Culture experiments on the toxic Nodularia spumigena strain NSGG-1 isolated from the Gulf of Gdańsk showed a significant effect of salinity on growth and nodularin production. Growth of the NSGG-1 strain, was optimal between 7 and 18 psu, lower at 3 and 24 psu and was significantly inhibited at the extreme salinities of 0 and 35 psu. Nodularin (NOD) content of N. spumigena, estimated by the NOD/Chla ratios, correlated positively with salinity and increased from 0 to 35 psu. The NOD/Chla ratio on day 10 of growth was high, and, reached the maximum at day 30. A sudden increase in salinity from 7 to 18 and 35 psu resulted in plasmolysis of Nodularia cells. Salinity was also observed to have other effects on NSGG-1; the filaments were longest at 7 psu, while an increased number of akinetes were formed at 35 psu. The number of heterocytes was markedly reduced at the extreme salinities. This latter finding might explain why Nodularia blooms do not occur outside a certain salinity range in nitrogen-deficient waters.  相似文献

Nodularia spumigena, like many cyanobacteria, produces specialised reproductive structures, known as akinetes, which are believed to allow survival under unfavourable conditions. This study investigated the effects of salinity, nitrogen and phosphorus concentration at two irradiances on akinete differentiation in a N. spumigena isolate from the Gippsland Lakes, Victoria, Australia. A computer image analysis program was used to photograph filaments and assess production of akinetes over time in separate experiments for each environmental parameter. Heterocyst production and cell morphology were also examined. The results suggest that akinete production increases over time. Production of akinetes is further increased at low and high salinities and with the addition of nitrate. Higher irradiance increases akinete differentiation, although in combination with different phosphorus concentrations causes varied effects. The development and sedimentation of akinetes may provide an inoculum for reoccurring blooms. Heterocysts were only observed during experiments with varying salinity and nitrogen exposures. Light quantity appeared to play a large role in heterocyst production. The ability of N. spumigena to produce akinetes and heterocysts is likely to be part of the reason for its success and continual occurrence in estuarine environments low in nitrogen, such as the Gippsland Lakes, Victoria, Australia. Factors known to reduce heterocyst and akinete production will provide new insight to possible management controls for this species.  相似文献

1. Sedimentary akinetes (resting stages) may represent significant potential inocula for nuisance blooms of cyanobacteria. We studied the effects of salinity and sediment source on the germination and subsequent growth of Anabaena flos‐aquae akinetes from a shallow, tidally influenced lake. 2. Surface sediments collected from littoral and open‐water sites were used as inocula to culture A. flos‐aquae akinetes in four salinities (0.1, 2.2, 4.4 and 6.5) over 22 days. Akinete germination and development was followed by counting developmental stages every second day. 3. Filament growth, but not akinete germination, was inhibited by salinity and there were significantly fewer filaments at 6.5 than at 0.1 and 2.2. Cultures inoculated with littoral sediment had more akinetes, germlings and filaments than those inoculated with open‐water sediment. 4. Sediment is a potential source of inocula for Anabaena blooms in the lake, which potentially could develop solely from this source because germination and subsequent filament growth do not depend on the existence of an initial pelagic Anabaena population.  相似文献

Clones of the filamentous green alga Ctenocladus circinnatus Borzi were isolated from algae collected at Abert Lake (Oregon) and Mono Lake (California). Stock cultures were exposed to varied salinities of natural lake water to examine the effects on growth rate, cell form, chlorophyll a, and water content. Growth rates were reduced in both clones with increased salinity over the range 25–100 g·L?1 and were almost completely inhibited at 150 g·L?1. Chlorophyll a increased between salinities of 25 and 100 g·L?1, reflecting slower growth, higher proportions of akinetes, and smaller cell sizes as salinity increased. Tissue water content remained essentially constant from 25 to 100 g·L?1 salinity. Shorter cell dimensions with increased salinity suggest that a lower surface-to-volume ratio may reduce the potential for passive loss of cell water. Prior acclimation of stock cultures to elevated salinity provided no enhancement of growth response at any salinity. The results indicate that environmental salinity can limit the productivity and distribution of Ctenocladus in nature.  相似文献

