The dielectric permittivity of alpha-elastin coacervate is reported over the frequency range of 1 MHz to 1000 MHz and the temperature dependence from 6.8 degrees C to 70 degrees C is also reported. A temperature-dependent simple Debye-type relaxation is observed with a correlation time of 8 nsec (40 degrees C) which is similar to that of the polypentapeptide of elastin (i.e. 7 nsec at 40 degrees C) where the band has been assigned to a peptide librational mode. By analogy this allows for the first assignment of a peptide librational mode in a naturally occurring polypeptide or protein. The strong spectrally localized band indicates a regularity of structure. The low temperature dependence of the correlation time, giving a 1.7 kcal/mole enthalpy of activation, is consistent with torsional motions associated with a peptide librational mode.  相似文献

Dextran, pullulan and amylose have been investigated by differential scanning calorimetry, thermogravimetric analysis, dynamic mechanical and dielectric spectroscopy over a wide range of temperatures and frequencies. No melting or glass transition is seen below the range of thermal degradation (about 300 degrees C) for either amylose or pullulan; only dextran shows a Tg at 223 degrees C (delta cp = 0.40 J/g deg). The viscoelastic spectrum of the 'dry' polysaccharides is characterized by a low temperature relaxation that occurs at -94, -73 and -59 degrees C, at 1 kHz, (activation energy 32, 39 and 52 kJ/mol) in dextran, pullulan and amylose respectively and is assigned to small entity local motions of the polysaccharide backbone. Absorbed water strongly modifies the relaxation spectrum, inducing a new relaxation below room temperature and dissipation regions associated with water loss above room temperature. The former appears at temperatures higher than the relaxation characteristic of the dry polymer and moves to lower temperature with increasing water content. In normal 'room humidity' conditions (about 10% absorbed water) the water-induced relaxation, attributed to the motion of complex polymer-water relaxing units, is the only observable feature in the dynamic mechanical and dielectric spectrum below room temperature.  相似文献

The relative permittivity and conductivity of aqueous solutions of oxyhaemoglobin and carboxyhaemoglobin were measured over the frequency range 150kHz-100MHz. To minimize errors of measurement the investigations were carried out with three different samples of each type of haemoglobin, independent apparatus being used in two different laboratories. The dielectric increment and relaxation time were calculated at each of several temperatures from the results. These lead to a dipole moment of 400 Debyes and an activation enthalpy of 17.6+/-1.4kJ.mol(-1), both of which were found to be independent of temperature to within experimental error over the range 3-35 degrees C. The value of the dipole moment shows that the distribution of charge throughout the haemoglobin molecule is nearly symmetrical with respect to the centre of charge. The magnitude of the activation enthalpy is similar to that of the viscosity of water, in accord with the common observation that dielectric relaxation and viscosity are related phenomena. No significant differences are found between the dielectric parameters of oxyhaemoglobin and carboxyhaemoglobin. Combining the results with those obtained from X-ray diffraction of the solid a hydration value of 0.45g of water/g of protein is suggested, subject to the limitations of the model used. Finally, the results indicate the presence of a subsidiary dispersion, which could be attributed to the above quantity of bound water having a static permittivity of about 100 and a relaxation frequency in the region 100-200MHz.  相似文献

The complex permittivities of L-alpha-lysolecithin in the absence and presence of the gramicidin A ion channel were measured over the temperature range 0-60 degrees C and over the frequency range 1-1000 MHz. One dielectric relaxation/loss has been observed. It is located at 103.3 MHz (1.54 ns) for a micellar 0.4 M L-alpha-lysolecithin solution at 20 degrees C, whereas it is shifted to 71.7 MHz (2.22 ns) for a lamellar L-alpha-lysolecithin-gramicidin A aqueous solution (0.4 M L-alpha-lysolecithin, 0.0308 M gramicidin A) at 20 degrees C. The dielectric relaxation decreases and the relaxation time increases when gramicidin A is incorporated into L-alpha-lysolecithin. These dielectric changes are related, in part, to the micellar-to-lamellar lipid phase transition induced by the incorporation of gramicidin A into lysolecithin. We suggest that the diffuse rotational motion of the polar head group of L-alpha-lysolecithin contributes to the dielectric relaxation/loss at around 100 MHz.  相似文献

