The effect of long-term mercury pollution on the soil microbial community   总被引:1,自引:0,他引:1  
The effect of long-term exposure to mercury on the soil microbial community was investigated in soil from three different sites along a pollution gradient. The amount of total and bioavailable mercury was negatively correlated to the distance from the center of contamination. The size of the bacterial and protozoan populations was reduced in the most contaminated soil, whereas there was no significant difference in fungal biomass measured as chitinase activity. Based on the number of colony morphotypes, moreover, the culturable bacterial population was structurally less diverse and contained a higher proportion of resistant and fast-growing forms. The profiles of amplified 16S rDNA sequences obtained from community DNA by denaturating gradient gel electrophoresis (DGGE) also reflected the altered community structure and decreased diversity along the mercury gradient as expressed in terms of the number and abundance of bands. The functional potential of the microbial population measured as sole carbon source utilization by Ecoplates((R)) differed between the soils, but there was no change in the number of substrates utilized. The observed changes in the different soil microbial populations are probably a combination of both direct and indirect effects of the mercury contamination.  相似文献

The relative efficacy of thiol-containing mercurial scavengers was assayed by using cellular suspensions of erythrocytes or isolated hepatocytes. The blood cells incubated in a buffer (pH 7.4) containing 1 mM glucose (10% hematocrit) were exposed to 5 μM methyl mercuric chloride. In the absence of extracellular thiols the red blood cells took up more than 90% of methyl mercury from the surrounding medium during 5–10 min. This uptake was almost completely inhibited by dimercaptosuccinic acid (DMSA) (1 mM) and the same chelant could rapidly remove 80% of the mercury from ‘pre-loaded’ erythrocytes. Hepatocytes prepared according to the method of Seglen [11] in a suspension of 106 cells/ml in a buffer containing 5 mM glucose and 5 mg/ml of bovine serum albumin were also exposed to methyl mercuric chloride (4 μM). Almost 50% of the mercurial was taken up by the cells slowly during the incubation period of 240 min. DMSA (1 mM) almost completely blocked the methyl mercury binding by the hepatocytes. 2-Mercaptopropionylglycin (Thiola) or mercaptosuccinic acid (MSA) was almost as effective mercurial scavengers as DMSA in hepatocytes and in red blood cells. Diethyldithiocarbamate (DDC) and dimercaptopropanol (BAL) were considerably less effective than DMSA to inhibit the mercurial binding to hepatocytes. Experiments in vivo have shown that DMSA is a better mercurial chelator than Thiola or MSA, whereas DDC and BAL may both be considered to be inapplicable in methyl mercury poisonings. Our cellular assay provides preliminary information of the efficiency of chelating thiols and may serve as a useful first approximation when planning further experiments.  相似文献

Successions are a central issue of ecological theory. They are governed by changes in community assembly processes that can be tracked by species’ traits. While single‐trait‐based approaches have been mostly promoted to address community assembly, ecological strategies actually encompass tradeoffs between multiple traits that are relevant to succession theory. We analyzed plant ecological strategies along a 140‐year‐long succession primary succession of 52 vertical outcrop communities after roadwork. We performed a RLQ analysis to relate six functional traits, associated with resource acquisition, competition, colonization ability and phenology, to the age of the outcrops. We found the prominence of two main axes of specialization, one related to resource acquisition and the other to reproduction and regeneration. We further examined the community‐level variation in ecological strategies to assess the abiotic and biotic drivers of community assembly. Using trait‐based statistics of functional richness, regularity and divergence, we found that different processes drove the variation in ecological strategies along the axes of specialization. In late succession, functional convergence was detected for the traits related to resource acquisition as a signature of habitat filtering, while the coexistence of contrasted strategies was found for the traits related to reproduction and regeneration as a result of spatial micro‐heterogeneity. We observed a lack of niche differentiation along the succession, revealing a weak importance of biotic interactions for the regulation of community assembly in the outcrops. Overall, we highlight a prominent role of habitat filtering and spatial micro‐heterogeneity in driving the primary succession governed by water and nutrient limitation.  相似文献

Rat amylin differs from human amylin (hIAPP) in that it lacks a fibril-forming capacity. As a consequence, toxic effects have been reported for human but not for rat amylin. This report demonstrates how a mercury contamination of commercial rat amylin imitates peptide-related vasoactive and cytotoxic effects on preparations of isolated blood vessels. The source of mercury contamination was believed related to the peptide synthesis. Thiol groups of cysteine-containing peptides are often protected by acetamidomethyl (Acm) which is cleaved by mercuric acetate.  相似文献

