Seeds of two lettuce cultivars (Lactuca sativa L., cv. Meikoninginand cv. Grand Rapids) were hermetically stored with constantmoisture contents ranging between 3.6 and 17.9 per cent (freshweight basis) at constant temperatures ranging between 5 and75 °C. The decline with time in percentage germination andpercentage normal seedlings was determined for each storagetreatment. The data were fitted to an equation which containsthe constants: K1, the probit of the initial percentage germinationor normal seedlings; KE, a species constant; CW, the constantof a logarithmic moisture term; CH, the constant of a lineartemperature term and CQ, the constant of a quadratic temperatureterm. Regression analysis of data from storage periods up to5.5 years at temperatures of 5–75 °C and seed moisturecontents of 3.6–13.6 per cent yielded the following values:KE= 8.218, CW=4.797±0.163, CH=0.0489±0.0050 andCQ=0.000365±0.000056. Although this equation consistentlyprovided a better fit, simplified equations, assuming eithera log-linear relationship between seed longevity and temperature,or a log-linear relationship between seed longevity and bothmoisture content and temperature, accounted for more than 94per cent of the variation at the restricted temperature rangeof 5–40 °C. Longevity of the same seed lots at sub-zero temperatures (–5,–10 and –20 °C) was studied in separate tests.Freezing damage, resulting in abnormal seedlings in the germinationtest, occurred at –20 °C when the moisture contentof the seeds exceeded 12 per cent. No decline in percentagenormal seedlings was observed after a storage period of 18 monthsor longer at –20 °C, provided the seed moisture contentdid not exceed 9.5 per cent. For seeds stored at –5 and–10 °C with 9.6–12.5 per cent moisture content,the observed rate of decline of percentage normal seedlingswas adequately predicted by the viability equation, using theabove values for the constants. This suggests that for low moisturecontents the viability equation can be applied to estimate longevityat sub-zero temperatures. Agave deserti, a semelparous, Crassulacean acid metabolism perennialoccurring in the northwestern Sonoran Desert, propagates primarilyvegetatively by ramets produced on rhizomes that extend lessthan 10 cm from the base of a parent plant. Carbon translocationfrom parents to ramets, measured after exposing leaves to 14CO2,was essentially complete in 7 d, with parents exporting 3·3%of their assimilated carbon to ramets. Shading ramets belowlight compensation for 6 weeks more than doubled the amountof carbon exported from the parent to shaded ramets, comparedwith unshaded ramets. The total amount of carbon imported bya ramet from its parent was independent of the mass of the ramet.Although the net movement of carbon is expected to be towardsthe ramets, parents also received carbon from labelled ramets,indicating bidirectional translocation. The physiological integrationof parents and ramets allows ramets to draw upon the reservesof the parent for up to 14 years, a longer period than for mostother reported clonal species, thereby facilitating ramet growthand establishment in a resource-limited environment. Chemical Composition of Bleeding Xylem Sap from Kiwifruit Vines   总被引:5,自引:0,他引:5  
A study of the chemical composition and charge balance was madeof bleeding xylem sap collected from excised one-year-old extensionshoots of healthy, Mn-deficient, Mn-toxic and Zn-deficient kiwifruitvines (Actinidia deliciosa (A. Chev.) C. F. Liang et A. R. Ferguson)immediately prior to leafburst. The exudates were analysed formacronutrient cations and anions, trace elements, amino acids,organic acids and sugars. Major charged species measured wereCa (13.3 mM), K (8.9 mM), Mg (5.6 mM), malate (12.5 mM) andphosphate (5.8 mM). Glutamine (12 mM) was the predominant Ncarrier identified, accounting for 58 per cent of the totalN followed by NO2-N (4.5 per cent), NH4+-N (3.5 per cent)and arginine-N (2.9 per cent). Approximately 22 per cent ofthe N was in a hydrolysable proteinaceous fraction comprisingmainly glutamine and glutamate. Eighteen free proteinaceousamino acids were idetified in sap, the most abundant being glutamine,glutamic