Mulberry Leaf Metabolism under High Temperature Stress   总被引:5,自引:0,他引:5  
Effects of high temperature on the activity of photosynthetic enzymes and leaf proteins were studied in mulberry (Morus alba L. cv. BC2-59). A series of experiments were conducted at regular intervals (120, 240 and 360 min) to characterize changes in activities of ribulose-1,5-bisphosphate carboxylase (RuBPC) and sucrose phosphate synthase (SPS), photosystem 2 (PS 2) activity, chlorophyll (Chl), carotenoid (Car), starch, sucrose (Suc), amino acid, free proline, protein and nucleic acid contents in leaves under high temperature (40 °C) treatments. High temperature markedly reduced the activities of RuBPC and SPS in leaf extracts. Chl content and PS 2 activity in isolated chloroplasts were also affected by high temperature, particularly over 360 min treatment. Increased leaf temperature affected sugar metabolism through reductions in leaf starch content and sucrose-starch balance. While total soluble protein content decreased under heat, total amino acid content increased. Proline accumulation (1.5-fold) was noticed in high temperature-stressed leaves. A reduction in the contents of foliar nitrogen and nucleic acids (DNA and RNA) was also noticed. SDS-PAGE protein profile showed few additional proteins (68 and 85 kDa) in mulberry plants under heat stress compared to control plants. Our results clearly suggest that mulberry plants are very sensitive to high temperature with particular reference to the photosynthetic carbon metabolism.  相似文献

Brassinosteroids (BRs) have been proposed to increase the resistance of plants to drought stress. The effect of foliar application of 0.1 μM 24-epibrassinolide (EBR) on chlorophyll (Chl) content, photosystem 2 (PS 2) photochemistry, membrane permeability, lipid peroxidation, relative water content (RWC), proline content, and the antioxidant system in drought-stressed Chorispora bungeana plants was investigated. The results showed that polyethylene glycol (PEG) induced water stress decreased RWC, Chl content and variable to maximum Chl fluorescence ratio (Fv/Fm) less in plants pretreated with EBR than in non-pretreated plants. In addition, lipid peroxidation, measured in terms of malondialdehyde content, membrane permeability and proline content in drought-stressed plants were less increased in EBR pretreated plants, while antioxidative enzyme activities and reduced ascorbate and glutathione contents were more increased in EBR pretreated than in non-pretreated plants. Diurnal variations in relative water content (RWC), nitrate reductase (NR) and proline content (PC) were studied at 3 h intervals during a 24 h cycle in the flag leaf of wheat (Triticum vulgare, v. Kalyansona) grown under stressed and non-stressed conditions. RWC was lower in stressed plants than in non-stressed ones throughout the 24 h period. Although it was lowest at 12 h, it recovered by 15 h. Non-stressed plants maintained higher NR activity compared to those under stress. The enzyme activity during night was about the same as during day time in both types of plants. Compared to non-stressed plants, stressed ones had lower NO3? content. Proline accumulation occurred under stress conditions and had a maximum at 12–15 h. Non-stressed plants exhibited higher PC during night than day time. Changes in temperature and relative humidity were noted during the period and their influence on RWC, NR and proline was discussed.  相似文献