The effects of two mercurial compounds, methylmercury and inorganic mercury, on lipids were examined by measuring permeability change of lipid bilayer, liposome. Both decrease in the cholesterol content and increase in the content of unsaturated fatty acid moieties in the lipid bilayers, augmented to susceptibility of the liposomes to the mercurial compounds. Inorganic mercury and methylmercury disrupted the lipid membrane to essentially the same extent. The influence on the permeability seems to be specific for mercury compounds. The significant increase in the permeability of some liposomal preparation noted even at the mercurial concentration of 10(-7) M strongly suggests that lipid in biomembrane could be one of the primary targets of these toxic substances.  相似文献

Exposure to contaminated soil and dust is an important pathway in human health risk assessment. Physical and chemical characteristics and biological factors determine the bioaccessibility/bioavailability of soil and dust contaminants. Within a single sample, contamination may arise from multiple sources of toxic elements that may exist as different species that impact bioavailability. In turn, the bioaccessibility/bioavailability of soil and dust contaminants directly impacts human health risk. Research efforts focusing on development and application of in vitro and in vivo methods to measure the bioaccessibility/bioavailability of metal-contaminated soils have advanced in recent years. The objective of this workshop was to focus on developments in assessing the bioaccessibility/bioavailability of arsenic-contaminated soils, metals’ contamination in urban Canadian residences and potential children's exposures to toxic elements in house dust, an urban community-based study (i.e., West Oakland Residential Lead Assessment), bioavailability studies of soil cadmium, chromium, nickel, and mercury and human exposures to contaminated Brownfield soils. These presentations covered issues related to human health and bioavailability along with the most recent studies on community participation in assessing metals’ contamination, studies of exposures to residential contamination, and in vitro and in vivo methods development for assessing the bioaccessibility/bioavailability of metals in soils and dusts.  相似文献

Binding of metal ions to the heteroatomic sites of proteins is undoubtedly fundamental to their observed physiological effects. In this paper, the interactions of inorganic mercury (Hg2+), methylmercury (MeHg+), ethylmercury (EtHg+), and phenylmercury (PhHg+) with human serum albumin (HSA) were studied from the electrophoretic behaviors, stoichiometry, thermodynamics, and kinetics by using a new hybrid technique, capillary electrophoresis on-line coupled with electrothermal atomic absorption spectrometry (CE-ETAAS), together with the consequent structural information from circular dichroism and Raman spectroscopy. The stoichiometry (mercurial species to HSA) for the interactions of Hg2+, MeHg+, EtHg+, and PhHg+ with HSA was found to be 6:1, 4:1, 4:1, and 3:1, respectively. Two types of binding sites in HSA were observed for the binding of mercurial species with the orders of magnitude of binding constants of 10(7) and 10(6) L mol-1, respectively, showing strong affinity of mercurial species for HSA. The interactions of mercurial species with both types of binding sites in HSA are exothermic and thermodynamically favorable and are both enthalpically and entropically driven. The binding of mercurial species to HSA follows the first-order kinetics for mercurial species and zero-order kinetics for HSA with the apparent activation energy of 57-59 kJ mol-1. Among the four mercurial species examined, only Hg2+ induces the secondary structure transition of HSA. Mercury-HSA adducts are formed mainly through metal-sulfur binding with participation of C=O and/or C-N groups of amino acid residues in HSA molecules. The present work represents the most comprehensive study on the interactions between various mercurial species with HSA and provides new evidence for and insights into the interactions of mercurial species with HSA for further understanding of the toxicological effects of mercurial species.  相似文献

Once the moult patterns have been taken into account, feather methylmercury levels can be used to accurately measure the mercury burdens of seabirds. We used body feathers from live seabirds and from museum collections to examine geographical and temporal patterns of mercury contamination in the North Sea. This approach identifies an increase in mercury concentrations in seabirds of the German North Sea coast during the last 100 years, especially high levels during the 1940s, and reduced contamination in the last few years. Comparisons among populations suggest that some increases in mercury levels are predominantly due to local pollution inputs, as on the German coast, while in other areas deposition from jet stream circulation of global contamination may be the major contributor. Mercury levels are far higher in seabirds from the German North Sea coast than in populations from the north and west North Sea or from most areas of the North Atlantic. We advocate the use of museum collections of birds for studies of long-term changes in levels of mercury contamination.  相似文献

The sediments and aquatic life of Tokuyama Bay, Japan, have been polluted by mercury effluent from chloro-alkali plants. In total, about 380 tons mercury were released from these plants and 6.64 tons of mercury were discharged into the bay in waste waters between 1952 and 1975, when mercury cells were employed. A number of surveys to study mercury pollution and the effectiveness of control measures in this area were conducted in the early 1970's by our laboroatory and other agencies. Analysis of human hair from Tokuyama Bay residents contained less mercury than those in Minamata and Agano districts, Japan, where serious mercury poisoning had occurred, but were contaminated with more mercury than those in other unpolluted areas. No occurrence of Minamata disease has been reported in the Tokuyama district.Reclamation of mercury contaminated sediments began in 1975; dredging of the bay continued until 1977. Since then, the levels of mercury contamination in sediments and aquatic life have gradually decreased. Today there are no problems with respect to mercury pollution.In this paper, we describe and discuss mercury pollution in Tokuyama Bay with regard to the following aspects of research and pollution control: the history of mercury pollution; mercury discharge and its accumulation in sediments; behaviour of mercury in sediments; mercury contamination of fish; mercury and the health of local residents; and remedial actions.  相似文献