acid, valine, isoleucine and phenylalanine. Computersimulation of the chemical composition predicted that in additionto hydrated cations, ion pairs formed between inorganic components(SO42–, HPO42–, H2PO4) and cations (Ca2+,Mg2+, Mn2+), plus metal-organic ligand complexes (Ca Malate,Zn Malate, FeCit, CuHis, CuGln) are important species involvedin translocation. The solubility product of hydroxyapatite wasexceeded in all exudates although in vitro precipitation wasnot observed. To achieve electroneutrality with the componentsmeasured, however, formation of precipitate precursors (hydroxyapatitenuclei) had to be assumed. Irregularities in Mn nutrition (butnot Zn) were clearly indicated by the elemental compositionof exudate suggesting the use of sap analysis as a possiblepre-season indicator of nutritional status for this species. The rate of carbon transport from an old tomato leaf (54 days),grown at 80 W m–2, was measured under light flux densitiesbetween 7 and 90 W m–2. Under low light, the rate of carbontransport over a 6 h period was about 1 mg C dm–2 h–1,well in excess of the concurrent photosynthetic rate. The lossfrom these leaves of 14C-leaf assimilate which was fixed beforethe experimental period amounted to 62 per cent of the totalinitial uptake and was higher than that from leaves with higherconcurrent photosynthetic rates. The higher loss of 14C fromleaves with low photosynthetic rates was due to a greater contributionof 14C from the starch and residue fractions. The rate of transportappeared to be determined by the concentration of the labilesucrose, not the total sucrose concentration. In comparisonwith young fully-expanded tomato leaves (Ho, 1976) the sizeof the labile sucrose pool appeared to decrease with age. Thephotosynthesistranslocation coefficient was low (k1k2=0•21)for an old tomato leaf. Based on these results a scheme of carbonpartitioning in relation to translocation is proposed. The specific respiration rates of nodulated root systems, ofnodules and of roots were determined during active nitrogenfixation in soya bean, navy bean, pea, lucerne, red clover andwhite clover, by measurements on whole plants before and afterthe removal of nodule populations. Similar measurements weremade on comparable populations of the six legumes, lacking nodulesbut receiving abundant nitrate-nitrogen, to determine the specificrespiration of their roots. All plants were grown in a controlled-environmentclimate which fostered rapid growth. The specific respiration rates of nodulated root systems ofthe three grain and three forage legumes during a 7–14-dayperiod of vegetative growth varied between 10 and 17 mg CO2g–1 (dry weight) h–1. This mean value consistedof two components: a specific root respiration rate of 6–9mg CO2 g–1 h–1 and a specific nodule respirationrate of 22–46 mg CO2 g–1 h–1. Nodule respirationaccounted for 42–70 per cent of nodulated root respiration;nodule weight accounted for 12–40 per cent of nodulatedroot weight. The specific respiration rates of roots lackingnodules and utilizing nitrate nitrogen were generally 20–30per cent greater than the equivalent rates of roots from nodulatedplants. The measured respiratory effluxes are discussed in thecontext of nitrogen nitrogen fixation, nitrate assimilation. Three-month ‘old’ and ‘fresh’ seedsof Talinum triangulare were subjected to various treatmentsto induce early and rapid germination. Scarification and activated carbon were the most effective treatmentsin improving total germination in fresh seeds, while the 3 and5 per cent thiourea treatments were most effective in improvingtotal germination in old seeds. Activated carbon, scarificationand 5 per cent thiourea treatments enhanced early germinationin both old and fresh seeds. Cumulative percentage germinationwas very high in fresh seeds after scarification or after treatmentwith activated carbon and 5 per cent thiourea, and lowest inseeds treated with 3 per cent thiourea and hot water. In oldseeds, highest cumulative percentage germination was obtainedwith 3 and 5 per cent thiourea treatments and scarification.Generally, higher germination was obtained with fresh seedsthan with old seeds. Partial seed-coat removal and