The dielectric permittivity and conductivity of muscle fibers from the giant barnacle, Balanus nubilus, have been measured at 1, 25, and 37 degrees C, between 10 MHz and 17 GHz. The dominant microwave dielectric relaxation process in these fibers is due to dipolar relaxation of the tissue water, which shows a characteristic relaxation frequency equal to that of pure water, ranging from 9 GHz (1 degree C) to 25 GHz (37 degree C). The total permittivity decrease, epsilon 0 -- epsilon infinity, due to this process accounts for approximately 95% of the water content of the tissue; thus, the major fraction of tissue water is dielectrically identical to the pure fluid on a picosecond time scale. A second dielectric process contributes significantly to the tissue dielectric properties between 0.1 and 1--5 GHz, and arises in part form Maxwell-Wagner effects due to the electrolyte content of the tissue, and in part from dielectric relaxation of the tissue proteins themselves.  相似文献

The complex permittivities of aqueous suspensions of 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) and of DMPC packaged gramicidin A' (DMPC-GA) have been determined over the frequency range of 1 MHz to 1 GHz and the temperature range of 0-60 degrees C. A dielectric relaxation/loss has been observed at about 66 MHz for the DMPC suspension (30 degrees C) and at about 57 MHz for the DMPC-GA suspension (30 degrees C). This dielectric relaxation/loss has been attributed to the rotational mobility of the zwitterionic group of DMPC. The temperature dependence (from 60 degrees C to 0 degrees C) of this dispersion/absorption process of the DMPC suspension indicates a sharp reduction of the dielectric relaxation at about 20 degrees C. This dielectric change is related to the conversions of shape and structure of bilayer aggregates. This sharp reduction of the dielectric relaxation disappears or broadens when GA is incorporated into the DMPC aqueous suspension. The interpretation of these results is that the GA addition into the DMPC aqueous suspension induces a small decrease of the rotational mobility of the zwitterionic group above the lipid phase transition, and a small increase of the rotational mobility of the zwitterionic group below the lipid phase transition.  相似文献

Isothermal dielectric loss spectra of neutralized and nonneutralized chitosan were acquired in successive runs from -130 degrees C up to increasing final temperatures, in a frequency range between 20 Hz and 1 MHz. Essentially, three relaxation processes were detected in the temperature range covered: (i) a beta-wet process, detected when the sample has a higher water content that vanishes after heating to 150 degrees C; (ii) a beta process, which is located at temperatures below 0 degrees C, becoming better defined and maintaining its location after annealing at 150 degrees C independently of the protonation state of the amino side group; and (iii) a sigma process that deviates to higher temperatures with drying, being more mobile in the nonneutralized form. Moreover, in dried neutralized chitosan, a fourth process was detected in the low frequency side of the secondary beta process that diminishes after annealing. Whether this process is a distinct relaxation of the dried polymer or a deviated beta-wet process due to the loss of water residues achieved by annealing is not straightforward. Only beta and sigma processes persist after annealing at 150 degrees C. The changes in molecular mobility upon drying of these two relaxation processes were evaluated.  相似文献

The complex permittivity of aqueous solutions at 20 degrees C has been measured at concentrations between 0.001 and 5 mol/L and over a frequency range 0.13-20 GHz. The results were combined with literature values to derive empirical equations to predict the dielectric behavior of sodium chloride solutions between 0 and 5 mol/L and 5 degrees C-35 degrees C.  相似文献