Abstract

Soil salinity is one of the major factors responsible for the low productivity of crop plants and has become an increasing threat for agriculture. In this context, the selection of tolerant genotype/s may be one of the remedies. Keeping this in view, the effect of NaCl (0–120 mM) stress on shoot length (SL) plant?1, area (A) leaf?1, leaf area index (LAI), fresh weight (FW) and dry weight (DW) plant?1, stomatal conductance (gs), net photosynthetic rate (P N), total chlorophyll (Chl) content, malondialdehyde (MDA) content, sensitivity rate index (SRI), leaf- nitrogen (N), potassium (K) and sodium (Na) content, leaf-K/Na ratio, nitrate reductase (NR: EC.1.6.6.1) and ATP-sulphurylase (ATP-S: EC.2.7.7.4) activities and proline (Pro) and glycinebetaine (GB) content of ten genotypes of Brassica juncea L. was studied at 55 and 65 days after sowing (DAS). NaCl treatments decreased all the above parameters, except Pro, GB, MDA, Na and SRI at both stages. Salt stress resulted in accumulation of Pro and GB, in all genotypes. The magnitude of increase in both osmolytes (Pro and GB) was higher in genotype G8 than the other genotypes. Salt stress increased MDA and Na content while it decreased Chl, N and K content and K/Na ratio, Chl content, NR and ATP-S activities in all genotypes. But the magnitude of increase in MDA and Na content and decrease in SL plant?1, A leaf?1, LAI, P N, gs, Chl content and NR and ATP-S activities in genotype G8 was more than that of other genotypes. These results suggest that the salt-tolerant genotype may have better osmotic adjustment and protection from free radicals by increasing the accumulation of Pro and GB content with overproduction of N and K and higher K/Na, NR and ATP-S activities under salinity stress.  相似文献

Cuttings from two contrasting Populus cathayana Rehder populations originating from Hanyuan (wet climate) and Ledu (dry climate) in western China were grown in a greenhouse to determine the effects of drought, shade and their interaction on the morphological and physiological traits of leaves. The dry climate population was more droughttolerant than the wet climate population, as indicated by smaller decreases in the leaf relative water content (RWC) and net photosynthetic rate (PN), as well as by greater increases in antioxidative enzyme activities and free proline content under drought. On the other hand, the negative effects of shade on leaf traits were more pronounced in the dry climate population, which suggested that the dry climate population was more light-demanding. In addition, moderate shade alleviated the drought stress of P. cathayana not only by improving the leaf RWC but also by maintaining the positive carbon balance. In contrast, severe shade aggravated drought stress as indicated by a pronounced decrease in leaf size, carbon and nitrogen contents, maximum PN, free proline content and antioxidative enzyme activities.  相似文献

Effect of grain soaking presowing in 1 mM salicylic acid (SA) and NaCl (0, 50, 100, 150 and 200 mM) on barley (Hordeum vulgare cv Gerbel) was studied. Increasing of NaCl level reduced the germination percentage, the growth parameters (fresh and dry weight), potassium, calcium, phosphorus and insoluble sugars content in both shoots and roots of 15-day old seedlings. Leaf relative water content (RWC) and the photosynthetic pigments (Chl a, b and carotenoids) contents also decreased with increasing NaCl concentration. On the other hand, Na, soluble sugars, soluble proteins, free amino acids including proline content and lipid peroxidation level and peroxidase activity were increased in the two plant organs with increasing of NaCl level. Electrolyte leakage from plant leaves was found to increase with salinity level. SA-pretreatment increased the RWC, fresh and dry weights, water, photosynthetic pigments, insolube saccharides, phosphorus content and peroxidase activity in the stressed seedlings. On the contrary, Na+, soluble proteins content, lipid peroxidation level, electrolyte leakage were markedly reduced under salt stress with SA than without. Under stress conditions, SA-pretreated plants exhibited less Ca2+ and more accumulation of K+, and soluble sugars in roots at the expense of these contents in the plant shoots. Exogenous application (Grain soaking presowing) of SA appeared to induce preadaptive response to salt stress leading to promoting protective reactions to the photosynthetic pigments and maintain the membranes integrity in barley plants, which reflected in improving the plant growth.  相似文献