treatment with 5 per cent thiouriaresulted in a higher rate of and cumulative percentage germinationcompared with seeds with the coat partially removed but nottreated with thiourea. Constantly high temperature (34 °C) increased both rateand total germination compared with seeds planted at room temperature(20–23 °C). Treatments that did not induce germinationwere 1 per cent thiourea, H2SO4, cold water soaking at roomtemperature, 6 per cent hydrogen peroxide and soil planting.These treatments effected less than 3 per cent germination. Respiratory activity and seasonal changes in carbohydrate contentof the storage organs of Rubus chamaemorus L. have been investigated.Leaf dark respiration rate increases in a non-linear mannerfrom 0·7 mg CO2 evolved dm–2 h–1 at 0 °Cto 4·6 rng CO2 evolved dm–2 hh–1 at 30 °C.Root and rhizome respiration rates increase from 1 µ1O2 uptake g–1 fresh weight h–1 at 0.7 ° C to10 µ10, uptake g–1 f. wt h–1 at 20 °C.Rhizome carbohydrate reserves decline from a September peakof 33 per cent alcohol insoluble d. wt to 16 per cent in May. Supplementing pollen suspension cultures with CO2 (3–5per cent) caused a marked increase in germination and tube growthin vitro in Brassica campestris L. cv. toria. A weakening ofself-incompatibility by increased CO2 levels from 3–5per cent was observed. The percentage of pollen tubes whichpenetrated the cuticle layer of stigmatic papilla cells in self-pollinatedpistils was high when CO2 level was 5 per cent. Phosphoenolpyruvate (PEP) carboxylase activity was greater in the pollengerminated in 4 per cent CO2 as compared to air (0.03 per cent).A possible role of CO2 for self-recognition and control of pollentube growth is proposed, proposed. Not all buds developed equally when 7-node rhizome fragmentsof Agropyron repens (L.) Beauv. were incubated in the dark at23 °C. Instead, after an initial flush of several shoots,buds were inhibited in a highly ordered sequence to leave onlyone dominant shoot growing. Applying an exogenous supply ofnitrogen KNO3) early during this sequence increased the meanshoot lengths and delayed the onset of dominance. Additionally,the application of nitrogen after eight days incubation alteredthe sequence of shoot growth such that, in some instances, smallrapidly-growing basal shoots ‘dominated’ largerand more slowly-growing apical ones. Dominance (correlativeinhibition) was maintained in untreated fragments for up to383 days Numbers of active budsand shoot extensionrate weremaximal intherange 13°to 23 °C where dominance was establishedwithin 30 days. In two experiments, the functioning and metabolism of nodulesof white clover, following a defoliation which removed abouthalf the shoot tissue, were compared with those of undefoliatedplants. In one experiment, the specific respiration rates of nodulesfrom undefoliated plants varied between 1160 and 1830 µmolCO2 g–1h–1, of which nodule ‘growth and maintenance’accounted for 22 ± 2 per cent, or 27 ± 3.6 percent, according to method of calculation. Defoliation reducedspecific nodule respiration and nodule ‘growth and maintenance’respiration by 60–70 per cent, and rate of N2 fixationby a similar proportion. The original rate of nodule metabolismwas re-established after about 5 d of regrowth; during regrowthnodule respiration was quantitatively related to rate of N2,fixation: 9.1 µmol CO2 µmol–1N2. With the possible exception of nodules examined 24 h after defoliation,the efficiency of energy utilization in nitrogenase functioningin both experiments was the same in defoliated and undefoliatedplants: 2.0±0.1 µmol CO2 µmol–1 C2H4;similarly, there was no change in the efficiency of nitrogenasefunctioning as rate of N2 fixation increased with plant growthfrom 1 to 22 µmol N2 per plant h–1. Exposure of nodulated white clover root systems to a 10 percent acetylene gas mixture resulted in a sharp peak in rateof ethylene production after 1.5–2.5 min; subsequently,rate of ethylene production declined rapidly before stabilisingafter 0.5–1 h at a rate about 50 per cent of that initiallyobserved. Regression of ‘peak’ rate of ethyleneproduction on rate of N2 fixation indicated a value of 2.9 µmolC2H4 µmol–1 N2, for rates of N2 fixation