The contamination of polar regions with mercury that is transported as inorganic mercury from lower latitudes has resulted in the accumulation of methylmercury in the food chain of polar environments, risking the health of humans and wildlife. This problem is likely to be particularly severe in coastal marine environments where active cycling occurs. Little is currently known about how mercury is methylated in polar environments. Relating observations on mercury deposition and transport through polar regions to knowledge of the microbiology of cold environments and considering the principles of mercury transformations as have been elucidated in temperate aquatic environments, we propose that in polar regions (1) variable pathways for mercury methylation may exist, (2) mercury bioavailability to microbial transformations may be enhanced, and (3) microbial niches within sea ice are sites where active microorganisms are localized in proximity to high concentrations of mercury. Thus, microbial transformations, and consequently mercury biogeochemistry, in the Arctic and Antarctic are both unique and common to these processes in lower latitudes, and understanding their dynamics is needed for the management of mercury-contaminated polar environments.  相似文献

Species abundance and community composition are affected not only by the local environment, but also by broader landscape and regional context. Yet, determining the spatial scales at which landscapes affect species remains a persistent challenge, hindering our ability to understand how environmental gradients shape communities. This problem is amplified by rare species and imperfect species detection. Here, we present a Bayesian framework that allows uncertainty surrounding the ‘true’ spatial scale of species’ responses (i.e. changes in presence/absence) to be integrated directly into a community hierarchical model. This scale‐selecting multispecies occupancy model (ssMSOM) estimates the scale of response, and shows high accuracy and correct levels of uncertainty in parameter estimates across a broad range of simulation conditions. An ssMSOM can be run in a matter of minutes, as opposed to the many hours required to run normal multispecies occupancy models at all queried spatial scales, and then conduct model selection – a problem that up to now has prohibited scale of response from being rigorously evaluated in an occupancy framework. Alternatives to the ssMSOM, such as GLM‐based approaches frequently fail to detect the correct spatial scale and magnitude of response, and are often falsely confident by favoring the incorrect parameter estimates, especially as species’ detection probabilities deviate from perfect. We further show how trait information can be leveraged to understand how individual species’ scales of response vary within communities. Integrating spatial scale selection directly into hierarchical community models provides a means of formally testing hypotheses regarding spatial scales of response, and more accurately determining the environmental drivers that shape communities.  相似文献

The spatial distributions of species, and the resulting composition of local communities, are shaped by a complex interplay between species’ climatic and habitat preferences. We investigated this interaction by analyzing how the climatic niches of bird species within given communities (measured as a community thermal index, CTI) are related to vegetation structure. Using 3129 bird communities from the French Breeding Bird Survey and an information theoretic multimodel inference framework, we assessed patterns of CTI variation along landscape scale gradients of forest cover and configuration. We then tested whether the CTI varies along local scale gradients of forest structure and composition using a detailed data set of 659 communities from six forests located in northwestern France. At landscape scale, CTI values decreased with increasing forest cover, indicating that bird communities were increasingly dominated by cold‐dwelling species. This tendency was strongest at low latitudes and in landscapes dominated by unfragmented forest. At local scale, CTI values were higher in mature deciduous stands than in conifer or early stage deciduous stands, and they decreased consistently with distance from the edge of forest. These trends underpin the assertion that species’ habitat use along forest gradients is linked with their climatic niche, although it remains unclear to what extent it is a direct consequence of microclimatic variation among habitats, or a reflection of macroscale correlations between species’ thermal preferences and their habitat choice. Moreover, our results highlight the need to address issues of scale in determining how habitat and climate interact to drive the spatial distribution of species. This will be a crucial step towards accurate predictions of changes in the composition and dynamics of bird communities under global warming.  相似文献

The relationship of mercury resistance to the concentration and chemical speciation of mercurial compounds was evaluated for microbial communities of mercury-polluted and control waters. Methodologies based on the direct viable counting (DVC) method were adapted to enumerate mercury-resistant communities. Elevated tolerance to Hg(II) was observed for the microbial community of one mercury-polluted pond as compared to the community of control waters. These results suggest an in situ acclimation to Hg(II). The results of the methylmercury resistance-DVC assay suggested that minimal acclimation to CH3Hg+ occurred since similar concentrations of CH3HgCl inhibited growth of 50% of organisms in both the control and polluted communities. Analyses of different mercury species in pond waters suggested that total mercury, but not CH3Hg+ concentrations, approached toxic levels in the polluted ponds. Thus, microbial acclimation was specific to the chemical species of mercury present in the water at concentrations high enough to cause toxic effects to nonacclimated bacterial communities.  相似文献