The glass transition temperature, T(g), and enthalpy relaxation of amorphous lactose glass were investigated by differential scanning calorimetry (DSC) for isothermal aging periods at various temperatures (25, 60, 75, and 90 degrees C) below T(g). Both T(g) and enthalpy relaxation were found to increase with increasing aging time and temperature. The enthalpy relaxation increased approximately exponentially with aging time at a temperature (90 degrees C) close to T(g) (102 degrees C). There was no significant change observed in the enthalpy relaxation around room temperature (25 degrees C) over an aging period of 1month. The Kohlrausch-Williams-Watts (KWW) model was able to fit the experimental enthalpy relaxation data well. The relaxation distribution parameter (beta) was determined to be in the range 0.81-0.89. The enthalpy relaxation time constant (tau) increased with decreasing aging temperature. The observed enthalpy relaxation data showed that molecular mobility in amorphous lactose glass was higher at temperatures closer to T(g). Lactose glass was stable for a long time at 25 degrees C. These findings should be helpful for improving the processing and storage stability of amorphous lactose and lactose containing food and pharmaceutical products.  相似文献

The electric permittivity of aqueous solutions of different synthetic polyelectrolytes have been measured as a function of frequency in the range 5 kHz up to 100 MHz in the absence of added salt. Solutions of polymethacrylic acid and polyacrylic acid of different degrees of polymerization, both partially neutralized with NaOH, were investigated as well as solutions of Na-polystyrenesulphonate at different concentrations.For all systems a dispersion profile with two separated dispersion regions was obtained with a molecular weight dependent value of the static electric permittivity. The low frequency dispersion region was found to be characterized by a molecular weight dependent mean relaxation time while for the high frequency dispersion region both the mean relaxation time and the dielectric increment are molecular weight independent. It is shown that the reciprocal values of the specific increments and of the relaxation times depend linearly on the macromolecular concentration. Extrapolation of the corresponding quantities to infinite dilution was found to be possible. A comparison of these extrapolated values with calculated ones according to the previously derived theory also applicable to flexible macromolecules establishes that this theory describes satisfactorily the dielectric behaviour of the systems investigated.The conclusion is reached that the high frequency dispersion and relaxation can be attributed to fluctuations in the distribution of bound counterions along limited parts of the macromolecule. The relaxation time of the low frequency dispersion region seems to be essentially determined by the rotation of the complete molecule and the static electric permittivity can he explained in terms of fluctuations in the counterion density extending over the whole macromolecule.  相似文献

Temperature-jump measurements were carried out on myosin subfragment 1 (S1) labeled at Cys-707 with 5-(iodoacetamido)fluorescein (S1-AF). The relaxation was monitored by following the increase in the fluorescence intensity of the attached probe after a jump of 5.8 degrees C. A single relaxation process was observed over a range of final temperatures, and the relaxation time decreased from 16.69 ms at 15 degrees C to 3.91 ms at 27 degrees C. The relaxation results are interpreted in terms of a two-state transition: (S1-AF)L K+ in equilibrium with K- (S1-AF)H, and the observed single relaxation time (tau) equals l/(k(+) + k-). The individual first-order rate constants, k+ and k-, were calculated from tau and the equilibrium constant previously determined. The activation energy was 21.9 kcal/mol for the forward reaction and 9.3 kcal/mol for the reverse reaction, corresponding to an enthalpy value of 12.6 kcal/mol for the two-state transition. The results provide, for the first time, direct kinetic evidence of a two-state transition of S1 in the absence of bound nucleotide, and support a two-state model of unliganded myosin subfragment 1.  相似文献

