The gene polymorphisms interferon-gamma (IFN-γ) +874 T/A and interleukin (IL)-4 −590 C/T have been associated with the altered production of cytokines. Therefore, they might be indicative of the occurrence of Paracoccidioidomycosis (PCM) caused by Paracoccidioides brasiliensis. The analysis of single nucleotide polymorphism (SNP) at position +874 IFN-γ showed an increase occurrence of A/T genotype in both PCM patients and healthy individuals as control (HIC) (56% and 45%, respectively), while the allelic distribution showed 82% of A allele in the patients and 80% in the controls. The SNP of −590 IL-4 showed that C/T genotype was significantly (p < 0.05) more prevalent (39%) in PCM group compared to the HIC group (19%), while IL-4 C/C genotype was significantly less frequent (59%) in the patient group compared to the control group (81%). Otherwise, 41% of PCM patients and 19% of HIC individuals carried the IL-4 T allele. Stimulation of peripheral blood mononuclear cells (PBMC) from PCM patients with cell extract antigenic preparations (PbAg) as well as secreted and surface antigens (MEXO) of P. brasiliensis evidenced that there is no difference in the IFN-γ production related to A and T alleles between PCM and HIC individuals. However, with IL-4 production, PCM patients classified as C phenotype showed two times more IL-4 production than PCM patients classified as T phenotype and HIC controls. In conclusion, our results suggest that functional genetic variants in the IL-4 promoter could influence the production of IL-4 in PCM. 相似文献

Paracoccidioides brasiliensis is a temperature-dependent dimorphic fungus and the agent of paracoccidioidomycosis (PCM), which is prevalent in rural workers
of Latin American countries. Until a decade ago, most of the studies involving P. brasiliensis used clinical isolates, since environmental samples from soil are difficult to obtain. More recently, P. brasiliensis has been isolated from infected wild and domestic animals, especially from the nine-banded armadillo Dasypus novemcinctus in Brazil. Over the years, diversity within the species has been observed at several phenotypic levels. The present review
will discuss the reports focusing on genetic polymorphism, which culminated with the detection of P. brasiliensis phylogenetic species as a result of a multilocus study. Polymorphism in the PbGP43 gene is detailed. This gene encodes fungal glycoprotein gp43, a dominant P. brasiliensis antigen largely studied in the last two decades for its importance in diagnosis, immune protection, and adhesive properties

In this paper we relate that sera from paracoccidioidomycosis patients inhibited the mitogen-induced proliferative responses of normal mononuclear cells. Treatment of these sera with 2.5% polyethyleneglycol (PEG), a method classically used to precipitate immune complexes, significantly reduced their inhibitory activity. Immunoblot analysis of the PEG precipitates identified a 34-kDa polypeptide, recognized by rabbit anti-P. brasiliensis IgG. Patient mononuclear cells showed partial restoration of their proliferative capacity after 24 h culture in medium alone, which suggests release of membrane-bound molecules in the culture medium. These findings indicate that circulating P. brasiliensis antigens, complexed or not with antibodies, may play a negative immunoregulatory effect in the mitogen-induced proliferative responses of paracoccidioidomycosis patients.Abbreviations CIC

A locus is described that controls levels of mitochondrial dihydroorotate dehydrogenase (EC 1.3.3.1) in Drosophila melanogaster. The effects of alleles of the locus, Dhod, are manifest in preparations from whole organisms as well as in partially purified mitochondrial preparations; however, other mitochondrial functions do not appear to be appreciably affected by Dhod genotypes. The locus maps near p in the proximal portion of the right arm of chromosome 3. Flies trisomic for a chromosome segment including that region display elevated enzyme levels, implying that an enzyme structural gene is in that vicinity. Furthermore, Dhod alleles are semidominant in heterozygotes, suggesting that the dosage-sensitive element detected in the trisomics is actually the Dhod locus. These findings are discussed relative to the role of dihydroorotate dehydrogenase in the de novo pyrimidine biosynthetic pathway and relative to other pathway mutants that have been described in Drosophila.This work was supported by NSF Grants PCM 76-17214 to W. The immune reconstitution inflammatory syndrome (IRIS) is a deregulated inflammatory response to invading microorganisms. It is manifested when there is an abrupt change in host immunity from an anti-inflammatory and immunosuppressive state to a pro-inflammatory state as a result of rapid depletion or removal of factors that promote immune suppression or inhibition of inflammation. The aim of this paper is to discuss and re-interpret the possibility of association of paracoccidioidomycosis (PCM) with IRIS in the central nervous system (CNS) in a case from Brazil published by Silva-Vergara ML. et al. (Mycopathologia 177:137–141, 6). An AIDS patient who was not receiving medical care developed pulmonary PCM successfully treated with itraconazole. The patient developed central nervous system PCM (NPCM) after starting the ARV therapy with recovery of immunity and control of HIV viral load, although it was not interpreted as IRIS by the authors, it fulfills the criteria for CNS IRIS. This could be the first case of NPCM associated with IRIS described. Although not frequent, IRIS must be considered in PCM patients and HIV, from endemic areas or patients that traveled to endemic areas, receiving ARV treatment and with worsening symptoms. 相似文献