between1 and 22 µmol N2 per plant h–1. Carbon dioxide efflux from 5- to 20-day-old pea fruits was measuredfor plants grown in controlled environment at 15 °C and600 µmol s–1 m–2 photon flux density in a16 h photoperiod. The rate of CO2 output per fruit increasedquickly from 0.005 to 0.018 mg CO2 min–1 during fruitelongation and subsequently more slowly to 0.030 mg CO2 min–1as the fruits inflated. On a d. wt basis the rate was highest,0.175 mg CO2 g–1 min–1, in the youngest fruits anddeclined curvilinearly with increasing fruit weight to 0.02mg CO2 g–1 min–1. Separation of maintenance andgrowth components was achieved by starvation methods and bymultiple regression analysis. From the latter method estimatesof the maintenance coefficient declined hyperbolically from150±8.7 mg carbohydrate g–1 d. wt day–1 inthe very young fruits (0.05 g) to 10.4±0.36 mg carbohydrateg–1 d. wt day–1 in older fruits (2.0 g). On a nitrogenbasis maintenance costs decreased from 2240 to 310 mg carbohydrateg–1 nitrogen day–1 while nitrogen concentrationfell from 6.7 to 3 per cent d. wt. A simple linear relationshipbetween maintenance cost per unit d. wt and nitrogen concentrationwas not observed. A growth coefficient of 50±6.7 mg carbohydrate g–1growth (equivalent to a conversion efficiency, YG, of 0.95)was estimated for all fruits examined. The overall efficiency, Y, increased from a mean of 0.70 to0.85 during fruit elongation and subsequently declined to 0.80.For a given fruit weight, efficiency increased asymptoticallywith relative growth rate; both asymptote and slope of the relationshipincreased as the fruits grew. Sporangia of Phytophthora palmivora germinated by either forminggerm tubes or producing zoospores. Two distinct modes of germ-tubedevelopment have been described. Sporangia in distilled waterformed zoospores at 10–34°C with an optimum at 22°Cbut germinated by means of germ tubes at 30 and 34°C only.Zoospore formation was inhibited to varying degrees by cocoapod extract, I.0 per cent (w/v) peptone and yeast extract, 100–500µg1-1 thiamine, and by very low concentrations of severalamino acids, carbohydrates, and inorganic salts. From homogenates prepared from surface-sterilized nodules ofseedlings of Casuarina cunninghamiana grown aeroponically, astrain of Frankia designated HFPCc13 was isolated and has beengrown in pure filamentous culture in a defined synthetic nutrientmedium. Vesicle and sporangium formation can be induced by removalof combined nitrogen from the medium.Frankia strain HFPCc13nodulates young seedlings of C. cunninghamiana and C. equisetifoliawithin three weeks of inoculation with an optimum root mediumpH of 6–7 for nodulation and optimum temperature of 30–35°C. The presence of combined nitrogen in the root mediuminhibits nodulation with NH4+ more inhibitory than NO3.Frankia HFPCc13 does not nodulate Allocasuarina species withinthe same family nor several other possible actinorhizal plantstested. Thus this strain is quite precise in its host specificity.The rate of acetylene reduction was greater in C. cunninghamianathan the closely related species C. equisetifolia. In neitherof these host species were vesicles observed to occur withinthe infected root nodules which had been demonstrated to beactively fixing dinitrogen. Root nodules were shown to be activein acetylene reduction over a range of O2 concentration in thegaseous environment with an optimum at about 20 per cent O2,the ambient PO2 of the air. The relationship between the induction of tracheary elementdifferentiation and exogenous L-methionine was examined in agar-growncultures of soya bean callus initiated from Glycine max L. ‘Wayne’and ‘Clark 63’. Although Wayne is a normal cultivarsoya bean, seedlings of Clark 63 exhibit abnormal growth at25 °C due to exessive ethylene biosynthesis at this temperature.Wayne callus showed increased xylogenesis in the presence ofexogenous L-methionine (3.7 µg 1–1) in comparisonto IAA–KN controls at both 20 and 25 °C. Clark 63callus produced greater numbers of tracheary elements in responseto exogenous L-methionine only at 25 °C. The induction ofxylem differentiation was independent of the maintenance temperatureof the stock cultures of both cultivars. Xylogenesis initiatedbyan IAA–KN medium was inhibited by the addition of AgNO3(20 mg 1–1) to the extent of 76.5 per cent in cv. Wayneand 6 per cent in cv. Clark 63. The inhibitory effect was partiallyreversed by the addition of L-methionine (3.7 µg 1–1)to the IAA–KN–AgNO2 medium. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown for 71 d in flowing nutrient solutions containingN as 10 mmol m–3 NH4NO3, under artificial illumination,with shoots at 20/15°C day/night temperatures and root temperaturereduced decrementally from 20 to 5°C. Root temperatureswere then changed to 3, 7, 9, 11, 13, 17 or 25°C, and theacquisition of N by N2 fixation, NH4+ and NO3 uptakewas measured over 14 d. Shoot specific growth rates (d. wt)doubled with increasing temperature between 7 and 17°C,whilst root specific growth rates showed little response; shoot:root ratios increased with root temperature, and over time at11°C. Net uptake of total N per plant (N2 fixation + NH4++ NO3) over 14 d increased three-fold between 3 and 17°C.The proportion contributed by N2 fixation decreased with increasingtemperature from 51% at 5°C to 18% at 25°C. Uptake ofNH4+ as a proportion of NH4+ + NO3 uptake over 14 d variedlittle (55–62%) with root temperature between 3 and 25°C,although it increased with time at most temperatures. Mean ratesof total N uptake per unit shoot f. wt over 14 d changed littlebetween 9 and 25°C, but decreased progressively with temperaturebelow 9°C, due to the decline in the rates of NH4+ and NO3uptake, even though N2 fixation increased. The results suggestthat N2 fixation in the presence of sustained low concentrationsof NH4+ and NO4 is less sensitive to low root temperaturethan are either NH4+ or NO3 uptake systems. The influence of salinity in the growing media on ribulose-1,5-bisphosphate (RuBP) carboxylase and on CO2 fixation by intactsugar beet (Beta vulgaris) leaves was investigated. RuBP carboxylase activity was mostly stimulated in young leavesafter exposure of plants for 1 week to 180 mM NaCl in the nutrientsolution. This stimulation was more effective at the higherNaHCO2 concentrations in the reaction medium. Salinity also enhanced CO2 fixation in intact leaves mostlyat rate-limiting light intensities. A 60 per cent stimulationin CO2 fixation rate was obtained by salinity under 450 µEm–2 s–1. At quantum flux densities of 150 µEm–2 s–1 (400–700 nm) this stimulation was280 per cent. Under high light intensities no stimulation bysalinity was found. In contrast, water stress achieved by directleaf desiccation or by polyethylene glycol inhibited enzymeactivity up to fourfold at –1.2 MPa. Lolium temulentum plants were grown at 20 °C, under an 8-hdaylength, in a controlled-environment chamber, and the kineticsof leaf expansion were observed by measuring the movement ofan optical grid attached to the fourth leaf. The leaf emerged23–24 d after sowing and was fully expanded 9–10d later. Extension rate was maximal between the second and fifthdays after emergence and declined markedly thereafter. Duringthe rapid growth phase the rate of elongation exhibited a distinctdiurnal rhythm, fluctuating between 1.9 to 2.3 mm h–1in the light period, and 1.3 to 1.7 mm h–1 in the dark.A circadian oscillation with a period of about 27 h was observedin leaves elongating in continuous darkness. When plants weretransferred to 5 °C soon after emergence of the fourth leafthere was an immediate reduction in rate of growth to about22 per cent of the rate at 20 °C: the Q10 for the mean elongationrate in the range 20–5 °C was 3.7. When plants weretransferred from 20 to 2 °C at fourth leaf emergence, meanextension rate declined to less than 5 per cent, correspondingto a Q10 in the range 5–2 °C of more than 300. Furthermore,growth at 2 °C was confined almost entirely to the darkphase of the photoperiod cycle. The responsive tissue was shownto be a small area of expanding leafless than 1.5 cm above theshoot apex and the possible mechanisms underlying low temperatureeffects in this region are discussed. Of the 560 leaf samples belonging to 259 species of green plantsexamined more than 50 per cent of the Angiosperms and 25 percent of the Pteridophytes and