The aim of the work was to investigate the temperature effect in physiological range on the dielectric properties of a fish (Cyprinus carpio) blood. By applying an empirical non-damaging method and statistical analysis of the dielectric data, it is observed that a heating of carp blood induces an almost linear value variation of the electrical conductivity and permittivity of the intracellular matter of carp red blood cells. The dielectric data, however, reveal anomalous behaviour for conductivity and permittivity of the interior of carp erythrocytes at 23 degrees C and 23 degrees, 37 degrees C, respectively and for both volume fraction and dielectric loss factor tg delta at 30 degrees, 35 degrees, 37 degrees C. For the presented dielectric parameters the temperature coefficient and the Arrhenius activation energy were estimated. Moreover, this work led to conclusion that the changes of the volume fraction of carp erythrocytes induce corresponding variations of the maximum of the dielectric loss factor tg delta determined for a whole blood.  相似文献

Dielectric spectroscopy has been applied to study the decomposition process of unmodified collagen and chondroitin sulfate (CS)- and hyaluronic acid (HA)-modified collagen. Measurements were performed over the frequency range from 10 Hz to 100 kHz and at temperatures from 22 to 260 degrees C. According to the Kramers-Kronig relationship a dispersion is apparent in both epsilon' and epsilon' for the three materials below 140 degrees C and at higher temperatures as a broad peak around 220-230 degrees C, respectively. The values of epsilon' and epsilon' at the same temperature for constant frequency are higher in HA-modified collagen than in the unmodified collagen. However, small differences are shown in these parameters between CS-modified collagen and unmodified collagen. The observed dispersion around 220-230 degrees C corresponds to the decomposition of unmodified and CS- and HA-modified collagen. Power-low responses are observed for the frequency dependence of ac conductivity for unmodified and modified collagen. The behaviour observed for temperature dependencies of the exponent n for the three materials is considered to be related to the proton polarization and conduction processes.  相似文献

The dynamic mechanical properties of purified collagen from bovine tendon were studied using a torsion pendulum in the temperature range of 120°-360°K at 0.3-1 cps. In the temperature range studied, two loss peaks were observed: a β-peak at about 200°K, and an α-peak approximately five times larger at about 280°K. The temperature of the α-transition is shown to be dependent on water content, decreasing with increasing amount of water and shifting to lower temperatures. Broad-line proton magnetic resonance results were also obtained on similar samples. A narrow nuclear magnetic resonance (NMR) line appears at about 250°C. The effects of shrinkage to form gelatin and of cross-linking on the relaxation behavior of collagen were also studied. The motions taking place in collagen over the 120°-360°K range are discussed.  相似文献

With an aim to reveal the mechanism of protein-water interaction in a predominantly two phase model protein system this study investigates the frequency and temperature dependence of dielectric constant epsilon' and loss factor epsilon' in cow horn keratin in the frequency range 30 Hz to 3 MHz and temperature range 30-200 degrees C at two levels of hydration. These two levels of hydration were achieved by exposing the sample to air at 50% relative humidity (RH) at ambient temperature and by evacuating the sample for 72 h at 105 degrees C. A low frequency dispersion (LFD) and an intermediate frequency alpha-dispersion were the two main dielectric responses observed in the air-dried sample. The LFD and the high frequency arm of the alpha-dispersion followed the same fractional power law of frequency. Within the framework of percolation cluster model these dispersions, respectively have been attributed to percolation of protons between and within the clusters of hydrogen-bonded water molecules bound to polar or ionizable protein components. The alpha-dispersion peak, which results from intra-cluster charge percolation conformed to Cole-Cole modified Debye equation. Temperature dependence of the dielectric constant in the air-dried sample exhibited peaks at 120 and 155 degrees C which have been identified as temperatures of onset of release of water bound to polar protein components in the amorphous and crystalline regions, respectively. An overall rise in the permittivity was observed above 175 degrees C, which has been identified as the onset of chain melting in the crystalline region of the protein.  相似文献

