largerwith abundant N. Total soluble protein, ribulose bisphosphatecarboxylase-oxygenase (RuBPc-o) protein and chlorophyll changedin proportion irrespective of nitrogen supply and leaf age.Photosynthesis per unit area of flag leaf and carboxylationefficiency in both winter and spring wheat were proportionalto the amount of total soluble protein up to 7.0 g m–2and to the amount of RuBPc-o protein up to 4.0 g m–2.However, photosynthesis did not increase in proportion to theamount of total soluble or RuBPc-o protein above these amounts.In young leaves with a high protein content the measured ratesof photosynthesis were lower than expected from the amount andactivity of RuBPc-o. Carboxylation per unit of RuBPc-o protein,measured in vitro, was slightly greater in N-deficient leavesof winter wheat but not of spring wheat. RuBPc-o activity perunit of RuBPc-o protein was similar in winter and spring wheatleaves and remained approximately constant with age, but increasedin leaves showing advanced senescence. RuBPc-o protein fromN-deficient leaves migrated faster on polyacrylamide gels thanprotein from leaves with high N content. Regulation of the rateof photosynthesis in leaves and chloroplasts with a high proteincontent is discussed. The conductance of the cell to the fluxof CO2 from intercellular spaces to RuBPc-o active sites iscalculated, from cell surface areas and CO2 fluxes, to decreasethe CO2 partial pressure at the active site by less than 0.8Pa at an internal CO2 partial pressure of 34 Pa. Thus the decreasein partial pressure of CO2 is insufficient to account for theinefficiency of RuBPc-o in vivo at high protein contents. Otherlimitations to the rate of photosynthesis are considered. Bot. 36: 1117–1125. Rates of net and gross photosynthesis of intact white cloverleaves were measured by infrared gas analysis and by short termuptake of 14CO2 respectively. Ribulose bisphosphate carboxylaseoxygenase (RuBPCO) was purified from young leaves and kineticproperties investigated in combined and separate assays. Theratio of carboxylase to oxygenase activities was compared withthe ratio of photosynthesis to photorespiration at various temperaturesand CO2 concentrations. The ratio of photosynthesis to photorespiration at 30 Pa p(CO2)was consistent with the ratio of carboxylase activity to oxygenaseactivity when each was measured above 20 °C. However, theratio of photosynthesis to photorespiration increased with decreasingtemperature, whereas the ratio of carboxylase to oxygenase activitywas independent of temperature. This resulted in a disagreementbetween the measurements on the purified enzyme and intact leafat low temperature. No disagreement between enzyme and leafat low temperature occurred, when the ratio of photosynthesisto photorespiration was determined at increased CO2 concentrations. The results suggest an effect of low temperature and low CO2concentration on the ratio of photosynthesis to photorespirationindependent of the enzyme. Photosynthesis by developing embryos of oilseed rape (Brassica napus L.)   总被引:1,自引:0,他引:1  
The aim of this study was to assess the photosynthetic potentialof developing seeds of oilseed rape (Brassica napus L.) andto compare photosynthetic properties of embryo plastids withthose of leaf chloroplasts from the same species. Measurementsof CO2-dependent O2 evolution show that developing seeds ofB. napus are photosynthetically active in vitro. Essentially,all of the photosynthetic activity of the developing seed isaccounted for by the embryo. The rate of photosynthesis by developingembryos increased until the onset of desiccation, after whichit declined, so that by maturity embryos were no longer photosyntheticallyactive. Photosynthetic activity was positively correlated withchlorophyll content throughout development. Comparison of thephotosynthetic characteristics of leaf and embryo chloroplastsrevealed that rates of uncoupled electron transport were 2.5-foldgreater in those from the embryo. Light-saturated rates of CO2-dependentO2 evolution, per unit chlorophyll, and CO2 saturation pointswere similar for chloroplasts from both tissues. However, light-saturationpoints and chlorophyll a/b ratios were lower for embryo thanfor leaf choroplasts. Embryos and embryo chloroplasts also containedconsiderably less ribulose 1,5-bisphosphate carboxylase/oxygenaseprotein per unit total protein, than leaves. Although excisedembryos were capable of high rates of CO2-dependent O2 evolution(90–100 mol mg–1 chlorophyll h–1) under