Somatic embryogenesis (SE) is expected to play an important role in the future of US forests by providing increased productivity, sustainability, and uniformity. For broad scale implementation to occur, SE technology must work with a variety of genetically diverse trees. Douglas fir (Pseudotsuga menziesii (Mirb) Franco) is the dominant tree in the Pacific Northwest and has great economic and recreational value. We have developed a highly effective medium for initiation of embryogenic tissue of Douglas fir that contains ABA, biotin, brassinolide, folic acid, MES, pyruvic acid and can be used as a gelled medium or in a gelled-liquid medium overlay system. When tested with many high-value crosses over 2 years, initiation tests averaged initiation in the range of 40–57%. Additionally, a time- and labor-saving tetrazolium chloride embryo staining technique was developed to evaluate seed health and screen out seed sources likely to perform poorly in the initiation process.  相似文献

Summary Effects of P fertilizers on growth of Douglas fir (Pseudotsuga menziesii var.menziesii (Mirb.) Franco.) seedlings were examined in pots and nursery beds. In pot experiments levels of P equivalent to 300 kg/ha were adequate for maximum growth over 14–18 weeks and resulted in available soil P levels of 80 ppm after 15 weeks' growth. Maximum growth in pots was obtained with shoot P concentrations of 0.18%–0.20%, with higher values at lower temperatures, but the optimum concentration for one-year old (1-0) nursery seedlings was 0.16% P. Growth of seedlings was greatly restricted at a soil temperature of 5°C and an air temperature of 12°C. At a soil temperature of 10°C and an air temperature of 14°C seedling P requirement was greater than at soil and air temperatures of 20°C.Comparison showed that monammonium phosphate was more effective than calcium superphosphate in stimulating growth in pots and nursery. Triple superphosphate was also effective in the nursery. Diammonium phosphate, potassium dihydrogen phosphate and phosphoric acid had no advantages as P sources in the nursery. Available P levels of 100–130 ppm, in the loamy sand and sandy loam nurseries studied, and needle P concentrations of 0.18%, when sampled in October, were associated with maximum growth of two-year old (2-0) seedlings.P fertilization decreased root/shoot ratio, but did not alter the allometric relationship of shoot to root. Improving P status from a low level increased root growth capacity in 2-0 seedlings and P fertilization of potted seedlings increased dry weight/height ratio. Uptakes per seed bed ha of 236 kg N, 31 kg P, 81 kg K and 73 kg Ca by 2-0 seedlings were comparable with, or greater than, uptake rates of agricultural crops. Recoveries of 6–11% of P from fertilizer were recorded in the nursery.  相似文献

Douglas fir seedlings grown under aseptic conditions in a peat-vermiculite substrate were inoculated with four pairs of ectomycorrhizal fungi to assess the relative inoculum dosages needed to establish two mycorrhizal fungi simultaneously in the same root system. The dual fungal combinations tested were: Pisolithus arhizus + Rhizopogon subareolatus, P. arhizus + R. roseolus, Laccaria bicolor + P. arhizus and L. bicolor + R. subareolatus. A total of 12 ml of inocula per plant was applied at the rates: 0+12, 3+9, 6+6, 9+3, 12+0, and 0+0 (v+v) for each combination. After 3 months growth, the number of mycorrhizas and uninfected short roots as well as the total plant biomass produced were recorded. Inoculations were successful with the fungal combinations P. arhizus + R. subareolatus and L. bicolor + P. arhizus. Plants developed P. arhizus and R. subareolatus mycorrhizas only at the rate 9Pa + 3Rs; at other rates tested, only monospecific mycorrhizas were formed. Plants developed L. bicolor and P. arhizus mycorrhizas at the three rates containing both fungi. L. bicolor behaved as an aggressive root colonizer and its level of root colonization remained constant at increasing rates of P. arhizus inoculum. L. bicolor displaced R. subareolatus at all inocula rates. P. arhizus displaced R. roseolus except at the rate 3Pa + 9Rr, with only a low number of mycorrhizas formed by either fungus. Total plant biomass was significantly increased by the presence of any fungal combination up to four times the values for uninoculated controls. P. arhizus and R. subareolatus were more effective in promoting plant growth and stimulating short root formation than either L. bicolor or R. roseolus.  相似文献