Neutralized and nonneutralized chitosan films subject to different thermal treatments were studied by dielectric relaxation spectroscopy from -130 to +150 degrees C in the frequency range between 20 Hz and 1 MHz. Two main relaxation processes, both arrhenian type, were detected: process I at temperatures below 0 degrees C with a mean activation energy of 49 +/- 1 kJ mol(-1), which has the characteristics of a secondary relaxation process related with local chain dynamics, and process II observable at higher temperatures with an activation energy of 94 +/- 2 kJ mol(-1), correlated with dc conductivity, which is found in dried polysaccharides systems. Process I is always observed in neutralized chitosan, but it is strongly depleted in the wet nonneutralized form. Although the location of process I is independent of NH2/NH3+ side group, process II deviates to higher temperatures with dryness in both chitosan forms, being located at lower temperatures in nonneutralized chitosan.  相似文献

In this work, detailed dielectric measurements are presented on aqueous electrolytic solutions of NaCl and KCl in a broad frequency range, typical for modern telecommunication techniques. The complex dielectric permittivity or equivalently the complex conductivity are systematically studied as function of frequency (100 MHz-40 GHz), temperature (10-60 degrees C) and molar concentration (0.001-1 mol/l). By a detailed analysis of the dielectric results using an asymmetrically broadened Cole-Davidson distribution of relaxation times, in addition to dc conductivity, the dielectric response as function of frequency, temperature, and molar concentration was fully parameterized by a total of 13 parameters. This model ansatz and the 13 parameters include an enormous predictive power, allowing a reasonable estimation of the dielectric constant, loss, and the conductivity for any set of external variables frequency, temperature and concentration. The proposed method is not only useful for rather simple electrolytic solutions, but also for cell suspensions and biological matter, if additional processes, especially at low frequencies, are adequately taken into account.  相似文献

The dielectric properties of biologically and pharmaceutically important low-molecular weight ethylene glycols H(-OCH2CH2-)n -OH (n = 1,2,4,6) were investigated to clarify the effect of chain length on the dielectric properties. The measurement of dielectric constant and dielectric loss was carried out over the frequency range 200 MHz to 20 GHz at temperatures of 25 degrees C to 55 degrees C. It is found that in these molecules microwave dielectric losses are significant. The dispersion behaviour of these molecules can be represented by Cole-Cole equation. The dielectric properties of these homologous ethylene glycols are discussed in terms of the effects of chain length and intermolecular hydrogen bonds regarding the molecular conformations. These wide frequency range dielectric data have also been discussed in view of the suitable selection of the oligomer of ethylene glycol for cosmetic preparations and other pharmaceutical applications with the intention of protection of the skin from weak microwave radiations present in the surrounding environment. These systematic microwave dielectric data with frequency and temperature variation are not available and are provided in this paper.  相似文献

The relative permittivity and dielectric loss of aqueous solutions of plasmid (pUC8.c1 and pUC8.c2) DNA have been measured at 20 degrees C over the frequency range 100 MHz-10 GHz. The solutions had a concentration of 0.1% DNA, and were studied both in the relaxed and the supercoiled form. The dielectric measurements were made using a variety of techniques including frequency domain and time domain methods of operation. No evidence of any resonance absorption, nor of any other kind of enhanced absorption, was observed.  相似文献

The electric permittivity of poly-L-glutamic acid (PGA) in salt-free aqueous solutions was measured in the frequency range 2.5 kHz – 100 MHz at different concentrations and degrees of ionization. Two samples of different molecular weight were investigated. The experimental results could under most circumstances be described by a superposition of two dispersion curves of the Cole-Cole type. The low-frequency dielectric parameters were strongly molecular weight dependent, the high-frequency ones not. Strong concentration effects were observed resulting in increasing specific dielectric increments and relaxation times with decreasing concentration. Using the theory proposed by Van der Touw and Mandel to interpret the experimental results these concentration effects could be ascribed to the influence of the polyion interactions on the average dimensions and the rigidity of the polyelectrolyte chains. The change in the total dielectric increment and low-frequency relaxation time with degree of ionization correctly reflects the helix-coil transition of PGA occurring in ths region α = 0.3–0.5. The effect of counterion size and charge on the dielectric behaviour was also found to be consistent with the theoretical model.  相似文献