asaturating photosynthetic photon flux density (PPFD), low transmittanceof light through the silique wall (30%), together with the highPPFD required to achieve light compensation points in developingseeds (500 mol m–2 s–1), suggests that photosynthesisin vivo is unlikely to make a net contribution to carbon economyunder normal environmental conditions. A comparison of the activity and properties of the enzyme phosphoenolpyruvatecarboxylase (PEPC) was made for plants of Sedum telephium L.grown under low (70 µmol m–2 s–1) or high(500µmol m–2 s–1) PPFD and subjected to varyingdegrees of water stress. Under well-watered conditions onlyplants grown under high PPFD accumulated titratable acidityovernight and the extractable activity of PEPC was almost 2-foldhigher in these plants than in plants grown under low PPFD.Increasing drought stress resulted in a substantial increasein the activity of PEPC extracted both during the light anddark periods and a decrease in the sensitivity to inhibitionby malic acid. The magnitude of these changes was determinedby the severity and duration of drought and by light intensity.A comparison of the kinetic properties of PEPC from severelydroughted plants revealed that plants droughted under high PPFDhad a lower Km for PEP than plants under low PPFD. Additionof 2·0 mol m–3 malate resulted in an increase inthe Km for PEP, with plants draughted under low PPFD havinga significantly higher Km in the presence of malic acid comparedto those under high PPFD. Response to the activator glc-6-P,which lowered the Km for PEP, also varied between plants grownunder the two light regimes. Under well-watered conditions PEPCextracted from plants under high PPFD was more sensitive toactivation by glc-6-P than those under low PPFD. After the severedrought treatment, however, the Km for PEP in the presence ofglc-6-P was similar for enzyme extracted from plants grown underboth light regimes. Soluble sugars and starch were depletedovernight and were both possible sources of substrate for PEPC.With increasing drought, however, the depletion of starch relativeto soluble sugars increased under both light regimes. The propertiesof PEPC and the characteristics of carbohydrate accumulation/depletionare discussed in relation to the regulation of CAM in S. telephiumgrown under different light and watering regimes. The relationship between the rate of CO2 fixation and sucroseconcentration in the leaves of rice (Oryza sativa L.) grownat different light intensities was investigated. Maximum sucrosecontent coincided with maximum rates of CO2 fixation, achievedat a photon flux density of 1600 µmol m–2 s–1.The levels of sucrose and fructose 2,6-bisphosphate were alsocompared in the leaves under different light intensities. Fructose2,6-Msphosphate accumulated during growth at low light. Theactivity of fructose-6-phosphate 2-kinase was high in the leavesgrown at low light while that of fructose-2,6-bisphosphatasewas low. The activities of phosphoglucose isomerase and phospho-glucomutasewere slightly increased by growth at low light The activitiesof UDP glucose pyrophosphorylase were adversely affected invitro with increased concentrations of fructose 2,6-bisphosphatewhile those of sucrose phosphate synthase were moderately affected.Phosphoglucose isomerase and phosphoglucomutase were activatedby fructose 2,6-bisphosphate (8-0 mmol m–3) by 12-15%.The results suggested that low light intensities during growthresult in an accumulation of fructose 2,6-bisphosphate whichmodulates the key enzymes of sucrose biosynthesis thus regulatingcarbon flow under conditions of limited photosynthesis. The specificity factors for ribulose 1, 5-bisphosphate carboxylase/oxygenase(rubisco) from six species of photosynthetic organisms are compared.The values were determined by measuring the oxygen uptake duringthe total consumption of ribulose-P2 in the presence of variousconcentrations of O2 and CO2. The specificity factors determinedin this way were similar to values previously published; smallbut significant differences were found between the specificityfactors of rubisco from C3 higher plant species. Photosynthetic carbon metabolism was studied with Chroomonassp. cells in which the rate of photosynthesis was inhibitedunder both an anaerobic condition and high concentrations ofoxygen. The time course of 14C-incorporation into photosyntheticproducts showed that 3-phosphoglycerate was the initial productof photosynthetic CO2 fixation in Chroomonas sp. cells. During5-min