Pollution of the root environment with excess of Pb retarded shoot growth, decreased chlorophyll (Chl) content and reduced Chl stability (CSI) to heat. Plants growing in Pb polluted soil accumulated much more free amino acids and less soluble sugars than the control plants. Stability of leaf membranes to heat (51 °C) or dehydration stresses (40% polyethylene glycol, 6000) decreased in response to Pb pollution where the membranes of leaf discs excised from Pb-treated plants were damaged more than those taken from plants growing in Pb free soil. Supplying kinetin ameliorated the deleterious effects of Pb pollution on the parameters tested. Kinetin-treated plants had higher Chl, soluble sugars content and produced more biomass in their shoots. Also, kinetin increased leaf membrane stability especially in Pb-treated plants, effectively protected chlorophyll degradation by heat and increased Chl a and b stability index; the most effective concentration was 10 mg L–1. The effects of Pb and kinetin as well as their interaction (Pb × Kin) on the parameters tested were statistically significant. Applied kinetin had a dominant role (as indicated by 2) in affecting shoot growth, soluble sugars, Chl a and b contents, stability of leaf membranes to dehydration stress as well as the Chl stability index. Pb had a dominant role on total free amino acids (TAA) and leaf relative water content (RWC). The interaction between Kin × Pb influenced the stability of leaf membranes to heat stress in a major way.  相似文献

Heat stress (HS) is the major constraint to crop productivity worldwide. The objective of the present experiment was to select the tolerant and sensitive genotype(s) on the basis of morpho-physiological and biochemical characteristics of ten Vicia faba genotypes. These genotypes were as follows: Zafar 1, Zafar 2, Shebam 1, Makamora, Espan, Giza Blanka, Giza 3, C4, C5 and G853. The experimental work was undertaken to study the effects of different levels of temperature (control, mild, and modest) on plant height (PH) plant−1, fresh weight (FW) and dry weight (DW) plant−1, area leaf−1, content of leaf relative water (RWC), proline content (Pro) and total chlorophyll (Total Chl), electrolyte leakage (EL), malondialdehyde level (MDA), hydrogen peroxide (H2O2), and activities of catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD) enzymes. HS significantly affected growth performance of all genotypes. However, the magnitude of reduction in genotypes ‘C5’ was relatively low, possibly due to its better antioxidant activities (CAT, POD and SOD), and accumulation of Pro and Total Chl, and leaf RWC. In the study, ‘C5’ was noted to be the most HS tolerant and ‘Espan’ most HS sensitive genotypes. It was concluded that the heat-tolerant genotypes may have better osmotic adjustment and protection from free radicals by increasing the accumulation of Pro content with increased activities of antioxidant enzyme.  相似文献

The influence of arbuscular mycorrhizal (AM) fungus Glomus deserticola (Trappe and John) on plant growth, nutrition, flower yield, water relations, chlorophyll (Chl) contents and water-use efficiency (WUE) of snapdragon (Antirhinum majus cv. butterfly) plants were studied in potted culture under well-watered (WW) and water-stress (WS) conditions. The imposed water stress condition significantly reduced all growth parameters, nutrient contents, flower yield, water relations, and Chl pigment content and increased the electrolyte leakage of the plants comparing to those of nonstressed plants. Regardless of the WS level, the mycorrhizal snapdragon plants had significantly higher shoot and root dry mass (DM), WUE, flower yield, nutrient (P, N, K, Mg, and Ca) and Chl contents than those nonmycorrhizal plants grown both under WW or WS conditions. Under WS conditions, the AM colonization had greatly improved the leaf water potential (??w), leaf relative water content (RWC) and reduced the leaf electrolyte leakage (EL) of the plants. Although the WS conditions had markedly increased the proline content of the leaves, this increase was significantly higher in nonmycorrhizal than in mycorrhizal plants. This suggests that AM colonization enhances the host plant WS tolerance. Values of benefit and potential dry matter for AM-root associations were highest when plants were stressed and reduced under WW conditions. As a result, the snapdragon plants showed a high degree of dependency on AM fungi which improve plant growth, flower yield, water relations particularly under WS conditions, and these improvements were increased as WS level had increased. This study confirms that AM colonization can mitigate the deleterious effect of water stress on growth and flower yield of the snapdragon ornamental plant.  相似文献