Certain cyanobacteria, including the noxious bloom-forming species Anabaena circinalis Rabenhorst, produce thick-walled reproductive structures (akinetes) which may serve as a resting stage and ensure survival during adverse growth conditions. The effect of certain environmental variables (temperature, salinity and desiccation) on akinete germination of A. circinalis was investigated under laboratory conditions, to determine the conditions under which germination was inhibited. The overall aims were to provide a broader understanding of the life history and ecology of this species and to assess suppression of akinete germination as a potential management strategy for control of cyanobacterial blooms in the lower Murray River, Australia. The results indicated a marked threshold of temperature and salinity tolerance for germination of A. circinalis, but the latter was not within a range that could be successfully manipulated in a natural ecosystem. However, it was found that desiccation of akinetes for moderately short periods can significantly impair their capacity to germinate. It is, therefore, speculated that allowing periodic drying of shallow wetlands adjacent to the Murray River and in other areas may reduce the size of the inoculum for population growth by reducing viability of akinetes in surface sediments.  相似文献

Nodularia spumigena is one of the dominating species during the extensive cyanobacterial blooms in the Baltic Sea. The blooms coincide with strong light, stable stratification, low ratios of dissolved inorganic nitrogen, and dissolved inorganic phosphorus. The ability of nitrogen fixation, a high tolerance to phosphorus starvation, and different photo-protective strategies (production of mycosporine-like amino acids, MAAs) may give N. spumigena a competitive advantage over other phytoplankton during the blooms. To elucidate the interactive effects of ambient UV radiation and nutrient limitation on the performance of N. spumigena, an outdoor experiment was designed. Two radiation treatments photosynthetic active radiation (PAR) and PAR +UV-A + UV-B (PAB) and three nutrient treatments were established: nutrient replete (NP), nitrogen limited (−N), and phosphorus limited (−P). Variables measured were specific growth rate, heterocyst frequency, cell volume, cell concentrations of MAAs, photosynthetic pigments, particulate carbon (POC), particulate nitrogen (PON), and particulate phosphorus (POP). Ratios of particulate organic matter were calculated: POC/PON, POC/POP, and PON/POP. There was no interactive effect between radiation and nutrient limitation on the specific growth rate of N. spumigena, but there was an overall effect of phosphorus limitation on the variables measured. Interaction effects were observed for some variables; cell size (larger cells in −P PAB compared to other treatments) and the carotenoid canthaxanthin (highest concentration in −N PAR). In addition, significantly less POC and PON (mol cell−1) were found in −P PAR compared to −P PAB, and the opposite radiation effect was observed in −N. Our study shows that despite interactive effects on some of the variables studied, N. spumigena tolerate high ambient UVR also under nutrient limiting conditions and maintain positive growth rate even under severe phosphorus limitation.  相似文献

Blooms of the toxic cyanobacterium Nodularia spumigena occur in various locations worldwide, but have not been observed in Brazil until recently. Three Nodularia strains were isolated from summer blooms in experimental shrimp production ponds of Penaeus vannamei in Rio Grande, in southern Brazil; these strains were characterized by morphology, phylogeny, growth rate and toxicity. The strains were identified as N. spumigena based on the size of vegetative cells, heterocytes and akinetes under a light microscope and based on the number of gas vesicles per μm2 under a transmission electron microscope. The 16S rRNA gene sequences of the three strains showed high identity (> 99%) with N. spumigena sequences available on the NCBI database but were grouped closer in the phylogenetic tree with N. spumigena strains from Australia and USA than those from the Baltic Sea. The growth rate in batch culture varied between 0.2 and 0.6 μ?d?1 based on cell density, optical density and chlorophyll-a content. The three strains produced the hepatotoxin nodularin (ELISA plate kit) with similar toxicity values (4.8–4.9?µg?l?1). We conclude that the three isolated strains are N. spumigena with similar rates of growth and nodularin production. The presence of N. spumigena now represents a potential problem in aquaculture and estuarine environments in Brazil.  相似文献