闽江口芦苇与短叶茳芏湿地土壤无机硫形态分布特征及其影响因素 总被引:3,自引:0,他引:3
选择闽江口鳝鱼滩的芦苇湿地、短叶茳芏湿地以及二者交错带湿地为对象,研究了不同类型湿地土壤无机硫赋存形态的分布特征及其主要影响因素。结果表明,芦苇与短叶茳芏在空间扩展过程中形成的交错带湿地土壤的水溶性硫(H_2O-S)、吸附性硫(Adsorbed-S)和盐酸可溶性硫(HCl-Soluble-S)含量在0—40 cm土层上整体要高于芦苇湿地和短叶茳芏湿地,而盐酸挥发性硫(HCl-Volatile-S)含量则与之相反。在垂直方向上,不同类型湿地土壤的H_2O-S、Adsorbed-S和HCl-Soluble-S含量整体均自表层向下呈先降低而后增加的变化,而盐酸挥发性硫(HCl-Volatile-S)含量则与之相反。3种类型湿地土壤中不同形态无机硫平均含量整体均表现为HCl-Soluble-S H_2O-S Adsorbed-S HCl-Volatile-S,且其总无机硫(TIS)平均含量分别占全硫(TS)含量的22.29%—39.99%(芦苇湿地)、32.39%—33.33%(交错带湿地)和30.20%—30.86%(短叶茳芏湿地)。与芦苇湿地和短叶茳芏湿地相比,交错带湿地土壤的H_2O-S、Adsorbed-S、HCl-Soluble-S和TIS平均含量整体均呈增加趋势,增幅分别为36.94%和54.31%、34.84%和13.03%、73.25%和67.59%以及45.72%和45.28%;与之不同,HCl-Volatile-S平均含量则呈降低趋势,降幅分别为2.78%和22.24%。不同类型湿地土壤中H_2O-S、Adsorbed-S和TIS含量的变化主要受有机质含量和细颗粒组成的控制,而HCl-Soluble-S和HCl-Volatile-S含量主要受土壤氧化还原环境和金属元素分布的影响。研究发现,芦苇与短叶茳芏的空间扩展明显增加了交错带湿地土壤的H_2O-S、Adsorbed-S、HCl-Soluble-S和TIS含量,但降低了HCl-Volatile-S含量,说明二者的空间扩展在整体提高交错带湿地土壤有效硫供给能力的同时,亦降低了挥发性硫化物可能对二者生长产生的不利影响,这对于保持二者在交错带湿地中的竞争力具有重要生态意义。 相似文献

Paracoccidioidomycosis (PCM) is a systemic mycosis caused by the fungus Paracoccidioides brasiliensis (Pb). The cyclosporin A (CsA) is an immunosuppressant drug that inhibits calcineurin and has been described as a potential
antifungal drug. The present study investigated the effect of CsA on the immune response, fungal load/antigenemia in experimental
murine PCM. It was used four groups of BALB/c mice: (a) infected with 1 × 105 Pb18 yeast cells (Pb), (b) infected and treated with CsA every other day 10 mg/kg of CsA (s.c.) during 30 days (Pb/CsA),
(c) treated with CsA (CsA) and (d) no infected/treated (PBS). The immune response was evaluated by lymphocyte proliferation,
DTH assays to exoAgs, ELISA for IgG anti-gp43 (specific immune responses) and cytokine serum levels (IFN-γ, TNF-α, IL-4 and
IL-10). Fungal load was determined by lung colony-forming units (CFU) counts, lung and liver histopathology analysis and antigenemia
determined by inhibition-ELISA. As expected, CsA was able to inhibit the specific cellular and humoral immune response (P < 0.05), with decrease in serum IFN-γ, TNF-α and IL-4 levels (P < 0.05). Cyclosporin A treatment also resulted in significantly decreased lung Pb CFU (P < 0.05) as well as a lower number of yeasts in the lung and liver (P < 0.05) by histopathology. In concordance, the decreased antigenemia was observed in Pb/CsA group (P < 0.05). In conclusion, even with immunosuppressive action, treatment with CsA results in decreased lung fungal load/antigenemia
in experimental PCM in BALB/c mice. Further study is required to determine whether this represents less severe disease or