Summary A factorial cross design was used to evaluate the influence of differences among pollen donors and recipients on variation in stylar attrition of pollen tubes in self-fertile plants of Petunia hybrida. Pollinations to flower buds were used to reduce the inhibitory ability of the style and these crosses were compared with flower pollinations to assess the degree of stylar influence on pollen fertilization ability. There was less pollen tube attrition after bud pollinations than after flower pollinations, indicating that styles of buds were less able to inhibit pollen tubes. The variance component for plants acting as pollen donors tended to be greater after flower pollinations than after bud pollinations. The lower variance in male success after bud pollinations indicates that differences among pollen donors after flower pollinations were due to stylar inhibition of pollen rather than differences in pollen vigor. Since the level of variation in pollen growth after pollination to flowers was greater among clones than among ramets within clones, the differences in pollen fertilization ability are probably genetically based.  相似文献

Purification and properties of glutamine synthetase from Douglas fir roots   总被引:2,自引:0,他引:2  
Glutamine synthetase (GS. EC 6.3.1.2) was purified to apparent electrophoretic homogeneity from roots of Pseudotsuga menziesii (Mirb) Franco by a three-step procedure involving diethylaminoethyl (DEAE)-Trisacryl chromatography, affinity chromatography on Matrex Gel Red A. and preparative polyacrylamide gel electrophoresis. The enzyme was purified 40-fold with a 16% recovery. The native enzyme had a molecular mass of 460 ± 5 kDa as estimated by gel filtration, interpolation of the Ferguson plots and non-denaturing gradient-PAGE. It was composed of two different subunits of 54 and 64 kDa. Affinity constants for glutamate (Glu), glutamine (Gln), ATP and ADP were 2.6, 10.5, 0.5 and 0.083 m M . respectively. The enzyme exhibited a negative cooperativity for ammonium (Hill number of 0.7) with two Km values which were 11 and 75 μ M in the presence of ammonium concentrations lower and higher than 1.3 m M , respectively. Glycine and ADP appeared as potential inhibitors of the GS activity. The optimum pH values were 7.2 and 7.6 for the transferase and the biosynthetic assays, respectively. The enzyme lost 30% of its activity within 25 days of storage at 4°C. The optimum temperatures of activity were 40°C and 45°C for the transferase and bio-synthetic activities, respectively.  相似文献

The effects on growth and nutrient status of an increased availability of all major nutrients was studied in a 40 year old Douglas fir stand. The nutrient amounts were applied by daily sprinkling and were in fixed, optimal proportions to the estimated annual uptake of N. Irrigation was included to avoid drought stress. The nutrient applications were also done in combination with a lower NH4 load, realized under a roof that was placed above the ground. According to nutritional standards nitrogen supply was optimal and was related to the moderately high atmospheric N inputs. Nutrition of P, K and in some years Ca seemed critical.In all four studied growing seasons water additions resulted in a growth increase, of 30 to 40%. Irrigation decreased needle fall. Nutrient applications increased the needle concentrations of P and K considerably, but this did not result in a growth enhancement. Fertilization also increased K and P in shedded needles. Only minor leaching losses were calculated. The recovery in trees of fertilizer K and P was maximally 15%. Input-output budgets suggested that P was strongly retained in the soil. The decrease in NH4 load had slightly decreased N availability and soil acidification, and seemed to enhance tree growth. The use of tree growth as a parameter to evaluate the effects of an improved nutritional balance is discussed.  相似文献

A pattern recognition system was developed to classify Douglas fir somatic embryos by employing an image analysis system and two neural network based classifiers. The contour of embryo images was segmented, digitalized and converted to numerical values after the discrete and fast Fourier transformation. These values, or Fourier features, along with some other shape factors, were used for embryo classification. The pattern recognition system used a hierarchical decision tree to classify Douglas fir embryos into three normal and one abnormal embryo classes. An accuracy of greater than 80% was achieved for normal embryos. This system provides an objective and efficient method of classifying embryos of Douglas fir. It will be a useful tool for kinetic studies and process optimization of conifer somatic embryogenesis.  相似文献

Nitrogen metabolism of the needles of 40-year-old Douglas fir and Scots pine trees, growing in two forest stands on cation-poor and acidic sandy soil with a relatively high atmospheric nitrogen deposition was studied. The composition of the free amino acid (FAA) pool, the concentrations of total nitrogen and soluble protein and the activities of glutamine synthetase (GS) and glutamate dehydrogenase (GDH) were determined in the needles. An excessive nitrogen supply by a high atmospheric nitrogen deposition in both forest stands was indicated by the high concentrations of total nitrogen and the amino acids arginine, glutamic acid, glutamine and aspartic acid in control trees. In addition the effect of optimal nutrition and water supply (fertigation) on the needle nitrogen metabolism was evaluated. The total concentration of the FAA pool in needles of both tree species was lower in the fertigated than in the non-fertigated (control) trees, except for 1-year-old needles of Scots pine, in which the concentration after fertigation did not differ from the control. The lower total FAA concentration in the fertigated trees could be attributed to arginine, the concentration of which was on average 60% lower than in the control. Neither the concentration of soluble protein nor the activity of GS were influenced by fertigation. The activity of GDH in fertigated trees only differed significantly from the control in October. Scots pine needles had higher concentrations of protein (50%) and higher activities of GS (44%) and GDH (25%) than Douglas fir needles. Possible explanations for the lower vitality of Douglas fir compared to Scots pine are given.  相似文献