photosynthesis, a considerable amount of 14C was incorporatedinto the insoluble fraction (mostly cryptomonad starch), andoxygen predominantly affected 14C-incorporation into this fraction.Although 14C-incorporation into intermediates of the photorespiratorypathway increased with increasing O2 concentration, the amountswere much less than expected from the degree of oxygen inhibition.It is noteworthy that 14C-dihydroxyacetone phosphate accumulatedduring photosynthesis only under the anaerobic condition, whereasthe levels of the other phosphate esters were scarcely affectedby the oxygen concentration. Ribulose-1,5-bisphosphate carboxylase from Chroomonas sp. wascompetitively inhibited by oxygen, and its Km(CO2) value wassimilar to those of terrestrial C3 plant enzymes. Photosynthetic rates of outdoor-grown soybean (Glycine max L.Merr. cv. Bragg) canopies increased with increasing CO2 concentrationduring growth, before and after canopy closure (complete lightinterception), when measured over a wide range of solar irradiancevalues. Total canopy leaf area was greater as the CO2 concentrationduring growth was increased from 160 to 990 mm3 dm–3.Photosynthetic rates of canopies grown at 330 and 660 mm3 CO2dm–3 were similar when measured at the same CO2 concentrationsand high irradiance. There was no difference in ribulose bisphosphatecarboxylase/oxygenase (rubisco) activity or ribulose 1,5-bisphosphate(RuBP) concentration between plants grown at the two CO2 concentrations.However, photosynthetic rates averaged 87% greater for the canopiesgrown and measured at 660 mm3 CO2 dm–3. A 10°C differencein air temperature during growth resulted in only a 4°Cleaf temperature difference, which was insufficient to changethe photosynthetic rate or rubisco activity in canopies grownand measured at either 330 or 660 mm3 CO2 dm–3. RuBP concentrationsdecreased as air temperature during growth was increased atboth CO2 concentrations. These data indicate that the increasedphotosynthetic rates of soybean canopies at elevated CO2 aredue to several factors, including: more rapid development ofthe leaf area index; a reduction in substrate CO2 limitation;and no downward acclimation in photosynthetic capacity, as occurin some other species. Changes in the activity and amount of ribulose 1,5-bisphosphate(RuBP)carboxylase (E.C. 4.1.1.39 [EC] ) were studied in well-watered plantsof Salix ‘aquatica gigantea’ and in similar plantsduring three different water stress treatments and after rewatering.The chloroplast ultrastructure of these plants was examinedby electron microscopy. The amounts of crystallized proteinin the chloroplast stroma were assessed according to the areaof crystal structure seen in the thin sections. RuBP carboxylase activity decreased with decreasing leaf waterpotentials but recovered upon rewatering, except when leaveshad been exposed to severe water stress. The percentage of totalchloroplast area made up of crystal inclusions decreased withdecreasing leaf water potentials. After rewatering, the crystalseither disappeared or the amount decreased markedly. Both RuBPcarboxylase activity and the area of crystal inclusions increasedinitially with increased extractable RuBP carboxylase proteinbut decreased with further increases above 6700–7000 µgRuBP carboxylase protein mg–1 chlorophyll. In well-wateredand water-stressed plants the activity of RuBP carboxylase,based on amount of chlorophyll, increased with an increasingamount of crystal inclusions in the chloroplast stroma. In rewateredplants no such correlation was observed, and the low percentageof crystal inclusions in the chloroplast area was independentof RuBP carboxylase activity. The effect of temporary droughts on photosynthesis, total conductanceto water vapour, intercellular CO2 concentration, CO2 compensationpoint, light-response curves, photorespiration, dark respiration,chlorophyll content, and ribulose 1,5-bisphosphate (RuBP) carboxylase(EC 4.1.1.39 [EC] ) activity has been examined in nodulated alfalfaplants (Medicago saliva cv. Aragón). Plants were subjectedto moderate (S1/RS1) or severe (S2/RS2) cycles of drought (drought/recovery).Photosynthetic light-response curves showed decreased light-saturatedphotosynthetic capacity and decreased apparent quantum yield.Upon rewatering, photosynthesis did not recover whereas conductancedid in moderately stressed plants. Calculated electron transportrate also declined in drought-stressed plants, but upon rewatering,moderately stressed plants exhibited a