Paracoccidioidomycosis (PCM) is a systemic mycosis caused by Paracoccidioides brasiliensis, which is endemic in many regions of Latin America. We describe the case of a 60-year-old man from a region endemic for PCM
who presented with a long history of left hip pain. He had been treated over the past 3 years with immunosuppressive drugs
(methotrexate, leflunomide, and adalimumab) for rheumatoid arthritis (RA). A hip radiograph showed lytic bone lesions, and
a chest radiograph showed an expansive excavated lesion in the left lung, suggestive of a lung cancer with bone metastases.
A left hip joint biopsy was inconclusive, but histological analysis of a surgical lung biopsy specimen was consistent with
P. brasiliensis infection. Treatment with intravenous amphotericin B (50 mg/day) and hydrocortisone (25 mg/day) was initiated. However, increasing
hip pain resulted in the amputation of the left lower limb, and the analysis of the surgical specimen revealed a diagnosis
of bone sarcoma. Postoperatively, the patient developed sepsis and died approximately 1 month later. To our knowledge, this
is the first report of PCM in a patient with RA who had been treated with immunosuppressive drugs, in particular TNF-α blocking
agents. The atypical presentation (left hip pain alone) emphasizes the importance of considering PCM in the differential diagnosis
of patients with pulmonary lesions and osteolytic lesions who live in a region endemic for PCM. This case report also demonstrates
that health professionals in these regions must pay close attention to patients receiving immunosuppressive drugs because
of the possibility of reactivating quiescent P. brasiliensis lesions. 相似文献

Though lacking adaptive immunity, insects possess a powerful innate immune system, a genome-encoded defence machinery used to confront infections. Studies in the fruit fly Drosophila melanogaster revealed a remarkable capacity of the innate immune system to differentiate between and subsequently respond to different bacteria and fungi. However, hematophagous compared to non-hematophagous insects encounter additional blood-borne infectious agents, such as parasites and viruses, during their lifetime. Anopheles mosquitoes become infected with the malaria parasite Plasmodium during feeding on infected human hosts and may then transmit the parasite to new hosts during subsequent bites. Whether Anopheles has developed mechanisms to confront these infections is the subject of this review. Initially, we review our current understanding of innate immune reactions and give an overview of the Anopheles immune system as revealed through comparative genomic analyses. Then, we examine and discuss the capacity of mosquitoes to recognize and respond to infections, especially to Plasmodium, and finally, we explore approaches to investigate and potentially utilize the vector immune competence to prevent pathogen transmission. Such approaches constitute a new challenge for insect immunity research, a challenge for global health. 相似文献

Paracoccidioidomycosis (PCM) is a severe systemic mycosis, endemic in Latin America and highly prevalent in Brazil, where
it ranks eighth as a mortality cause among infectious and parasitic diseases in humans. The disease in animals has been little
explored. It is observed that armadillos can harbor the fungus at high frequencies, although the active disease has not been
well documented in this wild mammal. Dogs are susceptible to experimental infection, and the naturally acquired PCM-disease
was reported only recently in a dog from Brazil. The present work reports the second case of naturally acquired PCM in a 6-year-old
female dog that presented emaciation, lymphadenomegaly, and hepatosplenomegaly. Biochemical and pulmonary radiographic evaluation
did not reveal any abnormalities. PCM was diagnosed by clinical findings, culturing, immunohistochemistry, and histopathology
of popliteal lymph node. The fungus was recovered from popliteal lymph node, and the molecular analysis showed respective
sequencing similarities of 99 and 100% for 803 nucleotides of the Gp43 gene and 592 nucleotides from the ITS-5.8S region of Paracoccidioides brasiliensis. Immunohistochemistry revealed severe lymphadenitis and presented numerous yeasts, which reacted against the gp43 antibody.
Histopathology revealed a severe granulomatous lymphadenitis associated with numerous single or multiple budding yeasts. After
diagnosis, the dog was successfully treated with itraconazol for 2 years. Veterinarians should be aware of the importance
of considering PCM for differential diagnosis, especially in dogs from PCM-endemic areas, whose monophagocytic system involvement

Cryptococcosis as an opportunistic infection in immunodeficiency secondary to paracoccidioidomycosis

We describe the case reports of two patients with immunodeficiency secondary to paracoccidioidomycosis (PCM) and opportunistic Cryptococcus neoformans infections. Secondary immunodeficiency likely occurred as a consequence of the intestinal loss of proteins and lymphocytes associated with malabsorption syndrome due to obstructed lymphatic drainage. Both patients had had severe abdominal involvement during the acute PCM disease. Immunological evaluation showed cellular and humoral immunity impairment. Cryptococcosis manifested as relatively well circumscribed lesions: osteolytic lesions of the skull in one patient, and pulmonary nodules in the other. The latter was treated surgically and with amphotericin B, whereas the other was treated with the combination amphotericin-B and flucytosine. Both patients had a good response to treatment with complete regression of the lesions. They have now 2 and 4 years of follow-up with maintenance therapy and no indication of reactivation of the infection. PCM also did not reactivate. The clinical and immunological characteristics of these patients are discussed and compared to the opportunistic C. neoformans infections of AIDS and transplant patients. 相似文献