Douglas fir (Pseudotsuga menziesii) seedlings in two bare-root forest nurseries were inoculated with the ectomycorrhizal fungus Laccaria laccata, together or not with one of five mycorrhization helper bacteria isolated from L. laccata sporocarps or mycorrhizas and previously selected by in vitro and glasshouse screenings. With the most efficient MHB isolates, when compared to the control with no bacteria, the percent of mycorrhizal short roots was increased from 60 to 90 or from 80 to 100, depending on the nursery, with inoculation doses as low as 106 living cells per m2. A dual inoculum made of calcium alginate beads containing the two microorganisms appears to be a valuable technique for increasing the efficiency of ectomycorrhizal inoculation of planting stocks in forest nurseries.  相似文献

Two recent studies have shown a relationship between male height and number of offspring in contemporary developed-world populations. One of them argues as a result that directional selection for male tallness is both positive and unconstrained. This paper uses data from a large and socially representative national cohort of men who were born in Britain in March 1958. Taller men were less likely to be childless than shorter ones. They did not have a greater mean number of children. If anything, the pattern was the reverse, since men from higher socioeconomic groups tended to be taller and also to have smaller families. However, clear evidence was found that men who were taller than average were more likely to find a long-term partner, and also more likely to have several different long-term partners. This confirms the finding that tall men are considered more attractive and suggests that, in a noncontracepting environment, they would have more children. There is also evidence of stabilizing selection, since extremely tall men had an excess of health problems and an increased likelihood of childlessness. The conclusion is that male tallness has been selected for in recent human evolution but has been constrained by developmental factors and stabilizing selection on the extremely tall. The validity of evolutionary explanations of vocal sexual dimorphism hinges upon whether or not individuals with more sexually dimorphic voices have higher reproductive success than individuals with less dimorphic voices. However, due to modern birth control methods, these data are rarely described, and mating success is often used as a second-rate proxy. Here, we test whether voice pitch predicts reproductive success, number of children born and child mortality in an evolutionarily relevant population of hunter-gatherers. While we find that voice pitch is not related to reproductive outcomes in women, we find that men with low voice pitch have higher reproductive success and more children born to them. However, voice pitch in men does not predict child mortality. These findings suggest that the association between voice pitch and reproductive success in men is mediated by differential access to fecund women. Furthermore, they show that there is currently selection pressure for low-pitch voices in men.  相似文献

A growth chamber experiment was conducted to determine if P fertilization to enhance the P nutrition of otherwise N and P deficient Douglas fir [Pseudotsuga menziesii (Mirb.) Franco] seedlings reduces water stress in the seedlings during drought periods. Seedlings were grown in pasteurized mineral soil under well-watered conditions and fertilized periodically with a small amount of nutrient solution containing P at either of three levels: 0, 20, or 50 mg P L-1. By age 6 mo, leaf nutrient analysis indicated that N and P were deficient in control (0 mg P L-1) seedlings. The highest level of P fertilization, which doubled leaf P concentration, did not affect plant biomass, suggesting that N deficiency was limiting growth. When these seedlings were subjected to drought, there was no effect of P fertilization on leaf water potential or osmotic potential. Furthermore, P fertilized seedlings had lower stomatal conductance and net photosynthesis rate. These results indicate that enhanced P nutrition, in the presence of N deficiency, does not reduce water stress in Douglas fir seedlings during drought periods.  相似文献

Formaldehyde (HCHO) concentrations of up to 479 mmol m–3 have been reported in urban fogwater, yet little is known about the phytotoxicity of HCHO at such concentrations. We report that formaldehyde at target concentrations of 300, 600, 900 and 1200mmol m–3 reduced germination of Douglas fir [ Pseudotsuga menziesii (Mirbel) Franco.] pollen in vitro . Formaldehyde concentrations and pH in media containing pollen decreased over 25 h. These effects were not seen with heat-killed pollen nor with media lacking pollen, which may suggest uptake or detoxification by live pollen. Stress was indicated by ion leakage from pollen, which increased within 20 h in all HCHO treatments. Reduced pollen viability was indicated by TTC staining of pollen, which decreased after HCHO treatment.  相似文献