In the Peel-Harvey estuary system, Western Australia, some 90%of riverflow and nutrient loading occurs in three winter months.Diatom blooms follow riverflow, but are replaced by blooms ofthe blue-green Nodularia spumigena Mert., especially in HarveyEstuary. By analysis of time series data from 1977–1983,it is shown that the magnitude of the Nodularia bloom in summeris related to the minimum salinity of the estuary (and hencetotal river flow), maximum phosphate concentration and totalriverine phosphorus loading, in the previous winter. The relationshipshave a predictive capacity. It is argued that diatom bloomstrap phosphorus, which is sedimented largely as faecal pellets;the phosphorus is recycled and supports Nodularia growth underwarmer conditions, and the amount available determines Nodulariabiomass. The effects of changes in diurnal light patterns, salinity, and phosphorus on nitrogen fixation (as measured by acetylene reduction) by Nodularia spumigena Mertens were examined. As well, the effects of added inorganic nitrogen on growth, nitrogen fixation and heterocyt frequencies, and changes in nitrogen fixation and heterocyst frequencies during the growth cycle of Nodularia in cultures were determined.The diurnal pattern of nitrogenase activity in Nodularia was primarily light-induced, though dark activity did occur. Nitrogenase activity following a period of darkness exceeded the normal light rate (> 90 compared to 50 nmol · C2H2 reduced · ml–1 · h–1). Nitrogen fixation was reduced by high and very low salinities (5 to 10 was the optimum range), and added phosphorus stimulated nitrogenase in P-starved cells. Added nitrogen (ammonium or nitrate) had no effect on the growth of Nodularia, but in short term studies, ammonium completely inhibited nitrogenase activity. Heterocyst frequencies were greatest in the log phase of growth (to 40 per mm). During stationary phase, nitrogenase activity was negligable.  相似文献

Effects of salinity on seed germination and growth of young (1 month old) and old (2-year old) seedlings of Excoecaria agallocha were investigated. Combined effects of salinity and nutrient level were also examined on old seedlings. Seed germination was best at 0 and 5 psu salinity. 15 psu salinity significantly delayed root initiation and decreased final establishment rate. All seeds failed to establish at 25 psu salinity. Young seedlings performed best at 0 and 5 psu, but growth was stunned at 15 psu, and all seedlings died within 90 days at 25 psu. Old seedlings grew best at salinities below 5 psu and they survived the whole cultivation at 25 psu. This indicated that E. agallocha increased salt tolerance over time. Gas exchange was significantly compromised by salinities above 15 psu but evidently promoted by high nutrient. Proline accumulated considerably at high nutrient, and its contents increased from 0 to 15 psu but decreased at 25 psu salinity. Lipid peroxidation was aggravated by increasing salinity beyond 15 psu but markedly alleviated by nutrient addition. These responses indicated that E. agallocha was intolerant to high salinity but it can be greatly enhanced by nutrient addition.  相似文献