Purification and Characterization of Two Distinct Isozymes of Protein Carboxymethylase from Bovine Brain 总被引:5,自引:2,他引:3
Protein carboxymethylase (EC 2.1.1.24) from cytosol of bovine brain was found to exist as two apparent isozymes that could be separated by chromatography on DEAE-cellulose at pH 8.O. Rechromatography of the two forms, designated PCM I and PCM II, indicated that they are not interconvertible. Both enzymes have a molecular weight of 24,300 by sodium dodecyl sulfate-polyacryl-amide gel electrophoresis. PCM I consists mainly of one isoelectric form, pI 6.5, whereas PCM II resolves into two forms of pI 5.6 and 5.7. The relative amounts of PCM I and PCM II show a marked tissue dependence. Brain has approximately twice as much PCM I as II, whereas liver contains only the type II enzyme. The two enzymes were found to have similar substrate specificities when tested with five different methyl-accepting proteins. Synapsin I, a basic protein associated with synaptic vesicles, was found to be an excellent methyl-accepting protein with regard to its Km (1.2 μM), but it exhibited a low stoichiome-try of methyl incorporation. 相似文献

Soluble starch branching enzymes and starch synthases from maize kernels of differing dosage of the ae locus were purified by DEAE-cellulose chromatography. A near-linear relationship between increasing dosage of the dominate amylose-extender allele (Ae) and branching enzyme IIb activity was found. In contrast, levels and properties of branching enzymes I and IIa, as well as the citrate-stimulated and primer-requiring starch synthases, remained unchanged. A genetically variable naphthylamidase enzyme, previously described as leucine aminopeptidase, was purified approximately fiftyfold, and its biochemical properties were investigated. The enzyme was renamed aminopeptidase I. Substrate affinities demonstrate that it is an -aminoacyl peptide hydrolase (E.C. 3.4.11.-). Aminopeptidase I had a monomer molecular weight of 65–68,000, average pI of pH 4.88, and broad pH optima between 6.5 and 8.0. The enzyme was inactivated rapidly between 40 and 50 C. Antibodies from purified enzyme did not cross-react with other naphthylamidases, but aminopeptidase I activity was inhibited by immune serum. The enzyme exhibited highest naphthylamidase activity for aromatic and hydrophobic aminoacyl naphthylamides. Aminopeptidase activity was highest for aromatic and hydrophobic N-terminal residues of tripeptides. Certain divalent metal cations, p-O H-mercuribenzoate, and N-ethylmaleimide were strongly inhibitory while chelating agents activated the enzyme.This work was supported by NIH Grant GM-21133, NSF Grant DEB 77-06074, USPHS Career Award GM-28963 to R. The present study investigated the protective activity of the P. brasiliensis high-molecular-mass (hMM) fraction (~380 kDa) in experimental murine PCM. In the first step, lymphocyte proliferation and
production of IFNγ (but not IL-4) were observed in “in vitro” spleen cells (from female BALB/c mice infected (i.v.) with P. brasiliensis) that were stimulated with hMM fractions. In the second step, female BALB/c mice were previously immunized (s.c.) with hMM
fraction (25 μg/protein = F-25 and 50 μg/protein = F-50), and the colony-forming units (CFU) of the lung and spleen, the histopathological
characteristics of the granulomatous lesions, and plasmatic gp43 soluble antigens and anti-hMM IgG levels were analyzed at
28 and 56 days after infection. The lung and liver CFU were lower in mice previously immunized with the hMM fraction (P < 0.05). The granulomatous lesions revealed a greater degree of compaction and organization, with no dissemination of the
fungus to other organs. Lower soluble antigen levels (P < 0.05) and higher IgG anti-hMM fraction (P < 0.05) were observed in immunized groups. The results for CFU, histopathology and antigenemia suggest that the hMM fraction

Glutamine-dependent CPSase, ATCase, and DHOase from Drosophila, the first three enzymes in pyrimidine biosynthesis, show coordinate variation in activity throughout development. The three activities were highest in first instar larvae and decreased as development proceeded. The three activities cosediment in sucrose gradients as a single peak with a relative sedimentation coefficient of approximately 30S. CPSase, ATCase, and DHOase copurify during (NH4) 2SO4 fractionation and during DEAE-cellulose and hydroxylapatite chromatography.This work was supported by a grant from the National Science Foundation (No. PCM75-22802).