We investigated the effects of jasmonic acid (JA) and gamma irradiation on the growth and metabolic responses to salt stress in rice (Oryza sativa L.) plants. The relative growth rate (RGR), relative water content (RWC), and chlorophyll (Chl) content were lower in NaCI-treated plants than in the control, whereas the malondialdehyde content (MDA), electrolyte leakage (EL), and contents of proline and abscisic acid (ABA) were higher in the treated plants. When induced by the salt stress, those effects, however, were somewhat alleviated by the application of JA or gamma irradiation. The most significant response was manifested by the proline content, with relatively lower values for alleviation being recorded for the contents of RGR, RWC, Chl, and MDA, as well as EL. Moreover, although total Chl content was not significantly influenced by JA or gamma irradiation in plants under salt stress, an increase in the level of Chl a resulted in a markedly changed ratio of Chl a/b. The degree of alleviation, in terms of growth and metabolic responses, was more extensive for JA-treated plants than for those exposed to gamma irradiation.  相似文献

The ameliorative effect of salicylic acid (SA: 0.5 mM) on sunflower (Helianthus annuus L.) under Cu stress (5 mg l−1) was studied. Excess Cu reduced the fresh and dry weights of different organs (roots, stems and leaves) and photosynthetic pigments (chlorophyll a, b and carotenoids) in four-week-old plants. There was a considerable increase in Chl a/b ratio and lipid peroxidation in both the roots and leaves of plants under excess Cu. Soluble sugars and free amino acids in the roots also decreased under Cu stress. However, soluble sugars in the leaves, free amino acids in the stems and leaves, and proline content in all plant organs increased in response to Cu toxicity. Salicylic acid (SA) significantly reduced the Chl a/b ratio and the level of lipid peroxidation in Cu-stressed plants. Under excess Cu, a higher accumulation of soluble sugars, soluble proteins and free amino acids including proline occurred in plants treated with 0.5 mM SA. Exogenous application of SA appeared to induce an adaptive response to Cu toxicity including the accumulation of organic solutes leading to protective reactions to the photosynthetic pigments and a reduction in membrane damage in sunflower.  相似文献

Three clones of tobacco transformed with the T-DNA of Agrobacterium rhizogenes Ri plasmid A4b cultivated in vitro on a solid agar medium neither showed pronounced morphological diversity nor significantly differed in chlorophyll (Chl) contents from control plants. However, the transformation caused a 27 to 83 % decay in leaf oxygen evolution and in both ribulose-1,5-bisphosphate carboxylase (RuBPC) and phosphoenolpyruvate carboxylase (PEPC) activities. Therefore, the transformation brought about a reduction of active PEPC as well as activable RuBPC amounts in plant tissues. Individual clones did not mutually differ. In tobacco transformed with A. rhizogenes strain TR101 and grown in soil only the mean leaf area tended to reduce. Chl contents, Chl a/b ratio, oxygen evolution, and activities of both RuBPC and PEPC were insignificantly affected by the transformation.  相似文献

The 7- to 8-day-old barley (Hordeum vulgare L.) seedlings grown in KNO3 solutions (1-40 mM) were characterized by the substrate activation of nitrate reductase (NR) in the apical leaf segments (1–2 cm in length), as well as by stimulated growth, broadened leaf blades, and by vigorously developed system of shortened roots. When the seedlings were grown in the presence of 20 mM KNO3, the ability of leaf segments to generate superoxide anion radical remained at the level typical of control plants grown in water. The content of 5-aminolevulinic acid (ALA) in plants grown in the presence of 20 mM KNO3 was 2.2–2.4 times higher than in control plants. The plants grown in the presence of nitrate had an elevated content of chlorophylls a and b, heme, and protein (by 42%). At the same time, the proline content was almost twofold lower than in control plants, which was due to substantial reduction (by 40%) in activity of Δ1-pyrroline-5-carboxylate synthetase (P5CS). It is concluded that the substrate activation of NR by KNO3 under normal growth conditions results in predominant utilization of glutamic acid (the primary product of inorganic nitrogen assimilation) for biosynthesis of tetrapyrroles and protein amino acids at the expense of inhibition of proline synthesis. When barley seedlings were grown in 150 mM NaCl solution, the plant growth and the root system development were suppressed to the levels of 63 ± 6% and 61 ± 11% of the control values, respectively. In the apical leaf tissues of plants adapted to NaCl, there was a slight decrease in the total NR activity (by 10%), a significant reduction in protein content (by 32%), and a parallel increase in the content of ALA (by a factor of 4.3), chlorophylls, heme, carotenoids, proline (2.2-fold) and P5CS (1.6-fold) with respect to the control values. It is proposed that the accumulation of ALA and proline under salinity-induced suppression of nitrogen assimilation results from the predominant allocation of glutamate for biosyntheses of ALA and proline at the expense of inhibition of growth-related processes requiring intense protein synthesis. The substrate activation of NR by KNO3 under salinity conditions was associated with prevailing allocation of the assimilated nitrogen for synthesis of proline and protein amino acids, which reinforced plant cell protection against salinity and stimulated plant growth.  相似文献