Akinetes of a clonal culture of Anabaena circinalis Rabenhorst from Mt. Bold reservoir (eutrophic), South Australia, were isolated and the effects of light, phosphorus, and nitrogen availability on their germination were investigated. Light was required but there was no significant difference in percentage of germination after 72 h if akinetes were incubated in ASM-1 medium at irradiances of 15, 30, or 50 μmol.m-2.s-1. Maximum akinete germination occurred by 48 h. Nitrogen was not required, as 88% of akinetes germinated in the flasks without combined nitrogen added to the medium and without N2 in the air. NH4+-N at 28 mg N.L-1 completely suppressed germination, whereas 28 mg NO3 N.L-1 had no effect relative to the controls without nitrogen. Phosphorus was required, and at 48 h percentage of germination in the flasks with 0.6 mg P.L-1 added (78%) was significantly greater than in the flasks with 0.06 P.L-1 (58%) and 0 mgP.L-1 (24%) added. Germlings in the 0 mg P.L-1 flasks were only 2–4 cells long and stunted in appearance, whereas germlings at all other P concentrations were 8–16 cells long. It is likely that the isolation process exposed some akinetes to intracellular phosphorus released from lysing vegetative cells, but this was insufficient to allow normal development in the 0 mg P.L-1 flasks. The plot of percentage of germination vs. initial phosphorus concentration, in the medium showed a relationship analogous to Michaelis-Menten nutrient uptake kinetics, suggesting that a specific membrane-bound enzyme system(s) is involved, with phosphorus as the substrate. Question: In seeds which are regularly consumed by waterbirds in the field, how does gut‐passage modify their response to salinity gradients? Location: Doñana National Park salt marsh, south‐west of Spain. Methods: Seeds of Scirpus litoralis and Scirpus maritimus were collected and force fed to mallards (Anas platyrhynchos). Both the ingested seeds (passage) and non‐ingested seeds (controls) were exposed, in germination chambers, to a salinity range similar to that observed in the field (0–32 dS/m). After 30 days, the total percentage germination, the duration of the dormancy period and the germination speed were computed. The response of the different germination parameters to ingestion and salinity was analyzed using generalized lineal models. Recovery tests on seeds that did not germinate in the various treatments and tests of the effect of ingestion on the intrinsic variability in seed response were also performed. Results: An increase in salinity reduced germinability and increased the length of dormancy, while gut pas sage increased the intrinsic variability of the temporal seed response in both species. In S. litoralis there was a significant interaction between the effects of salinity and passage on germination rate. Passage increased germination rate at low salinities (≤2 dS/m) but decreased it at high salinities (≥4 dS/m). Conclusion: Gut‐passage by ducks significantly changes seed response to salinity. The outcome of plant‐animal interactions can be influenced by environmental gradients. Studies of germination in response to gut passage that do not take such gradients into account may produce misleading results.  相似文献

Effects of salinity, temperature and their interactions on the rate and final percentage of germination were evaluated for two populations (Msarref, Oued dkouk) of the invasive glycophyte Lotus creticus Linné, grown under arid environmental conditions of the Tunisia. Seeds that were not treated with NaCl germinated well in a wide range of temperatures. For both populations, maximum germination occurred in distilled water at 25°C and lowest germination for all salinities was at 35°C. Germination was substantially delayed and significantly reduced with an increase in NaCl to levels above 300 mm . Compared to the Oued dkouk population, final germination and germination rate of the Msarref population was completely inhibited at 300 mm NaCl. The interactive effect of temperature and NaCl concentration on final germination and germination rate was significant (P < 0.01), indicating that the germination response to salinity depended on temperature. The inhibition of Oued dkouk population seed germination at high salt concentration was mostly due to osmotic effects while ionic effects were noted at Msarref population. The germination behaviour of the Oued dkouk population would therefore imply adaptive mechanisms to saline environments, while in the Msarref population such mechanisms seem to be absent. Since seed germination is more sensitive to salinity stress than the growth of established plants, the greater tolerance to salinity of Oued dkouk population would be an adaptive feature of this population to saline environment.  相似文献

The effects of salinity on the growth and valve morphology of five benthic estuarine diatoms (Nitzschia pusilla, N. frustulum, N. palea, N. filiformis var. conferta and Eolimna subminuscula), isolated from both freshwater and brackish/marine habitats, were investigated. The four Nitzschia strains grew well over a broad salinity range, though some (N. pusilla, N. frustulum) showed a broader salinity range tolerance (from fully saline down to at least 9.5 ppt) than others (N. palea, N. filiformis var. conferta had reduced growth at salinities of 16 ppt and above). Salinity significantly affected the valve morphology of the five strains studied. However, there was no consistent pattern in either the morphological characters affected or the direction of the effects. Although significant, the effects of salinity on valve morphology were very small and therefore it seems that the taxonomic usefulness of some of the classical taxonomical characters is not undermined.  相似文献