total recovery. Comparison of photosynthetic intercellular CO2 response curvesin well-watered and stressed leaves led to the assertion thateffects in chloroplast metabolism contribute significantly tophotosynthetic inhibition. Although the validity of this entireline of research has been questioned by some recent studiesbecause the occurrence of heterogeneous stomatal closure wouldaffect these curves, in our case, the effect of water stresswas investigated in experimental systems where stomata had beenremoved. Measurements of in vitro RuBP carboxylase activityand protein content showed a strong decline during drought treatmentsand upon rewatering no recovery was observed. Therefore, ourresults suggest the major implication of non-stomatal factorsin the decline of photosynthesis in alfalfa plants under cyclicdrought conditions. Bot. 36: 1835-1841. White clover was grown at 20 and100 Pa p(CO2). The CO2 response of net photosynthesis and differentialuptake of 14CO2 and 12CO2 by leaves were measured at varioustemperatures and at various O2 and CO2 partial pressures andcompared with predictions from ribulose bisphosphate carboxylase/oxygenasekinetics. Discrepancies between the observed gas exchange characteristicsfor the leaves and those predicted from the enzyme kineticswere interpreted as being due to a CO2 concentrating mechanism.Plants grown at 20 Pa p(CO2) showed a higher affinity for CO2than plants grown at 100 Pa p(CO2) when measured at 10 ?C. Nodifference in affinity was found at 30 ?C. The postulated CO2concentrating effect was greater in plants grown at low CO2than in plants grown at high CO2 concentration and occurredonly at low temperature and low CO2 partial pressure. It issuggested that plants grown at the lower CO2 partial pressurehave a higher affinity for CO2 due to a more efficient CO2 concentratingsystem than plants grown at the higher CO2 partial pressure. The effects of phosphate concentration on plant growth and photosyntheticprocesses in primary leaves of young sunflower (Helianthus annuusL.) plants were examined. Plants were grown for 3 weeks on half-strengthHoagland's solution containing 0, 0.1, 0.5, 1.0, and 3.0 molm–3 orthophosphate (Pi). It was shown that optimal photosynthesisand the highest light utilization capacity were achieved at0.5 mol m–3 Pi in the growth medium, which was in goodagreement with the maximum content of organic phosphorus inthe leaves. Low phosphate in the medium inhibited plant growthrate. Phosphate deficiency appreciably decreased photosyntheticoxygen evolution by leaves, the efficiency of photosystem two(PSII) photochemistry and quantum efficiency of PSII electrontransport. High oxidation state of PSII primary electron acceptorQA, at 0.1 mol m–3 Pi, however, indicates that photosyntheticelectron transport through PSII did not limit photosynthesisin Pi-deficient leaves. The results indicate that diminishedphotosynthesis under sub- and supra-optimal Pi was caused mainlyby a reduced efficiency of ribulose 1, 5-bisphosphate (RuBP)regeneration at high light intensities. These results suggestthat, under non-limiting C02 and irradiance, photosynthesisof the first pair of leaves could be diminished by both sub-and supra-optimal phosphorus nutrition of sunflower plants. Activities of photosynthetic and photorespiratory enzymes viz.,ribulose bisphosphate carboxylase, phosphoenol pyruvate carboxylaseand glycolate oxidase from jute (Corchorus olitorius L.; cv.JRO 632) leaves were compared with those from maize (C4) andsunflower (C3) leaves. The photosynthetic CO2 fixation products,the release of 14CO2 in light and dark following photosynthesisin 14CO2, chlorophyll a: b ratio, gross leaf photosyntheticrate and dry matter production rate were also studied. The native pyrenoid core matrix of the green alga Bryopsis maximawas isolated by diethyl ether treatment and sucrose densitygradient centrifugation using 1.8 M phosphate buffer. The purityof the pyrenoids was examined by microscopy, polyacrylamidegel electrophoresis and marker materials. The purified pyrenoidscontained the large subunit and the small subunit of ribulose1,5-bisphosphate carboxylase (RuBPCase) and more than 10 minorpolypeptides. They also showed RuBPCase activity when solubilizedon being transferred to a low-concentration buffer. The specificactivity was 0.62 µmol CO2 fixed (mg protein)–1min–1. This isolation method is suitable for obtainingintact pyrenoids not covered by starch sheaths or membraneswithout the need for chloroplast fixation.