Gymnosperms revealed the presenceof N2-fixing micro-organisms in their phyllosphere. Plants particularlyremarkable in this respect are orchids and several other epiphytes,Scindapsus officinalis, Ficus and cucurbits. Most of the isolatesappear to be biotypes of Klebsiella pneumoniae. The more activestrains fixed more than 5 mg N g–1 glucose utilized andreduced more than 100 nmol C2H2 mg–1 cell d. w h–1. The efficacy of the phyllosphere N2-fixing isolates for N-nutritionof host plants was studied by spraying suspensions of the culturesgrown on N-free media on rice and wheat seedlings. In IR-26rice or Sonalika and Janak wheat grown on soil in wooden flatsor earthenware pots, 22 per cent of the 161 cultures studiedcaused increased height and about three-quarters of the culturesenhanced dry weight by more than 50 per cent; chlorophyll andN-contents were enhanced more than 50 per cent by about halfand two-thirds of the cultures respectively. In N-free sandculture 26 of the 50 promising strains doubled N-content, and30 doubled dry weight of the tested plants. In some cases dryweight, number of grains per panicle, and 1000 grain weightwere increased by 300, 70–83 and 126–158 per centrespectively; N-content of straw and seed was increased three-or fourfold. In several cases the beneficial effects were foundto match closely the performance of plants receiving ammoniumsulphate. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown as simulated swards for 71 d in flowing nutrientsolutions with roots at 11 C and shoots at 20/15 C, day/night,under natural illumination. Root temperatures were then changedto 3, 5, 7, 11, 13, 17 or 25 C and the total N2, fixation over21 d was measured in the absence of a supply mineral N. Alltreatments were subsequently supplied with 10 mmol m–2NO2 in the flowing solutions for 14 d, and the relativeuptake of N by N2, fixation and NO3 uptake was compared.Net uptake of K+ was measured on a daily basis. Root temperature had little effect on root d. wt over the 35-dexperimental period, but shoot d. wt increased by a factor of3.5 between 3 and 25 C, with the sharpest increase occurringat 7–11 C. Shoot: root d. wt ratios increased from 25to 68 with increasing temperature at 7–25 C. N2-fixationper plant (in the absence of NO2 ) increased with roottemperature at 3–13C, but showed little change above13 C. The ratios of N2 fixation: NO2 uptake over 14d (mol N: mol N) were 0.47–0.77 at 3–7 C, 092–154at 11–17 C, and 046 at 25 C, reflecting the dominanceof NO3 uptake over N2 fixation at extremes of high andlow root temperature. The total uptake of N varied only slightlyat 11–25 –C (095–110 mmol N plant–1),the decline in N2 fixation as root temperature increased above11 C was compensated for by the increase in NO 3 uptake.The % N in shoot dry matter declined with decreasing root temperature,from 32% at 13 C to 15% at 3 C. In contrast, concentrationsof N expressed on a shoot water content basis showed a modestdecrease with increasing temperature, from 345 mol m–3at 3 C to 290 mol m–3 at 25 C. Experiments were made to determine the extent of reciprocaltransfer of products (derived from the assimilation of 14CO2)between various parts of the young vegetative grass plant (Loliummultiflorum Lam.). When individual laminae on different tillerswere supplied with 14CO2, 47–64 per cent of the fixedcarbon was exported after 24 h. The principal sinks were theroot system and the shoot or tiller to which the fed leaf wasattached. Other tillers also received significant quantitiesof radiocarbon. When whole tillers were supplied with 14CO2the percentage of fixed radiocarbon exported within 24 h rangedfrom 14–31 per cent. Of this, 50–74 per cent wasrecovered from the root system (except in the case of exportfrom the youngest tiller) but exchange of material between tillersalso occurred.A reciprocity diagram is presented and it is concludedthat despite the magnitude of the exchange no tiller showedan over-all net gain or loss. The main shoot and the tillersdiffered in the extent of their carbon exchange and in theirdegree of independence. The oldest daughter tiller of the mainshoot was the most independent and the main shoot most interdependent.  相似文献