2014年4-11月,选择黄河入海口北部滨岸高潮滩的碱蓬湿地为研究对象,基于野外原位氮负荷增强模拟试验(N0,无额外氮输入;N1,低氮输入;N2,中氮输入;N3,高氮输入),获取相应的不同氮基质种子(S0,S1,S2和S3),以研究其发芽率以及幼苗生长状况对不同盐分胁迫和氮浓度交互作用的响应。结果表明,不同氮负荷影响下碱蓬成熟种子中的氮含量整体表现为S2S0S1S3,中氮输入更利于种子中氮养分的累积。盐分和氮交互作用下4种氮基质种子的发芽率总体表现为S2S1S0S3(P0.05),S2在不同盐分胁迫下的发芽率最高,幼苗的生长状况也最好。随着盐分的增加,4种氮基质种子的发芽率及幼苗生长状况均受到一定程度的抑制,但较低的盐分有助于其幼苗长度的增长,且随着氮输入量的增加这种抑制作用可得到一定程度缓解。盐分胁迫、氮浓度和种子类型作为单独因素出现时对碱蓬的发芽率、幼苗长度、鲜重和干重均产生显著影响,除幼苗长度受氮浓度和盐分胁迫交互作用的影响达到显著水平外(P0.05),其他因子交互作用对诸生态指标的影响并不明显。研究发现,不同氮输入处理不仅改变了原生环境碱蓬种子的氮含量,而且也使这些具备不同氮基质的种子对不同盐分胁迫与氮浓度环境具有不同的生态适应对策,中氮输入下的碱蓬种子(S2)无论在萌发率还是在幼苗生长状况上均优于其他氮基质的种子。未来,随着黄河口新生湿地氮养分供给的不断增加,当湿地氮养分达到中氮水平时,将更有利于碱蓬种子的萌发以及幼苗的生长,当氮养分达到更高水平时,碱蓬种子的萌发以及幼苗生长可能会受到一定程度的抑制。  相似文献

By increasing water use efficiency and carbon assimilation, increasing atmospheric CO2 concentrations could potentially improve plant productivity and growth at high salinities. To assess the effect of elevated CO2 on the salinity response of a woody halophyte, we grew seedlings of the mangrove Avicennia germinans under a combination of five salinity treatments [from 5 to 65 parts per thousand (ppt)] and three CO2 concentrations (280, 400 and 800 ppm). We measured survivorship, growth rate, photosynthetic gas exchange, root architecture and foliar nutrient and ion concentrations. The salinity optima for growth shifted higher with increasing concentrations of CO2, from 0 ppt at 280 ppm to 35 ppt at 800 ppm. At optimal salinity conditions, carbon assimilation rates were significantly higher under elevated CO2 concentrations. However, at salinities above the salinity optima, salinity had an expected negative effect on mangrove growth and carbon assimilation, which was not alleviated by elevated CO2, despite a significant improvement in photosynthetic water use efficiency. This is likely due to non‐stomatal limitations to growth at high salinities, as indicated by our measurements of foliar ion concentrations that show a displacement of K+ by Na+ at elevated salinities that is not affected by CO2. The observed shift in the optimal salinity for growth with increasing CO2 concentrations changes the fundamental niche of this species and could have significant effects on future mangrove distribution patterns and interspecific interactions.  相似文献