Responses of a salt tolerant and a normal population of an oilseed crop,Eruca sativa Mill. were assessed after four weeks growth in sand culture salinized with 0 (control), 100, 200, or 300 mol m?3 NaCl. The salt tolerant plants produced significantly greater dry biomass than the normal population. The populations did not differ significantly in leaf osmotic potential, relative water content and leaf soluble proteins. However, the tolerant population accumulated significantly greater amounts of soluble sugars, proline and free amino acids in the leaves compared with the non-tolerant population. It is established that leaf soluble sugars, proline, and free amino acids are important components of salt tolerance inEruca sativa.  相似文献

The experiments were carried out with maize (Zea mays L.) seedlings, hybrid Kneja 530, grown hydroponically in a growth chamber. Twelve-day-old plants were foliar treated with putrescine, N1-(2-chloro-4-pyridyl)-N2-phenylurea (4-PU-30), and abscisic acid (ABA) at concentrations of 10−5 m. Twenty-four hours later the plants were subjected to a water deficit program, induced by 15% polyethylene glycol (PEG; molecular weight, 6,000). Three days after drought stress half of the plants were transferred to nutrient solution for the next 3 days. The effects of the water shortage, rewatering, and plant growth regulator (PGR) treatment on the fresh and dry weights, leaf pigment content, proline level, relative water content (RWC), transpiration rate, activities of catalase and guaiacol peroxidase, hydrogen peroxide content, and level of the products of lipid peroxidation were studied. It was established that the application of PGRs alleviated to some extent the plant damage provoked by PEG stress. At the end of the water shortage program the plants treated with these PGRs possessed higher fresh weight than drought-subjected control seedlings. It was found also that putrescine increased the dry weight of plants. Under drought, the RWC and transpiration rate of seedlings declined, but PGR treatment reduced these effects. The accumulation of free proline, malondialdehyde, and hydrogen peroxide was prevented in PGR-treated plants compared with the water stress control. The results provided further information about the influence of putrescine, 4-PU-30, and ABA on maize plants grown under normal, drought, and rewatering conditions. To understand the ecophysiological adaptation mechanisms of Calligonum roborovskii to altitude variation, this study analyzed chlorophyll a (Chl a), chlorophyll b (Chl b), Chl (a + b), carotenoid (Car), malondialdehyde (MDA), ascorbate (AsA), proline (Pro), membrane permeability (MP), reactive oxygen species (ROS), specific leaf area (SLA), leaf mass per area (LMA), leaf nitrogen content based on mass (Nmass), and the activities of peroxidase (POD), catalase (CAT), superoxide dismutase (SOD), and ascorbate peroxidase (APX) in leaves of plants inhabiting different altitudes (A1: 2100 m, A2: 2350 m, A3: 2600 m) on the northern slope of the Kunlun Mountains. The results showed that Chl a, Chl b, Chl (a + b), SLA, Nmass, and the activity of CAT increased with increasing altitude. LMA, MP, MDA, Car, Pro, AsA, O2, H2O2 and the activities of SOD, POD, and APX decreased with increasing altitude. The test results also showed that, changes in venvironmental factors along an altitudinal gradient are not obvious. Soil water content is the main ecological factor. With increasing altitude, soil water content increased significantly. More non-enzymatic and enzymatic antioxidants played an important role in eliminating intracellular ROS. They kept the cell membrane in a stable state and ensured the normal growth of C. roborovskii.  相似文献