The Allium micronucleus (MNC) assay was developed to monitor low levels of mercury in aquatic and terrestrial environments. Four mercurial derivatives namely mercuric chloride (MC), methyl mercuric chloride (MMC), phenyl mercuric acetate (PMA) and a methoxy ethyl mercuric chloride based fungicide, Emisan-6, were tested to assess the sensitivity and versatility of the Allium MNC assay. Allium bulbs were set directly on water and soil contaminated with known levels of mercurial derivatives (0.0001-10.00 ppm). On the 5th day the endpoints measured were root length, mitoses with spindle abnormality and cells with MNC in root meristems. The effective concentrations of the test chemicals that cause 50% of root length as compared to control (EC50) were determined from dose-response curves so obtained. The lowest effective concentration tested (LECT) and highest ineffective concentration tested (HICT) for each of the mercurial derivatives for the induction of spindle malfunction and MNC were determined. It was found that EC50, LECT and HICT values for mercurial derivatives in soil were higher than those in water. The frequencies of cells with MNC and mitoses with spindle abnormality were highly correlated indicating that MNC is a good parameter of spindle malfunction. The present approach increased the sensitivity of the Allium assay by 10-fold, the detection limit being 0.001-0.1 ppm and 0.1-1.0 ppm in aquatic and terrestrial environments respectively, depending on the species of mercury.  相似文献

In the 1980s, significant regional, interspecific and annual differences in contamination with toxic chemicals were found in eggs of breeding birds along the German North Sea coast. In some regions residue levels approached a range endangering breeding success, especially in terns. In 1987, a three-year monitoring-program for common ternSterna hirundo and oystercatcherHaematopus ostralegus was started, in order to recognize such contamination trends. Eggs from 9–14 areas along the coast are now being analysed for chlororganic residues and mercury. First results on geographical and year-to-year variation are presented here, and the advantages of seabird eggs as spatial and temporal monitors of marine pollution are discussed. The suggestion is made to include a shore-breeding bird species in the European monitoring programs. Insulin release in response to dextran-linked p-chloromercuribenzoic acid was studied in microdissected pancreatic islets of non-inbred ob/ob-mice. No contamination of the dextran-linked mercurial with free chloromercuribenzoic acid was detected before or after the incubation with islets. In comparison with free mercurial, of the same thiol-blocking activity, the dextran-linked compound had a weak insulin-releasing action with a different dose vs. response relationship. The dextran-linked mercurial had no demonstrable effect on the islet content of cyclic AMP. The results support the hypothesis that free organic mercurials mainly stimulate insulin release by blocking thiol groups that are embedded within the β-cell plasma membranes beneath their surfaces.  相似文献

Preserving biodiversity is a global challenge requiring data on species’ distribution and abundance over large geographic and temporal scales. However, traditional methods to survey mobile species’ distribution and abundance in marine environments are often inefficient, environmentally destructive, or resource‐intensive. Metabarcoding of environmental DNA (eDNA) offers a new means to assess biodiversity and on much larger scales, but adoption of this approach for surveying whole animal communities in large, dynamic aquatic systems has been slowed by significant unknowns surrounding error rates of detection and relevant spatial resolution of eDNA surveys. Here, we report the results of a 2.5 km eDNA transect surveying the vertebrate fauna present along a gradation of diverse marine habitats associated with a kelp forest ecosystem. Using PCR primers that target the mitochondrial 12S rRNA gene of marine fishes and mammals, we generated eDNA sequence data and compared it to simultaneous visual dive surveys. We find spatial concordance between individual species’ eDNA and visual survey trends, and that eDNA is able to distinguish vertebrate community assemblages from habitats separated by as little as ~60 m. eDNA reliably detected vertebrates with low false‐negative error rates (1/12 taxa) when compared to the surveys, and revealed cryptic species known to occupy the habitats but overlooked by visual methods. This study also presents an explicit accounting of false negatives and positives in metabarcoding data, which illustrate the influence of gene marker selection, replication, contamination, biases impacting eDNA count data and ecology of target species on eDNA detection rates in an open ecosystem.  相似文献

In soil, genetic structure modifications of indigenous bacterial community consecutively to a severe stress (mercury contamination) were delayed when the community was pre-exposed to various minor perturbations (heat, copper and atrazine). Such minor perturbations induced transitory community structure modifications leading to an increase of community stability towards a severe mercury stress. These results illustrated well the short-term pre-adaptation process for bacterial community hypothesizing that community submitted to perturbations become more resistant to withstand another stress.  相似文献