The purpose of the study was to compare the primary plankton productivities of lakes of different salinities and to determine the causative factors involved in their production rates. Four lakes (specific conductivity —mS cm−1 at 18°C) were initially chosen: Coragulac (9), Red Rock (25), Corangamite (38), Pink (250). Sampling and production measurements were made every two to three weeks. Three lakes were dominated by specific phytoplankton blooms: Red Rock (Anabaena spiroides), Corangamite (Nodularia spumigena). Pink (Dunaliella salina). Coragulac Lake had more diverse populations. Red Rock Tarn had some of the highest production values ever recorded. Extremely high soluble phosphate and inorganic carbon concentrations were the most important causative factors. Pink Lake had very low production rates. High salinity and low nutrient concentrations were limiting factors. The other lakes were intermediate in production and nutrient levels. Zooplankton populations were also determined.  相似文献

Toxic Pseudo-nitzschia australis strains isolated from French coastal waters were studied to investigate their capacity to adapt to different salinities. Their acclimation to different salinity conditions (10, 20, 30, 35, and 40) was studied on growth, photosynthetic capacity, cell biovolume, and domoic acid (DA) content. The strains showed an ability to acclimate to a salinity range from 20 to 40, with optimal growth rates between salinities 30 and 40. The highest cell biovolume was observed at the lowest salinity 20 and was associated with the lowest growth rate. Salinity did not affect the photosynthetic activity; Fv/Fm values and the pigment contents remained high with no significant difference among salinities. An enhanced production of zeaxanthin was, however, observed in the late stationary and decline phases in all cultures except for those acclimated to salinity 20. In terms of cellular toxin content, DA concentrations were 2 to 3-fold higher at the lowest salinity (20) than at the other salinities and were combined with a low amount of dissolved DA. The fact that P. australis accumulate more DA per cell in less saline waters, illustrates that climate-related changes in salinity may affect Pseudo-nitzschia physiology through direct effects on growth, physiology, and toxin content.  相似文献

Dietary supplementation with plant-derived polyphenolic compounds such as the flavonoids epigallocatechin-3-gallate (EGCG), quercetin and resveratrol can result in a beneficial effect on degenerative disease processes through both radical scavenging and activation of cellular ion channels. Preliminary investigations have shown that extracts from the halophyte species Carpobrotus rossii have high in vitro antioxidant and in vivo low-density-lipoprotein-lowering activities. In this study, we have investigated the environmental conditions responsible for inducing flavonoid production in C. rossii in an attempt to maximise production of these compounds. Both field surveys and controlled glasshouse experiments were conducted. Flavonoid production appears to be related to conditions known to cause oxidative stress in plants such as exposure to excessive light, reduced water availability, and low soil potassium levels. Flavonoid production was minimal under salinity levels optimal for C. rossi growth (around 50 mM NaCl) and increased dramatically at sub- and supra-optimal salinities. Flavonoids were clearly concentrated in metabolically active palisade mesophyll tissue rather than the spongy parenchyma. Non-optimal (outside 50–100 mM range) NaCl levels significantly increased flavonoid concentration on a per leaf basis. However, the reduction in biomass production at sub and supra optimal salinities diminished absolute flavonoid production per plant. As a result, saline conditions favouring optimal plant growth appear to be most suitable for maximising production of flavonoids in C. rossii.  相似文献

Seeds and seedlings of the halophyte Atriplex patula were exposed to 0–2% NaCl to determine the effect of salt stress on germination and growth. Seeds germinated and plants survived and grew in solutions of up to 2.0% NaCl. Both seed germination and dry mass production were negatively affected by increased salinity. Dry mass production declined to 1% of controls and seed germination to 17% of controls in the 2% NaCl treatments, indicating that seeds were less inhibited than growing plants. Also, recovery treatments indicated that high salinity did not permanently injure seeds. Percent ash, and Na+ and Cl ions increased in shoots with each salt increment, while the K+ ion content decreased sharply. Atriplex patula is a facultative halophyte, and is limited to low and moderately saline sites because both seed germination and growth are severely reduced at salinities > 1% NaCl.  相似文献