In mature and young leaves of sunflower (Helianthus annuus L. cv. Catissol-01) plants grown in the greenhouse, photosynthetic rate, stomatal conductance, and transpiration rate declined during water stress independently of leaf age and recovered after 24-h rehydration. The intercellular CO2 concentration, chlorophyll (Chl) content, and photochemical activity were not affected by water stress. However, non-photochemical quenching increased in mature stressed leaves. Rehydration recovered the levels of non-photochemical quenching and increased the Fv/Fm in young leaves. Drought did not alter the total Chl content. However, the accumulation of proline under drought was dependent on leaf age: higher content of proline was found in young leaves. After 24 h of rehydration the content of proline returned to the same contents as in control plants.  相似文献

The effect of benzyladenine (BA) and ascorbic acid (AA) on relative water content, proline accumulation, net photosynthetic rate (PN), chlorophyll (Chl) content and nitrate reductase (NR) activity, under sufficient water supply and moisture stress was studied in senna (Cassia angustifolia Vahl.) at seedling, vegetative, flowering and pod formation stages. AA treatment resulted in a higher accumulation of proline at all the stages of growth. Both BA and AA enhanced PN, Chl content and NR activity, and ameliorate the negative effect of water stress.  相似文献

The salt tolerance of peanut (Arachis hypogaea L.) seedlings was evaluated by analyzing growth, nutrient uptake, electrolyte leakage, lipid peroxidation and alterations in levels of some organic metabolites under NaCl stress. The plant height, leaf area and plant biomass decreased significantly in salt-treated seedlings as compared with control. The relative water content (RWC %) of leaf decreased by 16 % at high concentrations of NaCl. There was an increase in the lipid peroxidation level and decrease in the electrolyte leakage at high concentrations of NaCl. The total free amino acid and proline contents of leaf increased by 5.5- and 43-folds, respectively in 150 mM NaCl-treated plants as compared with control. Total sugar and starch content increased significantly at high concentrations of NaCl. Chl a, Chl b, total chlorophyll and carotenoid contents decreased significantly at high salinity. Na+ contents of leaf, stem and root increased in dose-dependent manner. K+ content remained unaffected in leaf and root and decreased in stem by salinity. The results from present study reveal that the peanut plants have an efficient adaptive mechanism to tolerate high salinity by maintaining adequate leaf water status associated with growth restriction. In order to circumvent the stress resulting from high salinity, the levels of some organic metabolites such as total free amino acids, proline, total sugars and starch were elevated. The elevated levels of the organic metabolites may possibly have some role in maintenance of osmotic homeostasis, nutrient uptake and adequate tissue water status in peanut seedlings under high-salinity conditions.  相似文献

A series of experiments were conducted to characterize the water stress-induced changes in the activities of RuBP carboxylase (RuBPCO) and sucrose phosphate synthase (SPS), photosystem 2 activity, and contents of chlorophylls, carotenoids, starch, sucrose, amino acids, free proline, proteins and nucleic acids in mulberry (Morus alba L. cv. K-2) leaves. Water stress progressively reduced the activities of RuBPCO and SPS in the leaf extracts, the chlorophyll content, and PS2 activity in isolated chloroplasts. Plants exposed to drought showed lower content of starch and sucrose but higher total sugar content than control plants. While the soluble protein content decreased under water stress, the amino acid content increased. Proline accumulation (2.5-fold) was noticed in stressed leaves. A reduction in the contents of DNA and RNA was observed. Reduced nitrogen content was associated with the reduction in nitrate reductase activity. SDS-PAGE protein profile showed few additional proteins (78 and 92 kDa) in the water stressed plants compared to control plants.  相似文献
